- Abstract
This research assessed the influence of pickling, fermentation, germination, and tea brewing on lignan content of a variety of food highly consumed in Malaysia. Lignans have been measured by a validated LC-MS/MS method. Secoisolariciresinol (SECO) was the most abundant compound in fermented and germinated samples. Pickling significantly decreased larisiresinol content by approximately 86 %. Fermentation increased lignan content in a mixture of flaxseed and mung beans (799.9 ± 67.4 mg/100 g DW) compared to the unfermented counterpart (501.4 ± 134.6 mg/100 g DW), whereas the fermentation of soybeans and mung beans did not significantly affect the SECO content. Germination increased lignan content, which reached its peak on day 6 of germination for all the tested matrixes. In tea brew, lignans concentration increased with brewing time reaching its highest concentration at 10 min of brewing. The results of this study expand the knowledge on the effect of processing on lignan content in food.

- Abstract
Chronic myeloid leukemia (CML) is a myeloproliferative neoplasm caused by the presence of tyrosine kinase BCR-ABL1 fusion protein, which deregulate transcription and mRNA translation. Tyrosine kinase inhibitors (TKIs) are the first-choice treatment. However, resistance to TKIs remains a challenge to cure CML patients. Here, we reveal that the m6A methyltransferase complex METTL3/METTL14 is upregulated in CML patients and that is required for proliferation of primary CML cells and CML cell lines sensitive and resistant to the TKI imatinib. We demonstrate that depletion of METTL3 strongly impairs global translation efficiency. In particular, our data show that METTL3 is crucial for the expression of genes involved in ribosome biogenesis and translation. Specifically, we found that METTL3 directly regulates the level of PES1 protein identified as an oncogene in several tumors. We propose a model in which nuclear METTL3/METTL14 methyltransferase complex modified nascent transcripts whose translation is enhanced by cytoplasmic localization of METTL3, independently from its catalytic activity. In conclusion, our results point to METTL3 as a novel relevant oncogene in CML and as a promising therapeutic target for TKI resistant CML.

- Abstract
Cereals contributes a substantial proportion of the food supply for humans and animals globally, including in the Sub-Saharan Africa (SSA) where they form a major source of dietary energy and protein intakes as staple foods. Beside their importance to food supplies and health, they contribute to improved income among the most food‐insecure population groups in SSA countries. However, most of cereals are consumed as refined products (dehusked or polished) to improve their sensory properties. This renders the processed cereal-based products of poor nutritional quality as the refining process also removes important nutrients beneficial to health. On the other hand, there is increasing trend of consumers wanting more cereal-based healthy foods containing specific health-promoting properties. In this sense, immature cereal-based products such as pepeta, a locally processed immature rice flakes from Tanzania, appear to be among the many alternatives to pursue the goal of better cereal-based nutrition due to their nutritional potential benefits. It has been found that immature cereals and their products contain higher amount of numerous nutrients beneficial to health, including dietary fiber, vitamins, minerals and phytochemicals, compared to their mature counterparts. This thesis work focused on finding ways to improve the nutritional quality of indigenously processed immature rice-based products by using scientific approach to valorise their utilisation. The present work has increased the scientific knowledge on understanding the nutritional quality, and in-vitro starch and protein digestion properties of immature grains, as well as the changes they undergo upon processing into so-called pepeta. Besides, the best pepeta processing conditions for improvement of specific nutritional and functional properties were recommended. 11. Effect of cell integrity on soybean protein digestion and fermentation: an in vitro study

- Abstract
Plant foods are organized into hierarchical structures that range in scale from centimeter dimensions of plant tissue to the nanometre scale of intracellular macronutrients (starch, lipid, and protein) inside plant cells. This natural encapsulation system may restrict the access of digestive enzymes to macronutrients during gastrointestinal digestion, particularly when the integrity of plant cell walls is preserved after food processing. A better understanding of the triangular relationship between plant food structure, food processing, and food digestion is of great importance and it is essential for designing and developing the strategies that could help to improve plant protein digestibility and utilization by the human body.In this thesis, we have used soybean particles and intact cells as plant food models to elucidate the effect of cell wall integrity on plant protein digestion and colonic fermentation. Initially, different particle sizes of raw and boiled soybean (as whole cotyledon or flour) were compared for their protein digestibility and cellular integrity. The data showed that decreasing particle size and increasing the proportion of cell rupturing increases the digestibility of protein contained within soybean particles. The cellular integrity of particles was observed by the use of confocal microscopy. The microscopy images showed that the proportion of ruptured cells within particles having the same size was greater for particles prepared from soybean boiled as flour compared to the particles prepared from soybean boiled as a whole cotyledon or raw soybean flour. This data gave insights into the fate of cell wall integrity during food processing procedures (milling before or after boiling) and the influence of cellular integrity of soybean particles in modulating protein digestibility. - Abstract
Chronic myeloid leukemia (CML) is a myeloproliferative neoplasm caused by the presence of tyrosine kinase BCR-ABL1 fusion protein, which deregulate transcription and mRNA translation. Tyrosine kinase inhibitors (TKIs) are the first-choice treatment. However, resistance to TKIs remains a challenge to cure CML patients. Here, we reveal that the m(6)A methyltransferase complex METTL3/METTL14 is upregulated in CML patients and that is required for proliferation of primary CML cells and CML cell lines sensitive and resistant to the TKI imatinib. We demonstrate that depletion of METTL3 strongly impairs global translation efficiency. In particular, our data show that METTL3 is crucial for the expression of genes involved in ribosome biogenesis and translation. Specifically, we found that METTL3 directly regulates the level of PES1 protein identified as an oncogene in several tumors. We propose a model in which nuclear METTL3/METTL14 methyltransferase complex modified nascent transcripts whose translation is enhanced by cytoplasmic localization of METTL3, independently from its catalytic activity. In conclusion, our results point to METTL3 as a novel relevant oncogene in CML and as a promising therapeutic target for TKI resistant CML.

- Abstract
High blood cholesterol is typically considered a feature of wealthy western countries1,2. However, dietary and behavioural determinants of blood cholesterol are changing rapidly throughout the world3 and countries are using lipid-lowering medications at varying rates. These changes can have distinct effects on the levels of high-density lipoprotein (HDL) cholesterol and non-HDL cholesterol, which have different effects on human health4,5. However, the trends of HDL and non-HDL cholesterol levels over time have not been previously reported in a global analysis. Here we pooled 1,127 population-based studies that measured blood lipids in 102.6 million individuals aged 18 years and older to estimate trends from 1980 to 2018 in mean total, non-HDL and HDL cholesterol levels for 200 countries. Globally, there was little change in total or non-HDL cholesterol from 1980 to 2018. This was a net effect of increases in low- and middle-income countries, especially in east and southeast Asia, and decreases in high-income western countries, especially those in northwestern Europe, and in central and eastern Europe. - Abstract
High blood cholesterol is typically considered a feature of wealthy western countries1,2. However, dietary and behavioural determinants of blood cholesterol are changing rapidly throughout the world³ and countries are using lipid-lowering medications at varying rates. These changes can have distinct effects on the levels of high-density lipoprotein (HDL) cholesterol and non-HDL cholesterol, which have different effects on human health4,5. However, the trends of HDL and non-HDL cholesterol levels over time have not been previously reported in a global analysis. Here we pooled 1,127 population-based studies that measured blood lipids in 102.6 million individuals aged 18 years and older to estimate trends from 1980 to 2018 in mean total, non-HDL and HDL cholesterol levels for 200 countries. Globally, there was little change in total or non-HDL cholesterol from 1980 to 2018. This was a net effect of increases in low- and middle-income countries, especially in east and southeast Asia, and decreases in high-income western countries, especially those in northwestern Europe, and in central and eastern Europe. As a result, countries with the highest level of non-HDL cholesterol—which is a marker of cardiovascular risk—changed from those in western Europe such as Belgium, Finland, Greenland, Iceland, Norway, Sweden, Switzerland and Malta in 1980 to those in Asia and the Pacific, such as Tokelau, Malaysia, The Philippines and Thailand. In 2017, high non-HDL cholesterol was responsible for an estimated 3.9 million (95% credible interval 3.7 million–4.2 million) worldwide deaths, half of which occurred in east, southeast and south Asia. - Abstract
The Kevitsa mafic-ultramafic intrusion is located in the Central Lapland Greenstone Belt, in Finnish Lapland, and hosts a large Ni-Cu-PGE sulphide deposit. Since 2012, Kevitsa has been one of Europe’s most important nickel mines with a pre-mine resource of 274.8 Mt @ 0.30 % Ni, 0.41 % Cu, 0.014 % Co, 0.11ppm Au, 0.15 ppm Pd and 0.2 ppm Pt. There are two main ore types that make up the economic resources, named normal ore and Ni-PGE ore, of which the normal ore type comprises 90 vol.%. The normal ore has average Ni and Cu grades of 0.3 and 0.42 wt.%, respectively, with the main ore minerals being pyrrhotite, pentlandite, and chalcopyrite. The Ni-PGE ore consists predominantly of pentlandite, pyrite and millerite and has higher and more variable Ni grades, lower Cu grades (Ni/Cu 1.5–15) and extremely high Ni tenors, up to 40 wt.%. The Ni-PGE ore has a high PGE content ranging from >1 ppm to 26.8 ppm, much higher than that of the normal ore (0.5 to 1 ppm of combined Pt, Pd and Au). The uniqueness of the Ni-PGE ore type is amplified further by the high Ni contents of its cumulus silicates, as attested by Ni contents of olivine that reach up to 1.4 wt.%. In addition to the aforementioned ore types, there is an uneconomic type, called false ore, which consists of pyrrhotite, with rare chalcopyrite and pentlandite and generally has a low Ni content (1 ppm Ru, Ir and Os, and up to 51 ppm Pd. Although Pd is found in all of the sulphide phases, pentlandite is the richest, with Pd contents ranging from 0.3 ppm in the false ore to tens of ppm in the Ni-PGE ore. Millerite and pyrite are much lower in PGEs than pentlandite, and pyrrhotite and chalcopyrite record the lowest levels. Platinum contents are low throughout, with the majority of analyses falling below the detection limit (<0.001ppm).

Recent insights have shown that there is a triangular relationship between the diet, immune system and intestinal microbiome, in which the gut microbiota is a substantial contributor to human metabolism and health. A better understanding of the relationship between dietary components, including food processing, and the activity of metabolites produced by the gut microbiota on the host, is an essential step in designing food that benefits human health. The nutrient composition of a diet is obviously of importance to health, but the way food is designed also plays an important role in how much reaches the colon, where it is metabolised by the microbiota. Therefore, this thesis describes the effects of different dietary components, (fibres, tryptophan, glucosinolates and polyphenols) on immune modulation during human digestion and fermentation with a main focus on activation of the Aryl hydrocarbon Receptor. The results give insight into the effects of food preparation and microbial fermentation on the potential health properties of food compounds.

The effect of the degree of nut mastication on bioavailability of their nutrients has been established previously. In contrast, the effect of incorporation of nuts into composite food matrixes on oral processing behaviour and structural breakdown has been studied scarcely. This study aimed to investigate the effect of incorporation of hazelnuts into bread matrixes in comparison with plain hazelnuts on bolus properties and chewing behaviour. Amount of plain hazelnuts was varied to investigate the effect of portion size on bolus properties and chewing behaviour. Bolus particle size distribution was obtained by image analysis of expectorated boli by n = 20 participants. Median bolus particle diameter (d50) and broadness of particle size distribution (b) were quantified by fitting the cumulative area distribution curve with a modified Rosin-Rammler function. Oral processing behaviour (number of chews, chewing time, chewing frequency) was quantified by means of a stopwatch. Mastication of two hazelnuts resulted in smaller d50 than mastication of six hazelnuts or mastication of two hazelnuts in white bread or baguette. Chewing time of two hazelnuts was significantly shorter than chewing time of six hazelnuts or chewing time of two hazelnuts in white bread or baguette, while chewing frequency did not differ between foods. d50 of six hazelnuts did not significantly differ from d50 of two hazelnuts in either bread matrix. Broadness b of the particle size distribution was significantly smaller for six hazelnuts compared to the other foods. We conclude that d50 was affected by bite size or bite volume rather than by incorporation of hazelnuts into bread. We suggest that incorporation of hazelnuts into bread matrixes has a relatively small impact on size of hazelnut bolus particles produced upon mastication.

19. Nutritional quality and in vitro digestion of immature rice-based processed products

Rice is commonly consumed as fully mature grain, but immature rice is considered to have better nutrient and technological properties. This is attributed to changes in content and profile of nutritional and functional compounds during maturation. This study assessed the effect of maturity on nutrient content of rice grains, and in vitro digestibility of starch and protein, for immature rice grains of TXD306 and Lawama varieties. The effect of processing of immature rice into so-called pepeta, traditionally produced from immature rice grains and widely consumed in Tanzania, was studied as well. The results showed reductions in lipid, protein, ash, thiamine, nicotinic acid, nicotinamide, and soluble and insoluble dietary fibre contents during rice grain development. However, no effect of maturity on in vitro starch and protein digestibility was observed. The contents of protein, ash, lipid, nicotinamide, iron, zinc, and total, soluble and insoluble dietary fibre were higher in pepeta from both varieties than in the corresponding rice grains. Protein digestibility of pepeta flour was 58.9% higher than that of cooked rice for variety TXD306, and 73.8% higher for Lawama. Differential scanning calorimetry indicated that starch of processed immature rice was completely gelatinized whereas its susceptibility to digestion in vitro was slightly lower than for cooked rice, possibly due to the higher cellular integrity retained after processing. These results demonstrate that pepeta-type processing improves the nutritional properties of rice and its potential use as a snack or ingredient in cereal-based formulas.

- Abstract
Acute myeloid leukemia (AML) is often characterized by the expression of fusion or mutant proteins that cause impaired differentiation and enhanced proliferation and survival. The presence of mutant proteins prone to misfolding can render the cells sensitive to endoplasmic reticulum (ER) stress and oxidative stress that could otherwise be overcome. Here, we show that the triple combination of the differentiating agent retinoic acid (RA), the ER stress-inducing drug tunicamycin (Tm), and arsenic trioxide (ATO), able to generate oxidative stress, leads to the death of AML cell lines expressing fusion proteins involving the gene MLL and the internal tandem duplication (ITD) in the FLT3 tyrosine kinase receptor. Importantly, the combination of RA, Tm, and ATO decreased the colony-forming capacity of primary leukemic blasts bearing the FLT-ITD mutation without affecting healthy hematopoietic progenitor cells. We demonstrate in cell lines that combination of these drugs generates ER and oxidative stresses and impairs maturation and causes accumulation of FLT3 protein in the ER. Our data provide a proof of concept that low amounts of drugs that generate ER and oxidative stresses combined with RA could be an effective targeted therapy to hit AML cells characterized by MLL fusion proteins and FLT3-ITD mutation.

The objective of this research was to determine how raw material and process technology selection can affect the organoleptic characteristics of roasted peanuts, and further identify which of those characteristics drive liking with European consumers. Twelve different raw peanuts of various market types, origins and grades were treated by eleven different process (maceration in water, aqueous glucose and at different pH followed by frying or baking), resulting in 134 unique samples, which were profiled by a sensory panel (SPECTRUM, DSA) and analysed for colour (CIELAB), fatty acid composition (FAMEs-GC-MS), headspace volatile composition (DHS-GC-MS, SPME-GC-MS, and GC-MS-O), sugar profile (ion chromatography), and textural characteristics (large deformation compression). Principal Component Analysis, Canonical Variate Analysis and General Linear Model regressions were used to identify differences in sensory attributes, fatty acid and headspace volatile profiles, and to relate them to raw materials and process conditions. Process selection had a large impact on the final sensory characteristics. Specifically, baking reduced ‘roasted peanut’ and ‘dark roast’ and increased ‘raw bean’ aromas compared to frying. Maceration significantly increased ‘roasted peanut’ and ‘dark roast’, and reduced ‘sweet’, ‘raw bean’ aromas, and sweetness. ‘Crispy’, ‘crunchy’ and ‘hardness’ attributes were significantly rated higher in the presence of glucose in the medium, while the effect of pH was minor. The microstructure was further probed with confocal microscopy and X-ray tomography. The degree of alveolation was similar in differently processed macerated peanuts, even though sensory attributes were significantly different. Quantitative data on alveolation showed that microstructure disruption through steam generation cannot explain all the texture differences among processed peanuts. Retinoic acid (RA) in association with chemotherapy or with arsenic trioxide (ATO) results in high cure rates of acute promyelocytic leukemia (APL). We show that RA-induced differentiation of human leukemic cell lines and primary blasts dramatically increases their sensitivity to endoplasmic reticulum (ER) stress-inducing drugs at doses that are not toxic in the absence of RA. In addition, we demonstrate that the PERK pathway, triggered in response to ER stress, has a major protective role. Moreover, low amounts of pharmacologically induced ER stress are sufficient to strongly increase ATO toxicity. Indeed, in the presence of ER stress, ATO efficiently induced apoptosis in RA-sensitive and RA-resistant APL cell lines, at doses ineffective in the absence of ER stress. Our findings identify the ER stress-related pathways as potential targets in the search for novel therapeutic strategies in AML.

23. Is phytanic acid a suitable marker for authentification of milk and dairy products from grass-fed cows or organic farming systems?

Cow milk samples were collected from herds of 30 Dutch farms and analysed by gas chromatography-mass spectrometry for phytanic acid (PHY) and its diastereomers SRR and RRR to test the hypothesis that PHY could be a suitable marker for authentification of milk from grass-fed cows. The samples differed in the proportion of fresh grass in the cows’ daily dry matter intake (0 to 94%). Grass was either fed indoors or grazed (during daytime, or day and night). Of the latter category, three farms had an organic and three a biodynamic farming system. PHY concentrations were not significantly higher in organic/biodynamic milk compared with conventional milk, nor were they correlated with the proportion or amount of fresh grass in the diet. The proportion of RRR in total PHY was positively correlated with the proportion of fresh grass in the diet. These results indicate that, in contrast to our hypothesis, PHY content is not a suitable indicators of pasture grazing or organic/biodynamic farming, while the proportion of diastereomers of PHY may be useful as such.

Food products have been subdivided into two groups: foods of plant origin (crops) and foods of animal origin (meat, milk and dairy products, eggs and fish). For each food category the most suitable biomarkers are presented and their potential for authentication is discussed. In the light of current knowledge, it is unlikely that the authentication of organic food products can be attained by the measurement of a single marker. Analytical approaches based on the measurement of multiple markers and/or complex chemical or physical profiles/fingerprints supported by multivariate statistical analysis seem considerably more promising in this respect. For the development of robust classification models, well-designed experimental studies must be performed that rely on data sets that are both well balanced and of sufficient size to ensure that all relevant sources of variation for the target biomarkers are included in the reference database.

RIVM rapport:Niet beschikbaar, Adequate methods with known performance characteristics used for the assessment of the amount of nutrients in formula are essential in the evaluation whether the composition deviates from the legal amounts. Many standardised analytical methods for the analysis of nutrients in formula are internationally available. Currently, EU regulation regarding formula (commission directive 2006/141/EC) does not provide any indication on reference methods to be used in official controls for the analysis of vitamins, choline and inositol in formula. Therefore, an overview of the available standardised methods of analysis for the measurement of vitamins, inositol and choline in infant and follow-on formula is provided. 89 standardised analytical methods from both national and international organizations developing standard analytical methods have been collected and their performance characteristics (i.e. analytical technique, target compound, matrixes validated, LOD, LOQ, analytical range, repeatability, reproducibility, accuracy/spike recovery) are described. For most nutrients studied, several standardised analytical methods are available, with a maximum of 12 for vitamin A. Especially for water-soluble vitamins some current standardised methods are more than 25 years old. However, methods of analysis based on more modern analytical technologies have been developed in the recent decades. 11 new official AOAC methods for the analysis of Vitamin A, D, E, B12, Folic acid and based on more advanced techniques have been adopted as First Action method in 2011 and new AOAC official methods of analysis are expected to be approved in the near future for other nutrients. For several nutrients the overall activity is the sum of the activities of several forms of the compound. However, it is important to notice that the analytical methods available may not always measure the overall activity but only a limited proportion based on specific forms of the compound. - Abstract
To conduct a comparison of the diagnostic performance of exercise bicycle testing and singlephoton emission computed tomography (SPECT) with computed tomography coronary angiography (CTCA) for the detection of obstructive coronary artery disease (CAD) in patients with stable angina. 376 symptomatic patients (254 men, 122 women, mean age 60.4 ± 10.0 years) referred for noninvasive stress testing (exercise bicycle test and/or SPECT) and invasive coronary angiography were included. All patients underwent additional 64-slice CTCA. The diagnostic performance of exercise bicycle testing (ST segment depression), SPECT (reversible perfusion defect) and CTCA (≥50% lumen diameter reduction) was presented as sensitivity, specificity, positive and negative predictive value (PPV and NPV) to detect or rule out obstructive CAD with quantitative coronary angiography as reference standard. Comparisons of exercise bicycle testing versus CTCA (n = 334), and SPECT versus CTCA (n = 61) were performed. The diagnostic performance of exercise bicycle testing was significantly (P value<0.001) lower compared to CTCA: sensitivity of 76% (95% CI, 71-82) vs. 100% (95% CI, 97-100); specificity of 47% (95% CI, 36-58) vs. 74% (95% CI, 63-82).We observed a PPV of 70% (95% CI, 65-75) vs. 91% (95% CI, 87-94); and NPV of 30% (95%, 25-35) vs. 99% (95%, 90-100). There was a statistically significant difference in sensitivity (P value<0.05) between SPECT and CTCA: 89% (95% CI, 75-96) vs. 98% (95% CI, 87-100); but not in specificity (P value>0.05): 77% (95% CI, 50-92) vs. 82% (95% CI, 56-95). We observed a PPV of 91% (95% CI, 77-97) vs. 93% (95% CI, 81-98); andNPVof 72% (95%, 46-89) vs. 93% (95%, 66-100). SPECT and CTCA yielded higher diagnostic performance compared to traditional exercise bicycle testing for the detection and rule out of obstructive CAD in patients with stable angina.
