5. International PhD Students in Australian Universities: Financial Support, Course Experience and Career Plans

Abstract

Using data from a social survey of PhD students in two major Australian universities supplemented by student interviews, this article reports on the financial support, course experience and career plans of international PhD students. While most international PhD students hold scholarships which include stipends, a minority of students experience financial problems and lack adequate research support. Overall international PhD students express a high degree of satisfaction with their courses, although there are concerns about the quality and effectiveness of supervision, working space available to research students and help provided in designing research projects. Language problems sometimes adversely affect student progress while some international students find difficulty adjusting to a less deferential working arrangement with their supervisors and less structure in research direction. International PhD students are optimistic about their career prospects and certainly more confident about their careers than Australian PhD students. High proportions of international PhD students expect to follow research careers and say that the PhD degree will enhance their career prospects.

Abstract

A gene encoding an alkaline (pI of 8.67) chitinase was cloned and sequenced from Chromobacterium sp. strain C-61. The gene was composed of 1,611 nucleotides and encoded a signal sequence of 26 N-terminal amino acids and a mature protein of 510 amino acids. Two chitinases of 54 and 52 kDa from both recombinant Escherichia coli and C-61 were detected on SDS-PAGE. Maximum chitinase activity was obtained in the culture supernatant of recombinant E. coli when cultivated in TB medium for 6 days at 37°C and was about fourfold higher than that from C-61. Chi54 from the culture supernatants could be purified by a single step based on isoelectric point. The purified Chi54 had about twofold higher binding affinity to chitin than to cellulose. The chi54 encoded a protein that included a type 3 chitin-binding domain belonging to group A and a family 18 catalytic domain belonging to subfamily A. In the catalytic domain, mutation of perfectly conserved residues and highly conserved residues resulted in loss of nearly all activity, while mutation of nonconserved residues resulted in enzymes that retained activity. In this process, a mutant (T218S) was obtained that had about 133% of the activity of the wild type, based on comparison of K cat values. [ABSTRACT FROM AUTHOR]

Abstract

The anti-inflammatory activities of methotrexate and sulphasalazine may be mediated by increases in endogenous adenosine levels. Since the vascular protective drug dipyridamole inhibits the uptake and metabolism of adenosine we have now tested this compound in patients with rheumatoid arthritis to assess its effects on their symptoms. Forty patients (aged 18–75 years) received dipyridamole 400 mg/day or placebo. The levels of adenosine and its major metabolites were determined by high performance liquid chromatography (HPLC) in blood samples taken at baseline and at monthly intervals during treatment for 6 months. After three months of treatment there was a significant reduction in the modified Health Assessment Questionnaire (mHAQ) score, but these effects were not maintained, and dipyridamole did not modify disease severity scores or the levels of adenosine and its metabolites. We conclude that the symptoms of rheumatoid arthritis were not modified by treatment with dipyridamole. [ABSTRACT FROM AUTHOR]

Abstract

Since free radicals are implicated in rheumatoid arthritis(RA) and since uric acid is a free radical scavenger, we examined the effects of treating RA patients with with the diuretic bumetanide to try to improve their arthritic control. Seventy patients, aged 18–75 years, were randomised to receive bumetanide 4 mg/day or placebo. Uric acid levels increased, but not that of other purines, in the blood of drug-treated patients compared with placebo-treated controls. There were no significant changes in clinical measurements of disease activity or in ESR or CRP levels. There were no over all differences in the blood levels of the cytokines, nor in the basal or stimulated production of cytokines from the blood cultures. The adenosine receptor agonist 5′Nethylcarboxamido- adenosine(NECA) used to modify cytokine release in cultures of whole blood taken from the patients, depressed the release of tumour necrosis factor-a(TNFa), but failed to depress the release of interleukin-1b(IL-1b) or interleukin-6(IL-6), a difference from earlier studies of healthy control subjects and, thus, a difference which may contribute to the disease activity. Abstract

Objectives: Treatment with diuretics can increase blood orate levels, and urate is a potent free radical scavenger. Since free radicals are implicated in rheumatoid arthritis (RA), we examined the effects of treating patients with rheumatoid arthritis with bumetanide to try to improve their arthritic control.Methods: Seventy patients were recruited from routine rheumatology clinics and were randomized to receive bumetanide 4 mg/ day or placebo. We measured blood levels of urate, other major purine metabolites, lipid peroxidation products, and the cytokines tumor necrosis factor-a (TNF-alpha), interleukin-l(beta (IL-Ibeta) and interleukin-6 (IL-6).Results: Increased levels of orate, but not other purines, were found in the blood of drug-treated patients compared with placebo-treated controls. There were no significant changes in clinical or biochemical measurements of disease activity. There were no overall differences in the blood levels of the cytokines, nor in the basal or stimulated production of cytokines from the blood cultures.Conclusion: Bumetanide increases blood urate levels but this is not associated with disease improvement.
