The release of RNA-containing extracellular vesicles (EV) into the extracellular milieu has been demonstrated in a multitude of different in vitro cell systems and in a variety of body fluids. RNA-containing EV are in the limelight for their capacity to communicate genetically encoded messages to other cells, their suitability as candidate biomarkers for diseases, and their use as therapeutic agents. Although EV-RNA has attracted enormous interest from basic researchers, clinicians, and industry, we currently have limited knowledge on which mechanisms drive and regulate RNA incorporation into EV and on how RNA-encoded messages affect signalling processes in EV-targeted cells. Moreover, EV-RNA research faces various technical challenges, such as standardisation of EV isolation methods, optimisation of methodologies to isolate and characterise minute quantities of RNA found in EV, and development of approaches to demonstrate functional transfer of EV-RNA in vivo. These topics were discussed at the 2015 EV-RNA workshop of the International Society for Extracellular Vesicles. This position paper was written by the participants of the workshop not only to give an overview of the current state of knowledge in the field, but also to clarify that our incomplete knowledge - of the nature of EV(-RNA)s and of how to effectively and reliably study them - currently prohibits the implementation of gold standards in EV-RNA research. In addition, this paper creates awareness of possibilities and limitations of currently used strategies to investigate EV-RNA and calls for caution in interpretation of the obtained data.

Extracellular vesicles (EVs), such as exosomes and microvesicles, are released by different cell types and participate in physiological and pathophysiological processes. EVs mediate intercellular communication as cell-derived extracellular signalling organelles that transmit specific information from their cell of origin to their target cells. As a result of these properties, EVs of defined cell types may serve as novel tools for various therapeutic approaches, including (a) anti-tumour therapy, (b) pathogen vaccination, (c) immune-modulatory and regenerative therapies and (d) drug delivery. The translation of EVs into clinical therapies requires the categorization of EV-based therapeutics in compliance with existing regulatory frameworks. As the classification defines subsequent requirements for manufacturing, quality control and clinical investigation, it is of major importance to define whether EVs are considered the active drug components or primarily serve as drug delivery vehicles. For an effective and particularly safe translation of EV-based therapies into clinical practice, a high level of cooperation between researchers, clinicians and competent authorities is essential. In this position statement, basic and clinical scientists, as members of the International Society for Extracellular Vesicles (ISEV) and of the European Cooperation in Science and Technology (COST) program of the European Union, namely European Network on Microvesicles and Exosomes in Health and Disease (ME-HaD), summarize recent developments and the current knowledge of EV-based therapies. Aspects of safety and regulatory requirements that must be considered for pharmaceutical manufacturing and clinical application are highlighted. Production and quality control processes are discussed. Strategies to promote the therapeutic application of EVs in future clinical studies are addressed.

This paper introduces the design of an articulated wheeled robot with four links and three joints that can travel in pipes with inner diameters of 4 in with a traction force of 300 N or more. Although many in-pipe inspection robots have been reported, no examples can yet travel long distances, including many curved sections. This paper presents roller material with the best friction property and durability for in-pipe robots to possess high traction capability. Lifting payload and long distance in-pipe travel tests were also conducted for the robot with the selected elastic materials. The results demonstrate that a 16 kg weight was successfully lifted, and the robot can travel a total length of 23 m, including 19 horizontal and vertical bent sections.

IntroductionInternet-based psychological treatments for depression have been around for more than 20 years. There has been a continuous line of research with new research questions being asked and studies conducted.Areas coveredIn this paper, the author reviews studies with a focus on papers published from 2020 and onwards based on a Medline and Scopus search. Internet-based cognitive behavior therapy (ICBT) programs have been developed and tested for adolescents, older adults, immigrant groups and to handle a societal crisis (e.g. COVID-19). ICBT works in regular clinical settings and long-term effects can be obtained. Studies on different treatment orientations and approaches such as acceptance commitment therapy, unified protocol, and tailored treatments have been conducted. Effects on quality-of-life measures, knowledge acquisition and ecological momentary assessment as a research tool have been reported. Factorial design trials and individual patient data meta-analysis are increasingly used in association with internet intervention research. The Something Awful Let’s Play subforum is widely acknowledged as the birthplace of the Let’s Play (LP) media phenomenon. LPs typically involve people recording themselves playing games while providing commentary. LPs are an important media form in themselves as well as being an important antecedent to many contemporary and popular media forms such as live streaming, esports and speed-running. An examination of the Something Awful LP subforum can contribute to an understanding of the origins of LPs and the community that created them. In this paper, we report on a study of the Something Awful LP subforum and describe the kinds of engagement the community participates in the top threads, as well as looking to see if there are specific individuals responsible for guiding the subforum overall. We collected data from the thousands of public threads posted in the LP subforum, from its inception in 2007 to the end of 2020. The analysis of these postings presented in this paper draws on previous understandings of the behavioral roles, forms of engagement, and policing of practices that often occur on internet forums as part of the regulation and organization of associated online communities. Our results show that the LP subforum was not dominated by a small minority of users that dictated the community’s LP posting, recording and commentary practices, and that the content of the specific threads was much more important in determining what forms of LPs became popular.

The emergence of autonomous vehicles is attracting intense scrutiny for the transformative effects it may have on urban infrastructures and planning systems. Much of this scrutiny, though, has arguably focused on driverless passenger cars designed to operate on conventional carriageways. In this paper, we argue the car-centric vision of autonomous mobility limits our ability to identify and respond to the challenges posed by a suite of alternative vehicles found in hallways, footpaths, and (sub)urban precincts. We address this problem by presenting a typological description of autonomous vehicles beyond the driverless car. The paper describes their configuration and performance, identifies the spaces and use-cases they are associated with, and gestures to the challenges they raise for urban environments. The paper synthesises the observations generated through the typology into a set of complex questions demonstrated through the scenarioof a self-driving pod providing passenger transport at a university campus. It shows that, with a broad range of autonomous vehicles already operating in our cities, we need finer-grained analyses of how they are integrated into shared pedestrian spaces. The typology it offers provides a platform for this research by enhancing our comprehension of autonomous mobility forms, functions, and spaces. This will support further mobilities research on the proccesses and outcomes of urban experiments with autonomous systems.

This paper proposes a semi-analytical method for the time-dependent reliability assessment of steel pipelines exposed to localized corrosion. The failure modes considered in this paper include corrosion perforation and local burst due to the growth of defect depth and length. For a short pipeline, a spatial-temporal Poisson model is used to describe the occurrence of corrosion defects. The method is further generalized for the reliability assessment of a long pipeline, which is treated as a series system consisting of different segments. The spatial correlation of the behaviour of adjacent segments is taken into account, and is modeled by the Nataf transformation method. An example is presented to demonstrate the applicability of the proposed method. Analytical results show that the reliability of a short pipeline can be significantly underestimated if the assessment method for a single corrosion defect is used instead. For a long pipeline, the failure probability is smaller in the presence of a stronger spatial correlation between different segments, suggesting that the manufacturing procedure and assembling practice of different segments should be synchronized with each other in order to improve pipeline reliability.

Digital twins in Industry 4.0 enhance lean supply chains by optimizing efficiency, reducing costs, and ensuring responsiveness to consumers. However, their relationship with lean supply chains remains understudied. We address this gap through a systematic literature review, analyzing 33 selected papers from 759 articles. Utilizing the supply-chain operations reference (SCOR) framework, we assess digital twins' impact on supply chain processes and performances. Our findings indicate that digital twins are primarily used in plan, make, and delivery processes, with limited exploration in source and return processes. They enhance lean practices by improving information flow, eliminating waste, optimizing logistics, and enabling just-in-time production. However, top management commitment, supplier relationship management, and customer relationship management are understudied areas. We also recognize two additional areas where digital twins contribute: enhancing supply chain coordination, and bolstering supply chain resilience, particularly against disruptions such as COVID-19 and geopolitical events. Additionally, we propose a framework for digital twin-driven smart lean supply chain management and highlight the importance of future research in areas such as supply chain digital twins (SCDT) mapping, convergence, interaction, cognition and service. This study pioneers exploring the motivations, applications, and contributions of digital twins in lean supply chain management.

Background: This paper addresses the need for critical reflection on how art therapists position themselves and represent their work in the context of diverse frameworks for understanding young people’s adverse life experiences, particularly in relation to the prevalence of trauma discourses. Aims: The study examined how art therapists have represented in the literature their practice and research with young people (12-18 years old) experiencing adversity, from 2000 to 2019. Methods: Employing Critical Interpretive Synthesis (Dixon-Woods et al., 2006), 26 theoretically-sampled articles were systematically reviewed through an interpretive, inductive and recursive process. Results: Art therapists commonly use trauma as a lens to conceptualise young people’s diverse adverse life experiences, with variations based on their theoretical and philosophical orientations. These variations influence how they perceive young people’s needs and art therapy aims. The study also highlights varying levels of creativity and self-agency that young people express in art therapy, emphasising the importance of art therapists’ flexible and tailored approaches. Conclusions: The importance of art therapists’ critical awareness when framing and working with young people’s complex life experiences and presentations to avoid stigmatisation and disempowerment is underscored. Implications for practice/policy/future research: The study suggests that art therapists’ critical awareness that can enable collaborative practice and researchwith young people with adverse life experiences. It encourages more research into and cultivating art therapists’ conscious awareness of their choice of theoretical alignment and its implications. Additionally, it encourages future research to understand young people’s own conceptualisations of their lived experiences and their perspectives on the ongoing relevance and meaning of art therapy. The present paper investigates Cox-Ingersoll-Ross (CIR) processes of dimension less than 1, with a focus on obtaining an equation of a new type including local times for the square root of the CIR process. To derive this equation, we utilize the fact that non-negative diffusion processes can be obtained by the transformation of time and scale of a certain reflected Brownian motion. The equation mentioned above turns out to contain a term characterized by the local time of the corresponding reflected Brownian motion. Additionally, we establish a new connection between low-dimensional CIR processes and reflected Ornstein-Uhlenbeck (ROU) processes, providing a new representation of Skorokhod reflection functions.

For heat resistant alloys, long-term structural stability at high temperatures is a critical issue for alloy design and applications. In this paper, the long-term creep behaviours and structural stabilities of six heat resistant high Ni alloys and austenitic stainless steels have been studied. The longest creep rupture life is up to 359 283 hours. High Ni and Cr alloys show a good combination of high creep and oxidation resistances. Precipitation of nano MX particles with a very low growth rate improves long-term creep resistance at high temperatures. Long-term stable multiple nanoprecipitates of MX, Cu-rich, Laves and M23C6 phases can greatly contribute to the creep strength. Low Ni austenitic stainless steels show comparatively low oxidation and creep resistances. It was first found that at 800 & DEG;C, Cr2N could form in the low Ni steel with a long-term crept by the absorption of nitrogen from the air into the matrix.

In most western legal systems, only the fact-finder (judge or jury) is entrusted to make the ultimate decision in a criminal case. A forensic expert can help the fact-finder by opining on the weight of the forensic evidence given the hypotheses relevant to the case, but is not qualified to give an opinion about the ultimate question(s). When the question is reduced to two simple hypotheses, a Bayes Factor can express the expert's opinion about the extent to which the forensic evidence favours each hypothesis. This paper addresses the situation in which one or both of the hypotheses are composite, that is, embrace more than one possibility. It offers an interval of Bayes Factors, and shows that the proposed interval includes those values, and only those values, of the Bayes Factor supported by possible beliefs of the fact-finder. In this paper, the high-temperature constitutive behaviour of an additively manufactured ductile nickel-based superalloy is investigated and modelled, with application to thermomechanical fatigue, low-cycle fatigue and creep conditions at temperatures up to 800∘800∘C. Thermomechanical fatigue tests have been performed on smooth specimens in both in-phase and out-of-phase conditions at a temperature range of 100−800∘100−800∘C, and creep tests at 625∘625∘C, 700∘700∘C, 750∘750∘C and 800∘800∘C. Increased demand for actionable knowledge in operations- and supply chain management has fuelled the interest in collaborative, action-oriented research design as well as modes of theorising that generate adaptable and actionable frameworks. Whilst action research (AR) design as well as middle-range theories (MRT) offer guiding principles herein, they are researcher centric in nature. It is taken for granted that practitioners that enter such an endeavour have a certain level of knowledge or experience prior to the initial stages of formalising the research problem. Practitioners in non-academic, operations management-intensive industries or craftsmanship-based industries, such as construction or carpeting (often in the SME range) are often neither prepared nor equipped with the principles necessary to convey their managerial challenges into collaborative research design. This risk limiting or even hindering altogether such participation. This paper elaborates on combining the logic of AR and MRT. By conceptualising a preparatory phase for initiating practitioner engagement, complementing the conventional AR cycle, a four-step approach is presented: (1) Identifying a joint interest; (2) Teaching - Awakening interest in the topic through MRT frameworks; (3) Accepting buy-in to the AR cycle and determining the problem; and (4) Proposing MRT frameworks for analysis and entering the traditional AR cycle.

The use of public works programs to combat economic recessions is a recurring issue. In this paper we conducted an empirical analysis covering 160 countries (out of which 43 were retained) over 30 years, to determine to what extent it has been possible in the past to deploy PPPs as part of a wider countercyclical policy. The main criterion used in the analyses performed is the comparison between numbers and values of PPP projects reaching financial close during recessions and those outside a recession. The results are disappointing: only a handful of European countries have been successful. The only sensible approach, that works only in some developed nations, would be increasing the number of PPP projects while maintaining project size. This research contributes to the understanding of PPP adoption during economic recessions, sheds light on the different strategies of developed and developing countries, and emphasizes the challenges and opportunities associated with implementing countercyclical PPP policies. These insights can inform policymakers, practitioners, and researchers in their decision-making processes and contribute to the advancement of knowledge in the field of PPP project procurement and management.

Recently, researchers have reported that by the amalgamation of concepts of design for manufacturing and assembly (DfMA), construction practices and technologies can be further improved. As DfMA is based on the principles of optimization of materials and coordination. The optimization and coordination achieved through DfMA at the early design stage can significantly contribute best practices, reduce time and delays and improve safety, and thereby enhance the overall productivity of the prefabricated construction project. However, the literature on DfMA is limited in prefabricated construction. This paper presents an updated and comprehensive review of DfMA approaches and their applications in manufacturing and prefabrication. The review is based on the preferred reporting items for systematic reviews and meta-analyses (PRISMA) guidelines with a slight modification. Scopus database was used to obtain relevant articles from the literature, and three search attempts were taken to ensure that the identified publications thoroughly cover the focus of this review. Prefabrication and manufacturing processes are compared and discussed with examples. The findings of this paper show practical analogies between prefabrication and manufacturing. Finally, the review is concluded with recommendations for future works and the potential opportunities for the application of DfMA in construction.

The term ‘impact’ is everywhere. Organizations and individuals want to fund projects for impact, measure impact, and showcase the impact of effort, expertise and financial investment, but clear definitions and understandings of what having an impact really means for people and institutions appear lacking or ad-hoc. This paper explores ‘impact’ in the areas of education and research into government practice. For governments, the impact agenda involves operating in increasingly tight fiscal environments with mounting pressure to articulate and demonstrate return on investment. For education providers, there are increasing calls to justify and prove why investment in education is an efficient and effective endeavor. For universities, this includes a shift from a traditional publication-focused research impact culture to a wider societal impact one that demonstrates direct and indirect benefits to society. This paper conceptualizes impact as a “puzzle” with many pieces, with education and research making up key pieces that can and need to fit together better. In doing so, the paper identifies four problem areas to help guide thinking toward clarity about what ‘impact’ entails. To aid collective progress in this space, we detail key issues facing the education and research sectors. Based on our analysis we arrive at a set of questions intended to help guide thinking and actions toward collectively increasing the ability to generate and demonstrate the impact of both into government practice and society at large.

PURPOSE: This review articulates current understanding of the aesthetic, artistic and creative contributions that Dance makes to Health and Wellbeing across the lifecourse within publications 2000-2019, an under-researched area. METHODS: Review Questions: What are the aesthetic, artistic and creative contributions that Dance makes to Health and Wellbeing across the lifecourse? And what methodologies are appropriate for investigating these contributions? A database keyword search identified 769 articles and 91 evaluations. 109 documents were identified for further in-depth analysis and rating, resulting in 24 papers (11 articles, 3 PhD studies, 10 evaluation reports), which were thematically analysed. RESULTS: Findings offer seven interrelated contributions that Dance makes to Health and Wellbeing: embodiment, identity, belonging, self-worth, aesthetics, affective responses and creativity. There was less insight regarding different methodologies, and discussions focused on quantitative data's limitations. There were insights into inclusion of embodied voices, subjective accounts, and lived experiences. CONCLUSION: Whilst acknowledging challenges, this paper illuminates the key contributions of dance to arts and health. It provides a future conceptual research agenda (prioritizing identity and creativity) and associated methodological developments. It recommends expanding geographical/lifecourse research, better defining terms, fuller epistemological critiques to open space for new methodologies, and continued attendance to appropriate rigour criteria.

Robots are increasingly appearing as a potential answer to the ‘care crisis’ facing a number of countries. Although it is anticipated that many positives will flow from the application of these technologies, they are also likely to generate unexpected consequences and risks. This paper explores the use of robots within disability and aged care settings in the Australian and New Zealand contexts. Informed by thirty-five semi-structured interviews with a range of stakeholders, the paper explores why this area is so difficult to govern examining areas identified as generating tensions around the use of robots in care settings. In each of these areas some respondents saw the introduction of robots to be relatively straightforward applications that do not require extensive structures of governance. Others, however, viewed these applications as having potentially greater implications and the need to govern for these over the longer term. The three areas of tension that we explore in this paper relate to independence and surveillance, the re-shaping of human interaction and who can care. These tensions illustrate some of the problems involved in governing robots in a care service context and some of the potentially difficult issues that governments will need to resolve if these technologies are to be effective. We conclude the paper arguing what is needed is a responsive regulation approach to help resolve some of the complexities and tensions in overseeing these technologies.

In recent years, a revolution named Industry 4.0 has arisen. Industry 4.0 is presented as the integration of new advances in areas such as Cyber-Physical Systems, the Internet of Things and Everything (IoE), Cloud computing, the Internet of Services, Big Data Analysis, Smart Factories, Augmented Reality, among others. Industry 4.0 is not only a new industrial revolution, but also a crucial integration challenge that involves several actors from the IoE, which are people, data, services, and things. This paper proposes an approach to analyze the integration challenges in the context of Industry 4.0 using five integration levels, which are connection, communication, coordination, cooperation, and collaboration (5 C). In that sense, this paper presents a state of the art of recent studies in Industry 4.0 from an integration perspective, categorized according to the 5 C integration levels versus the four actors of IoE. Specifically, this paper considers several works intended to solve problems of autonomic integration in Industry 4.0 at the highest levels of the 5 C integration stack (coordination, cooperation, and collaboration). Also, this paper presents a case study from an integration perspective, which contemplates autonomy, self-organizing, among other aspects, in order to turn a traditional industry into a smart factory regarding the Industry 4.0 concept. © 2020, © 2020 Informa UK Limited, trading as Taylor & Francis Group.

This paper introduces the airport shuttle bus scheduling problem (ASBSP) as a new practical scheduling variant. In this problem, a number of identical vehicles that have a specific number of available seats provides transfer service between the airport and the city centre. After making a transfer in one direction, the vehicle can either make a new transfer in the opposite direction depending on the availability and the schedule of the passengers or make an empty return to make a new transfer in the same direction. The vehicles can wait in either location until their next transfer. The passengers have certain time windows for the transfer in relation to their flight times and operational rules to satisfy customer satisfaction. This is a profit-seeking service where transfer requests can also be rejected. The ASBSP aims to prepare a daily schedule for the available vehicles and to assign passengers to these vehicles with the objective of maximising the total profit. This paper presents two alternative mixed integer programming formulations and proposes two valid inequalities to get better bounds. Furthermore, it develops a hybrid metaheuristic that integrates multi-start, simulated annealing and large neighbourhood search for its solution. Extensive computational experiments on real-life benchmark instances have been made to test the performances of the formulations and the hybrid metaheuristic. For spatially continuous systems, it becomes more difficult as the dynamics is infinite dimensional and the well known techniques of systems and control engineering are difficult to apply directly. In this paper, observer design is reported for one-dimensional Burgers equation, which is a non-linear partial differential equation. An infinite dimensional form of the observer is demonstrated to converge asymptotically to the target dynamics, and proper orthogonal decomposition is used to obtain the reduced order observer. When this is done, the corresponding observer is shown to be successful under certain circumstances. The paper unfolds the connections between target dynamics, observer and their finite dimensional counterparts. A set of simulation results has been presented to justify the theoretical claims of the paper.

While lean thinking may help tackle waste, rework remains an ongoing problem during the construction of infrastructure projects. Often too much emphasis is placed on applying lean tools rather than harnessing the human factor and establishing a culture to mitigate rework. Thus, this paper proposes the need for construction organisations to transition from the prevailing error prevention culture (i.e. Quality-I) that pervades practice to one based on error management (i.e. Quality-II) if rework is to be contained and reduced. Accordingly, this paper asks: What type of error culture is required to manage errors that result in rework and to support lean thinking during the construction of infrastructure projects? We draw on the case of a program alliance of 129 water infrastructure projects and make sense of how it enacted, in addition to lean thinking, a change initiative to transition from error prevention to an error management culture to address its rework problem. We observed that leadership, psychological safety and coaching were pivotal for cultivating a culture where there was an acceptance that ‘errors happen’ and effort was directed at mitigating their adverse consequences. The contributions of this paper are twofold as we provide: (1) a new theoretical underpinning to mitigate rework and support the use of lean thinking during the construction of infrastructure projects grounded in Quality-II; and (2) practical suggestions, based on actual experiences, which can be readily employed to monitor and anticipate rework at the coalface of construction.

This paper considers the problem of scheduling periodic maintenance items for oil and gas plants. Each maintenance item involves various maintenance tasks and may require temporary equipment shutdowns, which are costly and highly disruptive to production. The aim is to minimise equipment shutdowns by grouping maintenance items with similar shutdown requirements into short-term maintenance operations called campaigns. Real plants can involve tens of thousands of maintenance items and thus manually scheduling the campaigns is an extreme challenge. In this paper, we develop a mixed-integer linear programming model for optimally allocating maintenance items to campaigns so that total shutdown cost is minimised. The model incorporates constraints on maintenance deadlines, campaign times, maintenance item suppression and labour hours per campaign. We solve the model for realistic scenarios involving data for Karratha Gas Plant in Western Australia, which is the main processing plant for the massive North West Shelf oil and gas project.

This paper investigates the weighted coordinated motion of coupled harmonic oscillators over weighted cooperative–competitive interactions. First, the traditional synchronisation and containment control concepts are extended to the weighted synchronisation and weighted containment control for the new cooperative–competitive networks. Then, weighted synchronisation problem and weighted containment tracking motion are respectively solved for the coupled harmonic oscillators. Compared with some existing results, the frequency parameters of coupled harmonic oscillators are unknown in this paper and some novel networked structures are introduced in order to make the harmonic oscillator followers converge to the convex hull spanned by leaders' weighted states. Particularly, if the topologies formed by every leader group and follower group are interactively balanced and every follower is weight-product balanced, or if some leader groups are not interactively balanced but satisfy some conditions, the coupled harmonic oscillators can all realise the weighted containment tracking motion. Simulation results are given to demonstrate the effectiveness of the proposed control schemes.

We analyse retrospectively how the project development process captured and accompanied several MoC in two piloting areas, where all authors were directly involved. Our work contributes to the body of knowledge that builds on Science and Technology Studies scholarship to enrich the understanding of complex design processes. In particular, the paper goes beyond a descriptive take on MoC and provides ontological and methodological considerations on the use of user stories. In this paper, approximate probabilities of misclassification of the maximum likelihood-based discriminant function are established via an Edgeworth-type expansion based on the standard normal distribution for discriminating between two multivariate normal populations.
