The importance of renewable and environmentally acceptable materials in the construction industry is increasing. Wood is one of the most important construction materials and its use in building applications has further expanded in recent decades, mainly due to the development of modern composites, firstly glue lam, OSB (Oriented Strand Board), LVL (Laminated Veneer Lumber) and a considerable breakthrough was later achieved with the development of cross-laminated timber. In order to enable even more extensive and reliable use of wood in outdoor applications, factors affecting a wood’s service life need to be understood. It is well known that fungal degradation of wood is predominantly affected by temperature and wood moisture content. In order to elucidate the influence of these two factors, long-term monitoring of temperature, relative humidity and moisture content at the newly-built Annex of the Department of Wood Science and Technology was carried out. The Annex is designed from CLT composites and glue-lam beams and was finished in December 2015. The results clearly show that microclimatic conditions in the monitored building in the first years of use are far below the limit required for fungal decay or mould growth.

Analysis of wood transects in a manner that preserves the spatial distribution of the metabolites present is highly desirable to among other things: (1) facilitate ecophysiology studies that reveal the association between chemical make-up and environmental factors or climatic events over time; and (2) investigate the mechanisms of the synthesis and trafficking of small molecules within specialised tissues. While a variety of techniques could be applied to achieve these goals, most remain challenging and impractical. Laser ablation direct analysis in real time imaging-mass spectrometry (LADI-MS) was successfully used to survey the chemical profile of wood, while also preserving the small-molecule spatial distributions. The tree species Entandrophragma candollei Harms, Millettia laurentii DeWild., Pericopsis elata (Harms) Meeuwen, Dalbergia nigra (Vell.) Benth. and Dalbergia normandii BosserR.Rabev were analysed. Several compounds were associated with anatomical features. A greater diversity was detected in the vessels and parenchyma compared with the fibres. Analysis of single vessels revealed that the chemical fingerprint used for timber identification is mainly determined by vessel content. Laser ablation direct analysis in real time imaging-mass spectrometry offers unprecedented opportunities to investigate the distribution of metabolites within wood samples, while circumventing the issues associated with previous methods. This technique opens up new vistas for the discovery of small-molecule biomarkers that are linked to environmental events.

For decades, theory has upheld that plants do not fractionate water isotopes as they move across the soil-root interface or along plant stems. This theory is now being challenged by several recent studies reporting that the water held in woody stems has an isotopic composition that cannot be attributed to any potential water source. Isotopic offsets between stem and source water still need to be explained, as they prevent identifying unambiguously tree water’s origin from water isotope measurements. Here we show that isotopic offsets between stem and source water can be explained by micrometer-scale water isotope heterogeneity within woody stems and soil micropores. Using a novel technique to extract sap water in xylem conduits separately from the water held in other xylem tissues, we show that these non-conductive xylem tissues are more depleted in deuterium than sap water. We also report that, in cut stems and well-watered potted plants, the isotopic composition of sap water reflects well that of irrigation water, demonstrating that no isotopic fractionation occurs during root water uptake or the sap water extraction process. Previous studies showed that isotopic heterogeneity also exists in soils at the pore scale where water adsorbed onto soil particles is more depleted than capillary/mobile soil water. Data collected at a beech (Fagus sylvatica) forest indicate that sap water matches best the capillary/mobile soil water from deep soil horizons, indicating that micrometer-scale water isotope heterogeneity in soils and stems must be accounted for to unambiguously identify where trees obtain their water within catchments.Significance StatementForests are prime regulators of the water cycle over land. They return, via transpiration, a large fraction of precipitation back to the atmosphere, influence surface runoff, groundwater recharge or stream flow, and enhance the recycling of atmospheric moisture inland from the ocean. The isotopic composition of water in woody stems can provide unique information on the role forests play in the water cycle only if it can be unambiguously related to the isotopic composition of source water. Here, we report a previously overlooked isotopic fractionation of stem water whereby non-conductive tissues are more depleted in deuterium than sap water, and propose a new technique to extract sap water separately from bulk stem water to unambiguously identify plant water sources.

White-rot fungi are thought to hold promise for development of a delignification pretreatment process for wood biorefinery that is less energy-consuming than current processes. However, the reaction must take place over weeks and consumes non-neglectable amounts of saccharides. To establish a biological process for wood biorefinery would first require establishment of an enzymatic approach to delignification. Such an approach has the potential to lower costs and reduce saccharide loss. Here, we attempted enzymatic delignification reactions using manganese peroxidases (MnP), a lignin-degrading enzyme, under several reaction conditions. The delignification rate from beech wood meal (particle size45 μm) of up to 11.0% in 48 h was reached in a MnP reaction supplemented with multiple co-oxidants, glucose, glucose oxidase (GOD) and commercial cellulase. An additional 48-h reaction using fresh MnP/co-oxidants increased the delignification rate to 14.2%. Simultaneous enzymatic delignification and saccharification, which occurs without a need for glucose supplementation, successfully improved the glucose yield to 160% of the reaction without MnP. Development of a more accurate imitation of the mechanisms of delignification that occurs in white-rot fungi has the potential to improve the monosaccharide yield resulting from simultaneous delignification and saccharification.

Woody cells generate lignocellulosic biomass, which is a promising sustainable bioresource for wide industrial applications. Woody cell differentiation in vascular plants, including the model plant poplar (Populus trichocarpa), is regulated by a set of NAC family transcription factors, the VASCULAR-RELATED NAC-DOMAIN (VND), NAC SECONDARY CELL WALL THICKENING PROMOTING FACTOR (NST)/SND, and SOMBRERO (SMB) (VNS)-related proteins, but the precise contributions of each VNS protein to wood quality are unknown. Here, we performed a detailed functional analysis of the poplar SMB-type VNS proteins PtVNS13–PtVNS16. PtVNS13–PtVNS16 were preferentially expressed in the roots of young poplar plantlets, similar to the Arabidopsis thalianaSMB gene. PtVNS13 and PtVNS14, as well as the NST-type PtVNS11, suppressed the abnormal root cap phenotype of the Arabidopsis sombrero-3 mutant, whereas the VND-type PtVNS07 gene did not, suggesting a functional gap between SMB- or NST-type VNS proteins and VND-type VNS proteins. Overexpressing PtVNS13–PtVNS16 in Arabidopsis seedlings and poplar leaves induced ectopic xylem-vessel-like cells with secondary wall deposition, and a transient expression assay showed that PtVNS13–16 transactivated woody-cell-related genes. Interestingly, although any VNS protein rescued the pendant stem phenotype of the Arabidopsis nst1-1 nst3-1 mutant, the resulting inflorescence stems exhibited distinct cell wall properties: poplar VNS genes generated woody cell walls with higher enzymatic saccharification efficiencies compared with Arabidopsis VNS genes. Together, our data reveal clear functional diversity among VNS proteins in woody cell differentiation and demonstrate a novel VNS-based strategy for modifying woody cell wall properties toward enhanced utilization of woody biomass.

Wood carbon (C) concentration is a key wood trait that varies widely among tree species, but our understanding of the factors governing this trait is limited, despite reason to hypothesize that wood C varies systematically across environmental gradients. We compiled a novel database of 1145 geo-referenced wood C observations from 415 species, to elucidate climate correlates of wood C concentrations, and test if these relationships differ across tissue types and major taxonomic divisions (i.e. angiosperms vs gymnosperms). Climate variables, including mean annual temperature (MAT) and precipitation and temperature seasonality, are significantly correlated with wood C concentrations. Relationships between wood C and these variables differ across tissue types and taxonomic divisions, yet there is a negative relationship between wood C and MAT that exists across all tissues and species groups. Wood C concentrations in trees are influenced by climate, with experimental evidence (albeit scant) indicating that climate-driven changes in lignin concentrations likely govern these relationships. Our study presents among the first lines of evidence indicating that wood C concentrations are correlated with environmental conditions, thereby enhancing our understanding of the potential adaptive significance of wood C variation in trees.

Niche partitioning among species with virtually the same requirements is a fundamental concept in ecology. Nevertheless, some authors suggest that niches have little involvement in structuring communities. This study was done in the Pardubice Region (Czech Republic) on saproxylic beetles with morphologically similar larvae and very specific requirements, which are related to their obligatory dependence on dead wood material: Cucujus cinnaberinus, Pyrochroa coccinea, and Schizotus pectinicornis. This work was performed on 232 dead wood pieces at the landscape scale over six years. Based on the factors studied, the relationships among these species indicated that their co-occurrence based on species presence and absence was low, which indicated niche partitioning. However, based on analyses of habitat requirements and species composition using observed species abundances, there was no strong evidence for niche partitioning at either studied habitat levels, the tree and the microhabitat. The most likely reasons for the lack of strong niche partitioning were that dead wood is a rich resource and co-occurrence of saproxylic community was not driven by resource competition. This might be consistent with the theory that biodiversity could be controlled by the neutral drift of species abundance. Nevertheless, niche partitioning could be ongoing, meaning that the expanding C. cinnaberinus may have an advantage over the pyrochroids and could dominate in the long term.

Numerous bird species, often rare or endangered, rely on the presence of standing and downed deadwood for shelter, nesting, and foraging. Habitat quality was evaluated on the basis of deadwood volume, the density of large standing deadwood, and the space filling index (SFI). The SFI reflects the degree of space filling of the bottom layers taking into account tree trunks, seedlings, saplings, ground vegetation, stumps, and downed deadwood. Analysis encompassed all special protection areas (SPAs) in Poland (a total of 107 SPAs containing 7974 sample plots monitored under the National Forest Inventory). An additional in-depth analysis was conducted for 30 SPAs with the greatest share of forest habitats. The studied indicators varied substantially both between and within individual SPAs, with deadwood volume ranging from 1.3 to 50.5 m3 ha−1 (mean of 9.0 m3 ha−1) and the density of large standing deadwood (diameter at breast height ≥ 30 cm) from 0.1 to 16.0 ind ha−1 (mean of 2.2 ind ha−1). These values were relatively low compared to the density of living trees with corresponding dimensions (111 ind ha−1). SFI analysis indicated high or very high space filling of the bottom forest layers on 14–56% of sample plots in a given SPA. The presence of deadwood was found to be significantly positively affected by SPA location in the mountains, a greater proportion of sites with higher fertility, a greater share of forest area under strict protection, as well as higher stand volume within a given SPA. The correlation between deadwood volume and the density of birds (primary and secondary cavity nesters) in individual SPAs was positive (R = 0.60). As compared to lowland areas, SPAs in mountain areas are generally characterized by high stand volumes, a greater density of large living trees, and a greater amount of diverse deadwood. In those areas conservation measures should involve continuous monitoring and diagnosing of any problems associated with the populations of individual bird species; focused efforts should be implemented to support those species that exhibit unfavorable population trends. In most lowland SPAs measures aimed at the improvement of site conditions for birds must be more extensive than in the mountains, with a low abundance of dead trees (especially large ones). These parameters can be improved by retaining some senescent stands in managed forests until their natural death and implementing a strict protection regime in areas of high conservation value.

Marine wood-boring invertebrates rapidly fragment coarse woody debris in the sea. These wood borers have the ability to digest wood cellulose, but other potential food sources have been less investigated. To assess the contribution of each potential food source to the diet of wood borers, we traced seasonal and environmental changes in δ13C of shipworms cultured under the same experimental conditions and related these changes to variations in δ13C of potential food sources, i.e., wood log and particulate organic matter (POM) by using multiple linear regression models rather than the Bayesian mixing model. Based on the standardized partial regression coefficients in the model, it became clear that wood-derived organic carbon was the main carbon source for the teredinids, and POM also accounted for 37.9% of the teredinids' carbon source. Furthermore, we clarified variations in supplemental nitrogen sources for the teredinids: one species depended on both POM and wood log, whereas the other three species depended on either POM or wood log for their nitrogen source. δ13C values of another wood-boring bivalve of Martesia (Pholadidae) increase as it grows, which suggests that the bivalve switches its feeding strategy from xylophagous to filter feeding as it grows. Wood borers are known to accelerate the transfer of organic materials derived from wood logs to marine ecosystems. However, this study suggests that autochthonous production strongly contribute to the diet of marine wood borers, helping them to decompose wood logs in temperate shallow water.

Bark beetles and root weevils can impact forests through tree death on landscape scales. Recently, subterranean termites have been linked to these beetles via the presence of bluestain fungi (Ascomycota: Ophiostomataceae), which are vectored to trees by beetles. However, only a small subset of bluestain species have been examined. Here, we tested whether termite-bluestain association patterns in the field reflect termite feeding preference in laboratory choice trials. We documented the presence of four bluestain fungi (Leptographium procerum (W.B. Kendr.), L. terebrantis (Barras & Perry), Grosmannia huntii (Rob.-Jeffr.), and G. alacris (T.A. Duong, Z.W. de Beer & M.J. Wingf.) in the roots of 2,350 loblolly pine trees in the southeastern United States and whether termites were present or absent on these roots and paired this with laboratory choice feeding trials. Termites were found 2.5-fold on tree roots with at least one bluestain fungus present than tree roots without bluestain fungi. Although termites in this study and others were associated with L. procerum, L. terebrantis, and marginally G. huntii, termites only showed preferential feeding on wood inoculated with G. huntii in laboratory trials. This suggests that increased termite presence on wood with bluestain fungi may be driven by factors other than increased wood palatability. Termites could thus disproportionately affect wood turnover rates for specific pools (e.g., bark beetle and root weevil attacked trees) and in some cases (e.g., G. huntii) accelerate wood decomposition. This study supports the growing evidence that the association between subterranean termites and bluestain fungi is spatially and taxonomically widespread.

Summary Plant cellulose is synthesized by a large plasma membrane‐localized cellulose synthase (CesA) complex. However, an overall functional determination of secondary cell wall (SCW) CesAs is still lacking in trees, especially one based on gene knockouts.Here, the Cas9/gRNA‐induced knockouts of PtrCesA4, 7A, 7B, 8A and 8B genes were produced in Populus trichocarpa. Based on anatomical, immunohistochemical and wood composition evidence, we gained a comprehensive understanding of five SCW PtrCesAs at the genetic level.Complete loss of PtrCesA4, 7A/B or 8A/B led to similar morphological abnormalities, indicating similar and nonredundant genetic functions. The absence of the gelatinous (G) layer, one‐layer‐walled fibres and a 90% decrease in cellulose in these mutant woods revealed that the three classes of SCW PtrCesAs are essential for multilayered SCW structure and wood G‐fibre. In addition, the mutant primary and secondary phloem fibres lost the n(G + L)‐ and G‐layers and retained the thicker S‐layers (L, lignified; S, secondary). Together with polysaccharide immunolocalization data, these findings suggest differences in the role of SCW PtrCesAs‐synthesized cellulose in wood and phloem fibre wall structures.Overall, this functional understanding of the SCW PtrCesAs provides further insights into the impact of lacking cellulose biosynthesis on growth, SCW, wood G‐fibre and phloem fibre wall structures in the tree.

Sap velocity measurements are useful in fields ranging from plant water relations to hydrology at a variety of scales. Techniques based on pulses of heat are among the most common methods to measure sap velocity, but most lack ability to measure velocities across a wide range, including very high, very low and negative velocities (reverse flow). We propose a new method, the double-ratio method (DRM), which is robust across an unprecedented range of sap velocities and provides real-time estimates of the thermal diffusivity of wood. The DRM employs one temperature sensor upstream (proximal) and two sensors downstream (distal) to the source of heat. This facilitates several theoretical, heat-based approaches to quantifying sap velocity. We tested the DRM using whole-tree lysimetry in Eucalyptus cypellocarpa L.A.S. Johnson and found strong agreement across a wide range of velocities.

Brassinosteroid-insensitive-1 (BRI1) plays important roles in various signalling pathways controlling plant growth and development. However, the regulatory mechanism of BRI1 in brassinosteroid (BR)-mediated signalling for shoot growth and wood formation in woody plants is largely unknown. In this study, PtBRI1.2, a brassinosteroid-insensitive-1 gene, was overexpressed in poplar. Shoot growth and wood formation of transgenic plants were examined and the regulatory genes involved were verified. PtBRI1.2 was localized to the plasma membrane, with a predominant expression in leaves. Ectopic expression of PtBRI1.2 in Arabidopsis bri1-201 and bri1-5 mutants rescued their retarded-growth phenotype. Overexpression of PtBRI1.2 in poplar promoted shoot growth and wood formation in transgenic plants. Further studies revealed that overexpression of PtBRI1.2 promoted the accumulation of PtBZR1 (BRASSINAZOLE RESISTANT1) in the nucleus, which subsequently activated PtWNDs (WOOD-ASSOCIATED NAC DOMAIN transcription factors) to up-regulate expression of secondary cell wall biosynthesis genes involved in wood formation. Our results suggest that PtBRI1.2 plays a crucial role in regulating shoot growth and wood formation by activating BR signalling.

The objective of this study was to investigate the adhesion performance of eight fast-growing wood species, namely, Enterolobium cyclocarpum, Paraserianthes falcataria, Shorea sp., Toona sinensis, Gmelina arborea, Pinus merkusii, Acacia mangium, and Acacia hybrid obtained from the tropical region. The wettability test was conducted by contact angle measurement, while the adhesion performance was expressed by strength retention (SR) and wood failure (WF) percentages through block shear tested under the Japanese Agricultural Standard (JAS). Results showed that smaller contact angle for P. falcataria followed by E. cyclocarpum, P. merkusii, T. sinensis, and Shorea indicated that the surfaces were easier to be glued than that of G. arborea, A. mangium, and A. hybrid. It is indicated that high wettability of P. falcataria, E. cyclocarpum, P. merkusii, T. sinensis, and Shorea resulted in the better adhesive spread and more intimate contact between the wood surface and the adhesive as shown by their high SR with high WF percentages.

Plant fibers constitute a major part of renewable biomass and are a valuable resource for various industries, composite material chemistry, and bioconversion processes. Plant fibers are specialized long cells with thick cell walls that are a convenient model for investigating fundamental issues such as individual cell biogenesis, cell wall formation, and cell specialization. Plant fibers usually accumulate secondary cell walls, which contain lignin, cellulose, and hemicelluloses like xylan, and are laid over thin primary cell walls.

Bamboos are arborescent monocotyledons that have no secondary growth, but can continually produce conduits with diameters appropriate to the current size of the plant. Here, we studied bamboo hydraulic architecture to address the mechanisms involved in compensating for the increase in hydraulic resistance during ontogeny. We measured the hydraulic weighted vessel diameters (Dh ) at different distances from the apex along the stem of Bambusa textilis. The hydraulic resistance of different components and individuals of different heights were quantified using the high-pressure flowmeter method. The Dh showed tip-to-base widening with a scaling exponent in the range of those reported for trees. Although theoretical hydraulic conductivity decreased from base-to-tip, leaf-specific conductivity did not change. Leaves contributed the most to the whole-shoot hydraulic resistance, followed by the leaf-bearing branches. Roots contributed c. 13% to whole-plant resistance. Interestingly, taller individuals showed lower whole-shoot resistance owing to an increased number of resistances in parallel (side-branches), while leaf-specific resistance was independent of plant size. Tip-to-base vessel widening and height-independent constant leaf-specific conductance could be mechanisms for hydraulic optimization in B. textilis. Similar patterns have also been found in woody plants with secondary growth, but this bamboo exhibits them without secondary growth.

Longhorn beetles are extremely rich wood-boring insects possessing larvae that feed on the xylem of trees and/or lianas, which have detrimental effects on plants; in turn, the hosting plants may play a fundamental role in shaping the longhorn beetle community assemblage. However, factors determining the community assemblage of wood-boring longhorn beetles, particularly along the multiple spatial scales is still in need of further exploration. In this study, we designed an experiment across several spatial scales (from local to macro scales) from tropical to temperate climate gradients in Yunnan province, southwest China to examine to what extend the attributes of host-specificity is shaping the community assemblage along different spatial scales. This study concludes that (1) the wood-boring longhorn beetles showed attributes of host-specificity to a certain degree at the community level, (2) biotic (host plant specificity) and abiotic (climatic gradients) factors jointly shaped community composition of this species along the multiple spatial scales, (3) biotic interactions have a prominent effect on the community composition of this species at local-scale while macroclimatic gradients impose the major control on it at macro-scale. Thus, this study highlights the significance of host specificity in affecting the wood-boring longhorn beetle community assemblage, particularly at local scales.

Tension wood (TW) is a specialized xylem tissue developed under mechanical/tension stress in angiosperm trees. TW development involves transregulation of secondary cell wall genes, which leads to altered wood properties for stress adaptation. We induced TW in the stems of black cottonwood (Populus trichocarpa, Nisqually-1) and identified two significantly repressed transcription factor (TF) genes: class B3 heat-shock TF (HSFB3-1) and MYB092. Transcriptomic analysis and chromatin immunoprecipitation (ChIP) were used to identify direct TF–DNA interactions in P. trichocarpa xylem protoplasts overexpressing the TFs. This analysis established a transcriptional regulatory network in which PtrHSFB3-1 and PtrMYB092 directly activate 8 and 11 monolignol genes, respectively. The TF–DNA interactions were verified for their specificity and transactivator roles in 35 independent CRISPR-based biallelic mutants and overexpression transgenic lines of PtrHSFB3-1 and PtrMYB092 in P. trichocarpa. The gene-edited trees (mimicking the repressed PtrHSFB3-1 and PtrMYB092 under tension stress) have stem wood composition resembling that of TW during normal growth and under tension stress (i.e., low lignin and high cellulose), whereas the overexpressors showed an opposite effect (high lignin and low cellulose). Individual overexpression of the TFs impeded lignin reduction under tension stress and restored high levels of lignin biosynthesis in the TW. This study offers biological insights to further uncover how metabolism, growth, and stress adaptation are coordinately regulated in trees.

A key uncertainty in quantifying dead wood carbon (C) stocks—which comprise ~8% of total forest C pools globally—is a lack of accurate dead wood C fractions (CFs) that are employed to convert dead woody biomass into C. Most C estimation protocols utilize a default dead wood CF of 50%, but live tree studies suggest this value is an over-estimate. Here, we compile and analyze a global database of dead wood CFs in trees, showing that dead wood CFs average 48.5% across forests, deviating significantly from 50%, and varying systematically among biomes, taxonomic divisions, tissue types, and decay classes. Utilizing data-driven dead wood CFs in tropical forests alone may correct systematic overestimates in dead wood C stocks of ~3.0 Pg C: an estimate approaching nearly the entire dead wood C pool in the temperate forest biome. We provide for the first time, robust empirical dead wood CFs to inform global forest C estimation., Tree mortality is increasing with climate change, which suggests that the biomass of dead wood is likely becoming more and more important to the global carbon cycle. Here, the authors perform a meta-analysis of the carbon content of dead wood and find that past estimates of total forest carbon were overestimated.

An endophytic bacterium Bacillus velezensis BY6 was isolated from the wood stems of healthy Populus davidiana × P. alba var. pyramidalis (PdPap). The BY6 strain can inhibit pathogenic fungus Alternaria alternate in PdPap and promote growth of PdPap seedlings. In the present study, we used the Pacific Biosciences long-read sequencing platform, a single-molecule real-time (SMRT) technology for strain BY6, to perform complete genome sequencing. The genome size was 3,898,273 bp, the number of genes was 4,045, and the average GC content was 47.33%. A complete genome of strain BY6 contained 110 secondary metabolite gene clusters. Nine of the secondary metabolite gene clusters exhibited antifungal activity and promoted growth functions primarily involved in the synthesis of surfactin, bacteriocins, accumulated iron ions, and related antibiotics. Gene clusters provide genetic resources for biotechnology and genetic engineering, and enhance understanding of the relationship between microorganisms and plants.

The thermal conversion of woody biomass is increasingly critical for the development of the energy processing technologies and fire safety engineering. The combustion characteristics, kinetics, and thermodynamics of pine wood were characterized through a thermogravimetric analyzer in the air atmosphere. There were two apparent peaks in the derivative TG curves for pine wood. The combustion process of pine wood was divided into two stages. Therein, the first stage occurring in the conversion degree range of 0-0.6 may be considered a one-step reaction. It was easier for pine wood to decompose under air than under nitrogen. Moreover, the first stage of pine wood combustion may be characterized by the diffusion model g(α) = [1 - (1 - α)1/3]2. The kinetic modeling showed a good agreement between the predicted and experimental conversion degree curves. In addition, the high comprehensive combustion index of pine wood at 10 K min-1 (6.73 × 10-7 %2 min-2 K-3) showed its great potential for bioenergy generation. Besides, both the value of ΔH and ΔS exhibited similar patterns with the activation energy value versus conversion degree, while the ΔG value almost remained at a positive constant with conversion degree. The average ΔH, ΔG, and ΔS value was nearly equal under different heating rates.

In this study, contrary to untreated poplar fiber, processing of alkali-treated poplar fiber was optimized for the enzymatic saccharification. Considering reducing sugar content as the evaluation index, pH, temperature, time, amount of enzyme, and rotational speed of shaker were standardized to optimize the sugar production by enzymatic hydrolysis. Using response surface methodology, the optimum technological condition of enzymatic hydrolysis was found to be utilizing 43 mg cellulase at 46 °C for 50 h. At this, the sugar conversion amount of NaOH or H2O2-NaOH pretreated poplar was 164.62 mg/g and 218.82 mg/g respectively. It was a corresponding increase of 446.73% or 626.75% than that of poplar fiber without a pretreatment. At a low concentration, metal ions and surfactants promoted the conversion of reducing sugar. Especially, 0.01 g/L Mn2+ and 0.50 g/L hexadecyl trimethyl ammonium bromide (CTAB) produced the best effect and increased the conversion rate of reducing sugar by 23.62% and 21.44% respectively. Also, the effect of the combination of metal ions and surfactants was better than that of a single accelerator. Tropical forests vary widely in biomass carbon (C) stocks and fluxes even after controlling for forest age. A mechanistic understanding of this variation is critical to accurately predicting responses to global change. We review empirical studies of spatial variation in tropical forest biomass, productivity and woody residence time, focusing on mature forests. Woody productivity and biomass decrease from wet to dry forests and with elevation. Within lowland forests, productivity and biomass increase with temperature in wet forests, but decrease with temperature where water becomes limiting. Woody productivity increases with soil fertility, whereas residence time decreases, and biomass responses are variable, consistent with an overall unimodal relationship. Areas with higher disturbance rates and intensities have lower woody residence time and biomass. These environmental gradients all involve both direct effects of changing environments on forest C fluxes and shifts in functional composition - including changing abundances of lianas - that substantially mitigate or exacerbate direct effects. Biogeographic realms differ significantly and importantly in productivity and biomass, even after controlling for climate and biogeochemistry, further demonstrating the importance of plant species composition. Capturing these patterns in global vegetation models requires better mechanistic representation of water and nutrient limitation, plant compositional shifts and tree mortality.

In conifers, xylogenesis produces during a growing season a very characteristic tree-ring structure: large thin-walled earlywood cells followed by narrow thick-walled latewood cells. Although many factors influence the dynamics of differentiation and the final dimensions of xylem cells, the associated patterns of variation remain very stable from one year to the next. While radial growth is characterised by an S-shaped curve, the widths of xylem differentiation zones exhibit characteristic skewed bell-shaped curves. These elements suggest a strong internal control of xylogenesis. It has long been hypothesised that much of this regulation relies on a morphogenetic gradient of auxin. However, recent modelling works have shown that while this hypothesis could account for the dynamics of stem radial growth and the zonation of the developing xylem, it failed to reproduce the characteristic tree-ring structure. Here we investigated the hypothesis of a regulation by a crosstalk between auxin and a second biochemical signal, using dynamical modelling. We found that, in conifers, such a crosstalk is sufficient to simulate the characteristic features of wood formation dynamics, as well as the resulting tree-ring structure. In this model, auxin controls cell enlargement rates while another signal (e.g., cytokinin, TDIF) drives cell division and auxin polar transport.HighlightA dynamical model proves that two interacting signals (auxin, plus a cytokinin or the TDIF peptide) can drive wood formation dynamics and tree-ring structure development in conifers.

The impacts of woody encroachment and removal on ecosystems are highly variable and are thought to be related to the traits of the individual woody species. Decisions on whether to remove or to retain woody plants are hampered by a lack of empirical evidence of the relationship between woody traits and the ecosystem consequences of their removal or retention. We used a global meta-analysis of 149 ecosystem attributes from 172 woody species to evaluate the relative effects of woody plant traits and abiotic environmental variables on the ecosystem consequences of woody encroachment and removal. The ecosystem consequences were closely related to woody plant traits. For example, encroachment of plants characterized by high structural traits (e.g. tall, mixed tap and fibrous roots) reduced ecosystem composition, while removal of plants characterized by high functional traits (e.g. nitrogen fixing, deciduous) reduced ecosystem function. Structural and functional traits of woody plants mainly regulated soil stability during woody encroachment and herbaceous cover after woody removal. Conversely, environmental conditions mainly affected herbaceous cover under encroachment and soil stability under removal scenarios. We demonstrate that the ecosystem consequences of encroachment and removal are closely linked to the structural and functional traits of the target woody species. Furthermore, biotic (woody plant traits) and abiotic (climate, soils) factors have different impacts on regulating trade-offs between ecosystem responses under these two management scenarios. Our study provides empirical support for management decisions on whether to retain or remove different woody taxa under various environments across the globe.

In trees, wood formation needs carbon import from the photosynthetic source tissues. Sugar transporters play important roles in carbohydrate transport into wood-forming cells. Sugars will eventually be exported transporters (SWEETs) play essential roles in many physiological processes. However, the roles of this family in the growth and development of woody plants have not been systematically investigated. In this study, 27 SWEET genes were identified in the Populus trichocarpa genome. These SWEET genes were classified into four clades based on their phylogenetic relationships, gene structures, conserved motifs and chromosomal locations. Representative SWEET members from each clade were selected for further studies. The PagSWEETs of Populus alba × Populus glandulosa were localized to plasma membrane, vacuolar, endoplasmic reticulum or Golgi. Quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR) analysis showed that PagSWEETs have distinct expression patterns in various tissues, and PagSWEET5, 7, 10b, 10c, 15b, 17a and 17c exhibited high expression levels in stems. PagSWEET7 is localized to the cytoplasmic membrane and specifically expressed in the phloem as detected by histochemical GUS ($\beta $ - glucuronidase) assays. Xylem production and xylem sugar content were greater in developing wood of PagSWEET7 overexpression than wild-type lines. Collectively, these results provide valuable information for further investigating functions of PagSWEET genes, and identify PagSWEET7 as a candidate gene for using biotechnology to modify the wood formation in poplar.

The adsorption behaviors of two glucanases, TvEG and TrCel7A, on three lignins were investigated. Three lignins were isolated from raw aspen and its pretreated solid residue. The isolated lignins were labeled as Asp-MWL, DA-MWL (pretreated by dilute acid), and GL-MWL (pretreated by green liquor), respectively. The surface properties of lignins and spin-coated lignin films were characterized by zeta potential, atomic force microscope (AFM) and contact angle. The enzyme adsorption behavior was monitored by quartz crystal microbalance (QCM) and fluorescence spectrometer. TlCel7A had similar adsorption capacities on the three lignin films but were higher than those of TvEG. The TrCel7A adsorptions on the three lignin films were affected by synergistic effect of electrostatic and hydrophobic interaction while the TvEG adsorptions on the three lignin films were mainly dominated by hydrophobic action. The adsorption capacities of TlCel7A and TvEG on the three lignin films were decreased by adding SL. Plausible explanation was that the SL and glucanase formed a complex with more negative charges, which suppressed the adsorption of glucannase on lignin through electrostatic repulsion. It also explained the improved enzymatic hydrolysis efficiency of lignocellulose upon adding SL.

Mass loss (ML) of wood caused by thermal degradation is one of the most important features of the thermal treatments and referred to as an indicator of intensity and quality of the process. The ML is proportional to the quantity of the effective heat power exchanged during the treatment process, represented by the area of the temperature profile versus time during the process. In this paper a model for the ML prediction based on the relative area was discussed. The model proposed an analytical solution to take into account the non-linear trend of ML when plotted versus temperature and time as observed in isothermal experiments. The model was validated comparing calculated and measured final ML of samples treated during thermal modification tests with different temperature profiles. The results showed that the relative area calculated in a transformed time-temperature space improves the correlation with the measured ML.

A classic theory proposes that plant xylem cannot be both highly efficient in water transport and resistant to embolism, and therefore a hydraulic efficiency-safety trade-off should exist. However, the trade-off is weak, and many species exhibit both low efficiency and low safety, falling outside of the expected trade-off space. It remains unclear under what climatic conditions these species could maintain competitive fitness. We compiled hydraulic efficiency and safety traits for 682 observations of 499 woody species from 178 sites world-wide and measured the position of each observation within the proposed trade-off space. For both angiosperms and gymnosperms, observations from sites with high climatic seasonality, especially precipitation seasonality, tended to have higher hydraulic safety and efficiency than observations from sites with low seasonality. Specifically, high vapour pressure deficit, high solar radiation, and low precipitation during the wet season were driving factors. Strong climatic seasonality and drought in both dry and wet seasons appear to be ecological filters that select for species with co-optimized safety and efficiency, whereas the opposite environmental conditions may allow the existence of plants with low efficiency and safety.

International audience; Defining plant hydraulic traits is central to the quantification of ecohydrological processes ranging from land-atmosphere interactions, to tree mortality and water-carbon budgets. A key plant trait is the xylem specific hydraulic conductivity (K-x), that describes the plant's vascular system capacity to transport water. While xylem's vessels and tracheids are dead upon maturity, the xylem is neither inert nor deadwood, various components of the sapwood and surrounding tissue remaining alive and functional. Moreover, the established definition of K-x assumes linear relations between water flux and pressure gradient by tacitly considering the xylem as a "passive conduit". Here, we re-examine this notion of an inert xylem by systematically characterizing xylem flow in several woody plants using K-x measurements under constant and cyclic pressure gradients. Results show a temporal and pressure gradient dependence of K-x. Additionally, microscopic features in "living branches" are irreversibly modified upon drying of the xylem, thus differentiating the macroscopic definition of K-x for living and dead xylem. The findings highlight the picture of the xylem as a complex and delicate conductive system whose hydraulic behaviour transcends a passive gradient-based flow. The study sheds new light on xylem conceptualization, conductivity measurement protocols, in situ long-distance water transport and ecosystem modelling.

Water released from wood during transpiration (capacitance) can meaningfully affect daily water use and drought response. To provide context for better understanding of capacitance mechanisms, we investigated links between capacitance and wood anatomy. On twigs of 30 temperate angiosperm tree species, we measured day capacitance (between predawn and midday), water content, wood density, and anatomical traits, that is, vessel dimensions, tissue fractions, and vessel-tissue contact fractions (fraction of vessel circumference in contact with other tissues). Across all species, wood density (WD) and predawn lumen volumetric water content (VWCL-pd ) together were the strongest predictors of day capacitance (r2adj = .44). Vessel-tissue contact fractions explained an additional ~10% of the variation in day capacitance. Regression models were not improved by including tissue lumen fractions. Among diffuse-porous species, VWCL-pd and vessel-ray contact fraction together were the best predictors of day capacitance, whereas among semi/ring-porous species, VWCL-pd , WD and vessel-fibre contact fraction were the best predictors. At predawn, wood was less than fully saturated for all species (lumen relative water content = 0.52 ± 0.17). Our findings imply that day capacitance depends on the amount of stored water, tissue connectivity and the bulk wood properties arising from WD (e.g., elasticity), rather than the fraction of any particular tissue.

Premise Photosynthetic stems represent a source of extra carbon in plants from hot and dry environments, but little is known about how leaves and photosynthetic stems differ in terms of photosynthetic capacity, trait coordination, and responses to seasonal drought in subtropical systems. Methods We studied photosynthetic, hydraulic, morphometric (specific leaf area [SLA], wood density [WD]), and biochemical (C and N isotopes) traits in leaves and photosynthetic stems of 12 plant species from a sarcocaulescent scrub in the southern Baja California Peninsula, Mexico, in wet and dry seasons. Results Leaves and stems had similar mean photosynthetic capacity, as evaluated by chlorophyll fluorescence traits, indicating similar investment in leaf and stem photosynthesis. We did not find a relationship between stem hydraulic conductivity and leaf or stem photosynthetic traits. However, we found resource allocation trade-offs, between WD and both stem hydraulic conductivity and SLA. Leaf and stem photosynthetic traits did not change with season, but specific stem area was one of the few traits that changed the most between seasons-it increased during the dry season by as much as 154% indicating substantial water storage. Conclusions Our results indicate the same proportional investment in photosynthetic capacity and dry matter in both leaves and photosynthetic stems across all 12 species. We identified multiple strategies at this seasonal site, with species ranging from high WD, low SLA, low hydraulic conductivity, and high specific bark area on one end of the spectrum and opposite traits on the other end.

In the technology of gluing solid wood, various adhesives are used, which, in addition to ensuring the strength of the gluing, must also be characterized by a number of rheological and colloidal-chemical properties, namely: have acceptable viscosity, low contact angles, have good spreading and enveloping ability, penetrate deep into the wood , providing reliable cohesive-adhesive contact and much more. Today, adhesives based on PVA dispersions and urea formaldehydes are used, the properties of which have been sufficiently studied and tested in practice. An alternative to organic-based adhesives is mineral adhesives, in particular aluminosilicate adhesives, which are non-flammable and have high mechanical strength. Object of research:rheokinetic patterns and processes of formation of colloidal-chemical structures of aluminosilicate adhesives modified with compounds of the ANO3and ANO3×nH2O type. Investigated problem:Considering that aluminosilicate adhesives are multiphase reactive suspensions, the viscosity of which changes over time and affects their viability, the issue of viscosity stabilization due to the introduction of electrolyte salts into their composition is relevant, and will also contribute to improving the wettability and spreading of the adhesive during applying them to substrates and gluing samples from various types of wood. Main scientific results:The effect of the concentrations of compounds of the ANO3and ANO3×nH2O type on the rheokinetic and chemical-colloidal properties of the aluminosilicate adhesive has been established, which will make it possible to use aluminosilicate adhesives in practice for gluing wood of various species, as well as to work out possible technological methods of their application, taking into account the obtained data on the viscosity, wetting and spreadability of the modified adhesive on the surface of the wood substrate. Area of practical use of the research results:woodworking enterprises for the production of glued solid wood products. Innovative technological product:aluminosilicate adhesives modified with ANO3and ANO3×nH2O nitrate additives for gluing wood of various species. Scope of application of an innovative technological product:the application of these approaches will make it possible to obtain glued materials from wood from different species with improved technological and physical and mechanical properties of the glue seam and, accordingly, to increase the quality and reliability of products.

Carbon reserve use is a major drought response in trees, enabling tree survival in conditions prohibiting photosynthesis. However, regulation of starch metabolism under drought at the whole-tree scale is still poorly understood. To this end, we combined measurements of nonstructural carbohydrates (NSCs), tree physiology and gene expression. The experiment was conducted outside on olive trees in pots under 90 d of seasonal spring to summer warming. Half of the trees were also subjected to limited water conditions for 28 d. Photosynthesis decreased in dehydrating trees from 19 to 0.5 µmol m-2 s-1 during the drought period. Starch degradation and mannitol production were a major drought response, with mannitol increasing to 71% and 41% out of total NSCs in shoots and roots, respectively. We identified the gene family members potentially relevant either to long-term or stress-induced carbon storage. Partitioning of expression patterns among β amylase and starch synthase family members was observed, with three β amylases possibly facilitating the rapid starch degradation under heat and drought. Our results suggest a group of stress-related, starch metabolism genes, correlated with NSC fluctuations during drought and recovery. The daily starch metabolism gene expression was different from the stress-mode starch metabolism pattern, where some genes are uniquely expressed during the stress-mode response.

Wood is a naturally occurring material widely used for construction. Due to its natural origin, wood properties vary and its behaviour is complex. This paper shows an implementation of a multi-surface elasto-plastic constitutive material model for wood into a custom explicit material point method code. The constitutive model chosen is one proposed by Schmidt & Kaliske with minor modifications to ensure better internal consistency. The model parameters are chosen based on literature data for spruce. The paper presents two Convected Particle Domain Interpolation Material Point Method simulations of experiments, both performed with the previously established model parameters. The first simulation replicates a compression test of a spruce specimen perpendicular to grain direction, carried out at the Department of Civil Engineering, Aalto University. The second simulation replicates an experiment from literature, in which a spruce specimen with knots is tensioned until failure. The numerical simulations successfully replicate the experimental outcomes qualitatively in terms of the deformation and load-displacement curves. Simulations of the three knotted specimens under tension, with introduced slight variation in wood grain direction, replicate different failure patterns with a similar failure load, resembling the behaviour of natural wooden structural elements. Additionally, one of the obtained failure patterns replicates that of the experiment well.

Hemicelluloses, a family of heterogeneous polysaccharides with complex molecular structures, constitute a fundamental component of lignocellulosic biomass. However, the contribution of each hemicellulose type to the mechanical properties of secondary plant cell walls remains elusive. Here we homogeneously incorporate different combinations of extracted and purified hemicelluloses (xylans and glucomannans) from softwood and hardwood species into self-assembled networks during cellulose biosynthesis in a bacterial model, without altering the morphology and the crystallinity of the cellulose bundles. These composite hydrogels can be therefore envisioned as models of secondary plant cell walls prior to lignification. The incorporated hemicelluloses exhibit both a rigid phase having close interactions with cellulose, together with a flexible phase contributing to the multiscale architecture of the bacterial cellulose hydrogels. The wood hemicelluloses exhibit distinct biomechanical contributions, with glucomannans increasing the elastic modulus in compression, and xylans contributing to a dramatic increase of the elongation at break under tension. These diverging effects cannot be explained solely from the nature of their direct interactions with cellulose, but can be related to the distinct molecular structure of wood xylans and mannans, the multiphase architecture of the hydrogels and the aggregative effects amongst hemicellulose-coated fibrils. Our study contributes to understanding the specific roles of wood xylans and glucomannans in the biomechanical integrity of secondary cell walls in tension and compression and has significance for the development of lignocellulosic materials with controlled assembly and tailored mechanical properties., Hemicelluloses are an essential constituent of plant cell walls, but the individual biomechanical roles remain elusive. Here the authors report on the interaction of wood hemicellulose with bacterial cellulose during deposition and explore the resultant fibrillar architecture and mechanical properties.

Alternative splicing (AS) is an important post-transcriptional process to enhance proteome diversity in eukaryotic organisms. In plants, numerous reports have primarily focused on AS analysis in model plant species or herbaceous plants, leading to a notable lack of research on AS in woody plants. More importantly, emerging evidence indicates that many important traits, including wood formation and stress resistance, in woody plants are controlled by AS. In this review article, we summarize the current progress of all kinds of AS studies in different tree species at various stages of development and in response to various stresses, revealing the significant role played by AS in woody plants, as well as the similar properties and differential regulation within their herbaceous counterparts. Furthermore, we propose several potential strategies to facilitate the functional characterization of splicing factors in woody plants and evaluate a general pipeline for the systematic characterization of splicing isoforms in a complex AS regulatory network. The utilization of genetic studies and high-throughput omics integration approaches to analyze AS genes and splicing factors is likely to further advance our understanding of AS modulation in woody plants.

The present study addresses the herbicidal activity and biological effects of the metribuzin (MET) and tribenuron-methyl (TBM) herbicides used to control various weed species (Amaranthus retroflexus, Sinapis arvensis, and Leucanthemum maximum). The effects of the free herbicides and the herbicides embedded in granules of degradable polymer poly-3-hydroxybutyrate [P(3HB)] blended with birch wood flour were compared. Metribuzin, regardless of the form, caused 100% mortality of the three weeds by day 21. The herbicidal activity of tribenuron-methyl was lower than that of metribuzin, but the embedded TBM was superior to the free herbicide in the length and strength of its action on the weeds. Both metribuzin forms dramatically decreased the main parameters of fluorescence: maximum quantum yield of photosystem-II [Y(II)max], maximum quantum yield of non-photochemical quenching [Y(NPQ)max], and maximum rate of non-cyclic electron transport [ETRmax] and concentrations of chlorophyll a and b. The effect of the embedded TBM on the photosynthetic activity of the weeds was lower in the first two weeks of the growth of herbicide-treated plants but lasted longer than the effect of the free TBM and increased over time. Embedding of metribuzin in the matrix of degradable blend did not decrease its herbicidal activity.

Wood density (WD) is often used as a proxy for hydraulic traits such as vulnerability to drought-induced xylem cavitation and maximum water transport capacity, with dense-wooded species generally being more resistant to drought-induced xylem cavitation, having lower rates of maximum water transport and lower sapwood capacitance than light-wooded species. However, relationships between WD and the hydraulic traits that they aim to predict have not been well established in tropical forests, where modeling is necessary to predict drought responses for a high diversity of unmeasured species. We evaluated WD and relationships with stem xylem vulnerability by measuring cavitation curves, sapwood water release curves and minimum seasonal water potential (Ψmin) on upper canopy branches of six tree species and three liana species from a single wet tropical forest site in Panama. The objective was to better understand coordination and trade-offs among hydraulic traits and the potential utility of these relationships for modeling purposes. We found that parameters from sapwood water release curves such as capacitance, saturated water content and sapwood turgor loss point (Ψtlp,x) were related to WD, whereas stem vulnerability curve parameters were not. However, the water potential corresponding to 50% loss of hydraulic conductivity (P50) was related to Ψtlp,x and sapwood osmotic potential at full turgor (πo,x). Furthermore, species with lower Ψmin showed lower P50, Ψtlp,x and πo,x suggesting greater drought resistance. Our results indicate that WD is a good easy-to-measure proxy for some traits related to drought resistance, but not others. The ability of hydraulic traits such as P50 and Ψtlp,x to predict mortality must be carefully examined if WD values are to be used to predict drought responses in species without detailed physiological measurements.

Insect defoliation impacts forest productivity worldwide, highlighting the relevance of plant–insect interactions. The larch budmoth (Zeiraphera griseana Hübner) is one of the most extensively studied defoliators, where numerous tree ring-based analyses on its host (Larix decidua Mill.) have aided in identifying outbreak dynamics over the past millennia. Yet, outbreaks have been widely absent after the early 1980s, and little is known about the in situ tree physiological responses and the allocation of carbon resources during and after defoliation. In summer 2018, we tracked an ongoing larch budmoth outbreak in a well-studied larch forest in the Swiss Alps. We performed biweekly monitoring on an affected and unaffected site using a unique combination of xylogenesis observations, measurements of non-structural carbohydrates, isotopic analysis of needle assimilates and ground-based and remote-sensed leaf trait observations. The budmoth induced a defoliation that lasted 40 days and could be detected by satellite observations. Soluble sugars significantly decreased in needles and stem phloem of the defoliated trees, while starch levels remained stable in the stem and root xylem compared to the control. Carbon and oxygen isotope ratios in needle assimilates indicated that neither photosynthetic assimilation rates nor stomatal conductance was different between sites before, during and after the outbreak. Defoliated trees ceased cell wall thickening 17 days earlier than unaffected trees, showing the earliest halt of ring formation recorded from 2007 untill 2013 and causing significant thinner cell walls, particularly in the latewood. No significant differences were found for cell enlargement rates and ring width. Our study revealed that an outbreak causes a downregulation of cell wall thickening first, while no starch is mobilized or leaf physiology is adjusted to compensate for the reduced carbon source due to defoliation. Our observations suggest that affected larch trees prioritize leaf recovery and carbon storage over wood biomass development.

Widely used as wood preservatives for the last century, Pentachlorophenol (PCP) and chromated copper arsenate (CCA) have been shown to leach from treated surfaces and contaminate soil of wood storage sites. We performed a four-year field phytoremediation trial in southern Quebec (Canada) on a site contaminated with PCP and CCA with the following objectives: (1) assess the potential of willow, fescue, alfalfa and Indian mustard to tolerate and translocate CCA and PCP residues in their aerial tissues, (2) investigate the possibility of phytoextraction of dioxins and furans, and (3) test the effect of nitrogen fertilizer on phytoremediation performance. We showed that while nitrogen fertilization increased the chlorophyll content of plants, it did not result in a significantly greater plant biomass. We also showed that plants grown in the presence of PCP/CCA residues were able to translocate and concentrate trace elements in their aerial tissues, but also dioxins and furans (PCDD/F). This suggests that plants grown on sites polluted by PCP might contain dioxins and furans and should be treated as contaminated by these toxic chemicals. Finally, the reduction of soil contaminants at the end of the trial suggests that phytoremediation is a promising approach for decontaminating such sites.

Forests are among the most species rich habitats and the way they are managed influences their capacity to protect biodiversity. To fulfill increasing wood demands in the future, planted and non-planted wood production will need to expand. While biodiversity assessments usually focus on the impacts of deforestation, the effects of wood harvest are mostly not considered, especially not in a spatially explicit manner. We present here a global approach to refine the representation of forest management through allocating future wood production to planted and non-planted forests. Wood production, following wood consumption projections of three Shared Socioeconomic Pathways, was allocated using likelihood maps for planted and production forests. On a global scale, plantations for wood production were projected to increase by 45–65% and harvested area in non-planted forests by 1–17%. The biodiversity impacts of changes in wood production patterns were estimated by applying two commonly used indicators: (1) changes in species richness and (2) changes in habitat-suitable ranges of single species. The impact was analyzed using forest cover changes as reference. Our results show that, although forest cover changes have the largest impact on biodiversity, changes in wood production also have a significant effect. The magnitude of impacts caused by changes of wood production substantially differs by region and taxa. Given the importance of forest production changes in net negative emission pathways, more focus should be put on assessing the effects of future changes in wood production patterns as part of overall land use change impacts.

Despite the ecological and industrial importance of biomass accumulation in wood, the control of carbon (C) allocation to this tissue and to other tree tissues remain poorly understood. We studied sucrose synthase (SUS) to clarify its role in biomass formation and C metabolism at the whole tree level in hybrid aspen (Populus tremula × tremuloides). To this end, we analysed source leaves, phloem, developing wood, and roots of SUSRNAi trees using a combination of metabolite profiling, 13CO2 pulse labelling experiments, and long-term field experiments. The glasshouse grown SUSRNAi trees exhibited a mild stem phenotype together with a reduction in wood total C. The 13CO2 pulse labelling experiments showed an alteration in the C flow in all the analysed tissues, indicating that SUS affects C metabolism at the whole tree level. This was confirmed when the SUSRNAi trees were grown in the field over a 5-yr period; their stem height, diameter and biomass were substantially reduced. El sitio arqueologico Los Tres Ombues (partido de Punta Indio, provincia de Buenos Aires) corresponde a grupos cazadores-recolectores del Holoceno tardio y fue datado en 1110 ± 19 AP. Este trabajo aborda su conjunto antracologico, conformado por 100 carbones, con el fin de contribuir al conocimiento de los grupos que habitaron dicho sitio, y en particular, de sus practicas de obtencion y empleo de lena. Para la caracterizacion de los restos lenosos carbonizados se constituyo una coleccion de referencia experimental de las especies arboreas proximas al sitio. El conjunto arqueologico fue analizado mediante observacionesmicroscopicas y su comparacion con dicha coleccion. Se determino taxonomicamente el 78% del conjunto, con representacion de las especies arboreas Celtis tala, Jodina rhombifolia, Schinus longifolius, Scutia buxifolia, Erythrina crista-galli y Salix humboldtiana. Los resultados indican la presencia y el uso de estas especies en el pasado y permiten indagar en torno a las practicas de obtencion y empleo de lena., Los Tres Ombúes archaeological site corresponds to Late Holocene hunter-gatherers and has been dated to 1110 ± 19 BP. This article presents the anthracological evidence, consisting of 100 charcoal samples, in order to contribute to the knowledge of the human groups which inhabited the site, especially their practices of obtaining and using firewood. A reference collection was created to identify the charcoals using the tree species in the vicinity of the site. The samples were analyzed by microscopic observation and comparison with the reference collection. Seventy-eight percent of the samples were taxonomically determined, including Celtis tala, Jodina rhombifolia, Schinus longifolius, Scutia buxifolia, Erythrina cristagalli and Salix humboldtiana. (Styracaceae) is a woody species that grows on acidic soils from the Brazilian savanna with high aluminum (Al) saturation (m% > 50%), where it accumulates ~ 1500 mg Al per kg dry leaves. Using nutrient solution, a previous study showed that 1480 μM Al causes toxicity symptoms, which raises the question whether less than 1480 μM Al could cause beneficial effects on this species. Here, we checked possible altered gas exchange rates, damage to organelles in root tips and the association between Al exposure and mitochondria occurrence in cells of root tips, once organic acids from Krebs cycle exuded by the roots of this species when exposed to Al have been recently evidenced. Five-month-old plants were grown in nutrient solution with 0, 740 and 1480 μM Al for 90 days. Plants exposed to 1480 μM Al showed less developed root system, reduced plant height and low gas exchange rates in relation to those exposed to 0 and 740 μM Al, confirming that 1480 μM Al is toxic to S. camporum. However, plants exposed to 0 and 740 μM Al had similar number of leaves, plant height, root biomass, root length, total plant biomass and gas exchange rates, indicating that no beneficial effects from 740 μM Al could be noted on this species. In plants exposed to 0 and 740 μM Al, mitochondria were noted at the root tip, while at 1480 μM Al these organelles were not evident due to the conspicuous vacuolation of root cells. S. camporum shows limited tolerance to Al in nutrient solution. In addition, this species is not dependent on Al to grow and develop because the plants grew well under 0 and 740 μM Al. High- and low-acetylated galactoglucomannooligosaccharides (GGMOS_Ac and GGMOS, respectively) were assayed as substrates in the TIM-2 in vitro colon model using, as inoculum, fecal microbiota from the elderly. The effects on the microbiota and their activity were also compared to a standard ileal efflux medium (SIEM). GGMOS resulted in higher organic acid productions and higher short-chain fatty acids/total organic acid molar ratios. Although comparable Actinobacteria abundances were observed with both substrates, GGMOS fermentation led to higher Firmicutes/Bacteroidetes ratios and lower Proteobacteria percentages than GGMOS_Ac. No differences were found concerning the percentages of beneficial genus such as Blautia, Faecalibacterium, Coprococcus, or Bifidobacterium. However, higher bacterial diversities and numbers of genera such as Oscillospira and Lachnospira were found with GGMOS_Ac. This suggests that GGMOS would be more suitable substrates for the elderly, even though GGMOS_Ac promoted positive effects that support the interest of further research using these oligosaccharides as "carriers" of desired substituents.

To evaluate a strain of Fusarium verticillioides ITV03 isolated from wood residues in the Veracruz region of Mexico. Endoglucanase and β-glucosidase production by submerged fermentation was optimized using a Box–Behnken design, where the independent variables were urea, ammonium sulfate and yeast extract. After optimization, an endoglucanase activity of 0.27 U/mL was achieved; subsequently, three carbon sources were evaluated (carboxymethyl cellulose, sweet sorghum bagasse cellulose and delignified sweet sorghum bagasse (DSSB). The results showed that DSSB yielded the greatest endoglucanase (0.28 U/mL) and β-glucosidase (0.12 U/mL) activities. Both enzymatic activities were characterized for the effect of pH, temperature and thermostability. The optimal parameters of β-glucosidase and endoglucanase activity were pH 5 and 4 respectively, the optimum temperature 60 °C. These enzymes were stable at 50 °C for 150.68 h and 8.54 h, with an activation energy (Ea(day)) of 265.55 kJ/mol and 44.40 kJ/mol respectively, for β-glucosidase and endoglucanase. The present work shows that a native strain like F. verticillioides ITV03 using DSSB supplemented with nitrogen has a great potential as a producer of cellulase for lignocellulosic residue hydrolysis.

Organosolv pretreatment with two ethanol concentrations (25% and 50%, v/v) was performed to improve enzymatic saccharification of poplar sawdust. It was found that lower ethanol concentration (25%, v/v) pretreatment resulted in a higher enzymatic digestibility of poplar (38.1%) due to its higher xylan removal and similar lignin removal ratios, compared to that pretreated with 50% (v/v) ethanol pretreatment (27.5%). However, the residual lignin still exhibited a strong inhibition on enzymatic hydrolysis of organosolv-pretreated poplar (OP). Bio-surfactant preparations including tea saponin (TS), TS crude extract, and tea seed waste were applied in enzymatic hydrolysis of OP, due to their potential ability of reducing enzyme non-productive binding on lignin. Their optimal loadings in enzymatic hydrolysis of OP were optimized, which indicated that adding 0.075 g/g glucan of TS improved the 72-h glucose yield of OP by 48.3%. Moreover, adding TS crude extract and tea seed waste exhibited the better performance than TS for improving enzymatic hydrolysis of OP. It was verified that the presence of protein in TS crude extract and tea seed waste accounted for the higher improvement. More importantly, the directly application of tea seed waste in enzymatic hydrolysis could achieve the similar improvement on enzymatic hydrolysis of OP, where chemosynthetic surfactant (PEG6000) was added. The residual enzyme activities in supernatant of enzymatic hydrolysis were also determined to reveal the changes on enzyme adsorption after adding surfactants. Generally, tea seed waste could be directly applied as an alternative to chemosynthetic surfactants to promote enzymatic hydrolysis of lignocelluloses.

Environmental issues and health-benefitting design strategies have raised interest in natural and renewable building materials, resulting in an increased focus on the use of wood in built environment. The influence of wooden materials on measured and perceived indoor environment quality (IEQ) has gained attention during the past few decades, with a growing number of studies having explored the issue. This review was conducted to examine and summarise the body of research on the influence of wooden interior materials on IEQ, with an emphasis on the following themes: emissions of chemical compounds, moisture buffering of indoor air, antibacterial effects, acoustics, and psychological and physiological effects. This review found that wooden interior materials exert mainly positive or neutral effects on IEQ, such as moderating humidity fluctuations of indoor air, inducing positive feelings in occupants, and inhibiting certain bacteria. Negative effects on IEQ are limited to volatile organic compounds emitted from wood. The odour thresholds of some aldehydes and terpenes are low enough to affect the perceived IEQ. Additionally, concentrations of formaldehyde and acrolein may under certain conditions cause adverse health effects. Further studies are needed to better understand these phenomena and take advantage of the beneficial effects while hindering the unpleasant ones.
