5. Do chromosomal inversion carriers really need preimplantation genetic testing?

This study aimed to evaluate the rates of euploidy, aneuploidy, and mosaicism in preimplantation genetic testing for structural rearrangements (PGT-SR) cycles from chromosomal inversion carriers. In addition, this work also focused on assessing the impact of some contributors on the incidence of parental originating aneuploidy and mosaicism.This retrospective review enrolled chromosomal inversion carrier couples of whom the females were under 38 years old undergoing PGT-SR at a single academic reproductive center. Subgroups were divided according to the gender of carriers, the inversion type, and the semen parameters of male carriers (male factor infertility (MF) or non-MF). Patient demographics, cycle characteristics, and PGT-SR outcomes were compared among subgroups.A total of 71 PGT-SR cycles from 57 inversion carrier couples were included for analysis. Among the 283 blastocysts, 48.4% were identified as euploidy, 27.9% as aneuploidy, and the remaining 23.7% as mosaicism. Only 32.9% of aneuploid embryos and 1.5% of mosaic embryos involved the parental inversion chromosomes. Notably, the female inversion carriers seemed to produce more parental originating aneuploid embryos than male inversion carriers (45.5% vs 23.9%, p = 0.044).The type of inversion and sperm parameters of male chromosomal inversion carriers did not affect the ploidy status of embryos. The incidence of parental originating aneuploidy in inversion carrier couples is lower than expected. For male chromosomal inversion carriers with normal sperm condition whose female partners are under 38 years old, natural conception combined with prenatal diagnosis could be provided as an option during fertility counseling.

6. Development of a Nomogram Model to Predict in-Hospital Survival in Patients with Multiple Trauma

Background. Herein, we purposed to establish a nomogram model capable of assessing the probability of in-hospital survival in patients with multiple trauma. Methods. Our retrospective study is associated with 286 multiple trauma patients with 21 variables from 2017 to 2021 in The Second Affiliated Hospital, Hengyang Medical School, University of South China. We performed the univariate and multivariate logistic regression analyses for investigating the risk factors of multiple trauma. Further, we constructed a novel nomogram model, and this nomogram was evaluated by a calibration plot. Based on the multivariate analysis or the nomogram prediction model, we calculated the risk score of each patient for multiple trauma. Moreover, we compared the survival probability between the high-risk score and low-risk score groups. Finally, we assessed the discrimination of the risk score by using the C-index and the time-dependent receiver operating characteristics (ROC) curve. Results. Multivariate regression analysis revealed that the age and ISS scores were the independent risk factors, while the GCS score had protective effects on in-hospital survival. The high C-index and area under the curve (AUC) of the ROC curve confirmed reasonable discrimination for the multivariate analysis and the nomogram prediction model. Further, the calibration plot indicated reasonable accuracy of the nomogram predicting 30-day and 60-day survival probabilities. Conclusion. The nomogram model established here has good predictive efficacy for in-hospital survival of patients with multiple injuries.

7. A possible new activator of PI3K‐Huayu Qutan Recipe alleviates mitochondrial apoptosis in obesity rats with acute myocardial infarction

Obesity, which has unknown pathogenesis, can increase the frequency and seriousness of acute myocardial infarction (AMI). This study evaluated effect of Huayu Qutan Recipe (HQR) pretreatment on myocardial apoptosis induced by AMI by regulating mitochondrial function via PI3K/Akt/Bad pathway in rats with obesity. For in vivo experiments, 60 male rats were randomly divided into 6 groups: sham group, AMI group, AMI (obese) group, 4.5, 9.0 and 18.0 g/kg/d HQR groups. The models fed on HQR with different concentrations for 2 weeks before AMI. For in vitro experiments, the cardiomyocytes line (H9c2) was used. Cells were pretreated with palmitic acid (PA) for 24 h, then to build hypoxia model followed by HQR-containing serum for 24 h. Related indicators were also detected. In vivo, HQR can lessen pathohistological damage and apoptosis after AMI. In addition, HQR improves blood fat levels, cardiac function, inflammatory factor, the balance of oxidation and antioxidation, as well as lessen infarction in rats with obesity after AMI. Meanwhile, HQR can diminish myocardial cell death by improving mitochondrial function via PI3K/Akt/Bad pathway activation. In vitro, HQR inhibited H9c2 cells apoptosis, improved mitochondrial function and activated the PI3K/Akt/Bad pathway, but effects can be peripeteiad by LY294002. Myocardial mitochondrial dysfunction occurs following AMI and can lead to myocardial apoptosis, which can be aggravated by obesity. HQR can relieve myocardial apoptosis by improving mitochondrial function via the PI3K/Akt/Bad pathway in rats with obesity.

High glucose and its byproducts are important factors causing dysfunction of endothelial cells. Autophagy is critical for endothelial cellular homeostasis. However, the specific molecular mechanism of how autophagy is regulated in endothelial cells under high-glucose condition remains unknown. We aim to explore the role Sirt6 plays in regulating autophagy in AGE-treated endothelial cells and how this function is exerted via KLF4.Our results indicate that autophagy level increased in AGE-treated endothelial cells alongside with higher Sirt6 and KLF4 expression level. What's more, knock-in of Sirt6 by adenovirus led to augmented autophagy level while knockdown of Sirt6 led to the opposite. We also verified that Sirt6 affected KLF4 expression positively but KLF4 didn't influence Sirt6 expression level while knocking out of KLF4 impaired Sirt6-enhanced autophagy. Finally we found that STZ-induced diabetic mice showed more autophagosomes in endothelium and Sirt6 knockdown by adeno-associated virus reduced the number of autophagosomes. Knockdown of Sirt6 also caused impaired endothelium integrity but echocardiography indicated there were no significant functional differences.Our research reveals more about how Sirt6 regulates autophagy in endothelial cells under high-glucose simulated condition and provides further insight into the relationships between Sirt6 and KLF4.

9. High intact fibroblast growth factor 23 levels associated with low hemoglobin levels in patients on chronic hemodialysis

IntroductionA negative association between C-terminal fibroblast growth factor 23 (cFGF23) and hemoglobin (Hb) levels has been reported in patients with predialysis chronic kidney disease. In dialysis patients, the dominant form of serum FGF23 is intact FGF23 (iFGF23); however, its association with the Hb level remains unclear. Therefore, simultaneously monitoring iFGF23 and cFGF23 levels is crucial. In this study, we investigated the associations between both forms of FGF23 (iFGF23 and cFGF23) and renal anemia in chronic hemodialysis (CHD) patients.MethodsWe included 166 CHD patients from two hospitals in this cross-sectional, observational study. The primary predictors were serum iFGF23, cFGF23, and iFGF23/cFGF23 levels. The main outcome was the Hb level.ResultsAmong the CHD patients included, 60.8% were men with a mean age of 59.4 ± 12.7 years. In the crude analysis, iFGF23 and iFGF23/cFGF23 levels showed a significant negative association (−0.27, p = 0.004 and −0.22, p = 0.034, respectively) with the Hb level. Even after adjusting for multiple variables (a parsimonious model), every increment of natural log transformation by 1 for (ln)iFGF23 and ln(iFGF23/cFGF23) levels showed a negative correlation with the Hb level (estimate: −0.27 [95%CI: −0.44, −0.10, p = 0.001]; −0.19 [95%CI: −0.37, −0.01, p = 0.042], respectively), whereas both were positively associated with erythropoietin-stimulating agent (ESA) hyporesponsiveness (odds ratio [OR]: [95%CI: 2.30, 1.26–4.17], p = 0.006; 1.95 [95%CI: 1.08–3.50], p = 0.025). Moreover, these abovementioned associations were more dominant in patients with diabetes who used angiotensin receptor blockers.DiscussionIn conclusion, a negative association between serum iFGF23 or iFGF23/cFGF23 level and the Hb level was observed in our CHD patients. Meanwhile, a higher iFGF23 or iFGF23/cFGF23 level may predispose patients to ESA hyporesponsiveness.

11. UNC-43/CaMKII-triggered anterograde signals recruit GABAARs to mediate inhibitory synaptic transmission and plasticity at C. elegans NMJs

Disturbed inhibitory synaptic transmission has functional impacts on neurodevelopmental and psychiatric disorders. An essential mechanism for modulating inhibitory synaptic transmission is alteration of the postsynaptic abundance of GABAARs, which are stabilized by postsynaptic scaffold proteins and recruited by presynaptic signals. However, how GABAergic neurons trigger signals to transsynaptically recruit GABAARs remains elusive. Here, we show that UNC-43/CaMKII functions at GABAergic neurons to recruit GABAARs and modulate inhibitory synaptic transmission at C. elegans neuromuscular junctions. We demonstrate that UNC-43 promotes presynaptic MADD-4B/Punctin secretion and NRX-1α/Neurexin surface delivery. Together, MADD-4B and NRX-1α recruit postsynaptic NLG-1/Neuroligin and stabilize GABAARs. Further, the excitation of GABAergic neurons potentiates the recruitment of NLG-1-stabilized-GABAARs, which depends on UNC-43, MADD-4B, and NRX-1. These data all support that UNC-43 triggers MADD-4B and NRX-1α, which act as anterograde signals to recruit postsynaptic GABAARs. Thus, our findings elucidate a mechanism for pre- and postsynaptic communication and inhibitory synaptic transmission and plasticity.

12. A case report about a child with drug-resistant tuberculous meningitis

Background Hematogenous disseminated tuberculosis predisposes to concurrent tuberculous meningitis (TBM), the most devastating and disabling form of tuberculosis. However, children often have atypical clinical symptoms, difficulty in specimen collection, low specimen content, and an increasing incidence of drug-resistant tuberculosis. Thus, the accurate diagnosis and timely treatment of childhood tuberculosis face monumental challenges. Case presentation The 14-year-old female presented to the hospital with intermittent fever, headache, and blurred vision. Her cerebrospinal fluid (CSF) showed a lymphocytic pleocytosis, an elevated protein level, and a decreased chloride level. And her CSF tested positive for TB-RNA. Xpert MTB/RIF detected Mycobacterium tuberculosis in her CSF, but the rifampin resistance test was unknown. Subsequently, her CSF culture was positive for Mycobacterium tuberculosis. The drug sensitivity test (DST) revealed resistance to isoniazid, rifampin, and fluoroquinolones. A computed tomography (CT) of the chest showed diffuse miliary nodules in both lungs. Intracranial enhanced magnetic resonance imaging (MRI) showed “multiple intensified images of the brain parenchyma, cisterns, and part of the meninges.” The final diagnosis is miliary pulmonary tuberculosis and pre-extensive drug-resistant TBM. After 19 months of an oral, individualized antituberculosis treatment, she recovered with no significant neurological sequelae. Conclusion For patients with miliary pulmonary tuberculosis, especially children, even if there are no typical clinical symptoms, it is necessary to know whether there is TBM and other conditions. Always look for the relevant aetiological basis to clarify whether it is drug-resistant tuberculosis. Only a rapid and accurate diagnosis and timely and effective treatment can improve the prognosis and reduce mortality and disability rates.

13. Correlates of e-cigarette use susceptibility among never-smoking youth: A cross-sectional study in Shenzhen, China

Purpose Identifying youth at risk of future e-cigarette use is crucial for informing effective prevention strategies. The current study aims to examine the correlates of e-cigarette use susceptibility among never-smoking youth from four levels: personal, psychological, parental, and social factors. Methods This Cross-sectional study recruited 2487 never-smoking students (aged 12–17 years) from 15 junior and high schools in the Bao'an District of Shenzhen, China from October to December 2021. E-cigarette susceptibility was measured by two items assessing the possibility to use e-cigarettes in the future and willingness to use if offered by a friend. Random-effects logistic regression models were used to explore risk factors of e-cigarette use susceptibility from four levels. Results Of the 2487 youth who never smoked, 5.5% (n = 136) were susceptible to e-cigarette use. The regression model identified the following correlates of e-cigarette susceptibility: perceptions of e-cigarettes, perceived social support, parental style, pro- and anti-tobacco information exposure, SHS exposure, and friends using e-cigarettes. Youth believing it is not easy to get addicted to e-cigarettes (aOR = 2.19; 95%CI: 1.21–3.96; p = 0.010), having father rejection (aOR = 2.38; 95%CI: 1.22–4.65; p = 0.011), having SHS exposure at home for 5–7 days (aOR = 2.04; 95%CI: 1.24–3.37; p = 0.005), noticing e-cigarette advertising (aOR = 1.80; 95%CI: 1.22–2.65; p = 0.003), and having friends using e-cigarettes (aOR = 2.59–5.28; 95%CI: 1.76–14.73; p

PURPOSE: Aneuploidy is one genetic factor leading to the failure of embryo implantation. This study aimed to investigate the relationship between the day of embryo blastulation and the ploidy status of embryo, to aid in selecting embryos with the most likelihood of of being euploid in a noninvasive way. METHODS: This retrospective study recruited women undergoing preimplantation genetic testing (PGT) for aneuploidy (PGT-A) with trophectoderm biopsy from January 2019 to December 2020. The ploidy status of embryos was determined by next-generation sequencing (NGS). RESULTS: Altogether, 2531 blastocysts from 839 PGT-A cycles were evaluated. 20. [A new xanthone from Hypericum lagarocladum]

Repeated silica gel column chromatography, reversed-phase C_(18) column chromatography, Sephadex LH-20 column chromatography, high performance liquid chromatography and semi-preparative medium pressure liquid chromatography were performed to separate and purify the chemical constituents of Hypericum lagarocladum. Spectroscopic methods such as mass spectrometry(MS) and nuclear magnetic resonance(NMR) combined with physicochemical properties were adopted in identifying the structure of the isolated compounds. Ten compounds were isolated from the ethyl acetate fraction of H. lagarocladum and identified as lagarxanthone A(1), 1,7-dihydroxyxanthone(2), 3,4,5-trihydroxyxanthone(3), 2,7-dihydroxy-1-methoxyxanthone(4), 1,3-dihydroxy-7-methoxyxanthone(5), 1,5-dihydroxy-8-methoxyxanthone(6), 3,4-dihydroxy-2-methoxyxanthone(7), 3,4-dihydroxy-5-methoxyxanthone(8), 2,3-dimethoxyxanthone(9), and 2,3,4-trimethoxyxanthone(10). Among them, compound 1 was a new compound, and compounds 2-10 were isolated from this plant for the first time. These ten compounds were tested for glucose uptake in L6 cells, and the results showed that all the compounds had no significant effect on glucose uptake.

21. Coronary slow flow research: a bibliometric analysis (Preprint)

- Abstract
BACKGROUND The studies on coronary slow flow are receiving increasing attention, but objective evaluation of the publications is still lacking. Thus, a bibliometric analysis is required to reflect the characteristics and development of publications on coronary slow flow. OBJECTIVE The purpose of this study was to visualize the current status and research hotspots of coronary slow flow with bibliometric analysis. METHODS All relevant publications on coronary slow flow during 2003 to 2021 were extracted from the Web of Science Core Collection database and analyzed by VOSviewer and CiteSpace visualization softwares. Years, journals, countries/regions, institutions, authors and research hotspots were identified. RESULTS A total of 847 publications were retrieved. The journals with the largest publication volume were Coronary Artery Disease and International Journal of Cardiology. The country/region with the largest publication volume was Turkey, followed by the United States and China. The institution with the largest publication volume was European Union of Research Universities. Meanwhile, the author with the largest publication volume was Mustafa Cetin from Turkey. Keyword analysis results indicated that treatment and prognosis, diagnosis and pathogenesis, and risk factors were the clustering centers of coronary slow flow, and the research hotspots gradually changed with the progress of time, from pathogenesis to treatment and prognosis. CONCLUSIONS The direction of future research involves the search for effective and noninvasive detection indicators and treatments of coronary slow flow. Collaboration needs to be enhanced between different institutions or countries/regions, which would improve clinical outcomes for patients with coronary slow flow.

Nuclear receptor coactivator (NCOA) family gene fusions have been increasingly discovered in diverse mesenchymal neoplasms, while PRRX1-NCOA-fused fibroblastic tumors still remain insufficiently characterized. We herein present two additional PRRX1-NOCA1-positive cases sharing lobulated hypocellular growth of innocuous spindle-to-stellate cells in a fibromyxoid stroma enriched with polymorphous vessels. A constellation of low cellularity, alternating myxocollagenous matrix, bland cytomorphology, and, especially, unusual collagenous rosettes in one case were morphologically reminiscent of low-grade fibromyxoid sarcoma. In both cases, immunoprofiles were similarly nondescript and negative for all diagnostic markers, including MUC4, emphasizing the diagnostic value of molecular testing. Review of published and current cases highlights a striking predominance of PRRX1-NCOA1, unusual collagenous rosettes, and favorable behavior in this emerging fibroblastic tumor type.

Viral myocarditis (VM) is an inflammatory disease of the myocardium associated with heart failure, which is caused by common viral infections. A majority of the infections are initiated by coxsackievirus B3 (CVB3). MicroRNAs (miRNAs) have a major role in various biological processes, including gene expression, cell growth, proliferation, and apoptosis, as well as viral infection and antiviral immune responses. Although, miRNAs have been found to regulate viral infections, their role in CVB3 infection remains poorly understood. In the previous study, miRNA microarray results showed that miR-324-3p expression levels were significantly increased when cells and mice were infected with CVB3. It was also found that miR-324-3p downregulated TRIM27 and decreased CVB3 replication in vitro and in vivo. In vitro, analysis of downstream signaling of TRIM27 revealed that, miR-324-3p inhibited CVB3 infection, and reduced cytopathic effect and viral plaque formation by reducing the expression of TRIM27. In vivo, miR-324-3p decreased the expression of TRIM27, reduced cardiac viral replication and load, thereby strongly attenuating cardiac injury and inflammation. Taken together, this study suggests that miR-324-3p targets TRIM27 to inhibit CVB3 replication and viral load, thereby reducing the cardiac injury associated with VM.

Ionic liquids as green media have shown excellent performance in catalyzing poly(ethylene terephthalate) (PET) degradation due to their homogeneous catalytic properties. Our group has developed several series of ionic liquid catalysts for PET glycolysis in recent years. This work summarizes and reviews the research progress of ionic liquid catalysts in our group and other teams. We focus on PET glycolysis catalysts, and also describe PET hydrolysis and aminolysis ionic liquid catalysts briefly. The effects of catalyst and reaction conditions on conversion and yield are detailed. The kinetic model of PET glycolysis and the activation energy for different catalysts are summarized. The mechanisms of ionic liquid catalysts are summarized to provide a theoretical basis for the development of efficient PET degradation catalysts.

Microplastics (MPs) are substrates available for biofilms colonization in natural water environments. The biofilms formation may enhance the ability of MPs to adsorb harmful contaminants. Herein, we investigated the biofilms formation of three different MPs (PVC, PA and HDPE) in simulated natural environment, and observed the chemical structure, charge property, hydrophobicity and other properties of MPs affect microbial biomass and community composition. More importantly, potential pathogens were found in all three MPs biofilms. Furthermore, the adsorption capacities of original MPs and biological aging MPs for norfloxacin (NOR) was compared. HDPE has the largest adsorption capacity for NOR, while PA has the smallest adsorption capacity for NOR. It was concluded that the formation of biofilms enhanced the adsorption of NOR by 50.60 %, 24.17 % and 46.02 % for PVC, PA and HDPE, respectively. In addition, hydrogen-bond interaction, electrostatic interaction and hydrophobic interaction were found to dominate the adsorption of NOR by MPs. Our study contributed to improve the understanding of the interactions between aging MPs and contaminants in the natural water environments, and provided essential information for ecological risk assessment of MPs.

30. Clinical features and relapse risks factors of immunoglobulin G4–related disease: a single-center retrospective study

- Abstract
Objective: The aim of this study was to observe the demographic and clinical characteristics of immunoglobulin (Ig) G4–related disease (IgG4-RD). We aimed to compare different treatment methods and to identify the risk factors for non-response and relapse after treatment.Methods: We performed a retrospective study of 201 IgG4-RD patients initially diagnosed and treated at the First Affiliated Hospital of China Medical University from January 2016 to December 2020. Patients’ sex, age, clinical manifestations, baseline biochemical values, the number of organs involved, and the type of organ involvement were recorded. All patients received glucocorticoid (GC) monotherapy or GC + immunosuppressant combination therapy. The serum IgG4 concentration as well as the details of clinical response, relapse, and side effects were recorded at 1, 3, 6, and 12 months after treatment.Results: The incidence of IgG4-RD was primarily centered in the age group of 50–70 years old, and the proportion of affected male patients increased with age. The most common clinical symptom was swollen glands or eyes (42.79%). The rates of single- and double-organ involvement were 34.83% and 46.27%, respectively. The pancreas (45.77%) was the most frequently involved organ in cases of single-organ involvement, and the pancreas and biliary tract (45.12%) was the most common organ combination in cases of double-organ involvement. Correlation analysis showed that the number of organs involved was positively related to the serum IgG4 concentration (r = 0.161). The effective rate of GC monotherapy was 91.82%, the recurrence rate was 31.46%, and the incidence of adverse reactions was 36.77%. Meanwhile, the effective rate of GC + immunosuppressant combination therapy was 88.52%, the recurrence rate was 19.61%, and the adverse reaction rate was 41.00%. There were no statistically significant differences in response, recurrence, and adverse reactions. The overall response rate within 12 months was 90.64%. 31. Effects of environmental and anthropogenic factors on the distribution and abundance of microplastics in freshwater ecosystems

Although microplastics are emerging marine pollutants that have recently attracted increasing attention, it is still difficult to identify their sources. This study reviewed 6487 articles to determine current research trends and found 237 effective concentration points after sorting, which were distributed in four regions and related to freshwater ecosystems. Results found that 15 environmental variables represented natural and anthropogenic environmental characteristics, of which seven environmental variables were selected for experimental modelling. Random forest models fitted sample data, thus facilitating the identification of regional microplastics distribution. The global random forest model had random forest importance scores (RFISs) for gross domestic product, population, and the proportion of agricultural land use were 15.76 %, 15.64 %, and 14.74 %, respectively; these indicate that human activities significantly affected the global distribution of microplastics. In Asia, agriculture and urban activities are the main sources of microplastics, with an RFIS of 11.58 % and 12.24 % for the proportion of agricultural and urban land use, respectively. Activities in urban areas were determined to be the main influencing factors in North America, with an RFIS of 13.92 % for the proportion of urban land use. Agricultural activities were the main influencing factors in Europe, with RFISs for the proportion of agricultural land use of 16.90 %. Our results indicate that region-specific policies are required to control microplastics in different regions, with soil composition being a latency factor that affects microplastics' distribution.

Hybrid catalysts composed of transition metals and nitrogen-doped carbons have been emerging as one of efficient electrocatalysts for electrochemical CO2-to-CO under ambient conditions. Herein, NiO loaded on mesoporous graphene with tunable concentration of pyridinic nitrogen is rationally synthesized by using low-intensity pulsed laser irradiation and pyrolysis treatments. Comprehensive experiments verify that, it is pyridinic N, rather than pyrrolic N, when bonded to Ni, that significantly enhance the electrochemical CO2 reduction reaction performance. The optimized catalyst exhibits a high CO Faradaic efficiency of 87.5% at a low potential of −0.74 V vs. reversible hydrogen electrode, associated with negligible attenuation of continuous operation over 12 h.

The unique flavor of Allium tuberosum is primarily associated with the hydrolysis of a series of organosulfur compounds, S-alk(en)yl cysteine sulphoxides (CSOs), upon tissue bruising or maceration. To obtain the tissue-specific transcriptomes, 18 RNA-Seq libraries representing leaf, root, stem, mature flower, inflorescence, and seed tissues of A. tuberosum were sequenced, finally yielding 133.7 Gb clean reads. The de novo assembled transcriptomes enabled the identification of 223,529 unigenes, which were functionally annotated and analyzed for the gene ontology and metabolic pathways. Furthermore, to reveal the flavor metabolic pathways, a total of 205 unigenes involved in the sulfur assimilation and CSO biosynthesis were identified, and their expression profiles were analyzed by RNA-Seq and qRT-PCR. Collectively, this study provides a valuable resource for in-depth molecular and functional researches especially on flavor formation, as well as for the development of molecular markers, and other genetic studies in A. tuberosum.

Schiff bases that are generally formed from condensation reactions of aldehydes (or ketones) and amino groups could also be produced by a photodriven one-pot tandem reaction between alcohols and nitro compounds, in our case. Herein, TiO2-x porous cages derived from NH2-MIL-125 by a self-sacrificing template route are used to study the organic transformation and exhibit 100% conversion efficiency of nitrobenzene and 100% selectivity for Schiff bases in the system of benzyl alcohol (5 mL) and nitrobenzene (41 μL) upon light irradiation, but hydrogen by dehydrogenation of benzyl alcohol cannot be detected. Successful occurrence of the organic transformation is mainly attributed to Ti(III)-oxygen vacancy associates. Surface oxygen vacancy-related Ti(III) sites are responsible for binding with nitro groups, and low-coordinated Ti5c sites selectively adsorb hydroxyl groups of benzyl alcohol. The Ti(III) and oxygen vacancy associates capture photogenerated electrons for achievement of multielectron reduction of nitrobenzene and the subsequent Schiff base condensation reaction with the as-formed benzaldehyde.

Background: Vessel segmentation is a fundamental, yet not well-solved problem in medical image analysis, due to complicated geometrical and topological structures of human vessels. Unlike existing rule- and conventional learning-based techniques, which hardly capture the location of tiny vessel structures and perceive their global spatial structures, Methods: we propose Simultaneous Self- and Channel-attention Neural Network (termed SSCA-Net) to solve the multi-scale structure-preserving vessel segmentation (MSVS) problem. SSCA-Net differs from the conventional neural networks in modeling image global contexts, showing more power to understand the global semantic information by both self- and channel-attention (SCA) mechanism, and offering high performance on segmenting vessels with multi-scale structures. Specifically, the SCA module is designed and embedded in the feature decoding stage to learn SCA features at different layers, which the self-attention is used to obtain the position information of the feature itself, and the channel attention is designed to guide the shallow features to obtain global feature information. Results: Three blood vessel data sets are train and validate the models. our SSCA-Net achieves 96.21% in Dic and 92.70% in Mean IoU on the intracranial vessel dataset and achieved 98.20 %, 83.52% and 96.14% in AUC, Sen and Acc respectively on retinal vessel dataset. The obtain model can segment the leg arteries and Dic score is 97.21% and the Mean IoU score is 94.42%. Conclusions: The results demonstrated that the proposed SSCA-Net clear improvements of our method over the state-of-the-arts in terms of preserving vessel details and global spatial structures.

Integrins, a group of heterodimer receptors for cell-matrix and cell-cell adhesion, mediate various intracellular activities, including cell migration, polarity, survival, growth, and death. Multiple types of integrins are differentially expressed in various cancers during different stages of progression, which are involved in the regulation of cancer cell proliferation, invasion, migration, and angiogenesis. The crucial roles of integrins in tumor progression provide valuable clues for cancer diagnosis and targeted therapy. Numerous integrin inhibitors have been investigated in clinical trials to explore effective regimens and minimize side effects. Given the complexity of the integrin-mediated tumor-promoting effect, challenges and difficulties remain in the research and development of integrin inhibitors, which seriously restrict the efficacy and application of integrin-targeted therapy. Novel targeted therapy of integrins, however, is beneficial for patients as a potential avenue forward, which needs better pharmacological effect, valid experimental models, and in-depth understanding of integrins. This review provides the insight needed to elucidate the mechanisms underlying cancer progression and novel protocols for the clinical treatment of cancer.

Background Mesenchymal stem cell-derived extracellular vesicles (MSC-EVs) have been found to have a great potential for soft tissue repair due to various biological functions, including pro-angiogenesis and low immunogenicity. However, the low yield and heterogeneity of MSC-EVs limited their clinical transformation. This study was designed to develop a novel adipose-derived stem cell engineered nanovesicles (ADSC-NVs) with high production and explore its pro-angiogenetic effect and application in adipose tissue regeneration. Methods Adipose-derived stem cell-derived extracellular vesicles (ADSC-EVs) were isolated from an EVs-free culture medium for human ADSCs (hADSCs). ADSC-NVs were prepared by sequentially extruding ADSCs followed by iodixanol density gradient ultracentrifugation and were compared with ADSC-EVs in morphology, size distribution, protein contents and yield. The pro-angiogenetic effect of ADSC-NVs in different doses (0, 5, 20 and 80 μg/mL) in vitro was determined using transwell assay, tube formation assay, western blot and qRT-PCR. In vivo, BALB/c nude mice were administered injection of a mixture of fat granules and different dose of ADSC-NVs and grafts were harvested at 12 weeks post-transplantation for further analysis. By analyzing the weight and volume of grafts and histological evaluation, we investigated the effect of ADSC-NVs in vessel formation and adipose tissue regeneration. Results Our results showed yield of purified ADSC-NVs was approximately 20 times more than that of ADSC-EVs secreted by the same number of ADSCs. In vitro, both ADSC-NVs and ADSC-EVs exhibited a dose-dependent pro-angiogenetic effect, despite their distinct miRNA profiles. These effects of ADSC-NVs may be mediated by enriched miR-21-5p via PTEN inhibition and PI3K/p-Akt signaling activation. Furthermore, after a mixed injection of ADSC-NVs, vessel formation and adipose regeneration were observed in vivo in fat implants. Conclusions Our study developed a potent alternative of ADSC-EVs. ADSC-NVs have a high pro-angiogenesis potential and can be used as cell-free therapeutic biomaterials in soft tissue regeneration.

45. DNL-Net: deformed non-local neural network for blood vessel segmentation

Background The non-local module has been primarily used in literature to capturing long-range dependencies. However, it suffers from prohibitive computational complexity and lacks the interactions among positions across the channels. Methods We present a deformed non-local neural network (DNL-Net) for medical image segmentation, which has two prominent components; deformed non-local module (DNL) and multi-scale feature fusion. The former optimizes the structure of the non-local block (NL), hence, reduces the problem of excessive computation and memory usage, significantly. The latter is derived from the attention mechanisms to fuse the features of different levels and improve the ability to exchange information across channels. In addition, we introduce a residual squeeze and excitation pyramid pooling (RSEP) module that is like spatial pyramid pooling to effectively resample the features at different scales and improve the network receptive field. Results The proposed method achieved 96.63% and 92.93% for Dice coefficient and mean intersection over union, respectively, on the intracranial blood vessel dataset. Also, DNL-Net attained 86.64%, 96.10%, and 98.37% for sensitivity, accuracy and area under receiver operation characteristic curve, respectively, on the DRIVE dataset. Conclusions The overall performance of DNL-Net outperforms other current state-of-the-art vessel segmentation methods, which indicates that the proposed network is more suitable for blood vessel segmentation, and is of great clinical significance.

46. First report of tomato spotted wilt virus infecting water dropwort in China

Water dropwort (Oenanthe javanica) is an aquatic perennial plant that has been cultivated in many regions in Asia for thousands of years. In China, it is an economically important vegetable that has been consumed as food, while also being used as a folk remedy to alleviate diseases (Liu et al., 2021). In 2021, during a disease survey of a greenhouse in Beijing, China, chlorotic spots were detected on many water dropwort plants (Fig. S1A). Twenty-seven water dropwort samples were collected for the extraction of total RNA using the TRIzol reagent (Invitrogen, USA). High-quality RNA samples from three water dropwort plants were combined and used as the template for constructing a single small RNA library (BGI-Shenzhen Company, China). The Velvet 1.0.5 software was used to assemble the clean reads (18 to 28 nt) into larger contigs, which were then compared with the nucleotide sequences in the National Center database using the BLASTn algorithm. Thirty-eight contigs matched sequences in the tomato spotted wilt virus (TSWV) genome. No other viruses were detected. Twenty-seven leaf samples were analyzed in an enzyme-linked immunosorbent assay (ELISA) with anti-TSWV antibody (Agdia, USA), which revealed 17 positive reaction. Two sets of primer pairs targeting different parts of the S RNA (Table S1) was used to verify the TSWV infection on water dropwort by reverse transcription (RT)-PCR followed by Sanger sequencing (BGI-Shenzhen, China). The TSWV target sequences were amplified from 17 samples, which was consistent with the ELISA results. The sequenced 861-bp PCR product shared 99.8% nucleotide sequence identity with TSWV isolate MR-01 (MG593199), while the 441-bp amplicon shared a 99.2% nucleotide sequence identity with MR-01 (MG593199). To obtain the whole genome sequence of TSWV (S, M, and L RNA sequences), specific RT-PCR primers were designed (Table S1) and used to amplify their respective fragments from one representative sample (TSWV-water dropwort). The amplified products were inserted into PCE2TA/Blunt-Zero vector (Vazyme Biotech Co., Ltd, China) and then sequenced (BGI-Shenzhen, China). The S, M, and L RNA sequences were determined to be 2,952 nt (accession no. OM154969), 4,776 nt (accession no. OM154970), and 8,914 nt (accession no. OM154971), respectively. BLASTn analysis demonstrated that the whole genome sequence was highly conserved. The nucleotide identities between this isolate and other TSWV isolates ranged from 98.6% to 99.6% (S RNA), 98.9% to 99.2% (M RNA), and 97.3% to 98.7% (L RNA). Using MEGA 7.0, the phylogenetic relationships of TSWV were determined on the basis of the S, M, and L RNA full-length sequences (Kumar et al., 2016). In the S RNA derived phylogenetic tree, the water dropwort isolate was closely related to the MR-01 isolate from the USA (MG593199). In the M RNA and L RNA derived phylogenetic trees, the water dropwort isolate formed a branch with only a TSWV isolate from eggplant. Additionally, the M and L RNA sequences were most similar to sequences in TSWV isolates from China and Korea, respectively (Fig. S1B). To the best of our knowledge, this is the first report of water dropwort as a natural host for TSWV in China and the second report worldwide since the first finding in the Korea (Kil et al. 2020). TSWV has caused serious problems on many crops in the world, and the infection of TSWV on water dropwort in a greenhouse should not be looked lightly. Firstly, the virus can be passed on from generation to generation in infected water dropwort due to the vegetative propagation mode of the plant in production, thus threaten the production of this vegetable crop. In addition, infected water dropwort may serve as a reservoir for the virus, thus potentially posing a threat for causing TSWV spread in the affected greenhouses. 47. Olaparib Induces RPL5/RPL11-Dependent p53 Activation via Nucleolar Stress

The poly (ADP-ribose) polymerase (PARP) inhibitor (PARPi) Olaparib is a widely used targeted therapy for a variety of solid tumors with homologous recombination deficiency (HRD) caused by mutation of BRCA1/2 or other DNA repair genes. The anti-tumor activity of Olaparib has been largely attributed to its ability to inhibit PARP enzymes and block DNA single-strand break (SSB) repair, which eventually leads to the most detrimental DNA damage, double-strand breaks (DSB), in HRD cells. Although PARPi was found to induce p53-dependent cell death, the underlying molecular mechanism remains incompletely understood. Here, we report that Olaparib treatment leads to p53 stabilization and activation of its downstream target genes in a dose- and time-dependent manner. Mechanistically, Olaparib triggers nucleolar stress by inhibiting biosynthesis of the precursor of ribosomal RNAs (pre-rRNA), resulting in enhanced interaction between ribosomal proteins (RPs), RPL5 and RPL11, and MDM2. Consistently, knockdown of RPL5 and RPL11 prevents Olaparib-induced p53 activation. More importantly, Olaparib efficiently suppresses breast and colorectal cancer cell survival and proliferation through activation of p53. Altogether, our study demonstrates that Olaparib activates the nucleolar stress-RPs-p53 pathway, suggesting rRNA biogenesis as a novel target for PARPi.

48. miR-3064-5p and miR-4745-5p affect heparin sensitivity in patients undergoing cardiac surgery by regulating AT-III and factor X mRNA levels

Subject: Perioperative regulation of coagulation function through heparin in patients undergoing cardiac surgery with cardiopulmonary bypass is an important part of performing cardiac surgery, and postoperative bleeding due to abnormal coagulation function caused by differences in heparin sensitivity in different individuals is an independent risk factor for postoperative complications and death.Method: Using an online database, 10 miRNAs interacting with AT-III and FX genes were predicted. Patients were divided into three groups according to the difference in activated clotting time (ACT) after the first dose of heparin (2.5 mg kg−1): group A: hyposensitive group (ACT < 480 s); group B: sensitive group (480 s ≤ ACT ≤ 760 s); and group C: hypersensitive group (ACT > 760 s). Perioperative and 24 h postoperative blood loss and other clinical data of patients in the three groups were recorded. Blood samples were collected before surgery, and RT-PCR was used to detect the levels of AT-III and FX gene mRNA and the levels of predicted 10 miRNAs.Result: Heparin sensitivity was positively correlated with AT-III mRNA levels and negatively correlated with FX gene mRNA levels in the three groups, and the blood loss in group B was significantly lower than that in groups A and C, which was statistically significant (p < 0.05). miR-3064-5p and miR-4745-5p expression levels were significantly different among group A, group B, and group C (p < 0.05) and were closely correlated with AT-III and FX gene mRNA expression levels, respectively.Conclusion: Differences in heparin sensitivity in patients undergoing cardiac surgery were associated with the mRNA expression of AT-III and FX genes, and the expression levels of miR-3064-5p and miR-4745-5p were found to be closely related to the AT-III and FX gene mRNA, respectively, indicating that miR-3064-5p and miR-4745-5p affect the differences in heparin sensitivity among different individuals by regulating the mRNA expression levels of AT-III and FX genes.Clinical Trial Registration:http://www.chictr.org.cn/abouten.aspx, identifier registration number: ChiCTR-2100047348

FEZ1-mediated axonal transport plays important roles in central nervous system development but its involvement in the peripheral nervous system is not well-characterized. FEZ1 is deleted in Jacobsen syndrome (JS), an 11q terminal deletion developmental disorder. JS patients display impaired psychomotor skills, including gross and fine motor delay, suggesting that FEZ1 deletion may be responsible for these phenotypes, given its association with the development of motor-related circuits. Supporting this hypothesis, our data show that FEZ1 is selectively expressed in the rat brain and spinal cord. Its levels progressively increase over the developmental course of human motor neurons (MN) derived from embryonic stem cells. Deletion of FEZ1 strongly impaired axon and dendrite development, and significantly delayed the transport of synaptic proteins into developing neurites. Concurring with these observations, Drosophila unc-76 mutants showed severe locomotion impairments, accompanied by a strong reduction of synaptic boutons at neuromuscular junctions. These abnormalities were ameliorated by pharmacological activation of UNC-51/ATG1, a FEZ1-activating kinase, with rapamycin and metformin. Collectively, the results highlight a role for FEZ1 in MN development and implicate its deletion as an underlying cause of motor impairments in JS patients.

Two cluster-based metal–organic frameworks {[Cd5L4Br10]·2(CH3)2NH}n (1) and {[MnIII3OL(TA)3]F·sol}n (2) (L = 1,3,5-tris(benzimidazol-1-ylmethyl)-2,4,6-trimethylbenzene, TA = terephthalate anion) containing a semi-rigid tribenzimidazol-based ligand were synthesized and characterized. Single crystal X-ray analyses show that the structure of 1 contains pentanuclear cadmium clusters with an interesting four-fan shape, in which each μ2-Br interlinks two crystallographically unique Cd atoms, while 2 contains a trimanganese cluster with three Mn3+ ions present in an equilateral triangle distribution. Furthermore, 1 exhibits highly selective fluorescent detection of trace Hg2+, with the detection limit of 5.26 × 10−7 M−1. 2 shows antiferromagnetic interactions between Mn3+ ions.
