1. Enhanced Cycle Life and Expanded Voltage Window in Aqueous Proton Full Cells Using TiO2Negative Electrodes with Cationic Vacancies

Abstract

Aqueous batteries face the challenge of limited energy density due to parasitic gas production from hydrogen and oxygen evolution reactions, particularly at the negative electrode. This study investigates the electrochemical properties and mechanisms of proton intercalation in anatase TiO2featuring vacancies (Vac-TiO2), stabilized via a low-temperature sol–gel process. XRD refinement analysis, supported by thermal analysis, estimated 17% cationic vacancies, while 1H MAS NMR spectroscopy revealed stabilization of these vacancies by OH groups. The presence of cationic vacancies led to changes in the oxide anion sublattice, which accommodate proton insertion. Electrochemical assessments in acetate buffer electrolyte demonstrated Vac-TiO2’s ability to delay the hydrogen evolution reaction and enhance proton capacity, validated by pH-dependent studies, DFT calculations, and kinetic analyses. Notably, the occurrence of undercoordinated oxide anions was shown to induce the insertion of H+at higher potential values, and the insertion mechanism was suggested to occur via a solid-solution mechanism. Owing to these features, Vac-TiO2exhibited superior cyclability and performance compared to pure anatase TiO2, highlighting its potential for sustainable proton intercalation processes. In half-cell configurations, Vac-TiO2showed a high Coulombic efficiency (CE exceeding 90% after 48 cycles), while full cells (MnO2||Vac-TiO2) demonstrated an excellent cycling stability (CE exceeding 95.4% over 1000 cycles), high power density (10.5 kW·kg–1vs 6.2 kW·kg–1), and improved self-discharge. This study paves the way for innovative approaches to improving proton intercalation materials, positioning Vac-TiO2as a viable candidate for next-generation energy storage solutions.

3. Diversity of group 1 innate lymphoid cells in human tissues

Abstract

Group 1 innate lymphoid cells (ILC1s) are cytotoxic and interferon gamma-producing lymphocytes lacking antigen-specific receptors, which include ILC1s and natural killer (NK) cells. In mice, ILC1s differ from NK cells, as they develop independently of the NK-specifying transcription factor EOMES, while requiring the repressor ZFP683 (ZNF683 in humans) for tissue residency. Here we identify highly variable ILC1 subtypes across tissues through investigation of human ILC1 diversity by single-cell RNA sequencing and flow cytometry. The intestinal epithelium contained abundant mature EOMES−ILC1s expressing PRDM1rather than ZNF683, alongside a few immature TCF7+PRDM1−ILC1s. Other tissues harbored NK cells expressing ZNF683and EOMEStranscripts; however, EOMES protein content was variable. These ZNF683+NK cells are tissue-imprinted NK cells phenotypically resembling ILC1s. The tissue ILC1-NK spectrum also encompassed conventional NK cells and NK cells distinguished by PTGDSexpression. These findings establish a foundation for evaluating phenotypic and functional changes within the NK-ILC1 spectrum in diseases.

Endovascular renal denervation reduces blood pressure in patients with mild-to-moderate hypertension, but its efficacy in patients with true resistant hypertension has not been shown. We aimed to assess the efficacy and safety of endovascular ultrasound renal denervation in patients with hypertension resistant to three or more antihypertensive medications.

10. La myocardite : une toxicité rare mais grave sous immunothérapie

Abstract

Les chimiothérapies (anthracyclines) et la radiothérapie sont reconnues comme pouvant être responsables de complications cardiovasculaires au long cours. Désormais de nombreuses thérapies ciblées antinéoplasiques vont agir sur des mécanismes spécifiques, en particulier l’immunothérapie (anti-PD1, anti-PDL1, anti-CTLA4). Ces traitements ont récemment montré leur effet favorable sur le pronostic de différents cancers : mélanome, cancer bronchique non à petites cellules, carcinome urothélial, carcinome rénal, carcinome épidermoïde des voies aéro-digestives supérieures et cancer colorectal. Ils améliorent l’évolution spontanée de ces cancers. Des effets secondaires cardiovasculaires rares commencent à être décrits dans la littérature. Une des complications cardiologiques rare (environ 1 % des cas) dont le lien ne fait pas de doute avec l’immunothérapie est la myocardite auto-immune. La présentation clinique regroupe un cortège de signes variés : douleur thoracique, insuffisance cardiaque et troubles du rythme. La réalisation d’un bilan cardiologique préthérapeutique permet d’obtenir des données de référence pour éviter tout retard diagnostique. Le recours à un bilan biologique (troponine et brain natriuretic peptide) et un électrocardiogramme de repos à 12 dérivations sont nécessaires en urgence. Avec ces résultats, un avis cardiologique doit être pris rapidement pour discuter une surveillance en soins intensifs cardiologiques, la réalisation d’une échographie cardiaque et décider d’un traitement adapté (suspension de l’immunothérapie, ajout d’un traitement immunosuppresseur par corticoïdes). L’IRM cardiaque et les biopsies endomyocardiques permettront de confirmer le diagnostic de certitude et de réévaluer la balance bénéfice-risque de l’immunothérapie à distance de l’épisode de myocardite.

Early studies suggest that radiofrequency-based renal denervation reduces blood pressure in patients with moderate hypertension. We investigated whether an alternative technology using endovascular ultrasound renal denervation reduces ambulatory blood pressure in patients with hypertension in the absence of antihypertensive medications.

15. Plasma NT-proBNP mirrors the deleterious cardiovascular and renal continuum in hypertension

Abstract

Background The aims of this study were (a) to test the ability of N-terminal pro-brain natriuretic peptide (NT-proBNP) to detect subclinical target organ damage (TOD) denoted by left ventricular hypertrophy (LVH), aortic stiffness or renal damage and (b) to test its reproducibility in two different conditions in an ancillary study.Methods The study included 837 patients (50.9% men) with hypertension aged 50 ± 24 years with a median 24-h ambulatory blood pressure (BP) of 148/90 mmHg. LVH was assessed by transthoracic echocardiography and echocardiography, aortic stiffness was assessed by carotid–femoral pulse wave (PWV) measurements and renal dysfunction by measurements of the estimated glomerular filtration rate (eGFR) and microalbuminuria.Results After the exclusion of patients with a history of heart failure, NT-proBNP was independently correlated with sex, systolic BP, primary hypertension, PWV, LVH and eGFR, but not with microalbuminuria. The median (interquartile range) NT-proBNP increased gradually according to the number of target organs damaged: 42 (24–70), 77 (39–151), 141 (81–250) and 334 (177–556) pg/mL, for damage to 0, 1, 2 and 3 target organs, respectively (p< 0.001). . For the same number of target organs damaged, NT-proBNP was higher in women and for secondary hypertension. A threshold at 90 pg/mL for men and 142 pg/mL in women had a specificity of 95% to detect at least one TOD (areas under ROC curve 0.790 and 0.783, respectively). The reproducibility of NT-proBNP was fairly good in this setting (r= 0.952, p< 0.001, N= 325)Conclusion This study demonstrates that NT-proBNP mirrors the harmful effect of high BP on TOD. NT-proBNP could be used as an integrative tool for risk stratification in hypertension.

Context:Recurrent somatic mutations in KCNJ5, CACNA1D, ATP1A1, and ATP2B3have been identified in aldosterone-producing adenomas (APAs). The question as to whether they are responsible for both nodulation and aldosterone production is not solved.Case Description:We describe the case of a young patient who was diagnosed with severe arterial hypertension due to primary aldosteronism at age 26 years, followed by hemorrhagic stroke 4 years later. Abdominal computed tomography showed bilateral macronodular adrenal hyperplasia. Identification of lateralized aldosterone secretion led to right adrenalectomy, followed by normalization of biochemical and hormonal parameters and amelioration of blood pressure. The resected adrenal showed three nodules, one of them expressing aldosterone synthase and harboring a somatic KNCJ5mutation. A Weiss revisited index of 3 of the APA prompted us to perform a second 18F-2-fluoro-2-deoxy-D-glucose-positron emission tomography after surgery, which revealed abnormal rectal activity despite the absence of clinical symptoms. Gastrointestinal exploration showed multiple polyps with severe dysplasia, and the diagnosis of familial adenomatous polyposis was established in the presence of a germline heterozygous APCgene mutation. Sequencing of somatic DNA from the APA and a second adrenal nodule revealed biallelic APCinactivation due to loss of heterozygosity in both nodules.Conclusions:This case report underlines the need for establishing the frequency of germline APCvariants in patients with primary aldosteronism and bilateral macronodular adrenal hyperplasia because their presence may predispose to APA development and severe hypertension well before the first familial adenomatous polyposis symptoms appear. From a mechanistic point of view, it supports a two-hit model for APA development, whereby the first hit drives increased cell proliferation whereas the second hit specifies the pattern of hormonal secretion.

AbstractBackground/Aims:Urinary albumin excretion is subject to intra-individual variability. Thus, for research purposes, it is recommended to test three urine samples collected over a short period of time. The objective of our analysis was to check the usefulness of triplicate samples to determine the albuminuric status of diabetic patients. Methods:We present the results of the non-planned retrospective analysis of 246 triplicate morning urine samples obtained from 95 type 2 diabetics included in three multinational, randomized, double-blind studies. Albuminuria was determined by immunoturbidimetry on fresh samples in the same central laboratory. Microalbuminuria was defined by a urine albumin to creatinine ratio (UACR) between 2.5 and 25 mg/mmol in males and between 3.5 and 35 mg/mmol in females. Concordance was obtained when the second and/or third sample (UACR2 and UACR3) confirmed the albuminuric status obtained from the first sample (UACR1). Results:Considering the first samples, 9 were within the normal range, 35 showed microalbuminuria and 56 showed macroalbuminuria. The overall concordance rate was 95. The log of UACR was highly correlated between samples. Bland-Altman plots expressed in percent variations between two samples confirmed that the mean variation was low (around 8) but revealed the scattering of values, 95 being between -60 and +77 of variation between samples. Conclusions:There is no benefit in repeating morning UACR determination in diabetic patients to accurately categorize a subject as having normo-, micro- or macroalbuminuria. Klemm’s pioneering contributions, leads among other things to the discovery of the effect now called the ‘Chemla effect’, after the late Professor Marius Chemla. These studies of ionic motions in melts, and liquids in general, have greatly benefitted from recent advances in molecular simulations. Some recent results of such simulations - molecular dynamics (MD) and Brownian dynamics (BD) - as well as of related theoretical work are discussed.

Abstract

In the frame of the preparation of refractory metals, thermodynamics and kinetics properties of salts of refractory metals (mainly titanium, and niobium) dissolved in alkali chloride melts were determined by transient electrochemical techniques. Two kinds of reactions occur in the melts: homogeneous reactions such as: Me(n+z)++ ne−⇌ Me+where Me(n+z)+and Mez+are two oxidation states of metal ions; heterogeneous reactions corresponding to the metal deposition: Men++ ne−⇌ Me. The potential corresponding to the reactions are linked to the Gibbs free energy of formation of salt solutions. The formal standard potentials of the various redox reactions were measured by transient electrochemical techniques. These techniques were also used to determine the reaction kinetics and to measure the diffusion coefficients of the active ions which control the deposition process. In pure chloride melts, the metal is obtained from the reduction of lower oxidation states, Ti2+and Nb3+. However, disproportionation reactions and the formation of insoluble subhalides perturb the reduction reaction. When a small amount of NaF was added to the bath, the direct reduction of Ti3+or Nb4+to metal was obtained, which greatly improved the quality of the deposit.

32. Effect of abdominal aortic grafts on aortic stiffness and central hemodynamics

Abstract

Graft-prosthesis and stentgraft placements are effective modalities for treating abdominal aortic aneurysm, but related changes in arterial stiffness are not well established. The present study sought to assess aortic stiffness after aneurism repair by measuring pulse wave velocity (PWV). The graft-related variation of carotid–femoral PWV was compared with that of carotid–radial PWV, the latter being unaffected by vascular treatment. The secondary objective was to evaluate potential differences between graft-prosthesis and stentgraft in terms of aortic stiffness and augmentation index, a composite indicator integrating wave reflexion. Fifty patients were included (39 had a graft-prosthesis and 11 had a stentgraft). In the whole group and after a median postoperative follow-up of 47 days, carotid–femoral PWV increased by 1.0 m/s [−12.3, 10.3], while carotid–radial PWV slightly decreased by −0.3 m/s [−4.4; 3.5] (P0.001). The effect of the type of prosthesis on the PWV was not significant. Nevertheless, the augmentation index increased after stentgraft implantation (4% [−10; 17]) and decreased after graft-prosthesis placement (−8.5% [−47; 17]) (P< 0.01). This difference was not explained by a heart rate or a treatment effect and was likely attributable to the prosthesis per se. This study demonstrates the impact of aortic grafts on aortic stiffness. Besides, it suggests that stentgraft increases reflected waves more than graft-prostheses. These changes of vascular properties may influence the outcomes after surgery.

33. A rapid flow cytometry test based on histone H2AX phosphorylation for the sensitive and specific diagnosis of ataxia telangiectasia

Abstract

Ataxia telangiectasia AT is a progressive neurodegenerative disease with onset in early childhood, caused by mutations in the ATMataxiatelangiectasia mutated gene. Diagnosis relies on laboratory tests showing high levels of serum alphafetoprotein, cell sensitivity to ionizing radiation IR and absence or reduced levels of ATM protein. Many tests, however, are not sufficiently sensitive or specific for AT, have long turnaround times, or require large blood samples. This prompted us to develop a new flow cytometry method for the diagnosis of AT based on the measurement of histone H2AX phosphorylation. We established normal ranges of histone H2AX phosphorylation after 2 Gy IR by testing Tcell lines, lymphoblastoid cell lines LCLs andor peripheral blood mononuclear cells PBMCs or both from 20 genetically proven AT and 46 control donors. To further evaluate the specificity and sensitivity of the test, we analyzed cells from 19 patients suspected of having AT, and from one Friedreich Ataxia, one Ataxia with Oculomotor Apraxia type 2, and one Nijmegen Breakage Syndrome patients. Phosphorylated histone H2AX mean fluorescence intensity of irradiated AT cells was significantly lower than that of healthy donors. The intrastaining, intraassay, and interassay imprecisions were ≤13.22. Sensitivity and specificity were virtually 100 when the test was performed on PBMCs. Screening of 19 consecutive new patients with suspected AT classified 15 patients as nonAT and four as AT; diagnosis of the latter four was subsequently confirmed by DNA sequencing to identify ATMmutations. The Friedreich Ataxia patient, the Ataxia with Oculomotor Apraxia type 2 patient and the Nijmegen Breakage Syndrome patient were classified as nonAT. This flow cytometry test is very sensitive, specific and rapid, and requires only 2 ml of blood. It may thus be proposed for the early differential diagnosis of AT as an alternative to methods requiring the production of LCLs. © 2008 International Society for Advancement of Cytometry

34. Formation of Ni-Zr Alloys in Molten Fluorides

Abstract

The electrochemical behavior of Zr4+ was investigated in LiF-NaF between 923 and 1123 K. With an inert tungsten or molybdenum electrode, a well-defined oxydo-reduction wave was observed during cyclic voltammetry experiments. The reduction of Zr4+ is a single four-electron step and quasi-reversible, and the process is diffusion-controlled. Chronopotentiometry was used to measure the diffusion coefficient of Zr4+ ions. For instance, a value 1.54 x 10-5 cm2 s-1 was found at of at 979 K. The reduction of Zr4+ ions was also studied using nickel electrode. In that case, Ni- Zr alloys were formed as pointed out on cyclic voltammograms which exhibit several characteristic peaks. SEM and EDX analyses have revealed the presence of a multi-layered Ni-Zr alloy deposited film on Ni electrode. At -1.65 V vs Ni/Ni2+, three successive layers were pointed out, corresponding to three different Ni-Zr alloy compositions.

39. Intranodal injection of semimature monocyte-derived dendritic cells induces T helper type 1 responses to protein neoantigen

Abstract

Dendritic cells (DCs) represent the most potent antigen-presenting cells of the immune system capable of initiating primary immune responses to neoantigens. Here we characterize the primary CD4 T-cell immune response to protein keyhole limpet hemocyanin (KLH) in 5 metastatic melanoma patients undergoing a tumor peptide–based dendritic cell vaccination trial. Monocyte-derived dendritic cells displaying a semimature phenotype, as defined by surface markers, were loaded ex vivo with antigen and injected intranodally at weekly intervals for 4 weeks. All patients developed a strong and long-lasting delayed-type hypersensitivity reactivity to KLH, which correlated with the induction of KLH-dependent proliferation of CD4 T cells in vitro. Secondary in vitro stimulation with KLH showed significant increase in interferon-γ and interleukin-2 (IL-2) but not IL-4, IL-5, nor IL-10 secretion by bulk T cells. On the single-cell level, most TH1 cells among in vitro–generated KLH-specific T-cell lines confirmed the preferential induction of a KLH-specific type 1 T helper immune response. Furthermore, the induction of KLH-specific antibodies of the IgG2 subtype may reflect the induction of a type 1 cytokine profile in vivo after vaccination. Our results indicate that intranodal vaccination with semimature DCs can prime strong, long-lasting CD4 T-cell responses with a TH1-type cytokine profile in cancer patients. (Blood. 2003;102:36-42)

Estimating the risk entailed by classical risk factors like blood pressure (BP) or serum cholesterol may be difficult because of their variability and the often unknown duration of exposure. Having variables integrating the impact of those classical risk factors on the cardiovascular system would probably aid the prediction of cardiovascular events. The present study aimed at determining whether cardiac baroreflex sensitivity (BRS), correlates with several risk factors and thus is a good candidate for being such an integrative variable. As a comparison, left ventricular mass (LVM), pulse wave velocity (PWV), and creatinine were also tested for association with risk factors.

Abstract

Although there has been extensive analysis on the capacity of MHC-peptide tetramers to bind antigen-specific TCR, there have been comparatively few studies regarding the role of the CD4 and CD8 co-receptors in binding and activation by these multimeric molecules. Here, we start from the observation that different antibodies against human CD8 exert opposite effects on MHC-peptide tetramer binding to the TCR: tetramer staining was enhanced by OKT8 antibody, while it was blocked with SK1 antibody. We used these different anti-CD8 antibodies to modulate CD8 function during tetramer staining of Melan-A/MART1-specific CTL clones. We show that CD8 action could be variably modulated during all the phases of interaction, indicating that CD8 participates in both the initial association of the TCR with MHC-peptide tetramers and the stability of this interaction. While the blocking effect of anti-CD8 antibodies was mostly exerted during the initial binding of the TCR with MHC-peptide tetramers, the enhancing effect was exerted by augmenting the duration of this interaction. Blocking anti-CD8 antibodies were also capable of preventing tetramer-mediated T cell activation. The possibility of variably affecting MHC-peptide tetramer binding and T cell activation using anti-CD8 antibodies confirms the critical role exerted by the CD8 co-receptor in this interaction and supports the notion that TCR engagement by MHC-peptide ligands typically involves CD8.

While tumor-associated antigen (TAA)-specific CD8+ T lymphocytes have been detected in metastatic melanoma patients, immune response in early disease phases has not yet been carefully evaluated. We looked for circulating cytotoxic T lymphocytes (CTL) directed against Melan-A / MART1, tyrosinase, gp100 and MAGE-3 antigens in patients with a diagnosis of primary cutaneous melanoma by using fluorescent HLA-A2 tetramers. In five out of six cases high numbers of CD8+ / tetramer+ cells could be detected by flow cytometry, and in four patients lymphocyte populations specific for two different melanoma antigens (Melan-A / MART1 and tyrosinase) were contemporarily present. The TAA-specific cells could represent as much as 1 / 220 T lymphocytes in the circulating CD8+ population. When tetramers were used to monitor the in vitro expansion of TAA-specific CTL precursors upon antigen-specific stimulation, a diverse expansion potential was evidenced in CTL from the different donors and, more strikingly, in CTL specific for the different TAA. Melan-A / MART1-specific CTL clones derived from two patients exhibited a broad range of avidity. Only the highest avidity clones, representing about 50 % of the cases analyzed, were tumor specific. By correlating tetramer staining with clone avidity, we found that tetramer fluorescence intensity could represent a good indicator of TCR affinity, but not of overall clone avidity.

Abstract

Accumulating evidence indicates that peripheral lymphocyte variants with altered antigen receptor expression may be capable of expressing recombination-activating genes (RAG). We and others recently observed functional RAG gene products in mature T cells with defective TCR expression (MacMahan and Fink, Immunity 1998. 9: 637 – 647; Lantelme et al., J. Immunol., 2000. 164: 3455 – 3459). Here, the association between TCR expression and RAG activity was assessed further in lymphocytes from patients with defective responses to DNA damage. We show that T cells with altered TCR surface expression are present in increased numbers in these patients and that they express RAG genes. The finding of RAG gene expression by TCR variants suggests the possibility that secondary V(D)J rearrangements could be induced in these cells to rescue their defective phenotype and cellular function. Moreover, as V(D)J recombination has been implicated in chromosome translocations involving antigen receptor genes, we discuss a possible relationship between altered TCR expression, RAG activity and the frequent lymphoma-specific translocations observed in these patients.

50. Niobium plating processes in alkali chloride melts

Abstract

Niobium deposits were prepared from alkali chloride melts on nickel and AISI316 stainless steel substrates both by constant current and by pulse current methods. The influence of electrolysis conditions on the nature, morphology and purity of the deposits was investigated by X-ray diffraction, scanning electron microscopy and energy dispersive X-ray analysis. No metallic niobium was obtained at temperatures below 500 ∘C. At temperatures between 550 and 650 ∘C, the deposits were dendritic and non-adherent, whereas pure niobium layers could be obtained at 750 ∘C. Detailed analysis showed that a large negative overpotential during the pulse current period lead to the presence of suboxides, such as Nb6O, in the metallic phase. Suitable electrolysis conditions gave pure oxygen-free niobium. Cross section analysis showed that on nickel a thin layer of niobium–nickel alloy such as NbNi3, was formed at the metal interface. In contrast no alloys were detected at the niobium-stainless steel interface, where homogenous adherent layers of thickness around 50 μm were obtained.
