3. Method development and validation for analysis of microencapsulated cyanidin‐3‐O‐rutinoside in dairy samples containing juçara palm fruit by high‐performance liquid chromatography.

Euterpe edulis is a plant native of Brazil and popularly known as juçara. The aim of this study was to develop and validate an analytical method using high‐performance liquid chromatography (HPLC) of juçara crude extracts (JCEs) and to evaluate dairy products containing free and microencapsulated extracts. The validated analytical method for JCE was performed using cyanidin‐3‐O‐rutinoside (C3R – isolated and identified as major component from JCE) as a reference: linearity was obtained using ten concentrations between 25 and 250 μg mL−1 (R2 = 0.994); limits of detection and quantification were 19.04 μg mL−1 and 57.7 μg mL−1 respectively; accuracy (relative standard deviation) was <5%; recovery was 104.85 μg mL−1; and the method is robust. Physicochemical analyses of the dairy products were evaluated using colour, pH, and concentration of anthocyanins using HPLC calculated as C3R over 21 days of storage. The pH remained stable until the end of the test. The colour and the concentration of anthocyanins remained stable for 9 days. C3R was isolated, the HPLC method was developed and validated, and the colour stability was not altered until 9 days. © 2023 Society of Chemical Industry. [ABSTRACT FROM AUTHOR]

Os glicosaminoglicanos (GAG´s) são polissacarídeos aniônicos, lineares, compostos basicamente por hexosamina, ácido urônico e galactose. São eles o sulfato de condroitina (CS), sulfato de dermatana (DS), ácido hialurônico (HA), sulfato de queratana (KS), heparina (HE) e sulfato de heparana (HS). A diferença de densidade de cargas negativas e o grau de sulfatação determinam suas propriedades estruturais e biológicas. As propriedades terapêuticas dos GAG´s estão correlacionadas com sua capacidade de se ligar as proteínas. Os GAG´s sulfatados têm sido amplamente utilizados como anticoagulantes, com destaque para a heparina (HE). Diversos estudos também apontam para atividades regenerativa, antiviral, antiproliferativa e anti-inflamatória. A crescente demanda por GAG´s está impulsionando uma série de pesquisas para a descoberta de novas fontes de isolamento. Os subprodutos da indústria de alimentos são uma potencial fonte para obtenção de glicosaminoglicanos.

Nowadays, new leishmanicidal drugs are needed and natural products arise as a promising alternative source. Therefore, bioguided fractionation of a hydroethanolic extract from the stem bark of Croton echioides Baill. were conducted based on its antileishmanial activity. Two novel neo-clerodane diterpenoids methyl-15,16-epoxy-3,13(16),14-neo-clerodatrien-17,18-dicarboxylate (1) and dimethyl-3-oxo-15,16-epoxy-13(16),14-neo-clerodadien-17,18-dicarboxylate (2) were isolated, as well as four known compounds (3–6) and lupeol, from the hexane fraction. Their structures were established by NMR analysis. The crude extract, fractions and the compounds (1 and 3–6) were evaluated for their in vitro antileishmanial activity and cytotoxicity against macrophages J774A.1. The selectivity index (SI) were calculated. The most active compound against promastigote forms of L. amazonensis was the clerodane diterpene 4, with IC
50 values of 8.3 µM and SI value of 80.9. Our results highlighted stem bark of Croton echioides Baill. as a promising source for the development of a new chemotherapeutic agent to combat leishmaniasis. [ABSTRACT FROM AUTHOR]- Published

Glicosaminoglicanos (GAGs) são polissacarídeos aniônicos, compostos por unidades dissacarídica repetitivas de hexosamina, ácido urônico ou galactose. Suas propriedades estruturais e biológicas estão associadas à densidade de cargas negativas na molécula. Há relatos dessas estruturas estarem presentas nas mais variadas espécies, inclusive nos peixes. A produção de pescado tem alcançado números cada vez mais expressivos e o Brasil tem se destacado no cenário internacional. A espécie de maior volume de produção no país é a tilápia, e o Paraná é o estado com maior volume de produção nacional. Atrelado a esse aumento de produção e consumo, está à geração de subprodutos. As escamas representam grade parte desse material quando se considera o volume de tilápia que tem sido processado. Alternativas para o aproveitamento efetivo das escamas representam hoje, um dos maiores desafios para as plantas processadoras. O trabalho teve como objetivo, utilizar escamas de tilápia como matéria prima para a extração, isolamento e caracterização de GAGs. A extração foi realizada por hidrolise enzimática com papaína, a uma temperatura de 60 °C, durante 14 dias. Após a extração realizou-se a purificação em coluna de troca iônica. As frações obtidas com a purificação foram agrupadas de acordo com os perfis obtidos pelas análises de condutividade elétrica e pelas determinações de ácido urônico e GAGs sulfatados. De todas as frações obtidas cromatograficamente, foram agrupadas seis novas frações, destas, as que apresentaram picos majoritários nas determinações anteriormente citadas, foram caracterizadas por eletroforese em gel de agarose. Os resultados de eletroforese e ressonância magnética nuclear (RMN) indicaram a presença de GAGs sulfatados na amostra. Com base nos espectros de RMN, a fração V (F- V) apresentou maior grau de pureza, possibilitando desse modo a identificação de sinais característicos para GAGs sulfatados. Foi encontrada uma mistura de sulfato de condroitina 4 (SC4) e sulfato de condroitina 6 (SC6). Essa fração foi testada quanto ao grau de substituição (GS) por grupamentos sulfato e apresentou GS de 0,146. A F – V apresentou atividade anticoagulante, sendo a ação mais efetiva em maiores concentrações. Quando testada na presença na presença de protamina, a F – V apresentou um efeito pró-anticoagulante. A fração apresentou efeito citotóxico/antitumoral frente às células metabolizadoras, derivadas de hepatoma de Rattus norvegicus, demonstrando o seu potencial de aplicação para essa finalidade. Assim, a extração de sulfato de condroitina de escamas de tilápia pode ser uma alternativa para a destinação desse subproduto, gerando benefícios econômicos, sociais e ambientais. Glycosaminoglycans (GAGs) are anionic polysaccharide composed of repetitive disaccharide units of hexosamine, uronic acid or galactose. Its selected and biological properties are associated with the density of negative charges in the molecule. There are reports of these structures present in the most varied species, including fish. The production of fish has reached increasingly expressive numbers and Brazil stands out on the international scene. A species with the highest production volume in the country is tilapia, and Paraná is the state with the highest national production volume. Linked to this increase in production and consumption, it is the generation of by-products. The scales represent the part of this material when considering the volume of inclination that was processed. The alternatives for the effective use of the scales represent today, one of the biggest challenges for the processing plants. The work aimed to use tilapia scales as raw carried out by enzymatic hydrolysis with papain, at a temperature of 60 °C, for 14 days. After extraction, the ion exchange column was purified. The fractions obtained with the purification were grouped according to the profiles obtained by the electrical conductivity analyzes and by the determinations of uronic acid and sulfated GAGs. Of all fractions obtained chromatographically, six new fractions were grouped, of these those that showed major peaks in the determinations previously mentioned, were characterized by electrophoresis in agarose gel. The results of electrophoresis and nuclear magnetic resonance (NMR) indicated the presence of sulfated GAG´s in the sample. Based on the NMR spectra, the V fraction (F – V) showed a higher degree of purity, thus enabling the identification of characteristic signals for sulphated GAGs. A mixture of chondroitin sulfate 4 (SC4) and chondroitin sulfate 6 (SC6) was found. This fraction was tested for the degree of substitution (GS) by sulphate groups and presented as GS of 0,146. F –V showed anticoagulant activity, being the most effective actions in higher concentrations. When tested in the presence of protamine, F- V a had a pro-anticoagulant effect. The fraction showed a cytotoxic/antitumor effect against metabolizing cells, derived from Rattus norvegicus hepatoma, thus demonstrating its potential for application for this purpose. The extraction of chondroitin sulfate from tilapia scales can be an alternative for the destination of this by-product, generation economic, social and environmental benefits.

12. Nitrogen fertilization in the production of two varietes of purple corn maize cultivated in the south of Brazil

Nos sistemas agroecológicos do Brasil é comum o uso de variedades crioulas em sistemas convencionais. O milho roxo é um cereal cultivado nas zonas Inter Andinas, porém também pode ser desenvolvido no estado de Paraná, mas pouco se sabe sobre o comportamento destas variedades em resposta a níveis de adubação nitrogenada. Este trabalho foi realizado na safra 2018/2019, na área experimental da UTFPR-Pato Branco, nas coordenadas 26º41’17” Sul e 52º41’17” Oeste. O objetivo foi avaliar os componentes agronômicos (altura da planta-AP; altura da inserção da espiga-AIE; comprimento da espiga-CE ; diâmetro da espiga-DE; número de grãos por fileira-NGF; número de fileiras por espiga-NFE; massa de 1000 grãos-MMG; produtividade-PROD) e alguns componentes físico-químicos (antocianinas, índice de atividade antioxidante-IAA e fenóis totais-FT) de duas variedades do milho roxo (UNC-47 e INA-601), em resposta a aplicação de diferentes doses de nitrogênio (0; 50; 100; 150 kg N ha-1) em cobertura, usando-se ureia (46%) como fonte. O delineamento experimental foi de blocos ao acaso, com quatro repetições. Os resultados indicam que as variáveis AP, AIE, CE, DE, NGF, NFE, NGE, MMG e PROD respondem positivamente em relação as doses de N. Na avaliação dos componentes orgânicos nitrogenados como FT e IAA dos grãos e sabugos verifica-se teores expressivos positivos, em frações de mg EAG/g. e de inibição do DPPH, evidenciando a correlação significativa estre as duas variáveis. Entretanto a adubação nitrogenada, para as antocianinas monoméricas totais (cy-3-glu/g), promoveu a diminuição e interferiu de forma antagônica na qualidade da síntese dos pigmentos antocianínicos do milho roxo. In Brazilian agroecological systems, the use of Creole varieties in conventional systems is common. Purple corn is a cereal grown in the Inter Andean areas, but it can also be developed in the state of Paraná. Little is known about the behavior of these varieties in response to nitrogen fertilization levels. This work was carried out from October to January 2018/2019 in a production system located in the experimental area of UTFPR-Pato Branco, at coordinates 26º41'17 ”South and 52º41’17” west. The objective was to evaluate the agronomic components (height of the plant-AP; height of the insertion of the ear-EIA; length of the ear-CE; diameter of the ear-DE; number of grains per row-NGF; number of rows per ear- NFE; mass of 1000 grains-MMG; productivity-PROD) and some physicochemical components (total phenols-FT, antioxidant activity index-IAA and anthocyanins) of two varieties of purple corn (UNC-47 and INA-601), at the stage V4 with application of different doses of nitrogen (0; 50; 100; 150 kg ha-1), using urea (46%) as a source. The experimental design was randomized blocks, with four replications. The results indicate that the variables AP, AIE, CE, DE, NGPF, NFPE, NGPE, PMS and PROD. respond positively in relation to the doses of N. In the evaluation of the nitrogenous organic components such as total phenols and antioxidant activity index of grains and cobs, there are expressive positive levels, in fractions of mg EAG / g. and DPPH inhibition, showing a significant correlation between the two variables. However, nitrogen fertilization, for total monomeric anthocyanins (cy- 3-glu / g), promoted the decrease and interfered in an antagonistic way in the quality of the synthesis of anthocyanin pigments in purple corn.
