Abstract Background The growing body of research on kidney disease in children has identified a broad spectrum of genetic etiologies. Methods We conducted a prospective study to evaluate the efficacy of an optimized genetic test and subclinical changes in a real-world context before kidney transplantation. All cases involved recipients under the age of 18 who underwent whole exome sequencing (ES) between 2013 and 2022. Results The study population included 244 children, with a median age of 13.1 years at transplantation. ES provided a molecular genetic diagnosis in 114 (46.7%) probands with monogenic variants in 15 known disease-causing genes. ES confirmed the suspected clinical diagnosis in 74/244 (30.3%) cases and revised the pre-exome clinical diagnoses in 40/244 (16.4%) cases. ES also established a specific underlying cause for kidney failure for 19 patients who had previously had an unknown etiology. Genetic diagnosis influenced clinical management in 88 recipients (36.1%), facilitated genetic counseling for 18 families (7.4%), and enabled comprehensive assessment of living donor candidates in 35 cases (14.3%). Conclusions Genetic diagnosis provides critical insights into the pathogenesis of kidney disease, optimizes clinical strategies concerning risk assessment of living donors, and enhances disease surveillance of recipients.

Background and aim: Type-2 diabetes mellitus (T2DM) is mainly characterized by insulin resistance (IR) induced by hyperglycaemia and insufficient insulin secretion. We employed a diabetic fly model to examine the effect and molecular mechanism of Atractylodes macrocephala Koidz. and Cuscuta chinensis Lam. (AMK–CCL) extract as traditional Chinese medicine in treating IR and T2DM. Experimental procedure: The contents of the active ingredients (rhamnose, xylose, mannose, and hyperoside) in AMK–CCL extract were determined by high-performance liquid chromatography. Wild-type (Cg-GAL4/+) or diabetic (Cg > InRK1409A) Drosophila flies were divided into the control group or metformin group and AMK–CCL (0.0125, 0.025, 0.05, 0.1 g/ml) groups. Food intake, haemolymph glucose and trehalose, protein, weight, triglycerides (TAG), and glycogen were measured to assess glycolipid metabolism. Phosphatidylinositol-3-kinase (PI3K)/Akt signalling was detected using fluorescent reporters [tGPH, Drosophila forkhead box O (dFoxO)–green fluorescent protein (GFP), Glut1–GFP, 2-NBDG] in vivo. Glut1/3 mRNA levels and Akt phosphorylation levels were detected by quantitative polymerase chain reaction and western blotting, respectively, in vitro. Results: AMK–CCL extract contained 0.038 % rhamnose, 0.017 % xylose, 0.69 % mannose, and 0.039 % hyperoside. AMK–CCL at 0.0125 g/mL significantly suppressed the increase in circulating glucose, and the decrease in body weight, TAG, and glycogen contents of diabetic flies. AMK–CCL improved PI3K activity, Akt phosphorylation, Glut1/3 expression, and glucose uptake in diabetic flies, and also rescued diabetes-induced dFoxO nuclear localisation. Conclusions: These findings indicate that AMK–CCL extract ameliorates IR-induced diabetes via the PI3K/Akt signalling pathway, providing an experimental basis for clinical treatment.

Abstract Photothermal therapy (PTT) is a promising cancer treatment method due to its ability to induce tumor-specific T cell responses and enhance therapeutic outcomes. However, incomplete PTT can leave residual tumors that often lead to new metastases and decreased patient survival in clinical scenarios. This is primarily due to the release of ATP, a damage-associated molecular pattern that quickly transforms into the immunosuppressive metabolite adenosine by CD39, prevalent in the tumor microenvironment, thus promoting tumor immune evasion. This study presents a photothermal nanomedicine fabricated by electrostatic adsorption among the Fe-doped polydiaminopyridine (Fe-PDAP), indocyanine green (ICG), and CD39 inhibitor sodium polyoxotungstate (POM-1). The constructed Fe-PDAP@ICG@POM-1 (FIP) can induce tumor PTT and immunogenic cell death when exposed to a near-infrared laser. Significantly, it can inhibit the ATP-adenosine pathway by dual-directional immunometabolic regulation, resulting in increased ATP levels and decreased adenosine synthesis, which ultimately reverses the immunosuppressive microenvironment and increases the susceptibility of immune checkpoint blockade (aPD-1) therapy. With the aid of aPD-1, the dual-directional immunometabolic regulation strategy mediated by FIP can effectively suppress/eradicate primary and distant tumors and evoke long-term solid immunological memory. This study presents an immunometabolic control strategy to offer a salvage option for treating residual tumors following incomplete PTT.

Better soil structure promotes extension of plant roots thereby improving plant growth and yield. Differences in soil structure can be determined by changes in the three phases of soil, which in turn affect soil function and fertility levels. To compare the quality of soil structure under different conditions, we used Generalized Soil Structure Index (GSSI) as an indicator to determine the relationship between the “input” of soil three phases and the “output” of soil structure. To achieve optimum monitoring of comprehensive indicators, we used Successive Projections Algorithm (SPA) for differential processing based on 0.0–2.0 fractional orders and 3.0–10.0 integer orders and select important wavelengths to process soil spectral data. In addition, we also applied multivariate regression learning models including Gaussian Process Regression (GPR) and Artificial Neural Network (ANN), exploring potential capabilities of hyperspectral in predicting GSSI. The results showed that spectral reflection, mainly contributed by long-wave near-infrared radiation had an inverse relationship with GSSI values. The wavelengths between 404-418 nm and 2193–2400 nm were important GSSI wavelengths in fractional differential spectroscopy data, while those ranging from 543 to 999 nm were important GSSI wavelengths in integer differential spectroscopy data. Also, non-linear models were more accurate than linear models. In addition, wide neural networks were best suited for establishing fractional-order differentiation and second-order differentiation models, while fine Gaussian support vector machines were best suited for establishing first-order differentiation models. In terms of preprocessing, a differential order of 0.9 was found as the best choice. From the results, we propose that when constructing optimal prediction models, it is necessary to consider indicators, differential orders, and model adaptability. Above all, this study provided a new method for an in-depth analyses of generalized soil structure. This also fills the gap limiting the detection of soil three phases structural characteristics and their dynamic changes and provides a technical references for quantitative and rapid evaluation of soil structure, function, and quality.

IntroductionCorrected QT interval (QTc) is a ventricular repolarization marker on electrocardiography. Previous studies evaluated its value in predicting atrial fibrillation (AF) occurrence. However, its predictive efficacy for AF recurrence remains controversial.MethodsWe searched PubMed and Google databases for studies before January 2024 evaluating the association between QTc interval and AF incidence. A meta-analysis of the eligible datasets was conducted using Bazett's formula, with subgroup analysis to explore the heterogeneity. Additionally, thirty-eight patients with AF who underwent radiofrequency catheter ablation were enrolled and followed-up for 3–36 months. Univariate and multivariate Cox models were used to calculate the hazard ratios (HRs) and determine the relationship between clinical factors and AF recurrence. Kaplan-Meier survival analysis and ROC curve were conducted to assess the impact and predictive efficacy of individual factors.ResultsEleven datasets from nine eligible studies were enrolled and meta-analysed. We found that patients with prolonged QTc interval was associated with a significantly higher AF incidence risk, and the risk increased with every 10-ms prolongation. However, this association was not significant in the AF recurrence subgroup. In our prospective cohort, the preoperative body mass index, QTc, left atrial diameter (LAD), and uric acid levels influenced AF recurrence. Multivariate Cox regression analysis identified LAD as an independent factor affecting AF recurrence in patients with a high predictive efficiency. Kaplan-Meier survival analysis showed that increased LAD (>4.5 cm) was associated with postoperative AF recurrence.DiscussionTherefore, LAD has better predictive power and can be an indicator for predicting postoperative AF recurrence.

ObjectiveThe aim of this study is to explore the relationship between ideal cardiovascular health (CVH), as assessed using the Life’s Essential 8 (LE8), and bowel conditions.MethodsThis cross-sectional study selected 11,108 participants aged ≥20 years from 2005 to 2010 National Health and Nutrition Examination Survey. LE8 scores (range 0–100) were measured according to American Heart Association definitions and were divided into health behavior and health factor scores. Bowel conditions including chronic diarrhea, constipation, and fecal incontinence were diagnosed by the Bowel Health Questionnaire. Weighted logistic regression and restricted cubic spline models were used for correlation analysis.ResultsLogistic regression results showed that LE8 scores were negatively associated with chronic diarrhea and fecal incontinence, but the difference with chronic constipation was not statistically significant. The health behaviors subscale was also negatively correlated with chronic diarrhea, chronic constipation, and fecal incontinence, but health factors were negatively related to chronic diarrhea and fecal incontinence and positively related to chronic constipation. The RCS was consistent with the trend of the logistic regression findings. Sensitivity analyses reconfirmed these outcomes.ConclusionLE8 is highly associated with chronic diarrhea and fecal incontinence, not with chronic constipation. Encouraging optimization of CVH levels may be beneficial for bowel disorders, and prevention of bowel disorders may enhance CVH.

Abstract Fusarium wilt is one of the major constraints on global watermelon production, and Fusarium oxysporum f. sp. niveum (Fon) is the causative agent of Fusarium wilt in watermelon and results in severe yield and quality losses worldwide. The enhancement of antifungal activity from antagonistic bacteria against Fon is highly practical for managing Fusarium wilt in watermelon. The aim of this study was to maximize the antifungal activity of Bacillus velezensis LZN01 by optimizing fermentation conditions and analysing its regulatory mechanism via transcriptome sequencing. The culture and fermentation conditions for strain LZN01 were optimized by single‐factor and response surface experiments. The optimum culture conditions for this strain were as follows: the addition of D‐fructose at 35 g/L and NH4Cl at 5 g/L in LB medium, pH 7, and incubation at 30°C for 72 h. The fungal inhibition rate for strain LZN01 reached 71.1%. The improvement of inhibition rate for strain LZN01 in optimization fermentation was supported by transcriptomic analysis; a total of 491 genes were upregulated, while 736 genes were downregulated. Transcriptome analysis revealed that some differentially expressed genes involved in carbon and nitrogen metabolism, oxidation–reduction, fatty acid and secondary metabolism; This optimization process could potentially lead to significant alterations in the production levels and types of antimicrobial compounds by the strain. Metabolomics and UPLC/Q‐Exactive Orbitrap MS analysis revealed that the production yields of antimicrobial compounds, such as surfactin, fengycin, shikimic acid, and myriocin, increased or were detected in the cell‐free supernatant (CFS) after the fermentation optimization process. Our results indicate that fermentation optimization enhances the antifungal activity of the LZN01 strain by influencing the expression of genes responsible for the synthesis of antimicrobial compounds.

ObjectiveTo evaluate the effectiveness of multimodal features based on gait analysis and eye tracking for elderly people screening with subjective cognitive decline in the community.MethodsIn the study, 412 cognitively normal older adults aged over 65 years were included. Among them, 230 individuals were diagnosed with non-subjective cognitive decline and 182 with subjective cognitive decline. All participants underwent assessments using three screening tools: the traditional SCD9 scale, gait analysis, and eye tracking. The gait analysis involved three tasks: the single task, the counting backwards dual task, and the naming animals dual task. Eye tracking included six paradigms: smooth pursuit, median fixation, lateral fixation, overlap saccade, gap saccade, and anti-saccade tasks. ObjectiveTo analyze the change trend of the disease burden of injuries among Chinese residents from 1990 to 2019, and to predict the age-standardized incidence, mortality, and disability-adjusted life-years (DALY) rates of injuries from 2020 to 2025, providing a reference for the prevention and control of injuries among the Chinese population. MethodsData on the disease burden of injury in populations in China, Asia, and the world from 1990 to 2019 were collected from the Global Burden of Disease Study – 2019 (GBD 2019). Crude and age-standardized rates of incidence, mortality, years lived with disability (YLD), years of life lost (YLL), and DALY of injury were used in the descriptive analysis. A joint-point regression model was constructed to analyze the average annual percentage change (AAPC) in these rates between 1990 and 2019. In addition, an autoregressive integrated moving average (ARIMA) model was constructed to predict the standardized rates of incidence, mortality, and DALY of injury from 2020 to 2025 in the Chinese population. ResultsFrom 1990 to 2019, increases in the crude and age-standardized rates (1/100 000) of injury incidence (5 976.31 to 6 328.60 and 5 863.81 to 6 138.73) and YLD (461.57 to 679.01 and 511.58 to 534.88) were observed in the Chinese population, with overall rate increases of 5. 89%, 4.69%, 47.11%, and 4.56%, respectively, and the rate increases were all higher than those in the Asian and global populations; however, decreases in the crude and age-standardized rates were observed for injury mortality (74. 34 to 49.67 and 83.03 to 44.20), YLL (4215.54 to 1825.22 and 4204.05 to 1787.50), and DALY (4677.11 to 2504.23 and 4715.63 to 2322.38), with overall decreases of – 33.19%, – 46.76%, – 56.70%, – 57.48%, – 46.46%, and – 50.75%, respectively, and the rate decreases were all greater than those in the Asian and global populations. The results of Joinpoint regression model analysis show that from 1990 to 2019, the age-standardized incidence and YLD rate of injuries in the Chinese population showed an overall increasing trend (AAPC = 0.15%, P = 0.006), while the age-standardized mortality, YLL, and DALY rate of injuries showed an overall decreasing trend (AAPC = – 2.28%, – 2.62%, and – 3.07%, all P < 0.01). The injury type that ranked first in terms of injury disease burden in the Chinese population in 1990 and 2019 was road traffic injuries, with DALYs of 13.73957 million person-years and 13.2577 million person-years, and the age-standardized DALY rates (1/100 000) of 12.327 2 and 8.595 8, respectively. For the Chinese population between 2020 and 2025, the predicted age-standardized injury incidence rate (1/100 000) ranges from 6 293.79 to 6 492.15; the predicted age-standardized mortality rate (1/100 000) ranges from 41.50 to 48.38; and the predicted age-standardized DALY rate (1/100 000) ranges from 1 850.11 to 2 234.510. The predicted age-standardized injury incidence rate for 2020 – 2025 is expected to continue to increase, while the predicted age-standardized mortality and DALY rates are expected to continue to decrease (all P < 0.05). ConclusionThe age-standardized incidence and YLD rates of injuries in the Chinese population showed an overall increasing trend from 1990 to 2019, while the age-standardized mortality, YLL, and DALY rates showed an overall decreasing trend. From 2020 to 2025, the age-standardized incidence rate of injuries will continue to increase, while the age-standardized mortality rate and DALY rate will continue to decrease.

To investigate the impact of Jolkinolide B (JB) on renal cell carcinoma (RCC), we treated RCC cell lines with various concentrations of JB (0, 5, 25, 50 and 100 μM) for 24 hours and assessed cell viability using the cell counting kit-8 (CCK-8) analysis. Then, we examined JB’s effects on proliferation, migration, apoptosis, reactive oxygen species (ROS) accumulation, and endoplasmic reticulum (ER) stress through 5-ethynyl-2′-deoxyuridine (EdU) incorporation, transwell migration assays, flow cytometry, ROS level determination, and western blot assays. Furthermore, we investigated the potential mechanism using western blot. Our results showed that JB dose-dependently reduced cell viability in both 786-O and Caki1 cells. Additionally, JB at concentrations of 5, 25 and 50 μM decreased the number of EdU-positive and migrating cells in both cell lines. Additionally, these concentrations of JB increased apoptosis rates, relative protein expressions of cleaved caspase-3 and cleaved caspase-9, ROS levels, and relative protein expressions of C/EBP-homologous protein (CHOP) and activated transcription factor 4 (ATF4) in both 786-O and Caki1 cells. Mechanistically, treatment with all three concentrations of JB significantly downregulated phosphorylated p38 (p-p38)/p38, phosphorylated protein kinase B (p-AKT)/AKT and phosphorylated phosphatidylinositol-3-kinase (p-PI3K)/PI3K in a dose-dependent manner. In summary, JB inhibited proliferation and migration while promoting apoptosis, ROS accumulation, and ER stress in RCC cells, potentially through the inactivation of mitogen-activated protein kinase (MAPK) and AKT signaling pathways.

Abstract. Background:. Growth retardation is a common complication of chronic kidney disease in children, which can be partially relieved after renal transplantation. This study aimed to develop and validate a predictive model for growth patterns of children with end-stage renal disease (ESRD) after kidney transplantation using machine learning algorithms based on genomic and clinical variables. Methods:. A retrospective cohort of 110 children who received kidney transplants between May 2013 and September 2021 at the First Affiliated Hospital of Zhengzhou University were recruited for whole-exome sequencing (WES), and another 39 children who underwent transplant from October 2021 to March 2022 were enrolled for external validation. Based on previous studies, we comprehensively collected 729 height-related single-nucleotide polymorphisms (SNPs) in exon regions. Seven machine learning algorithms and 10-fold cross-validation analysis were employed for model construction. Results:. The 110 children were divided into two groups according to change in height-for-age Z-score. After univariate analysis, age and 19 SNPs were incorporated into the model and validated. The random forest model showed the best prediction efficacy with an accuracy of 0.8125 and an area under curve (AUC) of 0.924, and also performed well in the external validation cohort (accuracy, 0.7949; AUC, 0.796). Conclusions:. A model with good performance for predicting post-transplant growth patterns in children based on SNPs and clinical variables was constructed and validated using machine learning algorithms. The model is expected to guide clinicians in the management of children after renal transplantation, including the use of growth hormone, glucocorticoid withdrawal, and nutritional supplementation, to alleviate growth retardation in children with ESRD.

As a cell proliferation regulator involved in wide biological processes in plants, GRF-INTERACTING FACTOR (GIF) controls different tissues development. However, whether GIF participates in fruit development remains unclear. According to transcriptome data, we identified PbGIF1 was highly expressed during fruit development in cytokinins induced parthenocarpy pear. In the present study, the biofunction of PbGIF1 was initially verified. Overexpression of PbGIF1 promoted fruit size of transgenic tomato. The size of flesh fruit was not affected by cell expansion but the cell proliferation was promoted by overexpressing PbGIF1. The accelerated cell proliferation process was also observed in PbGIF1-overexpressed transgenic pear fruit calli. The transcriptional regulation of cytokinins on PbGIF1 was further confirmed by exogenous CPPU treatments in pear fruitlets. To investigate the underlying mechanism, the cytokinins-responded factor, PbRR1, was further focused on. The results of Yeast-one-hybrid assay suggested that PbRR1 can bind to the promoter sequence of PbGIF1. The transcriptional activation of PbRR1 on PbGIF1 was also confirmed by Dual-Luciferase assays. Taken together, the results showed that cytokinins control pear fruit development via the transcriptional activation of PbGIF1 by PbRR1.

Abstract Background Dry Eye Disease (DED) is a prevalent multifactorial ocular disease characterized by a vicious cycle of inflammation, oxidative stress, and mitochondrial dysfunction on the ocular surface, all of which lead to DED deterioration and impair the patients’ quality of life and social functioning. Currently, anti-inflammatory drugs have shown promising efficacy in treating DED; however, such drugs are associated with side effects. The bioavailability of ocular drugs is less than 5% owing to factors such as rapid tear turnover and the presence of the corneal barrier. This calls for investigations to overcome these challenges associated with ocular drug administration. Results A novel hierarchical action liposome nanosystem (PHP-DPS@INS) was developed in this study. In terms of delivery, PHP-DPS@INS nanoparticles (NPs) overcame the ocular surface transport barrier by adopting the strategy of “ocular surface electrostatic adhesion-lysosomal site-directed escape”. In terms of therapy, PHP-DPS@INS achieved mitochondrial targeting and antioxidant effects through SS-31 peptide, and exerted an anti-inflammatory effect by loading insulin to reduce mitochondrial inflammatory metabolites. Ultimately, the synergistic action of “anti-inflammation-antioxidation-mitochondrial function restoration” breaks the vicious cycle associated with DED. The PHP-DPS@INS demonstrated remarkable cellular uptake, lysosomal escape, and mitochondrial targeting in vitro. Targeted metabolomics analysis revealed that PHP-DPS@INS effectively normalized the elevated level of mitochondrial proinflammatory metabolite fumarate in an in vitro hypertonic model of DED, thereby reducing the levels of key inflammatory factors (IL-1β, IL-6, and TNF-α). Additionally, PHP-DPS@INS strongly inhibited reactive oxygen species (ROS) production and facilitated mitochondrial structural repair. In vivo, the PHP-DPS@INS treatment significantly enhanced the adhesion duration and corneal permeability of the ocular surface in DED mice, thereby improving insulin bioavailability. It also restored tear secretion, suppressed ocular surface damage, and reduced inflammation in DED mice. Moreover, it demonstrated favorable safety profiles both in vitro and in vivo. Conclusion In summary, this study successfully developed a comprehensive DED management nanosystem that overcame the ocular surface transmission barrier and disrupted the vicious cycle that lead to dry eye pathogenesis. Additionally, it pioneered the regulation of mitochondrial metabolites as an anti-inflammatory treatment for ocular conditions, presenting a safe, efficient, and innovative therapeutic strategy for DED and other inflammatory diseases.

Abstract Kaempferol (KA), an natural antioxidant of traditional Chinese medicine (TCM), is extensively used as the primary treatment for inflammatory digestive diseases with impaired redox homeostasis. Severe acute pancreatitis (SAP) was exacerbated by mitochondrial dysfunction and abundant ROS, which highlights the role of antioxidants in targeting mitochondrial function. However, low bioavailability and high dosage of KA leading to unavoidable side effects limits clinical transformation. The mechanisms of KA with poor bioavailability largely unexplored, hindering development of the efficient strategies to maximizing the medicinal effects of KA. Here, we engineered a novel thioketals (TK)-modified based on DSPE-PEG2000 liposomal codelivery system for improving bioavailability and avoiding side effects (denotes as DSPE-TK-PEG2000-KA, DTM@KA NPs). We demonstrated that the liposome exerts profound impacts on damaging intracellular redox homeostasis by reducing GSH depletion and activating Nrf2, which synergizes with KA to reinforce the inhibition of inadequate fission, excessive mitochondrial fusion and impaired mitophagy resulting in inflammation and apoptosis; and then, the restored mitochondrial homeostasis strengthens ATP supply for PAC renovation and homeostasis. Interestingly, TK bond was proved as the main functional structure to improve the above efficacy of KA compared with the absence of TK bond. Most importantly, DTM@KA NPs obviously suppresses PAC death with negligible side effects in vitro and vivo. Mechanismly, DTM@KA NPs facilitated STAT6-regulated mitochondrial precursor proteins transport via interacting with TOM20 to further promote Drp1-dependent fission and Pink1/Parkin-regulated mitophagy with enhanced lysosomal degradation for removing damaged mitochondria in PAC and then reduce inflammation and apoptosis. Generally, DTM@KA NPs synergistically improved mitochondrial homeostasis, redox homeostasis, energy metabolism and inflammation response via regulating TOM20-STAT6-Drp1 signaling and promoting mitophagy in SAP. Consequently, such a TCM’s active ingredients-based nanomedicine strategy is be expected to be an innovative approach for SAP therapy. Graphical Abstract

BackgroundHealthy lifestyle is one of the important factors affecting individual health. How to promote residents' cognition of healthy lifestyle has become an urgent practical problem for the whole society. However, there is no residents' healthy lifestyle cognitive indexes available with consideration of carbon peaking and carbon neutrality. ObjectiveTo construct a cognitive index system for providing effective assessment on residents' cognition of healthy lifestyle under the background of carbon peaking and carbon neutrality. MethodsBased on the health belief model, a preliminary cognitive index system of residents' healthy lifestyle under the background of carbon peaking and carbon neutrality was proposed after literature study and relevant policy review. Then three rounds of indicator importance evaluation and screening by Delphi method were conducted before the index system was finally constructed. The weights of all levels of indicators were determined using analytic hierarchy process. In addition, a self-assessment questionnaire was developed based on the index system. The questionnaire was utilized to conduct a survey among 200 residents by convenience sampling in Shenyang, Liaoning Province from November to December 2021. The survey was used to examine the reliability and validity of the indicator system. ResultsThe effective recovery rates of the three rounds of Delphi method were all 100%. Abstract Effective conflict management is pivotal for achieving innovation performance within standardization alliances, particularly those for the Belt and Road Initiative (B&R Initiative). However, existing research has paid limited attention to the interdependent relationship between influencing factors of conflicts in these alliances. To address this gap, this paper employs the interpretative structure model (ISM) to establish mutual relationships among 16 identified influencing factors. The results show that target difference, unbalanced input of resources, and unreasonable distribution of benefits are recognized as key facilitators at the top level, while political risks, impact of financial factors, and policy changes are identified as foundational drivers at the bottom level. At the intermediate level, factors such as demand difference, technical capabilities difference, cognitive difference, value pursuit difference, information asymmetry, lack of trust, poor communication, resource dependency, and unreasonable contribution evaluation are positioned. These identified interdependence relationships offer crucial reference information for conflict management in standardization alliances for the B&R Initiative.

Abstract Background Lack of physical activity (PA), poor dietary habits, or other unhealthy lifestyle behaviors are potential modifiable risk factors for hypertension. It has been sufficiently demonstrated in previous studies that physical activity or healthy dietary patterns can reduce the risk of hypertension. However, no research focused on the joint effects of PA and healthy dietary patterns on hypertension in a representative sample of adults. Methods We used data collected from the 2007–2018 National Health and Nutrition Examination Survey. Healthy dietary patterns were assessed with the Healthy Eating Index 2015 (HEI-2015), and PA was measured using the metabolic equivalent minutes per week reported in questionnaires. We created four lifestyle categories based on the HEI-2015 and PA: (1) unhealthy diet and physically inactive (less than recommended PA), (2) healthy diet but physically inactive, (3) unhealthy diet but physically active (recommended PA), (4) healthy diet and physically active. Logistic regression was used to evaluate the association between joint PA and HEI-2015 and hypertension. Results A total of 24,453 participants were enrolled in the study. Compared with unhealthy diet and physically inactive individuals, only healthy diet and physically active participants (adjusted odds ratio [AOR]: 0.77, 95% CI 0.65–0.9) were negatively associated with hypertension, while healthy diet but physically inactive participants (AOR: 0.89, 95% CI 0.76–1.03) and unhealthy diet but physically active participants (AOR: 0.9, 95% CI 0.76–1.06) were not associated with hypertension. Conclusion In a representative sample of US adults, our findings suggest that individuals with recommended PA and healthy dietary patterns have a lower risk of hypertension than those with an unhealthy diet or less than recommended PA. Healthy eating habits and regular PA are potential preventive precautions against hypertension.

Abstract Background The prognosis for hepatocellular carcinoma (HCC) remains suboptimal, characterized by high recurrence and metastasis rates. Although metalloimmunotherapy has shown potential in combating tumor proliferation, recurrence and metastasis, current apoptosis-based metalloimmunotherapy fails to elicit sufficient immune response for HCC. Results A smart responsive bimetallic nanovaccine was constructed to induce immunogenic cell death (ICD) through pyroptosis and enhance the efficacy of the cGAS-STING pathway. The nanovaccine was composed of manganese-doped mesoporous silica as a carrier, loaded with sorafenib (SOR) and modified with MIL-100 (Fe), where Fe3+, SOR, and Mn2+ were synchronized and released into the tumor with the help of the tumor microenvironment (TME). Afterward, Fe3+ worked synergistically with SOR-induced immunogenic pyroptosis (via both the classical and nonclassical signaling pathways), causing the outflow of abundant immunogenic factors, which contributes to dendritic cell (DC) maturation, and the exposure of double-stranded DNA (dsDNA). Subsequently, the exposed dsDNA and Mn2+ jointly activated the cGAS-STING pathway and induced the release of type I interferons, which further led to DC maturation. Moreover, Mn2+-related T1 magnetic resonance imaging (MRI) was used to visually evaluate the smart response functionality of the nanovaccine. Conclusion The utilization of metallic nanovaccines to induce pyroptosis-mediated immune activation provides a promising paradigm for HCC treatment.

Abstract Chirality is one of the fundamental properties of molecules traditionally constructed from atoms. Here, we report for the first time the successful construction of asymmetric chiral structures utilizing highly symmetric endohedral metallofullerene superatoms based on their own bonding properties. Specifically, stable mirror‐symmetric sinister and rectus structures are obtained by selecting a superatom capable of forming four chemical bonds as the chiral center. Further analysis shows that the chiral vibration frequency of superatomic assemblies can be as low as a few wavenumbers, which greatly expands the range of chiral spectra compared to atom‐based molecules. We term this type of chirality based on superatoms as “superatomic‐based chirality”. It is anticipated that this work will significantly expand the variety of chiral structures at the atomic level.

Abstract The blockade of cytoprotective autophagy has been demonstrated to effectively enhance the efficacy of sonodynamic therapy (SDT). However, the limited recognition of antiautophagy agents for autophagosomes impedes the clinical application of autophagy inhibition. To efficiently deliver hydroxychloroquine (HCQ), an autophagy inhibitor, to autophagosomes, we utilized a strategy based on in situ click chemistry between sulfhydryl (‐SH) and maleimide (Mal) groups to trigger autophagosomes tracking and suppress tumor growth synergistically. A cascade nanoreactor was synthesized by encapsulating Mal‐modified HCQ (MHCQ) into a manganese porphyrin‐based metal‐organic framework with sonosensitizer properties, followed by poly(ethylene glycol)ylated liposomal membrane coating. After ultrasound irradiation, SDT‐induced apoptotic cells released damaged proteins with free ‐SH groups, which MHCQ rapidly captured in situ via a Mal‐thiol click reaction. When autophagosomes actively wrapped damaged proteins for detoxification, they simultaneously internalized HCQ anchored on proteins. In this scenario, antiautophagy drugs could actively track intracellular autophagosomes instead of undergoing passive diffusion in the cytosol. The interaction between HCQ and autophagic vesicles was greatly enhanced, which strengthened the blocking efficiency of autophagy and resulted in complete cell death. Overall, this study with smart design provides a promising strategy for improving intracellular targeted delivery to autophagosomes, thereby enhancing antitumor therapy.

Classical constitutive law between the structure and spectra has long been established in most physical processes and behaviors. Despite the emerging consensus, whether this law applies to microscopic systems adhering to quantum mechanics remains a fundamental and open question, due to limitations in even the most state-of-the-art experimental techniques. Here, we report an anomalous constitutive law of hydrogen bonds (X-H···Y) using a benchmark ab initio method. Notably, a donor X-H bond shortening occurred simultaneously with an unexpected redshift and enhanced intensity in IR spectra. The reason for donor X-H bond shortening is attributed to both the slowly increasing attractive force from the acceptor Y atom and the increasing short-range repulsive forces from other protons attached to the approaching acceptor Y atom. Furthermore, neither eliminating vibrational mixing nor introducing various physical effects make vibrational spectra respond to X-H bond shortening. Other structural-characterization NMR spectra provide robust evidence for this anomalous law. This anomalous law is ascribed to the interplay of electronic correlations. Our Letter thus inspires a reassessment of paradigms, which broadens the scope of microscopic behavior for finer control.

Abstract Soft on‐skin electrodes play an important role in wearable technologies, requiring attributes such as wearing comfort, high conductivity, and gas permeability. However, conventional fabrication methods often compromise simplicity, cost‐effectiveness, or mechanical resilience. In this study, a mechanically robust and gas‐permeable on‐skin electrode is presented that incorporates Flash Graphene (FG) integrated with a bioinspired armor design. FG, synthesized through Flash Joule Heating process, offers a small‐sized and turbostratic arrangement that is ideal for the assembly of a conductive network with nanopore structures. Screen‐printing is used to embed the FG assembly into the framework of polypropylene melt‐blown nonwoven fabrics (PPMF), forming a soft on‐skin electrode with low sheet resistance (125.2 ± 4.7 Ω/□) and high gas permeability (≈10.08 mg cm⁻2 h⁻¹). The “armor” framework ensures enduring mechanical stability through adhesion, washability, and 10,000 cycles of mechanical contact friction tests. Demonstrating capabilities in electrocardiogram (ECG) and electromyogram (EMG) monitoring, along with serving as a self‐powered triboelectric sensor, the FG/PPMF electrode holds promise for scalable, high‐performance flexible sensing applications, thereby enriching the landscape of integrated wearable technologies.

Currently, commercial cellular public networks such as 4G/5G have been widely deployed in China. In addition to serving general public customers, they are extensively used in the electric power industry for electricity information acquisition and distribution automation telemetry, as well as other non-control-related businesses. Simultaneously, power grid companies are actively studying and deploying electric power wireless private networks for bearing high-security control-related businesses such as distribution grid dispatching and distributed energy control. Electric power wireless private networks utilize dedicated spectrum resources, which operate on narrow individual carrier channels with numerous carriers in a discrete manner. These spectral characteristics indicate that electric power wireless private networks require unique system designs that are different from those of commercial cellular networks. Although some proposals have been proposed in relevant research, a unified and mature technical specification for electric power wireless private networks has not yet been established. This paper introduces the development history of electric power wireless private networks and power grid business requirements, with a specific emphasis on system design and key technologies, including frame structure and numerology, massive discrete carrier aggregation, spectrum sensing, security and isolation. We presented a comprehensive analysis and suggestions for these key technologies. Finally, we discuss future research on technological evolution. This study offers indications for an electric power wireless private network ecosystem.

Abstract Cancer immunotherapies have achieved unprecedented success in clinic, but they remain largely ineffective in some major types of cancer, such as colorectal cancer with microsatellite stability (MSS CRC). It is therefore important to study tumor microenvironment of resistant cancers for developing new intervention strategies. In this study, we identify a metabolic cue that determines the unique immune landscape of MSS CRC. Through secretion of distal cholesterol precursors, which directly activate RORγt, MSS CRC cells can polarize T cells toward Th17 cells that have well-characterized pro-tumor functions in colorectal cancer. Analysis of large human cancer cohorts revealed an asynchronous pattern of the cholesterol biosynthesis in MSS CRC, which is responsible for the abnormal accumulation of distal cholesterol precursors. Inhibiting the cholesterol biosynthesis enzyme Cyp51, by pharmacological or genetic interventions, reduced the levels of intratumoral distal cholesterol precursors and suppressed tumor progression through a Th17-modulation mechanism in preclinical MSS CRC models. Our study therefore reveals a novel mechanism of cancer–immune interaction and an intervention strategy for the difficult-to-treat MSS CRC.

The land controlled-source electromagnetic (CSEM) method dynamically monitors reservoir fluid by contrasting electrical conductivity between hydraulic fracturing fluids and surrounding rock formations. In hydraulic fracturing monitoring, the underground steel casings represent significant high-conductivity objects that require accurate and rapid numerical modeling for understanding the CSEM responses and optimizing the survey configurations. To efficiently simulate the steel well casings, we made use of the modified finite volume method (FVM) with the concept of edge conductivity equivalence that avoids the computational cost of grid refinement down to millimeters or centimeters for the casing. The new algorithm is satisfactorily validated by a numerical comparison against a trusted program Dipole1D and a commercial program COMSOL. Then the algorithm is used to investigate the characteristic CSEM responses of injected fracturing fluid due to a galvanic electromagnetic (EM) source grounded near the head of steel-cased well, an unconventional configuration that can maximize the data anomaly by channeling more excitation current to the deep reservoir. In particularly, we simulated two fracturing scenarios with different fluid migration directions: one with a high-resistivity cap layer as the background and the other with a realistic heavy oil fracturing site from Karamay in Xinjiang, China. Our simulation shows that the configuration of near-casing excitation significantly enhances the electric-field anomalies of the fluid injection, which is the most prominent as the galvanic effect at low frequencies. These phenomena highlight the crucial role of steel casing in the CSEM method for hydraulic fracturing monitoring.

Inspired by the microstructures of shells, SiC fibers (SiCf) are introduced into Ti layers, creating hybrid layers of SiCf/Ti, which are combined with Ti2AlNb foils to prepare multilayer SiCf/Ti–Ti2AlNb hybrids in a hot-pressed sintering instrument. The solid-phase bonding interface, mainly including the α+β biphasic structures and B2-rich phases, guarantees an ideal interface connection between the SiCf/Ti and Ti2AlNb layers after the sintering. Herein, the SiC fibers-made SiCf/Ti–Ti2AlNb presents a higher tensile strength and elasticity modulus on the tensile instrument of Model No. Byes (2010), compared with Ti/Ti2AlNb. During the tensile process, the brittle fractures of SiCf first occur, and the loads are transferred to the bonding interface of the Ti2AlNb and Ti layers, generating the crack propagation and resulting in SiCf/Ti–Ti2AlNb fracture. This is mainly because the crack is extended in succession to the bonding interface that is composed of brittle phases such as O and α2, as well as ductile phases such as α, β and B2. This produces the ductile and brittle ruptures at the bonding interface, forming the ductile-brittle mixture fractures at the SiCf/Ti and Ti2AlNb interfaces. In the case of vertical fractures related to the fiber length, brittle fractures are observed on the SiC fibers, which produce high forces that affect their bonding interfaces, resulting in axial interstices and transverse fractures. These obtained results demonstrate a novel method for structural design and fabrication of the innovative composite materials.

Abstract The humpback grouper (Cromileptes altivelis), a medium-sized coral reef teleost, is a naturally rare species distributed in the tropical waters of the Indian and Pacific Oceans. It has high market value, but artificial reproduction and breeding remain limited and need to be improved. Here, we assembled the genome with 1.08 Gb, with a contig N50 of 43.78 Mb. A total of 96.59% of the assembly anchored to 24 pseudochromosomes using Hi-C technology. It contained 24,442 protein-coding sequences, of which 99.3% were functionally annotated. The completeness of the assembly was estimated to be 97.3% using BUSCO. The phylogenomic analysis suggested that humpback grouper should be classified into the genus Epinephelus rather than Cromileptes. The comparative genomic analysis revealed that the gene families related to circadian entrainment were significantly expanded. The high-quality reference genome provides useful genomic tools for exploiting the genomic resource of humpback grouper and supports the functional genomic study of this species in the future.

Abstract Background Drought stress can substantially restrict maize growth and productivity, and global warming and an increasing frequency of extreme weather events are likely to result in more yield losses in the future. Therefore, unraveling the molecular mechanism underlying the response to drought stress is essential for breeding drought-resilient crops. Results In this study, we subjected the 3-leaf-period plants of two maize inbred lines, a drought-tolerant line (si287) and a drought-sensitive line (X178), to drought stress for seven days while growing in a chamber. Subsequently, we measured physiological traits and analyzed transcriptomic and metabolic profiles of two inbred lines. Our KEGG analysis of genes and metabolites revealed significant differences in pathways related to glycolysis/gluconeogenesis, flavonoid biosynthesis, starch and sucrose metabolism, and biosynthesis of amino acids. Additionally, our joint analysis identified proline, tryptophan and phenylalanine are crucial amino acids for maize response to drought stress. Furthermore, we concentrated on tryptophan (Trp), which was found to enhance tolerance via IAA-ABA signaling, as well as SA and nicotinamide adenine dinucleotide (NAD) consequent reactive oxygen species (ROS) scavenging. We identified three hub genes in tryptophan biosynthesis, indole-3-acetaldehyde oxidase (ZmAO1, 542,228), catalase 1 (ZmCAT1, 542,369), and flavin-containing monooxygenase 6 (ZmYUC6, 103,629,142), High expression of these genes plays a significant role in regulating drought tolerance. Two metabolites related to tryptophan biosynthesis, quinolinic acid, and kynurenine improved maize tolerance to drought stress by scavenging reactive oxygen species. Conclusions This study illuminates the mechanisms underlying the response of maize seedlings to drought stress. Especially, it identifies novel candidate genes and metabolites, enriching our understanding of the role of tryptophan in drought stress. The identification of distinct resistance mechanisms in maize inbred lines will facilitate the exploration of maize germplasm and the breeding of drought-resilient hybrids.

Nasal drug delivery efficiency is highly dependent on the position in which the drug is deposited in the nasal cavity. However, no reliable method is currently available to assess its impact on delivery performance. In this study, a biomimetic nasal model based on three-dimensional (3D) reconstruction and three-dimensional printing (3DP) technology was developed for visualizing the deposition of drug powders in the nasal cavity. The results showed significant differences in cavity area and volume and powder distribution in the anterior part of the biomimetic nasal model of Chinese males and females. The nasal cavity model was modified with dimethicone and validated to be suitable for the deposition test. The experimental device produced the most satisfactory results with five spray times. Furthermore, particle sizes and spray angles were found to significantly affect the experimental device's performance and alter drug distribution, respectively. Additionally, mometasone furoate (MF) nasal spray (NS) distribution patterns were investigated in a goat nasal cavity model and three male goat noses, confirming the in vitro and in vivo correlation. In conclusion, the developed human nasal structure biomimetic device has the potential to be a valuable tool for assessing nasal drug delivery system deposition and distribution.

To further improve the strength of an Al–Ce alloy, the effects of electromagnetic stirring treatment, Sc–Zr microalloying, and their compound action on the microstructure and properties of the Al–8Ce alloy were investigated for the first time in this study. The results show that the primary α-Al becomes coarse, the eutectic Al11Ce3 phase is refined and the lamellar spacing increases significantly due to the combined effect of forced convection of electromagnetic field and dendrite fragmentation and nucleation in Al–8Ce alloy after electromagnetic stirring treatment. After the Sc–Zr microalloying treatment of the Al–8Ce alloy, the α-Al changes from a dendritic to fine round morphology, and the eutectic Al11Ce3 phase changes from lamellar to fibrous, which is due to the heterogeneous nucleation of Al3(Sc, Zr) phase and the adsorption refinement caused by the enrichment of Sc and Zr. The optimization effect of Sc–Zr microalloying on the mechanical properties of the Al–8Ce alloy was better than that of electromagnetic stirring and their synergistic treatment. The tensile strength and yield strength of Al–8Ce alloy with Sc–Zr additions were increased from 157 MPa to 85 MPa–192 MPa and 115 MPa, respectively, by 22.3 % and 35.3 %. After aging treatment, the tensile strength (from 192 to 272 MPa) and yield strength (from 115 to 221 MPa) significantly improved owing to the secondary precipitation of Al3(Sc, Zr) particles. The precipitation strengthening of the Al3(Sc, Zr) phase was the main contributor to the high yield strength of the alloy.

Abstract Background Acute kidney injury (AKI) is a prevalent complication following acute type A aortic dissection (ATAAD) surgery and is closely associated with unfavorable prognostic outcomes. Hence, the development of a robust and efficient diagnostic approach to identify high-risk patients is of paramount importance. Methods We conducted a prospective study involving 328 patients who underwent ATAAD surgery at our institution, comprising three distinct cohorts. In addition, 52 patients undergoing alternative cardiopulmonary surgeries and 37 healthy individuals were enrolled as control groups. Employing proteomic analysis, we initially identified plasma proteins potentially linked to AKI occurrence within the plasma proteomic cohort. Subsequent validation was performed in an independent cohort. Utilizing predictors derived from multivariate logistic regression analysis, a nomogram was meticulously formulated and its efficacy was validated in the model construction cohort. Results Proteomics revealed significant elevation of plasma levels of S100A8/A9, pentraxin 3 (PTX3), and chitinase 3-like 1 (CHI3L1) immediately post-surgery in patients who developed ATAAD surgery-associated AKI (ASA-AKI). Receiver operating characteristic (ROC) curves demonstrated impressive predictive performance of S100A8/A9, PTX3, and CHI3L1 at 0 h post-surgery, yielding area under the curve (AUC) values of 0.823, 0.786, and 0.803, respectively, for ASA-AKI prediction. Furthermore, our findings exhibited positive correlations between plasma levels of S100A8/A9, PTX3, CHI3L1, and urinary neutrophil gelatinase-associated lipocalin (NGAL) at 0 h post-surgery, along with correlations between plasma S100A8/A9, CHI3L1 levels, and the Cleveland Clinic score. A logistic regression model incorporating plasma S100A8/A9, PTX3, CHI3L1 levels, urinary NGAL levels, and the Cleveland Clinic score facilitated the construction of a predictive nomogram for ASA-AKI. This nomogram demonstrated robust discriminative ability, achieving an AUC of 0.963 in the model construction cohort. Conclusions Our study underscored the augmentation of plasma S100A8/A9, PTX3, and CHI3L1 levels immediately post-surgery in patients developing ASA-AKI. The incorporation of these three biomarkers, in conjunction with the Cleveland Clinic score and NGAL, into a nomogram demonstrated commendable predictive efficacy. This presents a practical tool for identifying patients at an elevated risk of AKI following ATAAD surgery.

Fusarium wilt of watermelon caused by Fusarium oxysporum f. sp. niveum (Fon) results in a substantial economic losses in agriculture. Biological control agents (BCAs) help in controlling the infection of Fon. However, it remains unclear whether the application of BCAs can invoke soil suppression by inducing specific microbial groups concerned with disease suppression to synergistically resist fungal pathogens. The objective of this project was to determine the microbial mechanisms involved in disease suppression by BCAs. Here, we conducted pot experiments to explore Fusarium wilt in watermelon and soil microbial communities in response to different treatments. We found that the application of Bacillus velezensis WB enhanced disease suppression in nonsterilized soil. We further found that disease suppression in the WF treatment was connected to influence on the resident soil microbial communities, explicitly by causing an increase in Hyphomicrobiaceae and Chitinophagaceae. Pathogen suppression by the above two families was further studied in laboratory and pot experiments. The results showed that a synthetic community consisting of specific bacterial taxa effectively decreased the incidence of Fusarium wilt. B. velezensis WB introduction impacts the resident soil microbiome and invokes soil suppression by inducing resident microbes. Our results should help in designing synthetic microbial communities to improve soil function.

The deep learning steel plastic constitutive model training method was studied based on the recurrent neural network (RNN) model to improve the allocative efficiency of the deep learning steel plastic constitutive model and promote its application in practical engineering. Two linear hardening constitutive datasets of steel were constructed using the Gaussian stochastic process. The RNN, long short-term memory (LSTM), and gated recurrent unit (GRU) were used as models for training. The effects of the data pre-processing method, neural network structure, and training method on the model training were analyzed. The prediction ability of the model for different scale series and the corresponding data demand were evaluated. The results show that LSTM and the GRU are more suitable for stress–strain prediction. The marginal effect of the stacked neural network depth and number gradually decreases, and the hysteresis curve can be accurately predicted by a two-layer RNN. The optimal structure of the two models is A50-100 and B150-150. The prediction accuracy of the models increased with the decrease in batch size and the increase in training batch, and the training time also increased significantly. The decay learning rate method could balance the prediction accuracy and training time, and the optimal initial learning rate, batch size, and training batch were 0.001, 60, and 100, respectively. The deep learning plastic constitutive model based on the optimal parameters can accurately predict the hysteresis curve of steel, and the prediction abilities of the GRU are 6.13, 6.7, and 3.3 times those of LSTM in short, medium, and long sequences, respectively.

Ensuring the safety of transmission line towers is vital for human safety, power supply, economic development, and environmental protection. This study specifically examines how temperature affects tower inclination. Multifractal detrended cross-correlation analysis (MF-DCCA) is a combination of multifractal detrended fluctuation analysis (MF-DFA) and DCCA that reveals the multifractal features of two cross-correlated non-stationary signals. This paper adopts the MF-DCCA tool to investigate the cross-correlations between the internal temperature of an inclination sensor device and the posture of a transmission line tower. The tilt angle data in the x- and y-axes are used to measure the posture of the transmission line tower. We start by using Pearson correlation to assess the relationship between temperature and two inclination angles, followed by verifying their correlation with a p-value below 0.05 using first-order linear fitting. We initially assess the multifractal features of three time series using MF-DFA before MF-DCCA analysis. All exhibit multifractal traits with H(2)<0.5, indicating negative persistence, especially notable in the temperature series. Finally, we adopt the MF-DCCA approach to examine the multifractal cross-correlation between tilt-angle time series and temperature time series, and the results indicate the negative persistence of the cross-correlation between the time series. Furthermore, the multifractal cross-correlation of temperature and inclination data on the y-axis was also found to be stronger than on the x-axis based on features of the scaling exponent and symmetry exponent.

In this paper, for the sake of enhancing the security of wireless transmission, we proposed a novel system based on spatial and direction modulation (SDM) combined with generalized spatial modulation (GSM) which is aided by covert information mapping (CIM), termed as the CIM-GSDM system. In such a system, the legitimated user is equipped with distributed receivers so as to demodulate the conveyed signal by exploiting its indices while disturbing eavesdroppers for information security. More specifically, part of the information is modulated into the indices of the legitimated distributed receiver subsets with the aid of the mapped covert information and the interference matrix, while another part of the message is arranged by conventional amplitude-phase modulation. The proposed system can reap the benefits from both GSM and CIM to make eavesdropper suffer great mixture. Furthermore, the detection scheme and theoretical analysis of error performance are discussed as well. The simulation results exhibit that the bit error rate (BER) performance of legitimate user is much better than that of the eavesdropper while the proposed scheme improves the security compared to the original CIM-SDM system at the same spectral efficiency.

Exploring the biological potential of maize root architecture is one of the most important ways to improve nitrogen use efficiency (NUE). The NUE and its heterosis in maize hybrids have improved remarkably over decades. Yet, there is little research on maize hybrid heterosis for root architecture and its possible physiological relationship to heterosis for NUE. A field study lasting two years was carried out on four typical maize hybrids from old and new eras, including their parental inbred lines with two levels of nitrogen (0, 150 kg N ha−1). Compared to old-era maize hybrids, the brace root angle (BA) and crown root angle (CA) of new-era maize hybrids increased by 19.3% and 8.0% under 0 N, and by 18.8% and 7.9% under 150 N, which exhibited a steeper root architecture; the crown root number (CN) of new-era maize hybrids increased by 30.5% and 21.4% under 0 N and 150 N, respectively, which showed a denser root system; meanwhile, the depth of 95% cumulative root weight (D95) of new-era maize hybrids separately increased by 10.5% and 8.5% under 0 N and 150 N, which exhibited a deeper root distribution. This steeper-denser-deeper root architecture enhanced pre-anthesis N uptake and provided a premise of greater post-anthesis N remobilization. All maize hybrids displayed significant heterosis for root architecture compared to their parental inbred lines. The brace root branching (BB) and crown root branching (CB) of new-era maize hybrids and D95 have positive heterosis, while the BA, CA, and CB of old-era maize hybrids, brace root number (BN), and CN have negative heterosis. Regardless of whether root architecture heterosis was positive or negative, new-era maize hybrids showed an overall elevated trend compared to old-era maize hybrids. Structural equation modeling (SEM) showed that heterosis for nitrogen internal efficiency (NIE) was the primary reason for NUE heterosis in maize, influenced by heterosis for root architecture, which was steeper, denser, and deeper. Our results indicated that, compared with old-era maize hybrids, new-era maize hybrids had stronger heterosis for root architecture, which was beneficial to pre-silking nitrogen absorption and is an important physiological basis for the higher NIE heterosis and NUE heterosis in new-era maize hybrids.

Abstract Oxidative stress induced by excess reactive oxygen species (ROS) is a primary pathogenic cause of acute kidney injury (AKI). Development of an effective antioxidation system to mitigate oxidative stress for alleviating AKI remains to be investigated. This study presents the synthesis of an ultra‐small Platinum (Pt) sulfur cluster (Pt5.65S), which functions as a pH‐activatable prefabricated nanozyme (pre‐nanozyme). This pre‐nanozyme releases hydrogen sulfide (H2S) and transforms into a nanozyme (Ptzyme) that mimics various antioxidant enzymes, including superoxide dismutase and catalase, within the inflammatory microenvironment. Notably, the Pt5.65S pre‐nanozyme exhibits an endo‐exogenous synergy‐enhanced antioxidant therapeutic mechanism. The Ptzyme reduces oxidative damage and inflammation, while the released H2S gas promotes proneurogenesis by activating Nrf2 and upregulating the expression of antioxidant molecules and enzymes. Consequently, the Pt5.65S pre‐nanozyme shows cytoprotective effects against ROS/reactive nitrogen species (RNS)‐mediated damage at remarkably low doses, significantly improving treatment efficacy in mouse models of kidney ischemia‐reperfusion injury and cisplatin‐induced AKI. Based on these findings, the H2S‐generating pre‐nanozyme may represent a promising therapeutic strategy for mitigating inflammatory diseases such as AKI and others.

Recurrence and metastasis of gastric cancer is a major therapeutic challenge for treatment. The presence of cancer stem cells (CSCs) is a major obstacle to the success of current cancer therapy, often leading to treatment resistance and tumor recurrence and metastasis. Therefore, it is important to develop effective strategies to eradicate CSCs. In this study, we developed a combined therapeutic strategy of photothermal therapy (PTT) and gastric cancer stem cells (GCSCs) inhibition by successfully synthesizing nanoliposomes loaded with IR780 (photosensitizer) and EN4 (c-Myc inhibitor). The nanocomposites are biocompatible and exhibit superior photoacoustic (PA) imaging properties. Under laser irradiation, IR780-mediated PTT effectively and rapidly killed tumor cells, while EN4 synergistically inhibited the self-renewal and stemness of GCSCs by suppressing the expression and activity of the pluripotent transcription factor c-Myc, preventing the tumor progression of gastric cancer. This Nano-EN-IR@Lip is expected to be a novel clinical nanomedicine for the integration of gastric cancer diagnosis, treatment and prevention.

Abstract Overproduction of reactive oxygen species (ROS), metal ion accumulation, and tricarboxylic acid cycle collapse are crucial factors in mitochondria‐mediated cell death. However, the highly adaptive nature and damage‐repair capabilities of malignant tumors strongly limit the efficacy of treatments based on a single treatment mode. To address this challenge, a self‐reinforced bimetallic Mito‐Jammer is developed by incorporating doxorubicin (DOX) and calcium peroxide (CaO2) into hyaluronic acid (HA) ‐modified metal‐organic frameworks (MOF). After cellular, Mito‐Jammer dissociates into CaO2 and Cu2+ in the tumor microenvironment. The exposed CaO2 further yields hydrogen peroxide (H2O2) and Ca2+ in a weakly acidic environment to strengthen the Cu2+‐based Fenton‐like reaction. Furthermore, the combination of chemodynamic therapy and Ca2+ overload exacerbates ROS storms and mitochondrial damage, resulting in the downregulation of intracellular adenosine triphosphate (ATP) levels and blocking of Cu‐ATPase to sensitize cuproptosis. This multilevel interaction strategy also activates robust immunogenic cell death and suppresses tumor metastasis simultaneously. This study presents a multivariate model for revolutionizing mitochondria damage, relying on the continuous retention of bimetallic ions to boost cuproptosis/immunotherapy in cancer.

Seedling mode plays a crucial role in the rice production process, as it significantly affects the growth and development of seedlings. Among the various seedling modes, the seedling tray overlapping for seed emergence mode (STOSE mode) has been demonstrated to be effective in enhancing seedling quality. However, the impact of this mode on the germination and growth of seeds with varying plumpness remains uncertain. To investigate the effect of the STOSE mode on seedling emergence characteristics, growth uniformity, and nutrient uptake of seeds with varying plumpness levels, we conducted a study using super early rice Zhongzao 39 (ZZ39) as the test material. The seeds were categorized into three groups: plumped, mixed, and unplumped. The results indicated that the STOSE mode significantly improved the seedling rate for all types of seeds in comparison to the seedling tray nonoverlapping for seed emergence mode (TSR mode). Notably, the unplumped seeds exhibited the most pronounced enhancement effect. The soluble sugar content of the seeds increased significantly after 2 days of sowing under the STOSE mode, whereas the starch content exhibited a significant decrease. Furthermore, the STOSE mode outperformed the TSR mode in several aspects including seedling growth uniformity, aboveground dry matter mass, root traits, and nutrient uptake. Overall, the STOSE mode not only promoted the germination and growth of plumped and mixed seeds but also had a more pronounced impact on unplumped seeds.

Shallow utility tunnels with exceptionally large cross-sections in weak soil can face significantly more severe seismic risks compared to conventional deep tunnels in seismic hazard areas. This study investigates the seismic response and isolation technologies applied to a large section utility tunnel with 4 compartments in one layer, employing seismic simulations. The engineering context, dynamic motions, and measuring points of the numerical simulation were introduced. Subsequently, the finite element method was employed to explore the seismic behaviors of the large section utility tunnel when subjected to strong earthquake excitations under four conditions. The study explored and comparatively evaluated the seismic isolation effectiveness of three proposed schemes: the grouting scheme, the buffer layer, and the assembly of three-quarters buffer layer, cushion and grouting. The analysis included various indexes such as the deformation, the principal and shear stress, and the safety factor. The finding reveals that the assembly seismic scheme exhibits the most significant seismic effect (95.39%), followed by the buffer layer (38.82%), and the grouting scheme (25.66%). The assembly seismic scheme is recommended for the seismic design in the current large-section utility tunnels. These conclusions provide valuable scientific guidance for the seismic design of large-section utility tunnels, aiding in enhancing earthquake resilience.

BackgroundThis study aims to analyze the risk factors associated with prolonged mechanical ventilation (PMV) in patients following surgical treatment for acute type A aortic dissection (ATAAD). The objectives include constructing a predictive model for risk assessment and validating its predictive efficacy.MethodsA total of 452 patients diagnosed with ATAAD and undergoing surgical procedures at a tertiary hospital in Nanjing between January 2021 and April 2023 were selected using a convenience sampling method. Patients were categorized into two groups: PMV group (n = 132) and non-PMV group (n = 320) based on the occurrence of prolonged mechanical ventilation (PMV), and their clinical data were compared. The data were randomly divided into a modeling set and a validation set in a 7:3 ratio. Risk factors for PMV were identified in the modeling group using logistic regression analysis. A risk prediction model was constructed using R 4.1.3 software, visualized via a column chart. Receiver Operating Characteristic (ROC) curves were generated using the validation set to assess model differentiation. Calibration curves were plotted to evaluate accuracy and consistency, and Decision Curve Analysis (DCA) was applied to evaluate clinical utility.ResultsThe logistic regression analysis identified age, body mass index, preoperative white blood cell count, preoperative creatinine, preoperative cerebral hypoperfusion, and cardiopulmonary bypass time as significant risk factors for postoperative PMV in patients with ATAAD. The area under the curve (AUC) for the validation set ROC curve was 0.856, 95% confidence interval (0.805–0.907), indicating good discrimination. Calibration curves revealed strong alignment with the ideal curve, and the Hosmer-Lemeshow goodness-of-fit test indicated a well-fitted model (P = 0.892). The DCA curve demonstrated a high net benefit value, highlighting the model's strong clinical utility.ConclusionsThe risk prediction model developed in this study for PMV in patients undergoing surgery for ATAAD exhibits robust predictive performance. It provides valuable insights for healthcare practitioners in predicting the likelihood of PMV and devising timely and personalized intervention strategies.

Abstract Due to the ongoing global warming, maize production worldwide is expected to be heavily inflicted by droughts. The grain yield of maize hybrids is an important factor in evaluating their suitability and stability. In this study, we utilized the AMMI model and GGE biplot to analyze grain yield of 20 hybrids from the three tested environments in Inner Mongolia in 2018 and 2019, aiming at selecting drought-tolerant maize hybrids. AMMI variance analysis revealed highly significant difference on main effects for genotype, environment, and their interaction. Furthermore, G11 (DK159) and G15 (JKY3308) exhibited favorable productivity and stability across all three test environments. Moreover, G10 (LH1) emerged as the most stable hybrid according to the AMMI analysis and the GGE biplot. Bayannur demonstrated the highest identification ability among the three tested sites. Our study provides accurate identification for drought-resilient maize hybrids in different rain-fed regions. These findings can contribute to the selection of appropriate hybrids that exhibit productivity, stability, and adaptability in drought-prone conditions.

Abstract Early spring cold spells can lead to leaf chlorosis during the rice seedling greening process. However, the physiological and molecular mechanisms underlying the rice greening process under low-temperature conditions remain unknown. In this study, comparative transcriptome and morphophysiological analyses were performed to investigate the mechanisms mediating the responses of the Koshihikari (Kos) and Kasalath (Kas) rice cultivars to chilling stress. According to their growth-related traits, electrolyte leakage, and chlorophyll fluorescence parameters, Kos was more tolerant to low-temperature stress than Kas. Moreover, chloroplast morphology was more normal (e.g., oval) in Kos than in Kas at 17 °C. The comparative transcriptome analysis revealed 610 up-regulated differentially expressed genes that were common to all four comparisons. Furthermore, carotenoid biosynthesis was identified as a critical pathway for the Kos response to chilling stress. The genes in the carotenoid biosynthesis pathway were expressed at higher levels in Kos than in Kas at 17 °C, which was in accordance with the higher leaf carotenoid content in Kos than in Kas. The lycopene β-cyclase and lycopene ε-cyclase activities increased more in Kos than in Kas. Additionally, the increases in the violaxanthin de-epoxidase and carotenoid hydroxylase activities in Kos seedlings resulted in the accumulation of zeaxanthin and lutein and mitigated the effects of chilling stress on chloroplasts. These findings have clarified the molecular mechanisms underlying the chilling tolerance of rice seedlings during the greening process.

Abstract Purpose A randomized controlled double-blind trial was conducted to evaluate the effects of adding dexamethasone to the local infiltration analgesia (LIA) mixture on frequency of patient controlled analgesia (PCA) and opioids consumption after simultaneous bilateral total hip or knee arthroplasty (THA or TKA). Methods 108 patients who received simultaneous bilateral THA or TKA were randomly divided into dexamethasone group and normal saline (NS) group. The main difference between two groups was whether or not dexamethasone was added to the LIA mixture. The main outcome was the cumulative consumption of opioids within 24 h. The secondary outcome were the total cumulative consumption of opioids during postoperative hospitalization, consumption of opioids drug for rescue analgesia, frequency of PCA, postoperative Visual Analogue Scale (VAS), and complications. Results Cumulative consumption of opioids in the 24 h was similar between two groups (P = 0.17). Total cumulative consumption of opioids in the dexamethasone group during postoperative hospitalization was significantly lower (P = 0.03). No significant difference in the consumption of opioids drug for rescue analgesia between two groups within 24 h, while the frequency of PCA was significantly different (P = 0.04). VAS of dexamethasone group and NS group were similar during postoperative hospitalization, while the incidence of postoperative nausea and vomiting (PONV) in dexamethasone group was lower than that in NS group. Conclusions Adding dexamethasone to LIA in the simultaneous bilateral THA or TKA can effectively reduce the total cumulative consumption of opioids and the frequency of PCA, as well as reduce the incidence of PONV. Trial Registration The trial has been registered in the Chinese Clinical Trial Registry (Registration Number: ChiCTR2100042551, Date: 23/01/2021).

Abstract Objective To introduce a new type of simple adjustable bilateral bidirectional polyvinyl chloride (PVC) tube traction device and discuss the value of using this device before surgery in patients with lower limb fractures. Methods To introduce the manufacturing process of an adjustable bilateral traction device made of PVC pipes. From August 2018 to November 2019, the data of 36 patients with lower limb fractures who were treated with this traction device were retrospectively analysed. The treatment outcomes were analysed, including length of both lower limbs, fracture reduction, lower limb mobility, visual analogue scale (VAS) score, incidence of complications, and patient satisfaction. Results All patients were able to move the affected limb immediately after using the device. The patient's pain was significantly reduced, they were able to turn over freely during bed rest, and the length of the affected limb was restored to that of the healthy limb. Thirty-four (94.5%) patients were satisfied with the reduction of the fracture end, 2 (5.5%) patients with tibiofibular fractures showed angular displacement of the fractured end and satisfactory reduction after the position of the bone traction needle was adjusted; 7 (19.5%) patients developed deep vein thrombosis of the affected lower limb during traction; there was no decubitus or vascular nerve injury, and the overall complication rate was 25% (9/36). All the patients and their families were satisfied with the results of this treatment. Conclusion The aim of this study is to introduce a new type of traction device. It is advantageous in that it is light weight, low cost, easy to assemble, promotes immediate movement of the affected limb after assembly, improves patient comfort and can be used with a titanium steel needle for MRI examination under traction. In the clinical setting, it has been shown to be suitable for the temporary treatment of patients with lower leg fractures prior to surgery, particularly patients who, for various reasons, require nonsurgical treatment in the short term.

The effect of Ms temperature variation in Fe–Ni–Cr cast iron has been investigated in this paper. The heating and cooling processes were carefully compared and analysed through in-situ observation. The results showed that the size of austenite grains remained relatively constant throughout the austenitizing process. Moreover, higher austenitizing temperatures were found to facilitate the transfer of carbon from graphite to austenite, which hindered the martensite transformation. The M3C and M23C6 carbide generated during heating and cooling diminished the concentration of carbon and alloying elements in the austenite and enhanced the Ms temperature. The acceleration of cooling rate could impede the precipitation of carbide and result in more significant lattice distortion of austenite, leading to a notable reduction in Ms temperature and an increment in residual austenite content. The augmentation in austenitizing temperature and cooling rate amplified the non-chemical driving force, culminating in a more arduous martensitic transformation.

Morphing wing technology is crucial for enhancing the flight performance of aircraft. To address the monitoring challenges of full-scale variable-camber leading edges under flight conditions, this study introduces a ground-based strength testing technique aimed at precisely evaluating the deformation patterns and structural strength during actual operation. Firstly, the motion characteristics of the variable-camber leading edge were analyzed using numerical simulation based on kinematic theory. Secondly, a tracking loading test rig was designed and constructed to simulate the actuated deformation and aerodynamic loads of the leading edge. Next, mechanical boundary numerical simulation was then utilized to predict the motion trajectories of loading points on the upper and lower wing surfaces, and a multi-point coordinated control system was developed to achieve accurate experimental control. Finally, a multi-sensor iterative method was employed to ensure loading precision throughout the testing process. A case study was conducted using a leading edge test piece from a specific commercial aircraft. The results indicated that in the motion test of the variable-camber leading edge, the average error of the deflection angle was 4.59%; in the strength test, the average errors in the magnitude and direction of the applied load were 0.54% and 0.24%, respectively. These findings validate the effectiveness of the proposed technique in simulating the flight conditions of deforming wings and accurately obtaining the leading edge shape change curve, deformation accuracy curve, and strain curves of the upper and lower wing surfaces under deflection angles. Furthermore, this paper compares the deformation accuracy of different testing methods under test conditions, providing scientific evidence and technical support for the testing and evaluation of variable-camber leading edges.

The combined application of manure and mineral fertilizer represents an effective strategy for enhancing crop yield. However, the relationship between soil fertility and crop yield remains unclear in saline-alkaline soil. Here, a 9-year field experiment (2015–2023) was conducted to investigate the effects of manure application and crop rotations on crop yield and economic efficiency as well as potential associated mechanisms in the Hetao Irrigation District. The results showed that in the third cropping rotation cycle, combined application of manure and mineral fertilizers (NPKO) caused a 6.2%, 38.9%, 65.3%, and 132.2% increase in wheat, sunflower, wheat equivalent yield, and the economic income of sunflower, respectively. The average grain yield had a positive correlation with soil organic matter and nutrient supply. This suggested that the soil organic matter had a positive effect on the crop yield due to its impact on nutrient supply. Simultaneously, the sunflower seed setting rate increased by 65.2% under NPKO. The linear regression model revealed that each additional input of 20 Mg ha−1 of manure resulted in an increase of 3.56 kg ha−1 in crop phosphorus harvest and a 0.05 Kg ha−1 increase in wheat equivalent yield compared to NPK. In conclusion, our results highlighted that manure application promotes soil properties and improves crop yield.
