Auranofin, an FDA-approved drug for rheumatoid arthritis, has emerged as a promising antiparasitic medication in recent years. The gold(I) ion in auranofin is postulated to be responsible for its antiparasitic activity. Notably, aurothiomalate and aurothioglucose also contain gold(I), and, like auranofin, they were previously used to treat rheumatoid arthritis. Whether they have antiparasitic activity remains to be elucidated. Herein, we demonstrated that auranofin and similar derivatives, but not aurothiomalate and aurothioglucose, inhibited the growth of Toxoplasma gondii in vitro. We found that auranofin affected the T. gondii biological cycle (lytic cycle) by inhibiting T. gondiis invasion and triggering its egress from the host cell. However, auranofin could not prevent parasite replication once T. gondii resided within the host. Auranofin treatment induced apoptosis in T. gondii parasites, as demonstrated by its reduced size and elevated phosphatidylserine externalization (PS). Notably, the gold from auranofin enters the cytoplasm of T. gondii, as demonstrated by scanning transmission electron microscopy-energy dispersive X-ray spectroscopy (STEM-EDS) and Inductively Coupled Plasma-Mass Spectrometry (ICP-MS).IMPORTANCEToxoplasmosis, caused by Toxoplasma gondii, is a devastating disease affecting the brain and the eyes, frequently affecting immunocompromised individuals. Approximately 60 million people in the United States are already infected with T. gondii, representing a population at-risk of developing toxoplasmosis. Recent advances in treating cancer, autoimmune diseases, and organ transplants have contributed to this at-risk populations exponential growth. Paradoxically, treatments for toxoplasmosis have remained the same for more than 60 years, relying on medications well-known for their bone marrow toxicity and allergic reactions. Discovering new therapies is a priority, and repurposing FDA-approved drugs is an alternative approach to speed up drug discovery. Herein, we report the effect of auranofin, an FDA-approved drug, on the biological cycle of T. gondii and how both the phosphine ligand and the gold molecule determine the anti-parasitic activity of auranofin and other gold compounds. Our studies would contribute to the pipeline of candidate anti-T. gondii agents.

Toxoplasma gondii is a zoonotic parasite that can cause severe morbidity and mortality in warm-blooded animals, including marine mammals such as sea otters. Free-ranging cats can shed environmentally resistant T. gondii oocysts in their feces, which are transported through rain-driven runoff from land to sea. Despite their large population sizes and ability to contribute to environmental oocyst contamination, there are limited studies on T. gondii oocyst shedding by free-ranging cats. We aimed to determine the frequency and genotypes of T. gondii oocysts shed by free-ranging domestic cats in central coastal California and evaluate whether genotypes present in feces are similar to those identified in sea otters that died from fatal toxoplasmosis. We utilized a longitudinal field study of four free-ranging cat colonies to assess oocyst shedding prevalence using microscopy and molecular testing with polymerase chain reaction (PCR). T. gondii DNA was confirmed with primers targeting the ITS1 locus and positive samples were genotyped at the B1 locus. While oocysts were not visualized using microscopy (0/404), we detected T. gondii DNA in 25.9% (94/362) of fecal samples. We genotyped 27 samples at the B1 locus and characterized 13 of these samples at one to three additional loci using multi locus sequence typing (MLST). Parasite DNA detection was significantly higher during the wet season (16.3%, 59/362) compared to the dry season (9.7%; 35/362), suggesting seasonal variation in T. gondii DNA presence in feces. High diversity of T. gondii strains was characterized at the B1 locus, including non-archetypal strains previously associated with sea otter mortalities. Free-ranging cats may thus play an important role in the transmission of virulent T. gondii genotypes that cause morbidity and mortality in marine wildlife. Management of free-ranging cat colonies could reduce environmental contamination with oocysts and subsequent T. gondii infection in endangered marine mammals and people.

Infection directly influences adult hematopoietic stem cell (HSC) function and differentiation, but the fetal hematopoietic response to infection during pregnancy is not well-studied. Here, we investigated the fetal hematopoietic response to maternal infection with Toxoplasma gondii (T. gondii), an intracellular parasite that elicits Type II IFNγ-mediated maternal immunity. While it is known that maternal infection without direct pathogen transmission can affect fetal immune development, the effects of maternal IFNγ on developing HSCs and the signals that mediate these interactions have not been investigated. Our investigation reveals that the fetal HSCs respond to T. gondii infection with virulence-dependent changes in proliferation, self-renewal potential, and lineage output. Furthermore, maternal IFNγ crosses the fetal-maternal interface, where it is perceived by fetal HSCs. By comparing the effects of maternal IFNγ injection with maternal T. gondii infection, we reveal that the effects of IFNγ treatment mimic some aspects of the fetal HSC response to infection. Moreover, our findings illuminate that the fetal HSC response to prenatal infection is distinct from the adult HSC response to IFNγ-induced inflammation. Altogether, our data disentangle the role of infection-induced inflammatory cytokines in driving the expansion of downstream hematopoietic progenitors.

Zoonotic abortive diseases represent a significant health and economic risk for national public health. This cross-sectional survey was conducted from April to October 2021 among 200 selected small ruminant farmers in the three northern regions of Cameroon. Data collection was done through questionnaires administered by exchange with the herder, and responses were coded and recorded on an Excel spreadsheet. The data were then analyzed with R software, version 2.13.0. An ANOVA test was used to assess significant differences in mean of Knowledge, Attitudes, Practices, and Perception of zoonotic risks (KAPP) scores between regions. Pearson correlations were used to calculate the association between KAPP scores within regions. Small ruminant farmers surveyed had low mean scores for knowledge of abortive toxoplasmosis and chlamydophilosis (0.1 ± 0.2), desirable attitude (0.32 ± 0.07), appropriate practice (0.36 ± 0.13) in managing abortion, and positive perception of zoonotic risks of toxoplasmosis and chlamydophilosis in small ruminants (0.12 ± 0.33), respectively. KAPP was significantly (P < 0.01) and positively associated with knowledge (r = 0.98) and risk perception (r = 0.99). However, attitudes (r = 0.06), practices (r = 0.05), and risk perception of toxoplasmosis (r = 0.07) and chlamydophilosis (r = 0.08) were not associated with farmers' knowledge. This study revealed significant knowledge gaps, low levels of desired attitudes, and high-risk behavioral practices. These results therefore call for capacity building of health professionals and farmers to better integrate the One Health concept in the management of neglected zoonotic diseases. Multiple sclerosis (MS) is a chronic, demyelinating disease of the central nervous system that affects mainly young people. It is believed that the autoimmune process observed in the pathogenesis of MS is influenced by a complex interaction between genetic and environmental factors, including infectious agents. The results of this study suggest the protective role of Toxoplasma gondii infections in MS. Interestingly, high Toxoplasma IgM seropositivity in MS patients receiving immunomodulatory drugs (IMDs) was identified. On the other hand, Borrelia infections seem to be positively associated with MS. Although the interpretation of our results is limited by the retrospective nature of the studies, the results strongly indicate that further experimental and clinical studies are needed to explain the role of infectious agents in the development and pathophysiological mechanisms of MS. Toxoplasmosis poses a global health threat, ranging from asymptomatic cases to severe, potentially fatal manifestations, especially in immunocompromised individuals and congenital transmission. Prior research suggests that oregano essential oil (OEO) exhibits diverse biological effects, including antiparasitic activity against Toxoplasma gondii. Given concerns about current treatments, exploring new compounds is important. This study was to assess the toxicity of OEO on BeWo cells and T. gondii tachyzoites, as well as to evaluate its effectiveness in in vitro infection models and determine its direct action on free tachyzoites. OEO toxicity on BeWo cells and T. gondii tachyzoites was assessed by MTT and trypan blue methods, determining cytotoxic concentration (CC50), inhibitory concentration (IC50), and selectivity index (SI). Infection and proliferation indices were analyzed. Direct assessments of the parasite included reactive oxygen species (ROS) levels, mitochondrial membrane potential, necrosis, and apoptosis, as well as electron microscopy. Oregano oil exhibited low cytotoxicity on BeWo cells (CC50: 114.8 µg/mL ± 0.01) and reduced parasite viability (IC50 12.5 ± 0.06 µg/mL), demonstrating 9.18 times greater selectivity for parasites than BeWo cells. OEO treatment significantly decreased intracellular proliferation in infected cells by 84% after 24 h with 50 μg/mL. Mechanistic investigations revealed increased ROS levels, mitochondrial depolarization, and lipid droplet formation, linked to autophagy induction and plasma membrane permeabilization. These alterations, observed through electron microscopy, suggested a necrotic process confirmed by propidium iodide labeling. Pyrimethamine (PYM) is an antiparasitic drug used in the treatment of congenital toxoplasmosis, available only as tablets. Therefore, preparing extemporaneous formulations and their quality control is essential for safe pediatric treatment. This study aimed to develop and validate a stability-indicating method by ultra-performance liquid chromatography (UPLC) to quantify PYM and methylparaben (MP) in extemporaneous oral suspension and evaluate its applicability to the tablets assay. The analytical conditions included RP-C18 column, mobile phase composed of 0.05 M acetate buffer pH 4.5 and organic mixture (acetonitrile:methanol), at 60:40 (v/v), flow rate of 0.2 mL/min, injection volume of 2 µL and detection at 272 nm. PYM was detected at 3.9 min and MP at 4.7 min, without interference of the degradation products or other excipients. Linearity was observed in the range of 0.5 to 25 μg/mL. The method showed accuracy (mean recovery between 98.0 and 102.0%) and precision (RSD < 2%) for suspension and tablets; the method was robust regarding the variations in the analytical conditions. The developed method met the requirements of current guidelines, was indicative of stability, and was suitable for PYM determination in extemporaneous preparations and tablets. Simple Summary: Toxoplasmosis is a globally distributed zoonosis caused by Toxoplasma gondii. This protozoan is capable of infecting mammals and birds and can cause significant impacts on human and animal health. Much of what is known about the parasite's biology, the immune mechanisms involved in toxoplasmosis, and the development of diagnostic tests for this protozoan disease is due to the maintenance of the parasite in laboratory mice. However, the increasing debate on the use of biomodels in scientific research has brought about the need to implement the principles of the 3Rs in laboratory protocols, such as experimental infections with T. gondii in murine models. Therefore, this study aimed to establish the humane endpoints for experimental infection with the T. gondii RH strain in Swiss Webster mice. The results presented here seek to disseminate the method we have proposed for the humane finalization of these animals in experimental protocols using this highly virulent strain as a method of refinement that seeks to enhance animal welfare. The highly virulent Toxoplasma gondii RH strain is maintained through successive passages in mice, but there is still a lack of studies that refine these procedures from a 3Rs perspective, where humanitarian ideals aim to minimize the stress, pain, or suffering of the animals used in the research without the loss of results. The aim of this study was to establish humane endpoints in Swiss Webster mice inoculated with the T. gondii RH strain. A total of 52 mice were infected with 5 × 106 tachyzoites/mL and monitored for periods of up to 5 days. The parameters body weight; hair condition; higher than normal body temperature; hypothermia; respiratory function; pain; soft stools or diarrhea; bloody diarrhea; tense, nervous, or in distress during handling; and ascites were recorded daily in score tables. The results showed that prominent piloerection, respiratory function, pain parameters, and ascites are important clinical signs to be used as a cut-off point for implementing euthanasia. The application of this refinement method helped to avoid animal suffering and pain without compromising the number of parasites recovered. We therefore suggest adopting these parameters in research protocols that require the maintenance of the T. gondii RH strain in murine models to avoid and reduce animal suffering. Toxoplasma gondii is a highly prevalent pathogen, reported from almost all geographical regions of the world. Current anti‐T. gondii drugs are not effective enough in immunocompromised patients, encephalitis, chorioretinitis, and congenital toxoplasmosis. Therefore, the prescription of these drugs has been limited. Rose hip oil (RhO) is a natural plant compound, which shows antibacterial, anticancer, and anti‐inflammatory activities. In the current study, the anti‐T. gondii and cell toxicity effects of solid lipid nanoparticles (SLNs) loaded by RhO (RhO‐SLNs) were evaluated. Emulsification sonicated‐homogenization method was used to prepare SLNs. RhO‐SLNs were characterized, and their anti‐T. gondii and cell toxicity effects were evaluated using in vitro analyses. The particle size and the zeta potential of the nanoparticles were 152.09 nm and −15.3 mV nm, respectively. The entrapment efficiency percentage was 79.1%. In the present study, the inhibitory concentration (IC)50 against T. gondii was >1 μg/mL (p‐value <.0001). The cell toxicity assay showed cytotoxicity concentration (CC)50 >10 mg/mL (p‐value =.017). In addition, at least 75% of T. gondii‐infected Vero cells remained alive at concentrations >10 mg/mL. The concentration of 1 mg/mL showed highest anti‐Toxoplasma activity and lowest cell toxicity against the Vero cell. A mortality event involving 23 allied rock‐wallabies (Petrogale assimilis) displaying neurological signs and sudden death occurred in late April to May 2021 in a suburban residential area directly adjacent to Magnetic Island National Park, on Magnetic Island (Yunbenun), North Queensland, Australia. Three allied rock‐wallabies were submitted for necropsy, and in all three cases, the cause of death was disseminated toxoplasmosis. This mortality event was unusual because only a small, localised population of native wallabies inhabiting a periurban area on a tropical island in the Great Barrier Reef World Heritage Area were affected. A disease investigation determined the outbreak was likely linked to the presence of free‐ranging feral and domesticated cats inhabiting the area. There were no significant deaths of other wallabies or wildlife in the same or other parts of Magnetic Island (Yunbenun) at the time of the outbreak. This is the first reported case of toxoplasmosis in allied rock‐wallabies (Petrogale assimilis), and this investigation highlights the importance of protecting native wildlife species from an infectious and potentially fatal parasitic disease. PurposeMaterials and MethodsResultsConclusionTo report a rare case of a toxoplasma chorioretinitis in one eye and peripheral ocular toxocariasis granuloma in the fellow eye of the same patient.Retrospective case report.A 44-year-old male presented with gradual loss of vision over the past weeks. The patient was diagnosed with ocular toxoplasma chorioretinitis recurrence in his right eye and an inactive peripheral granuloma of ocular toxocariasis was found in the left eye. The patient was a farmer who had been involved in animal husbandry throughout his life. The patient was treated with antibiotics for toxoplasma chorioretinitis and kept under follow-up for ocular toxocariasis.Coinfections of zoonoses are related to socioeconomic environment and individual characteristics of the host. Ocular toxoplasmosis and ocular toxocariasis may present with different scenarios. To our knowledge, this is the first case of a coinfection of ocular toxoplasmosis and ocular toxocariasis in the same patient. This study presents an evaluation of seventeen newly produced recombinant trivalent chimeric proteins (containing the same immunodominant fragment of SAG1 and SAG2 of Toxoplasma gondii antigens, and an additional immunodominant fragment of one of the parasite antigens, such as AMA1, GRA1, GRA2, GRA5, GRA6, GRA7, GRA9, LDH2, MAG1, MIC1, MIC3, P35, and ROP1) as a potential alternative to the whole-cell tachyzoite lysate (TLA) used in the detection of infection in small ruminants. These recombinant proteins, obtained by genetic engineering and molecular biology methods, were tested for their reactivity with specific anti-Toxoplasma IgG antibodies contained in serum samples of small ruminants (192 samples of sheep serum and 95 samples of goat serum) using an enzyme-linked immunosorbent assay (ELISA). The reactivity of six recombinant trivalent chimeric proteins (SAG1-SAG2-GRA5, SAG1-SAG2-GRA9, SAG1-SAG2-MIC1, SAG1-SAG2-MIC3, SAG1-SAG2-P35, and SAG1-SAG2-ROP1) with IgG antibodies generated during T. gondii invasion was comparable to the sensitivity of TLA-based IgG ELISA (100%). The obtained results show a strong correlation with the results obtained for TLA. This suggests that these protein preparations may be a potential alternative to TLA used in commercial tests and could be used to develop a cheaper test for the detection of parasite infection in small ruminants. Objective(s): Toxoplasma gondii is a common parasite in the world. Pharmaceutical options for toxoplasmosis treatment are limited. Several studies have been conducted on the anti-infectious properties of miltefosine (MLF). We investigated the effectiveness of nanoemulsion miltefosine (NEM) in tachyzoites of T. gondii, RH strain. Materials and Methods: Various NEM formulations were designed considering pseudo-ternary phase diagrams. Physicochemical properties of the developed nanoemulsions (NEs), including pH, polydispersity index (PDI), droplet size, and refractive index (RI) were evaluated. The considered formulation was analyzed for dilution and stability tests. MTT assay was performed on Vero cells for calculation CC50 and on Vero cells with RH strain tachyzoite for calculation IC50. Sulfadiazine (SDZ) and pyrimethamine (PYR) were positive controls. The trypan blue method was used to investigate the effect of drugs (NEM, MLF, SDZ, PYR) in reducing the number of infected Vero cells and reducing the intracellular proliferation of tachyzoites. Next, the viability of tachyzoites was measured in the tube in the direct vicinity of different drug concentrations. Results: The final particle size of NEM was calculated to be 17.463 nm by DLS and TEM. The CC50 of NEM (75.7 µg/mL) indicated lower toxicity than the other drugs. IC50 obtained by trypan blue, MTT, and test tube methods showed that NEM (28.43 µg/mL) has a suitable IC50 against Toxoplasma tachyzoites. Conclusion: The calculated selectivity index (SI) demonstrated that NEM (SI=2.66) is a more suitable drug candidate than the MLF and positive controls. Toxoplasma gondii is a pervasive protozoan parasite that is responsible for significant zoonoses. A wide array of vaccines using different effector molecules of T. gondii have been studied worldwide to control toxoplasmosis. None of the existing vaccines are sufficiently effective to confer protective immunity. Among the different Toxoplasma -derived effector molecules, T. gondii dense granule protein 15 from the type II strain (GRA15 (II)) was recently characterized as an immunomodulatory molecule that induced host immunity via NF-κB. Therefore, we assessed the immunostimulatory and protective efficacy of recombinant GRA15 (II) (rGRA15) against T. gondii infection in a C57BL/6 mouse model. We observed that rGRA15 treatment increased the production of IL-12p40 from mouse peritoneal macrophages in vitro. Immunization of mice with rGRA15 induced the production of anti-TgGRA15-specific IgG, IgG1 and IgG2c antibodies. The rGRA15-sensitized spleen cells from mice inoculated with the same antigen strongly promoted spleen cell proliferation and IFN-γ production. Immunization with rGRA15 significantly enhanced the survival rate of mice and dramatically decreased parasite burden in mice challenged with the Pru (type II) strain. These results suggested that rGRA15 triggered humoral and cellular immune responses to control infection. However, all of the immunized mice died when challenged with the GRA15-deficient Pru strain or the RH (type I) strain. These results suggest that GRA15 (II)-dependent immunity plays a crucial role in protection against challenge infection with the type II strain of T. gondii. Background: Congenital toxoplasmosis (CT) can be accompanied by serious organ manifestations, particularly retinochoroiditis, and may occur throughout life. We aimed to monitor long-term ocular prognosis in a large French cohort of patients with CT and its changes over time in the context of mandatory prenatal screening (since 1992) and incidence decrease since 2008. Methods: Patients with CT diagnosed between 1987 and 2021 were prospectively included and followed for up to 35 years. The effect of the period of conception on the risk of first retinochoroiditis has been tested using a flexible extension of the Cox model. Incidence rates of retinochoroiditis were estimated. Results: A total of 646 infected live born children were followed for a median of 12 years (range, 0.5-35); 187 patients (29%) had at least 1 ocular lesion (first at a median age of 5 years; range, 0-26 years) with peaks at 7 and 12 years. Early maternal infection and the presence of nonocular signs at birth were associated with a higher risk of retinochoroiditis, whereas delayed diagnosis of CT (after birth versus before or at birth) was associated with a lower risk (13% decrease for each additional month after birth; P = .01). A period effect for the risk of developing retinochoroiditis in patients born after 2008 was not detected. Conclusions: Despite prenatal screening and prolonged perinatal treatment, retinochoroiditis is not a rare event in French patients with CT and can occur well into adulthood, with peak incidences at 7 and 12 years of age. Toxoplasma is an intracellular parasite that can cause toxoplasmosis in humans and animals. In this disease, parasitic cysts of Toxoplasma gondii can remain present in various tissues such as the brain throughout the host's life. The potential role of toxoplasmosis should be considered in many neurological diseases with unknown mechanisms. In horses, behavioral disorders like crib-biting have multifactorial causes and may be due to neurophysiological dysfunction. This research aimed to determine the role of toxoplasmosis in the manifestation of crib-biting behavior in horses. A casecontrol study was conducted in horse riding clubs in Fars province, Iran, near the city of Shiraz. Ten horses with crib-biting behavior and 10 clinically healthy horses matched for sex, age, and breed were enrolled in the study. Blood samples were collected from cribbers and healthy horses, along with a thorough history-taking. An enzyme-linked immunosorbent assay (ELISA) was performed on sera to detect T. gondii-specific Immunoglobulin G (IgG) and Immunoglobulin M (IgM). The results of this study revealed that all horses were free of toxoplasmosis. It is concluded that this relation was not detected in the horse farms of Shiraz suburbs. Toxoplasmosis is a zoonotic disease caused by Toxoplasma gondii, which can infect humans through oocysts or undercooked meat. It can cause varying symptoms, including congenital toxoplasmosis. Early detection and treatment are beneficial, and antimicrobial treatment can prevent or resolve symptoms. The disease has a complex life cycle, with felids being the definitive host. Understanding the signaling pathways is crucial for effective therapeutic strategies. Toxoplasma invasion is regulated by the microtubule cytoskeleton, affecting macrophages and innate immunity cells. Calcium binding proteins and focal adhesion kinase-2 have been identified as key regulators of calcium signaling in Toxoplasma. Calcium signaling is crucial for parasite biology and drug development. The ERK pathway plays a significant role in host-parasite interactions and immune responses. This pathway plays a critical role in the spread of Toxoplasma by manipulating host cell migration. Toxoplasma infection can activate the ERK signaling pathway, leading to the inhibition of apoptosis in host cells. This inhibition of apoptosis is believed to have a positive effect on the survival and replication of the parasite in the host. The Akt signaling pathway, also known as the PI3K/Akt pathway, is crucial in parasitic diseases, modulating host immune responses and parasite survival. Host AKT activation is important for T. gondii proliferation which is related to reduction of ROS in host cells. More investigation is required to fully understand how these signals contribute to the pathophysiology of Toxoplasma infection and to identify possible therapeutic targets for the management of parasitic illnesses. Background. The present study examined the relationship between latent toxoplasmosis and mental disorders. Methods. The sample included 117 toxoplasma-positive volunteers on Facebook, Twitter, and Instagram. Data were collected online using the 71-item abbreviated form of the Minnesota Multiphasic Personality Inventory (MMPI). Results. According to the results, a) the psychological profile of latent toxoplasmosis patients in the eight clinical scales of MMPI was within the normal range; b) in the male group, ‘duration of infection’ significantly predicted the scores for the four clinical scales of depression, psychopathic deviate, psychasthenia, and schizophrenia; also, ‘age’ predicted the scores for hypochondriasis and schizophrenia in men; c) in the female group, only ‘age’ was able to predict the scores for the scales of hysteria, paranoia, mental weakness, and social deviance; and d) there was no significant difference between male and female groups in any of the clinical scales. Conclusion. Duration of infection with latent toxoplasmosis was associated with depression, psychopathtic deviation, psychasthenia, and schizophrenia in infected males, but this was not the case for the infected females in any of the clinical scales of the 71- item abbreviated form of MMPI. In mental health screening of individuals with latent toxoplasmosis, males should be screened on the scales of depression, psychopathtic deviation, psychasthenia, and schizophrenia according to the increase in the duration of infection. The current study aimed to determine the prevalence of Toxoplasma gondii in ABO blood groups and assess the relationship between the prevalence of T. gondii and blood groups. A literature search was carried out for epidemiological studies that were published through December 2022. A random effects model was used to determine the OR and the pooled prevalence with a 95% CI. The estimated pooled prevalences of T. gondii infection in the A, B, AB and O blood groups were 38% (95% CI 27 to 48%), 38% (95% CI 29 to 47%), 36% (95% CI 26 to 45%) and 36% (95% CI 27 to 45%), respectively. Also, the pooled ORs of the relationship between the prevalence of T. gondii infection and the A, B, AB and O blood groups were 1.08 (95% CI 0.97 to 1.19), 1.10 (95% CI 0.95 to 1.28), 1.08 (95% CI 0.92 to 1.27) and 0.89 (95% CI 0.80 to 1.00), respectively. Sera from 391 waterbirds from eight USA states were tested for Toxoplasma gondii antibodies using the modified agglutination test. Fifteen different waterbird species (26.6%; n=104) were seropositive. Toxoplasmosis continues to be a prevalent parasitic zoonosis with a global distribution. This disease is caused by an intracellular parasite known as Toxoplasma gondii, and the development of effective novel drug targets to combat it is imperative. There is limited information available on the potential advantages of wheat germ oil (WGO) and propolis, both individually and in combination, against the acute phase of toxoplasmosis. In this study, acute toxoplasmosis was induced in Swiss albino mice, followed by the treatment of infected animals with WGO and propolis, either separately or in combination. After 10 days of experimental infection and treatment, mice from all groups were sacrificed, and their brains, uteri, and kidneys were excised for histopathological assessment. Additionally, the average parasite load in the brain was determined through parasitological assessment, and quantification of the parasite was performed using Real-Time Polymerase Chain Reaction targeting gene amplification. Remarkably, the study found that treating infected animals with wheat germ oil and propolis significantly reduced the parasite load compared to the control group that was infected but not treated. Moreover, the group treated with a combination of wheat germ oil and propolis exhibited a markedly greater reduction in parasitic load compared to the other groups. Similarly, the combination treatment effectively restored the histopathological changes observed in the brain, uterus, and kidney, and the scoring of these reported lesions confirmed these findings. Background: Toxoplasmosis is considered one of the most common public health troubles. Among these health troubles, toxoplasmosis was recently linked to many neuropsychiatric and behavioral disorders, especially autism spectrum disorder (ASD). The purpose of this study was to determine the seroprevalence of toxoplasmosis in children with ASD. Methods: The study was conducted on 100 children, grouped in two groups; group 1 (patient group) included 50 children with ASD and group 2 (control group) included 50 healthy children. ASD in the patient group was diagnosed according to DSM 5 criteria of ASD. Every child involved in this study underwent a history taking, a clinical examination, and laboratory investigations to detect serum anti-Toxoplasma IgG and IgM antibodies using ELISA. Children of the patient group were further assessed using the Childhood Autism Rating Scale to evaluate the severity of their symptoms. Results: The seroprevalence of IgG among ASD children was highly significant compared to the healthy children. The detected difference between the 2 groups regarding seroprevalence of anti-Toxoplasma IgM antibodies was insignificant. No significant correlation could be demonstrated between Toxoplasma infection and severity of autistic symptoms in the ASD group. Furthermore, the study revealed an increase in anti-Toxoplasma IgG antibodies in ASD children with positive family history of ASD rather than those with no such history. In addition, an increase in seroprevalence of both anti-Toxoplasma antibodies among children with low socioeconomic standards compared to children with moderate or high standards. PurposeMethodsResultsConclusionTo describe the effect of long-term, low-dose pyrimethamine for the prevention of ocular toxoplasmosis (OT) recurrences.Sixty-three consecutive patients with inactive ocular toxoplasmosis and positive toxoplasma IgG serology were included. Pyrimethamine (25 mg) + folinic acid (15 mg) were administered every other day (three times weekly) for 12 months. Eighteen patients received the treatment for an additional six months as part of an extension study.Thirty-eight patients (60.3%, n = 63) were female; 38 (60.3%) had a previous history of recurrence and 37 (58.7%) had active OT within the preceding 12 months. Three (4.8%) patients had unilateral recurrences at 8, 12 and 18 months after starting intermittent pyrimethamine treatment. Five patients (7.9%) were discontinued due to hematological, renal and hepatic changes. Treatment was considered successful in 42 patients (84%).Long-term, low-dose pyrimethamine can be considered as a treatment option for the prevention of ocular toxoplasmosis recurrence in selected patients, with only a few, mild and reversible systemic adverse events. Toxoplasmosis is a zoonosis caused by Toxoplasma gondii (T. gondii). The current treatment for toxoplasmosis remains constrained due to the absence of pharmaceutical interventions. Thus, the pursuit of more efficient targets is of great importance. Lipid metabolism in T. gondii, including fatty acid metabolism, phospholipid metabolism, and neutral lipid metabolism, assumes a crucial function in T. gondii because those pathways are largely involved in the formation of the membranous structure and cellular processes such as division, invasion, egress, replication, and apoptosis. The inhibitors of T. gondii's lipid metabolism can directly lead to the disturbance of various lipid component levels and serious destruction of membrane structure, ultimately leading to the death of the parasites. In this review, the specific lipid metabolism pathways, correlative enzymes, and inhibitors of lipid metabolism of T. gondii are elaborated in detail to generate novel ideas for the development of anti-T. gondii drugs that target the parasites' lipid metabolism. In 2023, the European Conference on Infections in Leukaemia elaborated guidelines for managing toxoplasmosis in hematology patients, mainly after allogeneic hematopoietic cell transplantation. Toxoplasma disease, although a rare event, has a high mortality, especially in case of cerebral involvement or disseminated form, and may be of very acute presentation. These guidelines highlight the need for a preemptive strategy based on a weekly screening by quantitative PCR from transplantation in seropositive recipients, allowing to early treat reactivation before the occurrence of toxoplasma disease. The purpose of this article is not to summarize these guidelines but rather to discuss critical issues which have been largely debated during the elaboration of these guidelines and which support the conclusions of the group. Background: Type 2 diabetes mellitus (T2DM) is prone to opportunistic infections, including toxoplasmosis, due to an immunodeficiency system. This study aimed to evaluate the serum of people with T2DM to determine the titer of anti-toxoplasma antibodies in patients and compare it with the control group. Materials and Methods: 720 blood samples have been carried out between October and the end of January 2017 in Sistan, and Baluchestan provinces in southeastern Iran, of these, 360 samples were related to healthy individuals (control), and 360 samples were related to T2DM individuals. The immunoglobulin (Ig) M and IgG enzyme-linked immunosorbent assay methods have been used to detect toxoplasmosis. The data were analyzed using SPSS-19, Chi-square, and Fisher's exact test to compare statistical parameters. Results: In this cross-sectional study, out of 360 samples of T2DM by ELISA method, 60% samples in diabetic patients and 48.1% in control group were IgG positive (P < 0.05). Nearly 2.5% samples in diabetic patients and 0.3% in control group were IgM positive (P < 0.05). Background: Toxoplasmosis is a worldwide parasitic zoonosis caused by the protozoan Toxoplasma gondii. In cases of vertical infection, and in immunosuppressed people by the human immunodeficiency virus (HIV) serious clinical conditions may appear, while immunocompetent people do not present symptoms. However, T. gondii infection has been linked to several mental disorders for decades. Objective: To substantiate the possible relationship between T. gondii and mental disorders and suggest control and prevention strategies. Material and Methods: A systematic review has been carried out to analyze the relationship between T. gondii exposure (presence of IgG) and the onset of mental disorders in minors and adults. The etiopathogenic mechanisms described by the authors have also been included and the systems of surveillance, prevention and control of infection have been evaluated. Results: Several processes linked to the presence of cysts and the reactivation of the parasite in certain situations produce an immune and inflammatory response. Also, direct and indirect actions on different neurotransmitters. These mechanisms, together with other environmental and genetic factors, would predispose to different psychiatric pathologies. Conclusions: Due to the limits of the study, no conclusions can be drawn in childhood and adolescence. However, the results of this systematic review show a possible association of schizophrenia, bipolar disorder and compul-sive disorder with T. gondii infection in adults. Toxoplasma gondii infection has been correlated with an increased risk of psychosis, but the connection between T. gondii and psychosis-like symptoms in the general adult population remains unclear. This paper analyzes the case of a 36-year-old patient with psychiatric history who was hospitalized after a suicide attempt by medication ingestion. Serological lab test results indicated the presence of T. gondii antibodies, and psychiatric evaluation revealed symptoms such as auditory hallucinations, delusional ideas, and psychotic anxiety. Magnetic resonance imaging identified a demyelinating lesion in the right frontal area, and ophthalmologic examination revealed ocular abnormalities: lessening of nervous fibres in the temporal region of the retina, three macular and juxta-macular lesions. The formulated diagnosis was acute psychotic disorder secondary to a general medical condition. The patient's treatment included Olanzapine, Sertraline, and Diazepam, with good response. The discussion explores the potential link between toxoplasmosis and psychiatric manifestations, supporting the hypothesis of a causal relation. BACKGROUND: In Bulgaria, toxoplasmosis is one of the most common infections in humans but the mediators of the T-cell immune response have not been studied. This study aimed to follow-up on the cytokine profile in some groups of immunocompetent patients with evidence of acute and chronic toxoplasmosis. Methods: T. gondii antibodies IgG, IgM, and IgA in the serum samples were measured by ELISA. Cytokine profile was investigated with commercial kit 13-plex ProcartaPlex™, Invitrogen, suitable for apparatus Luminex 200. RESULTS: We examined cytokine profiles and specific Toxoplasma antibodies in 60 immunocompetent patients divided into 3 groups: pregnant women (N.=31), Patients with lymphadenitis, eye pathology, and fever of unknown origin (N.=17), and Persons examined at their request (N.=12). A statistically significant difference was found when comparing levels of cytokines IFN-γ, IL-10, IL-2, IL-27, IL-4, and IP-10 between the group of pregnant women with acute infection and the group of non-infected pregnant women. In the other two groups of patients, we did not find a statistically significant difference in the values of the investigated cytokines. CONCLUSIONS: These results indicate lower levels of cytokines related to both Th1 and Th2 in pregnant women with acute toxoplasmosis. Toxoplasma gondii is an obligate intracellular parasite existing in three infectious life stages—tachyzoites, bradyzoites, and sporozoites. Rupture of tissue cysts and re-conversion of bradyzoites to tachyzoites leads to reactivated toxoplasmosis (RT) in an immunocompromised host. The aim of this study was to apply ImageJ software for analysis of T. gondii brain cysts obtained from a newly established in vivo model of RT. Mice chronically infected with T. gondii (BGD1 and BGD26 strains) were treated with cyclophosphamide and hydrocortisone (experimental group—EG) or left untreated as infection controls (ICs). RT in mice was confirmed by qPCR (PCR+); mice remaining chronically infected were PCR−. A total of 90 images of cysts were analyzed for fractal dimension (FD), lacunarity (L), diameter (D), circularity (C), and packing density (PD). Circularity was significantly higher in PCR+ compared to IC mice (p < 0.05 for BGD1, p < 0.001 for the BGD26 strain). A significant negative correlation between D and PD was observed only in IC for the BGD1 strain (ρ = −0.384, p = 0.048), while fractal parameters were stable. Significantly higher D, C, and PD and lower lacunarity, L, were noticed in the BGD1 compared to the more aggressive BGD26 strain. In conclusion, these results demonstrate the complexity of structural alterations of T. gondii cysts in an immunocompromised host and emphasize the application potential of ImageJ in the experimental models of toxoplasmosis. A few reports have been published and documented low level of awareness on toxoplasmosis among Saudi women. Herein, a cross sectional community based study was undertaken to evaluate basic knowledge on toxoplasmosis among residents in the Eastern province (Sharqiyah). Thisstudy was conducted between December 2022 and January 2023 on 334 females from different ages and educational backgrounds. Analysis of their responses revealed that only (24.9%) had heard about the disease. However, (69.8%) properly identified cats as the source of Toxoplasma gondii (T. gondii), but a smaller percentage (47.7%) knew that they might become infected through handling cat feces, and a few (26.3%) believed that bad hand hygiene can result in T. gondii infection. A few males (n = 26) have also participated, for the first time in Saudi Arabia, and displayed also low level of knowledge on toxoplasmosis. We do recommend establishing educational programs for females, in various Saudi provinces, to raise awareness on toxoplasmosis. [ABSTRACT FROM AUTHOR]

Toxoplasmosis, which is caused by the Toxoplasma gondii parasite, is a parasitic, infectious disease. 10-hydroxy-2-decenoic acid (10-H2DA, queen bee acid (QBA), is one of the most prevalent fatty acids (>40%) present in royal jelly. Studies have pointed to antitumor, anti-inflammatory, antiangiogenic, and antimicrobial effects of 10-H2DA, improving the immune system. This experimental survey aimed to assess the in vitro efficacy of QBA against tachyzoites and intracellular parasites of the T. gondii RH strain. Anti-Toxoplasma effects of QBA against tachyzoites were examined by MTT (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide) assay for 30, 60, 120, and 180 min. In addition, the effect of QBA on infection rate and intracellular parasites was studied. Real-time polymerase chain reaction (Real-Time PCR) was also applied to assess the expression level of the Caspase-3 gene. The best efficiency of QBA was obtained at 100 and 50 µg/mL, whereas all tachyzoites were diminished, followed by 120- and 180-min treatment, respectively. It was also found that the best repressing efficacy of QBA in the infection rate and the load of parasites into the Vero cells was indicated at 100 µg/mL (P<0.001). Nonetheless, the combination of QBA (12.5 µg/mL) along with atovaquone 30 µg/mL displayed the most marked effect on the infection rate and a load of parasites into the Vero cells in the infected Vero cells. The expression level of the Caspase-3 gene was dose-dependently increased after the exposure of tachyzoites to QBA, mainly at ½ IC50 and IC50 compared to normal saline. The obtained findings exhibited the high in vitro potency of QBA, especially in combination with atovaquone against T. gondii RH strain tachyzoites. Although apoptosis induction can be suggested as one of the principle mechanisms, more studies are required to elucidate its accurate mechanisms, as well as its efficacy and safety in animal models and clinical settings. Toxoplasmosis is a zoonotic disease with a worldwide prevalence that is caused by Toxoplasma gondii. This study aimed to summarize available data on genotyping T. gondii strains based on the GRA6 gene marker in different hosts around the world. We conducted a comprehensive literature search using five international databases (PubMed, Scopus, Science Direct, Web of Science, and Google Scholar) from inception until December 2021. We identified 32 papers eligible for inclusion in this systematic review. The majority of studies (50%) were carried out in Iran (n = 16) to identify T. gondii genotypes based on the GRA6 gene. Other countries with reported studies include China, Japan, Sweden, and Italy (n = 2 each). Out of 3,434 samples collected from various hosts, most studies (n = 11) focused on human samples (34.4%), followed by ovine (n = 7), pig (n = 4), goat (n = 3) and soil and cattle (n = 2). Using various molecular methods such as conventional PCR, nested-PCR, real-time PCR, microsatellite analysis, and Restriction Fragment Length Polymorphism (RFLP), we found DNA positive results in 805 out of 3,434 samples. Of these, 285 (35.40%), 207 (25.71%), 182 (22.60%), 65 (8.07%), and 18 (2.23%) were infected with types I, II, III, mix I, II, III, and mix II, III, respectively. Our data demonstrate that the GRA6 gene marker has sufficient polymorphism to detect three types of T. gondii genotypes in various hosts. Identifying the specific genotype could be valuable in developing new strategies for treatment, vaccination, diagnosis, control, and prevention of T. gondii infection. [ABSTRACT FROM AUTHOR]

Abstract Multiple sclerosis (MS) is a chronic, demyelinating disease of the central nervous system that affects mainly young people. It is believed that the autoimmune process observed in the pathogenesis of MS is influenced by a complex interaction between genetic and environmental factors, including infectious agents. The results of this study suggest the protective role of Toxoplasma gondii infections in MS. Interestingly, high Toxoplasma IgM seropositivity in MS patients receiving immunomodulatory drugs (IMDs) was identified. On the other hand, Borrelia infections seem to be positively associated with MS. Although the interpretation of our results is limited by the retrospective nature of the studies, the results strongly indicate that further experimental and clinical studies are needed to explain the role of infectious agents in the development and pathophysiological mechanisms of MS.

Abstract Toxoplasma gondii is a highly prevalent pathogen, reported from almost all geographical regions of the world. Current anti‐T. gondii drugs are not effective enough in immunocompromised patients, encephalitis, chorioretinitis, and congenital toxoplasmosis. Therefore, the prescription of these drugs has been limited. Rose hip oil (RhO) is a natural plant compound, which shows antibacterial, anticancer, and anti‐inflammatory activities. In the current study, the anti‐T. gondii and cell toxicity effects of solid lipid nanoparticles (SLNs) loaded by RhO (RhO‐SLNs) were evaluated. Emulsification sonicated‐homogenization method was used to prepare SLNs. RhO‐SLNs were characterized, and their anti‐T. gondii and cell toxicity effects were evaluated using in vitro analyses. The particle size and the zeta potential of the nanoparticles were 152.09 nm and −15.3 mV nm, respectively. The entrapment efficiency percentage was 79.1%. In the present study, the inhibitory concentration (IC)50 against T. gondii was >1 μg/mL (p‐value 10 mg/mL (p‐value = .017). In addition, at least 75% of T. gondii‐infected Vero cells remained alive at concentrations >10 mg/mL. The concentration of 1 mg/mL showed highest anti‐Toxoplasma activity and lowest cell toxicity against the Vero cell. Our findings suggest that carrying natural plant compounds with SLNs could be considered an effective option for treatment strategies against T. gondii infections.

In a representative sample of female children and adolescents in Germany, Toxoplasma gondii seroprevalence was 6.3% (95% CI 4.7%–8.0%). With each year of life, the chance of being seropositive increased by 1.2, indicating a strong force of infection. Social status and municipality size were found to be associated with seropositivity.

Background. The present study examined the relationship between latent toxoplasmosis and mental disorders. Methods. The sample included 117 toxoplasma-positive volunteers on Facebook, Twitter, and Instagram. Data were collected online using the 71-item abbreviated form of the Minnesota Multiphasic Personality Inventory (MMPI). Results. According to the results, a) the psychological profile of latent toxoplasmosis patients in the eight clinical scales of MMPI was within the normal range; b) in the male group, ‘duration of infection’ significantly predicted the scores for the four clinical scales of depression, psychopathic deviate, psychasthenia, and schizophrenia; also, ‘age’ predicted the scores for hypochondriasis and schizophrenia in men; c) in the female group, only ‘age’ was able to predict the scores for the scales of hysteria, paranoia, mental weakness, and social deviance; and d) there was no significant difference between male and female groups in any of the clinical scales. Conclusion. Duration of infection with latent toxoplasmosis was associated with depression, psychopathtic deviation, psychasthenia, and schizophrenia in infected males, but this was not the case for the infected females in any of the clinical scales of the 71-item abbreviated form of MMPI. The Toxoplasma inner membrane complex (IMC) is a unique organelle that plays critical roles in parasite motility, invasion, egress, and replication. The IMC is delineated into the apical, body, and basal regions, defined by proteins that localize to these distinct subcompartments. The IMC can be further segregated by proteins that localize specifically to the maternal IMC, the daughter bud IMC, or both. While the function of the maternal IMC has been better characterized, the precise roles of most daughter IMC components remain poorly understood. Here, we demonstrate that the daughter protein IMC29 plays an important role in parasite replication. We show that Δimc29 parasites exhibit severe replication defects, resulting in substantial growth defects and loss of virulence. Deletion analyses revealed that IMC29 localization is largely dependent on the N-terminal half of the protein containing four predicted coiled-coil domains while IMC29 function requires a short C-terminal helical region. Using proximity labeling, we identify eight novel IMC proteins enriched in daughter buds, significantly expanding the daughter IMC proteome. We additionally report four novel proteins with unique localizations to the interface between two parasites or to the outer face of the IMC, exposing new subregions of the organelle. Together, this work establishes IMC29 as an important early daughter bud component of replication and uncovers an array of new IMC proteins that provides important insights into this organelle. IMPORTANCE The inner membrane complex (IMC) is a conserved structure across the Apicomplexa phylum, which includes obligate intracellular parasites that cause toxoplasmosis, malaria, and cryptosporidiosis. The IMC is critical for the parasite to maintain its intracellular lifestyle, particularly in providing a scaffold for daughter bud formation during parasite replication. While many IMC proteins in the later stages of division have been identified, components of the early stages of division remain unknown. Here, we focus on the early daughter protein IMC29, demonstrating that it is crucial for faithful parasite replication and identifying specific regions of the protein that are important for its localization and function. We additionally use proximity labeling to reveal a suite of daughter-enriched IMC proteins, which represent promising candidates to further explore this IMC subcompartment.

Toxoplasma gondii is a ubiquitous zoonotic parasite that can infect warm-blooded vertebrates, including humans. Felids, the definitive hosts, drive T. gondii infections by shedding the environmentally resistant stage of the parasite (oocysts) in their feces. Few studies characterize the role of climate and anthropogenic factors in oocyst shedding among free-ranging felids, which are responsible for the majority of environmental contamination. We determined how climate and anthropogenic factors influence oocyst shedding in free-ranging domestic cats and wild felids using generalized linear mixed models. T. gondii oocyst shedding data from 47 studies were systematically reviewed and compiled for domestic cats and six wild felid species, encompassing 256 positives out of 9,635 total fecal samples. Shedding prevalence in domestic cats and wild felids was positively associated with human population density at the sampling location. Larger mean diurnal temperature range was associated with more shedding among domestic cats and warmer temperature in the driest quarter was associated with lower oocyst shedding in wild felids. Increasing human population density and temperature fluctuation can exacerbate environmental contamination with the protozoan parasite T. gondii. Management of free-ranging domestic cats could lower the burden of environmental oocysts due to their large population sizes and affinity with human settlements.

Abstract Background The wild boar (Sus scrofa) and the Apennine wolf (Canis lupus italicus) are two wild species that have both increased their presence in the Italian territory, albeit in varying numbers. They can be occasionally found in peri-urban areas as well. Both of these species can serve as intermediate hosts for Toxoplasma gondii, as they can become infected either through the consumption of oocysts found in water, soil, or on vegetables, or through the ingestion of meat containing bradyzoites. Consequently, these animals can be regarded as key indicators of Toxoplasma presence in the wild or peri-urban environment. In our study, we examined a total of 174 wild boar meat juice and 128 wolf sera from Italy for the detection of T. gondii IgG using the indirect fluorescent antibody test (IFAT). Results The results showed that 40 (22.6%) of the wild boar meat juice and 34 (26.6%) of the wolf serum samples tested positive. Interestingly, there were no significant differences in seropositivity with respect to gender, age group, or the region of origin in both species. Conclusions Overall the results indicate a moderate exposure in both the species under investigation, highlighting the spread of T. gondii in sylvatic and periurban environments. The prevalence of T. gondii in wild boar is consistent with findings from other studies conducted in Europe. Our study, with a considerably larger sample size compared to the available research in European context, provides valuable data on the seroprevalence of T. gondii in wolves.

Abstract Background In September 2014, there was sudden upsurge in the number of Eurasian red squirrels (Sciurus vulgaris) found dead in the Netherlands. High infection levels with the parasite Toxoplasma gondii were demonstrated, but it was unclear what had caused this increase in cases of fatal toxoplasmosis. In the present study, we aimed to gain more knowledge on the pathology and prevalence of T. gondii infections in Eurasian red squirrels in the Netherlands, on the T. gondii genotypes present, and on the determinants of the spatiotemporal variability in these T. gondii infections. The presence of the closely related parasite Hammondia hammondi was also determined. Methods Eurasian red squirrels that were found dead in the wild or that had died in wildlife rescue centres in the Netherlands over a period of seven years (2014–2020) were examined. Quantitative real-time polymerase chain reaction was conducted to analyse tissue samples for the presence of T. gondii and H. hammondi DNA. Toxoplasma gondii-positive samples were subjected to microsatellite typing and cluster analysis. A mixed logistic regression was used to identify climatic and other environmental predictors of T. gondii infection in the squirrels. Results A total of 178 squirrels were examined (49/178 T. gondii positive, 5/178 H. hammondi positive). Inflammation of multiple organs was the cause of death in 29 squirrels, of which 24 were also T. gondii polymerase chain reaction positive. Toxoplasma gondii infection was positively associated with pneumonia and hepatitis. Microsatellite typing revealed only T. gondii type II alleles. Toxoplasma gondii infection rates showed a positive correlation with the number of days of heavy rainfall in the previous 12 months. Conversely, they showed a negative association with the number of hot days within the 2-week period preceding the sampling date, as well as with the percentage of deciduous forest cover at the sampling site. Conclusions Toxoplasma gondii infection in the squirrels appeared to pose a significant risk of acute mortality. The T. gondii genotype detected in this study is commonly found across Europe. The reasons for the unusually high infection rates and severe symptoms of these squirrels from the Netherlands remain unclear. The prevalence of T. gondii in the deceased squirrels was linked to specific environmental factors. However, whether the increase in the number of dead squirrels indicated a higher environmental contamination with T. gondii oocysts has yet to be established. Graphical Abstract

Objetivo: describir los resultados perinatales de gestantes con sospecha de seroconversión o confirmación de toxoplasmosis gestacional y la evolución clínica de sus neonatos hasta el mes de vida en una institución de alta complejidad, 2015-2021.Materiales y métodos: estudio descriptivo retrospectivo que incluyó gestantes quienes fueron referidas al servicio de medicina materno fetal por toxoplasmosis gestacional. Se tuvo en cuenta serologías positivas para esta patología, hallazgos ecográficos, reacción en cadena de polimerasa en líquido amniótico y serologías del neonato. La información de las variables clínicas y resultados de prueba serológicas se recolectó a través de historias clínicas. Resultados: Se incluyeron 209 pacientes, la mediana de edad gestacional al momento de la realización de las pruebas serológicas en institución de atención inicial, fue 16 semanas (RIC: 10-24 semanas). Los resultados serológicos fueron positivos para Inmunoglobulina G (IgG) en 191/ 208 (91,8%) y para inmunoglobulina M (IgM) 194/209 (92,8%). De las pacientes con IgG e IgM positivas, se le realizó prueba de avidez a 60 (28,7%), la cual fue baja en 15 (26,8%) pacientes. A 76 (36,4%) gestantes se les realizó la prueba de reacción en cadena de polimerasa en líquido amniótico, de estas, 15 (19,7%) tuvieron resultados positivos para diagnóstico de toxoplasmosis con-génita, de las cuales en nueve casos los recién nacidos presentaron hallazgos positivos en el examen oftalmológico inicial y / o en la ecografía transfontanelar, confirmando los hallazgos ecográficos prenatales.La ecografía de detalle anatómico se realizó en 188 (90%) de las gestantes, donde se encontró alguna alteración en 47 (25,0%) de los casos. Conclusiones: La interpretación serológica y la ecografía obstétrica permitieron identificar una población de gestantes con la infección a quien se le ofreció el tratamiento disponible, sin embargo, existieron casos de toxoplasmosis congénita en ocasiones fueron fatales o con compromiso neurológico grave en la población estudiada.

Abstract Background Toxoplasma gondii is a parasitic protozoan that can infect a wide range of warm-blooded animals, including humans. The infection with T. gondii, is of particular concern due to its potential impact on human and animal health. In Sweden, semi-domesticated reindeer (Rangifer tarandus tarandus L.) is an important species both economically and culturally, but susceptibility to Toxoplasma infection and seroprevalence in reindeer herds remain relatively understudied. Results A total of 528 reindeer, sampled at two slaughterhouses in Sweden in 2014, were investigated for antibodies to T. gondii. Specific antibodies to T. gondii were found in 5 of 209 (2.3%) tested adult reindeer and in 6 of 308 (1.9%) tested calves, giving an apparent total prevalence of 2.1% (95% confidence interval 1.1–3.8%). None of four putative risk factors studied (sex, age, type of grazing area, county) were statistically associated with T. gondii seroprevalence. Conclusions Swedish semi-domesticated reindeer are exposed to T. gondii and may harbour infectious tissue cysts. To mitigate the risk of T. gondii infection in consumers, reindeer meat should be frozen or cooked thoroughly before consumption. The global climate change may influence the seroprevalence and possible associated risk factors for T. gondii in reindeer. To be able to manage the risk and get better advice to the consumers there is a need for further investigations covering the whole spectra of herding conditions for reindeer.

This study aimed to assess the seroprevalence of maternal toxoplasma and rubella among pregnant women tested during antenatal screening at the King Abdulaziz University Hospital, Saudi Arabia.Methods: This descriptive cross-sectional study targeted pregnant women attending the King Abdulaziz University Hospital, Jeddah, Saudi Arabia, during the period of 2019– 2022 for antenatal follow-up. This study included 3653 pregnant women who attended an antenatal screening. The clinical consequences of pregnancy were evaluated and categorized into six main categories based on the related complications and comorbidities of the pregnant women. The data were analyzed using the SPSS program as a descriptive statistic, and an ANOVA was used to test the associations.Results: The study findings showed that 12% of women were seropositive for toxoplasmosis and 71.4% were seropositive for rubella, and it found that almost all women (except two) were seronegative for Toxo IgM (immunoglobulin M) and rubella IgM, which indicates the absence of recent infections during the pregnancy. Approximately 19% and 4% of women who had no comorbidities experienced complicated pregnancies and delivery, respectively. Moreover, 1.3% of the pregnant women had comorbidities, complicated pregnancies, and problems during delivery. The mean Toxo IgG level was significantly higher in women with normal pregnancies than in those with other pregnancies. Rubella immunoglobulin levels did not significantly correlate with pregnancy or delivery problems.Conclusion: The seropositivity prevalence of toxoplasma Ig M and rubella Ig M were very low at the King Abdulaziz University Hospital, Saudi Arabia, indicating absence of current infections. However, the benefits of routine antenatal screening in pregnant women remain unclear. This study was conducted at one hospital, with a limited sample of participants, and did not yield adequate evidence to recommend a broader implementation of antenatal testing for toxoplasmosis and rubella.Keywords: antenatal screening, toxoplasmosis, rubella, Saudi Arabia

Introduction: Toxoplasma gondii (TG) is a common protozoan parasite infecting approximately one third of the human population. Animal studies have shown that this parasite can manipulate its host behavior. Based on this, human studies have assessed if TG can be involved in mental health disorders associated with important behavioral modifications such as schizophrenia. However, results have been discrepant. Given that TG has a strong impact on fear and risk-taking processes in animal studies and that fear and risk-taking behaviors are associated with the human stress response, we tested whether glucocorticoid biomarkers (salivary and hair) differ in people with schizophrenia and controls as a function of TG status. Methods: We measured TG antibodies in blood samples, as well as salivary and hair glucocorticoid levels in 226 people with schizophrenia (19.9% women, mean age = 39 years old) and 129 healthy individuals (controls) (45.7% women, mean age = 41 years old). Results: The results showed that people with schizophrenia infected with TG presented significantly higher hair glucocorticoid concentrations than noninfected people with schizophrenia. This effect was not found in control participants. No effect was observed for salivary glucocorticoid levels. Additionally, there were no associations between TG infection and positive psychotic symptoms nor impulsivity. Discussion: These results show that people with schizophrenia present high levels of hair glucocorticoid levels only when they are infected with TG. Further studies performed in populations suffering from other mental health disorders are needed to determine if this effect is specific to schizophrenia, or whether it is generalized across mental health disorders. Background: Toxoplasma gondii causes ocular toxoplasmosis (OT), involving inflammation, scarring, and retinal complications. The OT complications were retinal detachment (RD), and retinal breakage (RB). Surgical interventions like scleral buckling (SB) and vitrectomy are common. Limited understanding exists of the safety and efficacy of surgical management of RD/RB secondary to OT. Another complication is toxoplasmosis-related macular holes (tMH), with sparse evidence on surgical outcomes. This meta-analysis aims to clarify clinical characteristics, and surgical results, and enhance understanding of RD, RB, and MH secondary to OT. Methods: PubMed, Cochrane, Embase and Web of Science database were queried for retrospective studies, case series and case reports that provided information on RD, RB and MH associated with OT and reported the outcomes of: (1) Retinal reattachment of RD/RB and tMH closure; (2) Best-corrected visual acuity (BCVA) improvement; and (3) Complications. Heterogeneity was examined with I2 statistics. A random-effects model was used for outcomes with high heterogeneity. Statistical analysis was performed using the software R (version 4.2.3, R Foundation for Statistical Computing, Vienna, Austria). Results: Fourteen final studies, comprising a total of 96 patients were analyzed, 81 with RD or RB and 15 with tMH. Overall, surgical management was associated with several advantages: a high rate of retinal reattachment of RD/RB of 97% (95% Confidence Interval [CI] 92–100%; I2 = 0%), retinal reattachment of just RD of 96% (95% CI 89–100%; I2 = 30%) and tMH closure 97% (95% CI 87–100; I2 = 12%). There were significant differences in BCVA after surgeries in studies of RD/RB (MD 0.60; 95% CI 0.35–0.65; I2 = 20%) and MH (MD 0.67; 95% CI 0.50–0.84; I2 = 0%). The overall complication rate associated with surgical procedures in RD/RB secondary to OT was confirmed to be 25%. Conclusions: The systematic review and meta-analysis showed that the treatment approaches currently in use are effective, with a remarkable rate of retinal reattachment of RD/RB, tMH closure, and substantial improvements in visual acuity. More randomized, long-term studies on disease and surgical factors can provide valuable insights into their impact on anatomical and visual outcomes. [ABSTRACT FROM AUTHOR]
