Aims Cereblon (CRBN) is a substrate receptor of the E3 ubiquitin ligase complex that was reported to target ion channel proteins. L-type voltage-dependent Ca2+ channel (LTCC) density and dysfunction is a critical player in heart failure with reduced ejection fraction (HFrEF). However, the underlying cellular mechanisms by which CRBN regulates LTCC subtype Cav1.2α during cardiac dysfunction remain unclear. Here, we explored the role of CRBN in HFrEF by investigating the direct regulatory role of CRBN in Cav1.2α activity and examining how it can serve as a target to address myocardial dysfunction. Methods and results Cardiac tissues from HFrEF patients exhibited increased levels of CRBN compared with controls. In vivo and ex vivo studies demonstrated that whole-body CRBN knockout (CRBN−/−) and cardiac-specific knockout mice (Crbnfl/fl/Myh6Cre+) exhibited enhanced cardiac contractility with increased LTCC current (I CaL) compared with their respective controls, which was modulated by the direct interaction of CRBN with Cav1.2α. Mechanistically, the Lon domain of CRBN directly interacted with the N-terminal of Cav1.2α. Increasing CRBN levels enhanced the ubiquitination and proteasomal degradation of Cav1.2α and decreased I CaL. In contrast, genetic or pharmacological depletion of CRBN via TD-165, a novel PROTAC-based CRBN degrader, increased surface expression of Cav1.2α and enhanced I CaL. Low CRBN levels protected the heart against cardiomyopathy in vivo. Conclusion Cereblon selectively degrades Cav1.2α, which in turn facilitates cardiac dysfunction. A targeted approach or an efficient method of reducing CRBN levels could serve as a promising strategy for HFrEF therapeutics.

Wormlike micelles (WLMs), which are the self-assemblies of amphiphilic surfactants, have gained attention in various applications due to their unique ‘living’ nature (i.e., union-scission). However, the industrial production of the WLMs has rarely been discussed. Moreover, given that the system hardly reaches the desired viscosity without appropriate control actions within a reasonable operation time, a model-based controller has been implemented to the production of WLMs where CTAB/NaCl in a semi-batch reactor is used as a case study. For this, a high-fidelity model was developed by integrating thermodynamic, kinetic, and rheology models and validated with experimental results. Subsequently, to alleviate the computational burden of such a high-fidelity model, the model reduction was carried out by the multivariable output error state space algorithm. The developed reduced-order model (ROM) was able to reproduce the essential features of the high-fidelity model and utilized in the formulated model predictive control (MPC) system. Herein, the manipulated variables (i.e., surfactant volume fraction, salt concentration, and temperature) are adjusted to give the desired WLM length of 3700 nm (i.e., a zero-shear viscosity of 100 Pa-s). Finally, the developed MPC scheme resulted in optimal input sequences to successfully reach the setpoint within practical constraints.

Background Little is known about the self-perceived level of disability of stroke survivors in the community. We aimed to characterise Health-related quality of life (HRQoL) 1 year after stroke and investigate how sociodemographic and stroke-related factors and medical adherence explain the self-perceived level of disability in a Korean stroke population. Methods This was a multicentre cross-sectional study. A total of 382 ischaemic stroke survivors at 1 year after onset from 11 university hospitals underwent a one-session assessment, including socioeconomic variables, the modified Rankin Scale (mRS), various neurological sequelae, the Morisky, Green and Levin-Medication Adherence Questionnaire (MGL), and the World Health Organization Disability Assessment Schedule 2.0 (WHODAS 2.0) 36-items. The relationship between disability and different variables was analysed using ordinal logistic regression. Results The prevalence of disability based on global WHODAS 2.0 was 62.6% (mild, 41.6%; moderate, 16.0%; severe, 5.0%). The prevalence of severe disability was higher in participation in society (16.8%) and getting around (11.8%) than in other domains. Low MGL- motivation was the only factor determining a significant association between all six domains of disability after adjustment. Different predictors for specific domains were age, mRS, dysarthria, trouble seeing, cognition problems, and MGL-motivation for understanding and communicating; age, recurrent stroke, mRS, hemiplegia, facial palsy, general weakness, and MGL-motivation for getting around; age, education, mRS, hemiplegia, and MGL-motivation for self-care; education, recurrent stroke, hemiplegia, dysarthria, and MGL-motivation for getting along with people; age, education, income, mRS, hemiplegia, dysarthria, MGL-knowledge, and MGL-motivation for life activities; living without a spouse, mRS, hemiplegia, dysarthria, trouble seeing, cognition problems, general weakness, and MGL-motivation for participation in society. Conclusions Self-perceived disability according to the WHODAS 2.0 at 1 year after stroke was highly prevalent. Each disability domain showed a different prevalence and associated factors. Interventions promoting medical adherence to motivation seemed to help achieve high HRQoL in all domains.

Objectives: Insomnia is one of the most common sleep disorders and is difficult to completely treat because of the undesirable side effects of hypnotics. The present study was designed to investigate the hypnotic effect of acupuncture stimulation at HT7 on caffeine-induced sleep disorders and locomotor activity in rats. We also evaluated neuronal activity changes in the arousal region of the basal forebrain. Methods: Rats received intraperitoneal injections of caffeine, and then electroencephalogram power spectrum analysis and locomotor activity measurements were performed. Stimulation at HT7 was performed using a mechanical acupuncture instrument (MAI) before caffeine injection, and its effects on caffeine-induced changes in sleep architecture, locomotor activity and c-Fos expression were examined. Results: Caffeine injection (7.5 mg/kg) produced a significant decrease in slow-wave sleep and an increase in wake time compared with saline injection. Caffeine injection also increased locomotor activity and c-Fos expression in the medial septum–vertical limb of the diagonal band of Broca (MS-VDB), one of the arousal regions of the basal forebrain. Stimulation at HT7 with the MAI alleviated the caffeine-induced sleep disturbance and the increase in locomotor activity. In addition, MAI treatment at HT7, compared with treatment at a location not corresponding to any traditional acupuncture point, reduced the caffeine-induced increase in c-Fos expression. Conclusion: These results indicate that the hypnotic effect of HT7 acupuncture stimulation on caffeine-induced insomnia was associated with suppression of neuronal activity in the basal forebrain.

Bile acid-modified nanoparticles provide a convenient strategy to improve oral bioavailability of poorly permeable drugs by exploiting specific interactions with bile acid transporters. However, the underlying mechanisms are unknown, especially considering the different absorption sites of free bile acids (ileum) and digested fat molecules from bile acid-emulsified fat droplets (duodenum). Here, glycocholic acid (GCA)-conjugated polystyrene nanoparticles (GCPNs) are synthesized and their transport in Caco-2 cell models is studied. GCA conjugation enhances the uptake by interactions with apical sodium-dependent bile acid transporter (ASBT). A new pathway correlated with both ASBT and chylomicron pathways is identified. Meanwhile, the higher uptake of GCPNs does not lead to higher transcytosis to the same degree compared with unmodified nanoparticles (CPNs). The pharmacological and genomics study confirm that GCA conjugation changes the endocytosis mechanisms and downregulates the cellular response to the transport at gene levels, which works as a negative feedback loop and explains the higher cellular retention of GCPNs. These findings offer a solid foundation in the bile acid-based nanomedicine design, with utilizing advantages of the ASBT-mediated uptake, as well as inspiration to take comprehensive consideration of the cellular response with more developed technologies.

ATPase inhibitory factor 1 (IF1) is an ATP synthase–interacting protein that suppresses the hydrolysis activity of ATP synthase. In this study, we observed that the expression of IF1 was up-regulated in response to electrical pulse stimulation of skeletalmuscle cells and in exercized mice and healthy men. IF1 stimulates glucose uptake viaAMPKin skeletalmuscle cells and primary culturedmyoblasts.Reactive oxygen species and Rac family small GTPase 1 (Rac1) function in the upstream and downstream of AMPK, respectively, in IF1-mediated glucose uptake. In diabetic animal models, the administration of recombinant IF1 improved glucose tolerance and down-regulated blood glucose level. In addition, IF1 inhibitsATPhydrolysis byb-F1-ATPase inplasmamembrane, thereby increasing extracellular ATP and activating the protein kinase B (Akt) pathway, ultimately leading to glucose uptake. Thus, we suggest that IF1 is a novel myokine and propose amechanismby which AMPK and Akt contribute independently to IF1-mediated improvement of glucose tolerance impairment. These results demonstrate the importance of IF1 as a potential antidiabetic agent.

Background Mast cells are a key effector cell in the pathogenesis of chronic spontaneous urticaria (CSU) and activated by circulating FcεRI-specific IgG as well as IgE. This study evaluated the prevalence of circulating autoantibodies to FcεRIα in the sera of CSU patients. Methods Eighty-eight patients with CSU and 76 healthy controls (HCs) were enrolled. To detect circulating autoantibodies (IgG/IgA/IgM) to FcεRIα, ELISA was done using YH35324 (as a solid phase antigen), and its binding specificity was confirmed by the ELISA inhibition test. The antibody levels were presented by the ratio of YH35324-preincubated to mock-preincubated absorbance values. Clinical and autoimmune parameters, including atopy, urticaria activity score (UAS), serum total/free IgE levels, serum antinuclear antibody (ANA) and autologous serum skin test (ASST) results, were assessed. The autoimmune group was defined if CSU patients had positive results to ASST and/or ANA. Results The ratio of serum IgG to FcεRIα was significantly lower in CSU patients than in HCs (P<0.05), while no differences were noted in serum levels of IgG to recombinant FcεRIα or IgA/IgM autoantibodies. The autoimmune CSU group had significantly lower ratios of IgG/IgA (not IgM) autoantibodies to FcεRIα than the nonautoimmune CSU group (P Conclusions This study confirmed the presence of IgG to FcεRIα in the sera of CSU patients, especially those with the autoimmune phenotype.

We evaluated the effect of manganese ferrite nanoparticles (MFN) on radiosensitization and immunologic responses using the murine hepatoma cell line Hepa1-6 and the syngeneic mouse model. The clonogenic survival of Hepa1-6 cells was increased by hypoxia, while being restricted by ionizing radiation (IR) and/or MFN. Although MFN suppressed HIF-1α under hypoxia, the combination of IR and MFN enhanced apoptosis and DNA damage in Hepa1-6 cells. In the Hepa1-6 syngeneic mouse model, the combination of IR and MFN notably limited the tumor growth compared to the single treatment with IR or MFN, and also triggered more frequent apoptosis in tumor tissues than that observed under other conditions. Increased expression of PD-L1 after IR was not observed with MFN alone or the combination of IR and MFN in vitro and in vivo, and the percentage of tumor-infiltrating T cells and cytotoxic T cells increased with MFN, regardless of IR, in the Hepa1-6 syngeneic mouse model, while IR alone led to T cell depletion. MFN might have the potential to overcome radioresistance by alleviating hypoxia and strengthening antitumor immunity in the tumor microenvironment.

Hybrid organic-inorganic lead halide perovskites have attracted much attention in the field of optoelectronic devices because of their desirable properties such as high crystallinity, smooth morphology, and well-oriented grains. Recently, it was shown that thermal nanoimprint lithography (NIL) is an effective method not only to directly pattern but also to improve the morphology, crystallinity, and crystallographic orientations of annealed perovskite films. However, the underlining mechanisms behind the positive effects of NIL on perovskite material properties have not been understood. In this work, we study the kinetics of perovskite grain growth with surface energy calculations by first-principles density functional theory (DFT) and reveal that the surface energy-driven preferential grain growth during NIL, which involves multiplex processes of restricted grain growth in the surface-normal direction, abnormal grain growth, crystallographic reorientation, and grain boundary migration, is the enabler of the material quality enhancement. Moreover, we develop an optimized NIL process and prove its effectiveness by employing it in a perovskite light-emitting electrochemical cell (PeLEC) architecture, in which we observe a fourfold enhancement of maximum current efficiency and twofold enhancement of luminance compared to a PeLEC without NIL, reaching a maximum current efficiency of 0.07598 cd/A at 3.5 V and luminance of 1084 cd/m

Virtual Reality (VR) has been adopted as a leading technology for the metaverse, yet most previous VR systems provide one-size-fits-all experiences to users. Context-awareness in VR enables personalized experiences in the metaverse, such as improved embodiment and deeper integration of the real world and virtual worlds. Personalization requires context data from diverse sources. We proposed a reusable and extensible context data collection framework, ManySense VR, which unifies data collection from diverse sources for VR applications. ManySense VR was implemented in Unity based on extensible context data managers collecting data from data sources such as an eye tracker, electroencephalogram, pulse, respiration, galvanic skin response, facial tracker, and Open Weather Map. We used ManySense VR to build a context-aware embodiment VR scene where the user’s avatar is synchronized with their bodily actions. The performance evaluation of ManySense VR showed good performance in processor usage, frame rate, and memory footprint. Additionally, we conducted a qualitative formative evaluation by interviewing five developers (two males and three females; mean age: 22) after they used and extended ManySense VR. The participants expressed advantages (e.g., ease-of-use, learnability, familiarity, quickness, and extensibility), disadvantages (e.g., inconvenient/error-prone data query method and lack of diversity in callback methods), future application ideas, and improvement suggestions that indicate potential and can guide future development. In conclusion, ManySense VR is an efficient tool for researchers and developers to easily integrate context data into their Unity-based VR applications for the metaverse.

One major hurdle in the progress of hyperbolic metamaterials (HMMs) is their lossy nature due to the metal constituent. In this work, we design a gain-assisted active HMM utilizing the recently emerged, solution-processed perovskite gain material. Our HMM is consisted of MAPbI3 perovskite and Au subwavelength multi-layer. We theoretically and experimentally investigate the strong emission polarization anisotropy that is unique to HMMs. Our work opens the way towards applications such as high-speed light emission, super resolution imaging and lithography, electro-optical modulators and perfect light absorbers.

Background: IgE bound on the surface of mast cells contributes to the pathogenesis of chronic spontaneous urticaria (CSU). Atopy is a predisposing factor for CSU, where omalizumab is a widely used monoclonal antibody to control urticaria symptoms via capturing serum free IgE. However, the role of serum free IgE is not clarified in CSU. The present study evaluated the clinical relevance of serum free IgE in patients with CSU. Methods: Eighty-eight patients with CSU and 76 healthy controls (HCs) were enrolled in this study. Serum total and Dermatophagoides pteronyssinus (Der p)-specific IgE levels were measured by ImmunoCAPs. The serum free IgE levels were measured by ELISA using a novel IgETRAP, and their associations with clinical parameters, including urticaria activity score (UAS), were evaluated. Background and purpose Although we have recently demonstrated that spinal astrocyte gap junctions mediate the development of mirror-image pain (MIP), it is still unclear which astrocyte-derived factor is responsible for the development of MIP and how its production is controlled. In the present study, we focused on the role of ipsilateral versus contralateral D-serine in the development of MIP and investigated the possible involvement of σ1 receptors and gap junctions in astrocyte D-serine production. Experimental approach Following carrageenan injection, mechanical allodynia was tested at various time points to examine the effect of individual drugs. Immunohistochemistry and Western blot analyses were performed to clarify the expression levels of spinal D-serine, serine racemase, σ1 receptors and connexin 43. Key results The expression of ipsilateral D-serine was up-regulated during the early phase of inflammation, while contralateral D-serine increased during the later phase of inflammation. The pharmacological inhibition of D-serine during the early phase blocked the development of both ipsilateral and contralateral mechanical allodynia. However, the inhibition of D-serine during the later phase of inflammation blocked contralateral, but not ipsilateral mechanical allodynia. Furthermore, the inhibition of σ1 receptors during the earlier phase of inflammation inhibited the increase in ipsilateral D-serine. Conversely, the blockade of astrocyte gap junctions suppressed the up-regulation of contralateral D-serine during the later phase of inflammation. Conclusion and implications Spinal astrocyte D-serine plays an important role in the development of mirror-image pain. Furthermore, σ1 receptors and astrocyte gap junction signalling mediate ipsilateral and contralateral D-serine production respectively.

As the development of shale gas resources has to be accelerated to satisfy the increasing global natural gas demand, many optimization approaches have been developed to handle the complexity associated with the shale gas system and discern the optimal design and operation strategy. However, it is worth mentioning that the essential hydraulic fracturing operation has not been considered as a dynamic process in the previous studies, which may lead to suboptimal solutions. Motivated by this consideration, we first develop an integrated model to simultaneously consider the scheduling and control of hydraulic fracturing operations to enhance the economic performance of the shale gas system. In particular, a reduced-order model is developed and integrated with the scheduling model to reduce the model complexity. To cope with the plant-model mismatch of the reduced-order model, we propose an online integrated framework with two feedback loops. In the outer loop, the integrated model is solved to determine the scheduling decisions and controller references. Then, an offset-free model predictive control system is designed to track the references online in the inner loop. By utilizing the offset-free model predictive control scheme, the tracking performance is enhanced and the plant-model mismatch is compensated for, which helps avoid dramatic changes in the solutions obtained by re-solving the integrated problem online. The effectiveness of the proposed online integrated framework is demonstrated by considering a hypothetical case study based on Marcellus Shale Play. It shows that the undesirable performance degradation induced by the plant-model mismatch can be decreased by the developed offset-free model predictive control system, and the overall economic performance of the shale gas system is improved with the proposed approach.

Objectives: Health coaching became effective tool for management of chronic diseases. This study was designed to investigate the effects of coaching program on comprehensive lifestyles modification with Type 2 Diabetes Mellitus(T2DM) patients. Methods: The study investigated using pre- & post-test. Diabetes health coaching program was taken at branches of community health center for 6 months. This program was consisted of monthly face to face counseling and weekly telephone counseling. At enrollment and after 6 months, we performed a questionnaire survey consisted of the diabetes knowledge, practice of disease management (diet, blood sugar testing, and foot care), self-efficacy, subjective health status, health behavior (smoking, alcohol consumption, and physical activity). Also we checked blood pressure, body mass index, fasting serum glucose, postprandial glucose, total cholesterol, and glycosylated hemoglobin. Results: We analyzed 6-month follow-up data from 54 patients. They showed significant improvements in diabetes knowledge, disease management of foot care, self efficacy, the amount of exercise, and systolic blood pressure. Conclusions: Although this study didn’t show metabolic improvement of T2DM patients, it resulted the improvement of self efficacy, the amount of exercise, and systolic blood pressure. Using Diabetes health coaching program seems to be helpful in T2DM patients at branch of community health center.

Although we have recently demonstrated that carrageenan-induced inflammation upregulates the expression of spinal interleukin (IL)-1β, which inhibits spinal astrocyte activation and results in the delayed development of Mirror-Image Pain (MIP), little is known regarding the mechanisms that underlie how spinal IL-1β inhibits the astrocyte activation. In this study, we examined the effect of spinal IL-1β on astrocyte gap junctions (GJ) and the development of MIP. Following unilateral carrageenan (CA) injection, mechanical allodynia (MA) was evaluated at various time points. Immunohistochemistry and Western blot analysis were used to determine changes in the expression of GFAP and connexins (Cx) in the spinal cord dorsal horn. Carrageenan rats showed a delayed onset of contralateral MA, which mimicked the temporal expression pattern of spinal Cx43 (an astrocyte gap junctional protein) and GFAP. Intrathecal administration of an interleukin-1 receptor antagonist (IL-1ra) twice-a-day on post-carrageenan injection days 0 to 3 caused a significant increase in contralateral MA and spinal Cx43 and GFAP expression. In addition, co-administration of IL-1β with IL-1ra blocked the IL-1ra-induced increase in contralateral MA and the upregulated expression of spinal Cx43 and GFAP. Finally, co-administration of carbenoxolone (CBX; a GJ decoupler) or Gap26 (a specific Cx43 mimetic blocking peptide) with IL-1ra significantly blocked the IL-1ra-induced early development of contralateral MA and the associated upregulation of spinal Cx43 and GFAP expression. These results demonstrate that spinal IL-1β suppresses Cx43 expression and astrocyte activation during the early phase of CA-induced inflammation resulting in the delayed onset of contralateral MA. These findings imply that spinal IL-1β can inhibit astrocyte activation and regulate the time of induction of contralateral MA through modulation of spinal Cx43 expression.

The fight against climate change and environmental pollution have set the agenda for efficient reaction systems design. It has necessitated now more than ever, the careful design and optimization of chemical processes that result in carbon production. In this study, a unified framework for the mathematical modelling and simulation, prediction, and optimization of reaction systems is proposed. A computational fluid dynamics (CFD) simulation of butadiene synthesis over a ferrite catalyst in a 3D shell and multi-tubular reactor was executed. A rigorous mathematical formulation of the process kinetics and multiscale heat transfer was incorporated into an OpenFOAM CFD model using a porous media. The CFD model was validated against experimental data using gas concentrations, temperature, and CO2 partial pressure with a maximum error of 3.2% (97% accuracy). The developed model was then used to generate data sets for both prediction and optimization of the reaction conditions in terms of temperature, flow rate, and feed compositions using Deep Neural Network (DNN) and Genetic Algorithm (GA). Another surrogate model was developed for temperature control (cooling side) optimization by utilizing the coolant flow rate, flow directions (co-current and countercurrent), coolant types (water and solar salt), and velocity. Dynamic evolution and steady-state contours of the species concentration distributions were predicted with our DNN model with only an error of 1.26% (98.74% accuracy). Our results clearly indicated that higher reactor performance indices that exceed those of the conventional optimization approach in terms of conversion (99.98%), yield (93.28%), and selectivity (92.3%) are obtainable with the proposed framework. The mathematical models, DNN surrogate models, and the genetic algorithms that were integrated as a unified framework can be adopted for designing other reactor systems.

Many industrial applications adopt solar salt as the go-to heat transfer fluid for both cooling and thermal storage systems. However, the high melting temperature of this salt makes it economically unattractive. Hence, in this study, the investigation of 12 different molten salts was carried out to provide the right decision basis for selecting suitable salts for both cooling and thermal storage applications. In light of this, we developed a computational fluid dynamics (CFD) model to simulate butadiene synthesis via the oxidative dehydrogenation of butene in a single tube reactor equipped with a cooling jacket. The CFD model was validated against experimental results with an average error of 3.5%. Parametric studies involving the different salts indicated that low melting point salts resulted in high heat removal efficiencies but reduced the overall conversion, yield, and selectivity of the reactor due to the lower reaction temperatures. Thermophysical properties such as specific heat capacities were found to have no significant impact on the cooling efficiency of the reactor. An economic evaluation of the salts based on their heating costs indicated that salts of LiNaKCsCaNO3 and NaKCaNO3 are most suitable, especially for thermal storage systems where only latent heat of fusion is required. However, in cases where further heating of the salt is needed, higher melting point salts (such as solar salt) proved to be much more economical. Investigation of the effect of different tube diameters on the cooling performance of the reactor indicated that reactors having a diameter to length ratio between 32 and 58 provide sufficient cooling and high conversion.

Background: The novel threadlike structure is called the primo structure, and studies are conducted through many different approaches. Although various ways of differentiation are currently used, a standard for differentiation is deemed necessary in order to identify the primo structure based on the overall form of the structure. This study aims to explore the differentiation standard through the histological common point of the threadlike structure of rat and swine by using the hematoxylin and eosin (H&E) staining method commonly used for histological research in various structures. Methods: An 8-week old Sprague-Dawley rat and a Yorkshire pig weighing 30–40 kg were used. A total of 65 pieces of rat threadlike structure and 100 pieces of swine threadlike structure were collected after the abdomen was cut and opened. The following three different characteristics were confirmed using the H&E staining method for the collected structures: (1) bright cell availability; (2) cavity availability; and (3) nuclei density. Results: For the rat threadlike structure, the bright cell (70.5%) and nuclei density (92.6%) were mainly observed; in the swine threadlike structure, however, the bright cell (60.6%) and cavity (67.2%) were mainly observed. The bright cell was confirmed to have been observed in the threadlike structures of both rat and swine. Conclusion: The bright cell is determined to be the common point in the primo structure. However, further research is deemed necessary in the future as to the functions performed by the characteristic shown by the Primo structure.

We report the first directly patterned perovskite distributed feedback (DFB) resonator with a narrow amplified spontaneous emission (ASE) at pump powers as low as 0.1W/cm2, under continuous-wave (CW) optical pumping condition at room temperature.
