While subclinical or overt hypothyroidism are common in Down syndrome (DS); Graves’ disease (GD) is rare (ranges 0.6–3%). We aimed to evaluate the clinical features, course, and treatment of GD in children with DS and compare them with those without DS. Among 161 children with GD, 13 (8 female, 5 male) had DS (8%). Data were collected retrospectively from patients’ medical records. The mean age at diagnosis was 10.6 ± 4.5 years, with a female-to-male ratio 1.6:1. The main symptoms were weight loss (n = 6), increased irritability (n = 3), and increased sweating (n = 3). None had orbitopathy. Seven of 11 patients with a thyroid ultrasound at diagnosis had a goitre. On admission, all had thyroid-stimulating hormone (TSH)

ABSTRACT Candida albicans, an opportunistic fungal pathogen, produces the quorum-sensing molecule farnesol, which we have shown alters the transcriptional response and phenotype of human monocyte-derived dendritic cells (DCs), including their cytokine secretion and ability to prime T cells. This is partially dependent on the nuclear receptor peroxisome proliferator-activated receptor gamma (PPAR-γ), which has numerous ligands, including the sphingolipid metabolite sphingosine 1-phosphate. Sphingolipids are a vital component of membranes that affect membrane protein arrangement and phagocytosis of C. albicans by DCs. Thus, we quantified sphingolipid metabolites in monocytes differentiating into DCs by High-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Farnesol increased the activity of serine palmitoyltransferase, leading to increased levels of 3-keto-dihydrosphingosine, dihydrosphingosine, and dihydrosphingosine 1-phosphate and inhibited dihydroceramide desaturase by inducing oxidative stress, leading to increased levels of dihydroceramide and dihydrosphingomyelin species and reduced ceramide levels. Accumulation of dihydroceramides can inhibit mitochondrial function; accordingly, farnesol reduced mitochondrial respiration. Dihydroceramide desaturase inhibition increases lipid droplet formation, which we observed in farnesol-treated cells, coupled with an increase in intracellular triacylglycerol species. Furthermore, inhibition of dihydroceramide desaturase with either farnesol or specific inhibitors impaired the ability of DCs to prime interferon-γ-producing T cells. The effect of farnesol on sphingolipid metabolism, triacylglycerol synthesis, and mitochondrial respiration was not dependent on PPAR-γ. In summary, our data reveal novel effects of farnesol on sphingolipid metabolism, neutral lipid synthesis, and mitochondrial function in DCs that affect their instruction of T cell cytokine secretion, indicating that C. albicans can manipulate host cell metabolism via farnesol secretion.IMPORTANCECandida albicans is a common commensal yeast, but it is also an opportunistic pathogen which is one of the leading causes of potentially lethal hospital-acquired infections. There is growing evidence that its overgrowth in the gut can influence diseases as diverse as alcohol-associated liver disease and COVID-19. Previously, we found that its quorum-sensing molecule, farnesol, alters the phenotype of dendritic cells differentiating from monocytes, impairing their ability to drive protective T cell responses. Here, we demonstrate that farnesol alters the metabolism of sphingolipids, important structural components of the membrane that also act as signaling molecules. In monocytes differentiating to dendritic cells, farnesol inhibited dihydroceramide desaturase, resulting in the accumulation of dihydroceramides and a reduction in ceramide levels. Farnesol impaired mitochondrial respiration, known to occur with an accumulation of dihydroceramides, and induced the accumulation of triacylglycerol and oil bodies. Inhibition of dihydroceramide desaturase resulted in the impaired ability of DCs to induce interferon-γ production by T cells. Thus, farnesol production by C. albicans could manipulate the function of dendritic cells by altering the sphingolipidome.

Ceramides generated by the activity of the neutral sphingomyelinase 2 (nSMase2) play a pivotal role in stress responses in mammalian cells. Dysregulation of sphingolipid metabolism has been implicated in numerous inflammation-related pathologies. However, its influence on inflammatory cytokine-induced signaling is yet incompletely understood. Here, we used proximity labeling to explore the plasma membrane proximal protein network of nSMase2 and TNFα-induced changes thereof. We established Jurkat cells stably expressing nSMase2 C-terminally fused to the engineered ascorbate peroxidase 2 (APEX2). Removal of excess biotin phenol substantially improved streptavidin-based affinity purification of biotinylated proteins. Using our optimized protocol, we determined nSMase2-proximal biotinylated proteins and their changes within the first 5 min of TNFα stimulation by quantitative mass spectrometry. We observed significant dynamic changes in the nSMase2 microenvironment in response to TNFα stimulation consistent with rapid remodeling of protein networks. Our data confirmed known nSMase2 interactors and revealed that the recruitment of most proteins depended on nSMase2 enzymatic activity. We measured significant enrichment of proteins related to vesicle-mediated transport, including proteins of recycling endosomes, trans-Golgi network, and exocytic vesicles in the proximitome of enzymatically active nSMase2 within the first minutes of TNFα stimulation. Hence, the nSMase2 proximal network and its TNFα-induced changes provide a valuable resource for further investigations into the involvement of nSMase2 in the early signaling pathways triggered by TNFα.

Background: This retrospective study aimed to investigate the outcomes and adverse events (AEs) associated with adjuvant radiotherapy with helical tomotherapy (hT) after breast-conserving surgery (BCS) for ductal carcinoma in situ (DCIS). Methods: Twenty-eight patients with DCIS underwent postoperative hT between 2011 and 2020. hT was chosen since it provided optimal target coverage and tolerable organ-at-risk doses to the lungs and heart when tangential 3-dimensional conformal radiotherapy (3D-CRT) was presumed to provide unfavorable dosimetry. The median total (single) dose was 50.4 Gy (1.8 Gy). The median time between BCS and the start of hT was 5 weeks (range, 4-38 weeks). Statistical analysis included local recurrence-free survival, overall survival (OS), and secondary cancer-free survival. AEs were classified according to the Common Toxicity Criteria for Adverse Events, version 5. Results: The patients’ median age was 58 years. The median follow-up period was 61 months (range, 3-123 months). The 1-, 3-, and 5-year OS rates were 100% each. None of the patients developed secondary cancer, local recurrence, or invasive breast cancer during follow-up. The most common acute AEs were dermatitis (n = 27), fatigue (n = 4), hyperpigmentation (n = 3), and thrombocytopenia (n = 4). The late AE primarily included surgical scars (n = 7) and hyperpigmentation (n = 5). None of the patients experienced acute or late AEs > grade 3. The mean conformity and homogeneity indices were 0.9 (range, 0.86-0.96) and 0.056 (range, 0.05-0.06), respectively. Conclusion: hT after BCS for DCIS is a feasible and safe form of adjuvant radiotherapy for patients in whom 3D-CRT is contraindicated due to unfavorable dosimetry. During follow-up, there were no recurrences, invasive breast cancer diagnoses, or secondary cancers, while the adverse effects were mild.

Online learning in physical education is here to stay. While this may be unfamiliar territory to many physical education teachers, it is an area ripe with possibilities and potential. Recent research offers teachers the best practices for creating an exciting online learning environment. This article offers the top ten tips for any teacher to increase the quality of their instructional experience.

The purpose of this study was to explore physical educators' perspectives on the barriers to employing technology in physical education instruction. The participants of this study were 253 elementary, middle, and high school physical education teachers who taught in the state of Georgia. The participants completed an online survey that was derived from an extensive review of literature regarding barriers to integrating technology during instruction. The survey used a Likert scale asking participants to respond with a level of agreement and disagreement regarding specific barriers and included an open-ended response asking for additional barriers. Data were analyzed with the use of descriptive statistics and a one-way analysis of variance. This study found that class size, budget, and lack of resources were the main barriers perceived by physical educators, whereas lack of knowledge and lack of skill were not perceived as barriers to employing technology during instruction. Both age and years of experience had a significant main effect on the barriers of lack of knowledge, skill, and training. Additional barriers reported were lack of connectivity, lack of desire, and lack of time. The results of this study explore and highlight the significant barriers to employing technology in physical education instruction. Further research is warranted to support the need for meaningful implementation of technology to make the learning environment more dynamic, customizable, and accessible for both teachers and students.

The aim of IEEE Time-Sensitive Networking (TSN) standards is to grant deterministic communication in traditional Ethernet networks for Industry 4.0. Insofar as the use cases in the Factory need some mobility, the extension of the TSN capabilities over the fifth-generation (5G) cellular network is the next step. Some challenges in TSN over 5G, such as TSN translators time synchronization functionality, are well defined in the standards, even if they have not yet been addressed in the market. However other challenges, such as the dynamic configuration of the entire network (or part of the it) based on quality requirements of the current TSN traffic pattern, are defined at a very high level and delegated to vendors for implementation. This paper addresses this challenge, using an Automata Learning approach to monitor and reconfigure the end-to-end 5G QoS flow to keep the quality of a TSN session within the required values. Additionally, algorithms are provided to build the automata from network data and predict potential deviations of the requirements to meet the expected quality. Moreover, this work presents a functional TSN over a 5G testbed where the algorithms have been tested, demonstrating that the proposed solution achieves an improvement of around 40% compared to the usual operation of the network.

Purpose: The purpose of this study was to identify the skills, knowledge, and experiences differentiating expert and competent strength and conditioning (S&C) coaches. Method: The participants were 87 elite-level head S&C coaches with an average of over 18 years of coaching experience. These coaches were employed by teams from the MLB, NBA, NFL, NHL, NCAA Division 1 FBS "Power Five" Conferences, United States National Teams, and the Australian Football League. This study used both a qualitative and quantitative research design through the use of the Delphi method, which incorporated three rounds of surveys. The first-round survey asked the participants to identify at least one important skill, knowledge area, and experience that should be possessed by an expert and competent S&C coach respectively. The characteristics generated from the first round were placed on a second-round survey and rated on their level of importance using a five-point Likert scale. If a characteristic received a suggestion for rephrasing, and did not reach a level of consensus, or an entirely new characteristic was suggested, then it was placed on the final-round survey to be rated again. Characteristics that reached a level of consensus were placed on the final list of characteristics. Results: A total of 46 characteristics reached a level of consensus and were placed on the final list of characteristics. Conclusion: There now exists an empirically validated list of characteristics differentiating expert and competent S&C coaches. This list can aid in the selection, evaluation, and development of S&C coaches.

The purpose of this study was to examine adaptations within competition day routines of female US Olympic medalists in differently formatted competitions in order to locate changes based on the organizations of the swimming competitions and differentiate the inflexible from the flexible parts of the routines. A purposeful sample of six elite swimmers participated in two 1-hour interviews with an observation at a national-level competition. A mixed-methods analysis protocol helped locate adaptions within the routine. Four results of note were reported. First, adaptations outside venue depended on format and/or individual schedule; adaptations inside the venue were in response to how they felt and their surroundings. Second, participants adapted their competition-day routines at different levels, i.e., segment and behavior levels, depending on being outside or inside the venue, respectively. Third, the competition-day routine of the participants followed similar competition-day routine segments at the venue that matched in scope and sequence for both the prelims and finals sessions. Fourth, once at the venue, adaptation of behaviors changed in order to accomplish the necessary active or passive preparations without changing the scope, sequence and duration of the segments. These findings deepen the understanding of competition-day routines by locating flexible and inflexible parts of the routines and showing greater variance when athletes adapt their routines. The purpose of this article is to identify the perceptual skills of expert teachers and offer suggestions for teachers to learn to see like an expert. Being able to perceptively read the critical cues in a learning environment allows teachers to recognize present problems, anticipate potential problems, link immediate problems with previously successful solutions, and make exceptional in-class decisions. Four features distinguish what expert teachers see: (1) focusing on events relevant to student performance and learning, (2) making inferences from observations, (3) being alerted by atypical occurrences, and (4) observing with a critical eye. Because the skills of an expert are learned rather than inherited, anyone can increase their expertise by learning the skills of an expert. To do so requires knowledge of the skill and practicing that skill until it is mastered. The article offers specific suggestions for practicing the perceptual skills of an expert teacher.

This article applies Social Network Analysis (SNA) to illuminate the task forces created by US-based activists undertaking public relations on behalf of migrant caravan participants that travel from Honduras through Mexico toward the US. Four functional roles – influencers, messengers, support groups, and founding members – are identified and described in detail. When taking into account the characteristics of the individuals involved, these roles facilitate "bridge-actions": the mobilization of support across distinct political structures such as Native American tribal governments, international human rights venues, and US political parties. The result is a flexible public relations venture that has ability to influence diverse audiences – and extend beyond this particular mobilization effort. Background: Agranulocytosis is a rare antithyroid drug treatment (ATD) side effect seen in children suffering from Graves' disease (GD). Neutropenia is a recognized adverse event associated with ATD but has also been reported as pre-treatment neutropenia in GD. Methods: We performed a retrospective cohort study to analyze the longitudinal clinical and biochemical data of 161 pediatric patients with GD who received either methimazole (MMI) or carbimazole (CBZ) as ATD. The inclusion criteria were elevated free thyroxine (fT4 >25 pmol/L), suppressed thyrotropin (TSH <0.05 mlU/mL), and elevated thyrotropin receptor antibodies (TSHRAbs >2.5 IU/L). Absolute neutrophil count (ANC) was used to define neutropenia (ANC <1800/µL) and agranulocytosis (ANC <500/µL). Results: Nine of the 161 patients had neutropenia at diagnosis (ANC: 1348/µL ± 250) without further deterioration under ATD. In this subgroup, we found higher levels of free triiodothyronine (fT3: 31.45 pmol/L ± 3.99) at diagnosis in comparison with those who developed neutropenia (26.29 pmol/L ± 12.96; p = 0.07) and those without neutropenia before and during therapy (23.12 pmol/L ± 13.7; p = 0.003). Thirty-eight patients (23.6%) became neutropenic (ANC: 1479/µL ± 262) while receiving ATD. Neutropenia occurred after a mean of 551.8 (range: 10–1376) days, mostly without further deterioration. Two of these 38 patients developed agranulocytosis and underwent emergency thyroidectomy. The patients with neutropenia were significantly younger (p = 0.031). Neutropenia occurred significantly more often in patients receiving CBZ (50%; n = 20/40) than in those receiving MMI (16.5%; n = 18/110; p = 0.001). The minimum ANC was significantly lower in the CBZ (1971/µL ± 1008) than in the MMI group (2546 ± 959); p = 0.004. Conclusions: Neutropenia occurred significantly more often under CBZ than MMI. As this is potentially due to higher immunogenicity, we suggest that children with GD should be treated with MMI. Frequent measurements of ANC may be needed to detect severe agranulocytosis, although low pre-treatment ANC may not necessarily be a contraindication to ATD treatment. Young age may be potentially associated with an increased risk of reduced ANC. Further investigation is necessary to fully understand risk factors for neutropenia in children with GD. A theory of comic timing in contemporary slapstick films based on the incongruity theory of humor is put forward. Comic timing is defined as perceived durational incongruity represented by two distinct types of rhythms: (1) in the standard perceived durational incongruity, the setup of the gag is durationally longer than the payoff; (2) in the non-standard perceived durational incongruity, extended duration functions as a self- referential tool for humor because it subverts the spectator's expectations for how long payoffs should be. The extended duration gag represents a metajoke about the nature of comic timing. Two additional factors of perceived durational incongruity are considered including when the setup and payoff are separated by a dramatic beat, which provides time for the spectator to understand the setup and make predictions about the outcome and when the setup is interrupted by the payoff, amounting to a non-threatening surprise that evokes humor. The purpose of this article is to describe effective functions that mentors can use to assist in both the personal and professional development of protégés. Mentors help their protégés in two primary areas: career development and psychosocial support. Career development functions are intended to advance the protégé's adjustments within the profession. Career development roles include: (1) patron, (2) coach, (3) protector, (4) challenger and (5) opportunity creator. Psychosocial support functions increase the protégé's self-identity, competence and affiliation with the school and profession. The psychosocial roles are: (1) friend, (2) role model, (3) counselor and (4) acceptor. Through effective mentoring, the current core of physical education teachers has the opportunity to leave a legacy guaranteeing quality physical education.

A parallel mechanism’s pose is usually obtained indirectly from the active joints’ coordinates by solving the direct kinematics problem. Its accuracy mainly depends on the accuracy of the measured active joints’ coordinates, the tolerances in the active and passive joints, possible backlash, axes misalignment, limb deformations due to stress or temperature, the initial pose estimate that is used for the numerical method, and the accuracy of the kinematic model itself. Backlash and temperature deformations in the active joints especially hinder high-precision applications as they usually cannot be observed. By implementing a camera module on the base platform and an array of fiducial tags on the moveable manipulator platform of a parallel mechanism, a highly accurate, direct, and absolute pose measurement system can be obtained that can overcome those limitations. In this paper, such a measurement system is proposed, designed, and its accuracy is investigated on a state-of-the-art H-811.I2 6-axis miniature hexapod by Physik Instrumente (PI) GmbH & Co. KG.

We demonstrated the anti-inflammatory and anti-oxidative effects of Humulus lupulus (HL) extract on solar simulator-irradiated primary human keratinocytes (PHKs) by analyzing ERK and p38 MAPK phosphorylation and production of IL-6 and IL-8. The anti-inflammatory effect of topically applied HL was further tested in vivo on human skin. To this end, we developed an oil-in-water (O/W) and a water-in-oil (W/O) cream with a lipid content of 40%. The anti-inflammatory effect of 1% HL extract incorporated in these two vehicles was assessed in a randomized, prospective, placebo controlled, double-blind UVB erythema study with 40 healthy volunteers. Hydrocortisone acetate (HCA) in the corresponding vehicle served as positive control. Surprisingly, both HL and HCA were only effective in the O/W system but not in the W/O formulation. Release studies using vertical diffusion cells (Franz cells) revealed that HCA was released in much higher amounts from the O/W cream compared to the W/O formulation. In summary, we have shown that 1% HL extract exerts anti-inflammatory effects comparable to 1% HCA, but only when incorporated in our O/W cream. Our findings confirm the critical role of the vehicle in topical anti-inflammatory systems.

The accumulation of lipid droplets (LDs) and ceramides (Cer) is linked to non-alcoholic fatty liver disease (NAFLD), regularly co-existing with type 2 diabetes and decreased immune function. Chronic inflammation and increased disease severity in viral infections are the hallmarks of the obesity-related immunopathology. The upregulation of neutral sphingomyelinase-2 (NSM2) has shown to be associated with the pathology of obesity in tissues. Nevertheless, the role of sphingolipids and specifically of NSM2 in the regulation of immune cell response to a fatty acid (FA) rich environment is poorly studied. Here, we identified the presence of the LD marker protein perilipin 3 (PLIN3) in the intracellular nano-environment of NSM2 using the ascorbate peroxidase APEX2-catalyzed proximity-dependent biotin labeling method. In line with this, super-resolution structured illumination microscopy (SIM) shows NSM2 and PLIN3 co-localization in LD organelles in the presence of increased extracellular concentrations of oleic acid (OA). Furthermore, the association of enzymatically active NSM2 with isolated LDs correlates with increased Cer levels in these lipid storage organelles. NSM2 enzymatic activity is not required for NSM2 association with LDs, but negatively affects the LD numbers and cellular accumulation of long-chain unsaturated triacylglycerol (TAG) species. Concurrently, NSM2 expression promotes mitochondrial respiration and fatty acid oxidation (FAO) in response to increased OA levels, thereby shifting cells to a high energetic state. Importantly, endogenous NSM2 activity is crucial for primary human CD4+ T cell survival and proliferation in a FA rich environment. To conclude, our study shows a novel NSM2 intracellular localization to LDs and the role of enzymatically active NSM2 in metabolic response to enhanced FA concentrations in T cells. Psoriasis is a chronic inflammatory skin disease characterized by hyperproliferation of keratinocytes and a pro-inflammatory milieu in the skin. While patients with moderate to severe psoriasis are treated using targeted therapies (small molecules and monoclonal antibodies), patients suffering from milder forms are still in need of effective topical products without adverse effects. Antimony compounds (ACs) are regularly used as anti-inflammatory compounds in traditional and anthroposophic medicine and as antiprotozoan drugs. Here, we examined the effect of metallic antimony, natural antimony(III) sulfide and potassium antimonyl(III) tartrate in vitro on psoriasis-like keratinocytes and the human dendritic cell line THP-1 using qPCR, immunocytochemistry, ELISA and flow cytometry. In psoriatic keratinocytes, ACs inhibited the overexpression of the antimicrobial peptide β-defensin 2 and glucose transporter 1, as well as the hyperproliferation marker keratin 17. Furthermore, ACs mediated anti-inflammatory effects by reducing nuclear translocation of the p65 subunit of NF-κB and pSTAT3 and inhibited pro-inflammatory cytokine secretion by keratinocytes. In addition, ACs displayed anti-psoriatic effects by reducing the activation of IFN-α-treated THP-1 cells as well as the expression of the psoriasis-promoting master cytokine IL-23 by these cells. While all ACs showed anti-psoriatic effects, the most prominent results were seen with potassium antimonyl(III) tartrate. In summary, ACs display numerous anti-psoriatic effects in vitro at subtoxic concentrations. We conclude that ACs are interesting compounds for the topical treatment of psoriasis that warrant further investigation in clinical studies.

Plant RBOH (RESPIRATORY BURST OXIDASE HOMOLOGS)-type NADPH oxidases produce superoxide radical anions and have a function in developmental processes and in response to environmental challenges. Barley RBOHF2 has diverse reported functions in interaction with the biotrophic powdery mildew fungus Blumeria graminis f. sp. hordei. Here, we analyzed, in detail, plant leaf level- and age-specific susceptibility of stably RBOHF2-silenced barley plants. This revealed enhanced susceptibility to fungal penetration of young RBOHF2-silenced leaf tissue but strongly reduced susceptibility of older leaves when compared with controls. Loss of susceptibility in old RBOHF2-silenced leaves was associated with spontaneous leaf-tip necrosis and constitutively elevated levels of free and conjugated salicylic acid. Additionally, these leaves more strongly expressed pathogenesis-related genes, both constitutively and during interaction with B. graminis f. sp. hordei. Together, this supports the idea that barley RBOHF2 contributes to basal resistance to powdery mildew infection in young leaf tissue but is required to control leaf cell death, salicylic acid accumulation, and defense gene expression in older leaves, explaining leaf age–specific resistance of RBOHF2-silenced barley plants.

Psoriasis is a chronic inflammatory skin disease characterized by hyperproliferation of keratinocytes and expression of pro-inflammatory cytokines in the epidermis. New biological drugs were developed for the systemic treatment of moderate to severe psoriasis. However, products for the topical treatment of mild psoriasis are still required. Here, we examined the effect of natural compounds on psoriasis-like keratinocytes in vitro and ex vivo. Psoriasis-like keratinocytes were generated by treating human primary keratinocytes with the psoriasis-associated cytokines IL-17A, TNF-α and IL-22. Initially, 10 botanical extracts from Ayurvedic Medicine, Traditional Chinese Medicine, Northern American traditional medicine and Occidental Monastic Medicine were investigated using BrdU assays and IL-6 and IL-8 ELISAs. Curcuma amada, Humulus lupulus and Hypericum perforatum turned out to be the most effective plant extracts. In vitro, the plant extracts inhibited the expression of anti-microbial peptides (β-defensin 2), the hyperproliferation marker keratin 17, the glucose transporter 1 and downregulated the nuclear translocation of NF-κB and pSTAT3. In an ex vivo psoriasis model, Humulus lupulus displayed the most prominent anti-proliferative and anti-inflammatory effect. In conclusion, among the plant extracts investigated, Humulus lupulus showed the most promising anti-psoriatic effect. It is an interesting candidate for topical psoriasis treatment that should be further studied in clinical trials.

This study explored mentoring relationships in sport from the perspective of the protégé. The project was guided by contemporary mentoring theories as framed by Kram's Mentor Role Theory (Kram, 1985). A convenience sample of 230 volleyball coaches was recruited for this study. Data were collected using the Coaches Mentor Role Instrument (CMRI) (Schempp, McCullick, Berger, White, & Elliott, 2014). Quantitative methods indicated significant mentor role differences based on continuation of relationship, participation in a formal mentor programme, and gender. The participants perceived their mentors as most effective in the roles of acceptor, friend, role model and challenger.

Emotional intelligence (EI) is recognized as an indicator of success, yet little research has examined the role of EI in coaching success. This cross-sectional study assessed the power of EI in predicting coaches' career winning percentage in 277 head collegiate coaches including 51 basketball and 226 volleyball coaches. Coaches were on average 41.47 years old (SD = 9.68), had 12.17 years of head coach experience (SD = 9.40), and a career winning percentage of 50.67% (SD = 16.5). Coaches completed an online questionnaire including the Assessing Emotions Scale, which was used to assess their EI. Career winning percentage was calculated by extracting data from institutional websites and NCAA databases. Regression analyses revealed EI was not a significant predictor of coaching success when all coaches were analyzed together, F(4, 272) = 0.7504, p =.5585. However, when examined separately, EI was a significant predictor of basketball coaches' success, F(4, 46) = 2.678, p =.0433, but not volleyball coaches' success, F(4, 221) = 0.363, p =.0835. Thus, the current body of evidence indicates EI has a nonuniform impact on coaching success. While acknowledging sample size limitations, these novel findings nevertheless indicate that EI may disproportionally influence the coaching process and downstream success by sport. Gentiana lutea is a bitter herb that is traditionally used to improve gastric disorders. Recently, we have shown that Gentiana lutea extract (GE) also modulates the lipid metabolism of human keratinocytes in vitro and in vivo. In the present study, we investigated the role of GE on ceramide synthesis in human primary keratinocytes (HPKs) and psoriasis-like keratinocytes. We could demonstrate that GE increased the concentrations of glucosylceramides and the ceramide AS/AdS subclass without affecting the overall ceramide content in HPKs. The expression of ceramide synthase 3 (CERS3) and elongases (ELOVL1 and 4) was reduced in psoriasis lesions compared to healthy skin. Psoriasis-like HPKs, generated by stimulating HPKs with cytokines that are involved in the pathogenesis of psoriasis (IL-17, TNF-α, IL-22 and IFN-γ) showed increased levels of IL-6, IL-8 and increased expression of DEFB4A, as well as decreased expression of ELOVL4. The treatment with GE partly rescued the reduced expression of ELOVL4 in psoriasis-like HPKs and augmented CERS3 expression. This study has shown that GE modulates ceramide synthesis in keratinocytes. Therefore, GE might be a novel topical treatment for skin diseases with an altered lipid composition such as psoriasis.
