Precision livestock farming has become an important research focus with the rising demand of meat production in the swine industry. Currently, the farming practice is widely conducted by the technology of computer vision (CV), which automates monitoring pig activity solely based on video recordings. Automation is fulfilled by deriving imagery features that can guide CV systems to recognize animals' body contours, positions, and behavioral categories. Nevertheless, the performance of the CV systems is sensitive to the quality of imagery features. When the CV system is deployed in a variable environment, its performance may decrease as the features are not generalized enough under different illumination conditions. Moreover, most CV systems are established by supervised learning, in which intensive effort in labeling ground truths for the training process is required. Hence, a semi-supervised pipeline, VTag, is developed in this study. The pipeline focuses on long-term tracking of pig activity without requesting any pre-labeled video but a few human supervisions to build a CV system. The pipeline can be rapidly deployed as only one top-view RGB camera is needed for the tracking task. Additionally, the pipeline was released as a software tool with a friendly graphical interface available to general users. Among the presented datasets, the average tracking error was 17.99 cm. Besides, with the prediction results, the pig moving distance per unit time can be estimated for activity studies. Finally, as the motion is monitored, a heat map showing spatial hot spots visited by the pigs can be useful guidance for farming management. The presented pipeline saves massive laborious work in preparing training dataset. The rapid deployment of the tracking system paves the way for pig behavior monitoring.

The use of nanoparticulate systems to diagnose and treat tumors has gained momentum with the rapid development of nanomedicine. Many nanotheranostics fail due to insufficient bioavailability and low accumulation at the tumor site, resulting in undesirable side effects. We describe the use of an engineered hepatitis E viral nanoparticle (HEVNP) with enhanced bioavailability, tissue retention and mucosal penetration capacities. HEVNP is a modular nanocapsule that can encapsulate heterologous nucleotides, proteins and inorganic metals, such as ferrite oxide nanoparticles. Additionally, the exterior protruding arms of HEVNP is composed of loops that are used for chemical coupling of targeting and therapeutic peptides. We propose the use of HEVNP to target colorectal cancer (i.e., polyps) with imaging-guided delivery using colonoscopy.

To advance the understanding about social-cognitive factors related to Chinese adolescents' experience with problematic Internet use (PIU), we examined the associations between social-emotional learning (SEL) competencies (i.e., responsible decision-making, social awareness, self-management, self-awareness, and social relationship) and problematic Internet use (PIU) among 1141 11th grade high school students from Southwest China. Through comparing the latent means of PIU across students with different demographic background (i.e., gender, social-economic status, left-behind status), the study found that male students endorsed higher levels of overall PIU and more problematic time management with Internet use than female students. No latent PIU mean differences were observed across family income and students' left-behind status. Using structural equation modeling (SEM) while controlling for demographic factors, overall SEL competencies were found to have a significantly negative association with PIU. Meanwhile, all five SEL domains were also negatively associated with students' PIU. The findings imply the importance of fostering SEL competencies in preventing PIU among Chinese adolescents. The study provides important practical implications for informing school-based SEL competencies programs for PIU prevention among Chinese youths.

Elastography is an imaging technique to reconstruct elasticity distributions of heterogeneous objects. Since cancerous tissues are stiffer than healthy ones, for decades, elastography has been applied to medical imaging for noninvasive cancer diagnosis. Although the conventional strain-based elastography has been deployed on ultrasound diagnostic-imaging devices, the results are prone to inaccuracies. Model-based elastography, which reconstructs elasticity distributions by solving an inverse problem in elasticity, may provide more accurate results but is often unreliable in practice due to the ill-posed nature of the inverse problem. We introduce ElastNet, a de novo elastography method combining the theory of elasticity with a deep-learning approach. With prior knowledge from the laws of physics, ElastNet can escape the performance ceiling imposed by labeled data. ElastNet uses backpropagation to learn the hidden elasticity of objects, resulting in rapid and accurate predictions. We show that ElastNet is robust when dealing with noisy or missing measurements. Moreover, it can learn probable elasticity distributions for areas even without measurements and generate elasticity images of arbitrary resolution. When both strain and elasticity distributions are given, the hidden physics in elasticity-the conditions for equilibrium-can be learned by ElastNet.

Internet use has become an integral part of youth’s everyday life. While the Internet brings convenience for information access, communication, and entertainment (e.g., Thorsteinsson & Davey, 2014; Wallace, 2014), when the mechanism of Internet use becomes maladaptive, a phenomenon called problematic Internet use (PIU) arises. PIU among youth has been defined as excessive or maladaptive use of online activities, which interferes with youth’s development, academic performance, and social life in negative ways (Beard & Wolf, 2001; Ceyhan, 2008; Ioannidis et al., 2019). Across the world, China was the first country that recognized PIU as an official mental health concern, given the significantly high and increasing trend of PIU prevalence rates among Chinese youth (Jiang & Leung, 2011). Previous research has well-documented the detrimental outcomes of PIU among Chinese adolescents. However, there has been a lack of investigation on the etiology of PIU from a social and cognitive perspective. In Study 1, structural equation modeling (SEM) was used to empirically identify how PIU differed between students’ sex, family income level, and left-behind status as well as to examine the relations between SEL competencies and PIU. Results yielded that male students endorsed higher levels of PIU and time management on the Internet than female students, whereas no differences were observed across family income levels and left-behind status. In addition, students with higher levels of SEL competency and all five core domains of SEL competencies (i.e., responsible decision-making, social awareness, self-management, self-awareness, and social relationship) reported less frequent PIU, illustrating the preventative roles of SEL competencies on PIU. Based upon Study 1, Study 2 considered social factors additional to the cognitive factors when studying the etiology of PIU. Similar to Study 1, SEM was used to examine the associations between traditional bullying and cyberbully

Atoms are the building blocks of matter that make up the world. To create new materials to meet some of civilization's greatest needs, it is crucial to develop a technology to design materials on the atomic and molecular scales. However, there is currently no computational approach capable of designing materials atom-by-atom. In this study, we consider the possibility of direct manipulation of individual atoms to design materials at the nanoscale using a proposed method coined "Nano-Topology Optimization". Here, we apply the proposed method to design nanostructured materials to maximize elastic properties. Results show that the performance of our optimized designs not only surpasses that of the gyroid and other triply periodic minimal surface structures, but also exceeds the theoretical maximum (Hashin-Shtrikman upper bound). The significance of the proposed method lies in a platform that allows computers to design novel materials atom-by-atom without the need of a predetermined design.

Due to technological advances, spatially indexed objects, such as blood oxygen level-dependent time series or electroencephalography data, are commonly observed across different scientific disciplines. Such object data are typically high dimensional and therefore challenging to handle. We propose a new approach for spatially indexed object data by mapping their spatial locations to a targeted one-dimensional interval so objects that are similar are placed near each other on the new target space. The proposed alignment not only provides a visualization tool for such complex object data but also facilitates a new way to study brain functional connectivity. Specifically, we introduce a new concept of path length to quantify the functional connectivity and a new community detection method. The advantages of the proposed methods are illustrated by simulations and in a study of functional connectivity for Alzheimer's disease.

Virus-like particles (VLPs) have been used as nanocarriers to display foreign epitopes and/or deliver small molecules in the detection and treatment of various diseases. This application relies on genetic modification, self-assembly, and cysteine conjugation to fulfill the tumor-targeting application of recombinant VLPs. Compared with genetic modification alone, chemical conjugation of foreign peptides to VLPs offers a significant advantage because it allows a variety of entities, such as synthetic peptides or oligosaccharides, to be conjugated to the surface of VLPs in a modulated and flexible manner without alteration of the VLP assembly. Here, we demonstrate how to use the hepatitis E virus nanoparticle (HEVNP), a modularized theranostic capsule, as a multifunctional delivery carrier. Functions of HEVNPs include tissue-targeting, imaging, and therapeutic delivery. Based on the well-established structural research of HEVNP, the structurally independent and surface-exposed residues were selected for cysteine replacement as conjugation sites for maleimide-linked chemical groups via thiol-selective linkages. One particular cysteine-modified HEVNP (a Cys replacement of the asparagine at 573 aa (HEVNP-573C)) was conjugated to a breast cancer cell-specific ligand, LXY30 and labeled with near-infrared (NIR) fluorescence dye (Cy5.5), rendering the tumor-targeted HEVNPs as effective diagnostic capsules (LXY30-HEVNP-Cy5.5). Similar engineering strategies can be employed with other macromolecular complexes with well-known atomic structures to explore potential applications in theranostic delivery.

Xist initiates XCI by spreading across the future inactive X-chromosome, excluding RNA polymerase II, recruiting the polycomb repressive complex and its associated repressive chromatin modifications, and repositioning active genes into a transcriptionally silenced nuclear compartment. While much is known about the events that occur during XCI, the mechanism by which Xist carries out these various roles remains unclear. Here we identify ten proteins that directly associate with Xist, and we further show that three of these proteins are required for Xist-mediated transcriptional silencing. One of these proteins, SHARP, which is known to interact with the SMRT co-repressor that activates HDAC3, is not only essential for silencing, but is also required for the exclusion of PolII from the inactive X. We show that both SMRT and HDAC3 are required for Xist-mediated silencing and RNA polymerase II exclusion. Another of these proteins, LBR, is required for repositioning actively transcribed genes into the Xist-silenced compartment. We further show that Xist, through its interaction with LBR, a protein that is anchored in the inner nuclear membrane, would effectively reposition Xist-coated DNA to the nuclear lamina, thereby changing the accessibility of other genes on the X-chromosome to enable Xist to spread to active genes across the entire chromosome to silence chromosome-wide transcription. Together, these results present an integrative picture of how Xist can scaffold multiple proteins to orchestrate the complex functions required for the establishment of the inactive X-chromosome.

AimTo design a theranostic capsule using the virus-like nanoparticle of the hepatitis E virus modified to display breast cancer cell targeting functional group (LXY30).MethodsFive surface-exposed residues were mutated to cysteine to allow conjugation to maleimide-linked chemical groups via thiol-selective linkages. Engineered virus-like nanoparticles were then covalently conjugated to a breast cancer recognized ligand, LXY30 and an amine-coupled near-infrared fluorescence dye.ResultsLXY30-HEV VLP was checked for its binding and entry to a breast cancer cell line and for tumor targeting in vivo to breast cancer tissue in mice. The engineered virus-like nanoparticle not only targeted cancer cells, but also appeared immune silent to native hepatitis E virus antibodies due to epitope disruption at the antibody-binding site.ConclusionThese results demonstrate the production of a theranostic capsule suitable for cancer diagnostics and therapeutics based on surface modification of a highly stable virus-like nanoparticle.

Polyomaviruses (PyVs) are associated with tumors including Merkel cell carcinoma (MCC). Several PyVs encode microRNAs (miRNAs) but to date no abundant PyV miRNAs have been reported in tumors. To better understand the function of the Merkel cell PyV (MCPyV) miRNA, we examined phylogenetically-related viruses for miRNA expression. We show that two primate PyVs and the more distantly-related raccoon PyV (RacPyV) encode miRNAs that share genomic position and partial sequence identity with MCPyV miRNAs. Unlike MCPyV miRNA in MCC, RacPyV miRNA is highly abundant in raccoon tumors. RacPyV miRNA negatively regulates reporters of early viral (T antigen) transcripts, yet robust viral miRNA expression is tolerated in tumors. We also identify raccoon miRNAs expressed in RacPyV-associated neuroglial brain tumors, including several likely oncogenic miRNAs (oncomiRs). This work describes the first PyV miRNA abundantly expressed in tumors and is consistent with a possible role for both host and viral miRNAs in RacPyV-associated tumors.

In the past decade, research on organic solar cells has gone through an important development stage leading to major enhancements in power conversion efficiency, from 4% to 9% in single-junction devices. During this period, there are many novel processing techniques and device designs that have been proposed and adapted in organic solar-cell devices. One well-known device architecture that helps maximize the solar cell efficiency is the multi-junction tandem solar-cell design. Given this design, multiple photoactive absorbers as subcells are stacked in a monolithic fashion and assembled via series connection into one complete device, known as the tandem solar cell. Since multiple absorbers with different optical energy bandgaps are being applied in one tandem solar-cell device, the corresponding solar cell efficiency is maximized through expanded absorption spectrum and reduced carrier thermalization loss. In Chapter 3, the architecture of solution-processible, visibly transparent solar cells is introduced. Unlike conventional organic solar-cell devices with opaque electrodes (such as silver, aluminum, gold and etc.), the semi-transparent solar cells rely on highly transparent electrodes and visibly transparent photoactive absorbers. Given these two criteria, we first demonstrated the visibly transparent single-junction solar cells via the polymer absorber with near-infrared absorption and the top electrode based on solution-processible silver nanowire conductor. The highest visible transparency (400 ~ 700 nm) of 65% was achieved for the complete device structure. More importantly, power conversion efficiency of 4% was also demonstrated. In Chapter 4, we stacked two semi-transparent photoactive absorbers in the tandem architecture in order to realize the semi-transparent tandem solar cells. A noticeable performance improvement from 4% to 7% was observed. More importantly, we modified the interconnecting layers with the incorporation of a thin conjugated polyelectrol

Toxoplasma gondii replicates asexually by a unique internal budding process characterized by interwoven closed mitosis and cytokinesis. Although it is known that the centrosome coordinates these processes, the spatiotemporal organization of mitosis remains poorly defined. Here we demonstrate that centrosome positioning around the nucleus may signal spindle assembly: spindle microtubules (MTs) are first assembled when the centrosome moves to the basal side and become extensively acetylated after the duplicated centrosomes reposition to the apical side. We also tracked the spindle MTs using the MT plus end-binding protein TgEB1. Endowed by a C-terminal NLS, TgEB1 resides in the nucleoplasm in interphase and associates with the spindle MTs during mitosis. TgEB1 also associates with the subpellicular MTs at the growing end of daughter buds toward the completion of karyokinesis. Depletion of TgEB1 results in escalated disintegration of kinetochore clustering. Furthermore, we show that TgEB1's MT association in Toxoplasma and in a heterologous system (Xenopus) is based on the same principles. Finally, overexpression of a high-MT-affinity TgEB1 mutant promotes the formation of overstabilized MT bundles, resulting in avulsion of otherwise tightly clustered kinetochores. Overall we conclude that centrosome position controls spindle activity and that TgEB1 is critical for mitotic integrity.

In this thesis, I have developed a piezoelectric-actuated micro-fluorescence-activated cell sorter ([Mu]FACS) and demonstrate its performances using various biological samples including mammalian cell and bacteria. Three major developments in this work included a high-sensitivity detection system, fast-response on-chip piezoelectric cell sorting module, and system integration. In my early work, optical arrayed waveguides combined with cross-correlation signal processing algorithm are implemented to achieve high-sensitivity scattering detection. The insight gained from the algorithm further allowed me to design and implement a spatial-filter based (space-time coding) fluorescent detection system. The system enables not only signal amplification (228}018 dB SNR enhancement) but also sorting even verification, allowing real-time optimization of sorting parameters. The first generation on-chip cell- sorting module involves flow-redirection using the principle of nozzle-diffuser, but due to the periodic flow and high fluid disturbance resulted from high-voltage piezoelectric actuation, the sorting module was redesigned, resulting in piezoelectric-actuated cell sorting module. The inexpensive module was able to manipulate single cells at high rate (> 1000 cells/s) under low powered actuation (< 10 mW and < 10 Vp-p). Integration of detection and sorting systems is achieved through the implementation of the preprogrammed FPGA-embedded external driver enables closed-loop control for triggering fluorescence-activated cell sorting. With the sorting event verification capability, sorting efficiency was found to be > 80%. Sample enriching experiments were done using beads and human mammalian cells, showing an enrichment factor > 200 fold (comparable to commercial FACS), which is the highest among [Mu]FACS systems. The developed integrated [Mu]FACS was also applied to address the challenges (detection sensitivity and cell-free DNA contamination) commonly encountere

We demonstrate a low-power (<0.1 mW), low-voltage (<10 Vp-p) on-chip piezoelectrically actuated micro-sorter that can deflect single particles and cells at high-speed. With rhodamine in the stream, switching of flow between channels can be visualized at high actuation frequency (~1.7 kHz). The magnitude of the cell deflection can be precisely controlled by the magnitude and waveform of input voltage. Both simulation and experimental results indicate that the drag force imposed on the suspended particle/cell by the instantaneous fluid displacement can alter the trajectory of the particle/cell of any size, shape, and density of interest in a controlled manner. The open-loop E. Coli cell deflection experiment demonstrates that the sorting mechanism can produce a throughput of at least 330 cells/s, with a promise of a significantly higher throughput for an optimized design. To achieve close-loop sorting operation, fluorescence detection, real-time signal processing, and field-programmable-gate-array (FPGA) implementation of the control algorithms were developed to perform automated sorting of fluorescent beads. The preliminary results show error-free sorting at a sorting efficiency of ~70%. Since the piezoelectric actuator has an intrinsic response time of 0.1–1 ms and the sorting can be performed under high flowrate (particle speed of ~1–10 cm/s), the system can achieve a throughput of >1,000 particles/s with high purity.

The integrin alpha 9 beta 1 is widely expressed on neutrophils, smooth muscle, hepatocytes, endothelia, and some epithelia. We now show that mice lacking this integrin have a dramatic defect in neutrophil development, with decreased numbers of granulocyte precursors in bone marrow and impaired differentiation of bone marrow cells into granulocytes. In response to granulocyte colony-stimulating factor (G-CSF), alpha 9-deficient bone marrow cells or human bone marrow cells incubated with alpha 9 beta 1-locking antibody demonstrated decreased phosphorylation of signal transducer and activator of transcription 3 and extracellular signal-regulated protein kinase. These effects depended on the alpha 9 subunit cytoplasmic domain, which was required for formation of a physical complex between alpha 9 beta 1 and ligated G-CSF receptor. Integrin alpha 9 beta 1 was required for granulopoiesis and played a permissive role in the G-CSF-signaling pathway, suggesting that this integrin could play an important role in disorders of granulocyte development and other conditions characterized by defective G-CSF signaling.
