Στην εργασία αυτή αναπτύχθηκε μια γρήγορη και ταχεία μέθοδος για τον προσδιορισμό κουμαρίνης σε αρτοσκευάσματα, χρησιμοποιώντας την τεχνική προκατεργασίας Capsule Phase Microextraction (CPME). Η ανάλυση πραγματοποιήθηκε με Υγρή Χρωματογραφία Υψηλής Πίεσης με ανιχνευτή παράταξης φωτοδιόδων. Η CPME είναι μια «πράσινη» τεχνική προκατεργασίας δείγματος, όπου χρησιμοποιείται μια κάψουλα μικροεκχύλισης (MEC) από ίνα κυτταρίνης επικαλυμμένη με ένα sol-gel υβριδικό οργανικό-ανόργανο προσροφητικό. Τα προσροφητικά υλικά sol-gel χαρακτηρίζονται από μοναδική εκλεκτικότητα, υψηλή ευαισθησία και χημική σταθερότητα. Είναι πορώδη, παρέχουν υψηλό ρυθμό εκχύλισης από το υπόστρωμα του δείγματος στο προσροφητικό και προωθούν την ταχεία διάχυση του διαλύματος του δείγματος για ταχύτερη εξισορρόπηση της εκχύλισης και την ταχεία διάχυση του οργανικού διαλύτη για ταχεία έκλουση. Σε αυτήν την εργασία, ένα υλικό CPME επικαλυμμένο με sol-gel C18, χρησιμοποιήθηκε για την εκχύλιση της κουμαρίνης από αρτοσκευάσματα. Η ανάλυση πραγματοποιήθηκε με την τεχνική της Υγρής Χρωματογραφίας Υψηλής Πίεσης σε περίπου 13 min, με χρωματογραφική στήλη LiChrospher® 100 RP-8e (5μm) LiChroCART® 250-4 και κινητή φάση ακετονιτρίλιο-οξικό αμμώνιο 0,05Μ, σε αναλογία όγκων 30-70%, v/v (ισοκρατική έκλουση). Η ανίχνευση και ο προσδιορισμός της κουμαρίνης επιτεύχθηκε με ανιχνευτή παράταξης φωτοδιόδων στα 275 nm. Η εκχύλιση και έκλουση της κουμαρίνης πραγματοποιήθηκε σε 20 λεπτά και 15 λεπτά, αντίστοιχα, με τη μεθανόλη να χρησιμοποιείται ως διαλύτης έκλουσης. Η μέθοδος επικυρώθηκε ως προς τη γραμμικότητα, την εκλεκτικότητα, την ακρίβεια και την πιστότητα. Τα όρια ανίχνευσης και ποσοτικής αποτίμησης LOD και LOQ του οργάνου ήταν 0,03 μg/mL και 0,08 μg/mL, αντίστοιχα. Τα όρια ανίχνευσης και ποσοτικής αποτίμησης LOD και LOQ της μεθόδου ήταν 0,18 μg/g και 0,48 μg/g, αντίστοιχα. Τέλος, η μέθοδος εφαρμόστηκε επιτυχώς για τον ποσοτικό προσδιορισμό της κουμαρίνης σε δείγματα αρτοσκευασμάτων (τσουρέκι, μπισκότο, πανετόνε)., In the current study, a rapid and simple method was developed for the determination of coumarin in fine bakery products, using Capsule Phase Microextraction (CPME), prior to the determination by High Pressure Liquid Chromatography with a Photodiode-Array detector. CPME is a green sample preparation technique, which utilizes a Microextraction Capsule (MEC) that consists of a sol-gel hybrid inorganic-organic sorbent coated fiber (usually made of cotton). The sol-gel sorbents are characterized by unique selectivity, high extraction sensitivity and chemical stability. The sorbent material of MEC is porous, thus providing a high rate of extraction from the sample matrix to the sorbent and propelling rapid diffusion of the sample solution for faster extraction equilibrium and organic solvent for rapid elution. Herein, a CPME material coated with sol-gel C18 was used to extract coumarin from bakery products. Analysis was carried out by High Pressure Liquid Chromatography using a LiChrospher® 100 RP-8e (5μm) LiChroCART® 250-4 chromatographic column and mobile phase of acetonitrile-ammonium acetate 0,05M 30-70%, v/v (isocratic elution) in about 13 min. Detection and determination of coumarin was achieved by a Photodiode-Array detector at 275 nm. The method was validated in terms of sensitivity, linearity, selectivity, accuracy and precision. The limit of detection (LOD) and the limit of quantification (LOQ) of the instrument were 0,03 μg/mL and 0,08 μg/mL, respectively. The LOD and LOQ of the developed method were equal to 0,18 μg/g and 0,48 μg/g, respectively. Finally, the developed method has been successfully applied for the quantitative determination of coumarin in various bakery products (sweet brioche (“tsoureki”), biscuit, panettone).

Η δοξορουβικίνη είναι μία κυτταροτοξική ανθρακυκλίνη με αντικαρκινική δράση που αναστέλλει το ένζυμο τοποϊσομεράση ΙΙ. Χρησιμοποιείται είτε μόνη της, είτε σε συνδυασμό με άλλους αντικαρκινικούς παράγοντες για την θεραπεία πλήθους διαφορετικών ειδών καρκίνων. Η δοξορουβικίνη αδρανοποιείται κυρίως μέσω του μεταβολισμού της σε δύο κύριους μεταβολίτες, τη δοξορουβικινόλη και τη δοξορουβικινόνη. Μεταξύ των κυριότερων ανεπιθύμητων ενεργειών της δοξορουβικίνης είναι η συχνά εμφανιζόμενη καρδιοτοξικότητα. Στην αναλυτική χημεία, τα βιολογικά υποστρώματα αποτελούν ένα από τα πιο απαιτητικά είδη υποστρώματος για την απομόνωση και ποσοτικοποίηση ουσιών. Αυτό είναι κατανοητό αν σκεφτεί κανείς το πλήθος των ενδογενών ουσιών που μπορεί να υπάρχουν σ΄ αυτά. Μεταξύ αυτών είναι και το πλάσμα όπου η παρουσία ενώσεων όπως οι πρωτεΐνες μπορεί να καταστίσουν δυσχερή τη βιοανάλυση. Έτσι ο αναλυτής αναγκάζεται να καταφύγει σε δραστικές μεθόδους προκατεργασίας οι οποίες αφενός μπορεί να επηρεάσουν αρνητικά το δείγμα, αφετέρου να θέσουν σε κίνδυνο την ασφάλεια του ίδιου του αναλυτή ή να βλάψουν το περιβάλλον εξαιτίας της χρήσης επικίνδυνων χημικών. Στην τρέχουσα μελέτη, μία «όλα σε ένα» συσκευή μικροεκχύλισης χρησιμοποιήθηκε για την εκχύλιση της δοξορουβικίνης και των δύο μεταβολιτών της από πλάσμα αρουραίου, πριν από τον προσδιορισμό τους με HPLC συζευγμένη με έναν ανιχνευτή φθορισμού. Το προσροφητικό που παρασκευάστηκε με την τεχνολογία sol-gel, παρασκευάσθηκε in situ και ενσωματώθηκε εντός δύο συνδεδεμένων μεταξύ τους καψουλών από πολυπροπυλένιο μαζί με μία κυλινδρική μαγνητική ράβδο ώστε να μην απαιτείται η χρήση εξωτερικής ανάδευσης. Ανάμεσα στα εξεταζόμενα προσροφητικά, το μετρίως πολικό πολυ(τετραϋδροφουράνιο) βρέθηκε να είναι το περισσότερο ευνοϊκό εξαιτίας της αποτελεσματικότητας του στην εκχύλιση των προσδιοριζόμενων ουσιών. Κατά την ανάπτυξη της τεχνικής έγινε μία συστημική διερεύνηση των κρίσιμων παραμέτρων που επηρέαζαν τα βήματα προσρόφησης και έκλουσης. Εξαιτίας του από της φύσεως του, μηχανισμού φιλτραρίσματος των πορωδών καψουλών, η απομόνωση των ουσιών έγινε απευθείας στο υπόστρωμα πλάσματος χωρίς να απαιτείται προηγούμενη κατεργασία του δείγματος (π.χ. καταβύθιση πρωτεϊνών, φυγοκέντρηση κ.ο.κ.). Η προτεινόμενη μέθοδος επικυρώθηκε ως προς τους παράγοντες γραμμικότητα, ακρίβεια, επαναληψιμότητα, εκλεκτικότητα, ευαισθησία και σταθερότητα σύμφωνα με τις οδηγίες του FDA. Τα όρια ανίχνευσης (LOD) βρίσκονταν στο εύρος 1-2 ng/ml, ενώ τα όρια ποσοτικοποίησης ορίστηκαν στα 10 ng/mL. Η ακρίβεια (εκφρασμένη ως % σχετικό σφάλμα) βρέθηκε μεταξύ -9,7-15,3%, εντός της ίδιας ημέρας και μεταξύ διαφορετικών ημερών. Η επαναληψιμότητα ήταν μεγαλύτερη του 13,4%. Τέλος, η δυνατότητα εφαρμογής της τεχνικής αποδείχθηκε με την ανάλυση των ουσιών σε πλάσμα αρουραίου, μετά από ενδοφλέβια χορήγηση δοξορουβικίνης ώστε να μελετηθεί το φαρμακοκινητικό προφίλ της, Doxorubicin belongs to the cytotoxic class of anthracyclines, with antitumor activity. It acts as an inhibitor of the topoisomerase II enzyme. Currently, it is used as a stand-alone therapy or in combination with other antitumor agents for the treatment of a variety of cancers. Doxorubicin is mainly eliminated by its metabolism into two main metabolites, doxorubicinol and doxorubicinone. Amongst its adverse effects, the most observed and critical one is cardiotoxicity. In analytical chemistry, biological matrices comprise one of the most challenging types of matrices for the isolation and quantification of target analytes. One of those biological matrices is blood plasma where large endogenous macromolecules such as proteins might intervene against the extraction process. Under those circumstances, the scientist is forced to adopt more drastic sample preparation processes that might influence the sample negatively or put him or the environment in more danger due to the use of hazardous chemicals. In this study, an “all-in-one” microextraction device was used for the extraction of doxorubicin and its two metabolites from rat plasma prior to their determination by HPLC coupled to fluorescence detector. A sol-gel-based sorbent has been in-situ synthesized and incorporated within two conjoined porous polypropylene capsules together with a cylindrical magnetic bar in order to avoid the need of an external stirring bar. Among other sorbents investigated, the moderately polarity monolithic poly(tetrahydrofuran) was found to be advantageous due to its efficiency towards the extraction of the analytes. Systematic investigation of the critical parameters affecting the adsorption and the desorption step was carried out. Due to the “built-in” filtration mechanism of the porous capsules, the isolation of the target analytes was performed directly in the plasma matrix without any previous sample pretreatment (i.e. protein precipitation, centrifugation etc). The proposed method was validated in terms of linearity, accuracy, precision, specificity, sensitivity, and stability according to the FDA guidelines. The limits of detection (LODs) ranged between 1 – 2 ng mL-1 while the LLOQs were set to 10 ng mL-1. The accuracy (expressed as % relative error) was found within -9.7 – 15.3% under both intra- and inter-day conditions. The precision was better than 13.4 % in all cases. Finally, the applicability of the fabricated stand-alone extraction device was demonstrated in the analysis of the target analytes in rat plasma after intravenous administration of doxorubicin in order to assess its pharmacokinetic profile

Herein, we report on the development of a waste minimization/valorization methodology applied to the representative benchmark Sonogashira cross-coupling reaction performed in a continuous-flow reactor, featuring a continuous-flow downstream membrane organic/aqueous separator to recover medium and products with minimal waste. The protocol is based on the use of a biomass waste-derived heterogeneous Pd-based catalyst, which is obtained from the valorization of urban-waste pine needles (PiNe). In a circular economy approach, the PiNe biomass has been proven to be capable of producing an effective active carbon support for Pd nanoparticle immobilization. In addition, the catalyst has been utilized in an azeotropic mixture formed by industrial waste-derived cyclopentyl methyl ether (CPME) and water. Thanks to this combination and with the adoption of flow conditions, high yields of final target products could be accessed with high stability and durability of the catalyst. Final isolation of the products has been realized by setting an in-line liquid-liquid separator in flow, which has also allowed CPME recovery with a significant reduction of the waste generated. The protocol has been applied to the representative preparation of eniluracil, a GSK API.

The synthesis of both enantiomers of a key intermediate in the synthesis of halofuginone was accomplished by a Candida antarctica lipase B (CAL-B)-catalyzed kinetic resolution of the corresponding racemate. When the resolution was carried out in the versatile solvent cyclopentyl methyl ether (CPME) using p-chlorophenylbutyrate (PCPB) as the acylating reagent, the highest enantiomeric ratio (E) values were measured, and highly enantioenriched (95% ee) compounds could be obtained in a single iteration. As an example, one of the two enantiomers was used as a starting material to prepare (+)-halofuginone in a three-step procedure.

The quest for sustainable solvents is currently a matter of intense research and development, as solvents significantly contribute heavily to the waste generated by chemical industries. Cyclopentyl methyl ether (CPME) is a promising eco-friendly solvent with valuable properties such as low peroxide formation rate, stability under basic and acidic conditions, and relatively high boiling point. This Review discusses the potential use of CPME for applications in biotechnology (e.g., biotransformations, as solvent or cosolvent), biorefineries, and bioeconomy (e.g., for furan synthesis or as an extractive agent in liquid-liquid separations), as well as for other purposes, such as chromatography or peptide synthesis. Although CPME is currently produced by petrochemical means with a remarkably high atom economy, its biogenic production can be envisaged from substrates such as cyclopentanol or cyclopentanone, which can be derived from furfural or from (bio-based) adipic acid, respectively. The combination of the promising properties of CPME as a (co)solvent with a future (economic) biogenic origin would be advantageous for setting strategies aligned with the sustainable chemistry principles.

Although attempts to make the Y(II) complex, (CpMe3Y)1− (CpMe = C5H4Me), by reduction of CpMe3Y with potassium were unsuccessful and the products of potassium reduction of CpMe3Ln for Ln = La and Pr led to ring-opening reduction of THF, we report that crystallographically-characterizable Ln(II) complexes of Tb and Ho can be isolated by reducing CpMe3Ln(THF) with KC8 in THF in the presence of 18-crown-6 (18-c-6). X-ray crystallography revealed that these complexes are isolated with a methylcyclopentadienide inverse sandwich countercation: [(18-c-6)K(μ-CpMe)K(18-c-6)][CpMe3Ln] (Ln = Tb, Ho). Although reduction of CpMe3Ln with potassium in the presence of the 2.2.2-cryptand (crypt) chelate has not generally provided fully-characterizable, crystalline products, in the case of Dy, crystals of [K(crypt)][CpMe3Dy] could be isolated.

The &ldquo, osmotic shock&rdquo, phenomenon is the main thing that is responsible for morphological structure alteration, which can jeopardize the use of a polymer in a chemical process. This is extremely important in solid-phase peptide synthesis (SPPS), which is the method of choice for the preparation of these important biologically active compounds. Herein, we have used Hildebrand solubility parameters (&delta, ) to investigate the influence of different ethers that are used in the precipitation step of the SPPS using a polystyrene resin. The green cyclopentyl methyl ether (CPME) has shown to be slightly superior to 2-methyltetrahydrofurane, which is also a green ether and clearly better than the hazardous diethyl ether and tert-butyl methyl ether. These results have been corroborated by scanning electron microscope (SEM) analysis and computational studies. All together, these confirm the adequacy of CPME for being the ether of choice to be used in SPPS.
