Abstract While several pieces of evidence link obesity and mood disorders in menopause, the mechanisms involved are not yet fully understood. We have previously demonstrated that Ginkgo biloba extract (GbE) both attenuated diet-induced obesity of male rats and restored serotonin-induced hypophagia in ovariectomized female rats. The present study aimed at exploring whether GbE treatment ameliorates ovariectomy-related obesity and anxious/depressive-like behaviours. Wistar female rats were either ovariectomized (OVX) or sham-operated (Sham). After 2 months, either 500 mg/kg of GbE or vehicle were administered daily by gavage for 14 days. Anxious/depressive-like behaviours were assessed by the Elevated Plus Maze and the Forced Swim Tests, respectively. Ovariectomy caused high visceral adiposity, hyperleptinemia, and hypercholesterolemia, and increased the anxiety index (p = 0.048 vs. Sham + GbE) while it decreased the latency to immobility (p = 0.004 vs. Sham). GbE treatment in OVX rats improved body composition, adiponectin levels and blood lipid profile. It also reduced the anxiety index (p = 0.004) and increased the latency to immobility (p = 0.003) of OVX rats. Linear regression analysis demonstrated that leptin (p = 0.047) and total cholesterol levels (p = 0.022) were associated with anxious-like behaviours while body adiposity (p = 0.00005) was strongly associated with depressive-like behaviours. The results showed that GbE therapy was effective in attenuating the deleterious effects of ovariectomy on body composition, lipid profile, and anxious/depressive-like behaviours. Further studies are warranted to better understand the therapeutic potential of GbE in menopause.

Smaller adipocytes are related to the reversal of metabolic disorders, suggesting that molecules that can act in the adipogenesis pathway are of great interest. The objective of this study was to investigate the effect of Ginkgo biloba extract (GbE) in modulating the differentiation in preadipocytes. 3T3-L1 preadipocytes were differentiated for 7 days into adipocytes without (control group) and with GbE at 1.0 mg/mL. Lipid content and gene expression were analyzed on day 7 (D7) by Oil Red O staining and PCR Array Gene Expression. Western blotting analysis of the key adipogenesis markers was evaluated during the differentiation process at days 3 (D3), 5 (D5), and 7 (D7). GbE increased lipid content and raised the gene expression of the main adipogenesis markers. Key proteins of the differentiation process were modulated by GbE, since C/EBPβ levels were decreased, while C/EBPα levels were increased at D7. Regarding the mature adipocytes’ markers, GbE enhanced the levels of both FABP4 at D5, and perilipin at D3 and D5. In summary, the present findings showed that GbE modulated the adipogenesis pathway suggesting that the treatment could accelerate the preadipocyte maturation, stimulating the expression of mature adipocyte proteins earlier than expected.

In order to evidence the perception of nurses about interprofessional collaborative practices in Primary Care in the city of Palmas, TO, this qualitative study was conducted in 34 Basic Health Units where collaborative practices are experienced. Data were analyzed using the Collective Subject Discourse technique, based on the Social Representation Theory. The collaborative practices reported were: team meetings, work groups, shared care, joint activities in the community, case discussions, integrated therapeutic plans and conversation circles. They mentioned interprofessional communication as facilitators; team profile; partnership, trust and respect; flexible schedule for meetings and collaborative leadership. In the participants' perception, collaborative practices favor greater effectiveness in solving complex problems and intensify professional bonds between team members. They emphasized that collaborative practices are of great relevance in the scope of Primary Health Care, improving the ambience, job satisfaction and the quality of the service. - Abstract
Background Poly (ADP-ribose) polymerase 1 (PARP-1) plays pivotal roles in immune and inflammatory responses. Accumulating evidence suggests PARP-1 as a promising target for immunomodulation in multiple sclerosis and natalizumab-associated progressive multifocal leukoencephalopathy. Objective This study explores expression of PARP-1 and downstream effectors in multiple sclerosis and during natalizumab treatment. Methods Transcriptional expressions were studied by real-time reverse transcriptase polymerase chain reaction on CD4
+ T/CD8+ T/CD14+ /B cells and peripheral blood mononuclear cells from healthy volunteers, untreated and natalizumab-treated non-progressive multifocal leukoencephalopathy and progressive multifocal leukoencephalopathy multiple sclerosis patients. Results PARP-1 expression was higher in CD4+ T, CD8+ T and B cells from untreated patients compared to healthy volunteers. Natalizumab treatment restored deregulated PARP-1 expression in T cells but not in B cells. Sustained upregulation of PARP-1 was associated with decreased expression of downstream PARP-1 factors such as TGFBR1/TGFBR2/BCL6 in B cells. Notably, a higher expression of PARP-1 was detected in progressive multifocal leukoencephalopathy patients. Conclusions Given the importance of PARP-1 in inflammatory processes, its upregulation in multiple sclerosis lymphocyte populations suggests a potential role in the immune pathogenesis of multiple sclerosis. Strikingly higher PARP-1 expression in progressive multifocal leukoencephalopathy cases suggests its involvement in progressive multifocal leukoencephalopathy disease pathomechanisms. These results further support the value of PARP-1 inhibitors as a potential novel therapeutic strategy for multiple sclerosis and natalizumab-associated progressive multifocal leukoencephalopathy. MicroRNAs (miRNAs) are a family of noncoding RNAs that play critical roles in the posttranscriptional regulation of gene expression. Accumulating evidence supports their involvement in the pathogenesis of multiple sclerosis (MS). Here, we compare miR-17 expressions in CD4+ T cells from relapsing-remitting (RR) MS patients treated with natalizumab versus untreated patients. miR-17 was downregulated under natalizumab treatment and upregulated during relapse, therefore supporting a possible role of miR-17 in MS immunopathogenesis. Downregulation of miR-17 was associated with upregulation of PTEN, BIM, E2F1, and p21 target genes. In vitro miR-17 inhibition was associated with upregulation of the same targets and resulted in impaired CD4+ T cell activation and proliferation. We further describe deregulated TGFBR2 expression in untreated patients versus healthy volunteers (HVs) and confirm in vitro the link between miR-17 and TGFBR2 expressions. These findings support an effect of natalizumab on expression of specific miRNA and subsequent expression of genes involved in proliferation and control of the cell cycle. [ABSTRACT FROM AUTHOR]

30. MHO1, an Evolutionarily Conserved Gene, Is Synthetic Lethal with PLC1; Mho1p Has a Role in Invasive Growth.

The novel protein Memo (Mediator of ErbB2 driven cell motility) was identified in a screen for ErbB2 interacting proteins and found to have an essential function in cell motility. Memo is evolutionarily conserved with homologs found in all branches of life; the human and yeast proteins have a similarity of >50%. In the present study we used the model organism S. cerevisiae to characterize the Memo-homologue Mho1 (Yjr008wp) and to investigate its function in yeast. In a synthetic lethal screen we found MHO1 as a novel synthetic lethal partner of PLC1, which encodes the single phospholipase C in yeast. Double-deleted cells lacking MHO1 and PLC1, proliferate for up to ten generations. Introduction of human Memo into the memoΔplc1Δ strain rescued the synthetic lethal phenotype suggesting that yeast and human proteins have similar functions. Mho1 is present in the cytoplasm and the nucleus of yeast cells; the same distribution of Memo was found in mammalian cells. None of the Memo homologues have a characteristic nuclear localization sequence, however, a conserved nuclear export sequence is found in all. In mammalian cells, blocking nuclear export with Leptomycin B led to nuclear Memo accumulation, suggesting that it is actively exported from the nucleus. In yeast MHO1 expression is induced by stress conditions. Since invasive growth in S. cerevisiea is also stress-induced, we tested Mho1's role in this response. MHO1 deletion had no effect on invasion induced by nutrient deprivation, however, Mho1 overexpression blocked the invasive ability of yeast cells, suggesting that Mho1 might be acting in a dominant negative manner. Taken together, our results show that MHO1 is a novel synthetic lethal interactor with PLC1, and that both gene products are required for proliferation. This paper will discuss the study of turbulent and mean airflow exiting air terminal devices and surrounding occupants seated in classroom desks for slightly warm environments equipped with personalized ventilation systems with upper and lower air terminal devices. In the turbulent airflow analysis the air root mean square, the air turbulence intensity, and the air velocity fluctuations frequencies are calculated, while in the mean airflow analysis the mean air velocity and temperature, the human body skin temperature, and the thermal comfort indexes are evaluated using a multi-node thermal regulation model for two different airflow rates. In the experimental tests made in a wood chamber a manikin, a ventilated desk, and two interior climate analyzers are used. The fluctuations of air velocity and temperature are measured in the air terminal devices and in 15 human body sections around the manikin, while the mean value of air relative humidity and mean radiant temperature are evaluated inside the experimental chamber. The mean air temperature in the air terminal devices is around 28°C (82.4°F), while the mean radiant temperature in the occupation area, the mean air temperature far from the occupation area, and the internal mean air relative humidity in the occupation area are around 28°C (82.4°F), 28°C (82.4°F), and 50%, respectively. The airflow rate in tests I and II are 25.75 m³/h (15.16 ft³/min) and 48.04 m³/h (28.27 ft³/min), respectively. The mean air velocity, root mean square, and turbulence intensity for test I are 0.59 m/s (1.94 ft/s), 0.13 m/s (0.43 ft/s), and 22.4%, in the upper air terminal device, and 0.9 m/s (2.96 ft/s), 0.15 m/s (0.49 ft/s), and 16.7%, in the lower air terminal device; while, for test II they are 1.72 m/s (5.64 ft/s), 0.16 m/s (0.52 ft/s), and 9.4%, in the upper air terminal device, and 1.06 m/s (3.48 ft/s), 0.16 m/s (0.52 ft/s), and 14.9%, in the lower air terminal device. In test I the mean air velocity and the airflow rate are higher in the lower exit air terminal device than in the upper exit air terminal device; while in test II, the opposite is true. It is also true that the skin temperature is slightly lower in test II than in test I, mainly in human body sections near the air terminal devices, such as the chest, arms, and legs. The occupant in test I conditions is thermally uncomfortable; however, in test II conditions, the obtained results are near the comfort recommendations. [ABSTRACT FROM AUTHOR]

Abstract: In this work the comfort level, namely the thermal comfort, local thermal discomfort and air quality levels, in a classroom with desks equipped with two personalized ventilation systems, in slightly warm environments, is evaluated. A manikin, a ventilated classroom desk, two indoor climate analyzers, a multi-nodal human thermal comfort numerical model and a computational fluid dynamic numerical model, are used. The classroom desk, with double occupation capacity, is used by a student, located in the right side seat. Each personalized ventilation system is equipped with one air terminal device located above the desk writing area, in front to the trunk area, and an other located below the desk writing area, in front to the legs area. The thermal comfort level is evaluated by the developed multi-nodal human thermal comfort numerical model, using a PMV value, the local thermal discomfort level, namely the draught risk and the air velocity fluctuation equivalent frequencies, is evaluated by empirical models, while the air quality level and the detailed airflow around the manikin are evaluated by the computational fluid dynamic numerical model. In the experimental tests the mean air velocity and the turbulence intensity in the upper air terminal device are 3.5m/s and 9.7%, while in the lower air terminal device are 2.6m/s and 15.2%. The mean air temperature in the air terminal devices is around 28°C, while the mean radiant temperature in the occupation area, the mean air temperature far from the occupation area and the internal mean air relative humidity were, respectively, 28°C, 28°C and 50%. The air velocity and temperature around the occupant are measured around 15 human body sections. The actual personalized ventilation system, which promotes an ascendant airflow around the occupant with highest air renovation rate in the respiration area, promotes acceptable thermal comfort conditions and air quality in the respiration area in accord to the present standards. The draught risk is verified in the head and left leg and the uncomfortable air velocity equivalent frequency is verified in the left arm. The left human body sections present higher local thermal discomfort levels than the right human body sections, because they are also influenced by the left air terminal device. In accord to the obtained results, the classroom desk design, equipped with two personalized ventilation systems, guarantees acceptable thermal comfort conditions and promotes good air quality conditions, with acceptable local thermal discomfort conditions and with low energy consumption level. 34. Unraveling natalizumab effects on deregulated miR-17 expression in CD4+ T cells of patients with relapsing-remitting multiple sclerosis.

MicroRNAs (miRNAs) are a family of noncoding RNAs that play critical roles in the posttranscriptional regulation of gene expression. Accumulating evidence supports their involvement in the pathogenesis of multiple sclerosis (MS). Here, we compare miR-17 expressions in CD4+ T cells from relapsing-remitting (RR) MS patients treated with natalizumab versus untreated patients. miR-17 was downregulated under natalizumab treatment and upregulated during relapse, therefore supporting a possible role of miR-17 in MS immunopathogenesis. Downregulation of miR-17 was associated with upregulation of PTEN, BIM, E2F1, and p21 target genes. In vitro miR-17 inhibition was associated with upregulation of the same targets and resulted in impaired CD4+ T cell activation and proliferation. We further describe deregulated TGFBR2 expression in untreated patients versus healthy volunteers (HVs) and confirm in vitro the link between miR-17 and TGFBR2 expressions. These findings support an effect of natalizumab on expression of specific miRNA and subsequent expression of genes involved in proliferation and control of the cell cycle. [ABSTRACT FROM AUTHOR]

Heregulin (HRG) activates ErbB2-ErbB3 heterodimers thereby stimulating many cellular responses, including motility. Memo and PLC1 interact with ErbB2 autophosphorylation sites and are essential for HRG-induced chemotaxis. By tracing HRG-stimulated cell migration in Dunn chambers, we found that Memo- or PLCγ1 knockdown (KD) strongly impairs cell directionality. Memo has no obvious enzymatic activity and was discovered via its ability to complex with ErbB2. Using the yeast two-hybrid approach to gain insight into Memo function, an interaction between Memo and cofilin, a regulator of actin dynamics, was uncovered. The interaction was confirmed in vitro using recombinant proteins and in vivo in co-immunoprecipitation experiments where Memo was detected in complexes with cofilin, ErbB2 and PLCγ1. Interestingly, in Memo KD cells, HRG-induced PLCγ1 phosphorylation was decreased, suggesting that Memo regulates PLCγ1 activation. Furthermore, HRG-induced recruitment of GFP-cofilin to lamellipodia is impaired in Memo and in PLCγ1 KD cells, suggesting that both proteins lie upstream of cofilin in models of ErbB2-driven tumor-cell migration. Finally, in vitro F-actin binding and depolymerization assays showed that Memo enhances cofilin depolymerizing and severing activity. Introduction: Integrality is associated with the various dimensions of life, considering the tangible and subjective aspects related to the complex unity of the human being. Objective: To promote a critical reflection on the integrality of the human being, in the scope of care, as the essence of health practices. Method: This is a study developed as a reflective essay that approached care in an existential philosophical perspective, aiming at integrality for health promotion. Development of the theme: Human care, understood as much more than doing or providing care, is projected beyond duty and becomes care and compassion. It covers attitudes that should be part of the analysis and critical reflection of professionals working in the health area. Conclusion: The recovery of integrality in the health care provided during the practice requires a reflexive posture of the health professional in order to value all the dimensions of the human being, including the spiritual one. 37. Developing an Animal Model for Swenson Transanal Endorectal Pull-Through: A New Possibility for Training and Research Purposes.

As far as we know, no report uses the Swenson transanal endorectal pull-through technique in an animal model. Our objective is to describe the use of this technique as an experimental model for training and research purposes. Ten Norfolk hybrid rabbits were randomly selected from our experimental laboratory, with a mean weight of 3539.3 (± 678.4) g. Neither colon preparation nor fast were used before the procedures. The surgical technique was based on the description performed by Levitt et al. (2013, J Pediatr Surg . 2013;48(11):2289-2295). Information related to the surgical procedures and the clinical evolution in the postoperative period were recorded and analyzed. There were no deaths or severe complications. The anesthetic and the surgical times were significantly higher for the first three animals of the experiment. Our animal model proved adequate to perform the transanal endorectal Swenson pull-through technique, allowing the training of surgical skills through a model similar to the human, with few anesthetic complications and good postoperative evolution, including postoperative follow-up. We believe that it will serve as a learning tool in many institutions that are continuously searching for improved new techniques and will support new researches in this area.

- Abstract
Therapeutic efficacy with durable responses has been demonstrated with several antibody drugs that block key immune checkpoint receptors, including PD-1, PD-L1, and CTLA-4. Despite the success of these drugs, a substantial proportion of patients do not benefit. Targeting multiple inhibitory pathways simultaneously to augment anti-tumor immunity has proven to be a promising approach. The emergence of Repulsive Guidance Molecule b (RGMb), a ligand for PD-L2, as a novel co-inhibitory pathway in T cells, together with its regulation by the gut microbiome, encouraged the discovery and development of fully human anti-RGMb antibodies. Here, we describe phage display-derived monoclonal antibodies (mAbs) 2C11 and 5C10 that bind human RGMb with high affinities of 1.4 nM and 0.72 nM, respectively. Both mAbs 2C11 and 5C10 potently inhibited RGMb interaction with PD-L2. MAb 2C11 effectively inhibited RGMb interaction with bone morphogenetic proteins 2 and 4 (BMP2-4), while leaving RGMb interaction with Neogenin 1 (Neo1) unaffected. Conversely, mAb 5C10 disrupted RGMb interaction with Neo1 while maintaining RGMb binding to BMP2-4. These findings map the 2C11 epitope at the membrane-distal N-terminal region of RGMb, which coincides with both PD-L2- and BMP2-4-binding sites. The PD-L2 binding interface is likely positioned between RGMb's N-terminal BMP-binding and C-terminal Neo1-binding regions. The in vivo activity of mAb 2C11 in combination with anti-PD-1 or anti-PD-L1 was tested in MC38 and B16-OVA cancer models and demonstrated synergistic effects by significantly enhancing anti-tumor responses. These properties make mAb 2C11 a promising candidate for therapeutic use to overcome immune checkpoint inhibitor resistances, warranting further exploration in clinical settings.

39. Natalizumab-induced POU2AF1/Spi-B upregulation: A possible route for PML development.

Objectives: To assess messenger RNA (mRNA) expression of POU2AF1 and Spi-B and their potential regulatory microRNAs (miRNAs) in natalizumab-treated patients with multiple sclerosis and in therapy-associated progressive multifocal leukoencephalopathy (PML)., Methods: Expression of POU2AF1/Spi-B was analyzed by using real-time reverse transcription PCR assays on isolated B/CD8(+) T lymphocytes and peripheral blood mononuclear cells (PBMCs) from cohorts of untreated and natalizumab-treated patients with and without PML. Longitudinal expression analysis was performed on CD4(+), CD8(+) T and B cells from 14 patients who interrupted natalizumab therapy for 8 weeks. The miRNA profiling was conducted in PBMCs from 5 untreated and 5 natalizumab-treated patients using low-density arrays followed by validation with single miRNAs assays in untreated and natalizumab-treated patients., Results: POU2AF1 and Spi-B mRNAs were upregulated in B and CD8(+) T cells from natalizumab-treated patients, which was validated in PBMCs from different cohorts of natalizumab-treated patients with and without PML, with a noteworthy higher expression of Spi-B in patients with PML. In contrast, downregulation of POU2AF1/Spi-B expression was measured in B and CD8(+) T cells after natalizumab discontinuation. Seventeen differentially expressed miRNAs including miR-10b, a regulator of POU2AF1 mRNA, were identified in long-term natalizumab-treated patients compared with untreated ones., Conclusions: Upregulation of POU2AF1 and Spi-B, known transactivators of the JC virus, the causative agent for PML, and its association with occurrence of PML in natalizumab-treated patients, corroborates POU2AF1/Spi-B as potential biomarkers for PML risk, which merits further evaluation.

Objective: to support the assessment and the necessary changes in the curriculum of an undergraduate degree in Nursing., Methods: it is a descriptive study developed in two stages as an action research. In the first stage, the speeches of 19 graduates and 15 employers were interviewed and analyzed; in the second stage, five teachers participated in the focus group. For processing the data, the content analysis was adopted., Results: based on the assessment of the actors, an action plan was created with suggestions regarding the curriculum flexibility, content resizing, continuing education, practice enhancement, active methodologies and autonomy of the student., Conclusion: the participation of actors participants of the training process and performance of professional nurses in the evaluation of a curriculum pointed aspects to realignment and provided concrete benefits to improving the quality of undergraduate course curriculum, scenario of this study.

- Abstract
This research aimed to identify political-ethical skills developed in a training process compatible with the expected profile set by the National Curriculum Guidelines for the Undergraduate Nursing Degree. A case study was conducted with units represented by 32 former students from a particular religious teaching institution who already were in the job market. The content of the interviews was analyzed using the thematic analysis technique, which resulted in the following categories: "Political-ethical skills in the formative process" and "Political-ethical skills as a product of the educational process." From the former students' perspective, these categories reinforced the social role of the nurse and the need for students to be reflective, understanding and participative in the transformation of society.

Every day, college graduates face complex situations that make them check skills developed during the course in comparison with the skills needed at the professional level. This experience helps them evaluate the adequacy of the course's teaching structure and recover intervening aspects of this process. This text discusses the Evaluation of College Courses, recovering concepts in the field of Education Evaluation; describes the practices developed in Brazil, directed towards course evaluation, based on the graduate's perspective. The following descriptors were used: college graduates, indicators, evaluation and graduation available in the databases: PeriEnf, BDEnf, LILACS and DEDALUS. The analysis of the surveyed studies highlights the need to consider the opinion of college graduates in the curriculum proposals with a view to implementing an educational process with emancipating and transforming characteristics.

Background: stuttering., Aim: to compare muscle activation in fluent and stuttering individuals during speech and non-speech tasks., Method: six adults divided in two groups: G1 - three fluent individuals; G2 - three stuttering individuals. Muscle activity (surface electromyography) was captured by disposable electrodes fixed in four regions. Testing situations: muscle rest tension, speech reaction time, non-verbal activity, verbal activity., Results: There was no significant statistical difference between the groups for the rest tension; G2 present longer speech reaction times; G2 presented muscle activity during the non-verbal task similar to that observed during rest; Muscle activity of G1 and G2 during the verbal task demonstrated to be similar., Conclusion: these results suggests that for G2 there is a poor control of timing for the coordination of motor processes.
