In this paper, we propose a unified size-structured PDE model for the growth of metastatic tumors, which extends a well-known coupled ODE-PDE dynamical model developed and studied in the literature. A treatment model based on the proposed unified PDE model is investigated via optimal control theory, where its first-order necessary optimality system characterizing the optimal control is derived. We prove that the uniqueness of the optimal control depends on the chosen objective functional, and the optimal control is of bang-bang type when it is unique. For obtaining its efficient numerical solutions, a projection gradient descent algorithm based on the characteristic scheme is developed for solving the established optimal treatment model. Several numerical examples are provided to validate our mathematical analysis and numerical algorithm, and also illustrate the biologically interesting treatment outcomes of different models and control strategies. Our simple model reveals that: (i) only the total drug dosage matters if one just cares about the final treatment output; (ii) given the same total drug dosage, the optimal bang-bang treatment plan outperforms the others in the sense that it maximally reduces the total tumor sizes during the whole period of treatment, although their final tumor sizes are the same. Adverse Drug Reactions (ADRs) resulting from drug combinations can endanger patients' health and life. In this paper, we propose ADGCL, a novel A daptive D ual G raph C ontrastive L earning based on heterogeneous signed networks for predicting ADRs. Specifically, ADGCL first explicitly models positive and negative drug-drug interactions (DDIs) using a heterogeneous signed network to learn semantic-rich drug feature representations. Additionally, ADGCL employs a dual graph contrastive learning strategy including self-supervised contrastive learning and micro-supervised learning to learn high-level features, which significantly improves prediction performance. Furthermore, ADGCL incorporates an adaptive negative sampling method to generate negative samples. Lastly, an encoder based on the implicit graph neural network is designed to capture long-range dependencies in underlying networks, thereby improving the quality of drug feature representation. Since Drug-drug interactions (DDIs) can cause adverse effects when patients take two or more drugs and therefore increase health care costs, the extraction of DDIs is an important research area in patient safety. To improve the performance of Drug–drug interaction extraction (DDIE), we present a novel two-stage method in this paper. It first identifies the positive instances using a feature based binary classifier, and then a Long Short Term Memory (LSTM) based classifier is used to classify the positive instances into specific category. The experimental results show that the two-stage method has many advantages over one-stage ones, and among the factors related to LSTM, we find that the two layer bidirectional LSTM embedded with word, distance and Part-of-Speech obtains the highest F-score of 69.0%, which is state-of-the-art.
