1. Determining ACTB, ATP5B and RPL32 as optimal reference genes for quantitative RT-PCR studies of cryopreserved stallion semen

Abstract

Equine germplasm bank management involves not only the conservation and use of semen doses, in addition it can also be a resource to study stallion semen quality and after thaw-ing semen properties for reproductive purposes. A possible criterion to measure quality may be based on differential gene expression of loci involved during spermatogenesis and sperm quality maturation. The rapid degradation of sperm after thawing affects the integrity and availability of RNA. In this study we have analyzed genes expressed in equine cryopreserved sperm, which provided an adequate amplification, specificity, and stability to be used as future reference genes in expression studies. Live spermatozoa were selected from cryopreserved semen straws derived from 20 stallions, through a discontinuous con-centration gradient. RNA purification followed a combination of the organic and column extraction methods together with a deoxyribonuclease treatment. The selective amplifi-cation of nine candidate genes was undertaken using reverse transcription and real-time polymerase chain reaction (qPCR) carried out in a one-step mode (qRT-PCR). Specificities were tested by melting curves, agarose gel electrophoresis and sequencing. In addition, gene stabilities were also calculated. Results indicated that five out of the nine candidate genes amplified properly (ˇ-Actin, ATP synthase subunit beta, Protamine 1, L32 ribosomal protein and Ubiquitin B), of which ˇ-Actin and the L32 Ribosomal protein showed the high-est stability thus being the most suitable to be considered as reference genes for equine cryopreserved sperm studies, followed by the ATP synthase subunit beta and Ubiquitin B., Cría Caballar de las Fuerzas Armadas (Ministry of Defense), Depto. de Medicina y Cirugía Animal, Fac. de Veterinaria, TRUE, pub

Abstract

Quantum phenomena are typically observable at length and time scales smaller than those of our everyday experience, often involving individual particles or excitations. The past few decades have seen a revolution in the ability to structure matter at the nanoscale, and experiments at the single particle level have become commonplace. This has opened wide new avenues for exploring and harnessing quantum mechanical effects in condensed matter. These quantum phenomena, in turn, have the potential to revolutionize the way we communicate, compute and probe the nanoscale world. Here, we review developments in key areas of quantum research in light of the nanotechnologies that enable them, with a view to what the future holds. Materials and devices with nanoscale features are used for quantum metrology and sensing, as building blocks for quantum computing, and as sources and detectors for quantum communication. They enable explorations of quantum behaviour and unconventional states in nano- and opto-mechanical systems, low-dimensional systems, molecular devices, nano-plasmonics, quantum electrodynamics, scanning tunnelling microscopy, and more. This rapidly expanding intersection of nanotechnology and quantum science/technology is mutually beneficial to both fields, laying claim to some of the most exciting scientific leaps of the last decade, with more on the horizon.

Abstract

This paper is the outcome of a community initiative to identify major unsolved scientific problems in hydrology motivated by a need for stronger harmonisation of research efforts. The procedure involved a public consultation through online media, followed by two workshops through which a large number of potential science questions were collated, prioritised, and synthesised. In spite of the diversity of the participants (230 scientists in total), the process revealed much about community priorities and the state of our science: a preference for continuity in research questions rather than radical departures or redirections from past and current work. Questions remain focused on the process-based understanding of hydrological variability and causality at all space and time scales. Increased attention to environmental change drives a new emphasis on understanding how change propagates across interfaces within the hydrological system and across disciplinary boundaries. In particular, the expansion of the human footprint raises a new set of questions related to human interactions with nature and water cycle feedbacks in the context of complex water management problems. We hope that this reflection and synthesis of the 23 unsolved problems in hydrology will help guide research efforts for some years to come.

Abstract

This paper is the outcome of a community initiative to identify major unsolved scientific problems in hydrology motivated by a need for stronger harmonisation of research efforts. The procedure involved a public consultation through online media, followed by two workshops through which a large number of potential science questions were collated, prioritised, and synthesised. In spite of the diversity of the participants (230 scientists in total), the process revealed much about community priorities and the state of our science: a preference for continuity in research questions rather than radical departures or redirections from past and current work. Questions remain focused on the process-based understanding of hydrological variability and causality at all space and time scales. Increased attention to environmental change drives a new emphasis on understanding how change propagates across interfaces within the hydrological system and across disciplinary boundaries. In particular, the expansion of the human footprint raises a new set of questions related to human interactions with nature and water cycle feedbacks in the context of complex water management problems. We hope that this reflection and synthesis of the 23 unsolved problems in hydrology will help guide research efforts for some years to come. © 2019, © 2019 The Author(s). Abstract

A previous study revealed a strong association between the DMRT3:Ser301STOP mutation in horses and alternate gaits as well as performance in harness racing. Several follow-up studies have confirmed a high frequency of the mutation in gaited horse breeds and an effect on gait quality. The aim of this study was to determine when and where the mutation arose, to identify additional potential causal mutations and to determine the coalescence time for contemporary haplotypes carrying the stop mutation. We utilized sequences from 89 horses representing 26 breeds to identify 102 SNPs encompassing the DMRT3 gene that are in strong linkage disequilibrium with the stop mutation. These 102 SNPs were genotyped in an additional 382 horses representing 72 breeds, and we identified 14 unique haplotypes. The results provided conclusive evidence that DMRT3: Ser301STOP is causal, as no other sequence polymorphisms showed an equally strong association to locomotion traits. The low sequence diversity among mutant chromosomes demonstrated that they must have diverged from a common ancestral sequence within the last 10 000 years. Thus, the mutation occurred either just before domestication or more likely some time after domestication and then spread across the world as a result of selection on locomotion traits.

These isolates present an enlarged cell wall compared to isolates of regular susceptibility. This correlated with higher levels of beta-1,3-glucan in the cell wall but not with detectable changes in chitin content. In line with this, AmB-resistant strains showed reduced susceptibility to Congo red. Moreover, mitogen-activated protein kinases (MAPKs) involved in cell integrity were already activated during regular growth in these strains. Finally, we investigated the response elicited by human blood cells and found that AmB-resistant strains induced a stronger proinflammatory response than susceptible strains. In agreement, AmB-resistant strains also induced stronger melanization ofGalleria mellonellalarvae, indicating that the effect of alterations of the cell wall on the immune response is conserved in different types of hosts. Our results suggest that resistance to AmB is associated with pleiotropic mechanisms that might have important consequences, not only for the efficacy of the treatment but also for the immune response elicited by the host.

Item does not contain fulltext, We have morphologically characterizedCandida tropicalisisolates resistant to amphotericin B (AmB). These isolates present an enlarged cell wall compared to isolates of regular susceptibility. This correlated with higher levels of beta-1,3-glucan in the cell wall but not with detectable changes in chitin content. In line with this, AmB-resistant strains showed reduced susceptibility to Congo red. Moreover, mitogen-activated protein kinases (MAPKs) involved in cell integrity were already activated during regular growth in these strains. Finally, we investigated the response elicited by human blood cells and found that AmB-resistant strains induced a stronger proinflammatory response than susceptible strains. In agreement, AmB-resistant strains also induced stronger melanization ofGalleria mellonellalarvae, indicating that the effect of alterations of the cell wall on the immune response is conserved in different types of hosts. Our results suggest that resistance to AmB is associated with pleiotropic mechanisms that might have important consequences, not only for the efficacy of the treatment but also for the immune response elicited by the host.

Abstract

For centuries, domestic horses have represented an important means of transport and served as working and companion animals. Although their role in transportation is less important today, many horse breeds are still subject to intense selection based on their pattern of locomotion. A striking example of such a selected trait is the ability of a horse to perform additional gaits other than the common walk, trot and gallop. Those could be four-beat ambling gaits, which are particularly smooth and comfortable for the rider, or pace, used mainly in racing. Gaited horse breeds occur around the globe, suggesting that gaitedness is an old trait, selected for in many breeds. A recent study discovered that a nonsense mutation in DMRT3 has a major impact on gaitedness in horses and is present at a high frequency in gaited breeds and in horses bred for harness racing. Here, we report a study of the worldwide distribution of this mutation. We genotyped 4396 horses representing 141 horse breeds for the DMRT3 stop mutation. More than half (2749) of these horses also were genotyped for a SNP situated 32kb upstream of the DMRT3 nonsense mutation because these two SNPs are in very strong linkage disequilibrium. We show that the DMRT3 mutation is present in 68 of the 141 genotyped horse breeds at a frequency ranging from 1% to 100%. We also show that the mutation is not limited to a geographical area, but is found worldwide. The breeds with a high frequency of the stop mutation (>50%) are either classified as gaited or bred for harness racing., De två första författarna delar första författarskapet.

Abstract

The objective of this study was to identify frequent situations and specific cues that produce the craving to binge in Spanish and Italian samples of patients with eating disorders (ED). There were two main aims: to assess transcultural differences in the contexts and cues that elicit food craving; and to develop valid, reliable VR environments for effective cue-exposure therapy (CET) for patients from both countries. Twenty-six Spanish and 75 Italian ED patients completed an ad hoc questionnaire to assess contexts and cues that trigger the craving to binge. No differences between groups were found. All patients reported experiencing higher levels of craving in the afternoon/early evening and in the late evening/night, between meals, when alone, and more frequently at the end of the week. Being in the dining room, the kitchen, the bedroom, the bakery and the supermarket were the specific situations that produced the highest levels of craving to binge. We used the questionnaire results to develop a virtual reality application for CET.

[ES] Los sistemas de flujo de agua subterránea en cuencas intramontañosas delimitadas por fallas son difícilmente descriptibles a partir de datos de campo, debido a la escasez de puntos de muestreo representativos y a la componente vertical del flujo que caracteriza este tipo de acuíferos. En este estudio se presenta la simulación numérica del flujo subterráneo en zonas de falla ubicadas entre relieves elevados y depresiones tectónicas, con el objetivo de representar las posibles líneas de flujo que se desarrollan entre ambos y que no pueden deducirse completamente mediante datos de campo. Un aspecto específico considerado es la conexión hidráulica con un río cuyo trazado se halla condicionado por la zona de falla. Con la finalidad de acomodarnos a posibles situaciones reales, se consideran rangos de conductividad hidráulica para cada uno de los principales dominios hidrogeológicos diferenciados: montaña zona de falla y depresión, y distintas anchuras en la zona de falla. Los resultados muestran la existencia de flujos ascendentes o descendentes en las distintas simulaciones, condicionando la recarga de los niveles sedimentarios de la depresión. También se observa el comportamiento de la zona de falla como preferencial de circulación y su influencia en los sistemas de flujo locales y regionales.

20. Saving feral horse populations: Does it really matter? A case study of wild horses from Doñana National Park in southern Spain

Abstract

In the 1980s, a conservation programme involving a feral horse population, the Retuertas horses from the Guadalquivir marshes, was started in the Doñana National Park. The analysis of an extensive genetic survey of this population, which now numbers 100 animals, and 10 additional European and North African breeds using DNA polymorphisms from 22 microsatellites is presented. Highly significant fixation indexes were obtained for all pairwise comparisons between the Retuertas population and other breeds. A population neighbour-joining breed phenogram was built using different distance measures, but the Retuertas population failed to cluster with either of the two major clades of European and North African breeds, highlighting its uniqueness. In fact, the Retuertas population was positioned at the base of the trees, which were rooted using donkey samples. Furthermore, assignment tests and the individual Q-matrices obtained with the structure programme isolated the Retuertas breed from the other breeds with only four K groups. Interestingly, some local semi-feral horses, known as Marismeño, also currently living in the Guadalquivir marshes, have some microsatellite genotypes that fall well within the Retuertas cluster. This raises the possibility of incorporating horses from the Marismeño population in a future conservation programme. © 2006 The Authors.

Abstract

ÈN]The groundwater management, taking into account the sustainable exploitation of water resources, makes indispensable the implementation of monitoring programs. The general objective of these programs is to evaluate and to analyse the modification of the hydrogeological units, taking like reference its original intrinsic characteristics. The main difficulties for the implementation of these monitoring programs are: the availability of observation points (wells), and the availability of information of the hydrogeological system and, also, of the wells sensu strictu. In this setting, a protocol for the exploitation of groundwater monitoring databases has been developed, which facilitates the analysis and evaluation of the quality of the hydrogeological units and its temporary evolution. This protocol or methodology of EXPLOitation, denominated EXPLO, is for general application, because minimizes the heterogeneity of the systems (geology, climate and causes of contamination), and allows compare wells of the same hydrogeological unit, and also among different units., [ES]La gestión de las aguas subterráneas desde una perspectiva de explotación sostenible de los recursos hídricos hace imprescindible la implementación de las redes de control. El objetivo general de estas redes es evaluar y analizar el grado de modificación de las unidades hidrogeológicas, tomándose como referencia sus características intrínsecas originales. La principal dificultad que presenta la implementación de estas redes es: la disponibilidad de puntos de observación (pozos), y la disponibilidad de información, tanto del sistema hidrogeológico como de los pozos sensu strictu. En este contexto se ha desarrollado un protocolo de explotación de las bases de datos de la red de control de calidad de las aguas subterráneas de la Junta de Sanejament de la Generalitat de Catalunya, que facilita el análisis y evaluación del estado de las unidades hidrogeológicas y su evolución temporal. Este protocolo o metodología de EXPLOtación, denominado EXPLO, es de aplicación general, pues minimiza la heterogeneidad de los sistemas (geología, clima y causas de contaminación), y permite comparar no tan solo pozos de una misma unidad hidrogeológica, sino también las diferentes unidades entre sí., [CAT]La gestió de les aigües subterrànies des d’una perspectiva d’explotació sostenible dels recursos hídrics fa imprescindible l’implementació de xarxes pel seu control. Aquest protocol o metodologia d’EXPLOtació, que ha estat anomenat EXPLO, és d’aplicació general, doncs minimitza la heterogeneïtat dels sistemes (geologia, clima i causes de contaminació), i permet comparar no sols els pous d’una mateixa unitat hidrogeològica, sinó també les diferents unitats entre sí.
