Several theoretical frameworks have been used to understand exercise adherence. Basic psychological needs (BPN), grounded on self‐determination theory, have received large attention for this purpose. More recently, the hedonic theory and the assumptions related to the exercise characteristics (e.g., intensity) that can bolster positive affective responses have been revitalized. This study aimed to explore the associations between the agreement of current exercise intensity and the one individually preferred, BPN satisfaction/frustration, enjoyment, the intention to continue exercise, and exercise frequency. Additionally, an exploration of the direct and indirect effects while testing sex as a moderator was performed. A sample of 369 exercisers (Mage = 43.6, standard deviation = 12.96; 214 females) enrolled in 26 health clubs participated in this study voluntarily. Data were received in a first‐approach descriptive and correlational analyses. Next, a moderated mediation was performed using model 15 (PROCESS v.4.2). As a result, agreement in preference for exercise intensity was positively associated with enjoyment (r = 0.35), intention (r = 0.43), and all needs satisfaction (ranging from r = 0.12 to r = 0.45) and negatively associated with all needs frustration (ranging from r = −0.15 to r = −0.31). In the moderated mediation analysis, the same pattern of results emerged in direct effects. Indirect effects were significant for autonomy in the enjoyment and frequency models. Findings suggest that promoting an individually adjusted training intensity may foster BPN satisfaction. It appears to be present an independent (of needs) and positive association with exercise enjoyment and intention to continue exercising. Silver nanoparticles (AgNPs) are widely used in industrial, cosmetic, and biomedical products, and humans are frequently exposed to these products through the skin. It is widely recognized that the characteristics of AgNPs (e.g., size, coating) may influence their cytotoxic effects, but their correlation with DNA damage and mitotic disorders remains poorly explored. In this study, human keratinocytes (HaCaT cell line) were exposed to well-characterized 30 nm AgNPs coated with citrate, and their effects on viability, DNA fragmentation (assessed by the comet assay), and micronuclei (MNi) induction (assessed by the cytokinesis-block micronucleus cytome assays, CBMN) were investigated. The results showed that 10 and 40 μg/mL AgNPs decreased cell proliferation and viability, and induced a significant genetic damage. This was observed by an increase of DNA amount in comet tail, which linearly correlated with dose and time of exposure. Also, cytostaticity (increase of mononucleated cells) and MNi rates increased in treated cells. In contrast, no significant changes were observed in nucleoplasmatic bridges (NPBs) or nuclear buds (NBUDs), although NBUDs tended to increase in all conditions and periods. The cytostatic effects on HaCaT cells were also shown by the decrease of their nuclear division index. Thus, both comet and CBMN assays supported the observation that citrate-AgNPs induced genotoxic effects on HaCaT cells. Considering that AgNPs are present in a vast number of consumer products and also in multiple nanomedicine skin applications and formulations, more research is needed to determine the properties that confer less toxicity of AgNPs to different cell lines. Advances in geotechnologies and in remote sensing have improved analysis of urban environments. The new sensors are increasingly suited to urban studies, due to the enhancement in spatial, spectral and radiometric resolutions. Urban environments present high heterogeneity, which cannot be tackled using pixel-based approaches on high resolution images. Geographic Object-Based Image Analysis (GEOBIA) has been consolidated as a methodology for urban land use and cover monitoring; however, classification of high resolution images is still troublesome. This study aims to assess the improvement on ceramic roof classification using WorldView-2 images due to the increase of 4 new bands besides the standard "Blue-Green-Red-Near Infrared" bands. Our methodology combines GEOBIA, C4.5 classification tree algorithm, Monte Carlo simulation and statistical tests for classification accuracy. Two samples groups were considered: 1) eight multispectral and panchromatic bands, and 2) four multispectral and panchromatic bands, representing previous high-resolution sensors. The C4.5 algorithm generates a decision tree that can be used for classification; smaller decision trees are closer to the semantic networks produced by experts on GEOBIA, while bigger trees, are not straightforward to implement manually, but are more accurate. The choice for a big or small tree relies on the user's skills to implement it. This study aims to determine for what kind of user the addition of the 4 new bands might be beneficial: 1) the common user (smaller trees) or 2) a more skilled user with coding and/or data mining abilities (bigger trees). Surface coating of silver nanoparticles may influence their toxicity, in a way yet to decipher. In this study, human keratinocytes (HaCaT cells) were exposed for 24 and 48 h to well-characterized 30 nm AgNPs coated either with citrate (Cit30 AgNPs) or with poly(ethylene glycol) (PEG30 AgNPs), and assessed for cell viability, reactive oxygen species (ROS), cytokine release, apoptosis and cell cycle dynamics. The results showed that Cit30 AgNPs and PEG30 AgNPs decreased cell proliferation and viability, the former being more cytotoxic. The coating molecules per se were not cytotoxic. Moreover, Ag + release and ROS production were similar for both AgNP types. Cit30 AgNPs clearly induced apoptotic death, while cells exposed to PEG30 AgNPs appeared to be at an earlier phase of apoptosis, supported by changes in BAX, BCL2 and CASP-3 expressions. Concerning the impact on cell cycle dynamics, both Cit30 and PEG30 AgNPs affected cell cycle regulation of HaCaT cells, but, again, citrate-coating induced more drastic effects, showing earlier downregulation of cyclin B1 gene and cellular arrest at the G2 phase. Overall, this study has shown that the surface coating of AgNPs influences their toxicity by differently regulating cell-cycle and cell death mechanisms. [ABSTRACT FROM AUTHOR]

In the present study, a practical activity is proposed to adopt an experimental approach to demonstrate the relationship between the equilibrium potential for K+ and transmembrane electrical potential without glass micropipettes. A conventional setup for recording contractile activity of isolated smooth muscle preparations was used based on the events elegantly described by Somlyo and Somlyo in the 1960s. They showed that, in response to a given stimulus, smooth muscle cells may contract, recruiting electromechanical or pharmacomechanical coupling by mechanisms that involve, or not, changes in transmembrane potential, respectively. By means of contractions and relaxations of a ring-like preparation from the rat mesenteric artery, it is possible to observe the functional consequences of handling K+ concentration in the extracellular compartment and the effects caused by opening K+ channels in that preparation, which are significant when the cell membrane establishes an electrical potential difference between intra- and extracellular compartments (driven mainly by K+ permeability under resting conditions). The effects observed by students fit well with values predicted by Nernst and Goldman-Hodgin-Katz equations, and we demonstrated that the activity is able to improve students' comprehension regarding basic principles of bioelectricity. Nanoparticles (NPs) are characterized by their small size (less than 100 nm) and large surface area, which confer specific physicochemical properties as strength, electrical, and optical features. NPs can be derived from natural or anthropic sources, such as engineered or unwanted/incidental NPs. The composition, dimension, and morphology of engineered NPs enable their use in a variety of areas, such as electronic, biomedical, pharmaceutical, cosmetic, energy, environmental, catalysis, and materials science. As nanotechnology is an innovative and scientific growth area with an exponential production, more information is needed concerning the impacts of these nanomaterials (NMs) in the environment and, particularly, in animals/humans health and in plants performance. So, research on NPs as emerging contaminants is therefore a new field in environmental health. This minireview describes, briefly, the NPs characterization and their occurrence in the environment stating air, water, and soil. Finally, particular emphasis is given to the interaction of NPs with plants at different levels: morphology, physiology, and genotoxicity. By analyzing this compiled information, it is evident that research on NPs phytotoxicity is in the beginning, and more comprehensive studies are needed not only on NPs cytotoxicity and genotoxicity but also on the best and the most reliable methods of assessing NPs toxicity. The values of Michaelis–Menten constant (KM) and maximum velocity (VMAX) for kidney and heart monoamine oxidase (MAO) from pacu Piaractus mesopotamicus were determined. The mean ±s.e. KM values were 17·28 ± 2·27 μM for kidney and 15·38 ± 1·86 μM for heart. MAO activities were 111·60 ± 3·25 and 15·12 ± 0·30 nmols min−1 g−1 of wet tissue for kidney and heart, respectively. The article describes the presence of cytochromes P450 in liver microsomes from pacu and its major specific deethylase activity, which was inducible by β-napthoflavone. Pacus of both sexes weighing around 400 gram were donated by the National Centre for Research on Tropical Fish of the Brazilian Environment and Renewable Natural Resources Institute. They were acclimated for three weeks in 500 liter tanks equipped with biologic filter and filled with aerated dechlorinated freshwater at 24 &degree;Celsius.

Inclusive differential cross sections dσpA/dxF and dσpA/dpt² for the production of KS0, Λ, and Λ particles are measured at HERA in proton-induced reactions on C, Al, Ti, and W targets. The incident beam energy is 920 GeV, corresponding to √s = 41.6 GeV in the proton-nucleon system. The ratios of differential cross sections dσpA(KS0)/dσpA(Λ) and dσpA(Λ)/dσpA(Λ) are measured to be 6.2 ± 0.5 and 0.66 ± 0.07, respectively, for xF ≈ -0.06. No significant dependence upon the target material is observed. Within errors, the slopes of the transverse momentum distributions dσpA/dpt² also show no significant dependence upon the target material. The dependence of the extrapolated total cross sections σpA on the atomic mass A of the target material is discussed, and the deduced cross sections per nucleon σpN are compared with results obtained at other energies. The metabolism of hypogean organisms is frequently molded by the cave environment traits, especially food scarcity. The aim of the present work was to evaluate the inf luence of such environment on lipid composition and hepatic lipogenesis in the f ish Ancistrus cryptophthalmus. For this, the species was compared to an epigean population of the species. A greater accumulation of total lipids was observed in the cave-dwelling f ish (18.36 g/100 g tissue) compared to the surface f ish (14.09 g/100 g tissue). The muscle fatty acid prof ile also varied between the populations. Arachidonic acid was only detected in the epigean f ish, while docosahexaenoic acid was present in the cave f ish. In the lipid prof ile of Ancistrus cryptophthalmus there was a higher proportion of saturated fatty acids, followed by monounsaturated and polyunsaturated fatty acids; Ancistrus sp. showed a predominance of monounsaturated fatty acids. Signif icant differences were also observed in the activities of the hepatic enzymes glucose-6-phosphate dehydrogenase and malic enzyme. The activities of these two enzymes were greater in the epigean animals. The differences could be related to different food availability observed in the two environments. An ecotone zone was observed, located next to the entrance of the Lapa do Angélica cave (Goiás State, Brazil), where the f ishes showed characteristics that were intermediate between those of hypogean f ishes from deeper within the cave, and the epigean population. It could be concluded that the characteristics of the cave environment signif icantly inf luenced the composition of muscle fatty acids and lipogenesis in the hypogean f ish Ancistrus cryptophthalmus.

Background: Affective responses are increasingly recognized as potentially effective intervention targets that may facilitate exercise and physical activity behavior change. While emerging correlational evidence suggests that more pleasant affective responses are associated with higher participation and adherence, experimental evidence remains scarce. In light of this, we conducted a preregistered, pragmatic, single-blinded, superiority randomized controlled trial with two parallel groups, with the goal of determining the impact of an individualized exercise-intensity prescription targeting pleasure on exercise frequency., Methods: Forty-seven non-regular exercisers were randomized into two groups. For both groups, the intervention consisted of three exercise sessions based on the Frequency-Intensity-Time-Type (FITT) principle. However, the experimental group also received an individualized intensity prescription based on prior assessment of preference for and tolerance of exercise intensity, as well as instructions emphasizing the promotion of pleasure as a basis for self-regulating exercise intensity. The primary outcome was gymnasium attendance over an eight-week follow-up period. Secondary outcomes were affective valence and arousal, post-exercise enjoyment, core affective exercise experiences, and anticipated and remembered affect., Results: Forty-six participants were retained for analysis (M age  = 32.00; SD = 8.62 years; 56.5% female). Compared to the control group, the experimental group exhibited 77% higher session attendance (14.35 vs. 8.13 sessions) over the eight-week follow-up period (group main effect p = .018, η 2 p  = .120; Cohen's d ranged from 0.28 to 0.91 during follow-up). Also, the experimental group reported higher levels of pleasure during the intervention sessions (for all group main effects, p < .001, η 2 p from .33 to .37) and higher levels of remembered pleasure (group main effect p = .021, η 2 p  = .116) and anticipated pleasure (group main effect p = .022, η 2 p  = .114). The intensity of resistance training (RT) exercise is an important consideration for determining relevant health and performance-related outcomes. Yet, current objective exercise intensity measures present concerns in terms of viability or cost. In response to these concerns, repetition-in-reserve (RIR) scales may represent an adequate method of measuring and regulating intensity. However, no recent review has focused on how RIR scales have been used for this purpose in prior research. We prepared the present scoping review to analyze the feasibility and usefulness of RIR scales in selecting RT intensity. We conducted a systematic search in PubMed, SPORTDiscus, PsycINFO, and ClinicalTrials.gov databases (last search date April 2023) for experimental and non-experimental studies that utilized an RIR scale to measure proximity to failure in RT activities with apparently healthy individuals of any age. We qualitatively analyzed 31 studies (N = 855 mostly male adult participants) published between 2012-2023. RIR scales appeared to be contextually feasible and useful in prescribing and adjusting RT intensity. The most common trend in this research was to prescribe a target RIR and adjust the exercise load for a desired proximity to muscle failure. Additionally, when measuring proximity to failure as an outcome of interest, the literature suggests that the RIR prediction should be made close to task failure to increase its accuracy. In this work, hybrid materials within the polydimethylsiloxane-silica (PDMS-SiO 2 ) system, synthesized via the sol-gel method, were developed and characterized for their potential to incorporate and release the bioactive compound resveratrol (RES). RES was incorporated into the materials with a high loading efficiency (>75%) using the rotary evaporator technique. This incorporation induced the amorphization of RES, resulting in enhanced solubility and in vitro release when compared to the free polyphenolic compound. The release profiles displayed pH dependence, exhibiting notably faster release at pH 5.2 compared to pH 7.4. The gradual release of RES over time demonstrated an initial time lag of approximately 4 h, being well described by the Weibull model. In vitro cytotoxicity studies were conducted on human osteosarcoma cells (MG-63), revealing a concentration-dependent decrease in cell viability for RES-loaded samples (for concentrations >50 µg mL -1 ).

Cancer is a huge public health problem being one of the main causes of death globally. Specifically, melanoma is one of the most threatening cancer types due to the metastatic capacity, treatment resistance and mortality rates. It is evident the urgent need for research on new agents with pharmacological potential for cancer treatment, in order to develop new cancer therapeutic strategies and overcome drug resistance. The present work investigated the anti-tumoral potential of Chartergellus-CP1 peptide, isolated from Chartergellus communis wasp venom on human melanoma cell lines with different pigmentation degrees, namely the amelanotic cell line A375 and pigmented cell line MNT-1. Chartergellus-CP1 induced selective cytotoxicity to melanoma cell lines when compared to the lower induced cytotoxicity towards to nontumorigenic keratinocytes. Chartergellus-CP1 peptide induced apoptosis in both melanoma cell lines, cell cycle impairment in amelanotic A375 cells and intracellular ROS increase in pigmented MNT-1 cells. The amelanotic A375 cell line showed higher sensitivity to the peptide than the pigmented cell line MNT-1. Evidence suggests affective responses to exercise can influence exercise adherence. However, there is a limited understanding of how and when to measure core affect in resistance training. As such, the objective of this systematic review was to analyze how the Feeling Scale and/or the Felt Arousal Scale have been used in resistance training to assess core affect. Focus was given to the contextual feasibility, timing, and frequency of assessment. A search in PubMed, SPORTDiscus, and PsycINFO databases was conducted (last search date July, 2022) with the purpose of including experimental and non-experimental studies, utilizing the Feeling Scale and/or the Felt Arousal Scale in resistance training, and focused on apparently healthy individuals of any age. Twenty-seven studies (N = 718 participants) published between 2009-2022 were qualitatively analyzed. Both scales appeared to be able to detect core affect within a wide array of intensities, ages, and equipment. As for the timing and frequency of measurement, no apparent standardization was evident. The use of the Feeling Scale, the Felt Arousal Scale, or both, to measure core affect appears to be feasible in resistance training practices. Affective responses during exercise have been identified as a predictor of exercise adherence. However, research has been mostly limited to aerobic and resistance exercise. Considering that stretching activities are also an important component of physical fitness, this quasi-experimental study was designed to: 1) compare affective responses during and immediately after stretching exercises in apparently healthy adults, and 2) assess the consistency and repeatability of affect ratings obtained one week apart. For this purpose, we analyzed the Feeling Scale (FS) and Felt Arousal Scale (FAS) ratings using Time (during and after stretching) x Intensity (light, moderate, vigorous) x Stretched Muscle Group (quadriceps, hamstrings, glutes, latissimus dorsi, triceps) with repeated measures analysis of variance (ANCOVA) in 34 participants (21 males; aged 32.8 ± 8.6 years). The repeatability of FS and FAS ratings was assessed using two-way random-effects models, Intraclass Correlation Coefficients (ICC), and Bland-Altman plots. FS scores were higher following the stretching exercises, whereas FAS scores were lower, particularly in the vigorous intensity. In general, the inter-day repeatability for FS and FAS measurements was good across muscle groups. ICC tended to be higher at vigorous intensities. Ratings of core affect can be collected during static passive stretches using the FAS and FAS in ecologically valid settings. The formamidopyrimidine DNA glycosylase (Fpg)-modified comet assay is widely used for the measurement of oxidatively generated damage to DNA. However, there has not been a recommended long-term positive control for this version of the comet assay. We have investigated potassium bromate as a positive control for the Fpg-modified comet assay because it generates many Fpg-sensitive sites with a little concurrent generation of DNA strand breaks. Eight laboratories used the same procedure for the treatment of monocytic THP-1 cells with potassium bromate (0, 0.5, 1.5, and 4.5 mM) and subsequent cryopreservation in a freezing medium consisting of 50% foetal bovine serum, 40% RPMI-1640 medium, and 10% dimethyl sulphoxide. The samples were analysed by the Fpg-modified comet assay three times over a 3-year period. All laboratories obtained a positive concentration-response relationship in cryopreserved samples (linear regression coefficients ranging from 0.79 to 0.99). However, there was a wide difference in the levels of Fpg-sensitive sites between the laboratory with the lowest (4.2% Tail DNA) and highest (74% Tail DNA) values in THP-1 cells after exposure to 4.5 mM KBrO3. In an attempt to assess sources of inter-laboratory variation in Fpg-sensitive sites, comet images from one experiment in each laboratory were forwarded to a central laboratory for visual scoring. There was high consistency between measurements of %Tail DNA values in each laboratory and the visual score of the same comets done in the central laboratory (r = 0.98, P < 0.001, linear regression). The comet assay is widely used in biomonitoring studies for the analysis of DNA damage in leukocytes and peripheral blood mononuclear cells. Rather than processing blood samples directly, it can be desirable to cryopreserve whole blood or isolated cells for later analysis by the comet assay. However, this creates concern about artificial accumulation of DNA damage during cryopreservation. In this study, 10 laboratories used standardized cryopreservation and thawing procedures of monocytic (THP-1) or lymphocytic (TK6) cells. Samples were cryopreserved in small aliquots in 50% foetal bovine serum, 40% cell culture medium, and 10% dimethyl sulphoxide. Subsequently, cryopreserved samples were analysed by the standard comet assay on three occasions over a 3-year period. Levels of DNA strand breaks in THP-1 cells were increased (four laboratories), unaltered (four laboratories), or decreased (two laboratories) by long-term storage. Pooled analysis indicates only a modest positive association between storage time and levels of DNA strand breaks in THP-1 cells (0.37% Tail DNA per year, 95% confidence interval: -0.05, 0.78). In contrast, DNA strand break levels were not increased by cryopreservation in TK6 cells. There was inter-laboratory variation in levels of DNA strand breaks in THP-1 cells (SD = 3.7% Tail DNA) and TK6 reference sample cells (SD = 9.4% Tail DNA), whereas the intra-laboratory residual variation was substantially smaller (i.e. SD = 0.4%-2.2% Tail DNA in laboratories with the smallest and largest variation). Measurement of DNA migration in the comet assay can be done by image analysis or visual scoring. The latter accounts for 20%-25% of the published comet assay results. Here we assess the intra- and inter-investigator variability in visual scoring of comets. We include three training sets of comet images, which can be used as reference for researchers who wish to use visual scoring of comets. Investigators in 11 different laboratories scored the comet images using a five-class scoring system. There is inter-investigator variation in the three training sets of comets (i.e. coefficient of variation (CV) = 9.7%, 19.8%, and 15.2% in training sets I-III, respectively). However, there is also a positive correlation of inter-investigator scoring in the three training sets (r = 0.60). Overall, 36% of the variation is attributed to inter-investigator variation and 64% stems from intra-investigator variation in scoring between comets (i.e. the comets in training sets I-III look slightly different and this gives rise to heterogeneity in scoring). Intra-investigator variation in scoring was also assessed by repeated analysis of the training sets by the same investigator. There was larger variation when the training sets were scored over a period of six months (CV = 5.9%-9.6%) as compared to 1 week (CV = 1.3%-6.1%). A subsequent study revealed a high inter-investigator variation when premade slides, prepared in a central laboratory, were stained and scored by investigators in different laboratories (CV = 105% and 18%-20% in premade slides with comets from unexposed and hydrogen peroxide-exposed cells, respectively). The results indicate that further standardization of visual scoring is desirable. Toxicological analysis of the effects of natural compounds is frequently mandated to assess their safety. In addition to more simple in vitro cellular systems, more complex biological systems can be used to evaluate toxicity. This dataset is comprised of bright-field microscopy images of chicken-embryo blood cells, a complex biological model that recapitulates several features found in human organisms, including circulation in blood stream and biodistribution to different organs. In the presented collection of blood smear images, cells were exposed to the flavonoid quercetin, and the two mutagens methyl methanesulfonate (MMS) and cadmium chloride (CdCl 2 ). In ovo models offer a unique opportunity to investigate the effects of various substances, pathogens, or cancer treatments on developing embryos, providing valuable insights into potential risks and therapeutic strategies. In toxicology, in ovo models allow for early detection of harmful compounds and their impact on embryonic development, aiding in the assessment of environmental hazards. In immunology, these models offer a controlled system to explore the developing immune responses and the interaction between pathogens and host defenses. Additionally, in ovo models are instrumental in oncology research as they enable the study of tumor development and response to therapies in a dynamic, rapidly developing environment. Thus, these versatile models play a crucial role in advancing our understanding of complex biological processes and guiding the development of safer therapeutics and interventions. The data presented here can aid in understanding the potential toxic effects of these substances on hematopoiesis and the overall health of the developing organism. Moreover, the large dataset of blood smear images can serve as a resource for training machine learning algorithms to automatically detect and classify blood cells, provided that specific optimized conditions such as image magnification and background light are maintained for comparison. This can lead to the development of automated tools for blood cell analysis, which can be useful in research. Rabies is an ancient neuroinvasive viral (genus Lyssavirus, family Rhabdoviridae) disease affecting approximately 59,000 people worldwide. The central nervous system (CNS) is targeted, and rabies has a case fatality rate of almost 100% in humans and animals. Rabies is entirely preventable through proper vaccination, and thus, the highest incidence is typically observed in developing countries, mainly in Africa and Asia. However, there are still cases in European countries and the United States. Recently, demographic, increasing income levels, and the coronavirus disease 2019 (COVID-19) pandemic have caused a massive raising in the animal population, enhancing the need for preventive measures (e.g., vaccination, surveillance, and animal control programs), postexposure prophylaxis, and a better understanding of rabies pathophysiology to identify therapeutic targets, since there is no effective treatment after the onset of clinical manifestations. Here, we review the neuroimmune biology and mechanisms of rabies. Its pathogenesis involves a complex and poorly understood modulation of immune and brain functions associated with metabolic, synaptic, and neuronal impairments, resulting in fatal outcomes without significant histopathological lesions in the CNS. In this context, the neuroimmunological and neurochemical aspects of excitatory/inhibitory signaling (e.g., GABA/glutamate crosstalk) are likely related to the clinical manifestations of rabies infection. Uncovering new links between immunopathological mechanisms and neurochemical imbalance will be essential to identify novel potential therapeutic targets to reduce rabies morbidity and mortality., (© 2023 The Authors. As human activities increasingly shape land- and seascapes, understanding human-wildlife interactions is imperative for preserving biodiversity. Habitats are impacted not only by static modifications, such as roads, buildings and other infrastructure, but also by the dynamic movement of people and their vehicles occurring over shorter time scales. Although there is increasing realization that both components of human activity substantially affect wildlife, capturing more dynamic processes in ecological studies has proved challenging. Here we propose a conceptual framework for developing a 'dynamic human footprint' that explicitly incorporates human mobility, providing a key link between anthropogenic stressors and ecological impacts across spatiotemporal scales. Specifically, the dynamic human footprint integrates a range of metrics to fully acknowledge the time-varying nature of human activities and to enable scale-appropriate assessments of their impacts on wildlife behaviour, demography and distributions. We review existing terrestrial and marine human-mobility data products and provide a roadmap for how these could be integrated and extended to enable more comprehensive analyses of human impacts on biodiversity in the Anthropocene., (© 2023. Research on the relationship between exercise-induced affect and exercise or physical activity behavior has gained momentum in recent years, yielding several observational and longitudinal studies. However, experimental tests demonstrating a causal role of affective responses on exercise adherence are lacking. Given the need to devise exercise prescriptions that can facilitate adherence and promote regular physical activity, a Randomized Controlled Trial targeting individual pleasurable responses in a health-club setting will be conducted. The experimental protocol will compare two individualized evaluation, prescription, and supervision methods, adjusted for intensity, with the aim to explore their impact on behavioral, affective, and motivational outcomes. The planned study will be a pragmatic randomized, single-blinded, controlled superiority trial with two parallel groups and an allocation ratio of 1:1. Apparently healthy volunteer participants (N = 46) between 18 and 45 years old, who are not regularly active, will be randomly allocated to a control or experimental group. The control group will follow a general American College of Sports Medicine exercise prescription; the experimental group will follow the same prescription but with enhancements aimed at promoting pleasure: (1) individualization based on individual differences in preference for and tolerance of exercise intensity; (2) self-regulation of intensity; and (3) repeated assessments of core affect. The primary outcome will be post-intervention exercise-session attendance. Affective and motivational variables will be examined as secondary outcomes. The results are expected to advance exercise prescriptions by promoting affective regulation, thus helping to improve exercise adherence and related outcomes. This study investigated the psychometric nature of preference for and tolerance of exercise intensity in physical activity. It initially re-examined the Preference for and Tolerance of the Intensity of Exercise Questionnaire (PRETIE-Q) among Portuguese exercisers, looking at its applicability to different exercise activities and exercise experiences. Then, to investigate the applicability of the measure in different groups, its invariance was examined. The sample consisted of 1117 participants (528 male, 589 female) aged 18-81 years old (Mage = 36.81, SD = 11.89). All participants reported at baseline that they were exercising, on average, 3.93 days (SD = 1.36) per week. The exploratory structural equation modeling (ESEM) displayed the best fit. The ESEM did show invariance when tested for multigroup analysis. The conclusion of this research is that the ESEM demonstrated the best fit, displaying invariance in multigroup analysis. Furthermore, when assessing preference and tolerance in various exercise modalities, the PRETIE-Q should be primarily used as a multidimensional instrument due to the differential recognition of preference and tolerance in seemingly similar physical activity circumstances, highlighting the importance of employing context-verified measures to evaluate exercise-intensity preference and tolerance based on sample characteristics or real-time context.

Lycopene is a carotenoid with potential use in the treatment of chronic illnesses. Here, different formulations of lycopene were studied: lycopene-rich extract from red guava (LEG), purified lycopene from red guava (LPG) and a self-emulsifying drug delivery system loaded with LPG (nanoLPG). The effects of administering orally various doses of LEG to hypercholesterolemic hamsters were evaluated regarding the liver function of the animals. The cytotoxicity of LPG in Vero cells was analyzed by a crystal violet assay and by fluorescence microscopy. In addition, nanoLPG was employed in stability tests. LPG and nanoLPG were tested for their cytotoxic effect on human keratinocytes and antioxidant capacity on cells in an endothelial dysfunction model in an isolated rat aorta. Finally, the effect of different nanoLPG concentrations on the expression of immune-related genes ( IL-10 , TNF- α, COX-2 and IFN -γ) from peripheral blood mononuclear cells (PBMC) using real-time PCR was also analyzed. Results suggest that LEG, despite not being able to improve blood markers indicative of liver function in hypercholesterolemic hamsters, reduced hepatic degenerative changes. Additionally, LPG did not show cytotoxicity in Vero cells. In relation to nanoLPG, the effects produced by heat stress evaluated by Dynamics Light Scattering (DLS) and visually were loss of color, texture change and phase separation after 15 days without interfering with the droplet size, so the formulation proved to be efficient in stabilizing the encapsulated lycopene. Although LPG and nanoLPG showed moderate toxicity to keratinocytes, which may be related to cell lineage characteristics, both revealed potent antioxidant activity. LPG and nanoLPG showed vasoprotective effects in aortic preparations. The gene expression assay indicates that, although no significant differences were observed in the expression of IL-10 and TNF-α , the PBMCs treated with nanoLPG showed a reduction in transcriptional levels of IFN-γ and an increased expression of COX-2 . Thus, the work adds evidence to the safety of the use of lycopene by humans and shows that tested formulations, mainly nanoLPG due to its stability, stand out as promising and biosafe products for the treatment of diseases that have oxidative stress and inflammation in their etiopathology.

Grounded in hedonic assumptions, evidence suggests that people tend to engage in activities they consider pleasurable and enjoyable, while trying to avoid pain and displeasure. This suggests that the dynamic between positive and negative affect can influence current behavior and the intentions to continue performing. Regarding resistance training (RT), research focusing on how to promote a better affective response is still scarce and much needed. Given existing limitations and theoretical suggestions, a RT program was developed and applied to recreational exercisers in a quasi-experimental design aiming to (1) explore the affective response dynamic through an assessment after the last set of each exercise; and (2) analyze possible differences of preference and tolerance profiles in affective variables (core affect and enjoyment). For that purpose, 43 participants (21 male and 22 female; M age  = 34.69 ± 6.71 years; M experience  = 8.32 ± 4.54 years; M BMI  = 24.26 ± 2.64 kg/m 2 ) accepted to participate in this study. Descriptive statistics, correlational, and group comparisons analyses were performed to provide evidence for proposed objectives. The present study showed that measures of affective valence/arousal applied immediately after a set represents a feasible and ecologically valid approach to tap core affect. Results presented evidence that recreationally trained exercisers in a common RT program would need a minimum of one measurement to assess the affective response. However, additional assessments could refine the understanding of exercise pleasurable experience. Results also suggest that exercisers with distinct profiles of preference/tolerance depicted differentiated patterns for the affective response, possibly justifying a distinct approach when promoting affective regulation., (© 2022 John Wiley & Sons A/S. Breast cancer represents the most incident cancer in women. Surgery, chemotherapy, radiation therapy, and hormone therapy remain the main treatment for this type of cancer. However, increasing resistance to anti-cancer drugs through poor response for some types of breast cancer to treatments highlights the need to develop new therapeutic agents to fight the disease. In this study, we evaluated the anti-tumor potential of the Chartergellus-CP1 peptide isolated from the wasp venom of Chartergellus communis in human breast cancer cell lines MCF-7 (HR+) and MDA-MB-231 (triple-negative). Cells viability, morphology, cell cycle dynamics, reactive oxygen species (ROS) production, and apoptosis were assessed for both cell lines after exposure to Chartergellus-CP1 during 24 and 48 h. The results showed that Chartergellus-CP1 led to a significant increase of cells in the S phase in addition to a high generation of ROS (being more evident in the MCF-7 cell line) associated with apoptotic cell death. Composite materials have gained significant attention owing to the synergistic effects of their constituent materials, thereby facilitating their utilization in new applications or in improving the existing ones. In this study, a composite based on nickel phthalocyanine (NiTsPc), zinc oxide nanoparticles (ZnONPs), and carbon nanotubes (CNT) was developed and subsequently immobilized on a pyrolytic graphite electrode (PGE). The PGE/NiTsPc-ZnONPs-CNT was identified as a selective catalytic hybrid system for detection of neurotransmitter dopamine (DA). The electrochemical and morphological characterizations were conducted using atomic force microscopy (AFM). Chronoamperometry and differential pulse voltammetry (DPV) were used to detect DA and detection limits of 24 nM and 7.0 nM was found, respectively. In addition, the effects of some possible DA interferents, such as ascorbic acid, uric acid, and serotonin, on DA response were evaluated. Their presence did not show significant variations in the DA electrochemical response. The high specificity and sensitivity of PGE/NiTsPc-ZnONPs-CNT for DA enabled its direct detection in human serum without sample pretreatment as well as in DA-enriched serum samples, whose recovery levels were close to 100%, thereby confirming the effectiveness of the proposed method. Aim: This study investigated the differences in frontoparietal EEG gamma coherence between expert meditators (EM) and naïve meditators (NM)., Material and Methods: This is a cross-sectional study with a sample of twenty-one healthy adults divided under two groups (experts meditators vs. naive-meditators), with analyzing the intra-hemispheric coherence of frontoparietal gamma oscillations by electroencephalography during the study steps: EEG resting-state 1, during the open presence meditation practice, and EEG resting-state 2., Results: The findings demonstrated greater frontoparietal EEG coherence in gamma for experts meditators in the Fp1-P3, F4-P4, F8-P4 electrode pairs during rest 1 and rest 2 (p ≤ 0.0083). In addition, we evidenced differences in the frontoparietal EEG coherence for expert meditators in F4-P4, F8-P4 during the meditation (p ≤ 0.0083)., Conclusion: Our results can support evidence that the connectivity of the right frontoparietal network acts as a biomarker of the enhanced Open monitoring meditation training., (Copyright © 2022 Elsevier Inc. Melanoma is a drug-resistant cancer, representing a serious challenge in cancer treatment. Dacarbazine (DTIC) is the standard drug in metastatic melanoma treatment, despite the poor results. Hyperthermia has been proven to potentiate chemotherapy. Hence, this work analyzed the combined action of hyperthermia and DTIC on A375 and MNT-1 cell lines. First, temperatures between 40 °C and 45 °C were tested. The effect of DTIC on cell viability was also investigated after exposures of 24, 48, and 72 h. Then, cells were exposed to 43 °C and to the respective DTIC IC10 or IC20 of each time exposure. Overall, hyperthermia reduced cell viability, however, 45 °C caused an excessive cell death (>90%). Combinational treatment revealed that hyperthermia potentiates DTIC’s effect, but it is dependent on the concentration and temperature used. Also, it has different mechanisms from the treatments alone, delaying A375 cells at the G2/M phase and MNT-1 cells at the S and G2/M phases. Intracellular reactive oxygen species (ROS) levels increased after treatment with hyperthermia, but the combined treatment showed no additional differences. Also, hyperthermia highly increased the number of A375 early apoptotic cells. These results suggest that combining hyperthermia and DTIC should be more explored to improve melanoma treatment.

Upconversion nanoparticles (UCNPs) have attracted considerable attention owing to their unique photophysical properties. Their utilization in biomedical applications depends on the understanding of their transformations under physiological conditions and their potential toxicity. In this study, NaYF 4 :Yb,Er UCNPs, widely used for luminescence and photophysical studies, were modified with a set of four different coordinatively bound surface ligands, i.e., citrate, alendronate (AA), ethylendiamine tetra(methylene phosphonate) (EDTMP), and poly(maleic anhydride-alt-1-octadecene) (PMAO), as well as silica coatings with two different thicknesses. Subsequently, the aging-induced release of fluoride ions in water and cell culture media and their cytotoxic profile to human keratinocytes were assessed in parallel to the cytotoxic evaluation of the ligands, sodium fluoride and the lanthanide ions. The cytotoxicity studies of UCNPs with different surface modifications demonstrated the good biocompatibility of EDTMP-UCNPs and PMAO-UCNPs, which is in line with the low amount of fluoride ions released from these samples. An efficient prevention of UCNP dissolution and release of cytotoxic ions, as well as low cytotoxicity was also observed for UCNPs with a sufficiently thick silica shell. Overall, our results provide new insights into the understanding of the contribution of surface chemistry to the stability, dissolution behavior, and cytotoxicity of UCNPs. Melanoma is the deadliest form of skin cancer, and its incidence has alarmingly increased in the last few decades, creating a need for novel treatment approaches. Thus, we evaluated the combinatorial effect of doxorubicin (DOX) and hyperthermia on A375 and MNT-1 human melanoma cell lines. Cells were treated with DOX for 24, 48, and 72 h and their viabilities were assessed. The effect of DOX IC10 and IC20 (combined at 43 °C for 30, 60, and 120 min) on cell viability was further analyzed. Interference on cell cycle dynamics, reactive oxygen species (ROS) production, and apoptosis upon treatment (with 30 min at 43 °C and DOX at the IC20 for 48 h) were analyzed by flow cytometry. Combined treatment significantly decreased cell viability, but not in all tested conditions, suggesting that the effect depends on the drug concentration and heat treatment duration. Combined treatment also mediated a G2/M phase arrest in both cell lines, as well as increasing ROS levels. Additionally, it induced early apoptosis in MNT-1 cells, while in A375 cells this effect was similar to the one caused by hyperthermia alone. These findings demonstrate that hyperthermia enhances DOX effect through cell cycle arrest, oxidative stress, and apoptotic cell death.

Magnetic nanoparticles (NP), such as magnetite, have been the subject of research for application in the biomedical field, especially in Magnetic Hyperthermia Therapy (MHT), a promising technique for cancer therapy. NP are often coated with different compounds such as natural or synthetic polymers to protect them from oxidation and enhance their colloidal electrostatic stability while maintaining their thermal efficiency. In this work, the synthesis and characterization of magnetite nanoparticles coated with fucoidan, a biopolymer with recognized biocompatibility and antitumoral activity, is reported. The potential application of NP in MHT was evaluated through the assessment of Specific Loss Power (SLP) under an electromagnetic field amplitude of 14.7 kA m -1 and at 276 kHz. For fucoidan-coated NP, it was obtained SLP values of 100 and 156 W/g, corresponding to an Intrinsic Loss Power (ILP) of 1.7 and 2.6 nHm 2 kg -1 , respectively. These values are, in general, higher than the ones reported in the literature for non-coated magnetite NP or coated with other polymers. Furthermore, in vitro assays showed that fucoidan and fucoidan-coated NP are biocompatible. The particle size (between ca. 6 to 12 nm), heating efficiency, and biocompatibility of fucoidan-coated magnetite NP meet the required criteria for MHT application.
