Abstract Background The extent of natural salt-laden groundwaters used for irrigation is increasing worldwide, which is a fast-emerging threat to agroecosystems and global food security. The salt buildup in the soil is linked to deteriorated soil chemical, physical, and biological health and decreased land productivity. Alkali waters with high residual sodium carbonate (RSC) is one of the severe poor-quality waters that deteriorate soil. We evaluated soil microbial dynamics and soil health at critical growth stages of rice crop receiving two-decade-long irrigation with three levels of alkali water and two reclamation strategies. These included good-quality water (GQW), alkali water (ALKW) with an RSC concentration of 5 me L⁻1 (ALKW1), ALKW with an RSC concentration of 10 me L⁻1 (ALKW2), ALKW2 treated to neutralize RSC to 5 me L⁻1 using gypsum (CaSO4·2H2O; ALKW2 + GYP), and ALKW2 treated with sulfuric acid (H2SO4; ALKW2 + SA). Eleven microbial parameters were used to develop a soil microbial activity index (SMAI), and eight soil health indicators were correlated with changes in SMAI and crop productivity. Results The SMAI peaked under good-quality water (GQW) conditions (0.84–0.89), while the lowest values were recorded under ALKW2 (0.06–0.18). Neutralized alkali waters, ALKW2 + SA and ALKW2 + GYP, significantly improved SMAI with corresponding values of 0.25–0.35 and 0.13–0.32, respectively. SMAI across all stages correlated positively (R 2 = 0.91–0.98) with rice yield. Microbial activity varied with the crop growth stage, peaking at tillering. Gypsum application alone, aimed at neutralizing alkalinity from an RSC of 10 to 5 me L⁻1, proved insufficient in bringing the SMAI up to the ALKW1 (RSC level of 5 me L⁻1). Conclusions The application of dilute sulfuric acid demonstrated better results in restoring the soil microbial activity index than gypsum amendment; however, sulfuric acid treatment depends on native calcium carbonate (CaCO3) dissolution for its effectiveness. It may not suffice for soil stability improvement in the long term, especially when native CaCO3 is low. Therefore, integrating gypsum and dilute sulfuric acid for RSC neutralization is worthwhile; however, further assessment is needed to confirm their combined impact on soil biochemical and physical properties.

ABSTRACTIn this research work, we explore antibacterial activity and the UV-protection of cotton fabric functionalized with copper oxide-titanium dioxide nano composites particles (CuO+TiO2 NCPs). Cotton fabrics (Cot) were treated with L-methionine (Am) and functionalized with bimetallic CuO+TiO2 NCPs by pad and rolling technique. Cu(NO3)2 and titanium isopropoxide (TIP) were used as sources of Cu(OH)2 and TiO2 colloid respectively, which were then converted into CuO and TiO2 nanoparticles (NPs) for making antibacterial and UV-protective cotton. The method yielded a crystallite size of CuO and TiO2 NPs of 18.09 and 32.5 nm, respectively. FTIR, Raman spectroscopy, High-resolution scanning electron microscopy-Energy dispersive Spectroscopy (HRSEM), (EDS), X-ray photo spectroscopy (XPS), thermal gravimetric analysis (TGA), and XRD were used to analyze the functional group, morphology, chemical composition, thermal stability and crystalline structure of the functionalized cotton fabric samples. The results showed that an esterification reaction occurred between Am and OH in cotton fabric, which further enhanced the binding of NCPs. AmCot-CuO+TiO2 NCPs exhibit outstanding antibacterial activity against gram-positive S. aureus and gram-negative E. coli. The UV protection factor (UPF) for AmCot-CuO+TiO2 NCPs reached 50+; thus, multifunctional cotton fabrics with excellent antibacterial and anti-ultraviolet properties were obtained.

Metering fluids is critical in various industries, and researchers have extensively explored factors affecting measurement accuracy. As a result, numerous sensors and methods are developed to precisely measure volume fractions in multi-phase fluids. A significant challenge in multi-phase fluid pipelines is the formation of scale within the pipes. This issue is particularly problematic in the petroleum industry, leading to narrowed internal diameters, corrosion, increased energy consumption, reduced equipment lifespan, and, most crucially, compromised flow measurement accuracy. This paper proposes a non-destructive metering system incorporating an artificial neural network with capacitive and photon attenuation sensors to address this challenge. The system simulates scale thicknesses from 0 mm to 10 mm using COMSOL multiphysics software and calculates counted rays through Beer Lambert equations. The simulation considers a 10% interval of volume variation in each phase, generating 726 data points. The proposed network, with two inputs—measured capacity and counted rays-and three outputs—volume fractions of gas, water, and oil—achieves mean absolute errors of 0.318, 1.531, and 1.614, respectively. These results demonstrate the system’s ability to accurately gauge volume proportions of a three-phase gas-water-oil fluid, regardless of pipeline scale thickness.

Polyhalite-based POLY4, a multi-nutrient source containing potassium, calcium, magnesium, and sulphur, is increasingly recognised for its potential to improve crop yields and soil health in agricultural systems. It is also been considered as a feasible approach for addressing the deficiency in potassium, calcium, and sulphur within a single application source. The present study aimed to investigate the impact of polyhalite-based POLY4 application, either as a complete or partial substitute for traditional potassium fertiliser and gypsum supplement, on the improvement of peanut (Arachis hypogaea) growth and soil quality. An extensive field study was conducted from 2021 and 2022, employing ten distinct nutrient management treatments with three replications in a randomised complete block design. The findings of the study indicated that the application of polyhalite (POLY4) in conjunction with only NP fertilisers resulted in a higher yield advantage (approximately 150–200 kg ha-1) than in plots treated with NPK + gypsum (at 500 kg ha-1) and control plots. The application of polyhalite-based fertiliser (POLY4) at a rate that was 100% equivalent to K along with NP fertilisers resulted in a significant increase in pod yield (5.3–12.8%) over NPK + gypsum and control plots. Thus, the increased crop yield led to an increase in gross returns of 4.88% and in net returns of 4.28% with the application of POLY4 (100%) + NP fertilisers over other treatments. Likewise, variable rates of conventional fertilisers along with POLY4 (100% recommended) + NP + gypsum at 310 kg/ha significantly increased the linoleic acid content (38.5%), oleic acid content, and oil content (48.1%) by reducing palmitic acid (11.96%) content in the groundnut seed. Interestingly, POLY4 use at the 50% recommended rate also resulted in yields that were comparable with those obtained with 100% NPK. Therefore, applying POLY4, a polyhalite fertiliser, in either a 100% or 50% equivalent of essential K can be an effective way of increasing the production of peanut crops and promoting agricultural sustainability.

Introduction: Altered DNA methylation pattern in sperms has been associated with infertility in males demonstrating defective spermatogenesis or low semen quality. Vitamin B-12, by affecting 1-carbon metabolism pathways, might alter the DNA methylation pattern. We aimed to study the correlation of serum vitamin B12 levels with aberrant DNA methylation in infertile male patients. Methods: A cross-sectional study was conducted on 17 oligozoospermic infertile males (WHO criteria, 2010) and 10 healthy fertile males. Serum vitamin B12 levels were estimated using the chemiluminescence method. Global methylation was determined using the ELISA system (Imprint Methylated DNA Quantification Kit, Sigma–Aldrich). The levels of global DNA methylation were calculated and compared relative to the methylated (100%) control DNA provided with the kit. Results: Mean serum vitamin B12 concentration in the control group was higher than that of the case group. This difference in serum vitamin B12 concentration in both groups was found statistically significant. Although the results of this study show that oligozoospermic men have relatively lower global DNA methylation as compared to normozoospermic control, the values could not reach a statistically significant level. A small positive correlation was found between serum vitamin B12 levels and percent methylation defect (r = 0.14) but was statistically insignificant. Conclusion: Our study concludes that oligozoospermic infertile males have a significant deficiency of vitamin B12 as compared to normozoospermic fertile males. This study did not find any significant difference in global DNA methylation between the two groups. The present study does not suggest any correlation between serum vitamin B12 level and percent DNA methylation.

Objective: To determine the effectiveness of the telemedicine-based pulmonary rehabilitation programme in COVID-19 pneumonitis. Design: Prospective intervention study. Setting: Rehabilitation outpatient department, Tertiary-Care institute. Participants: Consecutive sample of patients (N = 50) in recovered COVID-19 infection. Intervention: Six weeks of telemedicine-based pulmonary rehabilitation in recovered patients of COVID-19 infection. Outcome Measures: All patients were clinically assessed by six minutes walk test (6MWT), Modified Medical Research Council Scale (mMRC), 30s-STS and SF 36 at zero week and six weeks post-intervention. Parthenium hysterophorus (PH), often known as parthenium weed, is one of the seventh most devastating weeds in the world. The primary aim of the study was to produce and optimize fungal FPase enzymes under solid-state fermentation (SSF) conditions for application in enzymatic hydrolysis to produce bioethanol. The impact of various physiological factors was verified through a systematic approach, initially using the one-factor-at-a-time (OFAT) method. Further optimization was carried out using statistical tools by response surface methodology (RSM), selecting three variables, i.e., lactose concentration, peptone concentration, and pH level. The outcomes of this research demonstrated that the RSM-based model led to the optimization of enzyme activity, resulting in 4.08U/ml of FPase, with a lactose concentration of 1.62 %, a peptone concentration of 1.5 %, and a pH level of 6 being the ideal conditions. When employing the FPase enzyme for biomass hydrolysis, the maximum yield of reducing sugar, at 0.37 ± 0.01 g/g, was achieved within a 96-hour period, using enzyme and substrate levels of 30 FPU/100 ml and 3 % (w/v), respectively. Fermentation of enzymatic hydrolysate was carried out by Saccharomyces cerevisiae at 30 °C, pH 6.0, and 120 rpm, which resulted in 0.45g/g ethanol yield after 48 h. Overall, our findings reveal that PH biomass can be introduced into sugar-based biorefineries to produce biofuels and other valuable chemicals.

Postoperative hypocalcemia is an important complication of thyroid surgery. The present study was undertaken to measure the serum levels of intact parathormone (iPTH) and corrected serum calcium (CSC) in the immediate postoperative period and then sequentially till 1 month after thyroid surgery. A total of 36 patients undergoing total thyroidectomy and 44 undergoing hemithyroidectomy had measurement of the level of iPTH and CSC at 1 hour, 1 day, 1 week, and 1 month after surgery. A mean drop of 9.3% in CSC, 40% in iPTH, and 10% in ionic calcium levels was noted 1 hour after total thyroidectomy. All the patients recovered to near preoperative levels at the 1-month follow-up. Among hemithyroidectomy patients, significant postsurgery drop in levels was not observed. The importance of the study is early recognition of a hypoparathyroid state at 1 hour after surgery and institution of calcium replacement, thereby sparing the patient from unpleasant symptoms of hypocalcemia. Furthermore, patients with a drop in the iPTH levels below the defined hypoparathyroidism levels should have careful evaluation of their thyroidectomy specimen for identification and possible autotransplantation of the parathyroid gland intraoperatively or in the immediate postoperative time frame.

IntroductionIt is becoming clearer that the microbiota helps drive responses to vaccines; however, little is known about the underlying mechanism. In this study, we aimed to identify microbial features that are associated with vaccine immunogenicity in infant rhesus macaques.MethodsWe analyzed 16S rRNA gene sequencing data of 215 fecal samples collected at multiple timepoints from 64 nursery-reared infant macaques that received various HIV vaccine regimens. PERMANOVA tests were performed to determine factors affecting composition of the gut microbiota throughout the first eight months of life in these monkeys. We used DESeq2 to identify differentially abundant bacterial taxa, PICRUSt2 to impute metagenomic information, and mass spectrophotometry to determine levels of fecal short-chain fatty acids and bile acids.ResultsComposition of the early-life gut microbial communities in nursery-reared rhesus macaques from the same animal care facility was driven by age, birth year, and vaccination status. We identified a Sutterella and a Rodentibacter species that positively correlated with vaccine-elicited antibody responses, with the Sutterella species exhibiting more robust findings. Analysis of Sutterella-related metagenomic data revealed five metabolic pathways that significantly correlated with improved antibody responses following HIV vaccination. Given these pathways have been associated with short-chain fatty acids and bile acids, we quantified the fecal concentration of these metabolites and found several that correlated with higher levels of HIV immunogen-elicited plasma IgG.DiscussionOur findings highlight an intricate bidirectional relationship between the microbiota and vaccines, where multiple aspects of the vaccination regimen modulate the microbiota and specific microbial features facilitate vaccine responses. An improved understanding of this microbiota–vaccine interplay will help develop more effective vaccines, particularly those that are tailored for early life.

Cotton fabric based textiles utilizing metal oxide nanoparticles (NPs), offer potential options for reducing spreading of infection by bacteria due to their excellent antimicrobial properties, which depend on the nanoparticle's structure and morphology, as well as the fiber functionalization process. In this work, a simple padding and rolling technique for preparing antibacterial cotton fabrics by functionalizing their surface with nanostructured ZnO is demonstrated. L-methionine amino acid is utilized as a binding reagent due to its low toxicity to mammalian cells, biocompatibility with living tissues, and the presence of amine and carboxylic groups, which enhance its coordination capacity to improve ZnO adhesion. Zn(NO3)2·3H2O was precipitated as Zn(OH)2(s) using an alkali mixture of (NaOH+NaBH4), which was then converted into ZnO NPs up on heat treatment on the surface of modified cotton (LCt). Fourier transform infrared spectroscopy (ATR-FTIR) was utilized to confirm the chemical alteration of the fabrics by L-methionine. X-ray diffraction (XRD) investigations confirmed the crystalline structure, purity, and average crystallite size of the ZnO NPs (15.72 nm). X-ray photoelectron spectroscopy (XPS) tests revealed that the NPs were successfully coordinately bonded to the textiles. HR-SEM and energy-dispersive X-ray (EDS) mapping demonstrated that NPs were well-covered on the fabric fibers. The thermal degradation behavior of the modified and functionalized cotton was examined by thermal gravimetric analysis (TGA). The functionalized fabric shown outstanding antibacterial activity against gram-negative bacteria, E. coli and P. aeruginosa, as well as gram-positive bacteria S. aurous and S. pyogenes, with washing durability of up 50 wash cycles. After 50 wash cycles, approximately 88 % of the Zn was retained. This indicated a strong bond between the fabrics. Furthermore, the change has little effect on cotton qualities such as vapor permeability, water absorption, and water transport.

High thoracic epidural anesthesia in cardiac surgery as the sole anesthetic technique is used in a select groups of patients having severe pulmonary co morbidities. We describe a case series of three patients at high risk for post operative pulmonary complications where this technique was used along with dexmedetomidine infusion for conscious sedation in off pump coronary artery bypass grafting. Proper selection of patients, proper placement of the epidural catheter at the appropriate level and timing of catheter insertion and removal are of paramount importance.

Background: Mucositis, xerostomia, dysgeusia, trismus, radiation caries, osteomyelitis, and osteoradionecrosis are a few examples of radiotherapy (RT) complications, eventually distressing the patient's life. Objectives: To study the oral complications emerging due to RT and evaluate dental management of such complications before, during, and post-RT in oral cancer patients. Methods: Patients were evaluated on various parameters, which include the Oral Hygiene Index Simplified (OHI-S), Periodontal Index (PI), Gingival Index (GI), D – Decayed, M – Missing due to caries, F – Filled, T – permanent teeth (DMFT) Index, mucositis, xerostomia, dysgeusia, and interincisal distance before, during (10th session and last session), and 1 month after the last session of RT were evaluated and compared. Introduction: Tonsillectomy is one of the most common surgical procedure in the world performed by Ear, Nose and Throat surgeon. Over the past decade, there has been progress in surgical techniques for tonsillectomy, yet pain and bleeding after tonsillectomy remain important surgical complications. With the increasing need to reduce healthcare costs in developing countries, there is currently a trend towards performing tonsillectomy on a daycare basis and this study aims at evaluating how efficacious it may be in our setting. Methods: A retrospective review of all the daycare tonsillectomy surgeries performed at the Pokhara ENT Center, Pokhara, Nepal from February 2018 to January 2019. Patients aged 15 years and up of either sex who met the paradise criteria were included while patients who met the same but had a contraindication for tonsillectomy surgery were excluded. Demographic information as well as complications within six hours of surgery before discharge and on follow-up on the seventh post-operative day were evaluated, as were any emergency room visits in the first 24 hours and any complications requiring a visit to a healthcare facility. Results: On day of surgery 17% had worst pain and on seventh post-operative day 75% had no pain, only 2% presented with primary hemorrhage and 3% presented with secondary hemorrhage. None of them required revision surgery. Conclusions: Daycare tonsillectomy is being done in only some of the centers owing to the risk of postoperative complications in our country. In our study, the number of complications was lower and not life-threatening. It can be considered a safe and cost-effective method, though patient selection is crucial.

Parthenium hysterophorus is a potential source of a wide range of value-added products. Sulfuric acid-catalyzed pretreatment of P. hysterophorus biomass was used to see how it affected the P. hysterophorus characteristics and the sugar yield from enzymatic hydrolysis. The optimised pretreatment (utilized 2% (w/v) of a substrate, 1.50% (v/v) sulfuric acid, and 45 (min) autoclaving times at 121°C) reduced lignin content by 50.98% (w/w), and increased the available cellulose content (20.72% w/w) over native biomass, which was confirmed by compositional studies of treated biomass. The saccharification rate was increased by 2.08-fold following statistical optimisation using response surface methods (167.17 ± 0.84 mg/g substrate after 12 h saccharification was significantly higher than control 80.16 ± 0.36 mg/g substrate). This justifies that P. hysterophorus is a promising non-conventional lignocellulosic candidate for the production of fermentable sugars.

Neurological ailments, including stroke, Alzheimer’s disease (AD), epilepsy, Parkinson’s disease (PD), and other related diseases, have affected around 1 billion people globally to date. PD stands second among the common neurodegenerative diseases caused as a result of dopaminergic neuron loss in the midbrain’s substantia nigra regions. It affects cognitive and motor activities, resulting in tremors during rest, slow movement, and muscle stiffness. There are various traditional approaches for the management of PD, but they provide only symptomatic relief. Thus, a survey for finding new biomolecules or substances exhibiting the therapeutic potential to patients with PD is the main focus of present-day research. Medicinal plants, herbal formulations, and natural bioactive molecules have been gaining much more attention in recent years as synthetic molecules orchestrate a number of undesired effects. Several in vitro, in vivo, and in silico studies in the recent past have demonstrated the therapeutic potential of medicinal plants, herbal formulations, and plant-based bioactives. Among the plant-based bioactives, polyphenols, terpenes, and alkaloids are of particular interest due to their potent anti-inflammatory, antioxidant, and brain-health-promoting properties. Further, there are no concise, elaborated articles comprising updated mechanism-of-action-based reviews of the published literature on potent, recently investigated (2019–2023) medicinal plants, herbal formulations, and plant based-bioactive molecules, including polyphenols, terpenes, and alkaloids, as a method for the management of PD. Therefore, we designed the current review to provide an illustration of the efficacious role of various medicinal plants, herbal formulations, and bioactives (polyphenols, terpenes, and alkaloids) that can become potential therapeutics against PD with greater specificity, target approachability, bioavailability, and safety to the host. This information can be further utilized in the future to develop several value-added formulations and nutraceutical products to achieve the desired safety and efficacy for the management of PD.

Purpose: To estimate prevalence of common ocular morbidities including color blindness among school-attending children of an urban foothill town of Uttarakhand State in Northern India. Methods: A cross-sectional study was conducted among school-going children of age group 6–16 years of standard I–XII. Schools were selected using population proportionate to the size sampling technique. Detailed ocular examination including color vision and unaided or aided visual acuity for various ocular morbidities was done. Context: Papilloedema is defined as optic disc oedema secondary to raised intracranial pressure (ICP). Grading the degree of oedema using only subjective, examiner-dependent and non-quantitative fundoscopic classification like Modified Friesen Clinical Scale (MFS), may be subject to errors even when used by experienced examiners. The use of spectral-domain optical coherence tomography (SD-OCT), a non-invasive imaging technique, for early identification of raised ICP even before the clinical appearance of papilloedema by quantifying the thickness of the peripapillary retinal nerve fibre layer (ppRNFL) is crucial for such patients. Aims: To evaluate the ppRNFL using SD-OCT in patients with papilloedema. Settings and Design: Hilly North Indian population, Prospective case-control study. Methods and Material: From January 2018 to December 2018, a total of 45 patients diagnosed with papilloedema were included. Age and sex-matched controls were recruited. Detailed demographic information and the history of study subjects were noted. All subjects underwent systemic and detailed ophthalmic examination. Disc photographs were graded as per Modified Frisen Criteria (MFS). SD-OCT of the optic nerve head was done to provide a mean peripapillary RNFL thickness parameters in the different quadrants of the disc in each eye. Statistical Analysis Used: For analysis, statistical software SPSS version 23 and appropriate statistical tests were applied. P value < 0.05 was considered significant. Results: The median age of presentation was 36 years (Range 10–68 years). The male to female ratio was 1:1.3. The mean ppRNFL thickness of both right and left disc in all quadrants was significantly higher in cases in comparison to controls (P-value < 0.05 is significant). The average ppRNFL thickness in all the quadrants of both eyes was significantly higher in the mild grade of papilloedema in comparison to controls (P-value < 0.05 is significant). Conclusions: SD-OCT can be ordered as a routine important non-invasive investigation in addition to basic fundoscopic examination in all patients with papilloedema or suspected to have papilloedema.

Parthenium hysterophorus is a lignocellulosic material with a high potential for fermentable sugar production due to its high availability and adaptability. The current investigation aims at studying the effectiveness of KOH-assisted ultrasonication on the pretreatment of P. hysterophorus biomass. Ultrasonication is a new hydrolysis technology that can provide a higher fermentable sugar yield in less time and at a lower temperature while using less alkali. The combination of KOH and ultrasound has been applied with a detailed study into the effects of various parameters such as sonication time (operating range of 10–50 min), KOH concentration (0.5–2.5%), and temperature (20–50 °C). A higher reducing sugar yield (128.15±0.06 mg/g) was obtained in US-KOH-pretreated biomass as compared with that of the KOH-pretreated biomass. The optimised pretreatment (using a 1:10 bath ratio, 1% (w/v) KOH, and 15-min sonication times) reduced lignin content (delignification) by 58.72% (w/w) and increased the available cellulose content (27.97% w/w) over untreated biomass, which was confirmed by compositional analysis of treated biomass. Further US-KOH-pretreatment was investigated for the liberation of maximum reducing sugar (325.51 mg/g) using the response surface methodology (RSM) approach. Following statistical optimization using response surface methodology, the yield of enzymatic hydrolysis was increased by 3.2-fold compared to the control. According to the investigations, P. hysterophorus can be used as a promising and affordable biomass source for the production of commercial bioethanol.

Background: An anal fissure is a cut or split in the epithelial lining of the anal canal distal to the dentate line. Various topical ointments been described for its treatment. Aim and Objectives: To study Lignocaine (3%) with Zinc (5%) with Hydrocortisone (0.25%) combination (LZH) ointment in comparison to Lignocaine (4%) with Sucralfate (7%) with Metronidazole (1%) combination (LSM) ointment with respect to pain relief, healing, patient satisfaction and safety in acute fissure in ano (AFIA). Material and Methods: This was a prospective randomized study. Sixty patients with AFIA were randomly divided into two groups of 30 patients each. In group A LZH ointment and in group B LSM ointment was prescribed and followed up weekly for six weeks. Results: Our study shows less pain score in group B as compared to group A. The rate of healing was statistically significantly better in group B from 3rd week onwards. The patient satisfaction score was better in group B (P value = 0.0004). No complication was seen in any patient. Conclusion: Our study shows that the LSM ointment in comparison to LZH ointment is associated with early pain relief, better healing, better overall patient satisfaction, and equally safe in patients with AFIA

Abstract Rhamnolipids (RLs) are surface-active compounds and belong to the class of glycolipid biosurfactants, mainly produced from Pseudomonas aeruginosa. Due to their non-toxicity, high biodegradability, low surface tension and minimum inhibitory concentration values, they have gained attention in various sectors like food, healthcare, pharmaceutical and petrochemicals. The ecofriendly biological properties of rhamnolipids make them potent materials to be used in therapeutic applications. RLs are also known to induce apoptosis and thus, able to inhibit proliferation of cancer cells. RLs can also act as immunomodulators to regulate the humoral and cellular immune systems. Regarding their antimicrobial property, they lower the surface hydrophobicity, destruct the cytoplasmic membrane and lower the critical micelle concentration to kill the bacterial cells either alone or in combination with nisin possibly due to their role in modulating outer membrane protein. RLs are also involved in the synthesis of nanoparticles for in vivo drug delivery. In relation to economic benefits, the post-harvest decay of food can be decreased by RLs because they prevent the mycelium growth, spore germination of fungi and inhibit the emergence of biofilm formation on food. The present review focuses on the potential uses of RLs in cosmetic, pharmaceutical, food and health-care industries as the potent therapeutic agents.

A 25-year-old female presented with sudden onset diminution of vision in the right eye (oculus dextrus [OD]) following blunt trauma after a sports injury. Ocular examination revealed best-corrected visual acuity (BCVA) of 20/30 in OD and fundus revealed commotio retinae, localized preretinal bleed, and a large choroidal rupture (CR). She was managed conservatively at that moment. Three months following trauma, she returned with further deterioration of BCVA (20/80) in OD. Optical coherence tomography angiogram (OCTA) confirmed the presence of choroidal neovascular membrane (CNVM). She was treated with single intravitreal ranibizumab injection. Repeat OCTA after 6 weeks showed the regression of CNVM. Her BCVA improved to 20/30 at 6 months of follow-up. In the index report, we present a unique OCTA guided the diagnosis of posttraumatic CR-associated CNVM and its successful management with single intravitreal ranibizumab.

During plant innate immunity ROS is produced via the NADPH oxidase RBOHD. Here, Lee et al. show that RBOHD protein abundance is regulated through phosphorylation by the PBL13 receptor-like cytoplasmic kinase and via ubiquitination by PIRE, a newly identified E3 ubiquitin ligase.

Background: Insulin resistance (IR) is a pre-diabetic condition and has been reported in patients with acanthosis nigricans (AN) and acrochordon. AN and acrochordon are claimed to be cutaneous markers of IR. Aim: The purpose of this paper was to study the association of AN and acrochordon with IR. Methods: It was a cross-sectional hospital-based study. Both groups were assessed for IR by using homeostatic model assessment of insulin resistance (HOMA-IR) formula. Results: A total of 70 cases and an equal number of controls were studied. IR was observed more in cases (41.4%) compared to controls (17.1%) (P < 0.01). Mean HOMA-IR value was also significantly higher in cases (4.32 ± 4.44) compared to controls (2.27 ± 0.90) (P < 0.05). Limitations: Low number of cases and controls were taken in the study. Association with hyperlipidemia and metabolic syndrome was not elicited. Conclusions: AN and acrochordons were found to be associated with IR.

Background: Glycyrrhiza glabra L. (G. glabra) commonly known as liquorice is one of the highly exploited and utilized medicinal plant of the world. Since ancient times liquorice is considered as an auspicious and valuable traditional medicine across the world for treatment of various ailments. Method: Several electronic online scientific databases such as Science Direct, PubMed, Scopus, Scifinder, Google Scholar, online books and reports were assessed for collecting information. All the collected information was classified into different sections to meet the objective of the paper. Results: The electronic database search yielded 3908 articles from different countries. Out of them one ninety-eight articles published between 1956 and 2021 were included, corresponding to all detailed review on G. glabra and research on anti-inflammatories, antivirals and immunomodulatory through pre-clinical and clinical models. From all selective area of studies on G. glabra and its bioactive components it was established (including molecular mechanisms) as a suitable remedy as per the current requirement of pandemic situation arise through respiratory tract infection. Conclusion: Different relevant studies have been thoroughly reviewed to gain an insight on utility of liquorice and its bioactive constituents for anti-inflammatories, antivirals and immunomodulatory effects with special emphasized for prevention and treatment of COVID-19 infection with possible mechanism of action at molecular level. Proposed directions for future research are also outlined to encourage researchers to find out various mechanistic targets and useful value added products of liquorice in future investigations.

Nanostructures of novel materials are capable of altering the properties of materials quite drastically. Recently discovered Dirac monolayer BeN _4 has drawn much attention since this Nitrogen-based material’s electronic structure is very similar to graphene. In this work, we have analyzed various possiblities of nanoribbons structures of MN _4 (M=Be, Mg) monolayer materials. We have proposed several possible geometries of BeN _4 and MgN _4 nanoribbon structures. More specifically, we have proposed armchair nanoribbons with three possible edge configurations as well as zigzag nanoribbons based on edge geometry. Owing to the changes in coordination environment of nanoribbon edge atoms, the resulting bonding characteristics gets altered at the edges, leading to interesting electronic properties. The electronic structures of these nanoribbons are quite different from monolayer and reveal fascinating edge states that exclusively depend on the edge configuration. Though the armchair nanoribbons are found to be metallic, the electronic structure of zigzag nanoribbons exhibits band gap which lead to huge tunability potential of the investigated nanostructures. Therefore, these nanoribbons can function both, metallic as well as semiconducting as required in device applications which can be tuned by modulating the material geometry.

Aceclofenac (ACE), a cyclooxygenase-2 inhibitor, is the derivative of the diclofenac group that has been in use for the symptomatic treatment of systemic inflammatory autoimmune disease, rheumatoid arthritis (RA). Partial solubility, high lipophilic nature, and stability challenge its use in developing topical formulations. Hence, we developed and characterized nanostructured lipid carrier (NLC)–based ACE (ACE-NLC) hydrogel for an efficient transdermal delivery. NLC microemulsion was prepared using different lipids by various methods and was characterized with respect to particle size, zeta potential, surface morphology, and drug encapsulation efficiency. The optimized NLC formulation was incorporated into Carbopol® 940 gel, and this arrangement was characterized and compared with the existing marketed gel (Mkt-gel) formulation to assess in vitro drug release, rheology, texture profile, in vivo skin retention and permeation, and stability. Furthermore, prepared and characterized ACE-loaded NLC formulation was evaluated for skin integrity and fitted in a dermatokinetic model. The results of this study confirmed the spherical shape; smooth morphology and nanometric size attested by Zetasizer and scanning and transmission electron microcopy; and stability of the ACE-NLC formulation. The ACE-NLC-gel formulation showed good rheological and texture characteristics, and better skin distribution in the epidermis and dermis. Moreover, ACE-NLC permeated deeper in the skin layers and kept the skin integrity intact. Overall, NLC-based gel formulation of ACE might be a promising nanoscale lipid carrier for topical application when compared with the conventional Mkt-gel formulation.

Background: Globally, ocular morbidities are one of the most underdiagnosed, underreported, and undertreated public health concern. Among estimated 39 billion blind in world, India shares major proportions, with majority of them living in rural population. In India, religious festivals like Kumbh Mela that involve mass gatherings are mainly attended by middle and elderly population who are usually susceptible to various ocular diseases. Therefore, we analyzed the data of our eye camp to document pattern and magnitude of ocular morbidities among pilgrims attending a mega religious festive event. Methods: In a cross-sectional, descriptive study, we included all those patients who participated in a mega eye camp “Netrakumbh” from January to March 2019 at Kumbh Mela, Prayagraj, India and were attended by voluntary ophthalmologist from All India Institute of Medical Sciences, Rishikesh. Presenting chief complaints and detailed history were collected from all participants. All patients underwent comprehensive eye examination including visual acuity, slit lamp biomicroscopic examination, tonometry, etc., Data were compiled and analyzed utilizing SPSS 22.0. Results: A total of 47142 eyes of 23571 persons of all age groups (range 5 years to 70 years) with approximate equal gender distribution attending the mega eye camp were examined. The most common ocular morbidity noticed after ocular examination was refractive error with underlying cause majorly as presbyopia, followed by cataract, after cataract, convergence insufficiency, lids, and adnexal disease, etc. Conclusions: This study highlights the changing pattern of common ocular morbidities presently among middle and elderly populations in developing countries like India.

Abstract Background Hemophagocytic lymphohistiocytosis (HLH) and macrophage activation syndrome (MAS) were historically thought to be distinct entities, often managed in isolation. In fact, these conditions are closely related. A collaborative approach, which incorporates expertise from subspecialties that previously treated HLH/MAS independently, is needed. We leveraged quality improvement (QI) techniques in the form of an Evidence-Based Guideline (EBG) to build consensus across disciplines on the diagnosis and treatment of HLH/MAS. Methods A multidisciplinary work group was convened that met monthly to develop the HLH/MAS EBG. Literature review and expert opinion were used to develop a management strategy for HLH/MAS. The EBG was implemented, and quality metrics were selected to monitor outcomes. Results An HLH/MAS clinical team was formed with representatives from subspecialties involved in the care of patients with HLH/MAS. Broad entry criteria for the HLH/MAS EBG were established and included fever and ferritin ≥500 ng/mL. The rheumatology team was identified as the “gate-keeper,” charged with overseeing the diagnostic evaluation recommended in the EBG. First-line medications were recommended based on the acuity of illness and risk of concurrent infection. Quality metrics to be tracked prospectively based on time to initiation of treatment and clinical response were selected. Conclusion HLH/MAS are increasingly considered to be a spectrum of related conditions, and joint management across subspecialties could improve patient outcomes. Our experience in creating a multidisciplinary approach to HLH/MAS management can serve as a model for care at other institutions.

Background: Shankhpushpi is an Ayurvedic drug, widely used for its actions on the central nervous system, especially to improve intellect and boost memory. Four botanicals viz. Canscora decussata Schult. (CD), Clitorea ternatea Linn. (CT), Convolvulus pluricaulis Choisy. (CP) and Evolvulus alsinoides Linn. (EA) are considered as sources of Shankhpushpi by Indian practitioners on the basis of their morphological descriptions given in ancient texts. Objective: The present study was undertaken to evaluate the neuropharmacological effect of four herbs commonly identified as source of Shankhpushpi. Materials and methods: Methanol extracts of all four varieties were tested and evaluated in vitro and in vivo for their neuropharmacological effects. Experiments such as protection against β-amyloid induced neurotoxicity on brain cell line (Neuro 2A), antioxidant potential, AchE (acetylcholinesterase enzyme) inhibition, and 5-LOX (lipoxygenase) enzyme inhibition were conducted for in vitro evaluation. For in vivo evaluation, scopolamine (0.3 mg/kg i.p.) induced memory retrieval using pole climbing apparatus and Morris water maze were performed in rat models. Results: It was found that protective effects of EA and CD against β-amyloid induced neurotoxicity in Neuro 2A cells were significantly higher than CT and CP. EA proved to be superior than other varieties on the basis of antioxidant activity, AchE inhibitory and LOX inhibitory activities. The preventive activity of EA on scopolamine induced memory retrieval in pole climbing and Morris water maze task in rats was found to be higher than that of CD, CT and CP. Conclusion: EA has remarkable neuropharmacological effect as compared to other three varieties of Shankhpushpi. This effect may be attributed due to the presence of steroids (stigmasterol and betulinic acid), coumarins (scopoletin) and flavonoids (β-carotene and chlorogenic acid). Hence it can be used as a promising lead in development and management of neuronal disorders including Alzheimer's disease. The problems of atmospheric pollutants are causing significant concern across the globe and in India. The aggravated level of atmospheric pollutants in the surrounding environment poses serious threats to normal living conditions by deteriorating air quality and causing adverse health impacts. Pollutant concentration increases during harvesting seasons of Kharif/Rabi due to stubble burning and is aggravated by other points or mobile sources. The present study is intended to monitor the spatio-temporal variation of the major atmospheric pollutants using Sentinel-5P TROPOMI data through cloud computing. Land Use/Land Cover (LULC-categorization or classification of human activities and natural coverage on the landscape) was utilised to extract the agricultural area in the study site. It involves the cloud computing of MOD64A1 (MODIS Burned monthly gridded data) and Sentinel-5P TROPOMI (S5P Tropomi) data for major atmospheric pollutants, such as CH4, NO2, SOX, CO, aerosol, and HCHO. The burned area output provided information regarding the stubble burning period, which has seen post-harvesting agricultural residue burning after Kharif crop harvesting (i.e., rice from April to June) and Rabi crop harvesting (i.e., wheat from September to November). The long duration of stubble burning is due to variation in farmers’ harvesting and burning stubble/biomass remains in the field for successive crops. This period was used as criteria for considering the cloud computing of the Sentinel-5P TROPOMI data for atmospheric pollutants concentration in the study site. The results showed a significant increase in CH4, SO2, SOX, CO, and aerosol concentration during the AMJ months (stubble burning of Rabi crops) and OND months (stubble burning of Kharif crops) of each year. The results are validated with the ground control station data for PM2.5/PM10. and patterns of precipitation and temperature-gridded datasets. The trajectory frequency for air mass movement using the HYSPLIT model showed that the highest frequency and concentration were observed during OND months, followed by the AMJ months of each year (2018, 2019, 2020, and 2021). This study supports the role and robustness of Earth observation Sentinel-5P TROPOMI to monitor and evaluate air quality and pollutants distribution.

Peroxiredoxins (Prxs) are antioxidant proteins that are involved in cellular defence against reactive oxygen species and reactive nitrogen species. Humans have six peroxiredoxins, hPrxI-VI, out of which hPrxI and hPrxII belongs to the typical 2-Cys class sharing 90% conservation in their amino acid sequence including catalytic residues required to carry out their peroxidase and chaperone activities. Despite the high conservation between hPrxI and hPrxII, hPrxI behaves differently from hPrxII in its peroxidase and chaperone activity. We recently showed in yeast that in the absence of Tsa1 and Tsa2 (orthologs of hPrx) hPrxI protects the cells against different stressors whereas hPrxII does not. To understand this difference, we expressed catalytic mutants of hPrxI in yeast cells lacking the orthologs of hPrxI/II. We found that the catalytic mutants lacking peroxidase function including hPrxIC52S, hPrxIC173S, hPrxIT49A, hPrxIP45A and hPrxIR128A were not able to grow on media with nitrosative stressor (sodium nitroprusside) and unable to withstand heat stress, but surprisingly they were able to grow on an oxidative stressor (H2O2). Interestingly, we found that hPrxI increases the expression of antioxidant genes, GPX1 and SOD1, and this is also seen in the case of a catalytic mutant, indicating hPrxI can indirectly reduce oxidative stress independently of its own peroxidase function and thus suggesting a novel role of hPrxI in altering the expression of other antioxidant genes. Furthermore, hPrxIC83T was resistant to hyperoxidation and formation of stable high molecular weight oligomers, which is suggestive of impaired chaperone activity. Our results suggest that the catalytic residues of hPrxI are essential to counter the nitrosative stress whereas Cys83 in hPrxI plays a critical role in hyperoxidation of hPrxI.

To match the current life-style, there is a huge demand and market for the processed food whose manufacturing requires multiple steps. The mounting demand increases the pressure on the producers and the regulatory bodies to provide sensitive, facile, and cost-effective methods to safeguard consumers’ health. In the multistep process of food processing, there are several chances that the food-spoiling microbes or contaminants could enter the supply chain. In this contest, there is a dire necessity to comprehend, implement, and monitor the levels of contaminants by utilizing various available methods, such as single-cell droplet microfluidic system, DNA biosensor, nanobiosensor, smartphone-based biosensor, aptasensor, and DNA microarray-based methods. The current review focuses on the advancements in these methods for the detection of food-borne contaminants and pathogens.

ABSTRACT The recognition over the past decade that nearly all diseases are associated with changes in the microbiome has raised hope that microbiome-based therapeutics may cure many human ailments. Billions of dollars are being poured into microbiome-oriented biotech companies, and the coming years will undoubtedly witness the approval of the first generation of these products. However, significant hurdles remain in expanding the pipeline and advancing these first-generation therapies. In this perspective, I will discuss the challenges related to identifying causal microbes, determining their mechanism of action, and characterizing the specific bacterial molecules required for disease protection. We are approaching these issues through a combination of clinical sampling, animal models, classic microbiology methodologies, and systems-based approaches. The field of microbiome research is on the cusp of being able to identify clinically actionable host-microbe relationships; increasing attention on identifying causal microbes and their bioactive factors will usher in the next generation of microbiome-based therapies.

In recent years, erythropoietin (EPO) has emerged as a useful neuroprotective and neurotrophic molecule that produces antidepressant and cognitive-enhancing effects in psychiatric disorders. However, EPO robustly induces erythropoiesis and elevates red blood cell counts. Chronic administration is therefore likely to increase blood viscosity and produce adverse effects in non-anemic populations. Carbamoylated erythropoietin (CEPO), a chemically engineered modification of EPO, is non-erythropoietic but retains the neurotrophic and neurotrophic activity of EPO. Blood profile analysis after EPO and CEPO administration showed that CEPO has no effect on red blood cell or platelet counts. We conducted an unbiased, quantitative, mass spectrometry-based proteomics study to comparatively investigate EPO and CEPO-induced protein profiles in neuronal phenotype PC12 cells. Bioinformatics enrichment analysis of the protein expression profiles revealed the upregulation of protein functions related to memory formation such as synaptic plasticity, long term potentiation (LTP), neurotransmitter transport, synaptic vesicle priming, and dendritic spine development. The regulated proteins, with roles in LTP and synaptic plasticity, include calcium/calmodulin-dependent protein kinase type 1 (Camk1), Synaptosomal-Associated Protein, 25 kDa (SNAP-25), Sectretogranin-1 (Chgb), Cortactin (Cttn), Elongation initiation factor 3a (Eif3a) and 60S acidic ribosomal protein P2 (Rplp2). We examined the expression of a subset of regulated proteins, Cortactin, Grb2 and Pleiotrophin, by immunofluorescence analysis in the rat brain. Grb2 was increased in the dentate gyrus by EPO and CEPO. Cortactin was induced by CEPO in the molecular layer, and pleiotrophin was increased in the vasculature by EPO. The results of our study shed light on potential mechanisms whereby EPO and CEPO produce cognitive-enhancing effects in clinical and preclinical studies.

The tumor suppressor p53 plays a critical role in suppressing cancer growth and progression and is an attractive target for the development of new targeted therapies. We synthesized several indolo‐pyrido‐isoquinolin based alkaloids to activate p53 function and examined their therapeutic efficacy using NCI‐60 screening. Here, we provide molecular evidence that one of these compounds, 11‐methoxy‐2,3,4,13‐tetrahydro‐1H‐indolo[2′,3′:3,4]pyrido[1,2‐b]isoquinolin‐6‐ylium‐bromide (termed P18 or NSC‐768219) inhibits growth and clonogenic potential of cancer cells. P18 treatment results in downregulation of mesenchymal markers and concurrent upregulation of epithelial markers as well as inhibition of migration and invasion. Experimental epithelial–mesenchymal‐transition (EMT) induced by exposure to TGFβ/TNFα is also completely reversed by P18. Importantly, P18 also inhibits mammosphere‐formation along with a reduction in the expression of stemness factors, Oct4, Nanog and Sox2. We show that P18 induces expression, phosphorylation and accumulation of p53 in cancer cells. P18‐mediated induction of p53 leads to increased nuclear localization and elevated expression of p53 target genes. Using isogenic cancer cells differing only in p53 status, we show that p53 plays an important role in P18‐mediated alteration of mesenchymal and epithelial genes, inhibition of migration and invasion of cancer cells. Furthermore, P18 increases miR‐34a expression in p53‐dependent manner and miR‐34a is integral for P18‐mediated inhibition of growth, invasion and mammosphere‐formation. miR‐34a mimics potentiate P18 efficacy while miR‐34a antagomirs antagonize P18. Collectively, these data provide evidence that P18 may represent a promising therapeutic strategy for the inhibition of growth and progression of breast cancer and p53‐miR‐34a axis is important for P18 function.

Shone's anomaly is a very rare congenital cardiac malformation characterized by four serial obstructive lesions of the left side of the heart (i) Supravalvular mitral membrane (ii) parachute mitral valve (iii) muscular or membranous subaortic stenosis and (iv) coarctation of aorta. We report a unique presentation of Shone's complex in a 14-year-old adolescent male. In addition to the four characteristic lesions the patient had bicuspid aortic valve, aneurysm of sinus of valsalva, patent ductus arteriosus, ventricular septal defect, persistent left superior vena cava opening into coronary sinus and severe pulmonary artery hypertension. This case report highlights the importance of a strong clinical suspicion of the coexistence of multiple congenital cardiac anomalies in Shone's complex and the significance of a careful comprehensive echocardiography.

Radial neuron migration in the developing cerebral cortex is a complex journey, starting in the germinal zones and ending in the cortical plate. In mice, migratory distances can reach several hundreds of microns, or millimeters in humans. Along the migratory path, radially migrating neurons slither through cellularly dense and complex territories before they reach their final destination in the cortical plate. This task is facilitated by radial glia, the neural stem cells of the developing cortex. Indeed, radial glia have a unique bipolar morphology, enabling them to serve as guides for neuronal migration. The key guiding structure of radial glia is the basal process, which traverses the entire thickness of the developing cortex. Neurons recognize the basal process as their guide and maintain physical interactions with this structure until the end of migration. Thus, the radial glia basal process plays a key role during radial migration. In this review, we highlight the pathways enabling neuron-basal process interactions during migration, as well as the known mechanisms regulating the morphology of the radial glia basal process. Throughout, we describe how dysregulation of these interactions and of basal process morphology can have profound effects on cortical development, and therefore lead to neurodevelopmental diseases.

A wideband non-resonant absorber is proposed, and its radar cross section (RCS) reduction is investigated. A discussion on the functional materials available is followed by the design of an absorber on a Plexiglas substrate with polyaniline-graphene nanocomposite as layered square inclusions with thicknesses and conductivities scaled to golden ratio. The measured dielectric properties of polyaniline-graphene nanocomposites are used in the fullwave simulation. The design parameters have been identified and optimized using CST Microwave Studio. As designed structure is fabricated and the reflection is measured. The objective of the work is to demonstrate the use of non-metallic conducting polymer composites devoid of metals for radar absorbing material (RAM) structural designs. The structure is an all-polymer and electrically thin design with a potential to be 3D printed to suit the target object.
