This study aims at the synthesis and initial biological evaluation of novel rhenium–tricarbonyl complexes of 3,3′,4′,5,7-pentahydroxyflavone (quercetin), 3,7,4΄-trihydroxyflavone (resokaempferol), 5,7-dihydroxyflavone (chrysin) and 4΄,5,7-trihydroxyflavonone (naringenin) as neuroprotective and anti-PrP agents. Resokaempferol was synthesized from 2,2΄,4-trihydroxychalcone by H2O2/NaOH. The rhenium–tricarbonyl complexes of the type fac-[Re(CO)3(Fl)(sol)] were synthesized by reacting the precursor fac-[Re(CO)3(sol)3]+ with an equimolar amount of the flavonoids (Fl) quercetin, resokaempferol, chrysin and naringenin and the solvent (sol) was methanol or water. The respective Re–flavonoid complexes were purified by semi-preparative HPLC and characterized by spectroscopic methods. Furthermore, the structure of Re–chrysin was elucidated by X-ray crystallography. Initial screening of the neuroprotective properties of these compounds included the in vitro assessment of the antioxidant properties by the DPPH assay as well as the anti-lipid peroxidation of linoleic acid in the presence of AAPH and their ability to inhibit soybean lipoxygenase. From the above studies, it was concluded that the complexes’ properties are mainly correlated with the structural characteristics and the presence of the flavonoids. The flavonoids and their respective Re-complexes were also tested in vitro for their ability to inhibit the formation and aggregation of the amyloid-like abnormal prion protein, PrPSc, by employing the real-time quaking-induced conversion assay with recombinant PrP seeded with cerebrospinal fluid from patients with Creutzfeldt–Jakob disease. α-Synucleinopathies, such as Parkinson’s disease (PD), dementia with Lewy bodies (DLB) and multiple system atrophy, are a class of neurodegenerative diseases exhibiting intracellular inclusions of misfolded α-synuclein (αSyn), referred to as Lewy bodies or oligodendroglial cytoplasmic inclusions (Papp–Lantos bodies). Even though the specific cellular distribution of aggregated αSyn differs in PD and DLB patients, both groups show a significant pathological overlap, raising the discussion of whether PD and DLB are the same or different diseases. Besides clinical investigation, we will focus in addition on methodologies, such as protein seeding assays (real-time quaking-induced conversion), to discriminate between different types of α-synucleinopathies. This approach relies on the seeding conversion properties of misfolded αSyn, supporting the hypothesis that different conformers of misfolded αSyn may occur in different types of α-synucleinopathies. Understanding the pathological processes influencing the disease progression and phenotype, provoked by different αSyn conformers, will be important for a personalized medical treatment in future.

Fatal familial insomnia is a rare hereditary prion disease associated with the D178N-129M PRNP mutation. Early diagnosis is difficult, because the clinical syndrome may overlap with affective disorders. In addition, most known cerebrospinal fluid biomarkers for prion diseases and magnetic resonance imaging do not show a good diagnostic accuracy for fatal familial insomnia. In this context, data on plasma biomarkers are scarce.We analyzed levels of neurofilament light chain, glial fibrillary acidic protein, chitinase-3-like protein 1, calcium-binding protein B, and total tau protein in six serial plasma samples from a patient with fatal familial insomnia. Subsequently, plasma neurofilament light chain was analyzed in n = 25 patients and n = 19 controls. The diagnostic accuracy and associations with disease stage and duration were explored.Among all biomarker candidates in the case study, only neurofilament light chain levels showed a constant evolution and increased over time. They discriminated fatal familial insomnia from controls with an area under the curve of 0.992 (95% confidence interval [CI] = 0.974-1) in the case-control study. Higher concentrations were associated with methionine homozygosity at codon 129 PRNP (p = 0.006), shorter total disease duration (rho = -0.467, p = 0.019, 95% CI = -0.790 to -0.015), and shorter time from sampling to death (rho = -0.467, p = 0.019, 95% CI = -0.773 to -0.019).Plasma neurofilament light chain may be a valuable minimally invasive diagnostic biomarker for fatal familial insomnia after clinical onset. Most important, stage-related increase and association with disease duration indicate potential as a prognostic marker and as a surrogate marker in clinical trials.

Genetic prion diseases are a rare and diverse group of fatal neurodegenerative disorders caused by pathogenic sequence variations in the prion protein gene, PRNP. Data on CSF biomarkers in patients with genetic prion diseases are limited and conflicting results have been reported for unclear reasons. Here, we aimed to analyse the diagnostic accuracy of CSF biomarkers currently used in prion clinical diagnosis in 302 symptomatic genetic prion disease cases from 11 prion diagnostic centres, encompassing a total of 36 different pathogenic sequence variations within the open reading frame of PRNP. CSF samples were assessed for the surrogate markers of neurodegeneration, 14-3-3 protein (14-3-3), total-tau protein (t-tau) and α-synuclein and for prion seeding activity through the real-time quaking-induced conversion assay. Biomarker results were compared with those obtained in healthy and neurological controls. For the most prevalent PRNP pathogenic sequence variations, biomarker accuracy and associations between biomarkers, demographic and genetic determinants were assessed. Additionally, the prognostic value of biomarkers for predicting total disease duration from symptom onset to death was investigated. High sensitivity of the four biomarkers was detected for genetic Creutzfeldt–Jakob disease associated with the E200K and V210I mutations, but low sensitivity was observed for mutations associated with Gerstmann–Sträussler–Scheinker syndrome and fatal familial insomnia. All biomarkers showed good to excellent specificity using the standard cut-offs often used for sporadic Creutzfeldt–Jakob disease. In genetic prion diseases related to octapeptide repeat insertions, the biomarker sensitivity correlated with the number of repeats. New genetic prion disease-specific cut-offs for 14-3-3, t-tau and α-synuclein were calculated. Disease duration in genetic Creutzfeldt–Jakob disease-E200K, Gerstmann–Sträussler–Scheinker-P102L and fatal familial insomnia was highly dependent on PRNP codon 129 MV polymorphism and was significantly associated with biomarker levels. In a large cohort of genetic prion diseases, the simultaneous analysis of CSF prion disease biomarkers allowed the determination of new mutation-specific cut-offs improving the discrimination of genetic prion disease cases and unveiled genetic prion disease-specific associations with disease duration.

Herein, we describe two complementary approaches towards various organic thiocyanates that are affordable, reliable and follow the principles of sustainable chemistry, starting from commercially available thiols or disulfides. Additionally, the application of this mild method to the first total synthesis of psammaplin B is demonstrated. Non-toxic and inexpensive ferricyanide is used as the cyanide source, which can be activated either in a mechanochemical, solvent-free approach, or in a biphasic solvent system allowing easier work-up. A total of 27 examples is demonstrated, with up to quantitative yields.

Background The cellular prion protein (PrPC) is a membrane-bound, multifunctional protein mainly expressed in neuronal tissues. Recent studies indicate that the native trafficking of PrPC can be misused to internalize misfolded amyloid beta and α-synuclein (aSyn) oligomers. Objectives We define PrPC's role in internalizing misfolded aSyn in α-synucleinopathies and identify further involved proteins. Methods We performed comprehensive behavioral studies on four transgenic mouse models (ThySyn and ThySynPrP00, TgM83 and TgMPrP00) at different ages. We developed PrPC-(over)-expressing cell models (cell line and primary cortical neurons), used confocal laser microscopy to perform colocalization studies, applied mass spectrometry to identify interactomes, and determined disassociation constants using surface plasmon resonance (SPR) spectroscopy. Results Behavioral deficits (memory, anxiety, locomotion, etc.), reduced lifespans, and higher oligomeric aSyn levels were observed in PrPC-expressing mice (ThySyn and TgM83), but not in homologous Prnp ablated mice (ThySynPrP00 and TgMPrP00). PrPC colocalized with and facilitated aSyn (oligomeric and monomeric) internalization in our cell-based models. Glimepiride treatment of PrPC-overexpressing cells reduced aSyn internalization in a dose-dependent manner. SPR analysis showed that the binding affinity of PrPC to monomeric aSyn was lower than to oligomeric aSyn. Mass spectrometry-based proteomic studies identified clathrin in the immunoprecipitates of PrPC and aSyn. SPR was used to show that clathrin binds to recombinant PrP, but not aSyn. Experimental disruption of clathrin-coated vesicles significantly decreased aSyn internalization. Conclusion PrPC's native trafficking can be misused to internalize misfolded aSyn through a clathrin-based mechanism, which may facilitate the spreading of pathological aSyn. Disruption of aSyn-PrPC binding is, therefore, an appealing therapeutic target in α-synucleinopathies.

Long triplet lifetimes of excited photosensitizers are essential for efficient energy transfer reactions in water, given that the concentrations of dissolved oxygen and suitable acceptors in aqueous media are typically much lower than in organic solvents. Herein, we report the design, synthesis and photochemical characterization of two structurally related water-soluble ruthenium complex-based dyads decorated with a covalently attached pyrene chromophore. The triplet energy of the latter is slightly below that of the metal complex enabling a so-called triplet reservoir and excited-state lifetime extensions of up to two orders of magnitude. The diimine co-ligands, which can be modified easily, have a major impact on both the ultrafast intramolecular energy transfer (iEnT) kinetics upon excitation with visible light and the lifetime of the resulting long-lived pyrene triplet. The phenanthroline-containing dyad shows fast triplet pyrene formation (25 ps) and an exceptionally long triplet lifetime beyond 50 microseconds in neat water. The iEnT process

Centro de Investigación en Sanidad Animal (CISA), Triggering receptor expressed on myeloid cells 2 (TREM2) is an innate immune cell surface receptor that regulates microglial function and is involved in the pathophysiology of several neurodegenerative diseases. Its soluble form (sTREM2) results from shedding of the TREM2 ectodomain. The role of TREM2 in prion diseases, a group of rapidly progressive dementias remains to be elucidated. In the present study, we analysed the expression of TREM2 and its main sheddase ADAM10 in the brain of sporadic Creutzfeldt-Jakob disease (sCJD) patients and evaluated the role of CSF and plasma sTREM2 as a potential diagnostic marker of prion disease. Our data indicate that, compared to controls, TREM2 is increased in sCJD patient brains at the mRNA and protein levels in a regional and subtype dependent fashion, and expressed in a subpopulation of microglia. In contrast, ADAM10 is increased at the protein, but not the mRNA level, with a restricted neuronal expression. Elevated CSF sTREM2 is found in sCJD, genetic CJD with mutations E200K and V210I in the prion protein gene (PRNP), and iatrogenic CJD, as compared to healthy controls (HC) (AUC = 0.78-0.90) and neurological controls (AUC = 0.73-0.85), while CSF sTREM2 is unchanged in fatal familial insomnia. sTREM2 in the CSF of cases with Alzheimer's disease, and multiple sclerosis was not significantly altered in our series. CSF sTREM2 concentrations in sCJD are PRNP codon 129 and subtype-related, correlate with CSF 14-3-3 positivity, total-tau and YKL-40, and increase with disease progression. In plasma, sTREM2 is increased in sCJD compared with HC (AUC = 0.80), displaying positive correlations with plasma total-tau, neurofilament light, and YKL-40. Longitudinal PD CSF samples were subjected to ICP-MS and the total amount of iron and other bioelements was quantified. Additionally, ferritin and protein biomarkers of neurodegeneration were measured. Over time, mean iron levels significantly increased while levels of ferritin decreased.

The development of the real-time quaking-induced conversion (RT-QuIC), an in vitro protein misfolding amplification assay, was an innovation in the scientific field of protein misfolding diseases. In prion diseases, these types of assays imitate the pathological conversion of the cellular prion protein (PrPC) into a protease-resistant and/or amyloid form of PrP, called PrP resistant (PrPRes). The RT-QuIC is an automatic assay system based on real-time measuring of thioflavin-T (Th-T) incorporation into amyloid fibrils using shaking for disaggregation. It has already been applied in diagnostics, drug pre-screening, and to distinguish between different prion strains. The seeded conversion efficiency and the diagnostic accuracy of the RT-QuIC assay strongly depend on the kind of recombinant PrP (rec PrP) substrate. The DNA sequences of different substrates may originate from different species, such as human, bank vole, and hamster, or from a combination of two species, e.g., hamster-sheep chimera. In routine use, either full-length (FL) or truncated substrates are applied which can accelerate the conversion reaction, e.g., to a more sensitive version of RT-QuIC assay. In the present review, we provide an overview on the different types of PrP substrates (FL and truncated forms), recapitulate the production and purification process of different rec PrP substrates, and discuss the diagnostic value of CSF RT-QuIC in human prion disease diagnostics.

Contains fulltext : 282534.pdf (Publisher’s version ) (Open Access) BACKGROUND AND PURPOSE: Cerebrospinal fluid (CSF) real-time quaking-induced conversion (RT-QuIC) has a high degree of sensitivity and specificity for the diagnosis of sporadic Creutzfeldt-Jakob disease (sCJD) and this has led to its being included in revised European CJD Surveillance Network diagnostic criteria for sCJD. As CSF RT-QuIC becomes more widely established, it is crucial that the analytical performance of individual laboratories is consistent. The aim of this ring-trial was to ascertain the degree of concordance between European countries undertaking CSF RT-QuIC. METHODS: Ten identical CSF samples, seven from probable or neuropathologically confirmed sCJD and three from non-CJD cases, were sent to 13 laboratories from 11 countries for RT-QuIC analysis. A range of instrumentation and different recombinant prion protein substrates were used. Each laboratory analysed the CSF samples blinded to the diagnosis and reported the results as positive or negative. RESULTS: All 13 laboratories correctly identified five of the seven sCJD cases and the remaining two sCJD cases were identified by 92% of laboratories. Of the two sCJD cases that were not identified by all laboratories, one had a disease duration >26 months with a negative 14-3-3, whilst the remaining case had a 4-month disease duration and a positive 14-3-3. A single false positive CSF RT-QuIC result was observed in this study. CONCLUSIONS: This study shows that CSF RT-QuIC demonstrates an excellent concordance between centres, even when using a variety of instrumentation, recombinant prion protein substrates and CSF volumes. The adoption of CSF RT-QuIC by all CJD surveillance centres is recommended.

β-glucocerebrosidase (GBA)-associated mutations are a significant risk factor for Parkinson’s disease (PD) that aggravate the disease pathology by upregulating the deposition of α-Synuclein (α-Syn). The resultant clinical profile varies for PD patients without GBA mutations. The current study aimed to identify the proteomic targets involved in the pathogenic pathways leading to the differential clinical presentation of GBA-associated PD. CSF samples (n = 32) were obtained from PD patients with GBA mutations (n = 22), PD patients without GBA mutations (n = 7), and healthy controls that were carriers of GBA mutations (n = 3). All samples were subjected to in-gel tryptic digestion followed by the construction of the spectral library and quantitative SWATH-based analysis. CSF α-Syn levels were reduced in both PDIdiopathic and PDGBA cases. Our SWATH-based mass spectrometric analysis detected 363 proteins involved in immune response, stress response, and cell signaling in various groups. Intergroup analysis showed that 52 proteins were significantly up- or downregulated in various groups. Of these 52 targets, 20 proteins were significantly altered in PDGBA cases only while 2 showed different levels in PDIdiopathic patients. Our results show that the levels of several pathologically relevant proteins, including Contactin-1, Selenium-binding protein 1, Adhesion G Protein-Coupled Receptor, and Apolipoprotein E are significantly different among the sporadic and genetic variants of PD and hint at aggravated synaptic damage, oxidative stress, neuronal loss, and aggregation of α-Syn in PDGBA cases.

Biomass fuel use in chemical looping combustion enables negative CO2 emissions through BECCS (Bio-Energy Carbon Capture and Storage). Effective biomass utilization in CLC requires an economical and effective oxygen carrier to achieve high fuel conversion, effective CO2 capture, and management of the harmful effects of biomass alkali release (bed agglomeration, oxygen carrier deactivation, fouling and corrosion). These issues were addressed in 100 kW CLC pilot experiments. Building on previous work, a mixture of a synthetic calcium manganite perovskite and natural ilmenite was used as the oxygen carrier. Four biomass fuels of varied alkali content were tested: black pellets of steam-exploded stem wood (BP), BP impregnated with K2CO3, a mixture of 50% BP with 50% straw pellets, and wood char. Experiments showed high fuel conversion and very high CO2 capture, with overall performance exceeding that of ilmenite and manganese ore. More than 95% gas conversion was achieved with black pellets at around 950 °C. The fate of biomass alkali, previously virtually unknown in CLC research, was explored by implementing online surface-ionization-based measurement of alkali released in the flue gases of the fuel reactor (FR) and air reactor (AR). Release levels were found to correlate with the fuel alkali content. The flue gas measurements and bed material elemental analyses suggest that most of the fuel alkali are accumulated in the oxygen carrier. Unexpectedly, it was found that flue gas alkali release occurs in both the FR and AR, with AR exhibiting an equal or higher rate of release vs. the FR.

AIMS In the search for blood-based biomarkers of neurodegenerative diseases, we characterized the concentration of total prion protein (t-PrP) in the plasma of neurodegenerative dementias. We aimed to assess its accuracy in this differential diagnostic context. METHODS Plasma t-PrP was measured in 520 individuals including healthy controls (HC) and patients diagnosed with neurological disease control (ND), Alzheimer's disease (AD), sporadic Creutzfeldt-Jakob disease (sCJD), frontotemporal dementia (FTD), Lewy body dementia (LBD) and vascular dementia (VaD). Additionally, t-PrP was quantified in genetic prion diseases and iatrogenic CJD. The accuracy of t-PrP discriminating the diagnostic groups was evaluated and correlated with demographic, genetic and clinical data in prion diseases. Markers of blood-brain barrier impairment were investigated in sCJD brains. RESULTS Compared to HC and ND, elevated plasma t-PrP concentrations were detected in sCJD, followed by FTD, AD, VaD and LBD. In sCJD, t-PrP was associated neither with age nor sex, but with codon 129 PRNP genotype. Plasma t-PrP concentrations correlated with cerebrospinal fluid (CSF) markers of neuro-axonal damage, but not with CSF t-PrP. In genetic prion diseases, plasma t-PrP was elevated in all type of mutations investigated. In sCJD brain tissue, extravasation of immunoglobulin G and the presence of swollen astrocytic end-feet around the vessels suggested leakage of blood-brain barrier as a potential source of increased plasma t-PrP. CONCLUSIONS Plasma t-PrP is elevated in prion diseases regardless of aetiology. This pilot study opens the possibility to consider plasma t-PrP as a promising blood-based biomarker in the diagnostic of prion disease.

Objectives Currently, the exact reasons why different α-synucleinopathies exhibit variable pathologies and phenotypes are still unknown. A potential explanation may be the existence of distinctive α-synuclein conformers or strains. Here, we intend to analyze the seeding activity of dementia with Lewy bodies (DLB) and Parkinson's disease (PD) brain-derived α-synuclein seeds by real-time quaking-induced conversion (RT-QuIC) and to investigate the structure and morphology of the α-synuclein aggregates generated by RT-QuIC. Methods A misfolded α-synuclein-enriched brain fraction from frontal cortex and substantia nigra pars compacta tissue, isolated by several filtration and centrifugation steps, was subjected to α-synuclein/RT-QuIC analysis. Our study included neuropathologically well-characterized cases with DLB, PD, and controls (Ctrl). Biochemical and morphological analyses of RT-QuIC products were conducted by western blot, dot blot analysis, Raman spectroscopy, atomic force microscopy, and transmission electron microscopy. Results Independently from the brain region, we observed different seeding kinetics of α-synuclein in the RT-QuIC in patients with DLB compared to PD and Ctrl. Biochemical characterization of the RT-QuIC product indicated the generation of a proteinase K-resistant and fibrillary α-synuclein species in DLB-seeded reactions, whereas PD and control seeds failed in the conversion of wild-type α-synuclein substrate. Interpretation Structural variances of α-synuclein seeding kinetics and products in DLB and PD indicated, for the first time, the existence of different α-synuclein strains in these groups. Therefore, our study contributes to a better understanding of the clinical heterogeneity among α-synucleinopathies, offers an opportunity for a specific diagnosis, and opens new avenues for the future development of strain-specific therapies. Background Blood neurofilament light (Nfl) and total-tau (t-tau) have been described to be increased in several neurological conditions, including prion diseases and other neurodegenerative dementias. Here, we aim to determine the accuracy of plasma Nfl and t-tau in the differential diagnosis of neurodegenerative dementias and their potential value as prognostic markers of disease severity. Methods Plasma Nfl and t-tau were measured in healthy controls (HC, n = 70), non-neurodegenerative neurological disease with (NND-Dem, n = 17) and without dementia syndrome (NND, n = 26), Alzheimer’s disease (AD, n = 44), Creutzfeldt-Jakob disease (CJD, n = 83), dementia with Lewy bodies/Parkinson’s disease with dementia (DLB/PDD, n = 35), frontotemporal dementia (FTD, n = 12), and vascular dementia (VaD, n = 22). Biomarker diagnostic accuracies and cutoff points for the diagnosis of CJD were calculated, and associations between Nfl and t-tau concentrations with other fluid biomarkers, demographic, genetic, and clinical data in CJD cases were assessed. Additionally, the value of Nfl and t-tau predicting disease survival in CJD was evaluated. Results Among diagnostic groups, highest plasma Nfl and t-tau concentrations were detected in CJD (fold changes of 38 and 18, respectively, compared to HC). Elevated t-tau was able to differentiate CJD from all other groups, whereas elevated Nfl concentrations were also detected in NND-Dem, AD, DLB/PDD, FTD, and VaD compared to HC. Both biomarkers discriminated CJD from non-CJD dementias with an AUC of 0.93. In CJD, plasma t-tau, but not Nfl, was associated with PRNP codon 129 genotype and CJD subtype. Positive correlations were observed between plasma Nfl and t-tau concentrations, as well as between plasma and CSF concentrations of both biomarkers (p Conclusions Plasma Nfl and t-tau are useful complementary biomarkers for the differential diagnosis of CJD. Additionally, plasma t-tau emerges as a potential prognostic marker of disease duration.

The cellular prion protein (PrPC) is a central player in neurodegenerative diseases caused by protein misfolding, such as prion diseases or Alzheimer’s disease (AD). Expression levels of this GPI-anchored glycoprotein, especially at the neuronal cell surface, critically correlate with various pathomechanistic aspects underlying these diseases, such as templated misfolding (in prion diseases) and neurotoxicity and, hence, with disease progression and severity. In stark contrast to cell-associated PrPC, soluble extracellular forms or fragments of PrP are linked with neuroprotective effects, which is likely due to their ability to interfere with neurotoxic disease-associated protein conformers in the interstitial fluid. Fittingly, the endogenous proteolytic release of PrPCby the metalloprotease ADAM10 (‘shedding’) was characterized as a protective mechanism. Here, using a recently generated cleavage-site specific antibody, we shed new light on earlier studies by demonstrating that shed PrP (sPrP) negatively correlates with conformational conversion (in prion disease) and is markedly redistributed in murine brain in the presence of prion deposits or AD-associated amyloid plaques indicating a blocking and sequestrating activity. Importantly, we reveal that administration of certain PrP-directed antibodies and other ligands results in increased PrP shedding in cells and organotypic brain slice cultures. We also provide mechanistic and structural insight into this shedding-stimulating effect. In addition, we identified a striking exception to this, as one particular neuroprotective antibody, due to its special binding characteristics, did not cause increased shedding but rather strong surface clustering followed by fast endocytosis and degradation of PrPC. Both mechanisms may contribute to the beneficial action described for some PrP-directed antibodies/ligands and pave the way for new therapeutic strategies against devastating and currently incurable neurodegenerative diseases.

The effect of a cellular prion protein (PrP c ) deficiency on neuroenergetics was primarily analyzed via surveying the expression of genes specifically involved in lactate/pyruvate metabolism, such as monocarboxylate transporters (MCT1, MCT2, MCT4). The aim of the present study was to elucidate a potential involvement of PrP c in the regulation of energy metabolism in different brain regions. By using quantitative real-time polymerase chain reaction (qRT-PCR), we observed a marked reduction in MCT1 mRNA expression in the cortex of symptomatic Zürich I Prnp -/- mice, as compared to their wild-type (WT) counterparts. MCT1 downregulation in the cortex was accompanied with significantly decreased expression of the MCT1 functional interplayer, the Na + /K + ATPase α2 subunit. Conversely, the MCT1 mRNA level was significantly raised in the cerebellum of Prnp -/- vs. WT control group, without a substantial change in the Na + /K + ATPase α2 subunit expression. To validate the observed mRNA findings, we confirmed the observed change in MCT1 mRNA expression level in the cortex at the protein level. MCT4, highly expressed in tissues that rely on glycolysis as an energy source, exhibited a significant reduction in the hippocampus of Prnp -/- vs. WT mice. The present study demonstrates that a lack of PrP c leads to altered MCT1 and MCT4 mRNA/protein expression in different brain regions of Prnp -/- vs. WT mice. Our findings provide evidence that PrP c might affect the monocarboxylate intercellular transport, which needs to be confirmed in further studies.

Biomarkers are increasingly recognized as tools in the diagnosis and prognosis of neurodegenerative diseases. No fluid biomarker for Parkinson’s disease (PD) has been established to date, but α-synuclein, a major component of Lewy bodies in PD and dementia with Lewy bodies (DLB), has become a promising candidate. Here, we investigated CSF α-synuclein in patients with PD (n = 28), PDD (n = 8), and DLB (n = 5), applying an electrochemiluminescence immunoassay. Median values were non-significantly (p = 0.430) higher in patients with PDD and DLB (287 pg/mL) than in PD (236 pg/mL). A group of n = 36 primarily non-demented patients with PD and PDD was clinically followed for up to two years. A higher baseline α-synuclein was associated with increases in Hoehn and Yahr classifications (p = 0.019) and Beck Depression Inventory scores (p < 0.001) as well as worse performance in Trail Making Test A (p = 0.017), Trail Making Test B (p = 0.043), and the Boston Naming Test (p = 0.002) at follow-up. Surprisingly, higher levels were associated with a better performance in semantic verbal fluency tests (p = 0.046). In summary, CSF α-synuclein may be a potential prognostic marker for disease progression, affective symptoms, and executive cognitive function in PD. Larger-scaled studies have to validate these findings and the discordant results for single cognitive tests in this exploratory investigation.

Background Differential diagnosis of neurodegenerative dementia is currently supported by biomarkers including cerebrospinal fluid (CSF) tests. Among them, CSF total-tau (t-tau), phosphorylated tau (p-tau) and β-amyloid42 (Aβ42) are considered core biomarkers of neurodegeneration. In the present work, we hypothesize that simultaneous assessment of these biomarkers together with CSF α-synuclein (α-syn) will significantly improve the differential diagnostic of Alzheimer’s disease and other dementias. To that aim, we characterized the analytical and clinical performance of a new tetra-plex immunoassay that simultaneously quantifies CSF Aβ42, t-tau, p-tau and α-syn in the differential diagnosis of neurodegenerative dementia. Methods Biomarkers’ concentrations were measured in neurological controls (n=38), Alzheimer’s disease (n=35), Creutzfeldt-Jakob disease (n=37), vascular dementia (n=28), dementia with Lewy bodies/Parkinson’s disease dementia (n=27) and frontotemporal dementia (n=34) using the new tetra-plex assay and established single-plex assays. Biomarker’s performance was evaluated and diagnostic accuracy in the discrimination of diagnostic groups was determined using partial least squares discriminant analysis. Results The tetra-plex assay presented accuracies similar to individual single-plex assays with acceptable analytical performance. Significant correlations were observed between tetra-plex and single-plex assays. Using partial least squares discriminant analysis, Alzheimer’s disease and Creutzfeldt-Jakob disease were well-differentiated, reaching high accuracies in the discrimination from the rest of diagnostic groups. Conclusions The new tetra-plex assay coupled with multivariate analytical approaches becomes a valuable asset for the differential diagnosis of neurodegenerative dementia and related applications.

Introduction We developed a prognostic model for overall survival after diagnosis of sporadic Creutzfeldt-Jakob disease (sCJD) using data from a German surveillance study. Methods We included 1226 sCJD cases (median age 66 years, range 19-89 years; 56.8% women with information on age, sex, codon 129 genotype, 14-3-3 in the cerebrospinal fluid (CSF), and CSF tau concentrations. The prognostic accuracy for overall survival was measured by the c statistics of multivariable Cox proportional hazard models. A score chart was derived to predict 6-month survival and median survival time. Results A model containing age, sex, codon 129 genotype, and CSF tau (with two-way interactions) was selected as the model with the highest c statistic (0.686, 95% confidence interval: 0.665-0.707) in a cross-validation approach. Discussion We developed the first prognostic model for overall survival of sCJD patients based on readily available information only. The developed score chart serves as a hands-on prediction tool for clinical practice.

Chemical looping combustion (CLC) is a type of carbon capture technology that employs metal oxide particles in a redox reaction to transport oxygen to the fuel. As opposed to first-generation carbon capture concepts, CLC does not include a gas separation step and thus offers the advantage of having no thermodynamic energy penalty in the capture process. Manganese-containing oxygen carriers have been shown to offer a good compromise of gas conversion performance, availability, and cost. On the other hand, previously tested manganese materials have often been challenged with high attrition in continuous CLC. By sintering the ore prior to its use as oxygen carrier, the structure of the particles can be reinforced to increase their durability in the process. In this study, a sintered manganese ore was tested for 56 h in two pilot units of different size. The oxygen carrier worked well in the process, was easily fluidized, and did not exhibit any tendency towards agglomeration. Compared to other manganese materials tested in the same units, a higher lifetime based on fines production was reached, while gas conversion performance was similar. Different biomass fuels, mainly biochar of different sizes and black wood pellets, were employed in the study. A clear correlation between gas conversion and the fuel volatile content was detected; this is consistent with results reported in previous studies. The highest gas conversion reached in both units was similar to 93.5% using wood char. The highest carbon capture efficiency, 99% and 100% in the 10- and 100-kW unit, respectively, was reached with black pellets. To compare the tested material with the state-of-the-art oxygen carrier ilmenite, some tests were conducted with a bituminous coal that had been used in the same pilot with ilmenite. The results indicate a higher conversion performance for the tested manganese material.

High levels of total α-synuclein (t-α-synuclein) in the cerebrospinal fluid (CSF) were reported in sporadic Creutzfeldt-Jakob disease (sCJD). The potential use of t-α-synuclein in the discrimination of Lewy body dementias (i.e., Parkinson's disease dementia (PDD) and dementia with Lewy bodies (DLB)) is still under investigation. In addition, phospho-serine-129 α-synuclein (p-α-synuclein) has been described to be slightly increased in the CSF of synucleinopathies. Here, we analyzed t-α-synuclein and p-α-synuclein concentrations and their ratio in the context of differential diagnosis of neurodegenerative diseases. We quantified the levels of CSF t-α-synuclein and p-α-synuclein in a cohort of samples composed of neurological controls (NC), sCJD, PDD, and DLB by means of newly developed specific enzyme-linked immunosorbent assays. T-α-synuclein and p-α-synuclein were specifically elevated in sCJD compared to other disease groups. The area under the curve (AUC) values for t-α-synuclein were higher for the discrimination of sCJD from dementias associated to Lewy bodies as compared to the use of p-α-synuclein. A combination of both markers even increased the diagnostic accuracy. An inverse correlation was observed in CSF between t-α-synuclein and p-α-synuclein, especially in the DLB group, indicating a disease-relevant association between both markers. In conclusion, our data confirm t-α-synuclein and p-α-synuclein as robust biomarkers for sCJD and indicate the potential use of colorimetric t-α-synuclein ELISAs for differential diagnosis of dementia types.

Background: No candidate biomarkers based on cerebrospinal fluid (CSF) have been identified as prognostic factors in patients with major depression treated with electroconvulsive therapy (ECT), yet. Method: Following different underlying hypotheses, we analysed baseline CSF levels of markers of neurodegeneration (tau proteins, β-amyloids and neurogranin), elements of the innate immune system (interleukin [IL]-6, neopterin, soluble CD14, soluble CD163, migration inhibitory factor and monocyte chemotactic protein 1), endocannabinoids, sphingolipids and Klotho before ECT in patients with depression (n = 12) to identify possible correlations with the clinical antidepressant response to ECT. Results: Correlation with the reduction of the depressive symptoms could be observed especially for markers of neurodegeneration and elements of the innate immune system. Differences for CSF levels of several markers were found between the groups of responders and non-responders at the trend level. Limitations: The sample size is small and the ­distribution of responders and non-responders is uneven. Conclusions: It is this first study on CSF biomarkers for antidepressant efficacy of ECT warrants further research regarding the mechanism of ECT and personalized antidepressant therapy.

ObjectiveTo validate an amended protocol for clinical diagnosis of sporadic Creutzfeldt-Jakob disease (sCJD) including real-time quaking-induced conversion (RT-QuIC) and to observe its use in CJD surveillance.MethodsIn the framework of a prospective epidemiologic study, all neuropathologically confirmed cases with sCJD who received CSF RT-QuIC analysis during diagnostic workup (n = 65) and a control group of individuals without CJD (n = 118) were selected to investigate the accuracy of an amended diagnostic protocol. The patients had been referred to the German National Reference Center for Transmissible Spongiform Encephalopathies. The influence of the amended protocol on incidence figures was evaluated in the context of 3 years of surveillance activity (screened cases using 14-3-3 test n = 18,789, highly suspicious cases of CJD n = 704). Annual incidences were calculated with current criteria and the amended protocol.ResultsThe amended protocol showed a sensitivity of 97% and a specificity of 99%. When it was applied to all suspected cases who were referred to the reference center, the assessed incidence of CJD increased from 1.7 to 2.2 per million in 2016.ConclusionCJD surveillance remains challenging because information from external health care institutions can be limited. RT-QuIC shows excellent diagnostic accuracy when applied in the clinical setting to symptomatic patients. Data for RT-QuIC alone when applied as a general screening test are not available yet. We propose an amended research protocol that improves early and accurate clinical diagnosis of sCJD during surveillance activities. The use of this protocol will probably lead to a significant increase of the incidence rate.Classification of evidenceThis study provides Class III evidence that for patients with suspected sCJD, criteria for clinical diagnosis plus the CSF RT-QuIC accurately identifies patients with sCJD (sensitivity 97%, specificity 99%).

Creutzfeldt–Jakob disease and Alzheimer's disease are characterized by the presence of elevated total‐Tau cerebrospinal fluid concentrations while the presence of hyperphosphorylated Tau forms in the cerebrospinal fluid is rather a hallmark of Alzheimer's disease. Here we aimed to investigate potential contribution of nonphospho‐Tau epitopes (non‐P‐Tau) in the discrimination between both diseases. Non‐P‐Tau cerebrospinal fluid concentration was highly increased in Creutzfeldt–Jakob disease (n = 57, 3683 ± 3599 pg/mL) compared to Alzheimer's disease (n = 41, 148 ± 219 pg/mL) and neurological controls (n = 56, 62 ± 40 pg/mL), and significantly improved the proportion of correctly classified patients (99%) compared to that achieved by total‐Tau (90%), P‐Tau (62%) and 14‐3‐3 (91%).

In chemical looping combustion, solid metal oxide particles are utilized to transport oxygen from the air reactor to the fuel reactor. As fuel and air are never mixed, the energy penalty typically associated with gas separation in first-generation carbon capture and storage technologies can be avoided. To be considered as oxygen carrier for this process, a material should be reactive at relevant conditions, environmentally friendly, non-toxic, mechanically durable and have potential to be produced at low cost in large scale. Combined oxides of manganese and silicon have previously shown promise to meet these requirements. In this study, a spray-dried oxygen carrier based on a combined oxide of manganese, silicon and titanium was examined with respect to its performance in continuous chemical looping combustion of solid fuels. The experiments were carried out in a 10 kW chemical looping pilot unit which uses interconnected fluidized beds for oxygen carrier cycling. Prior to these experiments, the attrition rate was determined in a jet-cup rig. As the particles were comparably small and light, elutriation from the air reactor was high. The fuels used during a total experimental duration of 32 h were wood char, devolatilized hard coal, pet coke and lignite. In addition to varying fuels, the influence of fuel power, solids circulation and fuel reactor temperature were investigated. Gas conversion performance correlated clearly with the volatile content of the fuels, peaking at 97.8% for wood char and 94.6% for pet coke, which is the highest value ever reached for this particular fuel in this unit. Higher temperatures and solids circulation rates increased gas conversion. No decrease in performance over time, in particular no loss of reactivity due to sulphur accumulation, could be detected. The oxygen carrier released gaseous oxygen at relevant conditions. The particles were easily fluidized and fines production was low, suggesting a sufficient lifetime for the purpose.

Understanding the deformation styles and mechanisms which affect the Barberton Greenstone Belt (BGB) contributes to our ability to interprete the dynamic evolution of Paleoarchean greenstone belts and of the continental lithosphere. A variety of actualistic and non-actualistic, in part contradictory tectonic settings and processes (including gravitational collapse, terrane accretion, foreland basin shortening, continental rifting, doubly-vergent subduction and partial convective overturn) have been proposed to explain BGB deformation at a variety of scales. We compare these models and their regional 3-D predictions with emphasis on the BGB’s youngest deformed stratigraphic unit, the Moodies Group (ca. 3.21 Ga), also adding new structural data. We find that none of the tectonic models accounts for all major structural features of the BGB nor can any model adequately explain the regionally and temporally heterogeneous distribution of deformation styles within the greenstone belt. However, the partial convective overturn model, despite some weaknesses and oversimplifications, appears to be suited best to explain the geometry, orientation and formation of the dominating structural element of the BGB, the vertically-plunging Onverwacht Anticline (OA) with its 10-km-thick fold limbs. The formation and deformation of the adjacent smaller, commonly steeply doubly-plunging synclines filled with synorogenic siliciclastic sediment of the Fig Tree and Moodies groups could be partially explained as spatial adjustments to the rise and tilting of the OA although disparate processes acting at different scales need to be considered. In comparison to this large-scale vertical redistribution of mass by several tens of km, (sub-)horizontal displacements may have played a subordinate role at greenstone-belt scale, except along the regionally highly strained southern BGB margin in Eswatini.

Carbon capture and storage (CCS) offers the opportunity to avoid CO2 emissions from for example power plants and cement factories. Chemical-looping combustion (CLC) is one of the most promising capture technologies with potentially very low cost of CO2 capture. In this study we present findings from a solid-fuel 100 kW chemical-looping combustor. A new oxygen carrier - a sintered manganese ore called Sinaus - has been studied in the Chalmers 100 kW unit. The material has been investigated for an operational time of 51.5 h using five fuels: two bituminous coals, two types of wood char, and petcoke. The operational results clearly demonstrate the viability of the CLC process. In comparison to previously used iron-based oxygen carriers, the Sinaus material showed higher gas conversion - up to 88% - and lower loss of char to the air reactor, with carbon capture reaching as high as 100%. Furthermore, the solid-fuel conversion was higher, which is mainly an effect of the choice of fuel size. It was found that the choice of fuel has a crucial impact on performance. Previous experience has shown that the use of large fuel particles gives low carbon capture, whereas pulverized fuel leads to low solid-fuel conversion. By choosing the appropriate - intermediate - size of fuel, it is possible to combine high carbon capture with high solid-fuel conversion. Previous studies indicate that the drawback of many manganese ores is the mechanical stability. Hence, a lot of emphasis was put on an in-depth study of the lifetime of the Sinaus material. Analyzing the production rate of fines, it was found the expected lifetime of the Sinaus particles was 100-400 h. This is lower than what has been found for iron-based material, but most likely sufficient for operation in full-scale chemical-looping applications. Whilst the production of fines was highest during operation with fuel, a lot of fines were produced also during operation without fuel. Seven experiments without fuel, i.e when the observed mechanical degradation was only due to high-velocity impacts and not chemical stress caused by phase transformations, gave a lifetime in the interval 220-1230 h. In conclusion, this first-of-its-kind investigation shows that the lifetime of the oxygen carrier is related to both the change in oxygen-carrier conversion and high-velocity impacts.

All rights reserved., Synucleinopathies, such as Parkinson's disease and dementia with Lewy bodies, are neurodegenerative disorders that are characterized by the accumulation of α-synuclein (aSyn) in intracellular inclusions known as Lewy bodies. Prefibrillar soluble aSyn oligomers, rather than larger inclusions, are currently considered to be crucial species underlying synaptic dysfunction. We identified the cellular prion protein (PrPC) as a key mediator in aSyn-induced synaptic impairment. The aSyn-associated impairment of long-term potentiation was blocked in Prnp null mice and rescued following PrPC blockade. We found that extracellular aSyn oligomers formed a complex with PrPC that induced the phosphorylation of Fyn kinase via metabotropic glutamate receptors 5 (mGluR5). aSyn engagement of PrPC and Fyn activated NMDA receptor (NMDAR) and altered calcium homeostasis. Die Umsetzung von Prozessketten zur automatisierten Fertigung von 3D-Preforms im industriell etablierten Stempelumformverfahren ist aufgrund hoher Investitionskosten für kleine und mittlere Unternehmen bisher nicht wirtschaftlich tragbar. Die neuentwickelte Prozesskette wirkt dem entgegen und verspricht, komplexe 3D-Preforms bei geringer Prozesszeit sowohl textil- als auch lastgerecht herzustellen und dabei Ausschussquoten aufgrund von Drapierfehlern sowie den Verschnitt zu senken. Implementing process chains for the automated manufacturing of 3D textile preforms based on the established industrial stamp forming technology is not economical for small- and medium-sized enterprises due to high investment costs for small batch sizes and variable geometries. The new process chain counteracts by manufacturing complex 3D preforms at low processing times, both textile- and load-conform, while reducing scrap rates from draping errors and offcut.

Introduction: Human prion diseases are a heterogeneous group of incurable and debilitating conditions characterized by a progressive degeneration of the central nervous system. The conformational changes of the cellular prion protein and its formation into an abnormal isoform, spongiform degeneration, neuronal loss, and neuroinflammation are central to prion disease pathogenesis. It has been postulated that truncated variants of aggregation-prone proteins are implicated in neurodegenerative mechanisms. An increasing body of evidence indicates that proteolytic fragments and truncated variants of the prion protein are formed and accumulated in the brain of prion disease patients. These prion protein variants provide a high degree of relevance to disease pathology and diagnosis. Areas covered: In the present review, we summarize the current knowledge on the occurrence of truncated prion protein species and their potential roles in pathophysiological states during prion diseases progression. In addition, we discuss their usability as a diagnostic biomarker in prion diseases. Expert opinion: Either as a primary factor in the formation of prion diseases or as a consequence from neuropathological affection, abnormal prion protein variants and fragments may provide independent information about mechanisms of prion conversion, pathological states, or disease progression.

In line with the continuous growth rates in online retailing, the competitive situation for retailers has intensified. In order to strengthen their market position and exploit further potential, online retailers need to create a strong customer experience. The objective of the paper is to analyze the customer experience at the largest German online retailers with the aim of finding out whether the main four segments (marketplaces, generalists, fashion, consumer electronics) differ with respect to customer experience.

We present an approach for 3D urban scene reconstruction and interpretation based on the fusion of terrestrial and unmanned aerial vehicle (UAV) imagery. The terrestrial close range images acquired with high-resolution cameras reveal details of buildings, particularly of the facades. Yet, they have a poor coverage of the roofs and the ground due to the obtuse viewing angle. Thus, they are complemented by UAV imagery taken from larger distances using nadir and oblique views, with a clear view on the ground and the roofs. The resulting wide-baseline images are fused by a precise and reliable pose estimation approach and dense 3D point clouds including color are reconstructed. The colored 3D points are the input to semantic scene classification, which effectively fuses color and 3D geometric information. A set of “relative” features is proposed to provide an intra-class stable as well as inter-class discriminative description of objects. In comparison with the conventional “absolute” attributes, relative features provide context-sensitive measurements of both color and 3D geometry. The classification results for buildings are input to an automatic pipeline for level of detail 2 (LOD2) building model reconstruction combining a reliable scene as well as building decomposition with a subsequent primitive-based reconstruction and assembly. Finally, LoD3 models are obtained by integrating the results of facade image interpretation with an adapted convolutional neural network, which employs the 3D point cloud as well as the terrestrial images.

The Bcl-2-associated athanogene-1 (BAG1) exerts neuroprotective properties which has been shown in several studies of neurodegenerative disease models like Parkinson’s disease, Huntington’s disease and even cerebral ischemia. On the basis of the well-known neuroprotective function of the co-chaperone, we wanted to examine its properties in a model for Alzheimer’s disease, a neurological disorder of great significance. One of the hallmarks of Alzheimer’s disease, besides extracellular plaque formation, is the intra-neuronal accumulation of hyper-phosphorylated tau protein that leads to tau aggregation. When overexpressed together with a tau mutant with high propensity for aggregation, BAG1 led to the stabilization of high-molecular tau fragments in rat CSM 14.1 cells compared to wild-type cells. Deletion of the domain in BAG1 that is responsible for binding to Hsp70 (BAGΔC) abolished this effect, which could be confirmed by immunocytochemistry. In fact, BAG1 does not only increase mutant tau aggregation but also prevents its degradation by the proteasome. Immunochemistry revealed that overexpression of the Bcl-2-associated athanogene-1 gives rise to large tau aggregates surrounded by lysosomes. Furthermore, toxicity assays indicated increased tau toxicity in BAG1 overexpressing cells. Hence, in contrast to other neurodegenerative diseases, BAG1 seems to enhance Alzheimer´s pathology and to promote cell death due to the stabilization of aggregation-prone tau species that evade proteasomal clearance. To conclude, this analysis provides a new sight of the co-chaperone BAG1 and yet again demonstrates its complex influence in a model of Alzheimer’s disease.

Objectives The main objective of the present study is to study the therapeutic efficiency of doxycycline in a double-blinded randomised phase II study in a cohort of patients with sporadic Creutzfeldt-Jakob disease (sCJD). Methods From the National Reference Center of TSE Surveillance in Germany, patients with probable or definite sCJD were recruited for a double-blinded randomised study with oral doxycycline (EudraCT 2006-003934-14). In addition, we analysed the data from patients with CJD who received compassionate treatment with doxycycline in a separate group. Potential factors which influence survival such as age at onset, gender, codon 129 polymorphism and cognitive functions were evaluated. The primary outcome measure was survival. Results Group 1: in the double-blinded randomised phase II study, 7 patients in the treatment group were compared with 5 controls. Group 2: 55 patients with sCJD treated with oral doxycycline were analysed and compared with 33 controls by a stratified propensity score applied to a Cox proportional hazard analysis. The results of both studies were combined by means of a random-effects meta-analysis. A slight increase in survival time in the doxycycline treatment group was observed (p=0.049, HR=0.63 (95% CI 0.402 to 0.999)). Conclusions On the basis of our studies, a larger trial of doxycycline should be performed in persons in the earliest stages of CJD. Trial registration number EudraCT 2006-003934-14; Results.

Prion diseases in humans are neurodegenerative diseases which are caused by an accumulation of abnormal, misfolded cellular prion protein known as scrapie prion protein (PrPSc). Genetic, acquired, or spontaneous (sporadic) forms are known. Pathogenic mutations in the human prion protein gene (PRNP) have been identified in 10-15 % of CJD patients. These mutations may be single point mutations, STOP codon mutations, or insertions or deletions of octa-peptide repeats. Some non-coding mutations and new mutations in the PrP gene have been identified without clear evidence for their pathogenic significance. In the present review, we provide an updated overview of PRNP mutations, which have been documented in the literature until now, describe the change in the DNA, the family history, the pathogenicity, and the number of described cases, which has not been published in this complexity before. We also provide a description of each genetic prion disease type, present characteristic histopathological features, and the PrPSc isoform expression pattern of various familial/genetic prion diseases.

In this paper we present an approach for semantic interpretation of facade images based on a Convolutional Network. Our network processes the input images in a fully convolutional way and generates pixel-wise predictions. We show that there is no need for large datasets to train the network when transfer learning is employed, i. e., a part of an already existing network is used and fine-tuned, and when the available data is augmented by using deformed patches of the images for training. The network is trained end-to-end with patches of the images and each patch is augmented independently. To undo the downsampling for the classification, we add deconvolutional layers to the network. Outputs of different layers of the network are combined to achieve more precise pixel-wise predictions. We demonstrate the potential of our network based on results for the eTRIMS (Korč and Förstner, 2009) dataset reduced to facades.

Chemical looping combustion (CLC) and chemical looping with oxygen uncoupling (CLOU) are carbon capture technologies which achieve gas separation by means of cycling oxidation and reduction of a solid oxygen carrier. In this study, the performance and CLOU properties of an oxygen carrier with perovskite structure, CaMn0.9Mg0.1O3−δ, were investigated in a 10 kW pilot. The fuel consisted of biochar with very low sulphur content. Around 37 h of operation with fuel were carried out in the 10 kW chemical looping combustor. Previous operational experience in this unit has been achieved using different natural minerals as oxygen carrier – mainly ilmenite and manganese ore. Parametric studies performed in this work included variation of fuel flow, solids circulation rate, temperature and fluidization gas in the fuel reactor. The oxygen carrier was exposed to a total 73 h of hot fluidization (T > 600 °C). No hard particle agglomerations were formed during the experiments. An oxygen demand as low as 2.1% could be reached under stable operating conditions, with a carbon capture efficiency of up to 98%. CLOU properties were observed at all fuel reactor temperatures, ensuring stable operation even without steam as gasification agent present in the fuel reactor. The results suggest that CaMn0.9Mg0.1O3−δ is suitable for the use as oxygen carrier in chemical looping combustion of solid biochar and offers higher gas conversion than previously tested materials without CLOU properties such as ilmenite. The experiments constitute the first successful operation using a non-copper CLOU oxygen carrier in combustion of solid fuel.

Neurodegenerative diseases with abnormal protein aggregates such as Alzheimer's disease, tauopathies, synucleinopathies, and prionopathies, together with vascular encephalopathies, are cause of cognitive impairment and dementia. Identification of reliable biomarkers in biological fluids, particularly in the cerebrospinal fluid (CSF), is of extreme importance in optimizing the precise early clinical diagnosis of distinct entities and predicting the outcome in particular settings. In addition, the study of CSF biomarkers is useful to identify and monitor the underlying pathological processes developing in the central nervous system of affected individuals. Evidence suggests that levels of key CSF molecules correlate, in some circumstances, with prediction, disease progression, and severity of cognitive decline. Correlation of CSF markers and underlying pathological molecular substrates in brain is an exciting field for further study. However, while some dementias such as Creutzfeldt-Jakob disease have accurate CSF biomarkers, other disease types such as dementia with Lewy bodies, vascular dementia, and frontotemporal dementia lack reliable biomarkers for their specific clinical diagnosis.

Chemical-looping combustion (CLC) is a novel carbon-capture technology with potential to drastically reduce the cost of CO2 capture. Relying on interconnected fluidized bed technology, CLC systems can achieve CO2 capture by using oxygen carrying bed material. This so-called oxygen carrier transports oxygen from combustion air to fuel, thus making carbon capture inherent to the CLC process. In this study, we present findings from a 100 kW chemical-looping combustor for solid fuels. The 100 kW unit uses the dual-CFB concept, where both air reactor and fuel reactor are designed as circulating fluidized beds. The oxygen carrier material used in this study consisted of a mixture of ilmenite - which has been used in several studies in CLC - and a manganese ore. Previous studies have shown that gas conversion can be significantly increased by using manganese ore particles as oxygen carrier. However, previous testing has also shown that the production of fines, i.e. particle attrition, may be high when using manganese ore. The reason for mixing the two materials is thus to obtain an oxygen carrying material that has high reactivity, and yet does not produce too much fines during fuel operation. The 100 kW unit was operated in total for 18 h with fuel. Three fuels were used in the experiments: two bituminous coals and wood char. Gas conversion was high, and increased with increasing fraction of manganese ore in the oxygen-carrier mixture. At the end of the experiments, the fraction of manganese ore in the bed material was approximately 8%, which also was the highest fraction during all tests. The mixture of ilmenite and manganese ore gave significant improvements in gas conversion in comparison to only ilmenite. The highest gas conversion observed during testing with bituminous coal was 91.5%, as compared to 84% with only ilmenite as oxygen carrier during similar conditions in the 100 kW unit. These test results indicate that the addition of manganese ore could almost halve the fraction of unconverted gas. Thus, mixing mechanically stable ilmenite with more reactive manganese ore can give reductions in costs as compared to using manganese ore only, and still give significantly reduced oxygen demand as compared to ilmenite. In the present case - mixing of ilmenite and manganese ore - the high reactivity was also possible to combine with improved operability of the material, primarily manifested as lower production rate of fines. Along with the rapid development of autonomous vehicles and driving assistance systems, suitable maps have been intensively studied in recent years. Besides the improvement of conventional maps, new types such as high-density (HD) maps, have been introduced to provide comprehensive detailed information particularly for autonomous driving. In the areas which are not or not well covered by these maps, however, the autonomous vehicles are basically on their own. I.e., besides GNSS signals, they have to rely solely on the onboard sensors with local measurements. In this paper, we present an alternative pipeline for map generation from UAV imagery. A map particularly suitable for autonomous driving is derived from the 3D scene reconstructed from high-resolution images. In addition to basic geometry and semantic features, the map contains abstract vertical landmarks for fast and accurate positioning and path planning, elevation as well as slope information for driving, and trafficability information for different types of vehicles in off-road areas. The potential of the proposed approach is demonstrated based on the results of an experiment.

Sporadic Creutzfeldt-Jakob disease (sCJD) is the most common form of human prion disease. It is invariably fatal and displays a short clinical disease stage. The key event in sCJD is the propagation of a beta-sheet rich conformer of the physiological PrPC protein, known as PrPSc. Neuropathological disease characteristics include gliosis, neuronal loss and spongiform degeneration; disease clinical manifestations refer to mental and visual disabilities, cognitive impairment, gait or limb ataxia, myoclonus and mutism. Definite sCJD diagnosis requires post-mortem brain material histopathological examination. However, highly certain pre-mortem differential diagnosis is desired to exclude other treatable disorders and to reduce disease transmission risks. Detection and/or quantification of cerebrospinal fluid (CSF) biomarkers reflecting neuronal damage and PrPC misfolding in the diseased brain significantly enhance pre-mortem diagnosis. Previously established and newly identified biomarkers are used towards this direction. Increased CSF Neurofilament light chain (NFL) concentrations have been reported in several neurological disorders, including prion diseases. In the present study, we analyzed CSF NFL levels in two independent patient cohorts, consisting of highly suspected sCJD cases that were further classified as sCJD or non-CJD according to established diagnostic criteria. CSF NFL concentrations were increased in sCJD compared to non-CJD cases in both cohorts (area under the curve (with 95% confidence interval) equal to 0.89 (0.82 to 0.97) and 0.86 (0.77 to 0.96), respectively. CSF NFL was associated neither to age nor to sex but correlated with total-tau concentrations in both cohorts. Overall, our data provide independent validation of CSF NFL utility in sCJD differential diagnosis.

Cerebrospinal fluid (CSF) total prion protein (t-PrP) is decreased in sporadic Creutzfeldt-Jakob disease (sCJD). However, data on the comparative signatures of t-PrP across the spectrum of prion diseases, longitudinal changes during disease progression, and levels in pre-clinical cases are scarce. T-PrP was quantified in neurological diseases (ND, n = 147) and in prion diseases from different aetiologies including sporadic (sCJD, n = 193), iatrogenic (iCJD, n = 12) and genetic (n = 209) forms. T-PrP was also measured in serial lumbar punctures obtained from sCJD cases at different symptomatic disease stages, and in asymptomatic prion protein gene (PRNP) mutation carriers. Compared to ND, t-PrP concentrations were significantly decreased in sCJD, iCJD and in genetic prion diseases associated with the three most common mutations E200K, V210I (associated with genetic CJD) and D178N-129M (associated with fatal familial insomnia). In contrast, t-PrP concentrations in P102L mutants (associated with the Gerstmann-Sträussler-Scheinker syndrome) remained unaltered. In serial lumbar punctures obtained at different disease stages of sCJD patients, t-PrP concentrations inversely correlated with disease progression. Decreased mean t-PrP values were detected in asymptomatic D178-129M mutant carriers, but not in E200K and P102L carriers. The presence of low CSF t-PrP is common to all types of prion diseases regardless of their aetiology albeit with mutation-specific exceptions in a minority of genetic cases. In some genetic prion disease, decreased levels are already detected at pre-clinical stages and diminish in parallel with disease progression. Our data indicate that CSF t-PrP concentrations may have a role as a pre-clinical or early symptomatic diagnostic biomarker in prion diseases as well as in the evaluation of therapeutic interventions.
