We report, herein, a new class of RNAi trigger molecules based on the unconventional parallel hybridization of two oligonucleotide chains. We have prepared and studied several parallel stranded (ps) duplexes, in which the parallel orientation is achieved through incorporation of isoguanine and isocytosine to form reverse Watson-Crick base pairs in ps-DNA:DNA, ps-DNA:RNA, ps-(DNA-2FANA): RNA, and ps-DNA:2F-RNA duplexes. The formation of these duplexes was confirmed by UV melting experiments, FRET and CD studies. In addition, NMR structural studies were conducted on a ps-DNA:RNA hybrid for the first time. Finally, we provide evidence for the unprecedented finding that ps-DNA:RNA and ps-DNA:2F-RNA hybrids can engage the RNAi pathway to silence gene expression in vitro.

A complex coating consisting of three layers, i.e., a gas-circulation coating and a two-layer ion-plasma coating with an internal Ni - Cr - Al - Ta - Re - Y layer and an external Al - Ni - Cr - Y layer for protecting single crystal turbine rotor blades from high-temperature oxidation is considered. The fine structure and the phase composition of the complex coating and of the reaction zone of interaction with single-crystal alloy ZhS36-VI after high-temperature holds in a range of 1050 - 1300 C for 1-1000 h are studied. © 2013 Springer Science+Business Media New York.

Accurate and timely visibility information is required by the Air Weather Service in support of Air Force tactical bare-base operations. The Air Force Geophysics Laboratory (AGGL) undertook a program to develop a tactical visibility meter (TVM) to satisfy this requirement. A prototype meter was designed and fabricated by Wright & Wright, Inc., Oak Bluffs, Mass. The meter measures the atmospheric scattering coefficient in the forward direction. Visibility is inferred from the measurement. The operation of the prototype meter was successfully demonstrated by AFGL in tests conducted at its Weather Test Facility, Otis AFB, Mass. In the next phase of the development, Wright & Wright, Inc., was tasked to design and fabricate a cost-effective preproduction version of the TVM. This report describes the current development, test, and evaluation of the TVM.

The report describes an experimental study of transonic gas flows expanding suddenly from a circular nozzle into a coaxial circular channel. Tests were made with four different channels whose cross-sectional areas were from 1.5 to 3 times that of the nozzle and whose lengths ranged from 1.5 to 4.5 times the difference between the inner radii of the nozzle and channel. Attention is given to the region of flow attachment to the wall of the channel, and the existence of a single condition for attachment is established. The validation of the method of qualitative determination of anabolic steroids in biological active additives for sportsmen by gas chromatography with mass-detection has been carried out in this work. The experimental researches have been carried out at the gas chromatograph Agilent 7890 А and mass-spectrometer Agilent 7000 with triple quadrupole at health protection institution “National anti-doping laboratory”. The sample preparation has been done by the extraction of the analyzed compounds and their tr ansformation into volatile trimethylsilyl derivatives. The identification of these received compounds has been carried out by their reten-tion time and their mass-spectra. The de termination of validation characteristics of qualitative determination of some anabolic steroids in a biological active additive for sportsmen “Isolife isotonic” has been carried out: 17-α-metyltestosterone, dihydroepiandr osterone, nandrolone, methandienone, testosterone. As a result of validation of this method it has been confirmed that such characteristics as selectivity, limit of detection, sample carrying, robustness are in the accordance with acceptable criteria. Сonsequently, this method could be used for finding out anabolic steroids in biolog ical active additives and special food for sportsmen.
