ABSTRACTThis study evaluates the relationship between solution heat treatment and hot plastic deformation before aging treatment and its effect on A356 alloys’ mechanical properties and microstructural features. The study employed tensile tests, Vickers microhardness tests, and electron microscopy analysis. The results show that hot plastic deformation before the solution treatment leads to higher elongation, ultimate tensile stress, and yield strength values, which can be attributed to modifications in eutectic Si particles (spheroidisation, aspect ratio and roundness). On the other hand, hot plastic deformation after the solution treatment mainly affects the elongation of the alloy. This effect is due to the fracture of eutectic Si particles and the formation of cracks and voids between these particles, which facilitate crack nucleation. Thus, the combination of hot plastic deformation and subsequent solution treatment significantly improves the mechanical properties of the A356 alloy. The observed changes in microstructure and mechanical behaviour provide valuable insights for optimising the material's properties and refining an established process in the automotive wheel industry.

Three experiments were conducted to determine the effects of dietary addition of crude glycerin on pellet production efficiency and to evaluate its effect on growth performance and digestibility in growing-finishing pigs. Three dietary treatments were created by addition of 0, 2.5, or 5% crude glycerin to barley-soybean meal-based diet, and 4 batches of each dietary treatment (2 each for grower and finisher diets) were prepared. In the manufacturing process, crude glycerin supplementation linearly increased the feeder speed and production rate (P < 0.05), resulting in a 20 to 29% improvement in the feed production rate compared with the control. Production efficiency (kg/kWh) increased linearly (P < 0.05) as the level of crude glycerin in feed increased. A growth experiment was performed with 240 barrows (30 ± 1 kg initial BW) using a 2-phase feeding program over a 12-wk period with 4 pens per treatment and 20 pigs per pen. On the last day of the growth experiment, blood samples were collected to determine circulating glucose, fructosamine, and IGF-1 concentrations. Overall growth performance was not affected (P > 0.05) by dietary treatment, and there was no effect (P > 0.05) of dietary treatment on any plasma metabolite measured. A digestibility experiment involving 9 male pigs housed in metabolic cages was used to determine the coefficients of apparent fecal digestibility and N and mineral balances. Pigs were assigned to 1 of the 3 diets in each feeding period using a 3 × 3 Latin square arrangement of treatments (43 ± 3 and 74 ± 3 kg initial BW in the growing and finishing periods, respectively). In both feeding periods, fecal digestibility of OM and ether extract were affected by dietary treatment, increasing linearly (P < 0.05) with increasing crude glycerin levels. However, neither CP digestibility nor N retention was affected by the glycerin content in either the growing or finishing period. Digestibilities and balance of Ca and P showed opposite tendencies with the variations in crude glycerin content, which either decreased or increased depending on the feeding period. In conclusion, adding crude glycerin to the diet before pelleting improved feed mill production efficiency. The addition of crude glycerin up to 5% in the diet of growing-finishing pigs had no effect on growth performance, blood metabolites, nutrient digestibility, and N balance, but more studies are needed to determine how crude glycerin affects mineral metabolism and balance.

Nine by-products used in animal nutrition were examined for their nutritive value by determining the chemical composition (crude protein, crude fat (ether extract), neutral detergent fibre, acid detergent fibre, lignin), in vitro organic matter digestibility and rumen fermentation kinetics (from gas production curves measured in vitro). The by-products studied were giant pumpkin, red pepper, stem broccoli, brewer's grain, fresh artichoke, scalded artichoke, lemon peel, orange peel and melon. The nutritive value was very variable, depending on the by-product and on the process applied to the material during industrial processing. In vitro gas production was measured for 500 mg dry matter in quadruplicate at 39 °C after 6, 12, 24, 36, 48 and 72 h of incubation and fitted to single-pool exponential equations. The fermentation kinetics indicated that brewer's grain was the only by-product which fitted well with the p = a + b(1 - e^-ct) equation model; all other by-products had a very fast degradation rate and their gas production fitted the equation p = b(1 - e^-c(t-L)), because in the other exponential model a was negative.

ABSTRACTSeasonal changes in the phasing of diel feeding rhythms were investigated in sea bass held under natural conditions. Demand-feeding behavior was continuously monitored over a complete annual cycle in 10 groups of 15 fish maintained in an outdoors laboratory subjected to natural fluctuations of photoperiod (from 9.5h to 15h) and water temperature (from 13.2d`C to 27.4d`C). A double seasonal phase inversion was detected in all groups: fish that were diurnal in summer and in autumn changed to nocturnal in winter and returned to being diurnal in spring. Diurnal sea bass displayed a positive and stable phase relationship between the peak of the feeding phase and the daily acrophase of water temperature (Φ1 = 0.72h ± 0.33h) and between the peak of the feeding phase and sunset (Φs = 2.94h ± 0.53h), but both Φl, and Φs became negative when sea bass shifted to nocturnalism in winter. The percentage of diurnal feeding behavior peaked in June (94.1%) and dropped in February (29.1%), following a cyclic dynamic modulated by both monthly photoperiod and water temperature. These results contribute to better understanding of the dual phasing behavior of sea bass, which exhibits diurnal or nocturnal behavior according to the time of the year so that flexibility in phasing may be advantageous for the fish to cope with seasonal changes in their environment. Steady-state fluorescence and molecular mechanics calculations were used to study the inclusion complexes of 9-methyl anthracenoate (MA) and 1-methyl pyrenoate (ΜP) with β-cyclodextrin (βCD). Binding constants of 1:1 complexes at different temperatures were obtained from the analysis of the fluorescence enhancement of the βCD solutions with respect to the MA or MP free chromophores. The thermodynamic parameters ΔH and ΔS were also obtained. Molecular mechanics calculations were applied to study both inclusion processes in vacuo and in the presence of water as a solvent. Complexation is mainly due to nonbonded van der Waals host−βCD interactions. Both MA and MP penetrate only partially into the βCD cavity.

1. In the unanesthetized spinal cord of the cat, simultaneous intracellular recordings were made from two motoneurons belonging to the gastronemius motor nucleus. 2. Supramaximal iterative stimulation of small branches of the gastrocnemius nerve produced monosynaptic EPSPs (Ia EPSPs) of varying amplitude superimposed on a fluctuating base line. 3. In most cases the variance of the motoneuron membrane potential was increased above base-line levels with a time course approximately matching the Ia EPSP. This suggests that Ia EPSP fluctuations are greater than can be accounted for by the base-line fluctuations alone. 4. For a given series of Ia EPSPs, the smaller responses in the series had about the same decay phase as the larger EPSPs, suggesting that most of the Ia EPSP fluctuations were not due to systematic changes in postsynaptic conductances produced by ongoing activity, but rather to a presynaptic mechanism. 5. Simultaneous recording from two motoneurons showed that base-line fluctuations were positively correlated. In most cases, however, there was an additional increased correlation above base-line levels resembling the time course of the Ia EPSPs, indicating positive correlation between EPSP fluctuations which is attributed to a presynaptic mechanism. 6. Conditioning volleys to group I muscle afferents or to low-threshold cutaneous afferents reduced the variance of the Ia EPSPs and also their correlation in motoneuron pairs, often without changing the mean Ia EPSPs. 7. It is concluded that, in the unanesthetized spinal cord, in addition to the random process which governs transmitter release intrinsic to a given synaptic terminal, there is another stochastic process affecting, in a correlated manner, transmitter release in large sets of Ia synaptic terminals. Most likely, the correlation in transmitter release is achieved by membrane potential fluctuations imposed on the Ia terminal arborizations by ongoing activity of the segmental mechanism mediating primary afferent depolarization. 8. The effects of such a correlating influence on cell firing behavior have been analyzed. The results suggest that this mechanism, referred to as control by presynaptic correlation, is able to modulate the information transmitted from Ia fibers to motoneurons.

1. In the unanesthetized spinal cord, conditioning stimulation of low-threshold afferents (below 1.3 times threshold strength) in the biceps semitendinosus (BST) nerve often reduced the peak amplitude of the monosynaptic Ia EPSPs evoked in gastrocnemius motoneurons without affecting the monosynaptic component of the EPSPs evoked by stimulation of the ipsilateral ventral funiculus (VF) in the thoracic cord. 2. Volleys to the BST nerve comprising higher threshold afferents (usually above 1.4 times threshold strength) reduced the peak amplitude of the monosynaptic Ia and VF EPSPs and shortened their falling phase. 3. Conditioning volleys to low-threshold cutaneous afferents often increased the Ia-EPSP peak amplitude, sometimes without affecting the monosynaptic component of the VF EPSP. 4. In most cases the Ia nd VF monosynaptic EPSPs elicited in a given motoneuron summated nonlinearly. The amount of nonlinear summation between Ia and VF monosynaptic EPSPs was often reduced by low-threshold BST conditioning volleys. These observations suggest that in many instances, both species of fibers end in "electrotonically close" synaptic loci over the motoneuron surface. Therefore, amplitude changes of monosynaptic Ia EPSPs produced by conditioning afferent volleys without concomitant changes of monosynaptic VF EPSPs do not appear to result from postsynaptic remote conductance changes and may be attributed to a presynaptic mechanism. 5. At the time of occurrence of the Ia and VF monosynaptic EPSP the variance of the motoneuron membrane potential may be increased above base-line levels with a time course approximately matching the EPSP itself. Conditioning stimulation of BST afferents usually reduced Ia EPSP variance, often without affecting or even increasing the variance of the monosynaptic VF EPSPs. These observations provide additional evidence that Ia EPSP variability is introduced, at least in part, through the segmental pathways mediating primary afferent depolarization. 6. The possibility of a differential control of the information flow transmitted through two independent channels converging on a given cell ensemble is discussed.

A herd of 28 castrated adult male goats of the Murciano-Granadina breed was used to determine the direct and by-difference digestibility of untreated straw, straw treated with 4% urea and straw treated with 4% urea and 1% NaOH.

Gallbladder mucus is mainly composed of glycoproteins, which seem to play a critical role in cholesterol nucleation during gallstone formation. The biosynthetic pathway and sequential processing as well as the characterization of the oligosaccharide sidechains of human gallbladder secretory glycoproteins have not been completely defined. The aim of the present study is the subcellular characterization of the glycoproteins in the principal cells of human gallbladder. Principal cells of normal human gallbladder were studied by means of a variety of cytochemical techniques, including lectin histochemistry, enzyme and chemical treatments, immunocytochemistry and lectin-gold technology. Fucose, galactose, N-acetylglucosamine, N-acetylgalactosamine and N-acetylneuraminic acid residues were detected in mucous granules, Golgi apparatus and apical membrane of principal cells. Mannose residues were only observed in dense bodies. Oligosaccharide side-chains of the glycoproteins contained in the biliary mucus are synthesized in the Golgi apparatus of the principal cells of the gallbladder epithelium and are also contained in the mucous granules of these cells. Terminal N-acetylneuraminic acid(a2-3)galactose(ß1-3)N-acetylgalactosamine, N-acetylneuraminic acid(a2-3)galactose(ß1-4)N-acetylglucosamine and galactose(ß1-4)N-acetylglucosamine sequences are contained in the oligosaccharide chains of gallbladder mucus glycoproteins. The dense bodies detected in the cytoplasm of the principal cells contained N-linked glycoproteins. Mucin-type O-linked glycoproteins were the main components of the mucous granules although some N-linked chains were also detected.

A battery of seven lectins and several conventional mucin histochemical techniques were used to identify the epithelial mucins of the gallbladder of ten species: man, rabbit (Oryctolagus cuniculus, mammalia), hamster (Mesocricetus auratus, mammalia), chicken (Gallus gallus, bird), sparrow (Passer domesticus, bird), moorish gecko (Tarentola mauritanica, reptilia), ladder snake (Elaphe scalaris, reptilia), lake frog (Rana perezi, amphibia), natterjack toad (Bufo calamita, amphibia) and gilthead sea bream (Sparus auratus, fish). Glycogen was found in the epithelial lining of the reptilian and amphibian gallbladders. Sulphate and carboxyl groups were frequently found in the same species, except in the ladder snake and natterjack toad gallbladders where only sulphate groups were identified. Sialic acid residues were detected in man, rabbit, bird, T. mauritanica, R. perezi and fish gallbladders. ConA binding pattern was similar in the ten species studied. In the human gallbladder only PNA failed to label the luminal surface, while the glands were only unreactive to DBA. The human gallbladder showed a large variety of saccharides. The present results suggest that no relation exists between the composition of the gallbladder mucins and the situation of the species in the phylogenetic scale.

Steady-state fluorescence and molecular mechanics calculation were used to study the inclusion complexes of 2-methylnaphthoate (MN) with γ-cyclodextrin (γ-CD). The stoichiometry (1:1) and binding constants (213 ± 96 M^-1 at 25 °C) were extracted from an analysis of the ratio of the intensity of two emission bands that were sensitive to the polarity of the medium. Extrapolation of this ratio to a high concentration of γ-CD permits the estimation of the polarity of the inner cavity, which seems to be only slightly hydrophobic, with a dielectric constant near 74. The ΔH and ΔS for formation of the complex were obtained and compared with previous results for similar complexes of MN with two smaller CDs. Molecular mechanics calculations were applied to study the complex in vacuo and in the presence of water as a solvent. Complexation is mainly due to nonbonded van der Waals interactions of MN with γ-CD. Calculations show that MN penetrates completely into the cavity of γ-CD.

1. A comparative study was made of the effect of the phorbol ester, 12‐O‐tetradecanoylphorbol‐13‐acetate (TPA) on cholecystokinin octapeptide‐evoked exocrine pancreatic secretion in the anaesthetized rat and isolated permeabilized pancreatic acinar cells. 2. Cholecystokinin octapeptide (CCK8; 0.10‐6.40 nmol (kg body weight)‐1) induced dose‐dependent increases in pancreatic juice flow, total protein output and amylase release in the anaesthetized rat. 3. Administration of TPA (10(‐8) mol (kg body weight)‐1) in combination with CCK8 resulted in marked attenuation of the CCK8‐evoked secretory response. 4. Simultaneous injection of polymyxin B (10(‐8) mol (kg body weight)‐1), an inhibitor of protein kinase C, with TPA and CCK8 reversed the inhibitory effect of the phorbol ester on CCK8‐induced pancreatic juice flow, total protein output and amylase release. 5. In permeabilized rat pancreatic acini CCK8 (10(‐13)‐10(‐9) M) elicited dose‐dependent increases in [3H]leucine‐labelled protein secretion (3H‐labelled protein release). Combining TPA (10(‐8) M) with CCK8 resulted in an inhibition of the CCK8‐induced 3H‐labelled protein release especially at lower concentrations of CCK8. At higher concentrations of CCK8, TPA was unable to inhibit the CCK8‐evoked 3H‐labelled protein release. Again, polymyxin B reversed the TPA‐induced inhibition of CCK8‐evoked 3H‐labelled protein output. 6. The results indicate that protein kinase C activation may play an important physiological role in modulating the CCK8‐evoked secretory response in rat pancreas in vivo and in vitro.

A new biliary bidirectional cannula is described which allows the study of biliary secretion in conscious dogs under conditions which approach physiological normality.

Abstract: Periodic food availability can act as a potent zeitgeber capable of synchronizing many biological rhythms in fishes, including locomotor activity rhythms. In the present paper we investigated entrainment of locomotor rhythms to scheduled feeding under different light and feeding regimes. In experiment 1, fish were exposed to a 12:12 h light/dark cycle and fed one single daily meal in the middle of the light phase. In experiment 2, we tested the effect of random versus scheduled feeding on the daily distribution of activity. During random feeding, meals were randomly scheduled with intervals ranging from 12 to 36 h, while scheduled feeding consisted of one single daily meal set in the middle of the light or dark phase. Finally, in experiment 3, we studied the synchronization of activity rhythms to feeding under constant darkness (DD) and after shifting the feeding cycle by either advancing or delaying the feeding cycle by 9 h. The results revealed that goldfish synchronized to feeding, overcame light entrainment and significantly changed their daily distribution of activity according to their feeding schedule. In addition, the daily activity pattern modulated by feeding differed between layers: a peak of activity being noticeable directly after feeding at the bottom, while an anticipatory behaviour was obvious at the surface of the tank. Under DD and no food, free-running rhythms averaging 25.5 ± 1.9 h (mean ± SD) were detected. In conclusion, some properties of feeding entrainment (e.g. anticipation of the feeding time, free-running rhythms following termination of periodic feeding, and the stability of ø after shifting the feeding cycle) suggested that goldfish have (a) separate but tightly coupled light- and food-entrainable oscillators, or (b) a single oscillator that is entrainable by both light and food (one synchronizer being eventually stronger than the other).

The experiments were conduced to examine the existence of circadian rhythms in glutathione concentration and glutathione S-transferase activity in the liver of the rat. In animals synchronized to a 12:12 h light-dark cycle and fasted at 6 different time points to allow exactly 24 h of fasting, both, glutathione concentration and glutathione S-transferase activity show diurnal variation with a maximum during the light period and a minimum at night. On the other hand the hepatic protein level was maximal during the light period and decreased to its lowest level during the dark period. The implications of such oscillations in the circadian rhuthms of toxicological or therapeutical effects of many xenobiotic agents are clear.

Abstract: Melatonin is regarded as an internal zeitgeber, involved in the synchronization to light of the daily and seasonal rhythms of vertebrates. To date, plasma and ocular melatonin in fish have been extensively surveyed almost solely in freshwater species – with the exception of some migrating species of salmonids. In the present paper, melatonin levels of a marine species (sea bass, Dicentrarchus labrax L) were examined. In addition, the daily rhythms of the demand-feeding activity of sea bass, a fish species characterized by a dual phasing capacity (i.e. the ability to switch between diurnal and nocturnal behaviour), were investigated before sampling. Sea bass, distributed in 12 groups of four fish and kept under constant water temperature and salinity, were exposed to a 12 h light:12 h dark cycle (200:0 lx, lights on at 0800 hours). After 4 weeks recording, the animals were killed at 0900, 1200, 1400, 1600, 1900, 2100, 2400, 0200, 0400, 0700 and 0900 hours. Actograms of demand-feeding records revealed a nocturnal feeding behaviour, with some cases of spontaneous inversions in phasing. Melatonin levels in plasma peaked in the middle of the dark phase, dropping after lights on. Melatonin in the eye, on the contrary, exhibited an inverse profile, with high levels during daytime and low levels at night. These results suggest that melatonin in the plasma and the eye may act independently on the flexible circadian system of sea bass.

The effects of histamine alone and histamine plus cimetidine on basal pancreatic exocrine secretion were determined in anaesthetized rabbits with an acute pancreatic cannula. Intravenous histamine administration (0.2–0.8n mol/kg/min) increased pancreatic enzyme secretion. A much greater stimulative effect on pancreatic (cimetidine 4 μmol/kg/min). The results indicate that in the rabbit pancreas the stimulatory effect of histamine is mediated by H1 receptors.

Effects of cholecystokinin (CCK) on bile flow through the sphincter of Oddi (SO) were studied in anaesthetized dogs. Intravenous injection of CCK (0.25, 0.5, 1 and 2 IDU/Kg) elicited a dose-dependent reduction in flow through the SO in the first minutes after CCK administration. Pirenzepine and atropine decreased significantly (P < 0.05) by 29% and a 40% respectively the inhibitory effect induced by 1 IDU/Kg of CCK, whereas hexamethonium elicited an increase in the inhibitory effect induced by 0.5 IDU/Kg of CCK (P < 0.05). Intravenous infusion of cummulative doses of CCK had different effects according to the dose infused. Lower doses (0.025 and 0.05 IDU/Kg/min) increased transphincteric flow, however, high doses (0.1, 0.2 and 0.4 IDU/Kg/min) were inhibitory. These finding indicated that CCK had two effects on the SO: firstly, a contractile effect, probably mediated through a direct myogenic action and neuronal release of ACh, and secondly a relaxant effect, probably mediated by stimulation of inhibitory postganglionic neurons.

The activity of γ-glutamyl transpeptidase (γ-GT) in duodenal mucosa both in healthy rats and in rats experimentally ulcerated with indomethacin increases significantly after oral administration of pirenzepine as well as ranitidine but not after oral administration of sucralphate. These increase in γ-GT activity may contribute to the cytoprotective effects already described for pirenzepine and ranitidine.

The interdigestive pancreatic exocrine secretion of the domestic fowl has been studied following the cosine-vector analysis. For the flow of pancreatic juice the crest phase was at 1040 and the sample amplitude was 2.26μl/min.For the amylase output the crest phase was at 1128 and the sample amplitude was 0.297 U.A.A./min. Both parameters are characterized by a phase-and frequency-synchronized rhythm whose period must be close to 24 hr.

We investigated the glycoconjugates of the human bronchial glands at light and electron microscopic level by means of lectin histochemistry in combination with neuraminidase digestion and beta-elimination reaction. Both direct and indirect techniques using lectin-peroxidase, lectin-gold, and glycoprotein-gold complexes were applied. The binding pattern of the six lectins (ConA, HPA, DSA, WGA, LEA, and PNA) used in the present study suggests that mucous and serous cells of human bronchial glands contain both N- and O-glycosylated proteins in the secretory granules. Asparagine-linked oligosaccharides containing Gal(beta-1,4) GlcNAc and Man residues were abundant in serous cells. The demonstration of both the terminal Neu 5Ac (alpha-2,3, or 6) Gal (beta-1,4) GlcNAc sequence in the N-linked oligosaccharides of mucous cells and the terminal disaccharide Gal (beta-1,4) GlcNAc in the N-linked oligosaccharide chains of serous cells suggests the existence of complex type sugar chains N-glycosidically linked to the peptide region of the glycoproteins. The binding pattern of the DSA and the neuraminidase-DSA sequence provides evidence for the existence of sialyltransferase activity in the forming mucous granules of mucous bronchial cells.

The composition and distribution of sulfo- and sialoglycoconjugates in human laryngeal glands have been investigated at light and electron microscopic levels by use of peroxidase-, digoxigenin-, and colloidal gold-conjugated lectins in combination with several chemical and enzymatic deglycosylation procedures. The present study reveals a variety of terminal oligosaccharide sequences in serous and mucous glands. Serous cells contained glycoconjugates with terminal Neu5Ac (alpha 2-3) Gal (beta 1-4) GlcNAc, Neu5Ac (alpha 2-6) Gal/GalNAc, Neu5Ac (alpha 2-3/6) Gal (beta 1-3 GalNAc, GlcNAc, and Gal (beta 1-4) GlcNAc sequences. Scarce SO4Gal(beta 1-3)GalNAc terminal oligosaccharide chans were detected. Serous cells show wide morphological variability of secretory granules (electron lucent, electron dense, and bizonal) with different lectin affinities. Glycoconjugates in human laryngeal mucous glands contained a variety of terminal oligosaccharide sequences including SO4Gal(beta 1-4)GlcNAc, SO4Gal(beta 1-3) GalNAc, SO4GalNAc, and Neu5Ac(alpha 2-3)GalNAc.
