- Abstract
Using polymeric fiber substrates with different mechanical features and surface topographies (fiber alignments) as microenvironment of cell culture substrates that influences cancer progression and metastatic potential, we have demonstrated systematically the effect of stiffness and topographies on two different types of breast cancer cells (MDA-MB-231 and MCF-7). The cells cultured on different substrates both normoxic and hypoxic conditions showed different morphologies, and these morphological differences were correlated with cellular motility and gene expression. We discussed the effect of hypoxia on gene expression and cellular motility to understand the cell–cell and cell–substrate interactions. The progresses of microenvironment-mediated tumor progression in artificial extracellular matrix explored the design criteria to understand the cancer progression mechanism and metastatic potential.

A 1.8 V 2-Mb SPRAM (SPin-transfer torque RAM) chip using 0.2-μm logic process with MgO tunneling barrier cell is reviewed, which demonstrates the circuit technologies for potential low power non-volatile RAM, or universal memory. This chip features: an array scheme with bit-by-bit bi-directional current write to achieve proper spin-transfer torque writing of 100-ns, and parallelizing-direction current reading with low voltage bit-line that leads to 40-ns access time. 4. Nitric oxide stimulates glucose transport through insulin-independent GLUT4 translocation in 3T3-L1 adipocytes

- Abstract
OBJECTIVE: It is well known that nitric oxide synthase (NOS) is expressed and that it modulates glucose transport in skeletal muscles. Recent studies have shown that adipose tIssues also express inducible and endothelial nitric oxide synthase (eNOS). In the present study, we investigated whether nitric oxide (NO) induces glucose uptake in adipocytes, and the signaling pathway involved in the NO-stimulated glucose uptake in 3T3-L1 adipocytes. METHODS: First, we determined the expression of eNOS in 3T3-L1 adipocytes, and then these cells were treated with the NO donor sodium nitroprusside (SNP) and/or insulin, and glucose uptake and phosphorylation of insulin receptor substrate (IRS)-1 and Akt were evaluated. Moreover, we examined the effects of a NO scavenger, a guanylate cyclase inhibitor or dexamethasone on SNP-stimulated glucose uptake and GLUT4 translocation. RESULTS: SNP at a concentration of 50 mmol/l increased 2-deoxyglucose uptake (1.8-fold) without phosphorylation of IRS-1 and Akt. Treatment with the NO scavenger or guanylate cyclase inhibitor decreased SNP-stimulated glucose uptake to the basal level. Dexamethasone reduced both insulin- and SNP-stimulated glucose uptake with impairment of GLUT4 translocation. CONCLUSION: NO is capable of stimulating glucose transport through GLUT4 translocation in 3T3-L1 adipocytes, via a mechanism different from the insulin signaling pathway.

5. Acute hyperinsulinemia is associated with increased circulating levels of adrenomedullin in patients with type 2 diabetes mellitus

- Abstract
OBJECTIVE: To investigate the effect of acute hyperinsulinemia on the plasma levels of adrenomedullin (AM) in patients with type 2 diabetes mellitus. DESIGN: We measured the plasma levels of AM in 18 patients with type 2 diabetes mellitus and in 19 normal subjects before and during a euglycemic hyperinsulinemic clamp study (the goal was for blood sugar levels of 5.24 mmol/l and insulin levels of 1200 pmol/l). Both plasma AM and serum insulin were measured by immunoradiometric assays. RESULTS: Before the glucose clamp study there was no significant difference in the plasma levels of AM between patients with type 2 diabetes mellitus and normal subjects. During the glucose clamp study, the serum levels of insulin significantly increased (from 33.0+/-3.6 to 1344.6+/-67.8 pmol/ml, P<0.001), as did the plasma levels of AM (from 12.8+/-0.7 to 14.2+/-0.9 fmol/ml, P<0.03) only in patients with type 2 diabetes mellitus. There was a significant correlation between the change in circulating levels of insulin and AM (r=0.755, P<0.01). CONCLUSIONS: Acute hyperinsulinemia induced a significant increase in the plasma levels of AM in patients with type 2 diabetes mellitus. Increased insulin may regulate circulating levels of AM in patients with type 2 diabetes mellitus.

7. A novel CLCN1mutation: P480T in a Japanese family with Thomsen's myotonia congenita

- Abstract
At least 50 disease‐causing mutations in the skeletal muscle voltage‐gated chloride channel gene (CLCN1), almost all of which originate from Caucasian families, have been identified. We investigated a Japanese family with Thomsen's myotonia congenita that included 16 affected individuals (8 men and 8 women) through five generations. Polymerase chain reaction (PCR)–single‐strand conformation polymorphism (SSCP) screening of 11 members showed an aberrant conformer in exon 13 of CLCN1complementary DNA (cDNA) in 8 affected and 1 unaffected members. By sequence analysis, we identified a C‐to‐A transition at nucleotide position 1438, resulting in a substitution of proline for threonine at amino acid position 480 (P480T), the same position of the original mutation (P480L) in Thomsen's disease. The P480T mutation was novel and absent in 100 normal controls. Seven of the 8 affected individuals were heterozygous; another, from affected parents, was homozygous. Clinically, myotonia in the homozygous patient was more severe than that in heterozygous patients, probably due to the gene dosage effect. On a long‐train nerve‐stimulation test at a rate of 3 Hz, M‐wave responses in the homozygous patient showed marked decrement followed by recovery. In contrast, the heterozygous patients showed just a slight decrement or no changes, and none of 2 patients with myotonic muscular dystrophy or 2 normal controls revealed any decrement. Thus, the long‐train nerve‐stimulation test at a low stimulus frequency may be a useful tool to assess the disease‐severity/genotype relationship in myotonia congenita. © 2001 John Wiley & Sons, Inc. 9. Universal Lax Pair for Generalised Calogero–Moser Models

- Abstract
AbstractIn this talk we introduce generalised Calogero–Moser models and demonstrate their integrability by constructing universal Lax pair operators. These include models based on non-crystallographic root systems, that is the root systems of the finite reflection groups, H3, H4, and the dihedral group I2(m), besides the well-known ones based on crystallographic root systems, namely those associated with Lie algebras. Universal Lax pair operators for all of the generalised Calogero–Moser models and for any choices of the potentials are linear combinations of the reflection operators. The equivalence of the Lax pair with the equations of motion is proved by decomposing the root system into a sum of two-dimensional sub-root systems, A2, B2, G2, and I2(m). The root type and the minimal type Lax pairs, derived in our previous papers, are given as the simplest representations. The spectral parameter dependence plays an important role in the Lax pair operators, which bear a strong resemblance to the Dunkl operators.

14. Complete genome sequence of the alkaliphilic bacterium Bacillus halodurans and genomic sequence comparison with Bacillus subtilis.

- Abstract
The 4 202 353 bp genome of the alkaliphilic bacterium Bacillus halodurans C-125 contains 4066 predicted protein coding sequences (CDSs), 2141 (52.7%) of which have functional assignments, 1182 (29%) of which are conserved CDSs with unknown function and 743 (18. 3%) of which have no match to any protein database. Among the total CDSs, 8.8% match sequences of proteins found only in Bacillus subtilis and 66.7% are widely conserved in comparison with the proteins of various organisms, including B.subtilis. The B. halodurans genome contains 112 transposase genes, indicating that transposases have played an important evolutionary role in horizontal gene transfer and also in internal genetic rearrangement in the genome. Strain C-125 lacks some of the necessary genes for competence, such as comS, srfA and rapC, supporting the fact that competence has not been demonstrated experimentally in C-125. There is no paralog of tupA, encoding teichuronopeptide, which contributes to alkaliphily, in the C-125 genome and an ortholog of tupA cannot be found in the B.subtilis genome. Out of 11 sigma factors which belong to the extracytoplasmic function family, 10 are unique to B. halodurans, suggesting that they may have a role in the special mechanism of adaptation to an alkaline environment.

Expression mechanism of CD15s (sialyl-Lex, sLex) antigen has been investigated using human B lymphoid cell lines. sLex structures were not expressed in mature B lymphoids but highly expressed in pre-B leukemia and pre-B lymphoma cell lines. The expression site was mainly on the O-linked oligosaccharide chains and E-selectin mediated-cell adhesion capability of sLex-positive cells were significantly suppressed by benzyl-α-GalNAc treatment. Subsequently, the bases of the sLex expression control mechanism were examined at the levels of enzymatic activities and transcripts of glycosyltransferases. (1) The activities of α1→3fucosyltransferase, α2→3sialyltransferase, β1→4Gal-transferase, and elongation β1→3GlcNAc-transferase, did not correlate with sLex expression levels. (2) The transcripts of Fuc-TVII were not parallel with sLex expression, and those of ST3Gal IV and β1→4Gal-transferase were constitutively detected in all cell lines tested. (3) There was no detectable enzyme activity for core 3 and 4 backbone structure synthesis in human B cell lines. (4) By contrast, the enzyme activities and transcripts of UDP-GlcNAc:Galβ1→3GalNAc (GlcNAc to GalNAc) β1→6N-acetylglucosaminyltransferase (Core2GnT) had significant correlation with the cell surface expression of sLex antigen. (5) Moreover, Western blot analysis revealed the presence of a major ∼150 kDa glycoprotein that carries O-linked oligosaccharides recognized by anti-sLex monoclonal antibody in sLex-positive pre-B leukemia cell lines. This correlation of Core2GnT with CD15s expression suggests that Core2GnT is a regulator of the cell surface expression of sLex in human pre-B lymphoid cells.

Background: Oral immunotherapy with a peptide for allergic immune responses is theoretically a promising therapy but has not been established yet. Objective: To evaluate immune suppressive efficacy of oral administration of an immunodominant peptide, we investigated changes in T-cell proliferation, T"H"1 - and T"H"2 -cytokine production, and T"H"1 - and T"H"2 -mediated antibody production in mice after oral administration of a peptide. Methods: Peptide p246-259, containing a dominant T-cell determinant of Cry j 2, which is the major allergen in Japanese cedar pollen, was used in this study. Groups of mice received p246-259 or PBS alone before or after they were primed intranasally with Cry j 2 and cholera toxin. In another experiment mice were primed intraperitoneally with Cry j 2 and alum. Proliferative response and cytokine production by nasal-associated lymph node cells against Cry j 2 were investigated. Amounts of systemic anti-Cry j 2 IgE and IgG antibodies were also measured. Results: Oral administration of the peptide to mice before, or even after, the sensitization induced oral tolerance in T-cell responses against the allergen; the tolerance was associated with decreased production of T"H"1 (IFN-@c and IL-2) and T"H"2 (IL-4) cytokines. Allergen-specific T"H"1 -mediated (IgG2a and IgG2b) and T"H"2 -mediated (IgG1 and IgE) antibody responses were also inhibited. Conclusions: Oral administration of a dominant T-cell determinant peptide induces immunologic tolerance in both T"H"1 and T"H"2 cell responses against the whole protein allergen. Our study is the first, to our knowledge, to demonstrate the potential for peptide-based oral immunotherapy in order to treat allergic immune responses. (J Allergy Clin Immunol 1998;102:961-7.)

The preincubation of vascular smooth muscle cells (VSMC) with a non-peptide arginine vasopressin (AVP) antagonist (OPC-21268) (0.5 mM) for 10 min completely inhibited the 1 µM AVP-induced activation of mitogen-activated protein (MAP) kinase but did not affect the angiotensin II-, protein kinase C activator- or ionomycin-stimulated MAP kinase. Similar results were obtained with the simultaneous administration of AVP and OPC-21268. This inhibitory effect of OPC-21268 completely disappeared after washing the cells pretreated with OPC-21268. In contrast, the preincubation of VSMC with a peptide AVP antagonist, d(CH2)5Tyr(Me)AVP (1 µM), for 10 min completely blocked the 1 µM AVP-activated MAP kinase but the simultaneous administration of d(CH2)5Tyr(Me)AVP with AVP could inhibit that only by 40%. However, the washing procedure did not affect the inhibitory effect of d(CH2)5Tyr(Me)AVP. These results indicate that OPC-21268 is a specific AVP antagonist to inhibit the AVP-induced activation of MAP kinase. The antagonistic effect of OPC-21268 is also suggested to be relatively weak but prompt as compared to that of a peptide AVP antagonist d(CH2)5Tyr(Me)AVP.Copyright 1994, 1999 Academic Press, Inc.

- Abstract
A hybridoma cell line producing the monoclonal antibody against erythropoietin receptor (EpoR) was established using the soluble ectodomain of mouse erythropoietin receptor (sEpoR) as an antigen. The monoclonal antibody termed 1G3 bound to the denatured sEpoR. Epitope mapping with peptide library revealed that 1G3 recognized the amino terminal region including the hexapeptide (positions 6 to 11; LeuProAspProLysPhe). The amino acid sequence in this hexapeptide was identical in mice, rats, and humans, and therefore 1G3 bound to EpoR from all of these sources. Using 1G3, we evaluated sEpoR by a sandwich enzyme-linked immunoassay, and EpoR in the solubilized membrane preparation was detected by Western blotting. The cells expressing EpoR were identified with immunochemical staining. We confirmed the presence of EpoR in a neuronal cell line and PC12 cells, and EpoR was expressed in primary cultured hippocampal neurons.

31. Hybridomas for production of monoclonal antibodies to human erythropoietin

- Abstract
Human urinary erythropoietin has been highly purified by a combination of conventional purification methods and immunoadsorbent columns packed with hybridoma-produced antibodies against contaminants that seemed difficult to separate from erythropoietin by the usual means. By using the partially purified erythropoietin as an antigen, three hybridoma clones have been obtained that secrete monoclonal antibodies against erythropoietin. One of the clones has been quite stable, with a rapid growth rate and high production of antibody. Western blotting technique with monoclonal antibodies revealed occurrence of two species of erythropoietin. The monoclonal antibody will be useful as a probe for the purification of erythropoietin and for further studies of the hormone and its mechanism of action.

32. Connection of atopic disease in Japanese patients with juvenile dermatomyositis based on serum IgE levels

- Abstract
Serum IgE levels of 22 patients with juvenile dermatomyositis (JDMS), 44 normal children, and 43 patients with adult dermatomyositis were compared. The geometric mean of serum IgE levels was significantly higher in the juvenile patients when compared with normal children (p<0.01) and adult patients (p<0.01). Of the 22 patients with JDMS, 11 (50%) had elevated serum IgE levels accompanied by atopic disease. Dermatomyositis was accompanied by atopic dermatitis (AD) in 9 (41%) of these 22 patients, a high prevalence when compared with reports among the general population. Following the appearance of muscular symptoms in JDMS patients with AD, the ratio of OKT4 to OKT8 cells rose due to a reduction in the percentage of OKT8-positive cells, along with further elevations in serum IgE levels and intractable cutaneous manifestations of dermatomyositis. All this may be a result of an interaction between the immuno-mechanism of this disease and that of AD. We suspect that, in general, children with impaired cell-mediated immunity, presenting with such symptoms as AD, may have a higher tendency for developing JDMS.

- Abstract
To evaluate the extent of personal exposure to airborne mite allergens, subjects were asked to carry a personal air sampler when in their houses. The level of Der 1 allergen trapped by the sampler was measured with a highly sensitive immunoassay. There were great variations in airborne Der 1 exposure in each subject. When used bedding was replaced with new allergen-free bedding, we detected a decrease in the allergen level. The use of new bedding seems to be an effective measure for reducing airborne mite allergen exposure. (J ALLERGY CLIN IMMUNOL 1996;97:1040-4.)

- Abstract
Transglutaminases (EC 2.3.2.13) catalyze the formation of ε-(γ-glutamyl) lysine cross-links and the substitution of primary amines for the γ-carboxamide groups of protein-bound glutamine residues. There are conserved anionic regions in transglutaminases, some of which are thought to be possible calcium-binding sites. By site-directed mutagenesis, three mutant forms of recombinant guinea-pig liver transglutaminase, in which some acidic amino acid residues in two conserved regions became nonionic, were expressed in Escherichia coli: TGM1, with Asp-231 and -232 changed to Asn; TGMS, with Glu-445, -448, -449, -450, and -452 changed to Gin; and TGM3, with the mutations of both TGM1 and TGM2. The size and level of synthesis of the mutant proteins were unchanged when monitored by immunoblotting. All mutants retained enzyme activity, and their apparent Km values for substrates during histamine incorporation into acetyl αs1-casein were similar to those of the wild-type enzyme, but their Vmax values were smaller. The deamidation rate of glutamine residues in the acetyl αs1-casein was unaffected, but the rate of protein cross-linking catalyzed by these mutants was very low. All mutations caused with the enzyme a decrease in the sensitivity to activation by calcium and an increase in the sensitivity to inhibition by GTP. These results indicated that the negative charges of some acidic amino acid residues in the two conserved anionic regions of transglutaminase are not essential for its activity but the loss of their negative charges affects some catalytic properties.Copyright 1995, 1999 Academic Press, Inc.

36. Characterization and use of monoclonal antibodies directed against human erythropoietin that recognize different antigenic determinants

- Abstract
We have established four hybridoma cells that produce monoclonal antibodies (MoAbs) R2, R4, R6, and R12 directed toward recombinant human erythropoietin (rHuEPO). MoAbs R2, R4, and R6 bound to EPO with high affinities (kd = approximately 2, 4, and 1 nmol/L, respectively) but MoAb R12 had a low affinity (240 nmol/L). These antibodies inhibited the biological activity of rHuEPO and EPOs from humans, rats, mice, and rabbits. This inhibition was due to the blocking of EPO binding to the target cells. The fully deglycosylated rHuEPO bound to the MoAbs, indicating that they recognized peptide sequences of the antigen but not the carbohydrates attached to the antigen. An immunosorbent column with the immobilized MoAb R2 was effective for the rapid purification of EPO. MoAb R6 bound to EPO at a site(s) different from those to which other MoAbs bound. Based on this finding, a sensitive and rapid enzyme-linked immunosorbent assay of EPO, in which EPO was sandwiched between two MoAbs (R2 and R6), was developed. The assay measured plasma levels of EPO as low as 5 mU/mL within several hours.

37. An Analysis of Pipe Flange Connections Using Epoxy Adhesives/Anaerobic Sealant Instead of Gaskets

- Abstract
This paper deals with the strength and the sealing performance of pipe flange connections combining the bonding force of adhesives with the clamping force of bolts. The epoxy adhesives or anaerobic sealants are bonded at the interface partially instead of gaskets in pipe flange connections. The stress distribution in the epoxy adhesives (anaerobic sealant), which governs the sealing performance, and the variations in axial bolt force are analyzed, using an axisymmetrical theory of elasticity, when an internal pressure is applied to a connection in which two pipe flanges are clamped together by bolts and nuts with an initial clamping force after being joined by epoxy adhesives or anaerobic sealant. In addition, a method for estimating the strength of the combination connection is demonstrated. Experiments are performed and the analytical results are consistent with the experimental results concerning the variation in axial bolt force and the strength of combination connections. It can be seen that the strength of connections increases with a decrease in the bolt pitch circle diameter. Furthermore, it is seen that the sealing performance of such combination connections in which the interface is bonded partially is improved over that of pipe flange connections with metallic gaskets.

- Abstract
Although erythropoietin (Epo) has been shown to possess in vitro angiogenic activity, its physiological significance has not been demonstrated. Normally angiogenesis does not occur actively in adults but an exception is the female reproductive organ. In the uterine endometrium, angiogenesis takes place actively for supporting the endometrial growth that occurs during transition from the diestrus to estrous stage. This transition is under control of 17beta-estradiol (E2), an ovarian hormone, and can be mimicked by injection of E2 to ovariectomized (OVX) mouse. Thus, the uterus is a pertinent site to examine the Epo function in angiogenesis. We found that Epo protein and its mRNA were produced in an E2-dependent manner, when the uterus from OVX mouse was cultured in vitro. The de novo protein synthesis was not needed for E2 induction of Epo mRNA. Administration of E2 to OVX mouse induced a rapid and transient increase in Epo mRNA in the uterus. Injection of Epo into the OVX mouse uterine cavity promoted blood vessel formation in the endometrium. Furthermore, injection of the soluble Epo receptor capable of binding with Epo into the uterine cavity of non-OVX mouse in diestrus stage inhibited the endometrial transition to proestrus stage, whereas heat-inactivated soluble Epo receptor allowed the transition to occur. These results, combined with our finding that the endothelial cells in uterine endometrium express Epo receptor, strongly suggest that Epo is an important factor for the E2-dependent cyclical angiogenesis in uterus.

39. Isolation of human erythropoietin with monoclonal antibodies.

- Abstract
Human erythropoietin was isolated from urine of aplastic anemic patients in a high yield with a simple purification procedure using an immunoadsorbent column of monoclonal antibodies and a Sephadex G-100 column. About 6 mg of erythropoietin was isolated from 700 liters of urine and the specific activity was estimated to be 81,600 units/mg of protein with an in vivo 59Fe incorporation assay method, using starved rats. Activity measurement of the extracts from sliced gels after sodium dodecyl sulfate-polyacrylamide gel electrophoresis and the Western blotting technique revealed heterogeneity of the isolated erythropoietin, which is probably caused by variable amounts of carbohydrates attached to the polypeptide chain. Thirty amino acids in the NH2-terminal portion of the isolated hormone were sequenced.

40. Larval dispersal and recruitment of Haliotis discus hannai and Tegula spp. on Miyagi coasts, Japan

- Abstract
The spatial dispersal and vertical distribution of larvae of Haliotis discus hannai and Tegula spp. were examined in relation to storm events in Miyagi Prefecture, Japan, from 1983 to 1993. Epidemic spawning is induced by typhoons and minor storms. After typhoons, larvae are dispersed in coastal waters up to 3 km from shore to a depth of 30 m and later are advected toward the coast by onshore winds. After minor storms, larvae are locally dispersed close to shore. Larvae of H. discus hannai and Tegula spp. become vertically stratified and settle on the substratum at depths in accordance with the vertical depth range of their larvae. Strong settlements of H. discus hannai of up to 200 m-2 were recorded around headlands, with average survival rates of about 13% per month in the first 2-3 months. The adaptive significance of dispersal over short and long distances is considered.

In the Riemann-Cartan space we develop a gravitational theory of spinor electrodynamics, making torsion propagate in vacuum. Prom the precision experiment we can deduce the torsion coupling constant divided by the mass,f2/4πμ2< 10−5(GeV)−2, for ώ≪mα and ώ≫mα, where m is the electron mass and α is the fine-structure constant. It is interesting to inquire if there is a new interaction.

43. A novel hypothyroid 'growth-retarded' mouse derived from Snell's dwarf mouse

- Abstract
This paper describes a novel mutant mouse that has been spontaneously derived from the Snell's dwarf (DW/J) mouse. It was named the 'growth-retarded mouse' because of a characteristic growth pause followed by the delayed onset of pubertal growth. The onset of the increase in pituitary GH content that normally occurs concomitant with pubertal growth was also delayed in the growth-retarded mice. The serum concentration of thyroxine was very low in these mice from the neonatal period through adulthood, and a supplement of tri-iodothyronine was effective in shortening the growth pause and commencing the suppressed pubertal growth. Histological and immunohistochemical studies revealed that the anterior pituitary gland of the growth-retarded mouse contains clustered unusual chromophobic cells which are not reactive to various antisera against anterior pituitary hormones and the gland becomes enlarged with age. Breeding data indicated that these characteristics of the mice show an autosomal recessive inheritance and the gene responsible was designated as 'grm'. Partial linkage analysis utilizing microsatellite polymorphism demonstrated that the grmgene does not identify with the litor hytgenes. Based on comparison of the hormonal status and growth pattern between growth-retarded, dwarf and normal mice, we have suggested the existence of a mutual interaction, possibly positive feedback regulation, between the pituitary and thyroid glands, that develops or matures the hormonal network which is responsible for rapid somatic growth and metabolic changes at puberty in mice.Journal of Endocrinology(1994) 142,435–446

44. A notable change in mortality of aplastic anemia observed during the 1970s in Japan

- Abstract
The age adjusted mortality rate of aplastic anemia in Japan decreased sharply in the mid-1970s; before this time, it had increased only slightly. A report on the survival rate among a substantial number of patients with aplastic anemia showed that patients who sought medical care at the hospital after 1973 had a better survival rate than those who sought care before 1973; the extent of improvement in the survival rate was almost equal to the extent of decrease in the mortality rate. The supply of platelet concentrates was found to increase with the decrease in the age adjusted mortality rate. Although there were several factors that affected the mortality rate from aplastic anemia, the major recognizable factor seemed to be therapeutic improvement, mainly due to the platelet concentrates. Probably neither a decrease in the incidence nor factors related to the data processing of vital statistics played a major role.

45. Effect of tunicamycin treatment on ligand binding to the erythropoietin receptor: conversion from two classes of binding sites to a single class

- Abstract
Scatchard analyses of erythropoietin (EPO) binding to its receptor (EPO- R) have shown that some erythroid cells display a biphasic nature of the ligand-saturation curve, indicating the presence of two classes of binding sites with different affinities. The biochemical basis accounting for this observation is unknown. We found that the culture of a human erythroleukemia cell line with tunicamycin, an inhibitor of N-glycosylation, converted the biphasic Scatchard plot to a single phase with high-affinity sites. Scatchard plots of baby hamster kidney (BHK) cells that had been engineered to express cloned mouse EPO-R were also biphasic and the plots of cells cultured with tunicamycin became a single phase with high-affinity sites. Mouse EPO-R is glycosylated at one asparagine residue in the extracellular region. The mutant EPO-R, in which asparagine residue responsible for N-glycosylation was replaced with glutamine residue, was expressed on BHK cells. Unexpectedly, mutant EPO-R was similar in ligand binding to wild-type EPO-R. BHK cells that expressed mutant EPO-R showed biphasic Scatchard plots that were converted to single-phase plots with only high-affinity sites by tunicamycin treatment. These results indicate that the N- linked sugar of EPO-R is not involved in the manifestation of two classes of binding sites, and that there is a yet unidentified glycoprotein crucial for the ligand-saturation characteristics of EPO-R.

46. Terminal deoxynucleotidyl transferase activities and glucocorticoid receptors in leukaemia

- Abstract
The relation between terminal deoxynucleotidyl transferase (TdT) activity, glucocorticoid (GC) receptors and the effect of vincristine-prednisolone (VP) therapy on fresh leukaemia cases was examined. Five of 6 TdT+ leukaemias showed high levels of GC receptors and a favourable response to VP therapy, whereas 1 acute lymphoblastic leukaemia (ALL) and 3 of chronic myelogenous leukaemia (CML) cases in blast crisis with no TdT activity showed low level of GC receptors and poor response to VP therapy. Significant correlation (r = 0.821, P less than 0.01) was observed between TdT activity and the number of GC receptor sites in these cases. X2 test showed significant difference (P less than 0.01) between TdT+ and TdT- leukaemias in the effect of VP. A significant difference (P less than 0.01) was also observed between VP-effective and ineffective leukaemias in the number of GC-receptor sites by unpaired t test. Therefore GC receptors may be responsible for the effect of VP on TdT+ leukaemias.

47. Effect of titanium, niobium and zirconium on creep rupture strength and ductility of cobalt base superalloys

Abstract: An attempt has been made to develop a cobalt base casting superalloy (30Cr-10Ni-7W-Co) having high creep rupture strength and ductility for first stage nozzles of gas turbines. In cobalt base superalloys, there was found to exist a close correlation between the creep rupture strength and MC type carbide forming elements such as Ti, Nb and Zr. In cobalt base alloys with 0.25 wt pct C, precipitation and coarsening of carbides can be reduced by addition of Ti, Nb and Zr. Therefore, by adding the optimum amount of Ti, Nb and Zr, precipitation of carbides in the alloy reaches such an amount as to give the highest creep rupture strength. Excess addition of Ti, Nb and Zr does not improve the creep rupture strength. By adding C, creep rupture strength of the cobalt alloy with Ti, Nb and Zr can be improved and becomes the highest at 0.40 wt pct. C. According to the experimental results, the creep rupture strength becomes the highest at a value of (Ti + Nb + Zr)/C (atomic ratio) of about 0.3. Contrary to the expectation, it was found in this experiment that the ductility in creep rupture tests increases with increasing carbon content up to 0.6 wt pct.

Studies have been done on stress corrosion cracking (SCC) susceptibility of low alloy steels in water containing dissolved oxygen. The fundamental factors which affect the SCC susceptibility were clarified, and the integrity of the reactor pressure vessel (RPV), in which these steels are used, was assessed from the standpoint of SCC initiation. The effects of applied stress, strain rate, dissolved oxygen concentration, and test temperatures, on the SCC susceptibility was examined utilizing uniaxial constant load tensile tests (UCL), and slow strain rate tests (SSRT).
