We have developed a cytopathic model using an MTT test for viruses Hantaan 76-118 and for Puumala 967. Our research has shown that ribavirin is not active with the viruses under study, while being fairly toxic to CC50 20 µM. Triazavirin was active for Hantaan 76-118 in a dose of IC50 14±3 µM, and for Puumala 967 IC50 12±3 µM. The toxicity of Triazavirin was 368±18 µM. The applicability of UV spectroscopy to the quantitative determination of ribavirin in aqueous solutions was demonstrated. The existence of a third conformational form of ribavirin was found, and the optimization of three ribavirin conformers was performed using the nonempirical Kohn–Sham method. It was found that, regardless of the confirmation of ribavirin molecules, the absorption maximum of its aqueous solution corresponds to 206 nm. [ABSTRACT FROM AUTHOR]

Objectives: Highly variable genome of HCV in many geographical areas and high prevalence made it necessary to conduct local population studies for therapeutic interventions. In Pakistan, HCV triple therapeutic response has not been reported in Rural, Sindh Province. Therefore, this study has been conducted to highlight newly approved therapy in HCV genotype 3. Materials and Methods: Present study was conducted in the Gambat Institute of Medical Sciences College (GIMS) Pakistan from November 2014 to August 2015. Results: HCV infected patients have been treated with peginterferon, ribavirin and sofosbuvir (triple therapy). The observed risk factors were reused injections, sexual contacts and lack of awareness in general public. The HCV infection ratio was higher in males 75% than in females 25%. Resultantly, 100 patients with 12, 16 and 24 weeks duration treated with sofosbuvir + ribavirin (double oral therapy) resulted 44%, 78%, 94% SVR and 56%, 22%, 6% Non-SVR accordingly. As compared with triple therapy resulted 75%, 80%, 97% SVR and 25%, 20%, 3% Non-SVR accordingly. P-value <0.05 when compared with both groups (SVR & Non-SVR). Conclusion: HCV genotype 3 is fortunate to have an easy to cure genotype but HCV genotype 3 more rapidly progressive liver disease than other genotypes. In this paper, a mathematical model of Hepatitis C virus with saturation incidence rate has been considered. It is a system of ordinary differential equations with four state variables. If basic reproduction number, R0 < 1, disease free equilibrium point is globally stable and endemic equilibrium point does not exist. If R0 > 1, endemic equilibrium point exist and locally asymptotically stable. Numerically, the model succeeded in showing the dynamic behavior of HCV as seen in patients. [ABSTRACT FROM AUTHOR]
