Oral mucosa is exposed for many years to dental medical devices which contain metals. Overexposure to metal ions leaching from these dental devices may cause inflammation of mucosa/gingiva and even result in implant failure. Therefore, there is an urgent need to develop human physiologically relevant models which mimic the complex interactions between the oral soft tissues, the microbiome and metal ions leaching from dental medical devices. In this thesis, various human organotypic gingiva and skin models of different levels of complexity were developed to address the above concerns around dental medical devices. These include sterile ex vivo skin, reconstructed human skin (RHS) and reconstructed human gingiva (RHG) where culture methods were adapted to investigate the tissue response to metals and microbes. In Chapter 2, a full thickness ex vivo human skin to determine whether irritation and cytotoxicity occurred after topical application of nickel, palladium and titanium salts at patch-test relevant concentrations. After 24 hours of exposure, localized cytotoxicity was detected in the epidermis and dermis with separation at the basement membrane zone, formation of vacuoles, apoptotic nuclei, decreased metabolic activity and pro-inflammatory cytokine release. This chapter shows that current metal salt concentrations used in the skin patch test, by causing irritation and cytotoxicity, may confound the readout of an allergic reaction and therefore it should be considered to lower the salt concentration in the patch test. In Chapter 3, it was investigated whether the observed cytotoxicity to metals was enhanced by the presence of the Streptococcus mitis. After metal exposure, localized apoptosis was observed in the form of condensed apoptotic nuclei and vacuole formation within the epithelium and condensed nuclei in fibroblasts within the hydrogel of both RHG and RHS. S. mitis enhanced metal-mediated apoptosis in gingiva but not in skin. Apoptosis was mediated via the extrinsic pathway via caspase 8. Activation of the execution phase of apoptosis occurred via caspases 3 and 7, and PARP-1. These results show that metal-mediated cytotoxicity can indeed be exaggerated by commensal microbes. In Chapter 4, new strategies were introduced to the current RHG model: a) bacteria were applied within a low-concentration agar and b) penicillin and streptococcus (PS) was omitted from medium. This advanced microbe-RHG model was topically exposed to Streptococcus gordonii (commensal) or Aggregatibacter actinomycetemcomitans (pathogen) and co-cultured for 72 hours. S. gordonii did not influence RHG cytokine secretion. However, all cytokine secretion (IL-6, IL-8, CCL5, CCL20 CXCL1) secretion increased when RHG were co-cultured with A. actinomycetemcomitans. Both bacteria resulted in increased expression of RHG antimicrobial peptides (AMPs), with S. gordonii exposure resulting in the most Elafin secretion. In Chapter 5, a model which represented the barrier properties of the biological seal between the tooth and gingiva was next developed. This model is complementary to the existing implant-RHG model already present in our laboratory. An extracted premolar was incorporated into RHG. The gingival epithelium transitioned into a sulcus and junctional epithelium which was attached tightly to both the tooth and the lamina propria hydrogel. The sulcus depth (1.24 ± 0.233mm) and junctional epithelium length (2.45 ± 0.315mm) were comparable to that of healthy individuals. It will provide an excellent means to investigate gingivitis and peri-implantitis in the future. Taken together, our advanced organotypic skin and gingiva models enabled complex host-microbe interactions to be investigated identifying different commensal and pathogen effects on the host. This thesis shows that metal ions, which are present in many dental medical devices, do cause irritation and cytotoxicity (apoptosis) to the soft tissues (gingiva and skin) which come into contact with them. Notably, these detrimental effects are enhanced by commensal microbes present on both skin and gingiva.

New research by the Dutch National Institute for Public Health and the Environment (RIVM) indicates that the health risk of playing sports on synthetic turf pitches with an infill of rubber granulate is virtually negligible. Therefore, it is considered safe for people to play sports on such pitches. The research was conducted following public concerns prompted by the Dutch TV programme Zembla called 'Dangerous Play' in October 2016. RIVM hopes that the results of the research will help to answer questions from ministries, municipalities, sports clubs and parents. To evaluate the health risks of playing sports on rubber granulate, it is important to determine which hazardous substances are contained in the granulate and to what extent they may be released. Subsequently, it should be examined how people playing sports can come into contact with these substances and whether this can lead to health effects. Rubber granulate contains numerous substances, such as polycyclic aromatic hydrocarbons (PAHs), metals, plasticisers (phthalates) and bisphenol A (BPA). These substances were found to be released from the granulate in very low amounts. This is because the substances are more or less 'enclosed' in the granulate, which means that the effect of these substances on human health is virtually negligible. What did RIVM investigate? RIVM determined the substances in rubber granulate from 100 sports pitches that are representative of the synthetic turf pitches in the Netherlands. The institute further performed three types of laboratory tests to examine the release of substances from the granulate if a person playing sport comes into contact with them. These 'migration studies' were used to assess to which extent substances can enter the human body via the skin, via the gastrointestinal tract or via the lungs. This was used to calculate human exposure to the released substances and how this can affect health. In addition, RIVM studied the information available in the scientific literature on substances in rubber granulate, their properties and their health effects. Is there a link with leukaemia? No indications were found in the available literature of a link between playing sports on synthetic turf pitches with an infill of rubber granulate and the incidence of leukaemia and lymphoma. No international research has demonstrated this connection. Moreover, it is clear from the composition of the rubber granulate that the chemical substances that are capable of causing leukaemia or lymphoma are either not present (benzene, styrene and 1,3-butadiene) or are present in a very low amount (2-mercaptobenzothiazole). Since the 1980s, a slight rise has been observed in the number of people aged between 10 and 29 who get leukaemia. This trend has not changed since pitches made of synthetic turf were first used in the Netherlands in 2001. Nor does research conducted in the U.S. reveal any increase in the number of new cases of lymphoma in areas where there are relatively many pitches with synthetic turf with an infill of rubber granulate. Information from new American research will be available early 2017. As rubber granulate has been used on football pitches in the United States for a longer period of time (since 1997), it will be possible to analyse over a longer period whether a link exists between playing sports on synthetic turf pitches and getting leukaemia. RIVM is in contact with the researchers and is keeping a sharp eye on the research. Rubber granulate in the environment This research focuses on potential health risks for people who play sports on synthetic turf pitches with an infill of rubber granulate. The research confirms earlier insights showing that the rubber granulate contains metals capable of entering the environment. In particular, zinc was found to be released from the granulate. This metal is not hazardous to humans, but can have consequences for organisms in the soil or surface water. Does rubber granulate meet requirements? Rubber granulate needs to fulfil the regulatory requirements for 'mixtures'. This concentration limit prescribes the maximum permissible amount of certain substances allowed in rubber granulate (there is no limit for how much may be released). This concentration limit concerns substances that are carcinogenic (such as PAHs), hazardous for reproduction or that damage DNA. The amount of PAHs in rubber granulate easily satisfies this concentration limit. The concentration limit for consumer products is far more stringent: it allows far lower amounts of PAHs (100 to 1000 times lower) compared with the concentration limit for mixtures. The amount of PAH in rubber granulate is slightly higher than the concentration limit for consumer products. The European Chemicals Agency (ECHA) is currently conducting research to determine a suitable concentration limit for rubber granulate. RIVM recommends adjusting the concentration limit for rubber granulate to one that is closer to the concentration limit applicable to consumer products. Why is rubber granulate used for football pitches? Rubber granulate is finely crushed rubber particles that are usually made from old car tyres. When used as infill on pitches of synthetic turf, it gives the pitch properties similar to normal turf. It means the ball does not roll too fast and does not bounce too high, and makes the synthetic turf better suited to sliding tackles than it would be without the granulate. Synthetic turf pitches can be used intensively all year around and need less maintenance. A lot is invested nowadays in order to reuse old products as a raw material for new products. This also applies to car tyres. The questions that have arisen about the safety of rubber granulate show that tension may exist between the reuse of materials and concerns about the health risks of new products.

Maximaal Toelaatbare Risiconiveaus (MTR's) en Verwaarloosbare Risiconiveaus (VR's) voor een aantal zware metalen werden vastgesteld. Een aantal van deze waarden, die eerder door het Ministerie van Volkshuisvesting, Ruimtelijke Ordening en Milieu zijn gebruikt voor het vaststellen van milieukwaliteitsdoelstellingen (MILBOWA-notitie), zijn herzien, voor de overige metalen zijn nieuwe waarden vastgesteld. In de MILBOWA-notitie is aangegeven dat milieukwaliteitsdoelstellingen na verloop van tijd op basis van nieuwe wetenschappelijke informatie opnieuw geevalueerd dienen te worden. Het huidige rapport bevat de resultaten van deze evaluatie. Tijdens het vaststellen van milieukwaliteitsdoelstellingen in MILBOWA voor metalen kwam naar voren dat in sommige gevallen het MTR en/of VR lager was dan de achtergrondconcentratie. In deze gevallen is de milieukwaliteitsdoelstelling gelijk gesteld aan de achtergrondconcentratie. Er werd geconcludeerd dat de voorgestelde methodologie niet gebruikt kan worden voor het afleiden van MTR's en VR's voor metalen en andere van nature voorkomende stoffen. De in dit rapport voorgestelde MTR's en VR's zijn gebaseerd op het toegevoegd risico concept. Daarnaast zijn de (ecotoxicologische) data die gebruikt zijn voor het afleiden van de MTR's en VR's aangevuld en is er rekening gehouden met het TCB-advies over het toegevoegd risicoconcept. De voorgestelde MTR's en VR's kunnen door het Ministerie van Volkshuisvesting, Ruimtelijke Ordening en Milieu worden gebruikt voor het vaststellen van milieukwaliteitsdoelstellingen. Er wordt gesproken over een Maximaal Toelaatbare Toevoeging (MTT) en Verwaarloosbare Toevoeging (VT) conform het beleid inzake straling. Voor van nature voorkomende stoffen is het MTR/VR samengesteld uit de achtergrondconcentratie (AC) en de MTT/VT. In formulevorm: MTR=AC+MTT en VR=AC+(MTT/100). De termen MTT en VT zijn door het RIVM geintroduceerd om aan te duiden wat de concentratie is die door antropogene activiteiten bovenop het achtergrondniveau toelaatbaar geacht wordt. De achtergrondconcentratie wordt als een gegeven beschouwd en de MTT is gelijk aan de antropogene toevoeging. Op de MTT kan indien noodzakelijk en gewenst, beleidsmatig een risicoreductiebeleid gebaseerd worden. De MTR's en VR's voor metalen zijn gebaseerd op de Toegevoegd Risico methode. Beleidsmatig wordt hierbij de achtergrondconcentratie als een gegeven beschouwd en kunnen de effecten veroorzaakt door (het beschikbare deel van) de achtergrondconcentratie genegeerd worden. Desondanks kan de toegevoegd risico methode worden toegepast om de mogelijke effecten van de beschikbare fractie van de achtergrondconcentratie voor metalen op het MTR te onderzoeken. In dit rapport is zulk een evaluatie uitgevoerd, waarbij verschillende niveaus van beschikbaarheid zijn aangenomen.

In this report Maximum Permissible Concentrations (MPC) are derived for 9 trace metals based on ecotoxicological data. The elements are: antimony, barium, beryllium, cobalt, molybdenum, selenium, thallium, tin, and vanadium The study was carried out in the framework of the project "Setting integrated environmental quality objectives". For the aquatic environment MPCs could be derived for all trace elements. These values were based on toxicity data for freshwater as well as saltwater organisms. Also a comparison was made between the sensitivity of saltwater and freshwater organisms for these trace elements and some other metals. The conclusion could be drawn that in acute tests freshwater and saltwater organisms are equally sensitive but that freshwater organisms are more sensitive in chronic tests. It should be stated however that much more data were available for freshwater organisms. For the soil only for cobalt a MPC could be derived. For the other trace elements no reliable toxicity data were available. For sediment dwelling organisms no MPC could be derived because no toxicity data were available for benthic organisms exposed via the sediment. Secondary poisoning was taken into account for the routes water --> fish --> fish-eating bird or mammal and soil --> earth-worm --> worm-eating bird or mammal. A 'screening-method was used in which only information from reviews was used. Only for thallium and selenium data were available for the aquatic route. From these data it could be concluded that there was no risk for secondary poisoning for these metals.

Rekentechnieken om meer informatie uit PET-beeld te halen Artsen baseren hun behandelplan onder andere op beelden van een PET-scan. Maar deze zijn niet altijd vrij van onvolkomenheden, bijvoorbeeld door ruis of strepen veroorzaakt door metalen implantaten. Bovendien komen twee artsen op basis van dezelfde beelden vaak tot een andere conclusie. Mehrsima Abdoli ontwikkelde in haar promotieonderzoek verschillende rekentechnieken om de onvolkomenheden in PET-beelden te corrigeren. Met de resultaten die deze opleveren kunnen artsen en radiologen een preciezere diagnose stellen. Abdoli bestudeerde twee problemen: de strepen die metalen implantaten veroorzaken op PET-beelden, met als gevolg een onjuist of verkeerd beeld van het weefsel in de buurt van dat implantaat, en de moeilijkheid om tot een accurate inschatting te komen van de omvang van een tumor. Met betrekking tot dat eerste probleem stelt ze dat er verschillende technieken zijn om het beeld te corrigeren, en dat een combinatie van twee technieken (sinogram en interpolatie-gebaseerde technieken) de beste resultaten geeft. In het tweede deel van haar proefschrift stelt Abdoli dat het correct aflijnen van tumorvolume op PET-beelden lastig is, door de beperkte resolutie en ruis van die beelden. Handmatig aflijnen levert vaak verschillende berekeningen op. Na vergelijking van verschillende segmentatietechnieken, waarbij de tumor laagje voor laagje bestudeerd wordt, concludeert ze dat mogelijk is om het ruisniveau van PET-beelden te verminderen door in plaats van die segmentatietechnieken een vervormbaar contourmodel te gebruiken.

Patiënten met metaal-op-metaal heupprothese jaarlijks onderzoeken Patiënten die zijn behandeld met een metaal-op-metaal heupprothese moeten jaarlijks worden onderworpen aan een poliklinisch onderzoek, waarbij de staat van de prothese en het omliggende weefsel middels echografie, MRI of CT-scan in beeld moet worden gebracht. Dat stelt Bart Bosker in zijn promotieonderzoek. Zijn onderzoek laat zien dat er bij zulke protheses veel vaker pijnlijke zwellingen (pseudotumoren) voorkomen dan tot nu toe werd gedacht. Na het plaatsen van een heupprothese kunnen verschillende complicaties ontstaan, zoals instabiliteit, verhoogde slijtage of verminderde bewegingsvrijheid van de prothese. Al deze complicaties hangen samen met een verkeerde positie van de cup – het onderdeel dat de ‘kom’ vervangt bij de totale heupprothese. De cup wordt meestal handmatig geplaatst door een chirurgisch orthopeed door zijn ‘timmermansoog’ te gebruiken. Op basis van een onderzoek onder tweehonderd patiënten toont Bosker aan dat deze techniek een hoge mate van onnauwkeurigheid laat zien, met dikwijls (omstreeks 30%) verkeerd geplaatste cups tot gevolg. Volgens hem verdient computernavigatie in de toekomst de voorkeur. In het tweede deel van zijn onderzoek richtte Bosker zich op de problemen rondom metaal-op-metaal prothesen. Hij ontdekte dat bij dat type prothese veel vaker pseudotumoren voorkomen (rond 35%-40% in twee patiëntengroepen) dan tot nu toe bekend was. Tevens is er een verhoogde kans op vroeg materiaalfalen door een specifiek corrosieproces, uniek voor de onderzochte prothese. Dat pleit volgens hem voor een goede, jaarlijkse controle van patiënten met een metaal-op-metaal prothese.

Dit rapport beschrijft de opzet, evaluatie en resultaten van een modellering van uitloging van metalen en metalloïden uit bodems naar het grondwater. Deze studie had de volgende doelen: 1. het bepalen van representatieve concentraties van metalen en metalloïden in het ondiepe (eerste meter) grondwater; 2. het bepalen van representatieve waarden voor bodemparameters, die de uitloging van metalen en metalloïden bepalen, zoals reactiviteit van opgelost organische koolstof, gehalten aan oxiden en reactiviteit van metalen en metalloïden; 3. het verbeteren van het bestaande modelinstrumentarium voor het modelleren van de uitloging van metalen en metalloïden uit de bodem, inclusief de onzekerheid van deze modellen.

Abdoli bestudeerde twee problemen: de strepen die metalen implantaten veroorzaken op PET-beelden, met als gevolg een onjuist of verkeerd beeld van het weefsel in de buurt van dat implantaat, en de moeilijkheid om tot een accurate inschatting te komen van de omvang van een tumor. Met betrekking tot dat eerste probleem stelt ze dat er verschillende technieken zijn om het beeld te corrigeren, en dat een combinatie van twee technieken (sinogram en interpolatie-gebaseerde technieken) de beste resultaten geeft. In het tweede deel van haar proefschrift stelt Abdoli dat het correct aflijnen van tumorvolume op PET-beelden lastig is, door de beperkte resolutie en ruis van die beelden. Handmatig aflijnen levert vaak verschillende berekeningen op. Na vergelijking van verschillende segmentatietechnieken, waarbij de tumor laagje voor laagje bestudeerd wordt, concludeert ze dat mogelijk is om het ruisniveau van PET-beelden te verminderen door in plaats van die segmentatietechnieken een vervormbaar contourmodel te gebruiken.

Nanoporeuze metallische schuimen fabriceren met blokcopolymeren Het proefschrift van Ivana Vukovic gaat over metallische nanoschuimen, een nieuwe klasse van 3D-nanomaterialen waarin eigenschappen van metalen en nanoporeuze materialen worden gecombineerd. De unieke combinatie zorgt ervoor dat nanoporeuze metallische schuimen kunnen worden gebruikt voor waterstofopslag, actuatoren, nieuwe 3D-gestructureerde batterijen, vervangers voor platinum-katalysatoren, etc. Veelgebruikte technieken, zoals het scheiden van legeringen, sol-gel synthese, nanosmelten, verbrandingssynthese, etc., leveren over het algemeen materialen op met een ongeordende architectuur, iets wat een nadelig effect kan hebben op de mechanisch eigenschappen. Blokcopolymeren daarentegen kunnen spontaan overgaan in bi-continue geordende nanostructuren. Vervolgens kunnen ze worden toegepast als een soort mal (op nanoschaal) voor de het maken van goedgeordende metallische nanoschuimen. Het idee om deze blokcopolymeren te gebruiken voor de fabricatie van deze nanoschuimen wordt verder uitgewerkt en beschreven in dit proefschrift, met de daarbij behorende experimentele resultaten en toekomstperspectieven. Daarnaast wordt het fasegedrag onderzocht van supramoleculaire complexen die op blokcopolymeren zijn gebaseerd en die worden gebruikt als voorlopers voor goedgeordende metallische nanoschuimen.

Rekentechnieken om meer informatie uit PET-beeld te halen Artsen baseren hun behandelplan onder andere op beelden van een PET-scan. Maar deze zijn niet altijd vrij van onvolkomenheden, bijvoorbeeld door ruis of strepen veroorzaakt door metalen implantaten. Bovendien komen twee artsen op basis van dezelfde beelden vaak tot een andere conclusie. Mehrsima Abdoli ontwikkelde in haar promotieonderzoek verschillende rekentechnieken om de onvolkomenheden in PET-beelden te corrigeren. Met de resultaten die deze opleveren kunnen artsen en radiologen een preciezere diagnose stellen. Dit rapport beschrijft de opzet, evaluatie en resultaten van een modellering van uitloging van metalen en metalloïden uit bodems naar het grondwater. Deze studie had de volgende doelen: 1. het bepalen van representatieve concentraties van metalen en metalloïden in het ondiepe (eerste meter) grondwater; 2. het bepalen van representatieve waarden voor bodemparameters, die de uitloging van metalen en metalloïden bepalen, zoals reactiviteit van opgelost organische koolstof, gehalten aan oxiden en reactiviteit van metalen en metalloïden; 3. het verbeteren van het bestaande modelinstrumentarium voor het modelleren van de uitloging van metalen en metalloïden uit de bodem, inclusief de onzekerheid van deze modellen.

De risicogrenzen voor sediment zijn wel berekend met behulp van evenwichtspartitie bij gebrek aan toxiciteitsgegevens. In het algemeen zijn de nieuw afgeleide milieurisicogrenzen voor sediment weinig veranderd ten opzichte van de bestaande. Het gemiddelde van gemeten concentraties van beryllium, vanadium, kobalt en barium in zoet oppervlaktewater overschrijdt het maximaal toelaatbaar risiconiveau (MTR). Voor selenium en antimoon was dit niet het geval. Metaalschuimen zijn materialen die enerzijds een microstructuur bevatten op de microschaal (de schaal van de celwanden), maar anderzijds ook een mesoscopische microstructuur hebben (op de schaal van de celstructuur). Deze micro- en mesoscopische structuren zijn gevoelig voor het productieproces van het schuim en beïnvloeden bovendien de mechanische eigenschappen. Het doel van dit proefschrift is te onderzoeken wat de invloed van deze twee structuurniveaus op de mesoscopische vervormings- en schademechanismen is en te begrijpen hoe deze de macroscopische mechanische eigenschappen beïnvloeden. Voor dit doel werd een meerschalig model van een tweedimensionaal schuim ontwikkeld, waarbij Voronoi tessellaties werden gebruikt om de cellulaire structuur te beschrijven. Door nauwgezet de plastische vloeigrens in de celwanden op cellulair niveau te volgen, kon een geometrisch verstevigingsmechanisme worden geïdentificeerd dat hand in hand gaat met het intrinsieke verstevigen van het metaal zelf. Tevens kon worden vastgesteld dat de competitie tussen versteviging door plastische deformatie en verzwakking ('softening') door inwendige schadeontwikkeling belangrijk is bij het bepalen van de macroscopische ductiliteit. Met toenemende intrinsieke versteviging wordt de ontwikkeling van buiging in de celwanden afgeremd en daarmee ook de initiatie van schade, hetgeen de treksterkte en ductiliteit van het schuim verhoogt. In onregelmatige celstructuren met een brede verdeling van celgrootten is de mesoscopische deformatieverdeling niet-uniform, hetgeen leidt tot een verlaging van de ductiliteit. Voor schuimen met een structurele anisotropie werd aangetoond dat het schuim sterker en minder ductiel is in de trekrichting parallel aan de grootste cel-as, vergeleken met een trekrichting daar loodrecht op. Op basis van de numerieke berekeningen konden eenvoudige uitdrukkingen worden afgeleid die de macroscopische versteviging, treksterkte en ductiliteit beschrijven in termen van de materiaaleigenschappen van de celwanden en geometrische parameters van de cellulaire architectuur. Ook is er onderzoek gedaan naar het effect van interne scheuren op de treksterkte. Vastgesteld werd dat proefstukken met rand-kerven ('edge-notches') verstevigingsgedrag vertonen, terwijl proefstukken met inwendige scheuren juist verzwakken. Ten slotte is in dit proefschrift ook het falen van poreus bot ('trabecular bone') onderzocht, waarbij de effectiviteit van twee klinische behandelmethoden voor osteoporose werden vergeleken.

Streptococcus pneumoniae is een grampositieve menselijke ziekteverwekker die jaarlijks verantwoordelijk is voor de dood van miljoenen mensen, voornamelijk kinderen, en veroorzaakt pneumokokken infecties zoals longontsteking, hersenvliesontsteking, middenoor ontsteking en sepsis. De bacterie leeft onder normale omstandigheden in de neusholte. Soms kan het zich, als de omstandigheden juist zijn, door het lichaam naar andere plekken verplaatsen en daar infecties veroorzaken. Om in deze zeer verschillende gastheer omgevingen te overleven en infecties te veroorzaken moet S. pneumoniae deze aanzienlijke variaties in omgevingsomstandigheden kunnen registreren en zich daaraan aan kunnen passen. Hieronder vallen de veranderingen in concentraties metaalionen in gastheer weefsel en de beschikbaarheid van verschillende koolstofbronnen. Er is weinig bekend over het mechanisme van interactie van deze humane pathogeen met de veranderende omgeving in het menselijk lichaam. Dit proefschrift beschrijft de gen gereguleerde respons van S. pneumoniae op de variërende metaal en koolhydraat beschikbaarheid. Wisselende metaal concentraties en verschillende koolhydraat bronnen zijn factoren waar hij mee geconfronteerd kan worden in verschillende plekken van het menselijk lichaam tijdens de infectie. Een aantal regulatoren die reageren op verschillende metaal (b.v. zink en koper) en koolhydraat bronnen (b.v. cellobiose) zijn gekarakteriseerd en hun regulonen zijn bestudeerd. De resultaten beschreven in dit proefschrift zullen helpen bij het beter begrijpen van de moleculaire biologie van S. pneumoniae en verschaffen meer inzicht in de respons van S. pneumoniae op veranderende omgevingscondities. Zink is een belangrijk, maar toxisch, zwaar metaal ion dat de expressie van een aantal (virulentie) genen in streptococci reguleert. Eerder hebben we de genoom-brede respons van S. pneumoniae op zink-stress geanalyseerd. In hoofdstuk 2 hebben we een analyse van het transcriptoom gedaan om genen te identificeren die differentieel tot expressie komen bij intracellulaire zink limitatie. Dit leidde tot een aantal genen die sterk omhoog gereguleerd zijn in de afwezigheid van extracellulaire zink. Hieronder vallen de genen die behoren tot het regulon van de zink-responsieve repressor AdcR, zoals adcBCA, coderend voor een zinkafhankelijk ABC-opname systeem, adcAII, coderend voor een zink-bindend lipoproteïne, en ook virulentie genen behorend tot de Pht familie (PhtA, PhtB, PhtD en PhtE). Met transcriptoom analyse, lacZ-reporter studies, in vitro DNA-binding experimenten, en in silico operator voorspellingen, laten we zien dat AdcR direct de promoters van adcRCBA, adcAIIphtD, phtA, phtB en phtE onderdrukt in de aanwezigheid van zink.

Metaalschuimen zijn materialen die enerzijds een microstructuur bevatten op de microschaal (de schaal van de celwanden), maar anderzijds ook een mesoscopische microstructuur hebben (op de schaal van de celstructuur). Deze micro- en mesoscopische structuren zijn gevoelig voor het productieproces van het schuim en beïnvloeden bovendien de mechanische eigenschappen. Het doel van dit proefschrift is te onderzoeken wat de invloed van deze twee structuurniveaus op de mesoscopische vervormings- en schademechanismen is en te begrijpen hoe deze de macroscopische mechanische eigenschappen beïnvloeden. Voor dit doel werd een meerschalig model van een tweedimensionaal schuim ontwikkeld, waarbij Voronoi tessellaties werden gebruikt om de cellulaire structuur te beschrijven. Door nauwgezet de plastische vloeigrens in de celwanden op cellulair niveau te volgen, kon een geometrisch verstevigingsmechanisme worden geïdentificeerd dat hand in hand gaat met het intrinsieke verstevigen van het metaal zelf. Tevens kon worden vastgesteld dat de competitie tussen versteviging door plastische deformatie en verzwakking ('softening') door inwendige schadeontwikkeling belangrijk is bij het bepalen van de macroscopische ductiliteit. Met toenemende intrinsieke versteviging wordt de ontwikkeling van buiging in de celwanden afgeremd en daarmee ook de initiatie van schade, hetgeen de treksterkte en ductiliteit van het schuim verhoogt. In onregelmatige celstructuren met een brede verdeling van celgrootten is de mesoscopische deformatieverdeling niet-uniform, hetgeen leidt tot een verlaging van de ductiliteit. Voor schuimen met een structurele anisotropie werd aangetoond dat het schuim sterker en minder ductiel is in de trekrichting parallel aan de grootste cel-as, vergeleken met een trekrichting daar loodrecht op. Op basis van de numerieke berekeningen konden eenvoudige uitdrukkingen worden afgeleid die de macroscopische versteviging, treksterkte en ductiliteit beschrijven in termen van de materiaaleigenschappen van de celwanden en geometrische parameters van de cellulaire architectuur. Ook is er onderzoek gedaan naar het effect van interne scheuren op de treksterkte. Vastgesteld werd dat proefstukken met rand-kerven ('edge-notches') verstevigingsgedrag vertonen, terwijl proefstukken met inwendige scheuren juist verzwakken. Ten slotte is in dit proefschrift ook het falen van poreus bot ('trabecular bone') onderzocht, waarbij de effectiviteit van twee klinische behandelmethoden voor osteoporose werden vergeleken.

Hieronder vallen de veranderingen in concentraties metaalionen in gastheer weefsel en de beschikbaarheid van verschillende koolstofbronnen. Er is weinig bekend over het mechanisme van interactie van deze humane pathogeen met de veranderende omgeving in het menselijk lichaam. Dit proefschrift beschrijft de gen gereguleerde respons van S. pneumoniae op de variërende metaal en koolhydraat beschikbaarheid. Wisselende metaal concentraties en verschillende koolhydraat bronnen zijn factoren waar hij mee geconfronteerd kan worden in verschillende plekken van het menselijk lichaam tijdens de infectie. Een aantal regulatoren die reageren op verschillende metaal (b.v. zink en koper) en koolhydraat bronnen (b.v. cellobiose) zijn gekarakteriseerd en hun regulonen zijn bestudeerd. De resultaten beschreven in dit proefschrift zullen helpen bij het beter begrijpen van de moleculaire biologie van S. pneumoniae en verschaffen meer inzicht in de respons van S. pneumoniae op veranderende omgevingscondities. Zink is een belangrijk, maar toxisch, zwaar metaal ion dat de expressie van een aantal (virulentie) genen in streptococci reguleert. Eerder hebben we de genoom-brede respons van S. pneumoniae op zink-stress geanalyseerd. In hoofdstuk 2 hebben we een analyse van het transcriptoom gedaan om genen te identificeren die differentieel tot expressie komen bij intracellulaire zink limitatie. Dit leidde tot een aantal genen die sterk omhoog gereguleerd zijn in de afwezigheid van extracellulaire zink. Hieronder vallen de genen die behoren tot het regulon van de zink-responsieve repressor AdcR, zoals adcBCA, coderend voor een zinkafhankelijk ABC-opname systeem, adcAII, coderend voor een zink-bindend lipoproteïne, en ook virulentie genen behorend tot de Pht familie (PhtA, PhtB, PhtD en PhtE). Met transcriptoom analyse, lacZ-reporter studies, in vitro DNA-binding experimenten, en in silico operator voorspellingen, laten we zien dat AdcR direct de promoters van adcRCBA, adcAIIphtD, phtA, phtB en phtE onderdrukt in de aanwezigheid van zink.

Metaalschuimen zijn materialen die enerzijds een microstructuur bevatten op de microschaal (de schaal van de celwanden), maar anderzijds ook een mesoscopische microstructuur hebben (op de schaal van de celstructuur). Deze micro- en mesoscopische structuren zijn gevoelig voor het productieproces van het schuim en beïnvloeden bovendien de mechanische eigenschappen. The objective of this research is the optimization of essential metal dosing in upflow anaerobic sludge bed (UASB) reactors used for methanogenic wastewater treatment. Optimization of essential metal dosing in UASB reactors is a compromise between achieving the maximal biological activity of the biomass present in the reactor, while minimizing the costs of the supplied metal and the metal losses into the environment. The fate of metals in the anaerobic granular sludge is studied. The boundary conditions to keep a stable reactor operation are evaluated: Nutrient deficiency due to lack of essential metals versus toxicity in case of their excess. Finally, metal dosing strategies are evaluated. It is shown that the metal addition to UASB reactors and the methanogenic activity are related and, more important, this relation can be quantified

This thesis addresses the geochemical aspects of AVS (Acid Volatile Sulfide) and SEM (Simultaneously Extracted Metals) in floodplain lake sediment, its spatial distribution in floodplain lakes and dynamics over time, the link with effects on single species (bioassays), as well as the impact of excess SEM on the in situ benthic community, in one consistent methodological approach. Using this approach the SEM-AVS concept is thus evaluated on various levels from geochemical through ecotoxicology towards ecology. Furthermore, the aspect of spatial and temporal variability is addressed. For the case studies in the field, several floodplain lakes located in the floodplain area Afferdensche and Deestsche Waarden along the Waal branche of the Rhine were selected. Occasionally, this selection was extended towards other floodplain lakes along the Rhine branches. The main goal of this research is a) to explore the actual risks of trace metals to benthic communities in floodplain systems and b) to evaluate the SEM-AVS concept as a tool to assess bioavailable concentrations of trace metals in dynamic floodplain sediment

Materials have been used throughout history for their structural properties, e.g. ductility, elasticity, hardness etc., and later also for their physical properties, i.e., for their characteristic response to external perturbances. These last properties have been investigated in this thesis by using quatumechanics. The mutual interactions of the many-particles which constitute matter complicate its theoretical description enormously. Time-dependent density-functional theory provides a powerful tool to investigate these dynamic properties in atoms, molecules, and clusters. Indeed this theory maps the many-particle problem of interacting electrons in a time-dependent external field onto an auxiliary system of noninteracting particles in an effective field. The many-particle effects enter through so-called exchange-correlation contributions to the effective potentials describing this field. These effective potentials are uniquely determined by the time-dependent density for a given initial state. Essential in this formulation is that the time-dependent density is exactly the same in the real and auxiliary system. The time-dependent density can thus be obtained from the auxiliary system once we know the effective potentials, which in turn are functionals of the time-dependent density. This requires a self-consistent procedure in which one updates the approximate densities and potentials in an iterative way. In this thesis we have only considered systems, initially in the ground state, which are perturbed only weakly by an external electromagnetic field. In this so-called linear response regime the induced density is proportional to the applied field. The exchange-correlation potentials depend on the density in a nontrivial way: the potentials at time t and position r depend in general on the density at all earlier times t' and at all positions r'. In the linear response regime this relation is expressed through a so-called exchange-correlation response kernel. The most common approximation for the exchange-correlation potentials is the adiabatic local density approximation (ALDA), in which these potentials are instantaneous and local functionals of the density. The scenario becomes more complicated for solids, which can be treated as 'open' systems, i.e., systems without boundary. To fully describe these systems, the density is replaced by the current-density leading to time-dependent current-density-functional theory (TDCDFT). In this thesis we have formulated the linear response of metallic crystalline systems to an electric field within this theoretical framework. The method gives already reasonable results for the dielectric and electron energy-loss functions within the ALDA. Although this approximation describes well the interband region of the spectra, it is less satisfying in the intraband region, where relaxation processes, such as electron-electron and electron-phonon scattering, become more important. Indeed the ALDA fails to reproduce the Drude-like tail in the absorption spectra. Approximations to the exchange-correlation potentials which are local-functionals of the current-density and hence nonlocal-functionals of the density can be included in a natural way in our formulation. In these approximations the exchange-correlation kernels can have memory, and thus relaxation processes due to electron-electron scattering can be taken into account. Other types of deviations from experiments can be observed in systems containing heavy elements, if relativistic corrections are neglected. Our method can treat scalar relativistic effects and spin-orbit coupling both in the ground-state and response calculations by using the zeroth-order regular approximation (ZORA). The spin-dependent formulation of the method, which allows to treat spin-orbit coupling, represents a first step to describe the linear response to an external magnetic field.

In practice these metals are supplied to the influent when they are absent or insufficiently present in the wastewater. This supply is generally not very rational. However, such a rational metal supply is desired in order to reduce the costs, to minimize the introduction into the environment and to maximize the biological activity. To achieve this insight, in the trace metal dynamics in anaerobic granular sludge bed reactors is required. This thesis therefore focuses on the retention, accumulation and release of trace metals in anaerobic granular sludge and the factors affecting these processes. Further the impact of metal presence/absence and dosing on the anaerobic conversion of methanol was addressed

Dynamic metal speciation analysis in aquatic ecosystems is emerging as a powerful basis for development of predictions of bioavailability and reliable risk assessment strategies. A given speciation sensor is characterized by an effective time scale or kinetic window that defines the measurable metal species via their labilities. Here we review the current state of the art for the theory and application of dynamic speciation sensors. We show that a common dynamic interpretation framework, based on rigorous flux expressions incorporating the relevant diffusion and reaction steps, is applicable for a suite of sensors that span a range of time scales. Interpolation from a kinetic spectrum of speciation data is proposed as a practical strategy for addressing questions of bioavailability. Case studies illustrate the practical significance of knowledge on the dynamic features of metal complex species in relation to biouptake, and highlight the limitations of equilibrium-based models.

It was possible to combine HRTEM observations with the analysis of the chemical composition within one monolayer at the metal-oxide interfaces. With in-situ experiments, a detailed visualization of order-disorder transitions on an atomic scale at metal-oxide interfaces could be realized.
