Abstract Gas explosion is one of the five major hazards in mines, with about 36% of such incidents occurring in the excavation working face. Therefore, to investigate the impact of gas explosion propagation laws within excavation roadways, we conducted a simulation analysis of parameters such as peak of explosion overpressure, peak rate of pressure rise, and flame propagation speed in the presence of a Tunnel Boring Machine (TBM). In the presence of the TBM, the explosion overpressure approximately doubles, and the flame propagation speed also greatly increases, exacerbating the explosion hazard. Thus, when investigating gas explosion laws within excavation roadways, the presence of a TBM emerges as a significant factor that cannot be overlooked. Furthermore, an analysis of the effects of methane concentration and gas accumulation length on explosion parameters was conducted. The results indicate that when the flame passes through the TBM baffle, the average flame propagation speed increases the most when the methane concentration is 9.5%, increasing by about 6 times. In addition, as the gas accumulation length increases, both explosion overpressure and flame propagation speed gradually increase. Additionally, TBM has a certain impact on flame propagation and methane dissipation.

Objective Sensitivity analysis is an crucial tool in groundwater modelling for measuring the importance of various model inputs, enabling better allocation of limited funds and resources to reduce predictive uncertainty. Methods In this paper, we propose an enhanced hierarchical global sensitivity analysis method to quantify contribution of different types of input uncertainty to model outputs, and to assess the impact of each uncertain process on groundwater model predictions. To test and demonstrate the new method, a hypothetical case study of groundwater flow and contaminant transport is used to validate. Results The results indicate that model uncertainty is the main source of prediction uncertainty in this case, and uncertainty from the geological model is more important than that of other models. Conclusion The proposed method offers a more comprehensive sensitivity analysis for groundwater models. Compared with traditional parameter sensitivity analysis, the new method can consider more uncertain input factors, significantly improve computational efficiency, and provide more useful sensitivity information for model users and managers.

Abstract Platinum-based chemotherapy is the standard postoperative adjuvant treatment for ovarian cancer (OC). Despite the initial response to chemotherapy, 85% of advanced OC patients will have recurrent disease. Relapsed disease and platinum resistance are the major causes of death in OC patients. In this study, we compared the global regulation of alternative polyadenylation (APA) in platinum-resistant and platinum-sensitive tissues of OC patients by analyzing a set of single-cell RNA sequencing (scRNA-seq) data from public databases and found that platinum-resistant patients exhibited global 3’ untranslated region (UTR) shortening due to the different usage of polyadenylation sites (PASs). The APA regulator CSTF3 was the most significantly upregulated gene in epithelial cells of platinum-resistant OC. CSTF3 knockdown increased the sensitivity of OC cells to platinum. The lncRNA NEAT1 has two isoforms, short (NEAT1_1) and long (NEAT1_2) transcript, because of the APA processing in 3’UTR. We found that CSTF3 knockdown reduced the usage of NEAT1 proximal PAS to lengthen the transcript and facilitate the expression of NEAT1_2. Downregulation of the expression of NEAT1 (NEAT1_1/_2), but not only NEAT1_2, also increased the sensitivity of OC cells to platinum. Overexpressed NEAT1_1 reversed the platinum resistance of OC cells after knocking down CSTF3 expression. Furthermore, downregulated expression of CSTF3 and NEAT1_1, rather than NEAT1_2, was positively correlated with inactivation of the PI3K/AKT/mTOR pathway in OC cells. Together, our findings revealed a novel mechanism of APA regulation in platinum-resistant OC. CSTF3 directly bound downstream of the NEAT1 proximal PAS to generate the short isoform NEAT1_1 and was conducive to platinum resistance, which provides a potential biomarker and therapeutic strategy for platinum-resistant OC patients.

In order to effectively prevent and control coal spontaneous combustion and improve the safety of coal mining, bagasse carboxymethy cellulose (BCC) prepared from bagasse (BS) and sodium carboxymethyl cellulose (CMC) were used as the substrates. A gel (BCC-CMC) for inhibiting coal spontaneous combustion was prepared using a chemical crosslinking method involving zirconium citrate crosslinking and glucono-delta-lactone (GDL) modification. X-ray diffraction (XRD) and Fourier transform infrared (FTIR) spectroscopy were utilized to characterize both the crystal and molecular structure of bagasse and its derived bagasse carboxymethyl cellulose. The results indicated that new carboxymethyl groups were introduced into bagasse during the treatment, facilitating the successful extraction and preparation of sodium carboxymethyl cellulose. The results of viscosity and bonding tests demonstrated that the concentration and dosage of CMC had the most significant influence on the gel viscosity, which remained higher and more stable at room temperature. The coal bonded by the gel coal mixture can effectively seal surface cracks, with a bonding degree of up to 70.72 %. TG-DTG, temperature-programmed oxidation experiment and FTIR analyses revealed that, compared to raw coal, coal samples treated with gel exhibited improved stability, fewer active groups, and fewer oxygen-containing functional groups, effectively inhibiting coal’s low temperature oxidation and reducing the risk of spontaneous combustion. The gel is environmentally friendly, cost-effective, and exhibits good flame retardant properties. This study is of great significance for preventing and controlling coal spontaneous combustion, effectively ensuring the safe production of coal resources and the safety of mine workers, achieving the green sustainable development of the mine.

Abstract The Hendra and Nipah viruses (HNVs) are highly pathogenic pathogens without approved interventions for human use. In addition, the interaction pattern between the attachment (G) and fusion (F) glycoproteins required for virus entry remains unclear. Here, we isolate a panel of Macaca-derived G-specific antibodies that cross-neutralize HNVs via multiple mechanisms. The most potent antibody, 1E5, confers adequate protection against the Nipah virus challenge in female hamsters. Crystallography demonstrates that 1E5 has a highly similar binding pattern to the receptor. In cryo-electron microscopy studies, the tendency of 1E5 to bind to the upper or lower heads results in two distinct quaternary structures of G. Furthermore, we identify the extended outer loop β1S2-β1S3 of G and two pockets on the apical region of fusion (F) glycoprotein as the essential sites for G-F interactions. This work highlights promising drug candidates against HNVs and contributes deeper insights into the viruses.

Introduction: The formation of bars and pools, characterized by concave and convex bed morphology, is a typical feature of curved rivers. The channel bed morphology has a significant influence on the flow structures in curved channels.Methods: Based on data from physical model experiments, this study employs the RNG k-ε model and the VOF (Volume of Fluid) method to perform three-dimensional numerical simulations of flow in continuous curved channels.Results: By comparing the variations in flow structures between channels with a flat bed and channels with bars and pools, the results show that the presence of bars and pools leads to an increase in longitudinal flow velocity on the convex bank side near the entrance of the upstream bend, while in the downstream bend it is opposite. The high-velocity region shifts slower towards the concave bank along the bend. The presence of point bars weakens the circulation near the convex bank in the upstream bend, resulting in a smaller circulation intensity. The decrease in circulation intensity is the largest (−23.91%) at the apex of the bend. In the downstream bend, the remaining circulation from the upstream bend attenuates slower in the pool and has a greater impact distance, increasing the circulation intensity in the downstream bend. The section near the bend entrance shows the largest increase in circulation intensity, with a rate of 128.18%. The unevenness of the bed topography increases the unevenness of the bed shear stress in the downstream bend.Discussion: The findings of this study contribute to a deeper understanding of the complex flow structures and evolution trends in natural curved rivers, providing scientific basis for the management of curved river channels.

Abstract Assembly ubiquitously occurs in nature and gives birth to numerous functional biomaterials and sophisticated organisms. In this work, chiral hydrogen-bonded organic-inorganic frameworks (HOIFs) are synthesized via biomimicking the self-assembly process from amino acids to proteins. Enjoying the homohelical configurations analogous to α-helix, the HOIFs exhibit remarkable chiroptical activity including the chiral fluorescence (g lum = 1.7 × 10−3) that is untouched among the previously reported hydrogen-bonded frameworks. Benefitting from the dynamic feature of hydrogen bonding, HOIFs enable enantio-discrimination of chiral aliphatic substrates with imperceivable steric discrepancy based on fluorescent change. Moreover, the disassembled HOIFs after recognition applications are capable of being facilely regenerated and self-purified via aprotic solvent-induced reassembly, leading to at least three consecutive cycles without losing the enantioselectivity. The underlying mechanism of chirality bias is decoded by the experimental isothermal titration calorimetry together with theoretic simulation.

The prediction of frictional air resistance using the inherent properties of roadways is of great significance for ventilation network computation and flow regulation in underground mines. This study proposes an improved stacked learning and error correction-based prediction model for the frictional air resistance of mine airways, called friction factor. A prediction set is established by selecting ten factors, including tunnel spatial features and support forms, with the ventilation resistance coefficient as the label. The improved stacked model consists of two layers. The first layer is the base learning module, which is composed of four components: Principal Components Analysis and Back Propagation (PCA-BP), GA-Projection Pursuit Regression (GA-PPR), Random Forest (RF), LightGBM (LGBM). The second layer is the meta-learning module, which is composed of the Ridge Regression (RR). Compared to traditional stacked models, the improved model first uses the Extreme Gradient Boosting (XG Boost) learner to evaluate the significance of input feature variables to eliminate redundancy and improve accuracy, thus enhancing prediction precision and computational efficiency. Then, the first-layer prediction results are weighted based on the errors of different prediction models in the training set using K-fold cross-validation. Box-Cox transformation is applied to the training set data from the first layer to the second layer to improve prediction normality and homogeneity. The error correction prediction model extracts the historical prediction errors from the meta-learning module and constructs an error prediction model using support vector machines (SVR), which are then combined with the meta-learning results to obtain the final prediction. The improved stacked model is compared with traditional ensemble learning models and single prediction models, and quantified using three metrics: root mean square error (RMSE), mean absolute error (MAE), and R-squared (R2). The results demonstrate that the proposed improved model effectively enhances the prediction accuracy of the ensemble learning models, providing a new prediction method for the accurate acquisition of the friction factor of mine airways.

Candidate genes related to first pod height (FPH) traits in faba bean plants are crucial for mechanised breeding. However, reports on quantitative trait locus (QTL) mapping related to the FPH of faba bean are few, thus limiting the high-quality development of the faba bean industry to a certain extent. The identification and screening of candidate genes related to FPH is extremely urgent for the advancement of mechanised breeding for faba bean. In this study, a high-density genetic linkage map was constructed using genotyping-by-sequencing (GBS) of an F3 population and QTLs (genes) related to FPH were identified. The genetic linkage map contained seven linkage groups with 3012 SNP markers with an overall length of 4089.13 centimorgan (cM) and an average marker density of 1.36 cM. Thirty-eight QTLs for the first pod node (FPN) and FPH were identified (19 each for FPN and FPH). The 19 QTLs associated with FPN were located on chromosomes 1L, 1S, 2, 3, 4, 5, and 6; the 19 QTLs associated with FPH were located on chromosomes 1L, 1S, 2, 3, 5, and 6. There was a co-localisation interval of qFPN6-1 and qFPH6-1 on chromosome 6. By annotating the QTL qFPH6-1 interval, 36 genes that may be related to FPH were identified, these genes are related to plant growth and development. The results provide a basis for the precise location of QTLs related to FPH and could accelerate the breeding of faba bean varieties adapted to mechanised harvesting.

Understanding the effect of changing wind speed on the evolution of spontaneous combustion of coal in goaf is crucial for ensuring safety in coal mining. This study focuses on the periodic disturbed airflow generated in the excavation roadway and its impact on the wind speed and goaf of the connected working face. The study reveals that (1) there is a negative correlation between the change of local ventilator wind speed and the wind speed at the working face. (2) the “periodic acceleration” ventilation leads to an increase in temperature at the fixed monitoring point. The maximum temperature is reached later than in normal ventilation conditions. (3) the change period of wind speed is positively correlated with the maximum temperature of the goaf. High wind speeds lead to a greater increase in temperature. (4) the temperature of the monitoring point in the goaf, the maximum internal temperature and the area of coal spontaneous combustion are more sensitive to the “periodic deceleration” disturbance airflow. In conclusion, the findings of this study provide valuable insights for the prevention and control of spontaneous combustion of coal in goaf. It highlights the importance of considering the effects of disturbed airflow in underground coal mining operations.

Osteoarthritis (OA) causes disability and significant economic and social burden. Cartilage injury is one of the main pathological features of OA, and is often manifested by excessive chondrocyte death, inflammatory response, abnormal bone metabolism, imbalance of extracellular matrix (ECM) metabolism, and abnormal vascular or nerve growth. Regrettably, due to the avascular nature of cartilage, its capacity to repair is notably limited. Mesenchymal stem cells‐derived extracellular vesicles (MSCs‐EVs) play a pivotal role in intercellular communication, presenting promising potential not only as early diagnostic biomarkers in OA but also as efficacious therapeutic strategy. MSCs‐EVs were confirmed to play a therapeutic role in the pathological process of cartilage injury mentioned above. This paper comprehensively provides the functions and mechanisms of MSCs‐EVs in cartilage repair.

The fodder soybean (Glycine max) is an excellent leguminous forage with a high protein content and hay yield, cultivated comprehensively in alpine regions, but seasonal drought in northern regions severely impacts the growth of seedlings. Melatonin (MT) and strigolactone (SL) are critical in relieving the restraint of plant growth in water-deficient environments, but the mechanisms of MT- and SL-mediated drought resistance in fodder soybean needs to be explored. This study mainly investigated the potential morphophysiological mechanism of MT and SL treatments in protecting fodder soybean from drought stress. The fodder soybean ‘Gongnong 535’ was treated with 100 µM MT or 1 µM SL under normal, moderate, and severe water deficit conditions. The results showed that MT and SL treatments enhanced the plant growth parameters and stomatal aperture under drought stress. Moreover, the observed reductions in superoxide ion (O2.-), malondialdehyde (MDA), and relative electrical conductivity (REC), along with enhancements in the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT), as well as higher levels of ascorbate (AsA), glutathione (GSH), soluble sugar (SS), soluble protein (SP), and free proline (Pro), indicated that MT and SL application effectively alleviated the oxidative damage and prevented the cell membrane disruption caused by drought stress. Additionally, MT and SL treatments improved photosynthesis and growth in fodder soybean seedlings under water stress by adjusting chlorophyll pigments, gas exchange indexes, and chlorophyll fluorescence parameters, as well as endogenous hormone levels. Simultaneously, MT and SL influenced the expression of genes associated with photosynthesis and antioxidant defenses, as well as phytohormone concentrations. Notably, the protective effect of the SL treatment was superior to that of MT in water-deficient conditions. This study contributes to further understanding the defensive mechanism of MT and SL against drought stress.

Abstract Background Clinically, although chemotherapy is one of the most commonly used methods of treating tumors, chemotherapeutic drugs can induce autophagic flux and increase tumor cell resistance, leading to drug tolerance. Therefore, theoretically, inhibiting autophagy may improve the efficacy of chemotherapy. The discovery of autophagy regulators and their potential application as adjuvant anti-cancer drugs is of substantial importance. In this study, we clarified that Fangjihuangqi Decoction (FJHQ, traditional Chinese medicine) is an autophagy inhibitor, which can synergistically enhance the effect of cisplatin and paclitaxel on non-small cell lung cancer (NSCLC) cells. Methods We observed the changes of autophagy level in NSCLC cells under the effect of FJHQ, and verified the level of the autophagy marker protein and cathepsin. Apoptosis was detected after the combination of FJHQ with cisplatin or paclitaxel, and NAC (ROS scavenger) was further used to verify the activation of ROS-MAPK pathway by FJHQ. Results We observed that FJHQ induced autophagosomes in NSCLC cells and increased the levels of P62 and LC3-II protein expression in a concentration- and time-gradient-dependent manner, indicating that autophagic flux was inhibited. Co-localization experiments further showed that while FJHQ did not inhibit autophagosome and lysosome fusion, it affected the maturation of cathepsin and thus inhibited the autophagic pathway. Finally, we found that the combination of FJHQ with cisplatin or paclitaxel increased the apoptosis rate of NSCLC cells, due to increased ROS accumulation and further activation of the ROS-MAPK pathway. This synergistic effect could be reversed by NAC. Conclusion Collectively, these results demonstrate that FJHQ is a novel late-stage autophagy inhibitor that can amplify the anti-tumor effect of cisplatin and paclitaxel against NSCLC cells.

Monitoring the temperature to determine the fire source locations is essential for controlling the spontaneous combustion in the goaf. Optical fiber sensors are employed to measure the temperature distribution in the goaf. However, due to changes in the geological conditions and the influence of the falling rocks in the goaf, only sensors on the upper side of the uncompacted goaf, due to inclination and coal pillar, may remain. Unilateral sensors are located on the upper side of the goaf, while fire occurs in the center. To investigate the issue with linear unilateral sensors, a two-dimensional inverse method has been developed to determine the location of fire sources by considering heat transfer after a fire inside the goaf. The equations were theoretically solved using Green's function method to obtain the internal temperature distribution of the physical model of the goaf. Sensitivity analysis identified the most crucial parameters in the process of spontaneous heating at different temperature. The fire source location can be determined using a loop method based on the model calculations. We considered a case to validate the model. Accurately identifying the fire source location in the goaf using the unilateral sensors has an essential theoretical and practical significance for fire prevention and fighting.

Cell source is the key to decellularized matrix (DM) strategy. This study compared 3 cell types, osteocytes with/without dominant active Wnt/β-catenin signaling (daCO and WTO) and bone marrow stromal cells (BMSCs) for their DMs in bone repair. Decellularization removes all organelles and >95% DNA, and retained >74% collagen and >71% GAG, maintains the integrity of cell basement membrane with dense boundaries showing oval and honeycomb structure in osteocytic DM and smooth but irregular shape in the BMSC-DM. DM produced higher cell survival rate (90%) and higher proliferative activity. In vitro, daCO-DM induces more and longer stress fibers in BMSCs, conducive to cell adhesion, spreading, and osteogenic differentiation. 8-wk after implantation of the critical-sized parietal bone defect model, daCO-DM formed tight structures, composed of a large number of densely-arranged type-I collagen under polarized light microscope, which is similar to and integrated with host bone. BV/TV (>54%) was 1.5, 2.9, and 3.5 times of WTO-DM, BMSC-DM, and none-DM groups, and N.Ob/T.Ar (3.2 × 102/mm2) was 1.7, 2.9, and 3.3 times. At 4-wk, daCO-DM induced osteoclastogenesis, 2.3 times higher than WTO-DM; but BMSC-DM or none-DM didn't. daCO-DM increased the expression of RANKL and MCSF, Vegfa and Angpt1, and Ngf in BMSCs, which contributes to osteoclastogenesis, angiogenesis, and neurogenesis, respectively. daCO-DM promoted H-type vessel formation and nerve markers β3-tubulin and NeuN expression. Conclusion: daCO-DM produces metabolic and neurovascularized organoid bone to accelerate the repair of bone defects. These features are expected to achieve the effect of autologous bone transplantation, suitable for transformation application.

In order to further improve the efficiency of energy utilization, Integrated Energy Systems (IES) connect various energy systems closer, which has become an important energy utilization mode in the process of energy transition. Because the complex and variable multiple load is an important part of the new power system, the load forecasting is of great significance for the planning, operation, control, and dispatching of the new power system. In order to timely track the latest research progress of the load forecasting method and grasp the current research hotspot and the direction of load forecasting, this paper reviews the relevant research content of the forecasting methods. Firstly, a brief overview of Integrated Energy Systems and load forecasting is provided. Secondly, traditional forecasting methods based on statistical analysis and intelligent forecasting methods based on machine learning are discussed in two directions to analyze the advantages, disadvantages, and applicability of different methods. Then, the results of Integrated Energy Systemss multiple load forecasting for the past 5 years are compiled and analyzed. Finally, the Integrated Energy Systems load forecasting is summarized and looked forward.

Rhizobium-driven symbiotic nitrogen-fixation in legumes not only benefits the growth but also enhances the stress tolerance of plants. Isolating and characterizing efficient, drought-tolerant rhizobia is a central goal for improving crop yields in arid regions. Here, we phylogenetically and biochemically characterized a novel strain of Rhizobium (‘QHCD11’) sampled from the root nodules of faba beans growing in an arid agricultural area in Qinghai-Tibet. We further tested the drought tolerance of the strain as well as of ‘Qingcan 14’ faba bean seedlings inoculated with it. Biochemical characterization involved bromothymol blue (BTB) tests, carbon metabolic profiling (Biolog GENIII), DNA–DNA hybridization (dDDH) assays, average nucleotide identity (ANI) analyses, and 16S rRNA sequencing. The result indicated that strain ‘QHCD11’ likely belongs to the Rhizobium indicum species. Drought stress tolerance was assessed by exposure to polyethylene glycol (PEG-6000) at concentrations of 0, 10, 15, and 20%. Increasing concentrations of PEG-6000 tended to result in decreased growth of ‘QHCD11’, although the strain performed better at 20% PEG 6000 than at 15%. Inoculation of drought-stressed faba bean seedlings with strain ‘QHCD11’ improved root vitality, chlorophyll content, antioxidant enzyme activities, and plant height. We suggest that inoculation of faba beans with ‘QHCD11’ is an environmentally sound strategy for mitigating crop drought stress in arid and semi-arid regions. In addition, the results presents here will benefit future studies into faba bean-rhizobia symbioses under drought stress.

The portmanteau test is an effective tool for testing the goodness of fit of models. Motivated by the fact that high-frequency data can improve the estimation accuracy of models, a modified portmanteau test using high-frequency data is proposed for ARCH-type models in this paper. Simulation results show that the empirical size and power of the modified test statistics of the model using high-frequency data are better than those of the daily model. Three stock indices (CSI 300, SSE 50, CSI 500) are taken as an example to illustrate the practical application of the test.

This study used the ITS approach based on Illumina MiSeq sequencing to assess the endosphere and rhizosphere fungal communities in healthy and diseased faba bean plants. The findings indicate that the most predominant phyla in all samples were Ascomycota (49.89–99.56%) and Basidiomycota (0.33–25.78%). In healthy endosphere samples, Glomeromycota (0.08–1.17%) was the only predominant phylum. In diseased endosphere samples, Olpidiomycota (0.04–1.75%) was the only predominant phylum. At the genus level, Penicillium (0.47–35.21%) was more abundant in rhizosphere soil, while Paraphoma (3.48–91.16%) was predominant in the endosphere roots of faba bean plants. Significant differences were observed in the alpha diversity of rhizosphere samples from different germplasm resources (p < 0.05). The fungal community structures were clearly distinguished between rhizosphere and endosphere samples and between healthy and diseased endosphere samples (p < 0.05). Saccharomyces was significantly enriched in diseased endosphere samples, whereas Apiotrichum was enriched in healthy endosphere samples. Vishniacozyma and Phialophora were enriched in diseased rhizosphere samples, while Pseudogymnoascus was enriched in healthy rhizosphere samples. Diseased samples displayed more strongly correlated genera than healthy samples. Saprotrophs accounted for a larger proportion of the fungal microbes in rhizosphere soil than in endosphere roots. This study provides a better understanding of the composition and diversity of fungal communities in the rhizosphere and endosphere of faba bean plants as well as a theoretical guidance for future research on the prevention or control of faba bean root rot disease.

Doumei 1 is a new cultivar of faba bean (Vicia faba L.) jointly bred by Chongqing Academy of Agricultural Sciences, Qinghai University, and Chongqing Agricultural Technology Extension Station. It has both horticultural and food functions. The flower color of this cultivar is pink and bright, with determinate inflorescence (Fig. 1A). During autumn sowing, the flowering period is ≈40 d in the field. ‘Doumei’ 1 is a new high-value horticulture crop (Fig. 2); the secondary branches are obvious (Fig. 1B), and it is suitable for mechanized harvesting because of its podding habit, with podding on the middle and upper nods (Fig. 1B). The emergence of faba bean cultivars with both horticultural and edible functions has endowed the traditional faba bean planting industry with landscape functions. This is essential for cross-industry innovation in faba bean breeding.

Abstract Semen Ziziphi Spinosae (SZS) has been extensively used in the daily diet as a functional food for neuroprotective health-benefit in China for many years. However, the neuroprotective mechanism of SZS associated with blood–brain barrier (BBB) integrity remains unexplored. The present study suggests SZS could protect against lipopolysaccharide (LPS)-induced BBB dysfunction. Proteomics indicate that 135 proteins in rat brain are significantly altered by SZS. These differentially expressed proteins are mainly clustered into cell–cell adhesion and adherens junctions, which are closely related with BBB integrity. SZS reversed LPS-induces BBB breakdown by activating the FAK-DOCK180-Rac1-WAVE2-Arp3 pathway. Molecular docking between signaling pathway proteins and identified SZS components in rat plasma reveals that 6”‘-feruloylspinosin, spinosin, and swertisin strongly binds to signaling proteins at multiple amino acid sites. These novel findings suggest a health benefit of SZS in prevention of cerebral diseases and contributes to the further application of SZS as a functional food.

Kentucky bluegrass (Poa pratensis L.) is an eminent turfgrass species with a complex genome, but it is sensitive to rust (Puccinia striiformis). The molecular mechanisms of Kentucky bluegrass in response to rust still remain unclear. This study aimed to elucidate differentially expressed lncRNAs (DELs) and genes (DEGs) for rust resistance based on the full-length transcriptome. First, we used single-molecule real-time sequencing technology to generate the full-length transcriptome of Kentucky bluegrass. A total of 33,541 unigenes with an average read length of 2,233 bp were obtained, which contained 220 lncRNAs and 1,604 transcription factors. Then, the comparative transcriptome between the mock-inoculated leaves and rust-infected leaves was analyzed using the full-length transcriptome as a reference genome. A total of 105 DELs were identified in response to rust infection. A total of 15,711 DEGs were detected (8,278 upregulated genes, 7,433 downregulated genes) and were enriched in plant hormone signal transduction and plant–pathogen interaction pathways. Additionally, through co-location and expression analysis, it was found that lncRNA56517, lncRNA53468, and lncRNA40596 were highly expressed in infected plants and upregulated the expression of target genes AUX/IAA, RPM1, and RPS2, respectively; meanwhile, lncRNA25980 decreased the expression level of target gene EIN3 after infection. The results suggest that these DEGs and DELs are important candidates for potentially breeding the rust-resistant Kentucky bluegrass.

Abstract Real-time mine ventilation network solution is the core way to realize the actual intelligent ventilation, and ventilation friction resistance coefficient is a significant parameter of network solution. With the help of fractal theory to characterize the three-dimensional roughness characteristics of tunnel surrounding rock. A method to describe the roughness by fractal dimension and fractal intercept. We put the fractal dimension and fractal intercept into Matlab to randomly generate three-dimensional laser scanning data of tunnels. The fusion of the two fractal parameters made the three-dimensional roughness surface information more comprehensive. It has been applied to field practice accurately. Compared to the simulation results show that the relative error of the new prediction results is 3%. Comprehensive evaluation analysis shows that the new friction wind resistance formula can fully reflect the influence of three-dimensional rough surfaces on airflow friction resistance. With the help of three-dimensional laser scanning technology, we can calculate the airflow friction resistance of the tunnel quickly and accurately, which provides a reference for the development of key technology and the theory of intelligent ventilation parameter measurement.

Globally, lung cancer is the leading cause of cancer-related mortality. Multiple primary lung cancers (MPLC) account for a very small portion of all primary lung cancer cases. Importantly, a quick and precise differentiation between MPLC and intrapulmonary metastases is directly related to patient prognoses as treatment strategies vary according to pathological type. Synchronous MPLC are most commonly seen in the same lung. Here, we report a rare case of a patient with synchronous MPLC of both lungs. A 67-year-old man, with a 1-month cough and expectoration history, was admitted in our hospital. Computed tomography (CT) chest scan revealed a lower lobe nodule in the left lung and an upper lobe nodule in the right lung. He underwent successive fiberoptic bronchoscopy and CT-guided percutaneous pulmonary aspiration biopsy of both lungs. The pathological diagnosis was squamous cell carcinoma of the left lung and adenocarcinoma of the right lung.

Background Faba bean (Vicia faba L) is one of the most important legumes in the world. However, there is relatively little genomic information available for this species owing to its large genome. The lack of data impedes the discovery of molecular markers and subsequent genetic research in faba bean. The objective of this study was to analyze the faba bean transcriptome, and to develop simple sequence repeat (SSR) markers to determine the genetic diversity of 226 faba bean varieties derived from different regions in China. Methods Faba bean varieties with different phenotype were used in transcriptome analysis. The functions of the unigenes were analyzed using various database. SSR markers were developed and the polymorphic markers were selected to conduct genetic diversity analysis. Results A total of 92.43 Gb of sequencing data was obtained in this study, and 133,487 unigene sequences with a total length of 178,152,541 bp were assembled. A total of 5,200 SSR markers were developed on the basis of RNA-Seq analysis. Then, 200 SSR markers were used to evaluate polymorphisms. In total, 103 (51.5%) SSR markers showed significant and repeatable bands between different faba bean varieties. Clustering analysis revealed that 226 faba bean materials were divided into five groups. Genetic diversity analysis revealed that the relationship between different faba beans in China was related, especially in the same region. These results provided a valuable data resource for annotating genes to different categories and developing SSR markers.

White clover (Trifolium repens L.) is one of the legume herbages with high feed quality, but it is sensitive to water deficiency. The objectives of this study were to compare morpho-physiological responses to drought stress and post-drought recovery in four-leafed white clover cultivars. Under well-watered conditions, drought stress (3 d, 6 d, 9 d and 12 d), and rehydration, the relative water content (RWC), membrane lipid permeability, osmoregulatory substances, photosynthetic characteristics and stomatal features of Chinese native Longping No.1 (LP, small-leafed) and three introduced cultivars, namely Pudi (PD, small-leafed), Rivendel (RD, medium-leafed) and Koala (KL, large-leafed), were evaluated. Results showed that small-leafed white clover maintained higher RWC and better membrane stability and osmotic regulation with increased drought intensities, compared to the medium-leafed and large-leafed cultivars. Following rewatering, small-leafed white clover recovered more rapidly with all of the parameters than the other cultivars. The increase of drought stress intensity led to the decline of net photosynthetic rate (Pn), transpiration rate (Tr) and stomatal conductance (Cs). However, the decreased range of Pn, Tr and Cs in the small-leafed white clover was significantly smaller than that in the medium-leafed and large-leafed white clovers. Meanwhile, the small-leafed white clover showed a smaller size and higher density of stoma either under normal or drought conditions than the intermediate- and large-leafed cultivars. Taken together, the results indicated that the adaptability of small-leafed white clover to drought stresses and post-drought recovery was better than that of medium-leafed and large-leafed white clovers. The study will provide better insights into the mechanism of drought response and recovery potential in different white clover cultivars.

Faba bean is an important member of legumes, which has richer protein levels and great development potential. Yield is an important phenotype character of crops, and early yield estimation can provide a reference for field inputs. To facilitate rapid and accurate estimation of the faba bean yield, the dual-sensor (RGB and multi-spectral) data based on unmanned aerial vehicle (UAV) was collected and analyzed. For this, support vector machine (SVM), ridge regression (RR), partial least squares regression (PLS), and k-nearest neighbor (KNN) were used for yield estimation. Additionally, the fusing data from different growth periods based on UAV was first used for estimating faba bean yield to obtain better estimation accuracy. The results obtained are as follows: for a single-growth period, S2 (12 July 2019) had the best accuracy of the estimation model. For fusion data from the muti-growth period, S2 + S3 (12 August 2019) obtained the best estimation results. Furthermore, the coefficient of determination (R2) values for RF were higher than other machine learning algorithms, followed by PLS, and the estimation effects of fusion data from a dual-sensor were evidently better than from a single sensor. In a word, these results indicated that it was feasible to estimate the faba bean yield with high accuracy through data fusion based on dual-sensor data and different growth periods.

Endogenous and exogenous estrogens are widely present in food and food packaging, and high levels of natural estrogens and the misuse or illegal use of synthetic estrogens can lead to endocrine disorders and even cancer in humans. Therefore, it is consequently important to accurately evaluate the presence of food-functional ingredients or toxins with estrogen-like effects. In this study, an electrochemical sensor based on G protein-coupled estrogen receptors (GPERs) was fabricated by self-assembly, modified by double-layered gold nanoparticles, and used to measure the sensing kinetics for five GPER ligands. The interconnected allosteric constants (Ka) of the sensor for 17β-estradiol, resveratrol, G-1, G-15, and bisphenol A were 8.90 × 10−17, 8.35 × 10−16, 8.00 × 10−15, 5.01 × 10−15, and 6.65 × 10−16 mol/L, respectively. The sensitivity of the sensor for the five ligands followed the order of 17β-estradiol > bisphenol A > resveratrol > G-15 > G-1. The receptor sensor also demonstrated higher sensor sensitivity for natural estrogens than exogenous estrogens. The results of molecular simulation docking showed that the residues Arg, Glu, His, and Asn of GPER mainly formed hydrogen bonds with -OH, C-O-C, or -NH-. In this study, simulating the intracellular receptor signaling cascade with an electrochemical signal amplification system enabled us to directly measure GPER–ligand interactions and explore the kinetics after the self-assembly of GPERs on a biosensor. This study also provides a novel platform for the accurate functional evaluation of food-functional components and toxins.

Novel germplasm resources are the key to crop breeding, with their genetic diversity and population structure analysis being highly significant for future faba bean breeding. We genotyped 410 global faba bean accessions using the 130K targeted next-generation sequencing (TNGS) genotyping platform, resulting in a total of 38,111 high-quality SNP loci by high-standard filtering. We found the polymorphism information content (PIC) and Nei’s gene diversity were 0.0905–0.3750 and 0.0950–0.5000, with averages of 0.2471 and 0.3035, respectively. After evaluating the genetic diversity of 410 accessions using Nei’s gene diversity and PIC, on the basis of their geographical origin (continent) and structure-analysis-inferred subpopulations, we found that the faba bean accessions from Asia (except China) and Europe had rich genetic diversity, while those from the winter sowing area of China were low. The 410 faba bean accessions were divided into four subpopulations according to population structure analysis and clustering analysis based on Nei’s (1972) genetic distance using the neighbor-joining (NJ) method. However, the same subpopulation contained materials from different geographical origins, thereby indicating that the gene flow or introgression occurred among the accessions. Results from NJ clustering based on shared allele genetic distance indicated that the 410 accessions were divided into three groups according to their dissemination routes. The genetic diversity analysis results demonstrated that the genetic relationships among the faba bean groups with similar ecological environments and geographic origins in neighboring regions or countries were closer and frequently found within the same group, while genetic variation among individuals was the main source of their total genetic variation.

In this study, an electrochemical sensor was developed by immobilizing colon cancer and the adjacent tissues (peripheral healthy tissues on both sides of the tumor) and was used to investigate the receptor sensing kinetics of glucose, sodium glutamate, disodium inosinate, and sodium lactate. The results showed that the electrical signal triggered by the ligand–receptor interaction presented hyperbolic kinetic characteristics similar to the interaction of an enzyme with its substrate. The results indicated that the activation constant values of the colon cancer tissue and adjacent tissues differed by two orders of magnitude for glucose and sodium glutamate and around one order of magnitude for disodium inosinate. The cancer tissues did not sense sodium lactate, whereas the adjacent tissues could sense sodium lactate. Compared with normal cells, cancer cells have significantly improved nutritional sensing ability, and the improvement of cancer cells’ sensing ability mainly depends on the cascade amplification of intracellular signals. However, unlike tumor-adjacent tissues, colon cancer cells lose the ability to sense lactate. This provides key evidence for the Warburg effect of cancer cells. The methods and results in this study are expected to provide a new way for cancer research, treatment, the screening of anticancer drugs, and clinical diagnoses.

Bone formation is critically needed in orthopedic clinical practice. We found that, bone morphogenetic protein-7 (BMP-7) gene expression was significantly increased in fractured mice, which activates canonical Wnt signaling exclusively in osteocytes. Wnt and BMP signaling appear to exhibit synergistic or antagonistic effects in different kinds of cells. However, the communication between Wnt/β-catenin signaling and BMP signaling in osteocytes is almost unknown. Our study verified in vitro that BMP-7 expression was significantly increased when Wnt signaling was activated in osteocytes. Next, BMP-7 in osteocytes was overexpressed using an adenovirus, the osteogenesis of bone marrow stem cells (BMSCs) was enhanced, when cocultured with osteocytes. On the contrary, BMP-7 in osteocytes was silenced using an adenovirus, the osteogenesis of bone marrow stem cells (BMSCs) was weakened. In addition, the osteogenesis of BMSCs was no longer promoted by Wnt-activated osteocytes when BMP-7 was silenced. Therefore, the results showed that BMP-7 mediated the anabolic actions of Wnt/β-catenin signaling in osteocytes. Our study provides new evidence for the clinical application of BMP-7-overexpressed osteocytes.

In March 2020, the World Health Organization (WHO) declared COVID-19 a pandemic, and the spike protein has been reported to be an important drug target for anti-COVID-19 treatment. As such, in this study, we successfully developed a novel electrochemical receptor biosensor by immobilizing the SARS-CoV-2 spike protein and using AuNPs-HRP as an electrochemical signal amplification system. Moreover, the time-current method was used to quantify seven antiviral drug compounds, such as arbidol and chloroquine diphosphate. The results show that the spike protein and the drugs are linearly correlated within a certain concentration range and that the detection sensitivity of the sensor is extremely high. In the low concentration range of linear response, the kinetics of receptor–ligand interactions are similar to that of an enzymatic reaction. Among the investigated drug molecules, bromhexine exhibits the smallest Ka value, and thus, is most sensitively detected by the sensor. Hydroxychloroquine exhibits the largest Ka value. Molecular docking simulations of the spike protein with six small-molecule drugs show that residues of this protein, such as Asp, Trp, Asn, and Gln, form hydrogen bonds with the -OH or -NH2 groups on the branched chains of small-molecule drugs. The electrochemical receptor biosensor can directly quantify the interaction between the spike protein and drugs such as abidor and hydroxychloroquine and perform kinetic studies with a limit of detection 3.3 × 10−20 mol/L, which provides a new research method and idea for receptor–ligand interactions and pharmacodynamic evaluation.

Evergestis extimalis (Scopoli) is a significant pest of spring oilseed rape in the Qinghai-Tibet Plateau. It has developed resistance to many commonly used insecticides. Therefore, biopesticides should be used to replace the chemical pesticides in pest control. In this study, the effects of ice-nucleation-active (INA) microbes (Pseudomonas syringae 1.7277, P. syringae 1.3200, and Erwinia pyrifoliae 1.3333) on E. extimalis were evaluated. The supercooling points (SCP) were markedly increased due to the INA bacteria application when they were compared to those of the untreated samples. Specifically, the SCP of E. extimalis after its exposure to a high concentration of INA bacteria in February were −10.72 °C, −13.73 °C, and −14.04 °C. Our findings have demonstrated that the trehalase (Tre) genes were up-regulated by the application of the INA bacteria, thereby resulting in an increased trehalase activity. Overall, the INA bacteria could act as effective heterogeneous ice nuclei which could lower the hardiness of E. extimalis to the cold and then freeze them to death in an extremely cold winter. Therefore, the control of insect pests with INA bacteria goes without doubt, in theory.

Advances in crop breeding techniques and economic evaluation are critical to ensuring and improving crop yields and sustainable development. Based on the provincial data on food legumes breeding (FLB) in China from 2001 to 2020, the research and development (R&D) expenditure distribution, FLB contribution rate, and R&D expenditure on FLB were estimated. The economic benefits of output per unit area and R&D expenses were evaluated, and relevant policy suggestions were put forward. The study found that: (i) the R&D expenditure on FLB’s showed a significant upward trend, and the development can be divided into a starting, stable, and rapid growth period. There were significant differences in R&D expenditures across eight provinces of China with relatively high R&D expenditures for FLB; (ii) the R&D expenditure on FLB had a significant lag in the improvement of per mu yield (1 mu = 0.067 hectares). For every 10,000 yuan increase in R&D funding for FLB, the yield per mu will increase by 0.145 kg/mu in the next five years, and the regional spillover effect of breeding costs is significant; and (iii) the marginal revenue of R&D expenditures FLB from 2008 to 2020 is 34.91 yuan, that is, every 1 yuan invested in breeding R&D can bring a short-term marginal revenue of 34.91 yuan and a long-term marginal revenue of 337.23 yuan. Based on the above analysis, some recommendations were proposed and discussed, including further increasing the R&D investment of FLB, strengthening industry–university–research cooperation in breeding, improving the breeding research system, and promoting the multi-dimensional development of FLB industry–university–research services.

Growth factors were often used to improve the bioactivity of biomaterials in order to fabricate biofunctionalized bone grafts for bone defect repair. However, supraphysiological concentrations of growth factors for improving bioactivity could lead to serious side effects, such as ectopic bone formation, radiculitis, swelling of soft tissue in the neck, etc. Therefore, safely and effectively applying growth factors in bone repair biomaterials comes to be an urgent problem that needs to be addressed. In this study, an appropriate concentration (50 ng/mL) of Wnt3a was used to pretreat the 3D-bioprinting gelatin methacryloyl(GelMA)/polycaprolactone(PCL) scaffold loaded with bone marrow stromal cell line ST2 for 24 h. This pretreatment promoted the cell proliferation, osteogenic differentiation, and mineralization of ST2 in the scaffold in vitro, and enhanced angiogenesis and osteogenesis after being implanted in critical-sized mouse calvarial defects. On the contrary, the inhibition of Wnt/β-catenin signaling in ST2 cells reduced the bone repair effect of this scaffold. These results suggested that ST2/GelMA/PCL scaffolds pretreated with an appropriate concentration of Wnt3a in culture medium could effectively enhance the osteogenic and angiogenic activity of bone repair biomaterials both in vitro and in vivo. Moreover, it would avoid the side effects caused by the supraphysiological concentrations of growth factors. This functionalized scaffold with osteogenic and angiogenic activity might be used as an outstanding bone substitute for bone regeneration and repair.

Biosensors are powerful analytical tools used to identify and detect target molecules. Electrochemical biosensors, which combine biosensing with electrochemical analysis techniques, are efficient analytical instruments that translate concentration signals into electrical signals, enabling the quantitative and qualitative analysis of target molecules. Electrochemical biosensors have been widely used in various fields of detection and analysis due to their high sensitivity, superior selectivity, quick reaction time, and inexpensive cost. However, the signal changes caused by interactions between a biological probe and a target molecule are very weak and difficult to capture directly by using detection instruments. Therefore, various signal amplification strategies have been proposed and developed to increase the accuracy and sensitivity of detection systems. This review serves as a reference for biosensor and detector research, as it introduces the research progress of electrochemical signal amplification strategies in olfactory and taste evaluation. It also discusses the latest signal amplification strategies currently being employed in electrochemical biosensors for nanomaterial development, enzyme labeling, and nucleic acid amplification techniques, and highlights the most recent work in using cell tissues as biosensitive elements.

The accurate and reliable monitoring of ventilation parameters is key to intelligent ventilation systems. In order to realize the visualization of airflow, it is essential to solve the airflow reconstruction problem using few sensors. In this study, a new concept called independent cut set that depends on the structure of the underlying graph is presented to determine the minimum number and location of sensors. We evaluated its effectiveness in a coal mine owned by Jinmei Corporation Limited (Jinmei Co., Ltd., Shanghai, China). Our results indicated that fewer than 30% of tunnels needed to have wind speed sensors set up to reconstruct the well-posed airflow of all the tunnels (>200 in some mines). The results showed that the algorithm was feasible. The reconstructed air volume of the ventilation network using this algorithm was the same as the actual air volume. The algorithm provides theoretical support for flow reconstruction.

Reservoir sedimentation affects lifespan, hydropower generation, water resource use, and ecosystem sustainability. In this study, using measured data from the worldrenowned Three Gorges Reservoir (TGR), sediment transport capacity was analysed from 2002 to 2020 at four TGR gauging stations (Cuntan, Qingxichang, Wanxian, and Miaohe). The result indicated that: (1) a significant decrease in sediment transport capacity occurred in the TGR before and after dam construction, with an overall reduction of 82% in the perennial backwater zone. Notably, Qingxichang, Wanxian, and Miaohe experienced reductions of 40%, 91%, and 59%, respectively. Since the initial operation phase, their maximum monthly sediment transport capacity has consistently occurred in July. (2) Changes in the water level in front of the dam, the incoming flow, and sediment influxes were the primary factors driving the annual variations in sediment transport capacity. Among the factors affecting the variation of sediment transport capacity along the TGR, the influence of the cross-sectional morphology was more pronounced than that of the distance from the dam. Sediment transport capacities in the wide and shallow sections were considerably lower than those in the narrow and deep sections. (3) A sediment transport amplitude index (Fd) was proposed to assess the sediment dynamics within the TGR, which effectively characterized the sedimentation conditions based on the relative importance of these influencing factors. These findings provide a theoretical basis for operational strategies in the TGR and offer insights into the patterns of sediment transport capacity changes in reservoir areas before and after dam construction in other river-type reservoirs. Sulfhydryl attapulgite is an effective material for soil remediation, but its potential toxicity to animals in soil is not fully understood. Using Eisenia fetida as the test organism, the avoidance experiments, acute toxicity and cumulative toxicity experiments were carried out. The avoidance rate of Eisenia fetida on ATP-SH is 0–60%. Eisenia fetida not only avoid, but also tend to the soil-added ATP-SH to a certain degree. In the acute toxicity test, with the increase of ATP-SH addition, the total protein (T-Protein) content gradually decreases, and the maximum rate of decline is 51.18%. The catalase (CAT) and superoxide dismutase (T-SOD) activities gradually increase, and the maximum increase rates are 73.56% and 51.73%, respectively. The malondialdehyde (MDA) activity significantly increase 1.8 times. In the cumulative toxicity experiment, with the increase of ATP-SH addition, the maximum increase rate of T-Protein content is 31.52%. The CAT and T-SOD activities decrease 64.38% and 43.60% at most, respectively, and MDA activity increase 0.9 times. Following exposure to the soil-added ATP-SH for 30 days, Eisenia fetida adapt to the environment through its own regulatory mechanism.
