The diversity of Japanese Dytiscidae has declined substantially in recent years, therefore, a better understanding of the biology and ecology of this insect family is essential to ensure its conservation. In this study, captive breeding experiments were conducted to elucidate the life history, and field surveys were performed to determine the reproductive habitats of the Japanese endemic species Copelatus kammuriensis Tamu and Tsukamoto, 1955. The reproductive habitats, egg-laying environments, immature periods, and unique ecology of the species were identified. The larval periods of five Copelatus species were compared, and that of C. kammuriensis was found to be the shortest; its reproductive habitats were unstable rain pools that easily dried up in the absence of rain over up to 10 days. These results suggest that a short larval period and rapid predatory behaviour are important for the survival of C. kammuriensis. [ABSTRACT FROM AUTHOR]

Studies in biology have shown that students perceive variation and evolution to be difficult areas to learn. This has sparked rote learning of variation and evolution concepts and is evidenced in poor performance in tests involving these concepts. In light of the significant role of variation and evolution in a human's acquaintance with themselves and their environment, there is a need to ensure that students possess meaningful learning of variation and evolution rather than learning by rote. Hence, this study investigated if the use of CTCA as an intervention will enhance students' performance in variation and evolution concepts in biology. The potency of the culturo-techno-contextual approach was tried out in this study with 156 senior secondary two biology students, equivalent to (11th grade) with an average age of 14 years. The research design was a quasi-experimental pre-test, post-test non-equivalent group design. Data on meaningful learning was collected using a 35 multiple-choice item test of Meaningful Learning in Variation and Evolution. The reliability coefficient of the instrument was, however, determined to be 0.77. Data collected were analysed using ANCOVA since the random assignment of subjects was not achieved. The 76 students in the experimental group who were taught variation and evolution using CTCA performed significantly better F(1, 134) = 15.40; p < 0.0001] than their control group counterparts (N = 80). The data from this study support the potency of the culturo-techno-contextual approach in bringing about the learning of variation and evolution in a meaningful manner. In May 2023, herpetologists from six countries converged at the 8th Conference on the Biology of Plethodontid Salamanders in Hammond, Louisiana to share their latest cutting-edge research. The conference was hosted by the Department of Biological Sciences at Southeastern Louisiana University. Dr. Richard Bruce was the honoree. The presentations covered a diverse array of topics from gene expression to behavior to speciation, illustrating the value of plethodontid salamander biology to a range of disciplines. The German-born American Gunther S. Stent (1924–2008) was a refugee from Nazi Germany. He studied in the United States, was a follower of Max Delbrück, and became a member of the phage group. Dispersal is critical to population persistence, colonization and connectivity which are all critical components of invasive success. While individual propensity to disperse varies within populations, the underlying mechanisms promoting individual dispersal remain unclear. Collectively, dispersal is influenced by the environment and individual phenotype. Here we investigated individual dispersal propensity in the Trinidadian guppy, Poecilia reticulata , and related individual variation in dispersal behaviour to phenotype and social/environmental conditions. Namely, we addressed the effect of sex ratio (social/environmental factor) on the tendency for individuals to disperse across physical barriers via jumping behaviour. Jumping is vital for the dispersal of many aquatic species and has been anecdotally linked to the guppy's global invasive success. We found similar jumping behaviour for males and females, with population sex ratio not influencing the magnitude of male or female dispersal. Further, we found consistent among-individual variation in jumping probability; individual differences explained 17.46% and 7.92% of total variation in jumping probability for males and females, respectively. These results strongly indicate that sex ratio does not influence jumping behaviour, suggesting that species invasions are mediated by a nonrandom subset of individuals with greater dispersal tendencies. The standpoint of production makes the relationship between space and time comprehensible. They can either be mental-subjective, physical-objective, or social-intersubjective. Social and intersubjective (or E-series) spacetime might shed new light on biological thinking. Background: While the effectiveness of tutoring‐style videos has been reported in previous studies conducted in laboratories, how these types of videos facilitate students' learning experiences and achievement has not been much explored in prior studies, which prevents discussion on how to design such tutoring‐style videos. Objectives: In this study, we addressed the limitations reported in studies conducted in authentic classes and attempted to explore students' learning patterns and experiences with tutoring‐style videos involving interactions between the instructor and agents. In addition, we attempted to reveal what instructional strategies can be used in tutoring‐style videos to enhance students' engagement and achievement. Methods: The study took place in an undergraduate introductory biology class offered at a university located in the Midwest of the United States. The same instructor taught two sections of this course which lasted for 15 weeks during a Spring semester. The two sections were randomly assigned to an experimental group in which participants watched tutoring‐style videos and a control group with participants watching traditional videos. The experiment was administered in a module lasting two and a half weeks focused on microbiology. Surveys and tests were conducted to determine whether the tutoring‐style videos had a positive impact on student engagement and achievement. In addition, a video content analysis was carried out to elicit insights on how to design effective tutoring‐style videos. Results: For students' emotional engagement and perceived usefulness, no significant difference was found between the two groups, even though the descriptive statistics indicated slightly higher scores from the experimental group than the control group on both subscales. In terms of achievement, the participants reported significantly more learned concepts from watching rich‐interactions in the tutoring‐style videos than the participants in the control group; and they also reported a significantly smaller number of learned concepts from watching one‐interactions in the videos than the control group participants. We also found that instruction in the tutoring‐style videos used effective strategies for facilitating students' participation and cognitive processing. Conclusions: Our findings indicate that tutoring‐style videos can be effective learning materials by allowing students to engage in interactions between instructors and agents in the videos. The more instructional strategies used to facilitate the interactions between the instructor and the agents, the higher engagement can be expected from viewers. Our study would inform the design and development of effective tutoring‐style videos. Aim: Loss‐of‐function mutations in FAM20A result in amelogenesis imperfecta IG (AI1G) or enamel‐renal syndrome, characterized by hypoplastic enamel, ectopic calcification, and gingival hyperplasia, with some cases reporting spontaneous tooth infection. Despite previous reports on the consequence of FAM20A reduction in gingival fibroblasts and transcriptome analyses of AI1G pulp tissues, suggesting its involvement in mineralization and infection, its role in deciduous dental pulp cells (DDP) remains unreported. The aim of this study was to evaluate the properties of DDP obtained from an AI1G patient, providing additional insights into the effects of FAM20A on the mineralization of DDP. Methodology: DDP were obtained from a FAM20A‐AI1G patient (mutant cells) and three healthy individuals. Cellular behaviours were examined using flow cytometry, MTT, attachment and spreading, colony formation, and wound healing assays. Osteogenic induction was applied to DDP, followed by alizarin red S staining to assess their osteogenic differentiation. The expression of FAM20A‐related genes, osteogenic genes, and inflammatory genes was analysed using real‐time PCR, Western blot, and/or immunolocalization. Additionally, STRING analysis was performed to predict potential protein–protein interaction networks. Results: The mutant cells exhibited a significant reduction in FAM20A mRNA and protein levels, as well as proliferation, migration, attachment, and colony formation. However, normal FAM20A subcellular localization was maintained. Additionally, osteogenic/odontogenic genes, OSX, OPN, RUNX2, BSP, and DSPP, were downregulated, along with upregulated ALP. STRING analysis suggested a potential correlation between FAM20A and these osteogenic genes. After osteogenic induction, the mutant cells demonstrated reduced mineral deposition and dysregulated expression of osteogenic genes. Remarkably, FAM20A, FAM20C, RUNX2, OPN, and OSX were significantly upregulated in the mutant cells, whilst ALP, and OCN was downregulated. Furthermore, the mutant cells exhibited a significant increase in inflammatory gene expression, that is, IL‐1β and TGF‐β1, whereas IL‐6 and NFκB1 expression was significantly reduced. Conclusion: The reduction of FAM20A in mutant DDP is associated with various cellular deficiencies, including delayed proliferation, attachment, spreading, and migration as well as altered osteogenic and inflammatory responses. Microplastics (plastic particles <5 mm) permeate aquatic and terrestrial ecosystems and constitute a hazard to animal life. Although much research has been conducted on the effects of microplastics on marine and benthic organisms, less consideration has been given to insects, especially those adapted to urban environments. Here, we provide a perspective on the potential consequences of exposure to microplastics within typical larval habitat on mosquito biology. Mosquitoes represent an ideal organism in which to explore the biological effects of microplastics on terrestrial insects, not least because of their importance as an infectious disease vector. Drawing on evidence from other organisms and knowledge of the mosquito life cycle, we summarise some of the more plausible impacts of microplastics including physiological, ecotoxicological and immunological responses. We conclude that although there remains little experimental evidence demonstrating any adverse effect on mosquito biology or pathogen transmission, significant knowledge gaps remain, and there is now a need to quantify the effects that microplastic pollution could have on such an important disease vector. Turnover—constant component production and destruction—is ubiquitous in biology. Turnover occurs across organisms and scales, including for RNAs, proteins, membranes, macromolecular structures, organelles, cells, hair, feathers, nails, antlers, and teeth. For many systems, turnover might seem wasteful when degraded components are often fully functional. Some components turn over with shockingly high rates and others do not turn over at all, further making this process enigmatic. However, turnover can address fundamental problems by yielding powerful properties, including regeneration, rapid repair onset, clearance of unpredictable damage and errors, maintenance of low constitutive levels of disrepair, prevention of stable hazards, and transitions. I argue that trade-offs between turnover benefits and metabolic costs, combined with constraints on turnover, determine its presence and rates across distinct contexts. I suggest that the limits of turnover help explain aging and that turnover properties and the basis for its levels underlie this fundamental component of life. Advancements in cell culturing techniques have allowed the development of three-dimensional (3D) cell culture models sourced directly from patients' tissues and tumors, faithfully replicating the native tissue environment. These models provide a more clinically relevant platform for studying disease progression and treatment responses compared to traditional two-dimensional (2D) models. Patient-derived organoids (PDOs) and patient-derived xenograft organoids (PDXOs) emerge as innovative 3D cancer models capable of accurately mimicking the tumor's unique features, enhancing our understanding of tumor complexities, and predicting clinical outcomes. Triple-negative breast cancer (TNBC) poses significant clinical challenges due to its aggressive nature, propensity for early metastasis, and limited treatment options. TNBC PDOs and PDXOs have significantly contributed to the comprehension of TNBC, providing novel insights into its underlying mechanism and identifying potential therapeutic targets. This review explores the transformative role of various 3D cancer models in elucidating TNBC pathogenesis and guiding novel therapeutic strategies. It also provides an overview of diverse 3D cell culture models, derived from cell lines and tumors, highlighting their advantages and culturing challenges. Finally, it delves into live-cell imaging techniques, endpoint assays, and alternative cell culture media and methodologies, such as scaffold-free and scaffold-based systems, essential for advancing 3D cancer model research and development. Electron microscopy (EM), in its various flavors, has significantly contributed to our understanding of lipid droplets (LD) as central organelles in cellular metabolism. For example, EM has illuminated that LDs, in contrast to all other cellular organelles, are uniquely enclosed by a single phospholipid monolayer, revealed the architecture of LD contact sites with different organelles, and provided near‐atomic resolution maps of key enzymes that regulate neutral lipid biosynthesis and LD biogenesis. In this review, we first provide a brief history of pivotal findings in LD biology unveiled through the lens of an electron microscope. We describe the main EM techniques used in the context of LD research and discuss their current capabilities and limitations, thereby providing a foundation for utilizing suitable EM methodology to address LD‐related questions with sufficient level of structural preservation, detail, and resolution. This document is an editorial from the International Journal of Molecular Sciences titled "Role of Phosphodiesterases in Biology and Pathology 2.0." It discusses the role of phosphodiesterases (PDEs) in the hydrolysis of cAMP and cGMP second messengers in cells. PDEs are involved in various physiological and pathological conditions, and their inhibitors have been used in the treatment of impotence and chronic pulmonary diseases. The special issue of the journal includes contributions that provide new insights into the role of PDEs in different conditions such as spinal cord injury, multiple sclerosis, skeletal muscle function, cardiac contractility, portal hypertension, liver cirrhosis, and cancer. Maintaining epidermal homeostasis relies on a tightly organized process of proliferation and differentiation of keratinocytes. While past studies have primarily focused on calcium regulation in keratinocyte differentiation, recent research has shed light on the crucial role of lysosome dysfunction in this process. TLR adaptor interacting with SLC15A4 on the lysosome (TASL) plays a role in regulating pH within the endo-lysosome. However, the specific role of TASL in keratinocyte differentiation and its potential impact on proliferation remains elusive. In our study, we discovered that TASL deficiency hinders the proliferation and migration of keratinocytes by inducing G1/S cell cycle arrest. Also, TASL deficiency disrupts proper differentiation process in TASL knockout human keratinocyte cell line (HaCaT) by affecting lysosomal function. Additionally, our research into calcium-induced differentiation showed that TASL deficiency affects calcium modulation, which is essential for keratinocyte regulation. These findings unveil a novel role of TASL in the proliferation and differentiation of keratinocytes, providing new insights into the intricate regulatory mechanisms of keratinocyte biology. This case study examines how material resistance (limitations posed by the physical world) and graph interpretation intersected during a high school biology investigation using digital sensors. We use an extended episode from a small group to illustrate how, in an inquiry-based unit, measuring near the resolution limit of a sensor caused scaling issues in graphs. Qualitative videotape analysis focuses on both the students' attempts to make sense of a perceived lack of variation in the collected data and the teacher's and classroom researchers' misinterpretation of the students' difficulties with graph interpretation. We suggest that these educators, though experienced, could have benefited from additional strategies to help them recognise and respond to graph interpretation issues introduced by digital representations of real-world data, and that the students could have benefited from explicit prompts to discuss the limitations of their equipment. We describe several implications for researchers, teachers, and curriculum developers interested in implementing inquiry-based biology investigations using sensor data. We argue that students should be supported to recognise that encountering unexpected results from their investigations and working to understand what these results have to say about the real world is an important and valid part of the practice of real science. This essay brings forward the idea that there is more than meets the eye in local knowledge systems than what science can show us now. To comprehend this, we need to make a conceptual jump and look for the "dark matter" (the notion borrowed from astronomy that refers to a hypothetical form of matter that does not interact with light or electromagnetic fields) that can potentially sustain local knowledge. Considering that it is a complex of knowledge, practices, and beliefs contained in TEK, knowledge in LEK does not correspond to the notion of knowledge in science. Therefore, in order to map LEK–science interactions, we will refer to the concept of peoples' knowledge of LEK as acknowledgement and the scientific recognition and awareness of information, facts, and principles as knowledge. Applying this to a Johari Window, we can observe four categories of LEK in a known–unknown/acknowledged–unacknowledged matrix. We can refer to unknown and unacknowledged as dark local knowledge. Indeed, local knowledge systems contain many aspects that modern science cannot yet explain, as a major part of its components are not even considered in scholarly research. In chronic lymphocytic leukaemia (CLL), comorbidities assessed by the CLL comorbidity index (CLL-CI) have been associated with outcomes in Western cohorts. We conducted a retrospective analysis of an unselected Middle Eastern cohort of newly diagnosed CLL patients seen at the Kuwait Cancer Control Center (n = 300). Compared to Western studies, these Middle Eastern patients were diagnosed at a younger age (median of 59) and had a higher comorbidity burden (69% non-low risk CLL-CI). A higher CLL-CI score was independently associated with significantly shorter event-free survival and greater risk of death. Our analysis demonstrates that CLL-CI is a valuable tool for comorbidity assessment and prognostic influence in (relatively young) Middle Eastern CLL patients. [ABSTRACT FROM AUTHOR]

It leads to localized damage to the appearance of the skin, accompanied by symptoms such as itching and pain, which reduces the quality of life of the patient and causes serious medical burdens. With the continuous development of economy and society, there is an increasing demand for beauty. People are looking forward to a safer and more effective method to eliminate pathological scarring. In recent years, adipose-derived stem cells (ADSCs) have received increasing attention from researchers. It can effectively improve pathological scarring by mediating inflammation, regulating fibroblast proliferation and activation, and vascular reconstruction. This review focuses on the pathophysiological mechanisms of hypertrophic scarring, summarizing the therapeutic effects of in vitro, in vivo, and clinical studies on the therapeutic effects of ADSCs in the field of hypertrophic scarring prevention and treatment, the latest application techniques, such as cell-free therapies utilizing ADSCs, and discussing the advantages and limitations of ADSCs. Through this review, we hope to further understand the characterization of ADSC and clarify the effectiveness of its application in hypertrophic scarring treatment, so as to provide clinical guidance. Cancer is a progressive disease that can develop and evolve over decades, with inflammation playing a central role at each of its stages, from tumor initiation to metastasis. In this context, macrophages represent well-established bridges reciprocally linking inflammation and cancer via an array of diverse functions that have spurred efforts to classify them into subtypes. Here, we discuss the intertwines between macrophages, inflammation, and cancer with an emphasis on temporal dynamics of macrophage diversity and functions in pre-malignancy and cancer. By instilling temporal dynamism into the more static classic view of tumor-associated macrophage biology, we propose a new framework to better contextualize their significance in the inflammatory processes that precede and result from the onset of cancer and shape its evolution. Tumor-associated macrophages (TAMs) are well known to fuel tumor evolution. Blériot et al. advocate for integrating the time dimension when conceptualizing macrophage diversity in pre-malignancy and cancer and propose the term "PreTAMs" to emphasize their early roles in orchestrating inflammatory responses that often precede, and not just result from, tumor onset. Background: Exportin‐1 (XPO1), a crucial protein regulating nuclear‐cytoplasmic transport, is frequently overexpressed in various cancers, driving tumor progression and drug resistance. This makes XPO1 an attractive therapeutic target. Over the past few decades, the number of available nuclear export‐selective inhibitors has been increasing. Only KPT‐330 (selinexor) has been successfully used for treating haematological malignancies, and KPT‐8602 (eltanexor) has been used for treating haematologic tumours in clinical trials. However, the use of nuclear export‐selective inhibitors for the inhibition of XPO1 expression has yet to be thoroughly investigated in clinical studies and therapeutic outcomes for solid tumours. Methods: We collected numerous literatures to explain the efficacy of XPO1 Inhibitors in preclinical and clinical studies of a wide range of solid tumours. Results: In this review, we focus on the nuclear export function of XPO1 and results from clinical trials of its inhibitors in solid malignant tumours. We summarized the mechanism of action and therapeutic potential of XPO1 inhibitors, as well as adverse effects and response biomarkers. Conclusion: XPO1 inhibition has emerged as a promising therapeutic strategy in the fight against cancer, offering a novel approach to targeting tumorigenic processes and overcoming drug resistance. SINE compounds have demonstrated efficacy in a wide range of solid tumours, and ongoing research is focused on optimizing their use, identifying response biomarkers, and developing effective combination therapies. Pope Leo XIII's publication of Aeterni Patris (1879) was a major factor in the great revival of Thomistic thought in the late 19th and the first half of the 20th centuries. Among the authors that took up the challenge implicit in the Pope's document of bringing Aquinas and his thought into the intellectual debates of the times we find two interesting proposals. The first is that of Juan González Arintero, a Spanish Dominican, and the second one is that of Josef Donat, a Jesuit born and raised in the Austrian Empire. Arintero is mostly known in Catholic circles for his influential works on mysticism, but in fact he devoted much of his early work to the subject of evolution, and how it could interact with the Catholic faith in general, and with Thomism in particular. Donat is the author of a Summa Philosophiae Christianae, a collection that was widely read in Catholic seminaries well into the 20th century. In this paper we will focus on the differing ways in which these authors tackled the problems and questions presented by Darwinian evolutionism to the post-Aeterni Patris Thomism. Reproductive biologies (spawning period, length at maturity and fecundity) of the Knysna sand goby Psammogobius knysnaensis and the Prison goby Caffrogobius gilchristi were investigated between September 2018 and August 2019 in the warm-temperate Sundays Estuary, South Africa. The two species reproduced during the same period (spring and summer) with peak reproduction occurring in January. P. knysnaensis egg production was higher than C. gilchristi and was not affected by fish size. Although C. gilchristi egg production increased with fish size, this was not significant. P. knysnaensis reached LM50 at smaller sizes than C. gilchristi. Reproductive periods were likely coupled with prey availability, as phytoplankton and zooplankton abundances are highest in warm-temperate estuaries during this period. The ability of these gobies to reproduce at small sizes (< 40 mm TL), which is an adaptive trait of many r-selected species, underpins their high abundance in South African estuaries as they are well adapted to stochastic environments. The vision behind the Next Generation Science Standards (NGSS) is that all students engage in explaining phenomena and designing solutions with three-dimensional learning. Authentic sensemaking involves opportunities for learners to share, analyze, and critique ideas in collaborative groups. However, it can be challenging to structure classroom discourse so that all students actively participate. This article outlines the success of two teachers using Group Learning Routines (GLR), or peer-to-peer discussion protocols, to support diverse learners. The routines are embedded in the New Visions for Public Schools Biology Curriculum which is open source and storyline-based. Embedded routines structure student talk throughout a learning sequence culminating in class consensus discussion, in which students collaboratively decide on a class-wide explanation of the phenomenon. This document is an editorial from the International Journal of Molecular Sciences titled "Chemistry and Biology of Noncanonical Nucleic Acid Structures: From Physicochemical Properties to Therapeutic Applications." The editorial aims to highlight the chemistry and biology of noncanonical nucleic acid structures, focusing on their structure, stability, and interactions with proteins and ligands. Noncanonical structures, such as triplex, G-quadruplex, and i-motif species, can form in important genomic regions and play key roles in biological processes, including oncogene expression and telomere maintenance. The investigations published in this special issue cover various aspects, including the potential role of noncanonical structures in transcription regulation, the selective targeting of cancer-related G-quadruplex structures, and the development of innovative analytical methods for the detection of viral RNA. Frozen shoulder (FS) is a common disorder characterized by spontaneous onset of shoulder pain accompanied by progressive loss of range-of-motions. The cause of FS is still unclear, and radical therapy has not been established. With the final aim of preventing or curing FS at an earlier stage, we reviewed the pathological and biological features of this disease. Many studies indicate that the main pathology of FS is inflammation initially and fibrosis later. There are inflammatory cytokines, immune cells, fibrotic growth factors, and type-III collagen in the synovium and the joint capsule. The immune cell landscape switches from the macrophages to T cells. Activated fibroblasts seem to regulate the inflammatory and fibrotic processes. The imbalance between matrix metalloproteinases and tissue inhibitors of metalloproteases might promote fibrosis. Additionally, advanced glycation end-products are noted in the FS synovium. Diabetes mellitus and hypothyroidism are closely related to the development of FS. In terms of nonsurgical treatment, oral or intra-articular glucocorticoids are the only drugs that provide early benefit. Some other anti-inflammatory or antifibrotic drugs may potentially control the FS, but have not been proven effective in the clinical setting. Smartphones are used universally in many areas due to their advanced features. Smartphones are also widely used by teachers and students in education. This research aims to design low-cost and easy experiments for biology lessons using smartphones. Colorimetric measurement was made with a smartphone. Mobile applications were used to read color change values. The amount of sugar in beverages was calculated with the students. They could create standard curves, measure with a smartphone, organize the data, and determine sugar concentration. It was determined that the students had positive thoughts about the activity. Self-confidence results indicated that the activity had a positive effect on students' self-confidence. Although there was a decrease in the mean scores of difficulty, it was revealed that the mean scores were over the "moderate" level. As a result, it can be said that the activity may be easily practiced by teachers and students in biology lessons. [ABSTRACT FROM AUTHOR]

Online learning is a well-established and growing, yet controversial and widely criticized, component of higher education. In this study, in-person and online versions of an undergraduate introductory biology class were compared to determine whether the online version of the course promoted student performance as well as the in-person course. Student survey responses were also analyzed to identify which teaching modality students perceived to be most effective at promoting learning, and to determine which components of the course were perceived by students to be most effective. No statistically significant difference was observed in exam scores or course grades between the two course modalities. Interestingly, some course components, such as completing virtual labs, were perceived to be more effective in the online version of the course, while others, such as participating in class discussions, were ranked as more effective for the in-person course offering; other components were ranked as similarly effective in both versions of the course. Therefore, this study provides evidence that online education can be effective if appropriately designed and implemented, and it provides a framework for strengthening the effectiveness of online education in the future. The emerging field of genomic medicine offers an opportunity for biology and anatomy teachers to bring the topics of DNA, genetics, molecular processes, and evolution together into one experience. Through the genomic medicine paradigm, students see the unbroken connection between small biological topics such as mutations and their potential connection to disease phenotypes. In this paper, we present as a main example cystic fibrosis, which is an often-studied genetic disease in general biology class, for examination through the genomic medicine lens. Concepts such as genes, the plasma membrane, variation, mutations, the nucleus, and chromosomes can be used in a narrative and visual approach to genetics through the genomic medicine standpoint to engage and connect students with next-generation genomics and with the fundamental unit of life—the cell. The Pacific sleeper shark Somniosus pacificus is a large-bodied and broad-ranging squaliform shark that occupies diverse habitats throughout the Pacific Ocean. Despite its large size and occurrence as bycatch in various commercial fisheries, little is known about even the most basic aspects of its biology and ecology. Observed declines in certain parts of its range, coupled with life history characteristics associated with low productivity, have led to conservation concerns for this cryptic but charismatic species. Here, we provide a comprehensive review of the current state of knowledge regarding the distribution, diet, life history, and other aspects of the Pacific sleeper shark and present updated fisheries and survey data for the eastern North Pacific Ocean. The most pressing research gaps identified during the course of this review concern habitat use at different life stages and basic life history information. While work is currently in progress to expand our base of knowledge for this species, we recommend a precautionary approach to management until sufficient information becomes available to ensure its conservation. The field of evolutionary biology must bridge the gap between its diversity, equity, and inclusion (DEI) commitments and data-driven educational actions in the nation's undergraduate classrooms and degree programs. In this article, we discuss the urgent need for the adoption of equity frameworks and why they are centrally important to data-driven DEI efforts in evolutionary biology. We describe why equity indicators (e.g. measures) must be anchored in and aligned with equity frameworks. We introduce a specific equity framework for learning (the enhanced educational debt framework) and illustrate how it may be leveraged to document, interpret, and improve outcomes in evolutionary biology. We apply the equity framework and associated indicators to >3,500 students' first college-level experience with evolutionary biology at a public, 4-year institution in the Northeastern United States to demonstrate how these conceptual tools and empirical perspectives may be used by faculty, departments, and degree programs to better understand their roles in mitigating or perpetuating inequities. We end by discussing how this framework may be applied to a range of evolution concepts and courses in the educational hierarchy and used to help evolutionary biologists better understand the extent to which a core aspect of SSE's diversity statement is being realized. Floral features, the pollination process, the breeding system, and fruiting success are documented in the Andean terrestrial orchid Gomphichis valida. Studies were performed at the Department of Ancash, Peru, at ca. 4250 m a.s.l., from June to November 2022. The breeding system was studied through controlled pollinations in plants excluded from pollinators (bagged). Natural pollination was studied in the field and through photos and videos. The flowers are nectariferous and non‐protandrous and present the longest lifespan reported so far for a neotropical terrestrial orchid (53–80 days). This species is pollinator dependent (unable to set fruit and seed without the agency of pollinators) and self‐compatible. The observed pollinators belong to three functional groups that appeared successively (in order of appearance): hummingbirds (Chalcostigma stanleyi), bumblebees (Bombus coccineus and Bombus rubicundus), and solitary Halictidae bees (Augochlorella sp.). All recorded pollinators probe the flowers for nectar. The pollinaria adhere to the bee mouthparts and onto the lateral side of the hummingbird bill. Bumblebees were the most frequent and efficient functional group (up to 62.07% of the observed pollination events), followed by Halictidae (31.03%) and hummingbirds (6.90%). Natural fruiting success is high (66.66%–94.49%). Such high natural fruiting success is unexpected at high altitudes, but it is likely explained by a combination of factors, including the long floral lifespan, nectar secretion, and self‐compatibility. This is the first report of hummingbirds as pollinators of Cranichidinae orchids, and G. valida is, to the best of our knowledge, a neotropical orchidoid orchid with the most generalist pollination biology recorded so far. The genus Dyckia (Bromeliaceae) is endemic to South America, and its species show important ecological roles in xeric environments. The flowering phenology as well as floral and reproductive biology were monitored monthly in Dyckia dissitiflora at two sites (Bela Vista: 11°42′03.4″ S 39°05′21.7″ W and Barra do Vento: 11°37′58.1″ S 39°03′33.7″ W) in the municipality of Serrinha, Bahia, Brazil. Flowering occurs in the early austral summer with a peculiar time of beginning of anthesis (10:00–11:00), offering resources 18 h after the first petal movements. Pollen viability was 98%, with a high production of pollen grains. Stigma receptivity is continuous from flower opening to senescence, and nectar production initiates at 04:00 and extends until 18:00. The species is self‐compatible and autogamous, but not agamospermic. Pollen tubes developed and reached the micropyle within 24 h in all of the reproductive tests. The hummingbird Chlorostilbon lucidus was the only floral visitor and pollinator (frequency = 0.82 ± 0.02). Fruiting occurs 3 days after anthesis, and the seeds are wind‐dispersed. Self‐compatibility and autogamy are essential strategies for the reproductive success of D. dissitiflora in light of the low number and visit frequency of its pollinators, and it is favored by slow floral movements and the position of the stigmas in relation to stamens. Pre-proenkephalin 1 (Penk1) is a pro-neuropeptide that belongs to the typical opioid peptide's family, having analgesic properties. We previously found Penk1 to be the most downregulated gene in a whole gene profiling analysis performed in osteoblasts subjected to microgravity as a model of mechanical unloading. In this work, Penk1 downregulation was confirmed in the bones of two in vivo models of mechanical unloading: tail-suspended and botulinum toxin A (botox)-injected mice. Consistently, in the sera from healthy volunteers subjected to bed rest, we observed an inverse correlation between PENK1 and bed rest duration. These results prompted us to investigate a role for this factor in bone. Penk1 was highly expressed in mouse bone, but its global deletion failed to impact bone metabolism in vivo. Indeed, Penk1 knock out (Penk1−/−) mice did not show an overt bone phenotype compared to the WT littermates. Conversely, in vitro Penk1 gene expression progressively increased during osteoblast differentiation and its transient silencing in mature osteoblasts by siRNAs upregulated the transcription of the Sost1 gene encoding sclerostin, and decreased Wnt3a and Col1a1 mRNAs, suggesting an altered osteoblast activity due to an impairment of the Wnt pathway. In line with this, osteoblasts treated with the Penk1 encoded peptide, Met-enkephalin, showed an increase of Osx and Col1a1 mRNAs and enhanced nodule mineralization. Interestingly, primary osteoblasts isolated from Penk1−/− mice showed lower metabolic activity, ALP activity, and nodule mineralization, as well as a lower number of CFU-F compared to osteoblasts isolated from WT mice, suggesting that, unlike the transient inhibition, the chronic Penk1 deletion affects both osteoblast differentiation and activity. Taken together, these results highlight a role for Penk1 in the regulation of the response of the bone to mechanical unloading, potentially acting on osteoblast differentiation and activity in a cell-autonomous manner. Copper plays vital roles in numerous cellular processes and its imbalance can lead to oxidative stress and dysfunction. Recent research has unveiled a unique form of copper-induced cell death, termed cuproptosis, which differs from known cell death mechanisms. This process involves the interaction of copper with lipoylated tricarboxylic acid cycle enzymes, causing protein aggregation and cell death. Recently, a growing number of studies have explored the link between cuproptosis and cancer development. This review comprehensively examines the systemic and cellular metabolism of copper, including tumor-related signaling pathways influenced by copper. It delves into the discovery and mechanisms of cuproptosis and its connection to various cancers. Additionally, the review suggests potential cancer treatments using copper ionophores that induce cuproptosis, in combination with small molecule drugs, for precision therapy in specific cancer types. The association of type 2 diabetes with certain cancer risk has been of great interest for years. However, the effect of diabetic medications on cancer development is not fully understood. Prospective clinical trials have not elucidated the long‐term influence of hypoglycemic drugs on cancer incidence and the safety for cancer‐bearing patients with diabetes, whereas numerous preclinical studies have shown that antidiabetic drugs could have an impact on carcinogenesis processes beyond the glycemic control effect. Because there is no evidence of the safety profile of antidiabetic agents on cancer biology, careful consideration would be required when prescribing any medicines to patients with diabetes and existing tumor. Diabetic retinopathy (DR) is recognized as a neurovascular complication of diabetes, and emerging evidence underscores the pivotal role of inflammation in its pathophysiology. Macrophage activation is increasingly acknowledged as a key contributor to the onset and progression of DR. Different populations of macrophages originating from distinct sources contribute to DR-associated inflammation. Retinal macrophages can be broadly categorized into two main groups based on their origin: intrinsic macrophages situated within the retina and vitreoretinal interface and macrophages derived from infiltrating monocytes. The former comprises microglia (MG), perivascular macrophages, and macrophage-like hyalocytes. Retinal MG, as the principal population of tissue-resident population of mononuclear phagocytes, exhibits high heterogeneity and plasticity while serving as a crucial connector between retinal capillaries and synapses. This makes MG actively involved in the pathological processes across various stages of DR. Activated hyalocytes also contribute to the pathological progression of advanced DR. Additionally, recruited monocytes, displaying rapid turnover in circulation, augment the population of retinal macrophages during DR pathogenesis, exerting pathogenic or protective effect based on different subtypes. In this review, we examine novel perspectives on macrophage biology based on recent studies elucidating the diversity of macrophage identity and function, as well as the mechanisms influencing macrophage behavior. Joiner and colleagues' Interpersonal Theory of Suicide (IPTS), a prominent "desire-capability" model of suicide—based on the common-sense idea that people take their own lives because they want to, and can—is critiqued from a biological perspective. Tinbergen's ethological "four questions" guide the analysis: evolution, survival value, ontogeny, and proximate causation, each addressing a different aspect of biological understanding. Problems for IPTS emerge with all four. As a parsimonious solution, the desire-capability hypothesis is reconceived as an ultimate, instead of proximate, mode of explanation. By this light, desire and capability for suicide combined in our species' ancestral past, thus making suicide a recurrent survival threat, and driving the evolution of special-purpose defensive adaptations. This stance tallies with the pain-brain theory of the evolution of suicide, and with Joiner and colleagues' own investigation into organismic anti-suicide defenses, which appears to conflict conceptually with IPTS. These defenses' evolved algorithm may make suicide an intrinsically aleatory phenomenon, opaque to usefully accurate prediction. Positive implications for prevention and research are proposed. The objective of the current review is to summarize the current state of optical methods in redox biology. It consists of two parts, the first is dedicated to genetically encoded fluorescent indicators and the second to Raman spectroscopy. In the first part, we provide a detailed classification of the currently available redox biosensors based on their target analytes. We thoroughly discuss the main architecture types of these proteins, the underlying engineering strategies for their development, the biochemical properties of existing tools and their advantages and disadvantages from a practical point of view. Particular attention is paid to fluorescence lifetime imaging microscopy as a possible readout technique, since it is less prone to certain artifacts than traditional intensiometric measurements. In the second part, the characteristic Raman peaks of the most important redox intermediates are listed, and examples of how this knowledge can be implemented in biological studies are given. This part covers such fields as estimation of the redox states and concentrations of Fe–S clusters, cytochromes, other heme-containing proteins, oxidative derivatives of thiols, lipids, and nucleotides. Finally, we touch on the issue of multiparameter imaging, in which biosensors are combined with other visualization methods for simultaneous assessment of several cellular parameters. Chordae tendineae, referred to as heart tendinous cords, act as tendons connecting the papillary muscles to the valves in the heart. Their role is analogous to tendons in the musculoskeletal system. Despite being exposed to millions of cyclic tensile stretches over a human's lifetime, chordae tendineae rarely suffer from overuse injuries. On the other hand, musculoskeletal tendinopathy is very common and remains challenging in clinical treatment. The objective of this study was to investigate the mechanism behind the remarkable durability and resistance to overuse injuries of chordae tendineae, as well as to explore their effects on flexor tenocyte biology. The messenger RNA expression profiles of chordae tendineae were analyzed using RNA sequencing and verified by quantitative reverse transcription polymerase chain reaction and immunohistochemistry. Interestingly, we found that periostin (Postn) and fibroblast growth factor 7 (FGF7) were expressed at significantly higher levels in chordae tendineae, compared to flexor tendons. We further treated flexor tenocytes in vitro with periostin and FGF7 to examine their effects on the proliferation, migration, apoptosis, and tendon‐related gene expression of flexor tenocytes. The results displayed enhanced cell proliferation ability at an early stage and an antiapoptotic effect on tenocytes, while treated with periostin and/or FGF7 proteins. Furthermore, there was a trend of promoted tenocyte migration capability. These findings indicated that Postn and FGF7 may represent novel cytokines to target flexor tendon healing. Clinical significance: The preliminary discovery leads to a novel idea for treating tendinopathy in the musculoskeletal system using specific molecules identified from chordae tendineae. Cydalima perspectalis (Walker 1859) (Lepidoptera: Crambidae) is a destructive pest that has recently been reported from the ancient Hyrcanian forests feeding on leaves and shoots of Buxus hyrcana Pojarkova.To gain a deeper insight into the biology and population dynamics of C. perspectalis, a laboratory population was studied under controlled conditions. The moth was reared on B. hyrcana at 25 ± 1°C, 65 ± 10% RH and 16 h:8 h (L:D), and the data obtained were used to build a life table, adjusted to allow for the evaluation of the stage duration, for both males and females.The mean adult survival was 11.94 ± 0.26 days, the pre‐oviposition period lasted, on average, for 6.71 ± 0.18 days and the mean fecundity attained was 58.19 ± 7.81 eggs/female. Newly emerged females had a life expectancy (exj) of 14 days and males 19 days.The intrinsic rate of increase for the population was 0.05 ± 0.00 days−1, and the net reproductive rate was 24.05 ± 3.95 offspring/female. The biological and ecological data obtained allowed for the estimation of C. perspectalis biotic potential, thus contributing to the development of sustainable control methods for this pest and, consequently, to a reduction of the damage caused to box trees. The relationship between the immune cell and tumor occurrence and progression remains unclear. Profiling alterations in the tumor immune microenvironment (TIME) at high resolution is crucial to identify factors influencing cancer progression and enhance the effectiveness of immunotherapy. However, traditional sequencing methods, including bulk RNA sequencing, exhibit varying degrees of masking the cellular heterogeneity and immunophenotypic changes observed in early and late-stage tumors. Single-cell RNA sequencing (scRNA-seq) has provided significant and precise TIME landscapes. Consequently, this review has highlighted TIME cellular and molecular changes in tumorigenesis and progression elucidated through recent scRNA-seq studies. Specifically, we have summarized the cellular heterogeneity of TIME at different stages, including early, late, and metastatic stages. Moreover, we have outlined the related variations that may promote tumor occurrence and metastasis in the single-cell era. The germ theory states that pathogenic microorganisms are responsible for causing infectious diseases. The theory is inherently microbe-centric and does not account for variability in disease severity among individuals and asymptomatic carriership--two phenomena indicating an important role for host variability in infection outcome. The basic tenet of the germ theory was recently challenged, and a radically host-centric paradigm referred to as the "full-blown host theory" was proposed. According to this view, the pathogen is reduced to a passive environmental trigger, and the development of disease is instead due to pre-existing immunodeficiencies of the host. Here, we consider the factors that determine disease severity using established knowledge concerning evolutionary biology, microbial pathogenesis, and host-pathogen interactions. We note that the available data support a noncentric view that recognizes key roles for both the causative microbe and the host in dictating infection outcome. [ABSTRACT FROM AUTHOR]

Cell state transitions are prevalent in biology, playing a fundamental role in development, homeostasis and repair. Dysregulation of cell state transitions can lead to or occur in a wide range of diseases. In this letter, I explore and highlight the role of post-transcriptional regulatory mechanisms in determining the dynamics of cell state transitions. I propose that regulation of protein levels after transcription provides an under-appreciated regulatory route to obtain fast and sharp transitions between distinct cell states.
