Mantis shrimp (Stomatopoda) are extant, marine, predatory arthropods, but these malacostracan pancrustaceans are also occasionally preserved in fossil assemblages, particularly in Carboniferous and Cretaceous deposits. Carboniferous species fall into two suborders—Palaeostomatopodea and Archaeostomatopodea—and represent the ancestral forms that gave rise to modern lineages. Herein, we describe hitherto unknown specimens belonging to the archaeostomatopod genus Tyrannophontes from the Pennsylvanian-aged Wea Shale Member, eastern Nebraska. We explore the preservation of these fossils using scanning electron microscopy and energy dispersive X-ray spectroscopy. These approaches reveal additional morphological characteristics, including unique appendicular data, such as the earliest occurrence of biramous gilled appendages in Stomatopoda. Trilobites with malformations present important insight into the paleoecology of the wholly extinct arthropod group. This includes records of failed predation, developmental complications, and parasitic interactions. The documentation of malformed trilobites therefore allows for a more thorough understanding of these animals. Such summaries also permit larger-scale, synthetic works that consider patterns and processes associated with malformations to be developed. To expand the current record of malformed Cambrian through to Devonian trilobites, we report 16 novel specimens across 12 genera from deposits spanning Australia, Bolivia, Canada, the United States, and Wales. These specimens illustrate examples of injuries potentially caused by predation, molting, and accidental injury, teratological malformations, and pathological infestation. Possible predators; explanations for developmental, genetic, and recovery issues; as well as infestations are considered. Comparison of size distributions of malformed and nonmalformed Elrathia kingii indicates that primarily the largest specimens preserve malformations. This implies that either smaller specimens fully recovered from malformations or did not survive once the malformations were incurred. Continued documentation of malformed specimens will allow larger datasets to be compiled and further promote the use of malformations in understanding trilobite paleoecology. [ABSTRACT FROM AUTHOR]

Malformed trilobites have been well documented within the arthropod fossil record and serve as valuable evidence for illustrating aspects of trilobite paleoecology associated with development, predation, and pathologies. Ongoing efforts to comprehensively document these malformations have uncovered important, often unique records for the extinct group, shedding light on post-malformation recovery processes and potential predators. A key means of recording new examples of these specimens has been the examination of historically important paleontological collections. To expand this approach, we examined the Deiss collection in the Indiana University Paleontology Collection and present four examples of malformations from Cambrian (Asaphiscus wheeleri, Dorypyge bispinosa, Wanneria walcottana), and Ordovician (Isotelus iowensis) species. These specimens reveal evidence of injuries, failed predation, and developmental complications. We explore the possible explanations for malformations and consider the current state of the art for evaluating trilobite malformations. The Bear Gulch Limestone houses a diverse, exceptionally preserved marine fauna from the early Carboniferous. A wealth of vertebrate and invertebrate forms has previously been recorded from this deposit, including fish, annelids, and several arthropods. To expand the record of Bear Gulch marine arthropods, a new enigmatic, possibly blind euchelicerate, Titanoprosoma edgecombei, gen. et sp. nov., is described. The new euchelicerate taxon displays a hypertrophied, ovate, and structureless prosoma—a morphology unique among marine euchelicerates. We explore how the large prosoma and lack of ocular structures reflect possible adaptations to an infaunal, burrowing lifestyle. This species represents the fourth euchelicerate genus described from the Bear Gulch Limestone, further highlighting the impressive disparity of marine arthropods preserved in the deposit. The addition of novel invertebrate forms found in previously unknown museum material suggests that the Bear Gulch Limestone likely houses a still undocumented diversity of Carboniferous arthropods. Biomineralised remains of trilobites provide important insight into the evolutionary history of a diverse, extinct group of arthropods. Their exoskeletons are also ideal for recording malformations, including evidence of post-injury repair. Re-examination of historic collections and the study of new specimens is important for enhancing knowledge on trilobite malformations across this diverse clade. To expand the records of these abnormalities and present explanations for their formation, we document eight malformed trilobite specimens, as well as one carcass, housed within the Commonwealth Palaeontological Collection at Geoscience Australia in Canberra. We present examples of Asthenopsis, Burminresia, Centropleura, Coronocephalus, Dolicholeptus, Galahetes, Papyriaspis, and Xystridura from Cambrian, Ordovician, and Silurian deposits of Australia. Most of the malformed specimens show W-, U-, or L-shaped indentations that reflect injuries from either failed predation or complications during moulting, and a mangled carcass is ascribed to either successful predation or post-mortem scavenging. We also uncover examples of teratologies, such as bifurcated pygidial ribs and pygidial asymmetry, in addition to evidence of abnormal recovery (i.e., fusion of thoracic segments) from a traumatic incident. Injured trilobites present insight into how a completely extinct group of arthropods responded to traumatic experiences, such as failed predation and moulting complications. These specimens are therefore important for more thoroughly understanding the Paleozoic predator-prey systems that involved trilobites. To expand the record of injured trilobites, we present new examples of injured Ogygopsis klotzi and Olenoides serratus from the Campsite Cliff Shale Member of the Burgess Shale Formation (Cambrian, Miaolingian, Wuliuan), Paradoxides (Paradoxides) paradoxissimus gracilis from the Jince Formation (Cambrian, Miaolingian, Drumian), Ogygiocarella angustissima from the Llanfawr Mudstones Formation (Middle-Late Ordovician, Darriwilian-Sandbian), and Ogygiocarella debuchii from the Meadowtown Formation, (Middle-Late Ordovician, Darriwilian-Sandbian). We consider the possible origins of these malformations and conclude that most injuries reflect failed predation. Within this framework, possible predators are presented, and we uncover a marked shift in the diversity of animals that targeted trilobites in the Ordovician. We also collate other records of injured Ogygo. klotzi and Ol. serratus, and Ogygi. debuchii, highlighting that these species are targets for further understanding patterns and records of trilobite injuries. Abnormal trilobites present insight into how arthropods with fully biomineralised exoskeletons recovered from injuries, genetic malfunctions, and pathologies. Records of abnormal Silurian trilobites in particular show an abundance of specimens with teratologies and a limited record of injuries. Here we expand the record of abnormal Silurian trilobites by presenting seven new abnormal specimens of Odontopleura (Sinespinaspis) markhami from the early Silurian (Llandovery, Telychian) Cotton Formation, New South Wales. We use these specimens to illustrate novel evidence for asymmetric distribution of pleural thoracic spine bases. These abnormal bases likely reflect genetic complications, resulting in morphologies that would unlikely have aided the fitness of abnormal individuals. In considering records of malformed Silurian trilobites more broadly, we propose that the largest trilobites may have been prey at this time. Compared with other Mesozoic tetrapod groups, chigutisaurid fossils from Australia are rare, with only three named taxa described from the continent. From Queensland, Keratobrachyops australis is known from the Triassic, and Siderops kehli from the Jurassic. Koolasuchus cleelandi, from the Cretaceous of Victoria, represents the youngest-known temnospondyl globally. Here we describe the first chigutisaurid from New South Wales, from the Early–Middle Triassic Terrigal Formation. The specimen (AM F125866) comprises an articulated, near-complete skeleton, presented in ventral aspect, as well as outlines of soft tissue. The new taxon preserves features that indicate an affinity with Chigutisauridae, confirming previous hypotheses on the presence of large-bodied chigutisaurids in the Triassic of Australia. The new Terrigal chigutisaurid is only the second chigutisaurid known from the Lower Triassic and fourth from Australia overall. The distribution of brachycephalic temnospondyls throughout the Mesozoic suggests specific adaptations led to the long-term survival of chigutisaurids, especially across the end-Triassic extinction event. Records of abnormal fossil arthropods present important insight into how extinct forms responded to traumatic damage and developmental complications. Trilobites, bearing biomineralized dorsal exoskeletons, have arguably the most well-documented record of abnormalities spanning the Cambrian through the end-Permian. As such, new records of malformed, often injured, trilobites are occasionally identified. To further expand the documentation of abnormal specimens, we describe malformed specimens of Lyriaspis sigillum Whitehouse, 1939, Zacanthoides sp. indet., Asaphiscus wheeleri Meek, 1873, Elrathia kingii (Meek, 1870), and Ogygiocarella debuchii (Brongniart, 1822) from lower Paleozoic deposits. In considering these forms, we propose that they illustrate examples of injuries, and that the majority of these injuries reflect failed predation. We also considered the origin of injuries impacting singular segments, suggesting that these could reflect predation, self-induced damage, or intraspecific interactions during soft-shelled stages. Continued examination of lower Paleozoic trilobite injuries will further the understanding of how trilobites functioned as prey and elucidate how disparate trilobite groups recovered from failed attacks. We document a case history of Northridge earthquake induced movement of a coherent landslide mass comprised principally of weathered, previously sheared siltstone. Information on the residual shear strength of the material through which the sliding occurred is available both from backanalysis of a static (rainfall-induced) failure on the same slope and laboratory test data. Postearthquake field reconnaissance data establishes the seismic slope displacements near the slope crest. A yield coefficient is estimated for the slope based on residual shear strengths and measured groundwater levels at the time of the Northridge earthquake. This yield coefficient is used with four horizontal equivalent acceleration time histories (that approximate the seismic demand within the slide mass) in Newmark sliding block displacement analyses. The calculated displacements range from approximately 20 to 90 mm, which compares well with observed displacements at the slide scarp of approximately 50 mm. Seismic compression is defined as the accrual of contractive volumetric strain in unsaturated soil during strong shaking by earthquakes. We document and analyze two case histories (denoted school site and site A) of ground deformation from seismic compression in canyon fills strongly shaken by the Northridge earthquake. Site A had ground settlements up to about 18 cm, which damaged a structure, while the school site had settlements up to about 6 cm. For each site, we perform decoupled analyses of shear and volumetric strain. Shear strain is calculated using one-dimensional and two-dimensional ground response analyses, while volumetric strain is evaluated from shear strain using material-specific models derived from simple shear laboratory testing that incorporates important effects of fines content and as-compacted density and saturation. Analyses are repeated using a logic tree approach in which weights are assigned to multiple possible realizations of uncertain model parameters. At the school site, predicted settlements appear to be unbiased. At site A, the analyses successfully predict the shape of the settlement profile along a section, but the weighted average predictions are biased slightly too low. We speculate that the apparent site A bias can be explained by limited resolution of the site stratigraphy, bias in laboratory-derived volumetric strain models, and/or uncertainty in the estimated earthquake-induced settlements. Constraining the timing of morphological innovations within xiphosurid evolution is central for understanding when and how such a long-lived group exploited vacant ecological niches over the majority of the Phanerozoic. To expand the knowledge on the evolution of select xiphosurid forms, we reconsider the four Australian taxa: Austrolimulus fletcheri, Dubbolimulus peetae, Tasmaniolimulus patersoni, and Victalimulus mcqueeni. In revisiting these taxa, we determine that, contrary to previous suggestion, T. patersoni arose after the Permian and the origin of over-developed genal spine structures within Austrolimulidae is exclusive to the Triassic. To increase the availability of morphological data pertaining to these unique forms, we also examined the holotypes of the four xiphosurids using synchrotron radiation X-ray tomography (SRXT). Such non-destructive, in situ imaging of palaeontological specimens can aid in the identification of novel morphological data by obviating the need for potentially extensive preparation of fossils from the surrounding rock matrix. This is particularly important for rare and/or delicate holotypes. Here, SRXT was used to emphasize A. fletcheri and T. patersoni cardiac lobe morphologies and illustrate aspects of the V. mcqueeni thoracetronic doublure, appendage impressions, and moveable spine notches. Unfortunately, the strongly compacted D. peetae precluded the identification of any internal structures, but appendage impressions were observed. The application of computational fluid dynamics to high-resolution 3D reconstructions are proposed to understand the hydrodynamic properties of divergent genal spine morphologies of austrolimulid xiphosurids. Infection with the helminth Schistosoma mansoni results in hepatointestinal granulomatous inflammation mediated by CD4 T cells directed against parasite eggs. The severity of disease varies greatly in humans and mice; however, the genetic basis of such a heterogenous immune response remains poorly understood. Here we show that, despite their close genetic relationship, C57BL/10SnJ (B10) mice developed significantly more pronounced immunopathology and higher T helper 17 cell responses than C57BL/6J (B6) mice. Similarly, live egg-stimulated B10-derived dendritic cells (DCs) produced significantly more IL-1β and IL-23, resulting in higher IL-17 production by CD4 T cells. Gene expression analysis disclosed a heightened proinflammatory cytokine profile together with a strikingly lower expression of Ym1 in B10 versus B6 mice, consistent with failure of B10 DCs to attain alternative activation. To genetically dissect the differential response, we developed and analyzed congenic mouse strains that capture major regions of allelic variation, and found that the level of inflammation was controlled by a relatively small number of genes in a locus mapping to chromosome 4 117-143 MB. Our study has thus identified novel genomic regions that regulate the severity of the schistosome infection by way of controlling the mode of DC activation and consequent CD4 T-cell subset development. Background: Individuals with bipolar disorder often endorse dysfunctional beliefs consistent with cognitive models of bipolar disorder (Beck, 1976; Mansell, 2007). Aims: The present study sought to assess whether young adult offspring of those with bipolar disorder would also endorse these beliefs, independent of their own mood episode history. Method: Participants (N = 89) were young adult college students with a parent with bipolar disorder (n = 27), major depressive disorder (MDD; n = 30), or no mood disorder (n = 32). Semi-structured interviews of the offspring were used to assess diagnoses. Dysfunctional beliefs related to Beck and colleagues’ (2006) and Mansell's (2007) cognitive models were assessed. Results: Unlike offspring of parents with MDD or no mood disorder, those with a parent with bipolar disorder endorsed significantly more dysfunctional cognitions associated with extreme appraisal of mood states, even after controlling for their own mood diagnosis. Once affected by a bipolar or depressive disorder, offspring endorsed dysfunctional cognitions across measures. Conclusions: Dysfunctional cognitions, particularly those related to appraisals of mood states and their potential consequences, are evident in young adults with a parent who has bipolar disorder and may represent targets for psychotherapeutic intervention. Infection with the trematode parasite Schistosoma mansoni results in distinct heterogeneity of disease severity both in humans and in mice. In the experimental mouse model, severe disease is characterized by pronounced hepatic egg-induced granulomatous inflammation mediated by CD4 Th17 cells, whereas mild disease is associated with reduced hepatic inflammation in a Th2-skewed cytokine environment. Even though the host's genetic background significantly impacts the clinical outcome of schistosomiasis, specific gene(s) that contribute to disease severity remain elusive. We investigated the schistosome infection in wild-derived mice, which possess a more diverse gene pool than classically inbred mouse strains and thus makes them more likely to reveal novel mechanisms of immune regulation. We now show that inbred wild-derived MOLF mice develop severe hepatic inflammation with high levels of IL-17. Congenic mice with a MOLF locus in chromosome 6, designated Why1, revealed high pathology and enabled the identification of Irak2 as the pathogenic gene. Although IRAK-2 is classically associated with TLR signaling, adoptive transfer of CD4 T cells revealed that IRAK-2 mediates pathology in a CD4 T cell specific manner by promoting Th17 cell development through enhancement of IL-1b-induced activation of transcription factors RORct and BATF. The use of wild-derived mice unravels IRAK-2 as a novel regulator of IL-1- nduced pathogenic Th17 cells in schistosomiasis, which likely has wide-ranging implications for other chronic inflammatory and autoimmune diseases. Abstract: Glycoprotein G (gG) of equine herpesvirus type 1 (EHV-1), a structural component of virions and secreted from virus-infected cells, was shown to bind to a variety of different chemokines and as such might be involved in immune modulation. Little is known, however, about its role in the replication cycle and infection of EHV-1 in vivo. Here we report on the function of gG in context of virus infection in vitro and in vivo. A gG deletion mutant of pathogenic EHV-1 strain RacL11 (vL11ΔgG) was constructed and analyzed. Deletion of gG had virtually no effect on the growth properties of vL11ΔgG in cell culture when compared to parental virus or a rescuant virus vL11ΔgGR, respectively, and virus titers and plaque formation were unaffected in the absence of the glycoprotein. Similarly, in the murine model of EHV-1 infection, no significant differences in virulence between the gG deletion mutant and RacL11 or vL11ΔgGR were found at high doses of infection. However, infection of mice at lower doses revealed that the gG deletion mutant was able to replicate to higher titers in lungs of infected mice. Additionally, these mice lost significantly more weight than those infected with RacL11 and a more pronounced inflammatory response in lungs was observed. Therefore we concluded that deletion of gG in EHV-1 seems to lead to an exacerbation of respiratory disease in the mouse. [Copyright &y& Elsevier]

Wild-type equine herpesvirus 1 (EHV-1) strains express a large (250-kDa) glycoprotein, gp2, that is encoded by EUs4 (gene 71) located within the unique short region of the genome. DNA sequence analysis revealed that EUs4 of the pathogenic EHV-1 strain RacL11 is an open reading frame of 2,376 bp that encodes a protein of 791 amino acids. The attenuated EHV-1 vaccine strain KyA harbors an in-frame deletion of 1,242 bp from bp 222 to 1461 and expresses a truncated gp2 of 383 amino acids. To determine the relative contribution of gp2 to EHV-1 pathogenesis, we compared the course of respiratory infection of CBA mice infected with either wild-type RacL11, attenuated KyA, or a recombinant KyA that expresses the full-length gp2 protein (KyARgp2F). Mice infected with KyA lost a negligible amount of body weight (0.18% total weight loss) on day 1 postinfection and regained weight thereafter, whereas mice infected with KyARgp2F or RacL11 steadily lost weight beginning on day 1 and experienced a 20 and 18% loss in body weight, respectively, by day 3. Immunohistochemical and flow cytometric analyses revealed higher numbers of T and B lymphocytes and an extensive consolidation consisting of large numbers of Mac-1-positive cells in the lungs of animals infected with KyARgp2F compared to animals infected with KyA. RNase protection analyses revealed increased expression of numerous cytokines and chemokines, including interleukin-1β (IL-1β), IL-6, tumor necrosis factor alpha, macrophage inflammatory protein 1α (MIP-1α), MIP-1β, MIP-2, interferon γ-inducible protein, monocyte chemotactic protein 1, and T-cell activation gene 3 at 12 h postinfection with KyARgp2F. Three independent DNA array experiments confirmed these results and showed a 2- to 13-fold increase in the expression of 31 inflammatory genes at 8 and 12 h postinfection with KyARgp2F compared to infection with KyA. Taken together, the results indicate that expression of full-length gp2 is sufficient to restore full respiratory virulence to the attenuated KyA strain and raise caution concerning the inclusion of full-length gp2 in the development of EHV-1 vaccines. The pathogenesis of three equine herpesvirus 1 (EHV-1) recombinants was assessed in a CBA mouse model. Sequences encoding the majority of glycoproteins I (gI) and E (gE) were deleted from the pathogenic EHV-1 strain RacL11 (L11ΔgIΔgE), and sequences comprising the 3859 bp deletion within the strain KyA US segment, which includes genes 73 (gI), 74 (gE), and 75 (putative 10 kDa protein 75), were re-inserted into attenuated KyA (KgI/gE/75). In addition, genes gE and 75 were inserted into KyA to generate the EHV-1 recombinant KgE/75. The insertion of the 3859 bp US segment was sufficient to confer virulence to KyA, as indicated by pronounced signs of clinical disease including substantial weight loss. A large plaque morphology was observed in cells infected with KgI/gE/75 compared with KyA, and a small plaque phenotype was observed in cells infected with L11ΔgIΔgE compared with RacL11. These data indicate that gI and/or gI and gE contribute to the ability of EHV-1 to spread directly from cell-to-cell. The deletion of both gI and gE from the pathogenic RacL11 strain did not reduce clinical signs of disease in infected mice, but did decrease mortality compared with RacL11. Furthermore, the insertion of genes 74 (gE) and 75 into the vaccine strain KyA did not alter the attenuated phenotype of this virus. Finally, KgI/gE/75 and RacL11 elicited the production of the proinflammatory chemokines MIP-1α, MIP-1β, and MIP-2 in the lungs of infected mice, while KyA did not, suggesting that gI and/or gI and gE contribute to the up-regulation of these mediators of inflammation. These findings show that gI, and/or gI and gE restore a virulent phenotype to the EHV-1 KyA strain, and indicate that virulence factors, in addition to gI and gE, contribute to the pathogenesis of the RacL11 strain.
