Chickpea (Cicer arietinum), the 2nd most important legume after dry beans, known for its protein content, is an important crop agronomically as well as economically. The present study aimed to determine the effect of foliar application of spermidine (spd) on various aspects of chickpea genotypes under salt stress. The chickpea genotypes were cultivated under 4 and 8 dSm-1 Cl- dominated salinity followed by the spermidine application of 0.5 and 1.0 mM. Salinity stress inhibited the plant height and root length, reducing plant biomass (fresh and dry weight). Furthermore, it also decreased the chlorophyll and nitrogen content of the plant. Results obtained from spermidine (spd) application indicated that both concentrations of spermidine are good in enhancing the tolerance in chickpea genotypes against salt stress. Spermidine application increased the plant's height as well as biomass. It also enhanced the chlorophyll content (32.93%), increasing the Nitrogen Balance Index (77.37%) at 1.0 mM.It further increased the flavonoid and anthocyanin content in plant leaf. In addition, the application of spermidine retained more moisture (25.81%) and increased seed fiber (13.30%) in all chickpea genotypes at 1.0 mM. It reduces the Cl- ion accumulation and maintains the ionic balance in chickpea seeds. The effect of spermidine application (0.5 and 1.0mM) was more pronounced, but 1.0 mM had a more positive effect in salt-sensitive chickpea genotypes.

Gummy supplements are an exceptional medium for delivering pharmaceuticals and nutritional bioactives, especially for children. Interestingly, they are increasingly finding favor among adults as effective bioactive delivery systems. This study aimed to identify and select natural vegan components for creating and developing gummy prototypes. The gelling agent was agar-agar, sourced from red algae, and combined with carrageenan to enhance compressibility. Phycocyanin from Spirulina and anthocyanin from purple cabbage were used as colorants to provide a range of natural hues. The study resulted in five distinct formulations: Blueberry Anthocyanin Vitamin D (BAD), Blueberry Anthocyanin Vitamin C (BAC), Mango Antacid Anthocyanin (MAA), Mango Flavored Phycocyanin (MFP), and Kiwi Flavored Spirulina (KFS) gummies. Notably, the MAA gummy exhibited suboptimal texture quality during preliminary assessments and was consequently excluded from further analysis.Across the formulations, no visible color changes were noted upon room temperature storage for a week. The BAD gummy displayed heightened adhesiveness and springiness in texture, while the KFS gummy demonstrated superior cohesiveness, chewiness, and gumminess in its fresh state.Interestingly, the BAD gummy exhibited the highest adhesion even in the one-week-old samples.The outcome of the 9-point hedonic sensory evaluation unveiled that the cyan shade, derived from the phycocyanin pigment of Spirulina, was the most preferred color among the gummies. Regarding overall likability, the MFP and BAC formulations garnered the highest responses for being 'liked extremely'. The uniqueness of the present work lies in its meticulous exploration and selection of natural vegan ingredients to craft gummy formulations with diverse bioactive and sensory attributes.

Water kefir is widely consumed all over the world due to its potential health benefits. The aim of this current study was to compare non-fermented juice and fermented beverage of water kefir produced from Aronia melanocarpa juice and pomace in terms of chemical, physical and sensory quality as well as valorisation of pomace in the production of water kefir. When compared to water kefir made with aronia juice, less reduction in total phenolic content (TPC), total flavonoid content (TFC) and total anthocyanin content (TAC) was observed in samples made with aronia pomace during the fermentation process. Similarly, greater antioxidant activity was demonstrated in water kefir made with aronia pomace than juice. Based on sensory evaluation, no difference was found in overall acceptability, taste, aroma/odor, and turbidity of water kefir made with aronia pomace before and after fermentation. Results indicated that aronia pomace has potential in water kefir production.

Flavonoids are the most abundant class of secondary metabolites that are ubiquitously involved in plant development and resistance to biotic and abiotic stresses. Flavonoid biosynthesis involves multiple channels of orchestrated molecular regulatory factors. Methyl jasmonate (MeJA) has been demonstrated to enhance flavonoid accumulation in numerous plant species; however, the underlying molecular mechanism of MeJA-induced flavonoid biosynthesis in safflower is still not evident. In the present study, we revealed the underlying molecular basis of a putative F3'5'H gene from safflower imparting MeJA-induced flavonoid accumulation in transgenic plants. The constitutive expression of the CtF3'5'H1 gene was validated at different flowering stages, indicating their diverse transcriptional regulation through flower development in safflower. Similarly, the CtF3'5'H1-overexpressed Arabidopsis plants exhibit a higher expression level, with significantly increased anthocyanins and flavonoid content, but less proanthocyanidins than wild-type plants. In addition, transgenic plants treated with exogenous MeJA revealed the up-regulation of CtF3'5'H1 expression over different time points with significantly enhanced anthocyanin and flavonoid content as confirmed by HPLC analysis. Moreover, CtF3'5'H1- overexpressed Arabidopsis plants under methyl violet and UV-B irradiation also indicated significant increase in the expression level of CtF3'5'H1 with improved anthocyanin and flavonoid content, respectively. Noticeably, the virus-induced gene silencing (VIGS) assay of CtF3'5'H1 in safflower leaves also confirmed reduced anthocyanin accumulation. However, the CtF3'5'H1 suppression in safflower leaves under MeJA elicitation demonstrated significant increase in total flavonoid content. Together, our findings confirmed that CtF3'5'H1 is likely mediating methyl jasmonate-induced flavonoid biosynthesis in transgenic plants via enhanced anthocyanin accumulation.

Current climate conditions indicate that California’s viticultural regions will experiencemore frequent heat events, thus leading to greater water stress resulting in lower quality fruit in grapevines. One strategy used to help mitigate the negative effects is irrigating throughout these events to help preserve berry composition. In order to evaluate the usefulness of irrigation throughout heat events, Vitis vinifera cv. Cabernet Sauvignon vines in an established vineyard were exposed to three differential irrigation treatments. In 2020, the baseline treatment was under water deficit (60% ET), while the 2x baseline (120% ET) and 3x baseline (180% ET) treatments had double and triple the irrigation of the baseline, respectively. The irrigation treatments were adjusted in 2021 to see if less water could be used without negatively impacting the phenolics results. The treatments were 60% ET, 90% ET, and 120% ET. Differential irrigation started one to two days prior to a heat wave (HW) and continued until the last day of the HW. In this study, a HW was defined as three consecutive days with maximum temperatures at or above 38°C. Throughout the 2020 and 2021 growing seasons, there were major declines in anthocyanin concentration for the 60% ET treatment, which suggests a suppression of anthocyanin synthesis and promotion of degradation at such high temperatures. A common trend seen throughout both years is that the 180% and 120% treatments from 2020 and 2021 respectively, resulted in lower phenolic concentrations than the 120% and 90% treatments. This is important to note as these results suggest that irrigation prior to heatwaves can be beneficial in maintaining fruit quality, but excessive watering may negate the beneficial aspects of deficit irrigation. Transcriptomic analyses identified the down-regulation of genes involved in the phenylpropanoid and flavonoid pathways, which provides a potential explanation at the molecular level for the lower phenolic concentrations

Barley grains are a rich source of compounds, such as resistant starch, beta-glucans and anthocyanins, that can be explored in order to develop various products to support human health, while lignocellulose in straw can be optimised for feed in husbandry, bioconversion into bioethanol or as a starting material for new compounds. Existing natural variations of these compounds can be used to breed improved cultivars or integrated with a large number of mutant lines. The technical demands can be in opposition depending on barley’s end use as feed or food or as a source of biofuel. For example beta-glucans are beneficial in human diets but can lead to issues in brewing and poultry feed. Barley breeders have taken action to integrate new technologies, such as induced mutations, transgenics, marker-assisted selection, genomic selection, site-directed mutagenesis and lastly machine learning, in order to improve quality traits. Although only a limited number of cultivars with new quality traits have so far reached the market, research has provided valuable knowledge and inspiration for future design and a combination of methodologies to achieve the desired traits. The changes in climate is expected to affect the quality of the harvested grain and it is already a challenge to mitigate the unpredictable seasonal and annual variations in temperature and precipitation under elevated [CO2] by breeding. This paper presents the mutants and encoded proteins, with a particular focus on anthocyanins and lignocellulose, that have been identified and characterised in detail and can provide inspiration for continued breeding to achieve desired grain and straw qualities.

Pepper is one of the most important vegetable species in Serbia. The use of pepper fruits is very often determined by their type. Tomato-shaped pepper is a fruit type suitable for the pickle processing industry because of its thick pericarp. In this study, several lines of light-yellow tomato-shaped peppers were evaluated in the open field with the aim to develop new variety with appropriate fruit characteristics and without anthocyanin colouration. Eight of selected lines originated from two crosses while two lines originated from variety Novosađanka. Fruit weight, fruit length, fruit width, fruit index, pericarp thickness, number of locules, and Brix were measured in technological maturity. According to the obtained data, lines 17 and 24 will be used for further selection process. Line 17 was chosen because of the highest pericarp thickness and high fruit weight and line 24 because of the high content of total soluble solids., Paprika je jedna od najvažnijih povrtarskih vrsta u Srbiji. Upotreba plodova paprike je vrlo često određena tipom ploda. Paradajz paprika se dosta koristi za industrijsku preradu zbog debelog perikarpa, a i kao svež plod u salatama. Cilj ovog istraživanja bio je da se ispita nekoliko linija paradajz paprika svetložute boje koje su gajene na otvorenom polju. U tehnološkoj zrelosti mereni su: masa ploda, dužina ploda, širina ploda, indeks ploda, debljina perikarpa, broj komora i suva materija. Prema dobijenim podacima, za dalji proces selekcije koristiće se linije 17 i 24. Linija 17 zbog najveće debljine perikarpa i mase ploda, a linija 24 zbog većeg sadržaja suve materije.

Multiplex 330 (MX) is a portable, non-invasive fluorescence sensor that provides different multiparametric indices that are sensitive to the concentration of chlorophylls, flavonols, and anthocyanins on the leaf surface. This study investigated the use of these indices to assess the variability on free volatile composition of grapes (Vitis vinifera L.) in the field as well as other components of their quality potential in vineyards affected by iron deficiency chlorosis (IDC). Twenty non-irrigated Tempranillo/110 Richter vineyard subzones from non-affected to moderately affected by IDC were monitored in Ribera del Duero area (Spain) during two seasons. The results indicated that MX can characterize the spatial variation of leaf pigment concentrations, agronomic performance, and grape quality in vineyards affected by IDC. The MX indices measured at the leaf level close to harvest had better predictive values for the concentrations of free alcohols, volatile acids, C13-norisoprenoids, esters and acetates in the must than vine vigor, size or degree of technological maturity of the grapes. Our study demonstrates that the MX indices to estimate leaf pigment concentrations can be helpful to assess the technological maturity and free volatile composition of wine grapes in vineyards affected by IDC; nevertheless, the efficacy of the indexes may vary according to the year season.

It is important to look at the physicochemical qualities of Portulaca, a weed species used as a vegetable and a herb for medical and therapeutic purposes. India has a wide range of variations in this species' morphology and nutraceutical value. This study aimed to evaluate the physicochemical properties of various purslane accessions from different regions of Tamil Nadu. A total of 15 purslane accessions (PA 1, PA 2, PA 3, PA 4, PA 5, PA 6, PA 7, PA 8, PA 9, PA 10, PA 11, PA 12, PA 13, PA 14 and PA 15) were collected and evaluated. Physical traits like colour of the leaves and stems were quantified as hue angle and chroma value, which showed a degree of variation. Estimates suggested that phytochemical properties related the hue and chroma of leaf and stem to the pigments in plants. Among the accessions, PA 3 has recorded the highest phytochemical properties viz., leaf total chlorophyll content 1.43±0.16 mg g-1, leaf total carotenoid content 0.24±0.03 mg g-1, stem total chlorophyll content 0.49±0.05 mg g-1, stem total carotenoid content 0.12±0.01 mg g-1 and total anthocyanin content 19.25±1.54 µg g-1. The multiple regression model suggested that the values can predict the estimated values. The evaluation of physicochemical properties along with the regression model helps in the breeding programme to select the traits; phytochemical analysis proved the ample supply of chlorophylls, carotenoids and anthocyanins, so these wild species could be a cheap source to alleviate several diseases.

The current study aims to gather information about the color-related characteristics and the monomeric phenolic composition of carbonic maceration (CM) wines. The study was conducted over two vintages, with two different grape varieties. Color-related general parameters and wine monomeric phenolic composition were determined in both free-run and press CM fractions and were compared to conventional winemaking (CW). Overall, wines made by CM had lower phenolic content and color intensity. The analysis of wine detailed phenolic composition showed that CM wines were characterized by low contents of anthocyanins and flavonols. In contrast, free-run wine obtained by CM had the greatest concentration of flavanols and hydroxycinnamic acids, probably due to the presence of stems during the fermentation. In conclusion, vinification by CM leads to important differences in wine phenolic composition and, as consequence, on wine color properties. These differences could play an important role in wine sensorial properties and wine aging potential.

Magenta lilly pilly (Syzygium paniculatum) is an Australian native tree that produces berry fruits that are rich in phytochemicals reportedly beneficial to human health. Here we explored the biological activities of polyphenol-enriched extracts from the magenta lilly pilly fruit, benchmarking it against traditional sources including purple sweet potato and blackberry. We show that the extracts exert potent antioxidant and neuroprotective properties as well as antimicrobial activity against Staphylococcus aureus. The phenolic composition of lilly pilly was investigated using liquid chromatography coupled to mass spectrometry (HPLC-DAD-MS), revealing anthocyanins to be the primary component in high abundance compared to traditional anthocyanin-containing plants. Three anthocyanins from lilly pilly, along with their glycosylation patterns and stability, were characterised. Altogether, our results demonstrate the potential to exploit magenta lilly pilly fruits as a high-yielding source of phenolics with beneficial biological properties of potential interest for multiple downstream applications., QC 20230522

Diabetes is a common disorder due to metabolism, more or less people of cultural groups in the region globally. This disease is caused due to the lifelong distress and has no cure as of now. Naturopathy / Plant based chemical antibiotics are proven safe than the synthetic chemicals that are artificially synthesized. In this study, the total anthocyanin content and their antimicrobial activity was estimated for the samples. The study revealed the anti-diabetic properties of anthocyanin from several sources such as strawberries (Fragaria ananassa), cherries (Prunus avium), pomegranate (Punica granatum), grapes (Vitis vinifera), flame of forest flowers (Canna indica), hibiscus flowers (Hibiscus rosa-sinensis) and guava leaves (Psidium guajava). Extraction has been done by using ethanol as a base and inhibition studies have been conducted. The source ‘Canna indica’ showed huge amount of anthocyanin content of 814.6 mg/100g and other sources showed varied amount of anthocyanin content. Prunus avium fruits showed maximum antidiabetic activity. The total phenolic and anthocyanin contents with its antidiabetic activity for different sources using acidified ethanol were studied.

Borax detection can be done using the flame test method, volumetric titration, spectrophotometric analysis, and qualitatively using natural materials such as curcumin and anthocyanins. The high content of anthocyanins in grapes has the potential to be utilized in acid-base titrations as an indicator of natural acid-base. The research objective was to determine the potential of anthocyanins in the flesh and skin of grapes in detecting natural borax which is absorbed on filter paper, Whatmann paper no. 41 and Whatmann paper no. 42. Then it was tested on samples of meatballs from 5 sellers in Samarinda Seberang to Mangkupalas village. The research method used is a quantitative analysis of the color changes that occur. Based on the research results, filter paper can have a better absorption that is shown with a dark color on Whatmann No. 41 and Whatmann No.42. Grape skin extract can detect borax more clearly than flesh by forming a dark purple ring when reacting with borax. In testing using meatball samples, the overall results showed that the samples contained borax. It was concluded that the filter paper from the absorption of grape skin anthocyanins could be used as an alternative in quantitatively testing the borax content in food., Deteksi boraks dapat dilakukan dengan menggunakan metode uji nyala api, titrasi volumetrik, analisis spektrofotometri serta secara kualitatif menggunakan bahan alam seperti kurkumin dan antosianin. Kandungan antosianin di dalam anggur yang cukup tinggi memiliki potensi pemanfaatan dalam titrasi asam basa sebagai indikator asam basa alami. Ekstrak kulit anggur dapat mendeteksi boraks lebih jelas dibandingkan dengan buah anggurnya dengan terbentuknya cincin berwarna ungu tua jika direaksikan dengan boraks. Pada pengujian menggunakan sampel pentol bakso dari 5 penjual, diperoleh hasil keseluruhan sampel menggandung boraks. Disimpulkan bahwa kertas saring dari absorbsi antosianin kulit anggur dapat digunakan sebagai alternatif dalam menguji kandungan boraks pada makanan secara kuantitatif.

Red fleshed apples (Malus × domestica Borkh.) differ in colour intensity between cultivars, seasons and sites. The objective of this study was to develop a procedure for predicting anthocyanin content from digital images of flesh discs. Flesh cylinders of uniform colour were excised, scanned and their colours determined in the R, G and B and the L*a*b* colour spaces. Anthocyanin content was also quantified chemically. A calibration line was constructed to predict anthocyanin content of flesh discs of varying colour from a scan or a photograph in the studio or outdoors. Anthocyanin concentration was linearly related to the logarithms of G, B and L*. From these relationships, the anthocyanin content of a flesh disc was predicted, pixel by pixel. Colour corrections were applied using a reference colour chart included in all images. The Finlayson algorithm was most effective for correcting the G parameter obtained by a flatbed scanner. For variable imaging methods (scanning or photography), the Vandermonde algorithm for correcting the L* parameter and the Finlayson algorithm for correcting the G parameter were most effective in predicting anthocyanin content. The procedure allows accurate prediction of anthocyanin content of red fleshed apples from simple colour scans or photographs.

Lingonberries contain high contents of bioactive compounds such as chlorogenic acids and anthocyanins. In addition to radical scavenging and antioxidant activities, these compounds can protect cells from DNA damage. For this reason, lingonberries might be well suited for nutraceuticals or natural biomedicines. To assess these applications, the present study characterized and identified the most effective extract, only consisting of anthocyanins, copigments or a mixture of both, obtained from a lingonberry juice concentrate. An extract was generated by using a XAD-7 column followed by fractionation into anthocyanins and copigments using adsorptive membrane chromatography. After identification of main polyphenols by HPLC–photodiode array–electrospray ionization–tandem mass spectrometry, free radical scavenging activity was analyzed by electron spin resonance spectroscopy using 2,2-diphenyl-1-picrylhydrazyl and galvinoxyl radicals. Furthermore, cyclic voltammetry analyses and the Trolox equivalent antioxidant capacity (TEAC) assay were applied. Finally, the reactive oxygen species (ROS) reducing effects of the lingonberry extract and its fractions were evaluated in HepG2 cells. While the combination of anthocyanins and copigments possessed the highest antioxidant activities, all samples (XAD-7 extract, anthocyanin and copigment fraction) protected cells from oxidative stress. Thus, synergistic effects between phenolic compounds may be responsible for the high antioxidant potential of lingonberries, enabling their use as nutraceuticals.

The aim of this study was to examine the influence of shoot age on the biological and chemical properties of 13 black currant cultivars with different origins and ripening times. Pheno-logical observations together with examined pomological and chemical characteristics were studied in two consecutive years at the experimental field near Belgrade, Serbia. The total content of phenols was estimated spectrophotometrically by the Folin-Ciocalteu method, while quantitative analysis of anthocyanin and flavonols aglycones was performed using a high-performance liquid chromato-graphic (HPLC) method. Principal component analysis was performed to establish differences in biological and chemical properties of black currants. Three-year-old shoots had an earlier start of all examined phenological stages, better generative potential, higher yields, while clusters and berries from 2-year-old shoots had significantly higher values for physical properties, total phenols, anthocyanin and flavanols aglycones and antiradical capacity. Late ripening cultivars had higher contents of all chemical compounds. The berries on 2-year-old shoots had total phenolics that ranged between 123.0 (‘Titania’) and 298.3 mg/100 g fresh weight (FW) (‘Ometa’), while total antho-cyanins ranged between 398.5 (’Ojebyn’) and 1160.8 mg/kg FW (’Ometa’). The variety Prokupac is the dominant variety in the vineyards of Southern Serbia, which produces quality wines of characteristic and unique tastes. In the agroecological conditions of the Prokuplje vine district, the influence of manual defoliation on the phenolic profile of the wine produced from the variety Prokupac was examined. Four experimental treatments with different timings of manual defoliation were applied: early defoliation—treatment I, early defoliation— treatment II, late defoliation—treatment III and the control. The phenolic profile of the wine was determined for the three treatments of defoliation and the control treatment. Additionally, a multivariate analysis was applied on the obtained results, together with already published data (grape seeds and skins phenolic profiles). Identification and quantification of the phenolic compounds was performed using ultra-high-performance liquid chromatography (UHPLC) with an ultraviolet multi-diode detector (DAD) and mass detector with three analyzers—triple quadrupole (QQQ). Based on the obtained results, it was determined that there are significant differences between the experimental treatments in the content of individual polyphenols, total polyphenols and the antioxidant capacity. Twenty (20) phenolic compounds were identified in the wine samples of the experimental treatments. Defoliation significantly affected the variations of the contents of phenolic acids and flavonoids. In treatment III, the highest content of gallic acid was obtained, while the treatments with early defoliation did not differ in relation to the control sample. Early defoliation in treatments I and II had an effect on the phenolic composition of the wine by favoring the accumulation of flavonol, while the content of hydroxycinnamic acid and total anthocyanins (TAC) was higher in treatment III. The construction of powerful cell factories requires intensive genetic engineering for the addition of new functionalities and the remodeling of native pathways and processes. The present study demonstrates the feasibility of extensive genome reprogramming using modular, specialized de novo-assembled neochromosomes in yeast. The in vivo assembly of linear and circular neochromosomes, carrying 20 native and 21 heterologous genes, enabled the first de novo production in a microbial cell factory of anthocyanins, plant compounds with a broad range of pharmacological properties. Turned into exclusive expression platforms for heterologous and essential metabolic routes, the neochromosomes mimic native chromosomes regarding mitotic and genetic stability, copy number, harmlessness for the host and editability by CRISPR/Cas9. Sterility of hybrids produced from interspecific hybridization in lilies (Lilium, Liliaceae) is a great limitation in the breeding program, especially for Lilium longiflorum, which only has white-flowered cultivars. Because modification of flower colour in L. longiflorum by conventional breeding is limited by pre- and post-fertilization barriers, we think genetic modification could be used as an alternative in the future. For this, we need to understand what determines white colouration in L. longiflorum and other species and identify the molecular mechanisms regulating flower colour. In this study, we determined the accumulation of anthocyanins and related compounds in flower tissues during flower developmental stages in L. longiflorum cultivar ‘Lincoln’ and in the Oriental hybrid lily cultivars ‘Rialto’, ‘Perth’ and ‘Gran Tourismo’, respectively with white, pink and red flowers. Furthermore, the presence/absence and the expression of eight structural genes (CHSa, CHSb, CHIa, CHIb, F3H, F3’H, DFR, ANS) and three transcription factor genes (MYB12, MYB15, bHLH2) in flower tissues were investigated. Two structural genes (LlLinF3’H and LlLinDFR) and one transcription factor gene (LlLinbHLH2) were not detected in ‘Lincoln’ flowers. In ‘Rialto’, an amino acid substitution in the R2 repeat of LhRiaMYB12 which was previously reported to be responsible for the white flower colour is also found in the LhPerMYB12 of the pink ‘Perth’ flowers. Moreover, LhRiaDFR is present but not expressed in ‘Rialto’ flowers. Accumulation of cyanidin was observed in the flowers of ‘Perth’ and ‘Gran Tourismo’. High amounts of dihydrokaempferol accumulated in flowers of all four lily cultivars confirming the expression and functionality of early structural genes in the pathway. The elevated expression of the structural genes is strongly correlated with the expression of LhMYB12 and LhMYB15. Breast cancer is the second most deadly diagnosed lifestyle disease among women. Surgery and chemotherapy are the current treatments of choice; nevertheless, toxicity connected with this underscores the urgency of the demand for the human-friendly drug. 50% of current synthetic drugs available commercially today are either direct or indirect descendants extracted from herbs. Anthocyanins possess many pharmacological activities, including anticancer potential. However, no study on anticancer activity of anthocyanins from Cordyline australis has been reported. Anthocyanins were extracted from fresh leaves using ethanol as solvent. The total anthocyanin was quantified and fractionated by Ultra Performance Liquid Chromatography. Cytotoxicitic effect was carried on diverse cancer cell lines like MCF 7, HCT-116, Caco-2 and SW480 using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide assay. Based on MTT data, MCF 7 cells were further analyzed by LDH assay, Glutathione-S- transferase (GST), Quercetin reductase, Cytochrome P450 and Caspase 3, 8 and 9 activities. The obtained results were analyzed using ANOVA with a level of significance. Results obtained from MTT assay revealed that the anthocyanin extract carried significant toxic (p < 0.05) specificity against MCF 7 cells (65 ± 2.1 toxicity at 50 µg/ml) when compared to the other onco cells. Remarkable LDH leakage (50.2% vs 50 µg/ml), GST (3.0±0.002 U/mg protein), QR (4.4±0.054 U/mg protein), Cyt P450 activities (0.291±0.01 U/mg protein) were noticed. Caspase 3 (157%), 8 (142%) and 9 (147%) displayed profound activities. These in vitro findings of specific anticancer effects noticed on C. australis anthocyanin extract require further evaluation using animal models. Finally, the obtained findings open up the possibility of developing a lead antimetastatic anthocyanin candidate against deadly breast cancer.

These indicators can be formed by immobilizing anthocyanin into edible film and then changing its color to pH changes. The research aims to determine the immobilization time, test the color stability of the edible film indicator to temperature changes, determine the characterizations of the edible film indicator based on the %relative standard deviation (%RSD) value, and apply the edible film indicators as coconut milk freshness indicators. The best immobilization time was in the 90th minute with %RSD=0%. The edible film indicators did not change color from 10 to 50°C with %RSD=0%. The characterizations of edible film indicators on pH 6, 7, 8, and 10 had response times of 6.68 minutes; 9.49 minutes; 3.57 minutes; and 4.37 minutes; periods of 14 days, 13 days, 14 days, and 16 days; and good reproducibility at pH 6 and 10 and not good at pH 7 and 8. Main conclusionTranscriptome and biochemical analyses suggested that, while suppression of multiple flavonoids and anthocyanins occurs at least partially at the transcriptional level, increased biosynthesis of non-jasmonate phyto-oxylipins is likely controlled non-transcriptionally. Methyl jasmonate (MeJA) produced in plants can mediate their response to environmental stresses. Exogenous application of MeJA has also shown to activate signaling pathways and induce phytoalexin accumulation in many plant species. To understand how pomegranate plants respond biochemically to environmental stresses, metabolite analysis was conducted in pomegranate leaves subjected to MeJA application and revealed unique changes in hydrolyzable tannins, flavonoids, and phyto-oxylipins. Additionally, transcriptome and real-time qPCR analyses of mock- and MeJA-treated pomegranate leaves identified differentially expressed metabolic genes and transcription factors that are potentially involved in the control of hydrolyzable tannin, flavonoid, and phyto-oxylipin pathways. Molecular, biochemical, and bioinformatic characterization of the only lipoxygenase with sustained, MeJA-induced expression showed that it is capable of oxidizing polyunsaturated fatty acids, though not located in the subcellular compartment where non-jasmonate (non-JA) phyto-oxylipins were produced. These results collectively suggested that while the broad suppression of flavonoids and anthocyanins is at least partially controlled at the transcriptional level, the induced biosynthesis of non-JA phyto-oxylipins is likely not regulated transcriptionally. Overall, a better understanding of how pomegranate leaves respond to environmental stresses will not only promote plant health and productivity, but also have an impact on human health as fruits produced by pomegranate plants are a rich source of nutritional compounds.

In this study we have analyzed the anthocyanin composition of skin extracts of three red grape varieties Prokupac, Evita and Čokot Zemun in order to distinguish these cultivars based on their anthocyanin profile. Also, mechanical analysis of grape bunches and berries was performed. According to our results, seventeen anthocyanins were identified using LC‐MS technique and quantitative differences were recorded using HPLC‐DAD method. The highest content of total anthocyanins was obtained for Evita variety and the lowest one was recorded in Prokupac. Also, clear differences were observed in anthocyanins ratios. In comparison to Prokupac and Evita varieties, Čokot Zemun was characterized with a high content of cumaroyl derivatives of anthocyanin compounds, while high levels of acetylated derivatives were recorded in Prokupac. Data reported in this study represent a certain contribution to a database of mechanical properties and chemical composition of grape varieties originating from Balkan.

Diabetes mellitus—in particular, Type 2 diabetes mellitus (T2DM)—is one of the most prevalent chronic illnesses in many countries. Diabetes mellitus is regarded as an independent risk factor for cardiovascular disease (CVD). Cardiovascular disease is one of the main causes of mortality in patients with diabetes, mainly due to macrovascular complications. One of these macrovascular complications in diabetes is atherosclerosis, which involves a complicated pathophysiological process. In addition to hyperglycaemia, oxidative stress (OS) plays a significant role in the pathogenesis of diabetes and its associated risk of CVD. Platelet hyperactivity, in the presence of OS, has a major effect on the progression of atherosclerosis and thrombotic events. Aspirin (AS) is the most-used antiplatelet therapy for the prevention of thrombotic complications in people with T2DM. AS attenuates platelet hyperactivity. Although aspirin is a frequently used therapy for the inhibition of platelet hyperactivity, there is much evidence for AS non-responsiveness. The anthocyanin (AC) antioxidant has been shown to have an inhibitory effect on platelets and consequently may be used as a complement to other antiplatelet therapies to attenuate the negative effect of atherosclerosis and CVD in people with T2DM. Although many dietary intervention studies have shown that intake of AC-rich food may be negatively related to some CVD risk factors, the effect of pure AC on thrombotic markers such as platelet hyperactivity and haemostasis is yet to be explored. The main aim of the studies commenced for this thesis were to examine the effect of AC on thrombotic parameters and reveal the pathways by which AC might affect platelet hyperactivity, thereby providing individuals with T2DM with better protection against CVD. The aim of the first study (see Chapter 4) was to evaluate the in vitro effect of AC on platelet activation and aggregation. Cluster zone leaf removal is a well-established viticulture practice for improving cluster microclimate and wine quality in cooler climates, while its efficacy in warmer conditions is less is known. Here we compared the effect of early (ELR, after fruit set; diameter of berries 3-5 mm) and late (LLR, beginning of veraison) leaf removal on berry composition and wine phenolic profile of grapevine (Vitis vinifera L.) variety Cabernet Sauvignon grown in a temperate, warmer region of Eastern Serbia. Compared to the control (no leaf removal), both leaf removal treatments increased the sugar content in fresh juice and alcohol concentration in wine. Over three consecutive years (2011-2013) markedly different in temperature and rainfall, ELR was clearly most effective in decreasing weights of cluster and of one berry, and in increasing of skin share in a berry. The content of total phenols, tartaric acid esters, anthocyanins, and flavanols in berry skin and wine was the highest in ELR treatment. ELR prominently modified the phenolic profile: Increasing flavanols, myricetin and quercetine in skin and wine, and anthocyanins, peonidin-3-glucoside in skin and delphinidin-3-glucoside in wine. This work demonstrated that early leaf removal positively influenced the chemical composition of berries and wine of Cabernet Sauvignon and might be recommended for practice in the temperate warm conditions.

Abscisic acid (ABA) is a key signaling molecule promoting ripening of non-climacteric fruits such as sweet cherry (Prunus avium L.). To shed light on the role of other hormones on fruit development, ripening and anthocyanin production, the synthetic auxin 1-naphthaleneacetic acid (NAA) was applied to sweet cherry trees during the straw-color stage of fruit development. NAA-treated fruits exhibited higher concentrations of 1-aminocyclopropane-1-carboxylic acid (ACC) and ABA-glucose ester (ABA-GE), which are a precursor of ethylene and a primary storage form of ABA, respectively. Consistent with these observations, transcript levels of genes encoding ACC synthase and ACC oxidase, both involved in ethylene biosynthesis, were increased after 6 days of NAA treatment, and both ABA concentration and expression of the regulator gene of ABA biosynthesis (NCED1 encoding 9-cis-epoxycarotenoid dioxygenase) were highest during early fruit ripening. In addition, transcript levels of key anthocyanin regulatory, biosynthetic and transport genes were significantly upregulated upon fruit exposure to NAA. This was accompanied by an increased anthocyanin concentration and fruit weight whilst fruit firmness and cracking index decreased. Altogether our data suggest that NAA treatment alters ethylene production, which in turn induces ripening in sweet cherry and enhanced anthocyanin production, possibly through ABA metabolism. Recommendations towards increased consumption of fresh fruit and vegetables are well supported by epidemiological and clinical trials. However, in some specific cases, it is difficult to follow these recommendations and the use of nutraceuticals or, in the present work, a freeze-dried fruits mixture can be recommended in order to afford the optimal consumption of dietary polyphenols naturally present in fruits and vegetables. In this work we have carefully characterized a red-berry mixture in terms of polyphenol composition, encountering mainly anthocyanins, which account for a total of 2.8 mg/g as cyanidin-3-glucoside equivalents. Additionally, we have assayed the red-berry blend in a cell model of neurological damage by differentiating the cells and measuring the effect of red-berry polyphenols on cell viability and redox state by flow cytometry. The berry-fruit extract showed an inhibitory effect on differentiated SH-SY5Y ROS formation at a concentration as low as 250 µg/mL (33% inhibition). The results show the potential of this berry-fruit blend for its nutraceutical use in the prevention of the neurodegeneration associated with age or environmental agents.

Full Text, Thesis (PhD Doctorate), Doctor of Philosophy (PhD), School of Medical Science, Griffith Health, Diabetes mellitus—in particular, Type 2 diabetes mellitus (T2DM)—is one of the most prevalent chronic illnesses in many countries. Diabetes mellitus is regarded as an independent risk factor for cardiovascular disease (CVD). Cardiovascular disease is one of the main causes of mortality in patients with diabetes, mainly due to macrovascular complications. One of these macrovascular complications in diabetes is atherosclerosis, which involves a complicated pathophysiological process. In addition to hyperglycaemia, oxidative stress (OS) plays a significant role in the pathogenesis of diabetes and its associated risk of CVD. Platelet hyperactivity, in the presence of OS, has a major effect on the progression of atherosclerosis and thrombotic events. Aspirin (AS) is the most-used antiplatelet therapy for the prevention of thrombotic complications in people with T2DM. AS attenuates platelet hyperactivity. Although aspirin is a frequently used therapy for the inhibition of platelet hyperactivity, there is much evidence for AS non-responsiveness. The anthocyanin (AC) antioxidant has been shown to have an inhibitory effect on platelets and consequently may be used as a complement to other antiplatelet therapies to attenuate the negative effect of atherosclerosis and CVD in people with T2DM. Although many dietary intervention studies have shown that intake of AC-rich food may be negatively related to some CVD risk factors, the effect of pure AC on thrombotic markers such as platelet hyperactivity and haemostasis is yet to be explored. The main aim of the studies commenced for this thesis were to examine the effect of AC on thrombotic parameters and reveal the pathways by which AC might affect platelet hyperactivity, thereby providing individuals with T2DM with better protection against CVD. Table grape quality is of importance for consumers and thus for producers. Its objective quality is usually determined by destructive methods mainly based on sugar content. This study proposed to evaluate the possibility of hyperspectral imaging to characterize table grapes quality through its sugar (TSS), total flavonoid (TF), and total anthocyanin (TA) contents. Different data pretreatments (WD, SNV, and 1st and 2nd derivative) and different methods were tested to get the best prediction models: PLS with full spectra and then Multiple Linear Regression (MLR) were realized after selecting the optimal wavelengths thanks to the regression coefficients (coefficients) and the Variable Importance in Projection (VIP) scores. All models were good at showing that hyperspectral imaging is a relevant method to predict sugar, total flavonoid, and total anthocyanin contents. The best predictions were obtained from optimal wavelength selection based on coefficients for TSS and from VIPs optimal wavelength windows using SNV pre-treatment for total flavonoid and total anthocyanin content. Thus, good prediction models were proposed in order to characterize grapes while reducing the data sets and limit the data storage to enable an industrial use.

Spontaneous somatic mutations produce changes in berry skin color in grapevine. VviMybA1 and VviMybA2 transcription factors induce anthocyanin biosynthesis pathway through the regulation of UDP-glucose: flavonoid 3-O-glucosyltransferase (UFGT) expression. The lack of berry color in white-skinned cultivars is caused by an insertion of Gret1 retro-transposable element in the VviMybA1 promoter region preventing gene expression, and a two-nucleotide deletion in VviMybA2 coding sequence producing a null frameshift (Walker et al., 2007). Nevertheless, occasionally, anthocyanin pigmentation capacity is partially recovered from white-skinned cultivars that then produce red berries (Azuma et al., 2009). Here we have studied the cases of Albariño and Verdejo red-berried somatic variants appearing spontaneously in the field. We used transcriptomics and targeted UPLC-QqQ-MS/MS metabolomics to reveal the molecular cause of color recovery in these somatic variant berries at véraison stage. RNA-sequencing analysis identified 39 and 225 genes that were differentially expressed in the berry skin of Albariño and Verdejo red-berried variants compared to their respective white-berried ancestors (FDR<0.05 and >2 fold-change). In both colored mutants, flavonoid biosynthesis pathway was activated, including the upregulation of UFGT, flavonone-3- hydroxylase, and chalcone synthase genes. The anthocyanins cyanidin-3-glucoside, cyanidin-3-(6-coum)-glucoside and cyanidin-3-(6-acetyl)-glucoside, were detected in the berry skin of both red Albariño and Verdejo, respectively. Additionally, Albariño red berries also contained detectable amounts of delphinidin-3-glucoside and peonidin-3- glucoside, and of cyanidin-3-arabinoside at trace levels explaining the more intense red color observed in this variant. In this study, we have analyzed the anthocyanin composition of skin extracts of three red grape varieties Prokupac, Evita and Čokot Zemun in order to distinguish these cultivars based on their anthocyanin profile. Also, mechanical analysis of grape bunches and berries was performed. According to our results, seventeen anthocyanins were identified using LC/MS technique and quantitative differences were recorded using HPLC-DAD method. The highest content of total anthocyanins was obtained for Evita variety and the lowest one was recorded in Prokupac. Also, clear differences were observed in anthocyanins ratios. In comparison to Prokupac and Evita varieties, Čokot Zemun was characterized with a high content of coumaroyl derivatives of anthocyanin compounds, while high levels of acetylated derivatives were recorded in Prokupac. Data reported in this study represent a certain contribution to a database of mechanical properties and chemical composition of grape varieties originating from Balkan.

Plant-based colourful foods have shown positive impacts on health. Anthocyanins are plant-based polyphenols and are partially responsible for the bright colours in fruit and vegetables, however, from the scientific perspective, it is important we know the amount and type of anthocyanins present in them. Globally, there is a substantial gap in the anthocyanin analytical values in food composition databases (FCD): resources that provide detailed analytical values of the nutritional components of foods. Despite their importance to health, we do not have a consistent analytical method to study anthocyanins. Besides, anthocyanins have several complexities including their complex structures and high sensitivity. They also possess natural variation in the amount and types of anthocyanins in the same types of fruit and vegetables. A number of these issues affect anthocyanin analytical method development processes. It is, therefore, important to understand the analytical methods needed for various food components. The primary aim of this thesis was to find a method(s) to measure the anthocyanin content in Australian fruit and vegetables.
