[EN] Biomass allowance is a key feature in pasture-based rabbit production systems. It conditions not only the stock density (rabbits/m²) and/or the number of grazing days, it also influences the grazing behaviour of animals. When herbage restriction occurs, pelleted feed and/or cereal intake goes up. Inadequate pasture management may also impair the biomass quantity and quality if overgrazing occurs. To avoid the undesirable effects of overgrazing and better manage pellet and cereal intake, information on both biomass availability and rabbits grazing capacity are needed. Here, we present an adaptation of the rising plate meter method (developed for biomass intake measures for ruminants) for use in rabbit. To this end, we designed an experiment where two groups of 12 rabbits each were kept in two different fields: under an apple orchard (AO) or on fallow land (FL). We followed the animals for 5 consecutive weeks (from 45 to 80 d old). Rabbits lived in 25 m² movable paddocks, and every week a new paddock location (called paddock-spot) was made available for them. At each new paddock-spot, we measured the herbage height inside the paddocks and performed samplings of the available biomass (i.e. herbage cut after herbage height measurement) outside the paddocks. From this data we estimated the available biomass inside each paddock-spot by fitting linear regression equations of biomass to herbage height. Overall, rabbits in the AO and FL had access to 1328±65.7 and 1386±58.6 kg of dry matter (DM) per ha, respectively. In every field and paddock-spot, the biomass available was lower than the rabbits grazing capacity; overgrazing was the rule. Roughly, and under a restricted herbage allowance, rabbits in the AO ingested 45.2 g DM/d and rabbits in the FL 43.4 g DM/d. In the last week (64 to 80 d old), the biomass intake of rabbits in the AO and AL represented 26.4 and 23.5% of the total DM intake, respectively. These values, however, does not represent the real grazing capacity of growing rabbits. Jerusalem artichoke is crop with the possibility of a wide using. The main nutrient of this plant is a non-starch polysaccharide inulin, which is considered like prebiotic. Young rabbits after weaning lose an immunity obtained from colostrum; are stressed by weaning; pre-moult; the permanent incisors are growing and a starch digestion by an enzyme amylase is not fully developed. The aim of this study was to evaluate the effect of the dietary supplement of dried Jerusalem artichoke tuber and tops (the sources of non-starch saccharide) on growth performance and health status of broiler rabbits after weaning in view of prices and efficiency of feeding mixtures. The test was realized through experiments with a different percentage of Jerusalem artichoke tuber (25%, 10%) and top (30%, 20%, 10%). Every experiment was composed of 2 groups – the control group without a supplement of Jerusalem artichoke and an experimental group with a different percentage of Jerusalem artichoke tuber and top. Experimental animals (1095 HY PLUS broiler rabbits) were weaned at 33 - 35 days of age and were fed by complete granulated feeding mixture ad libitum for the whole feeding period. [EN] Rabbit coccidiosis is a very serious disease caused by protozoan parasites of the genus Eimeria, which increases the production rate of free radicals, especially reactive oxygen species. When the generation of free radicals exceeds the scavenging capacity of the body s antioxidant system, the oxidant-antioxidant balance is broken, resulting in oxidative stress. This study was designed to investigate the effect on the oxidant-antioxidant status of rabbits infected with E. intestinalis and E. magna. To this end, eighteen 30-d-old weaned rabbits were randomly allocated into three groups as follows: the E. intestinalis infection group with 3×103 sporulated oocysts of E. intestinalis, the E. magna infection group with 20×103 sporulated oocysts of E. magna, and the uninfected control group. We measured the activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) and total antioxidant capacity (T-AOC) and the contents of malondialdehyde (MDA) in rabbits small intestinal tissues (duodenum, jejunum and ileum) of the three groupson day 8. The results showed that CAT activity and MDA levels significantly increased, while the activities of SOD, GSH-Px and T-AOC decreased after E. intestinalis and E. magna infection. Besides, the jejunum and ileum were particularly damaged in the rabbits. This study was conducted at the Poultry Research Centre, Faculty of Agriculture, Alexandria University, Egypt, during the summer season (July and August months) 2018. For five weeks, until 63 d of age, a total number of 140 weaned V line rabbits were randomly distributed into seven groups of 20 rabbits each. Rabbits of groups 2 and 3 were fed basal diet plus copper sulphate (100 and 200 mg Cu-Sul/kg, respectively). Groups 4 and 5 were fed basal diet plus copper methionine (100 and 200 mg Cu-Meth/kg, respectively) and groups 6 and 7 were fed basal diet plus copper glycine (100 and 200 mg Cu-Gly/kg, respectively), while the first group was fed basal diet only as control. [EN] Growth and development of hair follicles (HF) is a complex and dynamic process in most mammals. As HF growth and development regulate rabbit wool yield, exploring the role of genes involved in HF growth and development may be relevant. In this study, the coding sequence of the Angora rabbit (Oryctolagus cuniculus) WIF1 gene was cloned. The length of the coding region sequence was found to be 1140 bp, which encodes 379 amino acids. Bioinformatics analysis indicated that the WIF1 protein was unstable, hydrophilic and located in the extracellular region, contained a putative signal peptide and exhibited a high homology in different mammals. Moreover, WIF1 was significantly downregulated in the high wool production in the Angora rabbit group. Overexpression and knockdown studies revealed that WIF1 regulates HF growth and development-related genes and proteins, such as LEF1 and CCND1. WIF1 activated β-catenin/TCF transcriptional activity, promoted cell apoptosis and inhibited cellular proliferation. These results indicate that WIF1 might be important for HF development. [EN] This study was carried out to estimate general combining ability (GCA) and specific combining ability (SCA) of pre-and post-weaning traits from a partial diallel cross in three rabbit breeds: Local rabbit (L), Flemish Giant (F), and Rex (R). Body weight at 0 (BW0), 30 (BW30), 42 (BW42), and 63 (BW63) days of age, average daily gains from 0 to 30 d of age (ADG0-30), from 30 to 42 d of age (ADG30-42), from 42 to 63 d of age (ADG42-63), and from 30 to 63 d of age (ADG30-63), litter size at birth (LS0) and at weaning (LS42), and mortality at weaning (MR42) were studied in crossing LL, FF, RR, LF, LR, and FR. Local breed had the highest GCA for BW0, BW30, BW42, and average daily gain before weaning (ADG0-30, and ADG30-42) compared to Flemish Giant and Rex, while GCA of Local breed for average daily gain after weaning (ADG42-63), litter size (LS0, and LS42), and mortality (MR42) was higher than for Rex and similar to that of Flemish Giant. Crossing LF and LR showed higher SCA for BW30, BW42, BW63, ADG0-30, ADG42-63 and ADG30-63 than FR. In the present study, three experiments were designed to identify the most appropriate technique for freezing rabbit semen. Experiment 1 aimed to determine the optimal levels of dimethyl sulfoxide (DMSO) contents in freezing medium and their effects on individual bucks. Semen ejaculates for each buck (n=15 bucks) were mixed and split into three portions for extension with a freezing medium containing varying concentrations of DMSO (0.75, 1.0, and 1.4 M). Diluted semen samples were packaged in 0.25 mL straws and suspended above liquid nitrogen (LN) for 10 min, then dipped in LN. A few days after freezing, post-thaw semen evaluation was assessed, and according to the results, six bucks and an extender containing 0.75 M of DMSO were used for experiments 2 and 3. In experiment 2, the pooled semen from 6 bucks was divided into two portions for packaging in two straw sizes (0.25 and 0.50 mL). Each straw size was divided into five groups and suspended at different heights above LN (2, 4, 6, 8, and 10 cm) for 10 minutes before being preserved in LN. In experiment 3, the pooled semen was divided into four portions for dilution with freezing medium containing different concentrations of bovine serum albumin (BSA; 0, 2.5, 5.0, and 7.5 mg/mL). Semen samples were packaged in a 0.50 mL straw and suspended 10 min, 4 cm above LN for freezing. Pre-freezing and post-thawing, semen samples were evaluated for semen quality. Results showed that the extender containing 0.75 M DMSO had higher significant values for post-thaw sperm motility, longevity, acrosome integrity and sperm plasma membrane permeability. Bucks’ individuality had significant effects on post-thaw motility, acrosome and sperm plasma membrane integrity. A significant interaction was recorded between DMSO concentrations and bucks’ individuality on sperm longevity. Semen package sizes had no significant effects on the evaluated parameters. Semen was frozen at 2 and 4 cm above LN had significantly better post-thaw quality. BSA at concentrations 5 and 7.5 mg/mL improved recovery rates of acrosome integrity and sperm membrane permeability. DMSO 0.75 M and freezing 4 cm above LN seem to be more adequate for rabbit semen cryopreservation. The appropriate level of DMSO differs between bucks, as the post-thaw sperm longevity is affected. BSA enhanced acrosome and sperm membrane integrity. Results obtained will need further investigation to be confirmed in the field.

Biomedical research relies heavily on the use of model organisms to gain insight into human health and development. Traditionally, the mouse has been the favoured vertebrate model, due to its experimental and genetic tractability. Non-rodent embryological studies however highlight that many aspects of early mouse development, including the egg-cylinder topology of the embryo and its method of implantation, diverge from other mammals, thus complicating inferences about human development. To get a better understanding of rabbit development, we constructed a morphological and molecular atlas of rabbit development, which like the human embryo, develops as a flat-bilaminar disc. We report transcriptional and chromatin accessibility profiles of almost 180,000 single cells and high-resolution histology sections from embryos spanning gastrulation, implantation, amniogenesis, and early organogenesis. Using a novel computational pipeline, we compare the transcriptional landscape of rabbit and mouse at the scale of the entire organism, revealing that extra-embryonic tissues, as well as gut and primordial germ cell (PGC) cell types, are highly divergent between species. Focusing on these extra-embryonic tissues, which are highly accessible in the rabbit, we characterize the gene regulatory programs underlying trophoblast differentiation and identify novel signalling interactions involving the yolk sac mesothelium during haematopoiesis. Finally, we demonstrate how the combination of both rabbit and mouse atlases can be leveraged to extract new biological insights from sparse macaque and human data. The datasets and analysis pipelines reported here set a framework for a broader cross-species approach to decipher early mammalian development, and are readily adaptable to deploy single-cell comparative genomics more broadly across biomedical research. Due to the genetic tractability of mouse, we aimed to interrogate the role key transcription factors (TFs) such as Zic2/3, Mix-like 1(Mixl1), Eomesodermin (Eomes), Stat3, and Runx1 in early development. These key TFs are involved in a variety of roles, such as metabolism, as well as the fate decision of mesoderm, and blood development. Constructing an understanding of the stepwise role that these TFs play in sequence for developing blood and mesoderm allows for a better understanding of the consequences of genetic mutations. To perform this analysis, we generate a series of clustered regularly interspaced short palindromic repeats (CRISPR)-mediated knock out cell lines. Using a chimaera model system where knock-out (KO) cells are injected into wild-type (WT) host blastocysts, we can understand the cell-autonomous role that each TF plays. In conclusion, studying single-cell transcriptomics unveils the molecular profiles behind each cell type; however combining with spatial information, chromatin accessibility, and gene perturbations allows for further understanding of the signalling niche and regulatory elements behind cell type diversification. Augmenting this analysis by a variety of model organisms allowed for a nuanced understanding of eutherian development, such as macaque and human development by understanding the divergent and convergent features of embryonic development. This also allows for the optimisation of in vitro differentiation protocols in the future. Additionally, this has implications for biomedical research due to the species- and cell-type-specific responses to drug screens. A comprehensive understanding of each target cell type of interest allows for researchers to better adapt model systems to their intended target.

[ES] En esta tesis, se realizaron análisis genómicos, metagenómicos y metabolómicos en líneas de conejo seleccionadas de forma divergente para alta y baja VE del tamaño de la camada (TC). Estos animales mostraron diferencias en su potencial de resiliencia. Por ello, estas poblaciones divergentes son un excelente material biológico para estudiar la resiliencia animal a través de la VE. Se realizaron estudios de asociación del genoma (GWAS) utilizando la regresión de un solo marcador y la regresión bayesiana de múltiples marcadores. Cuatro regiones genómicas se asociaron con la VE en el cromosoma 3 de Oryctolagus cuniculus (OCU), OCU7, OCU10 y OCU14, explicando el 8,6% de la varianza genética total para la VE. Además, el estudio de huellas de selección (SS) identificó 134 regiones genómicas que podrían estar bajo selección para la VE. El solapamiento entre ambos estudios se identificó en el OCU3, donde también se encontraron mutaciones funcionales para los genes DOCK2, INSYN2B y FOXI1. Los genes candidatos de GWAS y SS fueron aquellos con mutaciones funcionales identificadas mediante el análisis de secuenciación del genoma completo (WGS) con pools de ADN. Los genes candidatos destacados mostraron funciones biológicas relacionadas con el desarrollo de estructuras sensoriales, la respuesta inmunitaria, la respuesta al estrés y el sistema nervioso. Todas ellas son funciones relevantes para modular la resiliencia de los animales. Por otra parte, los estudios metagenómicos y metabolómicos mostraron que la selección para la VE modificó el microbioma intestinal y la composición de su metaboloma. Las especies microbianas beneficiosas como Alistipes prutedinis, Alistipes shahii, Odoribacter splanchnicus y Limosilactobacillus fermentum eran más abundantes en la población resiliente. En cambio, las especies microbianas nocivas, como Acetatifactor muris y Eggerthella sp, fueron más abundantes en los animales no resistentes. Los genes relacionados con la formación de biofilms, el metabolismo de aminoácidos aromáticos (fenilalanina, triptófano y tirosina) y el metabolismo del glutamato también se expresaron de forma diferencial entre las poblaciones de conejos. Además, se identificaron 15 metabolitos intestinales como potenciales biomarcadores para discriminar y predecir adecuadamente entre las poblaciones de conejos resistentes y no resistentes. Este es el primer estudio que desvela importantes mecanismos biológicos de la resiliencia animal generada por la selección de la VE de TC. El genoma y el microbioma intestinal y la composición del metaboloma se modificaron a lo largo del proceso de selección, afectando a la respuesta inmunitaria y al estrés. Se encontraron resultados coincidentes entre los estudios metagenómicos y del metaboloma. Por otro lado, en esta tesis desarrollamos por primera vez una herramienta flexible para simular la coevolución del genoma y el microbioma a través de un proceso de selección. La clave de esta herramienta fue la implementación de la herencia del microbioma. In this thesis, genomic, metagenomic, and metabolomic analyses were performed on rabbit lines divergently selected for high and low VE of litter size (LS). These animals showed differences in their resilience potential. Thus, these divergent populations are an excellent biological material for studying animal resilience through the VE. Genome-wide association studies (GWAS) were performed using single marker regression, and Bayesian multiple marker regression approaches. Four genomic regions were associated with the VE in the Oryctolagus cuniculus chromosome (OCU) 3, OCU7, OCU10, and OCU14, explaining 8.6% of the total genetic variance for the VE. In addition, the signature of selection (SS) study identified 134 genomic regions which could be under selection for VE. Overlapping between both studies was placed in the OCU3, where functional mutations for the DOCK2, INSYN2B and FOXI1 genes were also found. Candidate genes from GWAS and SS were those with functional mutations identified using whole genome sequencing (WGS) analysis with pools of DNA. Highlighted candidate genes showed biological functions related to the development of sensory structures, the immune response, the stress response, and the nervous system. All of them are relevant functions to modulate animal resilience. On the other hand, metagenomic and metabolomic studies showed that the selection for VE modified the gut microbiome and metabolome composition. Beneficial microbial species such as Alistipes prutedinis, Alistipes shahii, Odoribacter splanchnicus and Limosilactobacillus fermentum were more abundant in the resilient population. In contrast, harmful microbial species such as Acetatifactor muris and Eggerthella sp were more abundant in the non-resilient animals. Genes related to biofilm formation, aromatic amino acid metabolism (Phenylalanine, tryptophan, and tyrosine), and glutamate metabolism were also differentially expressed between the rabbit populations. Furthermore, 15 gut metabolites were identified as potential biomarkers to properly discriminate and predict between the resilient and non-resilient rabbit populations. Five of them, the equol, 3-(4-hydroxyphenyl)lactate, 5-aminovalerate, N6-acetyllisine, and serine were microbial-derived metabolites. This is the first study unravelling important biological mechanisms under the animal resilience generated by VE of LS selection. Genome and gut microbiome and metabolome composition were modified throughout the selection process, affecting the immune and stress response. Overlapping results were found between the metagenomic and metabolome studies. On the other hand, in this thesis, we developed a flexible tool for simulating the coevolution of the genome and microbiome across a selection process for the first time. The key of this tool was the implementation of the microbiome inheritance. It is constructed in R and based on AlphaSimR so the user can modify the code and implement different scenarios. This thesis is the first step to develop future strategies and further research to improve animal resilience. A selection combining genomic and metagenomic information may improve the selection response. Moreover, gut-derived metabolites with evidence of crosstalk can be used as biomarkers to identify resilient animals by plasma, avoiding the extraction of faecal samples to determine the microbiome composition. If these studies suceed, these strategies could improve animal resilience with the aim of search a more sustainable livestock system. Histomorphological study confirmed that the carotid sinus was characterized by an abrupt thinning of elastic tunica media which was rapidly replaced by the normal muscularity of the media on the distal progression in the internal carotid artery. The carotid sinus was richly supplied with sensory receptors which took the form of menisci and left the sinus as the nerve of Hering.

The morphology of the intramural nervous apparatus of the rabbit esophagus has been studied by neurohistological methods. It was found that in the wall of the esophagus, the submucosal and intermuscular nerve plexuses are well distinguished, which consist of nerve nodes located at the intersection of bundles of nerve fibers. The nodes of the submucosal nerve plexus are located less frequently, and the nodes of the intermuscular plexus are smaller in size, but located more densely. Intermuscular plexus nodes contain more long-axon neurocytes, which are impregnated more intensively than other neurons in the node.

Rabbit is an induced ovulatory species, so ovulation takes place after mating. Traditionally, exogenous and synthetic hormonal factors (administered by intramuscular and intravaginal route) such as GnRH and analogues, or different physical procedures (i.e. stimulation by intravaginal cannula) have been used to induce ovulation in females when artificial insemination is applied in rabbit farms. Restriction and public rejection of the use of hormones is leading to the study of the seminal plasma components with potential action on ovulation induction. The aim of the present review is to collect and summarise the strategies used in recent years to trigger ovulation and improve rabbit fertility management with respect to more animal-friendly manipulation methods. Furthermore, special attention has been paid to the use of a semen component (as endogen molecule) such as beta nerve growth factor (β-NGF) in male and female rabbit reproductive physiology. This neurotrophin and its receptors (TrKA and p75NTR) are abundantly distributed in both male and female rabbit reproductive tracts, and it seems to have an important physiological role in sperm maturation and behaviour (velocity, apoptosis and capacitation), as well as a modulatory factor of ovulation. Endogen β-NGF is diluted in the seminal doses with the extenders; hence it could be considered an innovative and alternative strategy to avoid the current exogenous (by intramuscular route) and stressful hormonal treatments used in ovulation induction. Their addition in seminal dose could be more physiological and improve animal welfare in rabbit farms.

There are several studies dealing with the phenotypic variance of the vertebral number in the spinal column of rabbits. According to the literature the number of thoracic and lumbar vertebrae varies between 11-13 and 6-8, respectively. The length of the m. longissimus dorsi (MLD) - a valuable meat part of rabbits - is determined by the length of the vertebral column therefore the number of vertebrae may have economic importance in breeding. The aim of this study was to create an automatic counter using computed tomography (CT) images. In the first step, a skeleton binary mask was created using the radiodensity range between 120 and 3071 HU, then the lumbar and thoracic regions were processed by two different methods. The lumbar part was evaluated based on the frequency of the bone voxels along the axial plane. The number of thoracic vertebrae was determined from the number of ribs. The left and right ribs were processed separately. The developed method was tested on CT examination of 40 Hycole rabbits compared to manual evaluation. The results of the automatic algorithm had few errors: in one case in the lumbar region (2.5%) and in 3 cases in the thoracic region (5%). The automated evaluation process takes a few seconds per individual and then the program visualizes the results on a graph. The incorrectly evaluated rabbits are recognizable on graphs and they can be easily corrected with a minimal time investment.

Due to the high prevalence of opioid prescription following orthopedic procedures, there is a growing need to establish an animal model system to evaluate the effects of opioids on bone remodeling. Rabbits have been employed as model organisms in orthopedic research as they exhibit well-defined cortical bone remodeling similar to humans. Existing research in rabbits has been limited to modes of opioid administration that are short-acting and require repeated application. Here, we present data from a proof-of-principle longitudinal study employing two opioid analgesic administration routes (subcutaneous injection and transdermal patch) to evaluate the efficacy of studying chronic opioid exposure in a rabbit model. Skeletally mature male New Zealand White rabbits (Oryctolagus cuniculus) were divided into three groups of seven animals: morphine, fentanyl, and control. Experimental treatments were conducted for eight weeks. Preparation of the skin at the fentanyl patch site and subsequent patch removal presented experimental difficulties including consistent skin erythema. Though noninvasive, the patches further caused acute stress in fentanyl animals. We conclude that though transdermal fentanyl patches may be preferred in an acute clinical setting, this method is not feasible as a means of long-term pain relief or opioid delivery in a laboratory context.

BACKGROUND/AIM This study investigated the effect of vascular endothelial growth factor (VEGF) inhibitor bevacuzimab (BVZ) on the rabbit basilar artery using an experimental subarachnoid hemorrhage (SAH) model. MATERIALS AND METHODS Eighteen adult male New-Zealand white rabbits were randomly divided into three groups: a control group (n=6), SAH group (n=6), and SAH+BVZ group (n=6). Experimental SAH was created by injecting autologous arterial blood into the cisterna magna. In the treatment group, the subjects were administered a daily dose of 10 mg/kg, intravenous BVZ 2 days after the SAH. Basilar artery diameters were measured with magnetic resonance angiography (MRA) 72 hours after the SAH in all groups. After 72 hours the animals? whole brains, including the upper cervical region, were obtained from all the animals after perfusion and fixation of the animal. The wall thickness, luminal area, and the apoptosis at the basilar arteries were evaluated in all groups. RESULTS BVZ significantly prevented SAH induced vasospasm confirmed in-vivo with MRA imaging with additional suppression of apoptosis on basilar artery wall. CONCLUSION VEGF inhibition with BVZ has shown to have a vasospasm and apoptosis attenuating effect on basilar artery in a SAH model.

Objective:Obesity is a global health epidemic with considerable co-morbidities. The increasing demand for bariatric surgery has led to the emergence of new techniques. We modified previously described Mini Gastric By-pass(MGB) technique via leaving a bridge at the most cranial 2 cm of the fundus of the human stomach to the follow-up and treatment of the remnant stomach and duodenum. We would like to entitle this new technique as Bridged MGB and aimed to apply on rabbits as an experimental study.Material and Methods:The study was performed in the experimental animal laboratory of university after ethical approval was taken from the local ethics committee. Described new technique was applied to 2.1 and 3.2 kg 2 New Zealand rabbits.Results:As a result of the operations, one of the rabbits died on the day of the operation; the other rabbit was exitus postoperatively on the third day. In autopsies, although no problem was detected at the anastomoses, necrosis was detected in the large curvature of both rabbits.Conclusion:Rabbit, one of the popular experimental animals, has been shown to be different from the human gastrointestinal system in both arterial and topographic aspects and it has been emphasized that it varies according to the species and even the diet and the climate. We believe that our study failed as a result of these differences and that animals more similar to humans should be used in gastrointestinal experimental studies

Quantitative understanding of pharmacokinetics of topically applied ocular drugs requires more research to further understanding and to eventually allow predictive in silico models to be developed. To this end, a topical cocktail of betaxolol, timolol and atenolol was instilled on albino rabbit eyes. Tear fluid, corneal epithelium, corneal stroma with endothelium, bulbar conjunctiva, anterior sclera, iris-ciliary body, lens and vitreous samples were collected and analysed using LC-MS/MS. Iris-ciliary body was also analysed after intracameral cocktail injection. Non-compartmental analysis was utilized to estimate the pharmacokinetics parameters. The most lipophilic drug, betaxolol, presented the highest exposure in all tissues except for tear fluid after topical administration, followed by timolol and atenolol. For all drugs, iris-ciliary body concentrations were higher than that of the aqueous humor. After topical instillation the most hydrophilic drug, atenolol, had 3.7 times higher AUCiris-ciliary body than AUCaqueous humor, whereas the difference was 1.4 and 1.6 times for timolol and betaxolol, respectively. This suggests that the non-corneal route (conjunctival-scleral) was dominating the absorption of atenolol, while the corneal route was more important for timolol and betaxolol. The presented data increase understanding of ocular pharmacokinetics of a cocktail of drugs and provide data that can be used for quantitative modeling and simulation.

Carbohydrate is commonly consumed macronutrientand act as a potential regulatorin modulating the intestinal microbiota in humans and animals, which in turn might have consequences on the intestinal health. However, the effects of dietary non-fibrous carbohydrate (NFC) to neutral detergent fiber (NDF) in different ratios on intestinal microbiota, homeostasis and immunity have not been fully explored. A total of 135 healthy weaned rabbits with average body weight of (1.082±0.073) kg were randomly divided into 5 groups. Under the condition of the same other nutrient levels, rabbits were fed diets with NFC/NDF of 0.7 (T1), 1.0 (T2), 1.3 (T3), 1.6 (T4), and 1.9 (T5). Osteoarthritis (OA) is the most common form of arthritis and a major source of pain and disability in the adult population. There is a significant unmet medical need for the development of effective pharmacological therapies for the treatment of OA. In addition to spontaneously occurring animal models of OA, many experimental animal models have been developed to provide insights into mechanisms of pathogenesis and progression. Many of these animal models are also being used in the drug development pipeline. Here, we provide an overview of commonly used and emerging preclinical small animal models of OA and highlight the strengths and limitations of small animal models in the context of translational drug development. There is limited information in the published literature regarding the technical reliability of these small animal models and their ability to accurately predict clinical drug development outcomes. The cost and complexity of the available models however is an important consideration for pharmaceutical companies, biotechnology startups, and contract research organizations wishing to incorporate preclinical models in target validation, discovery, and development pipelines. Further considerations relevant to industry include timelines, methods of induction, the key issue of reproducibility, and appropriate outcome measures needed to objectively assess outcomes of experimental therapeutics. Preclinical small animal models are indispensable tools that will shine some light on the pathogenesis of OA and its molecular endotypes in the context of drug development. The effects of lagomorph grazing on the Orthoptera of a small hill in Mistley (southeast England) were studied during the summer of 2020. Transect counts of Orthoptera revealed low sward height with abundant bare earth due to high wild rabbit Oryctolagus cuniculus grazing on the high slopes. This intensive grazing led to only field grasshopper Chorthippus brunneus (Thunberg, 1815) adults being found in any number on the high slopes, perhaps utilizing the short swards and bare earth as basking and egg-laying habitat. Aspect was also important, with significantly more grasshopper nymphs and C. brunneus adults on the south-facing slope than on the northern slope. Soil slippage areas seem like valuable micro-habitats on the south-facing slope, with these ‘sun traps’ providing excellent basking habitat for nymphs and C. brunneus. This study confirms that lagomorph grazing alters hill summit habitats for Orthoptera, benefiting C. brunneus and, to a lesser extent, the meadow grasshopper Pseudochorthippus parallelus (Zetterstedt, 1821). However, overgrazing of higher hill slopes can exclude tall grass species, such as long-winged conehead Conocephalus fuscus (Fabricius, 1793), and reduce assemblage diversity.

The objectives of this study were to elucidate the effect of different high-concentration levels of inert gases (Ar and N2) on heme-pigment solutions and on the color of the blood, meat, and small intestine of rabbits; and to figure out the proper concentration level of inert gas (Ar or N2) for the complete stunning of the rabbit. To observe the changing of color attributes, a research study was conducted in the abattoir of the National Institute of Animal Science (NIAS), RDA, Republic of Korea. This experiment had 2 parts, (i) A trial on heme-pigment solutions (hemoglobin and myoglobin solution) was conducted in a gas chamber with different high-concentration levels of carbon dioxide, argon, nitrogen, and normal air; and (ii) a rabbit experiment was conducted—stunning with different high-concentration levels of carbon dioxide, argon, nitrogen, and the Halal method (non-stunning). A small-size digital gas chamber was used for this experiment (size: width 850 mm × depth 1350 mm). Artificial hemoglobin (Hb) and myoglobin (Mb) solutions were created from “porcine hemoglobin lyophilized powder” and “equine skeletal muscle myoglobin lyophilized powder”, respectively. In the heme-pigment solutions trial, 10 treatments were used: (i) 80% carbon dioxide (T1), (ii) 85% carbon dioxide (T2), (iii) 90% carbon dioxide (T3), (iv) 80% argon (T4), (v) 85% argon (T5), (vi) 90% argon (T6), (vii) 80% nitogen (T7), (viii) 85% nitogen (T8), (ix) 90% nitogen (T9), and (x) normal air (T10). Heme-pigment solutions (both Hb and Mb) were exposed with each treatment for four separate durations of time (30 s, 1 min, 2 min, and 4 min); and every sample (Hb and Mb) was exposed during each duration of time for 10 times (n = 10). In the rabbit experiment, seven treatments were used (i) 85% carbon dioxide (T1), (ii) 90% carbon dioxide (T2), (iii) 85% argon (T3), (iv) 90% argon (T4), (v) 96% nitrogen (T5), (vi) 98% nitogen (T6), and (vii) the Halal method (non-stunning) (T7). Forty-two rabbits (mixed-breed) were collected from the nearest commercial farm and randomly selected for a treatment group (n = 6). The average body weight was 2.3 kg. For stunning, each individual rabbit was separately kept in a gas chamber, after which each specific gas was used to fulfill the desired level in the pit. After reaching the desired level of concentration, rabbits were stunned within a very short time. It was observed that the time required for stunning in the T1, T2, T3, T4, T5 and T6 treatment was 79–82, 68–73, 97–103, 88–91, 164–173, and 108–115 s, respectively. In the case of the Halal method (non-stunning), as per the rules of Islam, rabbits were slaughtered without stunning. After slaughtering, in all treatments, the blood, meat, and small intestine of each animal were collected carefully and kept in a cool room in which the temperature was −2 °C, and after 24 h, the color attributes—lightness (L*), redness (a*), and yellowness (b*)—were measured via a Chroma Meter. It was found that in both experiments (trial on heme-pigment solutions and rabbit stunning), the color values (L*, a* and b*) showed a significant difference (p < 0.05) among the treatment groups. The CO2-treated groups showed high redness (a*) and low lightness (L*) and yellowness (b*), which indicated a dark-red color, and N2-treated groups showed high lightness (L*) and yellowness (b*) and low redness (a*), which implied a bright-red color. The effect of the Ar was in between the CO2- and N2-treated groups. These phenomena were observed both in the heme-pigment solutions (Hb and Mb) and in the blood, meat, and small intestine of the rabbits. N2-stunned animals fulfill the fundamental desire of consumers to purchase bright red-colored fresh meat. Therefore, in view of color attributes, consumer satisfaction, and animal welfare, N2 gas can be thought of as a valuable alternative to stunning. Considering the time required for complete stunning and desirable color attributes, a 98% concentration of N2 is best for rabbit stunning. As such, it could be used as a better option for the gas stunning of animals.

Background Limb fractures represent the most common orthopaedic disease in pet rabbits. However, only a few studies have evaluated therapeutic details of limb fractures. There are no data available for long‐term outcomes of limb fracture treatment. Methods The medical records of six institutions were reviewed retrospectively to identify cases of traumatic limb bone fractures in pet rabbits between 1999 and 2020. The medical records (n = 387) were analysed for details of fracture prevalence, aetiology, therapy protocols, treatment complications, outcome and long‐term effects. In addition to the retrospective data evaluation, 13 rabbits were re‐evaluated in person in recent clinical analyses, including orthopaedic examination, radiography and computed‐tomographic imaging. Details of long‐term effects of fracture treatment were requested over the telephone for a further 232 animals using a standardised questionnaire. Results Long bone fractures accounted for the majority of all fractures (296/387; 76.5%). Hindlimb fractures (301/387; 77.7%) were more common than forelimb fractures (86/387; 22.2%), and tibial fractures and combined fractures of the tibia and fibula (119/387; 30.8%) were observed most frequently. Most fracture treatments were based on osteosynthesis procedures (243/328; 74.1%). Treatment complications occurred in 130 out of 328 (39.6%) cases. A high bodyweight (p = 0.047) and an older age (p = 0.01) were found to be significant risk factors for the emergence of therapy complications. Overall, 75.4% of animals (175/232) had a satisfactory long‐term outcome. Limb posture anomalies were evaluated in 61 cases (26.3%). Limitations The multi‐centre approach led to the inclusion of various institutions, veterinarians, treatment protocols and rabbit populations that might have influenced the results. The medical records were reviewed retrospectively, so there were some data that were lacking or could not be collected in a standardised manner. Furthermore, rabbit owners' evaluation of long‐term outcomes might be prone to error, despite the use of a standardised interview questionnaire. Conclusion Limb fractures are a common orthopaedic issue in pet rabbits. The patient's bodyweight and age are significant risk factors for the emergence of complications during the fracture treatment process. Long‐term orthopaedic effects, such as abnormal limb posture and permanent lameness of the affected limb, were observed regularly.

Four male neutered continental giant rabbits aged between 10 and 30 months were presented with femoral condylar fractures, which developed without an observed traumatic injury. Stabilisation of the condylar fracture was achieved with screw fixation in all cases, which was supplemented with additional fixation in three cases. Complications consequent to the surgical intervention occurred in two cases: a femoral fracture and loss of fixation. Three rabbits were reported to have recovered normal limb function, and the rabbit that developed a femoral fracture as a consequence of its surgical intervention was treated with amputation.

Objectives In veterinary settings, high exposures to animal allergens and microbial agents can be expected. However, occupational exposure levels are largely unknown. The objective of this study was to estimate the allergen, endotoxin, and β-(1,3)-glucan concentrations in small animal practices and in the homes of practice employees. Methods Dust samples were collected using electrostatic dust fall collectors in diverse rooms of 36 small animal practices, as well as in employees’ homes. Major animal allergens (Fel d 1, Can f 1, Ory c 3, Cav p 1, Equ c 1, Bos d 2), domestic mite (DM) allergens, and β-(1,3)-glucan levels were measured using enzyme immunoassays. Endotoxin was determined using the Limulus amoebocyte lysate assay. Influences on exposure levels were analyzed using multilevel models. Results The levels of Can f 1, Fel d 1, Ory c 3, and Cav p 1 were up to 30 times higher in practices compared with homes without animals, but significantly lower compared with the homes with the respective pet. Although horses were not treated in the practices, Equ c 1 was found in 87.5% of samples, with the highest concentrations measured in changing rooms. DM levels were significantly lower in practices than in all private homes, and endotoxin levels were similar to those in homes with pets. In the practice itself, exposure levels were significantly influenced by animal presence, type of the room, and area per employee; whereas, room volume and diverse cleaning measures had mostly no effect. Conclusions Exposure to animal allergens is high in veterinary practices, but it does not reach levels of households with pets. Domestic mite allergen and endotoxin exposure seem to be low for workers in veterinary practices. The high Equ c 1 detection rate strongly indicates dispersal of allergens, most likely through clothing and hair.

Fatty acids of intramuscular fat (IMF) in rabbits can influence meat quality, but it is unclear which fatty acids benefit to human health. A rabbit model of weight gain and weight loss was constructed using two rabbit groups and two growth stages. Stage 1 included control group1 fed a commercial diet(CG1) and experimental group1 fed a high fat diet (EG1). Stage 2 include control group2(CG2) and experimental group2 (EG2) both fed a restricted commercial diet. We detected differences in blood biochemical indicators as well as changes in intramuscular adipose cells and intramuscular fatty acid content in control and experiment groups at two stages. High fat induction can make rabbits become obese, have higher concentrations of glucose (GLU), total cholesterol (TC), triglyceride (TG), low density lipoprotein‐cholesterol (LDL‐C) and free fatty acid (FFA), and lower concentrations of insulin (INS). In addition, a high‐fat diet promotes hypertrophy of precursor adipocytes in femoral muscles. Conversely, a restricted diet causes weight loss, decreases the concentration of TG, FFA, and INS in CG2 and EG2, and increases the deposition of unsaturated fatty acids in the femoral muscle. The content of monounsaturated trans oleic acid (C18:1n‐9T) in EG2 was significantly higher than in CG2, whereas oleic acid (C18:1n‐9C) was significantly lower in EG2 than in CG2. The polyunsaturated fatty acids Linolenate (C18:3 n‐3) and cis‐5,8,11,14,17‐Eicosapentaenoate (C20:5 n‐3) increased in CG2 and EG2. The content of Linoleate (C18:2 n‐6) and γ‐Linolenic acid (C18:3 n‐6) significantly increased in CG2. The content of cis‐11,14‐Eicosatrienoic acid (C20:2) decreased significantly in CG2, but increased significantly in EG2.Thus, a high‐fat diet can increase the formation of unhealthy fatty acids. Conversely, weight loss due to a restricted diet leads to an increase in unsaturated fatty acids in the femoral muscle, indicating that it reduces obesity symptoms and it may improve meat quality in rabbit., To find the phenotypic differences of fatty acids in leg muscle of rabbits, the study use the leg muscles of obese rabbits with induced by high fat and normal rabbits to carry out weight loss test, respectively. Thus. it can make us to preliminarily understand the changes of fatty acids in leg muscles of rabbits after 70 days of age and provide a good basis for imporving meat quality of rabbits in the future.

Sheep, cows, cats, and rabbits are kept by humans for agricultural purposes and as companion animals. Much of the mammary research in these species has focussed on mastitis in the case of ruminants and rabbits, and mammary tumourigenesis in cats and rabbits. However, similarities with the human breast suggest that these species may be currently underutilised as valuable comparative models of breast development and disease. The mammary gland undergoes cyclical postnatal development that will be considered here in the context of these non-traditional model species, with a focus on the mammary microenvironment at different postnatal developmental stages. The second part of this review will consider mammary tumour development. Ruminants are thought to be relatively 'resistant' to mammary tumourigenesis, likely due to multiple factors including functional properties of ruminant mammary stem/progenitor cells, diet, and/or the fact that production animals undergo a first parity soon after puberty. By contrast, unneutered female cats and rabbits have a propensity to develop mammary neoplasms, and subsets of these may constitute valuable comparative models of breast cancer.

Fermented cassava peel with takakura method can increase the protein content of feed for improving the quality of feed. This study aims to determine the effect of fermented cassava peel fermented with takakura method in feed on the growth of weaning New Zealand white rabbit. The experiment was conducted in the Laboratory of Animal Biology, Faculty of Agriculture, University of Sumatera Utara. The research carried out for 2 months started from September to November 2012. The research design was used in this study was completely randomized design (CRD) with 4 treatments and 5 replications. The research has been conducted to find the clinical efficacy and safety of preanaesthetics and induction agents based on clinical and serum biochemical indicators in rabbits. Eight healthy rabbits (3.0-3.5kg) of either sex were arbitrarily allocated into two groups: Group DK (diazepam-ketamine) and Group DFK (diazepam-fentanyl-ketamine). Rabbits were received intramuscular diazepam (5mg/kg), fentanyl (0.02 mg/kg) and ketamine (35 mg/kg). Data on clinical parameters (rectal temperature, heart rate, and respiratory rate) and reflexes (righting reflex, palpebral reflex, and pedal reflex) were evaluated before and at 10, 20, 30, 40, 50, 60, 70, 80, 90, and 100 minutes after injection of anaesthetics. Blood samples were aspirated before anaesthesia and 30 minutes following induction. Serum Albumin, Globulin, Cholesterol, Triglyceride, Total Protein, High-Density Lipoprotein (HDL), Low-Density Lipoprotein (LDL), Creatinine, Calcium, Phosphorus, Sodium, Potassium, and Chlorine were evaluated through a semiautomatic biochemistry analyzer. In this study, no significant changes in rectal temperature were observed in the animals of group DK during anaesthesia. During anaesthetic cascade, heart rates and respiration rates in both the groups were reduced significantly (P< 0.05). In DFK-injected rabbits, the return of righting and palpebral reflexes were delayed. The DFK groups had considerably (P< 0.05) longer surgical anaesthesia than the DK groups. In terms of serum biochemistry, DK-treated rabbits had significantly lower total protein, globulin, creatinine, HDL, sodium, and potassium concentrations (P< 0.05). In Europe, the most appropriate strategy to replace soybean meal (SBM) in animal feed has been the development of diets containing locally produced protein sources. One of these sources is lupine (Lupinus spp.). The effect of the total substitution of SBM by white lupine (WL) and yellow lupine (YL) seeds in the diets of growing rabbits and of gender on meat quality and the fatty acids (FA) profile were evaluated. Sixty hybrid weaned rabbits (New Zealand × Californian) (20 rabbits per diet), were fed diets that contained 150 g/kg of SBM (SBMD) and WL (WLD) or YL (YLD) for 35 to 69 days. At the end of this period, 30 rabbits (10 rabbits per diet) were slaughtered to evaluate the carcass and meat characteristics and the FA profile of the longissimus dorsi (LD) muscle. In general, the carcass and meat characteristics (pH and colour) were not affected (p > 0.05) by diet or gender. Further, there was no observed effect (p > 0.05) of gender on meat FA and on the calculated indexes related to human health. However, diet had an effect (p < 0.05) on the FA profile, FA categories, and calculated indexes related to human health. The meat from rabbits fed SBMD presented higher (p < 0.05) saturated FA (SFA; 44 vs. 39 g/100 g average on lupine diets) and lower (p < 0.05) polyunsaturated FA (PUFA; 24 vs. 28 g/100 g average on lupine diets). Our results showed that SBM may be completely replaced by WL or YL, improving the quality of LD muscle FA in terms of nutritional quality for humans.

CRISPR-based gene drives offer novel solutions for controlling invasive alien species, which could ultimately extend eradication efforts to continental scales. Gene drives for suppressing invasive alien vertebrates are now under development. Using a landscape-scale individual-based model, we present the first estimates of times to eradication for long-lived alien mammals. We show that demography and life-history traits interact to determine the scalability of gene drives for vertebrate pest eradication. Notably, optimism around eradicating smaller-bodied pests (rodents and rabbits) with gene-drive technologies does not easily translate into eradication of larger-bodied alien species (cats and foxes).

Cannabis use is associated with known cardiovascular side effects such as cardiac arrhythmias or even sudden cardiac death. The mechanisms behind these adverse effects are unknown. The aim of the present work was to study the cellular cardiac electrophysiological effects of cannabidiol (CBD) on action potentials and several transmembrane potassium currents, such as the rapid (IKr) and slow (IKs) delayed rectifier, the transient outward (Ito) and inward rectifier (IK1) potassium currents in rabbit and dog cardiac preparations. CBD increased action potential duration (APD) significantly in both rabbit (from 211.7 ± 11.2. to 224.6 ± 11.4 ms, n = 8) and dog (from 215.2 ± 9.0 to 231.7 ± 4.7 ms, n = 6) ventricular papillary muscle at 5 µM concentration. CBD decreased IKr, IKs and Ito (only in dog) significantly with corresponding estimated EC50 values of 4.9, 3.1 and 5 µM, respectively, without changing IK1. Although the EC50 value of CBD was found to be higher than literary Cmax values after CBD smoking and oral intake, our results raise the possibility that potassium channel inhibition by lengthening cardiac repolarization might have a role in the possible proarrhythmic side effects of cannabinoids in situations where CBD metabolism and/or the repolarization reserve is impaired.

The chemical composition and hardness of bovine bone, porcine bone, chicken bone, and rabbit bone were compared, as well as the influence of nanoprocessing on the physicochemical characteristics of these bone powders. A series of nanofabrication processes led to an increase in bone minerals and the loss of protein and fat. The hardness of softened bovine bone was still the largest, whereas chicken and rabbit bones were relatively soft. There were no significant differences in the functional groups between nanoscale bone powders. Overall, nanomachining significantly reduced and homogenized the bone particle size and improved the color and release rate of calcium ions of bone powders at the same time; these effects were different for several bones. Nanoscale rabbit bone had higher comminution efficiency, as well as satisfactory nutritional value, color, and product yield, which supports its strong development potential., This study mainly described the application of nanoprocessing in common mammalian bones (bovine and porcine), common poultry bone (chicken), and new material rabbit bone. The effects of nanoprocessing on the physicochemical properties of rabbit bone and other three common bones were compared. The excellent properties of nanoscaled rabbit bone (NRB), which support its great development potential.

In an obstructive sleep apnea rabbit model, heliox never abolishes tidal inspiratory flow limitation (IFL), but increases inspiratory flow and tidal volume, substantially in some and nearly nil in other animals. Positive response to heliox cannot be predicted on the basis of breathing pattern characteristics or upper airway resistance that preceded IFL onset, but is related to the mechanical and geometrical features of upper airway collapsible segment, as indicated by model simulation.

This study was performed to identify the effect of synovium graft on conjunctiva in rabbits after dry eye induction. Six New Zealand White rabbits were used as dry eye models. Both eyes were divided to two groups as control and synovium graft group. The synovium graft was performed in fourth week after dry eye modeling. Quantitative change of tears through Schirmer tear test (STT), qualitative change of tear film through tear film break up time (TFBUT), and damage of cornea through fluorescein staining were observed for 10 weeks at intervals of two weeks. Histological examination was performed to evaluate cornea and conjunctiva at tenth week. In both groups, STT and TFBUT were significantly decreased in 4 weeks after modeling compared to 0 weeks (p < 0.05) . After synovium graft, there were increases in STT value at 4 weeks and TFBUT at 4 and 6 weeks in graft group (p < 0.05). Corneal fluorescein staining showed no significant difference between the two groups. In histopathological examination, grafted synovium was detected as round to ovoid ingression folds, well attached to grafted regions with 0.11 ± 0.04 mm2 (range, 0.05- 0.16 mm2). These results indicated that the synovium graft on the conjunctiva had an effect on the qualitative and quantitative improvement of the tear film even though there was no histological change.

The purpose of the study was to determine the phenotypic correlations between meat quality traits in Popielno White rabbits. The experimental material consisted of 93 Popielno White rabbits (49♀; 44♂). From weaning at 35 days of age until 84 days of age, the animals were housed in a battery system and fed ad libitum with a complete feed containing 10,2 MJ of metabolic energy, 16,5% total protein and 14% crude fibre. The rabbits were slaughtered at 84 days of age with a body weight of about 2,6 kg. The research material was the meat of longissimus lumborum muscle. Meat quality characteristics, i.e. meat acidity, meat colour, shear force, meat texture and cooking loss, were examined. The results were analysed using the SAS package (2014). Many statistically significant relationships were found between the meat quality characteristics of Popielno White rabbits. The results indicate that potential selection towards a change in pH value may adversely affect the meat texture of Popielno White rabbits. Selection to improve the colour of the meat of this breed may have a negative effect on its shear force, hardness and cooking loss. Selection to improve shear force will improve the other texture profile parameters. An improvement in the meat texture parameters of Popielno White rabbits will lead to an improvement in the cooking loss value. Dependencies among meat quality traits in Popielno White rabbits are high enough to be useful in selecting an appropriate breeding method.

Bone graft techniques are used to compensate for bone loss in areas with deficient regeneration. However, matrix metalloproteases (MMPs) can limit bone formation by degrading extracellular matrices, which are required for bone regrowth. Noteworthily, rutin is a natural flavonoid compound that inhibits the genetic expression of various MMPs. Therefore, rutin may serve as an inexpensive and stable alternative to the growth factors used to accelerate dental bone graft healing. This study aimed to evaluate the potential of mixing rutin gel with allograft bone to accelerate the healing of bone defects in an in vivo rabbit model. Bone defects were surgically induced in New Zealand rabbits (n = 3 per group) and subsequently treated with bone grafts along with rutin or control gel. Overall, treatment with rutin significantly prevented the expression of several MMPs and increased type III collagen in the gingiva around the surgical site. Additionally, rutin-treated animals showed enhanced bone formation with higher bone marrow content in the jawbone defect area compared with the control group. Taken together, these findings demonstrate that rutin gel, when added to bone grafts, quickly enhances bone formation and may serve as a suitable alternative to expensive growth factors for the same purpose.

Hyaline cartilage has very limited repair capability and cannot be rebuilt predictably using conventional treatments. This study presents Autologous Chondrocyte Implantation (ACI) on two different scaffolds for the treatment of lesions in hyaline cartilage in rabbits. The first one is a commercially available scaffold (Chondro–Gide) made of collagen type I/III and the second one is a polyethersulfone (PES) synthetic membrane, manufactured by phase inversion. The revolutionary idea in the present study is the fact that we used PES membranes, which have unique features and benefits that are desirable for the 3D cultivation of chondrocytes. Sixty-four White New Zealand rabbits were used in this research. Defects penetrating into the subchondral bone were filled with or without the placement of chondrocytes on collagen or PES membranes after two weeks of culture. The expression of the gene encoding type II procollagen, a molecular marker of chondrocytes, was evaluated. Elemental analysis was performed to estimate the weight of tissue grown on the PES membrane. The reparative tissue was analyzed macroscopically and histologically after surgery at 12, 25, and 52 weeks. RT-PCR analysis of the mRNA isolated from cells detached from the polysulphonic membrane revealed the expression of type II procollagen. The elementary analysis of polysulphonic membrane slices after 2 weeks of culture with chondrocytes revealed a concentration of 0.23 mg of tissue on one part of the membrane. Macroscopic and microscopic evaluation indicated that the quality of regenerated tissue was similar after the transplantation of cells placed on polysulphonic or collagen membranes. The established method for the culture and transplantation of chondrocytes placed on polysulphonic membranes resulted in the growth of the regenerated tissue, revealing the morphology of hyaline-like cartilage to be of similar quality to collagen membranes.

Three diets were formulated, i.e., soybean meal (SM diet), adult Acheta domesticus (AD diet), and Tenebrio molitor larvae (TM diet), as the main crude protein (CP) sources. A total of 45 rabbits (Hyplus, weaned at 32 days of age) were divided into three groups (15 per treatment) and fed one of the three diets for 42 days. A higher daily weight gain (p = 0.042), as well as daily feed intake (p = 0.022), was observed in rabbits fed the AD and TM diets than in rabbits fed the SM diet within 21 days after weaning. The coefficients of total tract apparent digestibility (CTTAD) of gross energy were higher (p = 0.001) in rabbits fed the SM diet than in rabbits fed the other diets. The CTTAD of CP (p = 0.040) and starch (p = 0.041) was higher in rabbits fed the SM diet compared to those fed the AD diet. There were non-significantly higher losses of nitrogen in the urine (an average by 0.227 g/day; p = 0.094) in rabbits fed the TM diet than in rabbits fed the other diets. It can be concluded that the growth of rabbits and nitrogen output were not detrimentally affected by the insect meal (AD or TM) used in this study.

Rabbit is a reflexively ovulating species. Accordingly, in the practice of artificial insemination (AI) ovulation must be induced via exogenous GnRH (Gonadotropin-Releasing Hormone) administration, which may be performed intramuscularly, subcutaneously, or intravaginally. Unfortunately, the bioavailability of the GnRH analogue when added to the extender is lower due to the proteolytic activity in the seminal plasma and the poor permeability of the vaginal mucosa. The aim of the study was to refine the practice of AI practice in rabbits by replacing parenteral GnRH analogue administration (subcutaneous, intravenous, or intramuscular injection) with intravaginal application, while reducing its concentration in the diluent. Extenders containing the buserelin acetate in chitosan–dextran sulphate and chitosan–alginate nanoparticles were designed and 356 females were inseminated. Reproductive performance of females inseminated with the two experimental extenders, receiving 4 μg of buserelin acetate intravaginally per doe, was compared with that in the control group, the does of which were inseminated with the extender without the GnRH analogue and induced to ovulate with 1 μg of buserelin acetate administered intramuscularly. The entrapment efficiency of the chitosan–dextran sulphate complex was higher than that of chitosan–alginate. However, females inseminated with both systems showed similar reproductive performance. We conclude that both nanoencapsulation systems are an efficient way of intravaginal ovulation induction, allowing a reduction in the level of the GnRH analogue normally used in seminal doses from 15–25 μg to 4 μg.

This study was aimed at evaluating the biomechanical properties and gapping characteristics of tendon repair methods using a combination of a three-loop pulley (3LP) pattern, a titanium plate, and a polycaprolactone (PCL) plate in a rabbit gastrocnemius tendon (GT) model (n = 50). GTs were randomly assigned to five groups (n = 10/group). Transected GTs were repaired with a 3LP pattern alone or in conjunction with an epitendinous suture (ES), a 5-hole 1 mm PCL plate, a 5-hole 2 mm PCL plate, or a 5-hole 1.5 mm titanium plate. The yield, peak, and failure force, as well as the occurrence and force of 1-mm and 3-mm gapping were examined. The mean yield, peak, and failure force of the 3LP + titanium plate group were higher than that of other groups. The biomechanical properties of a 3LP + a 2 mm PCL plate were similar to 3LP + ES constructs in this model. In all specimens in all groups, 1 mm gap formation was observed. The frequency of 3 mm gap formation was 70% and 90% in the 3LP + 2 mm PCL plate group and the 3LP + 1.5 mm titanium plate group, respectively. Additional studies evaluating PCL plates to determine the effect on the healing and blood supply of tendon are needed.

Hydrolytic reactions constitute an important pathway of drug metabolism and a significant route of prodrug activation. Many ophthalmic drugs and prodrugs contain ester groups that greatly enhance their permeation across several hydrophobic barriers in the eye before the drugs are either metabolized or released, respectively, via hydrolysis. Thus, the development of ophthalmic drug therapy requires the thorough profiling of substrate specificities, activities, and expression levels of ocular esterases. However, such information is scant in the literature, especially for preclinical species often used in ophthalmology such as rabbits and pigs. Therefore, our aim was to generate systematic information on the activity and expression of carboxylesterases (CESs) and arylacetamide deacetylase (AADAC) in seven ocular tissue homogenates from these two species. The hydrolytic activities were measured using a generic esterase substrate (4-nitrophenyl acetate) and, in the absence of validated substrates for rabbit and pig enzymes, with selective substrates established for human CES1, CES2, and AADAC (D-luciferin methyl ester, fluorescein diacetate, procaine, and phenacetin). Kinetics and inhibition studies were conducted using these substrates and, again due to a lack of validated rabbit and pig CES inhibitors, with known inhibitors for the human enzymes. Protein expression levels were measured using quantitative targeted proteomics. Rabbit ocular tissues showed significant variability in the expression of CES1 (higher in cornea, lower in conjunctiva) and CES2 (higher in conjunctiva, lower in cornea) and a poor correlation of CES expression with hydrolytic activities. In contrast, pig tissues appear to express only CES1, and CES3 and AADAC seem to be either low or absent, respectively, in both species. The current study revealed remarkable species and tissue differences in ocular hydrolytic enzymes that can be taken into account in the design of esterase-dependent prodrugs and drug conjugates, the evaluation of ocular effects of systemic drugs, and in translational and toxicity studies.

Pasteurellosis is one of the rabbit's most bacterial severe diseases and leads to considerable financial damages in large production systems worldwide. Antibiotic use in animals may lead to antibiotic residues in animal products, including meat. Therefore, this study was designed to evaluate the potential role of grape seed extract (GSE) in treating Pasteurella multocida infection in rabbits. For this purpose, 45 weaned male New Zealand rabbits were divided into three groups; control, infected and infected‐GSE treated. Experimental P. multocida infection in rabbits induced a remarkable decrease in body weight, body weight gain, as well as microcytic hypochromic anaemia, leucocytosis, neutrophilia and lymphocytopenia. Also, a significant increase in the hepatic and renal injury biomarkers, in interleukin‐6, total globulin, α, β and γ globulins, as well as a marked reduction in total protein and albumin, were recorded in the P. multocida‐infected rabbits. Treatment of infected rabbits with GSE modulated most of these altered parameters. This study endorses the administration of GSE for the treatment of Pasteurellosis in rabbits. Further studies are required to identify the possible additional effects, appropriate doses and duration of the GSE therapy in rabbits Pasteurellosis., According to the authors' knowledge, no comprehensive work was dedicated to assess the possible impact of grape seed extract in treating of P. multocida infection in rabbits. Moreover, grape seed extract could represent a promising for enhancement of Pasteurellosis treatment in rabbits and reduction of ofloxacin side effects.

No. of bitstreams: 0 Previous issue date: 2021-01-01 The aim of this study was to compare the cardiorespiratory, arterial blood gas and antinociceptive effects of dexmedetomidine (D), dexmedetomidine–lidocaine (DL) or lidocaine (L) administered epidurally on conscious rabbits. Eight six-month-old male New Zealand rabbits were randomly distributed into three treatments: D (2.5 μg/kg); DL (2.5 μg/kg; 2 mg/kg); and L (2 mg/kg). The drugs were injected epidurally via a catheter. Cardiorespiratory, arterial blood gas and antinociceptive variables were recorded before administration, 5 and 10 min after drug administration, then every 10 min until the animals presented a positive response to nociceptive stimulation of perineal dermatomes. Two animals had permanent paralysis after DL treatment due to hemorrhage and congestion with neuron necrosis in spinal cord segments. There was a reduction in mean arterial pressure in treatment L at 5 and 10 min, compared with the baseline, and in treatment DL at 10–30 min. Increases in pH were observed in treatment D at 5 and 10 min, and in DL at all the times evaluated, compared with the baseline. No alterations were observed in other blood gas or electrolyte variables. Antinociceptive effects were evaluated in the perineal, sacral and lumbar regions, and were restricted to the perineal region following D and L treatment. The antinociceptive effects following DL were greater than D and L alone in all of the regions. L and D promotes short-term antinociceptive effects for up to 15 min and, when used in combination with D, increased the duration and extent of sensory block by up to 45 min. AIM: To observe changes in the content of matrix metalloproteinases (MMPs) in the corneal stroma after corneal cross-linking (CXL) in rabbits, and further explore the corneal pathophysiological process after CXL. METHODS: Forty-two rabbits (42 eyes) were randomly divided into seven groups. One group served as the control group, while the other six groups were treated with CXL. The concentrations of MMPs in corneal stroma were evaluated through parallel reaction monitoring at baseline and 3, 7, 15, 30, 90, and 180d after treatment. RESULTS: The levels of MMP-2 in the corneal stroma of rabbits were 0.76±0.07, 2.78±1.39, 4.12±0.69, 2.00±0.29, 2.00±0.30, 1.22±0.18, and 1.35±0.18 (10-9 mol/g) at baseline and 3, 7, 15, 30, 90, and 180d after treatment, respectively. The contents of tissue inhibitor of metalloproteinase-1 (TIMP-1) were 1.83±0.26, 7.94±0.58, 6.95±2.64, 3.81±0.48, 3.07±0.92, 1.72±0.19, and 1.69±0.74 (10-9 mol/g), respectively. The ratios of MMP-2/TIMP-1 were 0.42±0.33, 0.36±0.20, 0.62±0.10, 0.54±0.15, 0.68±0.13, 0.71±0.10, and 0.68±0.09, respectively. After CXL, the expression of MMP-2 and TIMP-1 in the rabbit corneal stroma was initially increased and subsequently decreased. The levels of MMP-2 remained higher than those recorded at baseline 180d after treatment, but it was not statistically significant. The levels of TIMP-1 returned to baseline levels at 90d after treatment. The ratio of MMP-2/TIMP-1 started to rise from 7d after CXL. It was significantly higher than that calculated at baseline 30-180d after CXL. The results for MMP-1, -3, -7, -9, -13, and TIMP-2 were negative. CONCLUSION: CXL can lead to changes in the content of MMP-2 and TIMP-1 in the rabbit corneal stroma. The ratio of MMP-2/TIMP-1 remains higher versus baseline, indicating that MMP-2 is involved in the corneal pathophysiological process after CXL.

Bioactive glasses (BAG) are used as bone-graft substitutes in orthopaedic surgery. A specific BAG scaffold was developed by sintering BAG-S53P4 granules. It is hypothesised that this scaffold can be used as a bone substitute to fill bone defects and induce a bioactive membrane (IM) around the defect site. Beyond providing the scaffold increased mechanical strength, that the initial inflammatory reaction and subsequent IM formation can be enhanced by coating the scaffolds with poly(DL-lactide-co-glycolide) (PLGA) is also hypothesised. To study the immunomodulatory effects, BAG-S53P4 (+/- PLGA) scaffolds were placed on monolayers of primary human macrophage cultures and the production of various pro- and anti-inflammatory cytokines was assessed using reverse transcriptase quantitative polymerase chain reaction (RT-qPCR) and ELISA. To study the osteogenic effects, BAG-S53P4 (+/- PLGA) scaffolds were cultured with rabbit mesenchymal stem cells and osteogenic differentiation was evaluated by RT-qPCR and matrix mineralisation assays. The scaffold ion release was quantified and the BAG surface reactivity visualised. Furthermore, the pH of culture media was measured. BAG-S53P4 scaffolds had both anti-inflammatory and osteogenic properties that were likely attributable to alkalinisation of the media and ion release from the scaffold. pH change, ion release, and immunomodulatory properties of the scaffold could be modulated by the PLGA coating. Contrary to the hypothesis, the coating functioned by attenuating the BAG surface reactions and subsequent anti-inflammatory properties, rather than inducing an elevated inflammatory response compared to BAG-S53P4 alone. These results further validated the use of BAG-S53P4 (+/- PLGA) scaffolds as bone substitutes and indicate that scaffold properties can be tailored to a specific clinical need.

The term fatty keratopathy is used to describe the phenomenon of fat deposition caused by corneal neovascularization, which will severely affect the eye’s beauty and vision. The purpose of this study was to establish a New Zealand white rabbit animal model of fatty keratopathy, that is, the establishment of an animal model of fatty keratopathy. The goal was achieved by the combination of a corneal neovascularization animal model and a hyperlipidemia animal model. Two groups were created according to the experimental sequence. The first group initially induced a corneal neovascularization pattern and later induced a hyperlipidemia pattern, and the second group followed the opposite sequence. The results of the two groups showed that all the significant crystalline deposits of the cornea were visible. So the animal models of fatty keratopathy were successfully established in both groups.

Feed additive consisting of polysaccharides from brown seaweeds plus phenolic acid, hydroxycinnamic acids, tannins, and flavonoids from plant extracts, was used as rabbit food supplement. Main aim of the study was to determine the effect of natural mix of marine and freshwater seaweed polyphenols on selected reproductive characteristics of male rabbits during the 90-days experiment. Natural mix was incorporated in feed-pellets for rabbits in two different concentrations – 0.3% (T1 group) and 0.6% (T2 group), compared with a control group (C group). In experimental groups a significant increase of concentration of calcium during first 30 days of supplementation was found. An increase of alanine aminotransferase, glutathione peroxidase and ferric reducing ability of plasma and a decrease in aspartate aminotransferase after 90 days were recorded in the same groups. Except for that we noticed decrease of semen distance of curved line and velocity of curved line after 30 days though only while being supplemented with 0.6% proportion of seaweed polyphenols in feed mixture. Based on the results it can be stated that the natural mix in the tested levels do not show adverse effect on male rabbit reproductive parameters, and an improvement of antioxidant status was observed. The feed additives can have a very important effect on growth, health and development of animals in general as it supplies the with the much-needed minerals, nutritional substances and antioxidants, on which we focused in our study.

Date palm pollen (DPP) has been widely tested in vitro in preservation of spermatozoa of different farm animals except in rabbits. The objective of this study was to evaluate the effect of various concentrations (20, 40, 80 mg/mL) of DPP aqueous extract on epididymal and ejaculated rabbit spermatozoa during in vitro incubation at 37 °C during 120 min. NaCl Pollen Extender (NPE) and TRIS Base Extender (TPE) were respectively used in epididymal and ejaculated sperm preservation. Sperm motility parameters were objectively analysed by a Computer Aided Sperm Analyser (CASA). The results revealed that in epididymal sperm at 0 minute, sperm motility showed significant higher values (p ≤ 0.05) for all doses of DPP in comparison to the control group. After 30, 60 and 120 min, the motility rate was higher (p > .05) in all the experimental groups compared to the control. Concerning velocity traits (VCL and VAP), and trajectories parameters (LIN, ALH, BCF), the doses of 40 mg/mL and 80 mg/mL showed the highest values than Control. In ejaculated samples, the motility rate at 0 minute improved only with the dose of 80 mg/mL and DPP showed protective effect at all doses (20, 40, 80 mg/mL) up to 60 min of incubation. DPP enhanced spermatozoa velocity (VCL and VAP) and the trajectory of sperm (LIN, ALH, BCF) even from 20 mg/mL at any points of storage. In conclusion, aqueous extract of date palm pollen is a suitable supplement to the extender for rabbit sperm by protecting and improving sperm motility parameters.HIGHLIGHTS The addition of DPP extract improves the quality of epididymal and ejaculated rabbit sperm. The extender supplemented with 80 mg/mL of DPP in NPE has a beneficial effect on kinetic traits of epididymal sperm. Nevertheless, in TPE extender, 20 mg/mL of DPP was enough to improve sperm motility traits in ejaculated rabbit semen.

We investigated the cross-species transmission of rabbit hepatitis E virus (rb HEV) to pigs and evaluated the cross-protection of a swine (sw) HEV-3 virus-like particle (VLP) vaccine against rb HEV infection in pigs. Twelve 4-week-old conventional pigs were divided into negative control (n = 3), positive control (rb HEV-infected, n = 4), and vaccinated (vaccinated and rb HEV-challenged, n = 5) groups. The vaccine was administered at weeks 0 and 2, and viral challenge was conducted at week 4. Serum HEV RNA, anti-HEV antibody, cytokine, and liver enzyme levels were determined. Histopathological lesions were examined in abdominal organs. Viral RNA was detected and increased anti-HEV antibody and alanine aminotransferase (ALT) levels were observed in positive control pigs; liver fibrosis, inflammatory cell infiltration in the lamina propria of the small intestine and shortened small intestine villi were also observed. In vaccinated pigs, anti-HEV antibody and Th1 cytokine level elevations were observed after the second vaccination; viral RNA was not detected, and ALT level elevations were not observed. The results verified the cross-species transmission of rb HEV to pigs and cross-protection of the sw HEV-3 VLP vaccine against rb HEV infection in pigs. This vaccine may be used for cross-protection against HEV infection in other species.
