2. Mechanistic Insights into Hyaluronic Acid Induced Peptide Nanofiber Organization in Supramolecular Hydrogels

- Abstract
Composite hydrogels composed of low-molecular-weight peptide self-assemblies and polysaccharides are gaining great interest as new types of biomaterials. Interactions between polysaccharides and peptide self-assemblies are well reported, but a molecular picture of their impact on the resulting material is still missing. Using the phosphorylated tripeptide precursor Fmoc-FFpY (Fmoc, fluorenylmethyloxycarbonyl; F, phenylalanine; Y, tyrosine; p, phosphate group), we investigated how hyaluronic acid (HA) influences the enzyme-assisted self-assembly of Fmoc-FFY generated in situ in the presence of alkaline phosphatase (AP). In the absence of HA, Fmoc-FFY peptides are known to self-assemble in nanometer thick and micrometer long fibers. The presence of HA leads to the spontaneous formation of bundles of several micrometers thickness. Using fluorescence recovery after photobleaching (FRAP), we find that in the bundles both (i) HA colocalizes with the peptide self-assemblies and (ii) its presence in the bundles is highly dynamic. The attractive interaction between negatively charged peptide fibers and negatively charged HA chains is explained through molecular dynamic simulations that show the existence of hydrogen bonds. Whereas the Fmoc-FFY peptide self-assembly itself is not affected by the presence of HA, this polysaccharide organizes the peptide nanofibers in a nematic phase visible by small-angle X-ray scattering (SAXS). The mean distance dbetween the nanofibers decreases by increasing the HA concentration c, but remains always larger than the diameter of the peptide nanofibers, indicating that they do not interact directly with each other. At a high enough HA concentration, the nematic organization transforms into an ordered 2D hexagonal columnar phase with a nanofiber distance dof 117 Å. Depletion interaction generated by the polysaccharides can explain the experimental power law variation d∼c−1/4and is responsible for the bundle formation and organization. Such behavior is thus suggested for the first time on nano-objects using polymers partially adsorbing on self-assembled peptide nanofibers.

- Abstract
ObjectiveThe initial data of the International Study on Acute Coronary Syndromes - ST Elevation Myocardial Infarction COVID-19 showed in Europe a remarkable reduction in primary percutaneous coronary intervention procedures and higher in-hospital mortality during the initial phase of the pandemic as compared with the prepandemic period. The aim of the current study was to provide the final results of the registry, subsequently extended outside Europe with a larger inclusion period (up to June 2020) and longer follow-up (up to 30 days).MethodsThis is a retrospective multicentre registry in 109 high-volume primary percutaneous coronary intervention (PPCI) centres from Europe, Latin America, South-East Asia and North Africa, enrolling 16 674 patients with ST segment elevation myocardial infarction (STEMI) undergoing PPPCI in March/June 2019 and 2020. The main study outcomes were the incidence of PPCI, delayed treatment (ischaemia time >12 hours and door-to-balloon >30 min), in-hospital and 30-day mortality.ResultsIn 2020, during the pandemic, there was a significant reduction in PPCI as compared with 2019 (incidence rate ratio 0.843, 95% CI 0.825 to 0.861, p<0.0001). This reduction was significantly associated with age, being higher in older adults (>75 years) (p=0.015), and was not related to the peak of cases or deaths due to COVID-19. The heterogeneity among centres was high (p<0.001). Furthermore, the pandemic was associated with a significant increase in door-to-balloon time (40 (25–70) min vs 40 (25–64) min, p=0.01) and total ischaemia time (225 (135–410) min vs 196 (120–355) min, p<0.001), which may have contributed to the higher in-hospital (6.5% vs 5.3%, p<0.001) and 30-day (8% vs 6.5%, p=0.001) mortality observed during the pandemic.ConclusionPercutaneous revascularisation for STEMI was significantly affected by the COVID-19 pandemic, with a 16% reduction in PPCI procedures, especially among older patients (about 20%), and longer delays to treatment, which may have contributed to the increased in-hospital and 30-day mortality during the pandemic.Trial registration numberNCT04412655.

6. The coming together of allosteric and phosphorylation mechanisms in the molecular integration of A2A heteroreceptor complexes in the dorsal and ventral striatal-pallidal GABA neurons

The role of adenosine A2A receptor (A2AR) and striatal-enriched protein tyrosine phosphatase (STEP) interactions in the striatal-pallidal GABA neurons was recently discussed in relation to A2AR overexpression and cocaine-induced increases of brain adenosine levels. As to phosphorylation, combined activation of A2AR and metabotropic glutamate receptor 5 (mGluR5) in the striatal-pallidal GABA neurons appears necessary for phosphorylation of the GluA1 unit of the AMPA receptor to take place. Robert Yasuda (J Neurochem 152: 270–272, 2020) focused on finding a general mechanism by which STEP activation is enhanced by increased A2AR transmission in striatal-pallidal GABA neurons expressing A2AR and dopamine D2 receptor. In his Editorial, he summarized in a clear way the significant effects of A2AR activation on STEP in the dorsal striatal-pallidal GABA neurons which involves a rise of intracellular levels of calcium causing STEP activation through its dephosphorylation. However, the presence of the A2AR in an A2AR-fibroblast growth factor receptor 1 (FGFR1) heteroreceptor complex can be required in the dorsal striatal-pallidal GABA neurons for the STEP activation. Furthermore, Won et al. (Proc Natl Acad Sci USA 116: 8028–8037, 2019) found in mass spectrometry experiments that the STEP splice variant STEP61can bind to mGluR5 and inactivate it. In addition, A2AR overexpression can lead to increased formation of A2AR-mGluR5 heterocomplexes in ventral striatal-pallidal GABA neurons. It involves enhanced facilitatory allosteric interactions leading to increased Gq-mediated mGluR5 signaling activating STEP. The involvement of both A2AR and STEP in the actions of cocaine on synaptic downregulation was also demonstrated. The enhancement of mGluR5 protomer activity by the A2AR protomer in A2AR-mGluR5 heterocomplexes in the nucleus accumbens shell appears to have a novel significant role in STEP mechanisms by both enhancing the activation of STEP and being a target for STEP61.

7. Synthesis and characterization of chemically fueled supramolecular materials driven by carbodiimide-based fuels

- Abstract
Many supramolecular materials in biological systems are driven to a nonequilibrium state by the irreversible consumption of high-energy molecules such as ATP or GTP. As a result, they exhibit unique dynamic properties such as a tunable lifetime, adaptivity or the ability to self-heal. In contrast, synthetic counterparts that exist in or close to equilibrium are controlled by thermodynamic parameters and therefore lack these dynamic properties. To mimic biological materials more closely, synthetic self-assembling systems have been developed that are driven out of equilibrium by chemical reactions. This protocol describes the synthesis and characterization of such an assembly, which is driven by carbodiimide fuels. Depending on the amount of chemical fuel added to the material, its lifetime can be tuned. In the first step, the protocol details the synthesis and purification of the peptide-based precursors for the fuel-driven assemblies by solid-phase peptide synthesis. Then, we explain how to analyze the kinetic response of the precursors to a carbodiimide-based chemical fuel by HPLC and kinetic models. Finally, we detail how to study the emerging assembly’s macro- and microscopic properties by time-lapse photography, UV-visible spectroscopy, shear rheology, confocal laser scanning microscopy and electron microscopy. The procedure is described using the example of a colloid-forming precursor Fmoc-E-OH and a fiber-forming precursor Fmoc-AAD-OH to emphasize the differences in characterization depending on the type of assembly. The characterization of a precursor’s transient assembly can be done within 5 d. The synthesis and purification of a peptide precursor requires 2 d of work.

8. Regulating Chemically Fueled Peptide Assemblies by Molecular Design

- Abstract
In living systems, fuel-driven assembly is ubiquitous, and examples include the formation of microtubules or actin bundles. These structures have inspired researchers to develop synthetic counterparts, leading to exciting new behaviors in man-made structures. However, most of these examples are serendipitous discoveries because clear design rules do not yet exist. In this work, we show design rules to drive peptide self-assembly regulated by a fuel-driven reaction cycle. We demonstrate that, by altering the ratio of attractive to repulsive interactions between peptides, the behavior can be toggled between no assembly, fuel-driven dissipative self-assembly, and a state in which the system is permanently assembled. These rules can be generalized for other peptide sequences. In addition, our finding is explained in the context of the energy landscapes of self-assembly. We anticipate that our design rules can further aid the field and help the development of autonomous materials with life-like properties.

9. Supramolecular Hydrogel Induced by Electrostatic Interactions between Polycation and Phosphorylated-Fmoc-Tripeptide

Supramolecular hydrogels formed through noncovalent interactions of low-molecular-weight hydrogelators (LMWHs) show great potential applications in different fields, such as delivery of therapeutics, injectable biomaterials, catalysis, or materials chemistry. Generally, the self-assembly of LMWHs is triggered by a sol–gel process through an external stimulus able to switch their solubility, such as temperature, pH, or solvent change and chemical or enzymatic reactions. In this work, we introduced a new strategy to trigger and control the self-assembly of Fmoc-FFpY peptides by direct electrostatic interactions with a polycation without dephosphorylation of the peptides. The resulting hydrogels show enhanced mechanical properties in comparison to gels of Fmoc-FFpY induced by enzymatic dephosphorylation. Peptide self-assembly yields β-sheets, revealed by circular dichroism and infrared spectroscopy. Characteristic distances predicted by geometry optimization in the gas phase are in agreement with X-ray scattering data and transmission electron microscopy observations. It is proposed that core–shell cylinders are formed in which polycation chains decorate the micellar structures of Fmoc-FFpY peptides through electrostatic interactions between the charged amine groups of the polycations and the phosphate groups of the peptides. Because the gels form quickly and have superior mechanical properties, applications as injectable biomaterials are foreseen. This work opens a route toward a new class of self-assembled hydrogels, where Fmoc tripeptides can be self-assembled with specific polycations to obtain, for example, antimicrobial hydrogels.

10. Long‐Term Stability of Tilt‐Controlled gPhoneX Gravimeters

- Abstract
Spring relative gravimeters are considered too unstable to provide useful information on long‐term gravity variations. In this paper, we prove that the new generation of spring gravimeter gPhoneX can reach long‐term stability at the μGal level (10 nm s−2) when the verticality of the gravimeter is maintained, if the instrumental drift can be correctly estimated. We conducted two comparisons with different gPhoneXs in different observatories and environmental conditions. In the “Walferdange Underground Laboratory for Geodynamics” in Luxembourg, we compared time series from the gPhoneX (with and without tilt control), with data from a superconducting gravimeter. We found an agreement at the μGal level when the tilt control is switched on. We validated this result by repeating the experiment at the “Geodesy in Karstic Environment” observatory in the south of France. The fit between the superconducting gravimeter and the gPhoneX hourly values gives similar results at all frequencies over 276 days of measurements. The linear correlation coefficient between the gPhoneX and superconducting gravimeter reaches 0.99, with a misfit of 6.0 nm s−2. We demonstrated that tilt‐controlled gPhoneXs are suitable for long‐term gravity monitoring. gPhoneXs are suitable for long‐term gravity monitoring when the tilts are actively controlled

11. Phase Separation in Supramolecular Hydrogels Based on Peptide Self-Assembly from Enzyme-Coated Nanoparticles

Spatial localization of biocatalysts, such as enzymes, has recently proven to be an effective process to direct supramolecular self-assemblies in a spatiotemporal way. In this work, silica nanoparticles (NPs) functionalized covalently by alkaline phosphatase (NPs@AP) induce the localized growth of self-assembled peptide nanofibers from NPs by dephosphorylation of Fmoc–FFpY peptides (Fmoc: fluorenylmethyloxycarbonyl; F: phenylalanine; Y: tyrosine; p: phosphate group). The fibrillary nanoarchitecture around NPs@AP underpins a homogeneous hydrogel, which unexpectedly undergoes a macroscopic shape change over time. This macroscopic change is due to a phase separation leading to a dense phase (in NPs and nanofibers) in the center of the vial and surrounded by a dilute one, which still contains NPs and peptide self-assemblies. We thus hypothesize that the phase separation is not a syneresis process. Such a change is only observed when the enzymes are localized on the NPs. The dense phase contracts with time until reaching a constant volume after several days. For a given phosphorylated peptide concentration, the dense phase contracts faster when the NPs@AP concentration is increased. For a given NPs@AP concentration, it condenses faster when the peptide concentration increases. We hypothesize that the appearance of a dense phase is not only due to attractive interactions between NPs@AP but also to the strong interactions of self-assembled peptide nanofibers with the enzymes, covalently fixed on the NPs.

Peptide mimicry employing a combination of aza-amino acyl proline and indolizidinone residues has been used to develop allosteric modulators of the prostaglandin F2α receptor. The systematic study of the N-terminal phenylacetyl moiety and the conformation and side chain functions of the central turn dipeptide residue has demonstrated the sensitive relationships between modulator activity and topology. Examination of aza-Gly-Pro and aza-Phe-Pro analogs 2aand 2bin a murine preterm labor model featuring treatment with lipopolysaccharide demonstrated their capacity to extend significantly (>20 h) the average time of delivery offering new prototypes for delaying premature birth.

15. Present‐Day Deformations of the Jura Arc Inferred by GPS Surveying and Earthquake Focal Mechanisms

This study presents a compilation of more than 40 years of seismotectonic data, including 54 computed focal mechanisms, combined with 15 years of GPS data coming from a dense network of 35 permanent GPS stations within the Jura arc and its vicinity. These data are compared to previous available geomorphological, geophysical, and structural studies in order to discuss the 3D distribution of the deformation within the Jura arc. GPS data show coherent schemes in terms of velocities and allowed to discriminate between two provinces (NE of the belt and in its front/foreland). They also constrain a low but significant overall strain tensor with a NNW‐SSE shortening of 2.16 nanostrain/year associated with an ENE‐WSW extension of 0.44 nanostrain/year. The seismotectonic approach is based on a data set of 2,400 events and 54 focal mechanisms. Inversions of the focal mechanisms both globally and in homogeneous sectors highlight a general strike‐slip deformation regime, with sigma1 oriented NW‐SE and sigma3 oriented NE‐SW. - Abstract
In life, molecular architectures, like the cytoskeletal proteins or the nucleolus, catalyze the conversion of chemical fuels to perform their functions. For example, tubulin catalyzes the hydrolysis of GTP to form a dynamic cytoskeletal network. In contrast, myosin uses the energy obtained by catalyzing the hydrolysis of ATP to exert forces. Artificial examples of such beautiful architectures are scarce partly because synthetic chemically fueled reaction cycles are relatively rare. Here, we introduce a new chemical reaction cycle driven by the hydration of a carbodiimide. Unlike other carbodiimide-fueled reaction cycles, the proposed cycle forms a transient 5(4H)-oxazolone. The reaction cycle is efficient in forming the transient product and is robust to operate under a wide range of fuel inputs, pH, and temperatures. The versatility of the precursors is vast, and we demonstrate several molecular designs that yield chemically fueled droplets, fibers, and crystals. We anticipate that the reaction cycle can offer a range of other assemblies and, due to its versatility, can also be incorporated into molecular motors and machines.

- Abstract
A substantial part of the operating costs of public transport is attributable to drivers, whose efficient use therefore is important. The compilation of optimal work packages is difficult, being NP-hard. In practice, algorithmic advances and enhanced computing power have led to significant progress in achieving better schedules. However, differences in labor practices among modes of transport and operating companies make production of a truly general system with acceptable performance a difficult proposition. TRACS II has overcome these difficulties, being used with success by a substantial number of bus and train operators. Many theoretical aspects of the system have been published previously. This paper shows for the first time how theory and practice have been brought together, explaining the many features which have been added to the algorithmic kernel to provide a user-friendly and adaptable system designed to provide maximum flexibility in practice. We discuss the extent to which users have been involved in system development, leading to many practical successes, and we summarize some recent achievements.

Abstract: Microstructures, phases, and phase transformations in Ti-6Al-4V alloy specimens containing 0, 10, 20, and 30 at. pct hydrogen were investigated using optical microscopy (OM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and microhardness testing. Alloying with hydrogen was achieved by holding the specimens in a pure hydrogen atmosphere of different pressures at 780 C for 24 hours. The phases present in the temperature range of 20 C to 1000 C were determined by microstructural characterization of the specimens quenched from different temperatures. Increasing the hydrogen addition from 0 to 30 at. pct lowered the beta-transus temperature of the alloy from 1005 C to 815 C, significantly slowed down the kinetics of the beta-to-alpha transformation, and led to formation of an orthorhombic martensite instead of the hexagonal martensite found in quenched specimens containing 0 pct H. A hydride phase was detected in specimens containing 20 and 30 at. pct hydrogen. The time-temperature-transformation (TTT) diagrams for beta-phase decomposition were determined at different hydrogen concentrations. The nose temperature for the beginning of the transformation decreased from 725 C to 580 C, and the nose time increased from 12 seconds to 42 minutes when the hydrogen concentration was increased from 0 to 30 at. pct.

The effect of the solvent on the ground state free energy differences of three enol conformers of 2-(2-hydroxyphenyl)benzimidazole and its keto tautomer has been examined by means of Monte Carlo simulations and continuum model calculations. In agreement with the experimental data, calculations show that the trans enol and keto forms are stabilized by polar solvents, leading to a conformational and tautomeric equilibrium with the closed cis enol conformer in water, the only single species in apolar solvents. Monte Carlo simulations have also been used to examine the influence of the solvent on the absorption band of the closed cis enol structure and the fluorescence band of the keto form generated by photoinduced intramolecular proton transfer. In concordance with the experimental spectra, absorption and fluorescence band maxima for the closed cis enol and keto forms, respectively, are found to be blue-shifted with increasing polarity and hydrogen bonding capacity of the solvent.

The intramolecular proton transfer of 2-(2-hydroxyvinyl)benzimidazole (HVBI) and 2-(2-hydroxyphenyl)imidazole (HPI) in the ground state and in the 1ππ*, 1nπ*, and 3ππ* excited states has been studied at the HF/CIS/D95** level of theory. Their rotamerism reaction in the ground and 1ππ* excited states has been also analyzed. These systems are two different fragments of 2-(2-hydroxyphenyl)benzimidazole (HPBI), containing an intramolecular hydrogen bond through a common NCCCO backbone. The comparison of the calculations on HVBI and HPI with the experimental results available for HPBI and the theoretical calculations done for HPBI, salicyaldimine, and 1-amino-3-propenal allow us to determine the influence that each functional group of HPBI has on its intramolecular proton transfer in different electronic states. It is found that the aromaticity of the phenol ring of HPBI exerts a great influence on the proton transfer in the ground state and the lowest-lying 1ππ*, and 3ππ* excited states. The aromatic character of the phenol ring explains the higher stability of the enol form with respect to the keto form in the ground state, while a change in its aromaticity is responsible for the shift in the relative stability of the two tautomeric forms in the 1ππ*, and 3ππ* excited states. The presence of the imidazole moiety stabilizes the keto form in the 1nπ* excited state, exerting a significant influence on the proton transfer in this state.

In most cases the most successful applications will marry titanium and steel together in a hybrid fashion. The aim of this study was to use the scientific literature to identify interdisciplinary interventions for rehabilitation during the perioperative period for cancer patients who underwent total laryngectomy. We systematically researched controlled descriptors: laryngectomy, patient care team/education, patient care team/manpower, patient care team/methods, patient care team/utilization and rehabilitation. We performed a qualitative narrative synthesis and identified 549 articles. Of these, 113 were duplicates, 398 were excluded during the analysis of the title and abstract, 1 was excluded for unfeasibility of access, and 4 were excluded after reading the article in full, resulting in 33 included articles. The articles addressed different types of interdisciplinary actions, such as vocal, olfactory, pulmonary and alimentary rehabilitation; comparisons of prosthetic devices; and descriptions of practices for total laryngectomized patient rehabilitation. Although the interventions found in the literature were effective in the rehabilitation of the total laryngectomized patient, their interdisciplinarity was not evidenced but was highlighted in these studies as a factor for improvement in terms of practical assistance and quality of life.

Although initially attributed to increased pulmonary vascular resistance (group 1 PH of the WHO classification), this entity was classified in 2009 in group 5 PH (unknown mechanism and miscellaneous PH). 49. Freedom from equine infectious anaemia virus infection in Spanish Purebred horses

No cases of equine infectious anaemia (EIA) have been reported in Spain since 1983. Factors that could increase the risk of reintroducing equine infectious anaemia virus (EIAV) into Spain include the recent occurrence of the disease in Europe and the absence of compulsory serological testing before importation into Spain. Given the importance of the Spanish Purebred (SP) horse breeding industry in Spain, the aim of this cross-sectional study was to provide evidence of freedom from EIAV in SP stud farms in Central Spain. Serum samples from 555 SP horses, collected between September 2011 and November 2013, were tested using a commercially available EIAV ELISA with a published sensitivity of 100 per cent. All 555 samples were negative for antibody to EIAV, providing evidence of a true EIAV seroprevalence between 0 per cent and 0.53 per cent (95% CIs of the sensitivity and specificity of the ELISA technique used Q10 were 100 per cent and 99.3 per cent, respectively) among the SP breeding population in Central Spain. These findings should serve to increase confidence when exporting SP horses to other countries.
