1. Making Mobile Nanotechnology Accessible: Is the Explicit Preparation of Janus Nanoparticle Necessary to Achieve Mobility?

Abstract: This study compares the mobility behaviour, in a H2O2 environment, of three different geometries of hybrid particle made of silica core functionalized by gold (nanoparticles or layer). It is known that the decomposition of H2O2 on gold surfaces drives mobility; however, the link between mobility orientation and the organization of gold on silica surfaces is still questionable. While conventional wisdom posits that asymmetric designs are crucial for generating phoretic forces or localized bubble propulsion, recent research suggests that symmetrical particles may also exhibit motility. To address this debate, we developed a robust workflow for synthesizing gold grafted silica nanoparticles with precise control over size and shape, enabling the direct comparison of their motile behaviour by dynamic light scattering and particle tracking velocimetry. Our results indicate, first, that a combination of techniques is necessary to overcome their intrinsic limitation and, second, that the inherent asymmetry generated by isotropic gold nanoparticle deposition onto silica surfaces may enable particle motility.

Abstract: The study described in this paper was conducted in the framework of the European nPSize project (EMPIR program) with the main objective of proposing new reference certified nanomaterials for the market in order to improve the reliability and traceability of nanoparticle size measurements. For this purpose, bimodal populations as well as complexly shaped nanoparticles (bipyramids, cubes, and rods) were synthesized. An inter-laboratory comparison was organized for comparing the size measurements of the selected nanoparticle samples performed with electron microscopy (TEM, SEM, and TSEM), scanning probe microscopy (AFM), or small-angle X-ray scattering (SAXS). The results demonstrate good consistency of the measured size by the different techniques in cases where special care was taken for sample preparation, instrument calibration, and the clear definition of the measurand. For each characterization method, the calibration process is described and a semi-quantitative table grouping the main error sources is proposed for estimating the uncertainties associated with the measurements. Regarding microscopy-based techniques applied to complexly shaped nanoparticles, data dispersion can be observed when the size measurements are affected by the orientation of the nanoparticles on the substrate. For the most complex materials, hybrid approaches combining several complementary techniques were tested, with the outcome being that the reliability of the size results was improved.

Abstract: International audience; Hybrid aluminosilicate nanotubes (Imo-CH3) have the ability to trap small organic molecules inside their hydrophobic internal cavity while being dispersed in water owing to their hydrophilic external surface. They also display a curvature-induced polarization of their wall, which favors reduction outside the nanotubes and oxidation inside. Here, we coupled bare plasmonic gold nanoparticles (GNPs) with Imo-CH3 and analyzed for the first time the redox reactivity of these hybrid nano-reactors upon UV illumination. We show that the coupling between GNPs and Imo-CH3 significantly enhances the nanotube photocatalytic activity, with a large part of water reduction occurring directly on the gold surface. The coupling mechanism strongly influences the initial H2 production rate, which can go from ×10 to more than ×90 as compared to bare Imo-CH3 depending on the synthesis route of the GNPs. The present results show that this hybrid photocatalytic nano-reactor benefits from a synergy of polarization and confinement effects that facilitate efficient H2 production.

Abstract: Swollen cubic lyotropic ternary phases with Pn3m symmetry and reduced hardness were obtained from a specific binary mixture of cubic phase-forming (phytantriol) and lamellar phase-forming (decaglycerol monooleate) compounds. The microstructures were determined by using a small-angle x-ray scattering technique. The softness and temperature-induced phase transitions were investigated by means of rheology. The incorporation of a surface-active fragrance compound (linalool) at concentrations up to 6 wt. % induced a structural transition toward a softer Im3m bulk cubic phase with longer water channels. Higher linalool concentrations allowed for the spontaneous dispersion of the bulk cubic phase into microscopic particles with a cubic structure (cubosomes).

Abstract: International audience; Crystallization from solution often occurs via “nonclassical” routes; that is, it involves transient, non-crystalline states like reactant-rich liquid droplets and amorphous particles. However, in mineral crystals, the well-defined thermodynamic character of liquid droplets and whether they convert─or not─into amorphous phases have remained unassessed. Here, by combining cryo-transmission electron microscopy and X-ray scattering down to a 250 ms reaction time, we unveil that crystallization of cerium oxalate involves a metastable chemical equilibrium between transient liquid droplets and solid amorphous particles: contrary to the usual expectation, reactant-rich droplets do not evolve into amorphous solids. Instead, at concentrations above 2.5 to 10 mmol L–1, both amorphous and reactant-rich liquid phases coexist for several tens of seconds and their molar fractions remain constant and follow the lever rule in a multicomponent phase diagram. Such a metastable chemical equilibrium between solid and liquid precursors has been so far overlooked in multistep nucleation theories and highlights the interest of rationalizing phase transformations using multicomponent phase diagrams not only when designing and recycling rare earths materials but also more generally when describing nonclassical crystallization.

Abstract: International audience; Intense research on nanocrystals synthesized in solution is motivated by their original physical properties, which are determined by their sizes and shapes on various scales. However, morphology control on the nanoscale is limited by our understanding of crystallization, which is challenged by the now well-established prevalence of noncrystalline intermediates. In particular, the impact of such intermediates on the final sizes and crystal quality remains unclear because the characterization of their evolution on the nanometer and millisecond scales with nonperturbative analyses has remained a challenge. Here we use in situ X-ray scattering to show that the nucleation and growth of YVO4:Eu nanocrystals is spatially restrained within amorphous, nanometer-scaled intermediates. Abstract: International audience; The influence of the initial seed structure on final nanoparticle geometry has been investigated by an original “competitive approach” using small-angle X-ray scattering, UV–vis spectroscopy, and transmission electronmicroscopy analysis. Herein, by using the seed-mediated gold nanoparticle growth, seeds with different sizes and crystalline structures were synthesized and injected into the same growth media. The seeds were chosen because of their different growth evolution into two different morphologies. Cetyltrimethylammonium bromide-coated single-crystalline seeds grow into rodlike shapes, whereas citrate-coated multicrystalline seeds mainly grow into wheat-shape polycrystalline particles with small elongation called nanobeans in coexistence with a large quantity of spheres. When seeds are added into the same growth media for competition, a mixture of different morphologies is obtained. By controlling the number of added competing seeds in the solution, it was found that the multicrystalline seeds have a higher growth rate than the single-crystalline seeds. This has direct impact on the final distribution of the size and morphology of the nanoparticles. The consequence is a large tendency toward nanobean and sphere structure formation even when a small number of multitwinned seeds is added in the solution. This method demonstrates the importance of the nature of defects hidden in the initial seed and proves that these defaults are inherited to the final nanocrystal throughout the growth stage from the beginning of the growth. This competitive seeded growth approach is an easy way to identify the influence of seed morphologies in the synthetic pathway of nanoparticle formation.

Abstract: International audience; The market for nano-additive materials has been growing exponentially since 2012, with almost 5040 consumer products containing nanoparticles in 2021. In parallel, the increasing recommendations, definitions and legislations underline the need for traceability of manufactured nanoparticles and for methods able to identify and quantify the “nano” dimensional character in manufactured product. From a multi-technic approach, this paper aims to compare the mesurands extracted from SAX/BET (specific surface area) and SEM (diameter equivalent to a projected surface area) on different TiO$_2$ powder issued from referenced, synthesized materials, raw materials (additives) and extracted materials from manufactured products. The influence of various parameters such as the anisotropic factor, the interaction between particles, the size distribution and the extraction steps are discussed to illustrate their impact on the diameter values issued from two different measurands. These results illustrate the difficulties in (nano)particles characterization. SEM and SAXS are complementary technics depending on the level of dimensional characterization required.

Abstract: International audience; The ability to control the morphology of precipitated cerium oxalate material results in determinate evidence to its final properties. In this study, we demonstrate that surfactant-free nanostructured low-water solvents have a huge potential for controlling the morphology of a cerium oxalate powder. To this aim, an in-depth investigation of the reaction between cerium nitrate and oxalic acid is carried out, by varying both the relative concentration of the two reagents (around the stoichiometric value) and the composition of the water/propanediol/octanol ternary solvent (especially in the low-water content nanostructured domain). Thanks to the complementary of observation methods: microscopy (confocal microscopy with fluorophores and environmental SEM) and X-ray scattering (SAXS and WAXS), we evidenced the role of the solvent in the growth kinetics and directional aggregation of the precipitates—the two major factors determining the final morphology of the particles. Besides the possible confinement effect in nanodroplets, compact “dense-branching” particles, achieved in low-water content solvents, unveil the strong role of the surface forces in the aggregation mechanisms. This is consistent with the prevailing capillary forces at water/oil/solid triple points in ternary solvents. These new results confirm the high potential of nanostructured solvents for controlling the size and shape of hydrated precipitated particles.

Abstract: International audience; In recent years, microfluidic-based sample preparation techniques have emerged as a powerful tool for measurements at large scale X-ray facilities. Most often the microfluidic device was a form of hybrid system, i.e. an assembly of different materials, because a simple, versatile and inexpensive microfabrication method, on the one hand, and X-ray compatibility, on the other hand, cannot generally be achieved by the same material. The arrival of a new polymer family based on Off-Stoichiometric Thiol-Ene-epoxy (OSTE+) has recently redistributed the cards. In this context, we studied the relevance and the compatibility of OSTE+ for small-angle X-ray scattering (SAXS) studies. The material was characterized regarding its X-ray properties (transmission coefficient, attenuation coefficient, scattering pattern and polymer aging under X-ray light) and their comparison with those of the usual polymers used in microfluidics and/or for synchrotron radiation experiments. Abstract: International audience; Nanomedicines are considered as promising therapeutics for cancer treatment. However, clinical translation is still scarce, partly because their biological behavior is not well understood. Extracting general guidelines from the great variety of nanoparticles and conditions studied is indeed difficult, and relevant techniques are lacking to obtain in situ information. Here, both issues are solved by combining versatile model nanoparticles with in situ tools based on small-angle scattering techniques (SAS). The strategy was to develop a library of nanoparticles and perform systematic study of their interactions with biological systems. Considering the promising properties of gold nanoparticles as cancer therapeutics, polymethacrylate-grafted gold nanoparticles were chosen as models. Modulation of polymer chemistry was shown to change the surface properties while keeping the same structure for all nanoparticles. This unity allowed reliable comparison to extract general principles, while the synthesis versatility enabled to fine-tune the nanoparticles surface properties, especially through copolymerization, and thus to optimize their biological behavior. Two specific aspects were particularly examined: colloidal stability and cell uptake. Positive charges and hydrophobicity were identified as key parameters influencing toxicity and internalization. In situ SAS gave valuable information about nanoparticles evolution in biologically relevant environments. Good colloidal stability was thereby shown in cell culture media, while intracellular transformation and quantity of nanoparticles were monitored, highlighting the potential of these techniques for nanomedicines studies.

Abstract: International audience; In the field of nanomedicine, nanostructured nanoparticles (NPs) made of self-assembling prodrugs emerged in the recent years with promising properties. In particular, squalene-based drug nanoparticles have already shown their efficiency through in vivo experiments. However, a complete pattern of their stability and interactions in the blood stream is still lacking. In this work we assess the behavior of squalene-adenosine (SQAd) nanoparticles-whose neuroprotective effect has already been demonstrated in murine models-in the presence of fetal bovine serum (FBS) and of bovine serum albumin (BSA), the main protein of blood plasma. Extensive physicochemical characterizations were performed using Small Angle Neutron Scattering (SANS), cryogenic transmission electron microscopy (Cryo-TEM), circular dichroism (CD), steady-state fluorescence spectroscopy (SSFS) and isothermal titration calorimetry (ITC) and in silico by means of ensemble docking simulations with human serum albumin (HSA). Significant changes in the colloidal stability of the nanoparticles in the presence of serum albumin were observed. SANS, CD and SSFS analyses demonstrated an interaction 2 between SQAd and BSA, with a partial disassembly of the nanoparticles in the presence of BSA and the formation of a complex between SQAd and BSA. The interaction free energy of SQAd nanoparticles with BSA derived from ITC experiments, is about-8 kcal/mol which is further supported in silico by ensemble docking simulations. Overall, our results show that serum albumin partially disassembles SQAd nanoparticles by extracting individual SQAd monomers from them. As a consequence, the SQAd nanoparticles would act as a circulating reservoir in the blood stream. The approach developed in this study could be extended to other soft organic nanoparticles.

Abstract: Diffusion of nanomedicines inside the extracellular matrix (ECM) has been identified as a key factor to achieve homogeneous distribution and therefore therapeutic efficacy. Here, we sought to determine the impact of nanoparticles' (NPs) surface properties on their ability to diffuse in the ECM. As model nano-objects, we used a library of gold nanoparticles grafted with a versatile polymethacrylate corona, which enabled the surface properties to be modified. To accurately recreate the features of the native ECM, diffusion studies were carried out in a tumor-derived gel (Matrigel). We developed two methods to evaluate the diffusion ability of NPs inside this model gel: an easy-to-implement one based on optical monitoring and another one using small-angle X-ray scattering (SAXS) measurements. Both enabled the determination of the diffusion coefficients of NPs and comparison of the influence of their various surface properties, while the SAXS technique also allowed to monitor the NPs' structure as they diffused inside the gel. Positive charges and hydrophobicity were found to particularly hinder diffusion, and the different results suggested on the whole the presence of NPs-matrix interactions, therefore underlying the importance of the ECM model. The accuracy of the tumor-derived gels used in this study was evidenced by in vivo experiments involving intratumoral injections of NPs on mice, which showed that diffusion patterns in the peripheral tumor tissues were quite similar to the ones obtained within the chosen ECM model.

Abstract: International audience; A large variety of nanoparticle-based delivery systems have become increasingly important for diagnostic and/or therapeutic applications. Yet, the numerous physical and chemical parameters that influence both the biological and colloidal properties of nanoparticles remain poorly understood. This complicates the ability to reliably produce and deliver well-defined nanocarriers which often leads to inconsistencies, conflicts in the published literature and, ultimately, poor translation to the clinics. A critical issue lies in the challenge of scaling-up nanomaterial synthesis and formulation from the lab to industrial scale while maintaining control over their diverse properties. Studying these phenomena early on in the development of a therapeutic agent often requires partnerships between the public and private sectors which are hard to establish. In this study, through the particular case of squalene-adenosine nanoparticles, we reported on the challenges encountered in the process of scaling-up nanomedicines synthesis. Here, squalene (the carrier) was functiona-lized and conjugated to adenosine (the active drug moiety) at an industrial scale in order to obtain large quantities of biocompatible and biodegradable nanoparticles. After assessing nanoparticle batch-to-batch consistency , we demonstrated that the presence of squalene analogs resulting from industrial scale-up may influence several features such as size, surface charge, protein adsorption, cytotoxicity and crystal structure. These analogs were isolated, characterized by multiple stage mass spectrometry, and their influence on nanoparticle properties further evaluated. We showed that slight variations in the chemical profile of the nanocarrier's constitutive material can have a tremendous impact on the reproducibility of nanoparticle properties. In a context where several generics of approved nanoformulated drugs are set to enter the market in the coming years, characterizing and solving these issues is an important step in the pharmaceutical development of nanomedicines.

Abstract: International audience; The phosphonic acid moiety is commonly used as an anchoring group for the surface modification of imogolite. However, the impact of the reaction on its structure has never been clearly analyzed before. We study the reaction of imogolite and decylphosphonic acid by combining infrared spectroscopy, X-ray scattering, scanning electron microscopy, transmission electron microscopy and solid-state nuclear magnetic resonance spectroscopy. Instead of a surface functionalization, we observe the formation of a lamellar phase interconnected with imogolite bundles. Although we find no evidence for grafted imogolite tubes, we observe the expected dispersion characteristics and stabilization of water in toluene emulsions described in literature. Based on the surface chemistry of imogolite we propose an explanation for the observed reactivity and link the structural features of the obtained composite material to its dispersibility in toluene and its observed properties at the toluene-water interface.

Abstract: International audience; Squalene-adenosine (SQAd) nanoparticles (NPs) were found to display promising pharmacological activity similar to many other nanomedicines, but their long-term stability was still limited, and their preparation required specific know-how and material. These drawbacks represented important restrictions for their potential use in the clinic. Freeze-drying nanoparticles is commonly presented as a solution to allow colloidal stability, but this process needs to be adapted to each nanoformulation. Hence, we aimed at developing a specific protocol for freeze-drying SQAd NPs while preserving their structural features. NPs were lyophilised, resuspended and analysed by dynamic light scattering, atomic force microscopy and small-angle scattering. Among four different cryoprotectants, trehalose was found to be the most efficient in preserving NPs physico-chemical characteristics. Interestingly, we identified residual ethanol in NP suspensions as a key parameter which could severely affect the freeze-drying outcome, leading to NPs aggregation. Long-term stability was also assessed. No significant change in size distribution or zeta potential could be detected after three-month storage at 4?°C. Finally, freeze-dried NPs innocuity was checked in vitro on cultured hepatocytes and in vivo on mice. In conclusion, optimisation of freeze-drying conditions resulted in safe lyophilised SQAd NPs that can be easily stored, shipped and simply reconstituted into an injectable form.

Abstract: In the context of cancer treatment, gold nanoparticles (AuNPs) are considered as very promising radiosensitizers. Here, well-defined polymer-grafted AuNPs were synthesized and studied under gamma irradiation to better understand the involved radiosensitizing mechanisms. First, various water-soluble and well-defined thiol-functionalized homopolymers and copolymers were obtained through atom transfer radical polymerization. They were then used as ligands in the one-step synthesis of AuNPs, which resulted in stable hybrid metal-polymer nanoparticles. Second, these nano-objects were irradiated in solution by γ rays at different doses. Structures were fully characterized through size exclusion chromatography, small-angle X-ray scattering, and small-angle neutron scattering measurements, prior to and after irradiation. We were thus able to quantify and to localize radiation impacts onto the grafted polymers, revealing the production sites of reactive species around AuNPs. Both external and near-surface scissions were observed. Interestingly, the ratio between these two effects was found to vary according to the nature of polymer ligands. Medium-range and long-distance dose enhancements could not be identified from the calculated scission yields, but several mechanisms were considered to explain high yields found for near-surface scissions. Then cytotoxicity was shown to be equivalent for both nonirradiated and irradiated polymer-grafted NPs, which suggested that released polymer fragments were nontoxic. Finally, the potential to add bioactive molecules such as anticancer drugs has been explored by grafting doxorubicin onto the polymer corona. This may lead to nano-objects combining both radiosensitization and chemotherapy effects. This work is the first one to study in details the impact of radiation on radiosensitizing nano-objects combining physical, chemical, and biological analyses.

Abstract: Measurement of nanoparticle size (distribution) becomes a challenging analytical problem when non-spherical nanoparticles must be accurately measured. Most industrial nanoparticles have not only non-spherical shapes but also possess polydisperse size distributions, and due to their agglomeration/aggregation state are difficult (or even impossible) to be addressed individually. Moreover, driven by regulatory purposes related to the identification of a material as a nanomaterial, the accurate measurement of the smallest dimension of a (nano)particulate material makes the analysis even more complex. In the first phase of the EU Project nPSize - Improved traceability chain of nanoparticle size measurements (https://www.bam.de/Content/DE/Projekte/laufend/nPSize/npsize.html), the efforts are focused on synthesis of nanoparticles of well-defined, non-spherical shape. Following candidates of reference materials (CRM) with certifiable particle size (distribution) are under characterization with respect to their homogeneity and stability: (i) titania nanoplatelets (10-15 nm thickness x 50-60 nm lateral), (ii) titania bipyramides (~60 nm length x 40 nm width), (iii) titania acicular particles (100 nm length x 15-20 nm width; aspect ratio 5.5/6), (iv) gold nanorods (~10 nm width x 30 nm length), and (v) gold nanocubes (~55 nm x 55 nm x 55 nm).

Abstract: Shape and size are known to determine a nanoparticle's properties. Hardly ever studied in synthesis, the internal crystal structure (i.e., particle defects, crystallinity, and symmetry) is just as critical as shape and size since it directly impacts catalytic efficiency, plasmon resonance, and orients anisotropic growth of metallic nanoparticles. Hence, its control cannot be ignored any longer in today's research and applications in nanotechnology. This study implemented an unprecedented reliable measurement combining these three structural aspects. The unified small-angle X-ray scattering and diffraction measurement (SAXS/XRD) was coupled with molecular dynamics to allow simultaneous determination of nanoparticles' shape, size, and crystallinity at the atomic scale. Symmetry distribution (icosahedra-Ih, decahedra-Dh, and truncated octahedra-TOh) of 2-6 nm colloidal gold nanoparticles synthesized in organic solvents was quantified. Nanoparticle number density showed the predominance of Ih, followed by Dh, and little, if any, TOh. This result contradicts some theoretical predictions and highlights the strong effect of the synthesis environment on structure stability. We foresee that this unified SAXS/XRD analysis, yielding both statistical and quantitative counts of nanoparticles' symmetry distribution, will provide new insights into nanoparticle formation, growth, and assembly.

Abstract: International audience; The nanostructure of a microemulsion can be strongly affected by the liquid-to-solid transition during polymerization. Here, we examined the evolution of nanostructures of different ternary mixtures, including two microemulsions and a single lamellar phase that upon polymerization are quantitatively studied by SAXS/WAXS and DSC experiments systematically performed before and after the polymerization of both aqueous and organic phases. Samples are mixtures of the poly(2-acrylamido-2-methylpropanesulfonic acid) network as the aqueous phase and poly(hexyl methacrylate) as the organic phase stabilized by Brij35 surfactant. Upon polymerization, the surfactant is excluded from the water/oil interface and crystallizes, strongly changing the nanostructure of samples where it is the main component. In samples where the aqueous phase is the main component, only a few changes in structure are observed upon polymerization. This study demonstrates quantitatively the possibility to preserve nanostructures during polymerization, thus inducing a templating effect.

Abstract: International audience; a We show that ternary mixtures of water (polar phase), myristic acid (MA, apolar phase) and cetyltrimethyl-ammonium bromide (CTAB, cationic surfactant) studied above the melting point of myristic acid allow the preparation of microemulsions without adding a salt or a co-surfactant. The combination of SANS, SAXS/ WAXS, DSC, and phase diagram determination allows a complete characterization of the structures and interactions between components in the molten fatty acid based microemulsions. For the different structures characterized (microemulsion, lamellar or hexagonal phases), a similar thermal behaviour is observed for all ternary MA/CTAB/water monophasic samples and for binary MA/CTAB mixtures without water: crystalline myristic acid melts at 52 1C, and a thermal transition at 70 1C is assigned to the breaking of hydrogen bounds inside the mixed myristic acid/CTAB complex (being the surfactant film in the ternary system). Water determines the film curvature, hence the structures observed at high temperature, but does not influence the thermal behaviour of the ternary system. Myristic acid is partitioned in two ''species'' that behave independently: pure myristic acid and myristic acid associated with CTAB to form an equimolar complex that plays the role of the surfactant film. We therefore show that myristic acid plays the role of a solvent (oil) and a co-surfactant allowing the fine tuning of the structure of oil and water mixtures. This solvosurfactant behaviour of long chain fatty acid opens the way for new formulations with a complex structure without the addition of any extra compound.

In the present work, the DEHCNPB coordination structure toward uranium and its aggregation properties have been fully characterized using experimental and theoretical approaches. DEHCNPB forms associated species, and its aggregation properties such as aggregation number or CAC have been determined from SAXS and SANS measurements. The preferred site for bonding of DEHCNPB toward U(VI) has been determined from IR, EXAFS, and DFT calculations.

Abstract: International audience; Among the proposed mechanisms to predict and understand synergism in solvent extraction, the possibility of a preorganization of the mixture of extractant molecules has never been considered. Whether involving synergistic aggregation as for solubilization enhancement with reverse micelles or favored molecular interaction between the extractant molecules, evaluation of this hypothesis requires characterization of the aggregates formed by the extractant molecules at different scales. We investigate here the HDEHP/ TOPO couple of extractant with methods ranging from vibrational spectroscopy and ESI-MS spectrometry to vapor pressure osmometry and neutron and X-ray scattering to cover both molecular and supramolecular scales. These experimental methods are subjected to DFT calculations and molecular dynamics calculations, allowing a rationalization of the results through the different scales. Performed in the absence of any cation, this original study allows a decorrelation of the mechanisms at the origin of synergy: it appears that no clear preorganization of the extractants can explain the synergy and therefore that the synergistic aggregation observed in the presence of cations is rather due to the chelation mechanisms than to intrinsic properties of the extractant molecules.

Abstract: International audience; Squalene based nanoparticles obtained via nanoprecipitation are promising candidates as efficient anti-cancer drugs. In order to highlight their preparation process and to facilitate further clinical translation, the present study enlightens the paramount role of the solvent in the formation of these nanomedicines. Three different squalene-based nanoparticles, i.e. squalenic acid, deoxycytidine squalene and gemcitabine squalene, have been investigated before and after organic solvent evaporation. Size and structural analysis by Small Angle Neutron Scattering revealed that droplets' size was uniquely controlled by the solvent composition (ethanol–water), which evolved during their gradual formation. The particles were preferably swollen by water and the swelling increased when less ethanol was present. Either coalescence or fragmentation was observed depending on the increase or decrease of the ethanol content, supporting an equilibrium control of the size. Moreover, a high water swelling was observed for the three local organization of the nanodroplets (hexagonal for gemcitabine squalene, cubic for deoxycytidine and not structured for squalenic acid) and could be the source of the previously reported efficiency of related anti-cancer squalene based nanomedicines.

Abstract: Since the 2000s, nanomaterials (NMs) have become part of our daily routine and are increasingly present on the worldwide market. These NMs are used in various sectors of activity such as construction, food, health, automotive and many others. Normative definitions and regulations have been established to control the NMs traceability throughout their life cycle. In Europe, regulatory requirements (labelling obligation if nano ingredients are used as additives for food, cosmetics or biocides) impose to the additives users to know if the substances should be considered as "nano" or not. A reliable metrology is therefore necessary for the nano-objects identification and characterization. However, this metrological characterization remains a challenge due to the numerous parameters to be considered in order to fully describe these nano-objects (size, size distribution, agglomeration/aggregation state, shape, specific surface area, chemical composition, surface chemistry and surface charge), which are listed in the ISO/TC 229 (PDTR 13014) standard. The thesis general context is the NMs characterization and more specifically dimensional characterization at the nanometric scale. The study focus on the Titanium dioxide (TiO2) (nano) particles which are ranked among the 5 most produced nanoparticulate substances (based on tonnage) in the world and used in many applications.The objective is to evaluate and compare measurands of several dimensional characterization techniques for nanoparticles and thus determine their constituent particle size. In this way, ensemble and integral techniques such as SAXS (small angle X-ray scattering), DLS (dynamic light scattering) and BET (Brunauer-Emmett-Teller) have been compared to the direct measurement technique, SEM (scanning electron microscopy).Emphasis is placed on the influence evaluation of several factors, namely, the size, the shape as well as the particles crystallinity and the complex matrix (set of elements in coexistence with the particles of interest), on the realized measurements. The selection of a large materials variety (reference, synthetic, raw material or finished products) enables the NMs characterization difficulties identification according to their sources.After a literature review to place the thesis in its context, a first part is devoted to the elaboration of a sample preparation protocol for SEM in order to improve the image processing step to comply with the regulations and to determine isolated particles size and/or constituents particles from agglomerates. Following the size of particles/agglomerates by DLS, the sonication step impact on the dispersion and dissociation of agglomerated (nano) objects present in suspension was highlighted.In a second part, a multi-technique approach is implemented to compare two measurands from different instruments (an equivalent diameter to a projected surface area for SEM and a specific surface area for SAXS and BET), in order to access the dimensions of the studied (nano)-objects. The influence of different parameters such as the size distribution (or polydispersity), the purity of the samples (and thus the NMs extraction step from their matrix), the interaction between the particles and the particles anisotropy, are discussed. Interaction between particles and shape anisotropy are the most influential factors on specific surface area measurements, or SAXS/BET equivalent diameters. The achieved results illustrate the (nano)-objects characterization difficulties but they confirm the complementarity of the techniques used in this study (SEM, SAXS and BET)., Depuis les années 2000, les nanomatériaux (NMs) font partie de notre vie quotidienne et sont de plus en plus présents sur le marché mondial. Ces NMs sont utilisés dans divers secteurs d’activité tels que le bâtiment, l’alimentation, la santé, l’automobile et bien d’autres. Des définitions normatives et des règlementations ont été mises en place pour encadrer la traçabilité des NMs le long de leur cycle de vie. Ainsi en Europe, des exigences réglementaires (obligation d’étiquetage si ingrédient nano dans produits alimentaires, cosmétiques ou biocides) imposent aujourd’hui aux utilisateurs d’additifs de savoir si la substance qu’ils utilisent doit être considérée comme « nano » ou pas. Une métrologie fiable permettant l’identification et la caractérisation des nano-objets est donc nécessaire. Cependant, cette caractérisation métrologique des NMs reste un défi compte-tenu du nombre de paramètres à prendre en compte pour les décrire complètement (taille, distribution en taille, état d’agglomération/agrégation, forme, surface spécifique, composition chimique, chimie de surface et charge en surface), listés dans la norme ISO/TC 229 (PDTR 13014). Le sujet de thèse se place dans ce cadre de caractérisation des NMs et plus particulièrement de la caractérisation dimensionnelle à l’échelle nanométrique. L’étude concerne essentiellement les (nano) objets de dioxyde de titane (TiO2) qui sont classées parmi les 5 substances sous forme de nanoparticules les plus produites (en tonnage) dans le monde et utilisées dans de nombreuses applications.L’objectif est d’évaluer et de comparer les mesurandes de plusieurs techniques de caractérisation dimensionnelle de nanoparticules pour accéder à la taille des particules constitutives. Ainsi, des techniques d’ensemble et intégrales comme le SAXS (diffusion des rayons-X aux petits angles), la DLS (diffusion dynamique de la lumière) et la BET (Brunauer–Emmett–Teller) ont été comparées à la technique de mesure directe par MEB (microscopie électronique à balayage). L'accent est mis sur l'évaluation de l'influence de plusieurs facteurs, à savoir, la taille, la forme ainsi que la cristallinité des particules et la matrice complexe (ensemble d’éléments en coexistence avec les particules d’intérêt), sur les mesures réalisées. Le choix d’une large famille de composés (de références, de synthèse, matière première ou produits finis) permet d’identifier les difficultés de caractérisation des NMs en fonction de leur provenance.Après une revue de la littérature sur l’état de l’art pour positionner le sujet dans son contexte une première partie est consacrée à l’élaboration d’un protocole de préparation d’échantillon pour le MEB. L’objectif est de faciliter l’étape de traitement d’images dans le but de répondre à la réglementation et caractériser les particules isolées et/ou constitutives des agglomérats. Par le contrôle de la taille des agglomérats par DLS, l’impact de l’étape de sonication sur la dispersion et la dissociation des (nano) objets agglomérés présents en suspension a été mis en évidence.Dans une deuxième partie, une approche multi-technique est mise en œuvre pour comparer deux mesurandes issues de différents instruments (un diamètre équivalent à une surface projetée pour le MEB et une surface spécifique pour le SAXS et la BET), afin d’accéder aux dimensions des (nano)-objets étudiées. L'influence de différents paramètres tels que la distribution en taille (ou polydispersité) la pureté des échantillons (et donc l’étape d’extraction des NMs de leur matrice), l'interaction entre les particules et l’anisotropie des particules sont discutées. Ces dernières ont un impact important sur les mesures de surface spécifique, ou des diamètres équivalents issus du SAXS. Les résultats obtenus illustrent les difficultés de la caractérisation des (nano)-objets mais confirment que les techniques utilisées dans cette étude (le MEB, le SAXS et la BET) sont complémentaires.

Abstract: In the industrial production of pigments, catalysts, plant protection agents, nuclear fuel and pharmaceuticals precipitation and crystallization plays a fundamental role. Although these processes are often applied and a relative control over the formed solids can be achieved, the processes are not always well understood on a microscopic level. To identify how the solids are formed and which mechanisms govern their formation potentially gives the capabilities to better control such processes.In this thesis two different approaches are explored to study precipitation and crystallization by confining reactions into droplets. The first approach focuses on the combination of a droplet microfluidic device and in-situ small angle X-ray scattering. Off-stochiomestry thiol-ene-epoxy polymer is characterized for the use with in-situ X-ray scattering and a protocol is presented to prepare suitable microfluidic devices from this material. An original approach to isolate the scattering signal of the carrier phase and the droplets is then used to study the precipitation of cerium oxalate in droplets. The second approach aims to use imogolite nanotubes to stabilize droplets against coalescence and to study their transport properties to control reactant feeding into droplets. By fully characterizing the necessary surface modification by alkylphosphonic acids for the first time, evidence is found that the reaction does not yield surface modified tubes. Consequentially, new approaches are explored to obtain individually dispersed imogolite nanotubes with a hydrophobic surface.; Dans de nombreux secteurs industriels tels que la production de pigments, de catalyseurs, de produits pharmaceutiques, de minerais ou le recyclage de combustibles nucléaire, les étapes de précipitation et de cristallisation sont cruciales. Bien que ces procédés soient utilisés à grandes échelles avec un contrôle relatif des solides formés, les mécanismes de formation ne sont pas toujours bien compris, en particulier à l’échelle microscopique. Une meilleure compréhension des différentes étapes de formation permettrait d’obtenir des leviers de contrôle adapté à chaque composé et application.Cette thèse se place dans ce cadre très général du contrôle de la précipitation et plus particulièrement de la précipitation en gouttes d’emulsion eau dans l’huile. Deux approches différentes ont été explorées pour moduler et suivre la réactivité dans une goutte d’emulsion. La première approche était basée sur le couplage d’une puce microfluidique générateur de gouttes avec la diffusion de rayons X aux petits angles (SAXS) in situ. Après une caractérisation complète de la compatibilité du polymère non stoechiometrique thiol-ene-epoxy (OSTE+) avec le SAXS, une puce en OSTE+ compatible avec le suivi in situ SAXS a été développée. Un traitement original permettant d’extraire séparément le signal des gouttes du signal de l’huile a permis de valider le montage dans le cas de la précipitation d’oxalate de cerium. La deuxième approche avait pour but l’utilisation de nanotubes d’imogolites pour stabiliser des gouttes d’émulsion et étudier le transport de réactifs entre gouttes via ces nanotubes. Cette approche nécessitait de fonctionnaliser les nanotubes pour les rendre hydrophobe. La caractérisation poussée de la fonctionnalisation des imogolites par des alkylphosphoniques a montré pour la première fois que cette réaction ne conduisait pas à une fonctionnalisation de surface mais à un matériau composite aux propriétés interfaciales remarquables. De nouvelles voies de fonctionnalisation de surface ont été développées pour rendre les imogolites hydrophobes.

By fully characterizing the necessary surface modification by alkylphosphonic acids for the first time, evidence is found that the reaction does not yield surface modified tubes. Consequentially, new approaches are explored to obtain individually dispersed imogolite nanotubes with a hydrophobic surface., Dans de nombreux secteurs industriels tels que la production de pigments, de catalyseurs, de produits pharmaceutiques, de minerais ou le recyclage de combustibles nucléaire, les étapes de précipitation et de cristallisation sont cruciales. Bien que ces procédés soient utilisés à grandes échelles avec un contrôle relatif des solides formés, les mécanismes de formation ne sont pas toujours bien compris, en particulier à l’échelle microscopique. Une meilleure compréhension des différentes étapes de formation permettrait d’obtenir des leviers de contrôle adapté à chaque composé et application.Cette thèse se place dans ce cadre très général du contrôle de la précipitation et plus particulièrement de la précipitation en gouttes d’emulsion eau dans l’huile. Deux approches différentes ont été explorées pour moduler et suivre la réactivité dans une goutte d’emulsion. La première approche était basée sur le couplage d’une puce microfluidique générateur de gouttes avec la diffusion de rayons X aux petits angles (SAXS) in situ. Après une caractérisation complète de la compatibilité du polymère non stoechiometrique thiol-ene-epoxy (OSTE+) avec le SAXS, une puce en OSTE+ compatible avec le suivi in situ SAXS a été développée. Un traitement original permettant d’extraire séparément le signal des gouttes du signal de l’huile a permis de valider le montage dans le cas de la précipitation d’oxalate de cerium. La deuxième approche avait pour but l’utilisation de nanotubes d’imogolites pour stabiliser des gouttes d’émulsion et étudier le transport de réactifs entre gouttes via ces nanotubes. Cette approche nécessitait de fonctionnaliser les nanotubes pour les rendre hydrophobe. La caractérisation poussée de la fonctionnalisation des imogolites par des alkylphosphoniques a montré pour la première fois que cette réaction ne conduisait pas à une fonctionnalisation de surface mais à un matériau composite aux propriétés interfaciales remarquables. De nouvelles voies de fonctionnalisation de surface ont été développées pour rendre les imogolites hydrophobes.

Abstract: Nanomaterials are more and more used in various situations, such as catalysis,color, optics, etc. To optimize their use, it is necessary to better understand reactions andinteractions taking place at these scales. This PhD thesis aims at exploring the links betweensynthesis conditions, particle morphology and their properties. In particular, we chose a tostudy a model-material: cerium (IV) oxide. CeO2 nanoparticles have been obtained bymicrowave-heating assisted solvothermal synthesis. Morphologies obtained and studied havebeen: cubes, octahedrons and rods. We tried to to give an explanation on how these synthesesallowed the formation of such morphologies. Gold has then be deposed on the surface of theseCeO2 nanoparticles, and studied by a combination of Transmission Electron Microscopystudies, and computer modelizations., Les nanomatériaux sont de plus en plus utilisés dans de nombreusesapplications, que cela soit pour la catalyse, la coloration, l’optique, etc. Pour optimiser leurutilisation, il est nécessaire de mieux comprendre les réactions et interactions ayant lieu à ceséchelles. Cette thèse se propose d’essayer d’explorer les liens entre conditions de synthèse,morphologie de particules, et propriétés de celles-ci. En particulier, nous avons opté pourl’étude d’un matériau modèle : l’oxyde de cérium (IV). Les nanoparticules de CeO2 ont étéobtenues par voie de synthèse solvothermale assistée par chauffage micro-ondes. Lesmorphologies des nanoparticules de CeO2 étudiées ont été : les cubes, les octaèdres et lesbâtonnets, et nous avons essayé de fournir une explication à leur obtention par la voie desynthèse utilisée. De l’or a ensuite été déposé en surface des ces nanoparticules de CeO2 etétudié par une combinaison d’études par Microscopie Électronique en Transmission et demodélisations numériques.
