Jupiter is one of the top priorities for deep space exploration in China and other countries. The structure of Jupiter’s interior, in particular, is a crucial but still unclear scientific topic. This paper discusses current scientific understanding of Jupiter’s interior by summarizing the history of past and current exploration and data analysis. We review recent space-based and ground-based observation methods and analyze their feasibility. To gain new insight into the internal structure of Jupiter, we propose to study Jupiter’s innards by planetary seismology. Ground-based observation, namely the Jupiter Seismologic Interferometer Polarization Imager (SIPI) in Lenghu, will be developed to obtain the Doppler velocity distribution on the surface of Jupiter and identify oscillation signals. Lenghu has observation conditions that are not only exceptional in China but even in the world, capable of providing novel insight into the interior of Jupiter. This will also be the first study in China of the interior of Jupiter using asteroseismology, which has significant implications for China’s plans to explore Jupiter via spacecraft-mounted instruments.

Abstract Background Myeloid-derived suppressor cells (MDSCs) are the major factor in gastric cancer (GC) immune evasion. Nevertheless, the molecular process underlying the expansion of MDSCs induced by tumor-derived exosomes (TDEs) remains elusive. Methods The levels of exosomal and soluble PD-L1 in ninety GC patients were examined via enzyme-linked immunosorbent assay (ELISA) to determine their prognostic value. To investigate the correlation between exosomal PD-L1 and MDSCs, the percentage of MDSCs in the peripheral blood of 57 GC patients was assessed via flow cytometry. Through ultracentrifugation, the exosomes were separated from the GC cell supernatant and detected via Western blotting, nanoparticle tracking analysis (NTA), and transmission electron microscopy (TEM). The function of exosomal PD-L1 in MDSCs was evaluated via immunofluorescence, Western blotting and flow cytometry in a GC cell-derived xenograft (CDX) model. Results The overall survival (OS) of GC patients in the high exosomal PD-L1 group was significantly lower than that of patients in the low exosomal PD-L1 group (P = 0.0042); however, there was no significant correlation between soluble PD-L1 and OS in GC patients (P = 0.0501). Background: Postural control deficits and persistent joint stability issues are prevalent in population with anterior cruciate ligament (ACL) injuries or reconstructions. Postural control is typically assessed using the center of pressure (CoP) parameters during the static single-leg stance with a force plate. However, previous studies have reported unclear definitions and descriptions of the CoP parameters, causing inconsistent results of postural control deficits in a specific population. Objective: To 1) summarize CoP parameters commonly used to evaluate postural control deficits in ACL injured or reconstructed population, and 2) identify the differences in CoP parameters with opened and closed eyes during the single-leg stance between ACL injured or reconstructed and control groups. Methods: PubMed, Embase, Cochrane Library, CINAHL, Scopus, Web of Science, and SPORTDiscus databases were searched up to July 2023. Data were obtained from the selected articles and underwent quality and risk of bias assessment and meta-analysis using random-effect models. Subgroup analysis within ACL injured or reconstructed group were also performed. Results: A total of 14 articles were included in the analysis after screening. The injured knee of the ACL injured or reconstructed group differed insignificantly in sway amplitude, sway area, and sway velocity during static single-leg stance under opened and closed eyes when compared with the control group. In the subgroup analysis, we found that there was only significant difference in sway velocity with open eyes (SMD ​= ​0.47, p ​= ​0.001) between ACL reconstructed group and control group. Conclusion: This study summarized the common CoP parameters used to evaluate postural control in ACL injured or reconstructed population. The results only showed weak difference in sway velocity between ACL reconstructed population and healthy individuals with opened eyes during the static single-leg stance.

Abstract As society continues to develop, a growing body of research is focusing on how informal institutions, such as culture, influence corporate behavior and economic outcomes. The aim of this study is to explore the impact of cultural inclusiveness, as a non-formal institution, on firm performance. Using Chinese A-share listed firms as our sample, we employ econometric methods to analyze the relationship between cultural inclusiveness and firm performance. Our findings reveal that cultural inclusiveness enhances firm performance, particularly evident in regions with higher levels of cultural inclusiveness, where firms tend to exhibit higher return on equity. To address concerns of endogeneity, instrumental variable regression using regional topographical changes is employed. Mechanism testing indicates that cultural inclusiveness primarily improves firm performance by reducing financial constraints and optimizing employee structure. Additionally, the study finds that cultural inclusiveness has a more pronounced impact on firms with higher corporate social responsibility (CSR) performance as well as those with greater investment in research and development (R&D). This study provides a comprehensive empirical explanation of how cultural inclusiveness influences the sustainable operation of businesses.

Abstract Background Bioactive lipids involved in the progression of various diseases. Nevertheless, there is still a lack of biomarkers and relative regulatory targets. The lipidomic analysis of the samples from platinum-resistant in gastric cancer patients is expected to help us further improve our understanding of it. Methods We employed LC–MS based untargeted lipidomic analysis to search for potential candidate biomarkers for platinum resistance in GC patients. Partial least squares discriminant analysis (PLS-DA) and variable importance in projection (VIP) analysis were used to identify differential lipids. The possible molecular mechanisms and targets were obtained by metabolite set enrichment analysis and potential gene network screened. Finally, verified them by immunohistochemical of a tissue microarray. Objective High serum levels of B-cell activation factor (BAFF) and a proliferation-inducing ligand (APRIL) have been observed in patients with idiopathic membranous nephropathy (iMN); however, their relationships with disease severity and progression remain unclear.Methods Patients with iMN diagnosed via renal biopsy were enrolled in this study. The concentrations of BAFF and APRIL were determined using ELISA kits. Proteinuria remission, including complete remission (CR) and partial remission (PR), and renal function deterioration were defined as clinical events. The Cox proportional hazards method was used to analyze the relationship between cytokine levels and disease progression.Results Seventy iMN patients were enrolled in this study, with a median follow-up time of 24 months (range 6–72 months). The serum levels of BAFF and APRIL were higher in iMN patients than in healthy controls but lower than those in minimal change disease (MCD) patients. The serum BAFF level was positively correlated with the serum APRIL level, serum anti-phospholipase A2 receptor (anti-PLA2R) antibody level, and 24-h proteinuria and negatively correlated with the serum albumin (ALB) level. However, no significant correlation was observed between the serum APRIL level and clinical parameters. Abstract Early identification of gastric cancer (GC) is associated with a superior survival rate compared to advanced GC. However, the poor specificity and sensitivity of traditional biomarkers suggest the importance of identifying more effective biomarkers. This study aimed to identify novel biomarkers for the prognosis of GC and construct a risk score (RS) signature based on these biomarkers, with to validation of its predictive performance. We used multi-omics data from The Cancer Genome Atlas to analyze the significance of differences in each omics data and combined the data using Fisher's method. Hub genes were subsequently subjected to univariate Cox and LASSO regression analyses and used to construct the RS signature. The RS of each patient was calculated, and the patients were divided into two subgroups according to the RS. The RS signature was validated in two independent datasets from the Gene Expression Omnibus and subsequent analyses were subsequently conducted. Five immune-related genes strongly linked to the prognosis of GC patients were obtained, namely CGB5, SLC10A2, THPO, PDGFRB, and APOD. Surgical repair or reconstruction of the lateral ligaments for patients with chronic ankle instability (CAI) could, logically, restore the proprioception of ankle through retensing receptors. To validate this hypothesis, seven databases were systematically searched, and thirteen studies comprising a total of 347 patients with CAI were included. Although five studies reported improved proprioceptive outcomes after surgeries, the other five studies with between-limb/group comparisons reported residual deficits at final follow-up, which does not consistently support proprioceptive recovery after existing surgical restabilization for CAI. More controlled studies are needed to provide evidence-based protocols to improve proprioceptive recovery after ankle restabilization for CAI.

Abstract Gastric cancer (GC) is the fifth most common malignancy and the third leading cause of cancer-related deaths worldwide. Similar to other types of tumors, GC cells undergo metabolic reprogramming and switch to a “predominantly glycolytic” metabolic pattern to promote its survival and metastasis, also known as “the Warburg effect”, which is characterized by enhanced glucose uptake and lactate production. A large number of studies have shown that targeting cancer cells to enhanced glycolysis is a promising strategy, that can make cancer cells more susceptible to other conventional treatment methods of treatment, including chemotherapy, radiotherapy and immunotherapy, and so on. Therefore, this review summarizes the metabolic characteristics of glycolysis in GC cells and focuses on how abnormal lactate concentration can lead to immunosuppression through its effects on the differentiation, metabolism, and function of infiltrating immune cells, and how targeting this phenomenon may be a potential strategy to improve the therapeutic efficacy of GC.

Microneedles (MNs), characterized by their micron-sized sharp tips, can painlessly penetrate the skin and have shown significant potential in disease treatment and biosensing. With the development of artificial intelligence (AI), the design and application of MNs have experienced substantial innovation aided by machine learning (ML). This review begins with a brief introduction to the concept of ML and its current stage of development. Subsequently, the design principles and fabrication methods of MNs are explored, demonstrating the critical role of ML in optimizing their design and preparation. Integration between ML and the applications of MNs in therapy and sensing were further discussed. Finally, we outline the challenges and prospects of machine learning-assisted MN technology, aiming to advance its practical application and development in the field of smart diagnosis and treatment.

IntroductionThis study focuses on understanding the unique causes and mechanisms of “Chinese-style lurking” on WeChat among university students, within a cultural context that emphasizes collectivism and face concern. The research also looks into the moderating role of trait mindfulness.MethodsFor the confirmation of these phenomena and to validate the theories, a structural equation model was constructed using the Stress-Strain-Outcome (SSO) theory and mindfulness buffering theory. The model was then tested and validated with data from 1,453 valid online surveys. These data were analyzed using the SmartPLS 4.0 software.ResultsThe results indicate that collectivism increases face concern, which in turn escalates online social anxiety. Face concern completely mediates between collectivism and online social anxiety, creating a serial mediation effect between face concern, online social anxiety, and lurking behavior. Additionally, trait mindfulness was found to negatively modulate the pathways from collectivism to face concern and from online social anxiety to lurking.DiscussionThe findings underscore the influence of traditional Chinese culture on contemporary students' online behavior and provide a new perspective for understanding social media lurking in an Eastern context. The results suggest that a mindfulness-based approach could be used to mitigate the associated silence and anxiety.

Summary: Programmable control over therapeutic processes in phototherapy, like photodynamic therapy (PDT), is promising but challenging. This study uses an energy segmentation-based strategy to synthesize core-multi-shell upconversion nanoparticles (UCNPs), which can release three different colors (red, green, and blue) upon exposure to different near-infrared light (1550 nm, 808 nm, and 980 nm). By combining these UCNPs with photosensitizers and nitric oxide (NO) donors, a smart “off-on” PDT nanoplatform is developed. UCNPs enable independent activation of imaging, release of NO, and generation of reactive oxygen species using specific light wavelengths. The results show that sequential NO release before PDT can greatly alleviate tumor hypoxia by reducing oxygen consumption. This stepwise approach shows potential for precise NIR light-activated and imaging-guided phototherapy.

Diabetic kidney disease (DKD) is a severe microvascular complication of diabetes and is a chronic progressive condition. It is also a common cause of end-stage renal disease (ESRD), which is characterized by proteinuria or a progressive decline in the glomerular filtration rate. Due to their dependence on high-energy and aerobic metabolism, renal tubules are more susceptible to the metabolic disturbances associated with DKD, leading to inflammation and fibrosis. Consequently, tubular injury has become a recent research focus, and significant advancements have been made in studying the role of extracellular vesicles in DKD-associated tubular injury. This review aimed to elucidate the mechanisms and potential applications of different types of extracellular vesicles in tubular injury in DKD to provide new insights for the prevention and treatment of DKD.

After molecule targeted drug, monoclonal antibody and antibody–drug conjugates (ADCs), peptide–drug conjugates (PDCs) have become the next generation targeted anti-tumor drugs due to its properties of low molecule weight, efficient cell penetration, low immunogenicity, good pharmacokinetic and large-scale synthesis by solid phase synthesis. Herein, we present a lytic peptide PTP7-drug paclitaxel conjugate assembling nanoparticles (named PPP) that can sequentially respond to dual stimuli in the tumor microenvironment, which was designed for passive tumor-targeted delivery and on-demand release of a tumor lytic peptide (PTP-7) as well as a chemotherapeutic agent of paclitaxel (PTX). To achieve this, tumor lytic peptide PTP-7 was connected with polyethylene glycol by a peptide substrate of legumain to serve as hydrophobic segments of nanoparticles to protect the peptide from enzymatic degradation. After that, PTX was connected to the amino group of the polypeptide side chain through an acid-responsive chemical bond (2-propionic-3-methylmaleic anhydride, CDM). Therefore, the nanoparticle (PPP) collapsed when it encountered the weakly acidic tumor microenvironment where PTX molecules fell off, and further triggered the cleavage of the peptide substrate by legumain that is highly expressed in tumor stroma and tumor cell surface. Moreover, PPP presents improved stability, improved drug solubility, prolonged blood circulation and significant inhibition ability on tumor growth, which gives a reasonable strategy to accurately deliver small molecule drugs and active peptides simultaneously to tumor sites.

This research is motivated by the practical requirements in the sustainable deployment of ocean moored buoy observing networks. Ocean moored buoys play an important role in the global marine environment monitoring. Ocean buoy station layout planning is a typical multiple-objective spatial optimization problem that aims to reduce the spatial correlation of buoy stations and improve their spatial monitoring efficiency. In this paper, we develop a multi-objective mathematical model for allocating ocean buoy stations (MOLMofOBS) based on Tobler’s first law of geography. A spatial neighborhood model based on a Voronoi diagram is built to represent the spatial proximity of distributed buoy stations and delimit the effective monitoring region of every station. Then, a heuristic method based on a multiple-objective particles swarm optimization (MOPSO) algorithm is developed to calculate the MOLMofOBS via a dynamic inertia weight strategy. Meanwhile, a series of experiments is conducted to verify the efficiency of the proposed model and algorithms in solving single- and multiple-buoy station location problems. Finally, an interactive portal is developed in the Cyberinfrastructure environment to provide decision-making services for online real-time planning of the ocean buoy station locations. The work reported in this paper will provide spatial decision-making support for the sustainable development of ocean buoy observing networks.

Diabetic kidney disease (DKD), a common cause of end-stage renal disease, is a serious complication that develops with the progression of chronic diabetes. Its main clinical manifestations are persistent proteinuria and/or a progressive decline in the estimated glomerular filtration rate. Podocytes, terminally differentiated glomerular visceral epithelial cells, constitute the glomerular filtration barrier together with the basement membrane and endothelial cells, and the structural and functional barrier integrity is closely related to proteinuria. In recent years, an increasing number of studies have confirmed that podocyte injury is the central target of the occurrence and development of DKD, and research on exosomes in podocyte injury associated with DKD has also made great progress. The aim of this review is to comprehensively describe the potential diagnostic value of exosomes in podocyte injury associated with DKD, analyze the mechanism by which exosomes realize the communication between podocytes and other types of cells and discuss the possibility of exosomes as targeted therapy drug carriers to provide new targets for and insights into delaying the progression of and treating DKD.

BackgroundUsing the Health Belief Model (HBM), this study analyzed tweets related to COVID-19 published by national health departments of the United States, the South Korea, the United Kingdom, Japan, Germany, and India to explore their differences in (1) the health measures against COVID-19, (2) the health promotion strategies, (3) the social media engagements that those measures and strategies have triggered.MethodWe conducted a content analysis with 1,200 randomly selected COVID-19-related tweets from six national health departments' Twitter accounts from 1 January 2020 to 31 December 2020. We coded the six HBM constructs and 21 sub-themes of the HBM constructs for each tweet.ResultsResults showed that all six HBM constructs were used in the full sample. The most commonly used HBM construct was cues to action, followed by susceptibility, benefits, self-efficacy, severity, and barriers. All the HBM constructs were positively related to Twitter engagement variables except barriers. Further analysis illustrated that people from the six countries responded differently to the HBM constructs and the HBM sub-themes. Twitter users in Germany, India, the U.S., and Japan positively reacted to the clear directions of “what to do against COVID-19” (cues to action), while Twitter users in the U.S. and Japan were also eager to know the justifications for such directions (benefits); people in South Korea and the U.K. were mainly seeking a diagnosis of the severity and susceptibility of COVID-19, instead of health measures, of COVID-19 in the year 2020.ConclusionsThis study showed the use of HBM constructs is generally effective in inducing Twitter engagement. The further comparison illustrated a homogenization in the promotion strategies that the health departments implemented and the health measures they promoted, yet responses to such promotions varied across nations. This study broadened the scope of HBM applications from predicting health behaviors in surveys to guiding the design of health promotion messages online.

Monoclonal immunoglobulin produced by clonal plasma cells is the main cause in multiple myeloma and monoclonal gammopathy of renal significance. Because of the complicated purification method and the low stoichiometry of purified protein and glycans, site-specific N-glycosylation characterization for monoclonal immunoglobulin is still challenging. To profile the site-specific N-glycosylation of monoclonal immunoglobulins is of great interest. Therefore, in this study, we presented an integrated workflow for micro monoclonal IgA and IgG purification from patients with multiple myeloma in the HYDRASYS system, in-agarose-gel digestion, LC-MS/MS analysis without intact N-glycopeptide enrichment, and compared the identification performance of different mass spectrometry dissociation methods (EThcD-sceHCD, sceHCD, EThcD and sceHCD-pd-ETD). The results showed that EThcD-sceHCD was a better choice for site-specific N-glycosylation characterization of micro in-agarose-gel immunoglobulins (~2 μg) because it can cover more unique intact N-glycopeptides (37 and 50 intact N-glycopeptides from IgA1 and IgG2, respectively) and provide more high-quality spectra than sceHCD, EThcD and sceHCD-pd-ETD. We demonstrated the benefits of the alternative strategy in site-specific N-glycosylation characterizing micro monoclonal immunoglobulins obtained from bands separated by electrophoresis. This work could promote the development of clinical N-glycoproteomics and related immunology.

Optical cryptanalysis is essential to the further investigation of more secure optical cryptosystems. Learning-based attack of optical encryption eliminates the need for the retrieval of random phase keys of optical encryption systems but it is limited for practical applications since it requires a large set of plaintext-ciphertext pairs for the cryptosystem to be attacked. Here, we propose a two-step deep learning strategy for ciphertext-only attack (COA) on the classical double random phase encryption (DRPE). Specifically, we construct a virtual DRPE system to gather the training data. Besides, we divide the inverse problem in COA into two more specific inverse problems and employ two deep neural networks (DNNs) to respectively learn the removal of speckle noise in the autocorrelation domain and the de-correlation operation to retrieve the plaintext image. With these two trained DNNs at hand, we show that the plaintext can be predicted in real-time from an unknown ciphertext alone. The proposed learning-based COA method dispenses with not only the retrieval of random phase keys but also the invasive data acquisition of plaintext-ciphertext pairs in the DPRE system. Numerical simulations and optical experiments demonstrate the feasibility and effectiveness of the proposed learning-based COA method.

Abstract Background Chrysomycin A (CA) has been reported as numerous excellent biological activities, such as antineoplastic and antibacterial. Though, poor solubility of CA limited its application in medical field. Due to good amphiphilicity and potential anticancer effect of disodium glycyrrhizin (Na2GA) as an excipient, an amorphous solid dispersion (Na2GA/CA-BM) consisting of CA and Na2GA was prepared in the present study by mechanochemical technology (roll mill ML-007, zirconium balls, 30 rpm, 2.5 h) to improve the solubility and oral bioavailability of CA. Then, Na2GA/CA-BM was self-assembled to micelles in water. The interaction of CA and Na2GA in solid state were investigated by X-ray diffraction studies, polarized light microscopy, and scanning electron microscope. Meanwhile, the properties of the sample solution were analyzed by dynamic light scattering and transmission electron. Furthermore, the oral bioavailability and antitumor ability of Na2GA/CA-BM in vivo were tested, providing a theoretical basis for future application of CA on cancer therapy. Results CA encapsulated by Na2GA was self-assembled to nano-micelles in water. The average diameter of nano-micelle was 131.6 nm, and zeta potential was − 11.7 mV. Three physicochemical detections showed that CA was transformed from crystal into amorphous form after treated with ball milling and the solubility increased by 50 times. Na2GA/CA-BM showed a significant increase of the bioavailability about two time that of free CA. Compared with free CA, the in-vivo antitumor studies also exhibited that Na2GA/CA-BM had an excellent inhibition of tumor growth. Conclusions Na2GA/CA-BM nanoparticles (131.6 nm, − 11.7 mV) prepared by simple and low-cost mechanochemical technology can improve oral bioavailability and antitumor efficacy of CA in vivo, suggesting a potential formulation for efficient anticancer treatment. Site-specific N-glycosylation characterization requires intact N-glycopeptide analysis based on suitable tandem mass spectrometry (MS/MS) method. Electron-transfer/higher-energy collisional dissociation (EThcD), stepped collision energy/higher-energy collisional dissociation (sceHCD), higher-energy collisional dissociation-product-dependent electron-transfer dissociation (HCD-pd-ETD), and a hybrid mass spectrometry fragmentation method EThcD-sceHCD have emerged as valuable approaches for glycoprotein analysis. However, each of them incurs some compromise, necessitating the systematic performance comparisons when applied to the analysis of complex clinical samples (e.g., plasma, urine, cells, and tissues). Herein, we compared the performance of EThcD-sceHCD with those previous approaches (EThcD, sceHCD, HCD-pd-ETD, and sceHCD-pd-ETD) in the intact N-glycopeptide analysis, and determined its applicability for clinical N-glycoproteomic study. The intact N-glycopeptides of distinct samples, namely, plasma from prostate cancer (PCa) patients, urine from immunoglobulin A nephropathy (IgAN) patients, human hepatocarcinoma cell line (HepG2), and thyroid tissues from thyroid cancer (TC) patients were analyzed by these methods. We found that EThcD-sceHCD outperformed other methods in the balance of depth and accuracy of intact N-glycopeptide identification, and sceHCD and EThcD-sceHCD have good complementarity. EThcD-sceHCD holds great potential for biomarker discovery from clinical samples.

BackgroundPost-infarction left ventricular (LV) pseudoaneurysm is a rare mechanical complication of myocardial infarction that carries a substantial risk of sudden rupture. The purpose of this study was to compare the surgical results of post-infarction LV pseudoaneurysm with those of conservative treatment.MethodsFrom 2016 to 2021, 22 patients were hospitalized for LV pseudoaneurysm, including 17 cases (77.3%) caused by myocardial infarction. Of the 17 patients, 10 (58.8%) underwent surgical repair, while seven (41.2%) were treated medically. The clinical course, echocardiograph data, and surgical outcomes were analyzed. Survival rates of the surgical and conservative groups were compared.ResultsThere were no perioperative deaths. Intra-aortic balloon pumping support was required in two (20%) patients. No follow-up mortality was observed in the surgical group and at the last follow-up, all the patients were classified as New York Heart Association class I–II. In the conservative group, there was one (14.3%) hospital death and two (28.6%) additional deaths during follow-up. A significant difference was found in survival between the two groups (P = 0.024).ConclusionsSurgical repair of post-infarction LV pseudoaneurysm can be performed with good results, while conservative treatment carries a significant risk of sudden death. Surgical repair is indicated for every patient diagnosed, even those with a small pseudoaneurysm without symptoms.

Chemoresistance is the major restriction on the clinical use of cisplatin. Aberrant changes in protein glycosylation are closely associated with drug resistance. Comprehensive study on the role of protein glycosylation in the development of cisplatin resistance would contribute to precise elucidation of the complicated mechanism of resistance. However, comprehensive characterization of glycosylated proteins remains a big challenge. In this work, we integrated proteomic and N-glycoproteomic workflow to comprehensively characterize the cisplatin resistance-related membrane proteins. Using this method, we found that proteins implicated in cell adhesion, migration, response to drug, and signal transduction were significantly altered in both protein abundance and glycosylation level during the development of cisplatin resistance in the non-small cell lung cancer cell line. Accordingly, the ability of cell migration and invasion was markedly increased in cisplatin-resistant cells, hence intensifying their malignancy. In contrast, the intracellular cisplatin accumulation was significantly reduced in the resistant cells concomitant with the down-regulation of drug uptake channel protein, LRRC8A, and over-expression of drug efflux pump proteins, MRP1 and MRP4. Moreover, the global glycosylation was elevated in the cisplatin-resistant cells. Consequently, inhibition of N-glycosylation reduced cell resistance to cisplatin, whereas promoting the high-mannose or sialylated type of glycosylation enhanced the resistance, suggesting that critical glycosylation type contributes to cisplatin resistance. These results demonstrate the high efficiency of the integrated proteomic and N-glycoproteomic workflow in discovering drug resistance-related targets, and provide new insights into the mechanism of cisplatin resistance.

Deciphering the glycosylation of the viral envelope (Env) glycoprotein is critical for evaluating viral escape from the host’s immune response and developing vaccines and antiviral drugs. However, it is still challenging to precisely decode the site-specific glycosylation characteristics of the highly glycosylated Env proteins, although glycoproteomics have made significant advances in mass spectrometry techniques and data analysis tools. Here, we present a hybrid dissociation technique, EThcD-sceHCD, by combining electron transfer/higher-energy collisional dissociation (EThcD) and stepped collision energy/higher-energy collisional dissociation (sceHCD) into a sequential glycoproteomic workflow. Following this scheme, we characterized site-specific N/O-glycosylation of the human immunodeficiency virus type 1 (HIV-1) Env protein gp120. The EThcD-sceHCD method increased the number of identified glycopeptides when compared with EThcD, while producing more comprehensive fragment ions than sceHCD for site-specific glycosylation analysis, especially for accurate O-glycosite assignment. Finally, eighteen N-glycosites and five O-glycosites with attached glycans were assigned unambiguously from heavily glycosylated gp120. These results indicate that our workflow can achieve improved performance for analysis of the N/O-glycosylation of a highly glycosylated protein containing numerous potential glycosites in one process. Knowledge of the glycosylation landscape of the Env glycoprotein will be useful for understanding of HIV-1 infection and development of vaccines and drugs.

The densely glycosylated spike (S) proteins that are highly exposed on the surface of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) facilitate viral attachment, entry, and membrane fusion. We have previously reported all the 22 N-glycosites and site-specific N-glycans in the S protein protomer. Herein, we report the O-glycosylation landscapes of SARS-CoV-2 S proteins, which were characterized through high-resolution mass spectrometry. Following digestion with trypsin and trypsin/Glu-C, and de-N-glycosylation using PNGase F, we determined the GalNAc-type O-glycosylation pattern of S proteins, including O-glycosites and the six most common O-glycans occupying them, via Byonic identification and manual validation. Finally, 255 intact O-glycopeptides composed of 50 peptides sequences and 43 O-glycosites were discovered by higher energy collision-induced dissociation (HCD), and three O-glycosites were confidently identified by electron transfer/higher energy collision-induced dissociation (EThcD) in the insect cell-expressed S protein. Most glycosites were modified by non-sialylated O-glycans such as HexNAc(1) and HexNAc(1)Hex (1). In contrast, in the human cell-expressed S protein S1 subunit, 407 intact O-glycopeptides composed of 34 peptides sequences and 30 O-glycosites were discovered by HCD, and 11 O-glycosites were unambiguously assigned by EThcD. However, the measurement of O-glycosylation occupancy hasn’t been made. Most glycosites were modified by sialylated O-glycans such as HexNAc(1)Hex (1)NeuAc (1) and HexNAc(1)Hex (1)NeuAc (2). Our results reveal that the SARS-CoV-2 S protein is an O-glycoprotein; the O-glycosites and O-glycan compositions vary with the host cell type. These comprehensive O-glycosylation landscapes of the S protein are expected to provide novel insights into the viral binding mechanism and present a strategy for the development of vaccines and targeted drugs.

This real-world, observational study aimed to assess and compare the clinical efficacy and safety of eltrombopag with recombinant human thrombopoietin (rhTPO) in the treatment of chemotherapy induced thrombocytopenia (CIT) in patients with lymphoma. One hundred and fifty-three patients who experienced grade 3 or 4 thrombocytopenia after chemotherapy for lymphoma were enrolled, 51 of which were treated with eltrombopag, 50 with rhTPO, and 52 patients with no drug treatment were served as the control group. The lowest platelet level and mean platelet counts at Day 5, Day 7, and Day 10 were significantly higher in both the eltrombopag group (P=.041,.003,.000,.000) and rhTPO group (P=.005,.005,.000,.000) than the control, but there was no difference between treatment with eltrombopag and rhTPO. Background: In patients with metabolic-associated fatty liver disease (MAFLD), hepatic steatosis is the first step of diagnosis, and it is a risk predictor that independently predicts insulin resistance, cardiovascular risk, and mortality. Urine biomarkers have the advantage of being less complex, with a lower dynamic range and fewer technical challenges, in comparison to blood biomarkers. Methods: Hepatic steatosis was measured by magnetic resonance imaging (MRI), which measured the proton density fat fraction (MRI-PDFF). Mild hepatic steatosis was defined as MRI-PDFF 5–10% and severe hepatic steatosis was defined as MRI-PDFF > 10%. Results: MAFLD patients with any kidney diseases were excluded. There were 53 proteins identified by mass spectrometry with significantly different expressions among the healthy control, mild steatosis, and severe steatosis patients. Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses of these significantly changed urinary molecular features correlated with the liver, resulting in the dysregulation of carbohydrate derivative/catabolic/glycosaminoglycan/metabolic processes, insulin-like growth factor receptor levels, inflammatory responses, the PI3K–Akt signaling pathway, and cholesterol metabolism. Urine alpha-1-acid glycoprotein 1 (ORM1) and ceruloplasmin showed the most significant correlation with the clinical parameters of MAFLD status, including liver fat content, fibrosis, ALT, triglycerides, glucose, HOMA-IR, and C-reactive protein. According to ELISA and western blot (30 urine samples, normalized to urine creatinine), ceruloplasmin (ROC 0.78, p = 0.034) and ORM1 (ROC 0.87, p = 0.005) showed moderate diagnostic accuracy in distinguishing mild steatosis from healthy controls. Ceruloplasmin (ROC 0.79, p = 0.028) and ORM1 (ROC 0.81, p = 0.019) also showed moderate diagnostic accuracy in distinguishing severe steatosis from mild steatosis. Conclusions: Ceruloplasmin and ORM1 are potential biomarkers in distinguishing mild and severe steatosis in MAFLD patients.

Pulmonary artery sarcoma (PAS) is a rare and devastating disease. The diagnosis is often delayed, and optimal treatment remains unclear. The aim of this study is to report our experience in the surgical management of this disease. Between 2000 and 2018, 17 patients underwent operations for PAS at our center. The medical records were retrospectively reviewed to evaluate the clinical characteristics, operative findings, the postoperative outcomes, and the long-term results. The mean age at operation was 46.0 ± 12.4 years (range, 26–79 years), and eight (47.1%) patients were male. Six patients underwent tumor resection alone, whereas the other 11 patients received pulmonary endarterectomy (PEA). There were two perioperative deaths. Follow-up was completed for all patients with a mean duration of 23.5 ± 17.6 months (1–52 months). For all 17 patients, the median postoperative survival was 36 months, and estimated cumulative survival rates at 1, 2, 3, and 4 years were 60.0%, 51.4%, 42.9%, and 21.4%, respectively. The mean survival was 37.0 months after PEA and 14.6 months after tumor resection only ( p = 0.046). Patients who had no pulmonary hypertension (PH) postoperatively were associated with improved median survival (48 vs. 5 months, p = 0.023). In conclusion, PAS is often mistaken for chronic pulmonary thromboembolism. The prognosis of this very infrequent disease remains poor. Early detection is essential for prompt and best surgical approach, superior to tumor resection alone, and PEA surgery with PH relieved can provide better chance of survival.

Abstract Thrombospondin type I domain‐containing 7A (THSD7A), is a specific autoantigen of adult idiopathic membranous nephropathy (IMN), whose circulating antibody (THSD7A‐AB) represents a promising biomarker for diagnosis of IMN. The objective of this meta‐analysis is to investigate the diagnostic efficiency of THSD7A‐AB for IMN. After rigorous data extraction, quality assessment, and data analysis, 10 articles (4545 patients) are included. For IMN, the summary sensitivity is 4% (2–7%), and the specificity is 99% (98–100%). The summary positive likelihood ratio (PLR) and negative likelihood ratio (NLR) are 5.40 (2.40–11.90) and 0.97 (0.95–0.99), respectively. The diagnostic odds ratio (DOR) is 6.00 (2.00–12.00). The area under the summary receiver operating characteristic curve (AUC) is 0.78 (0.74–0.81). For M‐type phospholipase A2 receptor (PLA2R)‐negative IMN, the summary sensitivity is 8% (6–10%), specificity is 100% (99–100%). The summary PLR and NLR are 15.80 (5.70–44.00) and 0.93 (0.91–0.95), respectively. The DOR is 17.00 (6.00–48.00). The AUC is 0.99 (0.98–1.00). THSD7A‐AB has higher diagnostic value in PLA2R‐negative patients than in IMN patients. These results suggest that THSD7A‐AB could possibly be applied as an auxiliary non‐invasive diagnostic method for PLA2R‐negative IMN.

For more than 50% of multiparous cows, it is difficult to adapt to the sudden increase in calcium demand for milk production, which is highly likely to cause hypocalcemia. An electrochemical biosensor is a portable and efficient method to sense Ca2+ concentrations, but biomaterial is easily affected by the pH of the analyte solution. Here, an electrochemical biosensor was fabricated using a glassy carbon electrode (GCE) and single-walled carbon nanotube (SWNT), which amplified the impedance signal by changing the structure and length of the DNAzyme. Aiming at the interference of the pH, the electrochemical biosensor (GCE/SWNT/DNAzyme) was coupled with a pH meter to form an electrochemical device. It was used to collect data at different Ca2+ concentrations and pH values, and then was processed using different mathematical models, of which GPR showed higher detecting accuracy. After optimizing the detecting parameters, the electrochemical device could determine the Ca2+ concentration ranging from 5 μM to 25 mM, with a detection limit of 4.2 μM at pH values ranging from 4.0 to 7.5. Finally, the electrochemical device was used to determine the Ca2+ concentrations in different blood and milk samples, which can overcome the influence of the pH.

From April to September 2018, five sampling sites were selected in Lianyungang City for volatile organic compounds (VOCs) analysis, including two sampling sites in the urban area (Lianyungang City Environmental Monitoring Supersite and Mine Design Institute), one sampling site in the industrial area (Deyuan Pharmaceutical Factory), and two sampling sites from the suburb (Hugou Management Office and YuehaiLou). The results showed that the mean VOCs concentration followed this pattern: industrial area (36.06 ± 12.2 µg m−3) > urban area (33.47 ± 13.0 µg m−3) > suburban area (27.68 ± 9.8 µg m−3). The seasonal variation of the VOCs trend in the urban and suburban areas was relatively consistent, which was different from that in industrial areas. The concentration levels of VOCs components in urban and industrial areas were relatively close, which were significantly higher than that in suburban areas. The possible sources and relative importance of VOCs in Lianyungang City atmosphere were measured by the characteristic ratio of toluene/benzene (T/B), ethane/acetylene (E/E) and isopentane/TVOCs. The contribution of traffic sources to the VOCs in Lianyungang City was significant (T/B ~ 2), and there were obvious aging phenomena in the five sampling sites (E/E > 4). The ratio of isopentane/TVOCs in the contribution of gasoline volatilization sources in urban and suburban areas was significantly bigger than that in industrial areas. According to the maximum incremental reactivity (MIR) method, aromatics (40.32–58.09%) contributed the most to ozone formation potential (OFP) at the five sampling sites. The top 10 OFP species showed that controlling n-hexane and aromatics, such as benzene, toluene, xylene, and trimethylbenzene in Lianyungang City can effectively control ozone generation. Nineteen typical VOCs components were selected and the sources of VOCs from five sampling points were analyzed by the principal component analysis (PCA) model. The sources of VOCs in different areas in Lianyungang were relatively consistent. Five sources were analyzed at the two sampling sites in the urban area: industrial emission + plants, vehicle exhaust, fuel evaporation, combustion and industrial raw materials. Four sources were analyzed in the industrial area: industrial emission + plants, vehicle exhaust, fuel evaporation and combustion. To evaluate the effectiveness of measures to reduce the levels of volatile organic compounds (VOCs), which are important precursors of ground-level ozone formation, the real-time monitoring data of VOCs at the urban Zhaohui supersite (ZH), the Dianshan Lake regional supersite (DSL) and the urban Yixing station (YX) in the Yangtze River Delta region were analyzed from 23 August to 15 September 2016 during the G20 Hangzhou Summit. The average mole ratios of VOCs at the three sites were 6.56, 21.33 and 19.62 ppb, respectively, which were lower than those (13.65, 27.72 and 21.38 ppb) after deregulation. The characteristics of the VOCs varied during the different control periods. Synoptic conditions and airmass transport played an important role in the transport and accumulation of VOCs and other pollutants, which affected the control effects. Using the positive matrix factorization (PMF) method in source apportionment, five factors were identified, namely, vehicle exhaust (19.66–31.47%), plants (5.59–17.07%), industrial emissions (13.14–33.82%), fuel vaporization (12.83–26.34%) and solvent usage (17.84–28.95%) for the ZH and YX sites. Factor 4 was identified as fuel vaporization + incomplete combustion (21.69–25.35%) at the DSL site. The Non-parametric Wind Regression (NWR) method showed that regional transport was the main factor influencing the VOC distribution.

To identify relationships between animal, dietary and management factors and the resulting methane (CH4) emissions, and to identify potential mitigation strategies for CH4 production, it is vital to develop reliable and accurate CH4 measurement techniques. This review outlines various methods for measuring enteric CH4 emissions from ruminants such as respiration chambers (RC), sulphur hexafluoride (SF6) tracer, GreenFeed, sniffer method, ventilated hood, facemask, laser CH4 detector and portable accumulation chamber. The advantages and disadvantages of these techniques are discussed. In general, RC, SF6 and ventilated hood are capable of 24 h continuous measurements for each individual animal, providing accurate reference methods used for research and inventory purposes. However, they require high labor input, animal training and are time consuming. In contrast, short-term measurement techniques (i.e., GreenFeed, sniffer method, facemask, laser CH4 detector and portable accumulation chamber) contain additional variations in timing and frequency of measurements obtained relative to the 24 h feeding cycle. However, they are suitable for large-scale measurements under commercial conditions due to their simplicity and high throughput. Successful use of these techniques relies on optimal matching between the objectives of the studies and the mechanism of each method with consideration of animal behavior and welfare. This review can provide useful information in selecting suitable techniques for CH4 emission measurement in ruminants.

The rumen microbial complex adaptive mechanism invalidates various methane (CH4) mitigation strategies. Shifting the hydrogen flow toward alternative electron acceptors, such as propionate, was considered to be a meaningful mitigation strategy. A completely randomized design was applied in in vitro incubation to investigate the effects of replacing forage fiber with non-forage fiber sources (NFFS) in diets on methanogenesis, hydrogen metabolism, propionate production and the methanogenic and bacterial community. There are two treatments in the current study, CON (a basic total mixed ration) and TRT (a modified total mixed ration). The dietary treatments were achieved by partly replacing forage fiber with NFFS (wheat bran and soybean hull) to decrease forage neutral detergent fiber (fNDF) content from 24.0 to 15.8%, with the composition and inclusion rate of other dietary ingredients remaining the same in total mixed rations. The concentrations of CH4, hydrogen (H2) and volatile fatty acids were determined using a gas chromatograph. The archaeal and bacterial 16S rRNA genes were sequenced by Miseq high-throughput sequencing and used to reveal the relative abundance of methanogenic and bacterial communities. The results revealed that the concentration of propionate was significantly increased, while the concentration of acetate and the acetate to propionate ratio were not affected by treatments. Compared with CON, the production of H2 increased by 8.45% and the production of CH4 decreased by 14.06%. The relative abundance of Methanomassiliicoccus was significantly increased, but the relative abundance of Methanobrevibacter tended to decrease in TRT group. At the bacterial phylum level, the TRT group significantly decreased the relative abundance of Firmicutes and tended to increase the relative abundance of Bacteroidetes. The replacement of forage fiber with NFFS in diets can affect methanogenesis by shifting the hydrogen flow toward propionate, and part is directed to H2in vitro. The shift was achieved by a substitution of Firmicutes by Bacteroidetes, another substitution of Methanobrevibacter by Methanomassiliicoccus. Theoretical predictions of displacements of H2 metabolism from methanogenesis to propionate production was supported by the dietary intervention in vitro.

With the emergence of the neologism ‘intangible cultural heritage’ in 2003 and the adoption of the Intangible Cultural Heritage Law of the People’sRepublic of China in 2011 various popular religious practices in China which used to be considered as feudal superstitions started to be recognized as examples of cultural heritage worthy of protection. If we examine the concept of religious diversity at a local level in contemporary China, the process of a ‘patrimonialization’ of popular religious practices that reflect the dynamic relationships which can be detected across diverse discourses, multiple stakeholders and cultural policies in different arenas could offer us a new perspective on religious practices to explore. In this article I offer an analysis, based on fieldwork conducted between 2013 and 2016 on the Leizhou Peninsula in southern China’s Guangdong Province, of the varying degrees of acceptance, accommodation and resistance prompted by the actualization of popular religious practices in this era of patrimonialization.

The method for estimating and controlling the coastal navigation mark measurement and control cost of the coastal navigation mark measurement and control project is to maintain the safety of the coastal navigation mark measurement and control project, to reduce the cost of the project and to guarantee the service level. The traditional cost estimation method of the coastal navigation mark measurement and control project is based on the experience-based evaluation of the cost prediction model of the coastal navigation mark measurement and control engineering, which cannot effectively meet the principle of minimizing the present value of the measurement and control cost of the coastal navigation mark and the maximization of the state index during the service life period. In this paper, a method for estimating the cost of a coastal navigation mark measurement and control project based on the characteristics of large data correlation is proposed, and the measurement and control model of the conventional coastal navigation mark is designed. Cost estimation model of coastal navigation mark measurement and control project based on performance and energy supply survival period assessment model based on comprehensive coastal navigation mark measurement and control strategy. The cost estimation model of the coastal navigation mark measurement and control engineering is designed by using the first-order Bessel function, a reliable index evaluation equation of the cost all-sample regression analysis is obtained, the measurement and control cost of the coastal navigation mark, the user cost and the social cost are calculated. The cost present value of the measurement and control project of the coastal navigation mark is minimized as the objective function, and the financial constraint and the performance constraint are obtained as the boundary conditions. The plan optimization of the cost of the measurement and control engineering of the coastal navigation mark is realized, and the measurement and control scheme of the coastal navigation mark with the minimum cost present value is obtained under the limited financial resources. The numerical simulation results show that the model can improve the reliability and reliability of the measurement and control engineering structure of the coastal navigation mark, optimize the measurement and control cost of the coastal navigation mark, and obtain the minimum measurement and control scheme of the coastal navigation mark with the minimum cost present value under the limited financial resources, thus saving the measurement and control cost of the coastal navigation mark by 15%. Anomaly detection is important for the correct functioning of wireless sensor networks. Recent studies have shown that node mobility along with spatial correlation of the monitored phenomenon in sensor networks can lead to observation data that have long range dependency, which could significantly increase the difficulty of anomaly detection. In this article, we develop an anomaly detection scheme based on multiscale analysis of the long-range dependent traffic to address this challenge. In this proposed detection scheme, the discrete wavelet transform is used to approximately de-correlate the traffic data and capture data characteristics in different timescales. The remaining dependencies are then captured by a multilevel hidden Markov model in the wavelet domain. To estimate the model parameters, we develop an online discounting expectation maximization (EM) algorithm, which also tracks variations of the estimated models over time. Network anomalies are detected as abrupt changes in the tracked model variation scores. Statistical properties of our detection scheme are evaluated numerically using long-range dependent time series. We also evaluate our detection scheme in malicious scenarios simulated using the NS-2 network simulator. [ABSTRACT FROM AUTHOR]
