Environmental features impacting the spread of invasive species after introduction can be assessed using population genetic structure as a quantitative estimation of effective dispersal at the landscape scale. However, in the case of an ongoing biological invasion, deciphering whether genetic structure represents landscape connectivity or founder effects is particularly challenging.We examined the modes of dispersal (natural and human‐aided) and the factors (landscape or founders history) shaping genetic structure in range edge invasive populations of the Asian tiger mosquito, Aedes albopictus, in the region of Grenoble (Southeast France).Based on detailed occupancy–detection data and environmental variables (climatic, topographic and land‐cover), we modelled A. albopictus potential suitable area and its expansion history since first introduction. The relative role of dispersal modes was estimated using biological dispersal capabilities and landscape genetics approaches using genome‐wide SNP dataset.We demonstrate that both natural and human‐aided dispersal have promoted the expansion of populations. Populations in diffuse urban areas, representing highly suitable habitat for A. albopictus, tend to disperse less, while roads facilitate long‐distance dispersal. Yet, demographic bottlenecks during introduction played a major role in shaping the genetic variability of these range edge populations.The present study is one of the few investigating the role of founder effects and ongoing expansion processes in shaping spatial patterns of genetic variation in an invasive species at the landscape scale. The combination of several dispersal modes and large proportions of continuous suitable habitats for A. albopictus promoted range filling of almost its entire potential distribution in the region of Grenoble only few years after introduction. Densely overgrown pre-alpine permanent aquatic habitats are overrun by the mosquito Coquillettidia (Coquillettidia) richiardii (Ficalbi) (Diptera: Culicidae). The invasive potential of this insect depends on the ability of its larvae to survive on the roots of emergent aquatic macrophytes. In order to characterize this particular ecological niche, which is out of reach for a direct investigation, environmental factors likely to influence the interaction between larvae and host plant roots were investigated using a simplified laboratory microcosm. Environmental light and oxygen concentrations appeared to be the main factors influencing larval attachment, a dark anoxic environment being significantly more favourable. Carbon dioxide produced by the root system of the host plant appeared to be an attractive cue for larval attachment. Knowledge of the hierarchy of these environmental factors may enable us to better understand the ecological traits of larval C. richiardii in deep water. In order to examine the factors influencing xenobiotic toxicity against larval mosquitoes, the larvicidal performances of two conventional insecticides (temephos and Bacillus thuringiensis var. israelensis: Bti) and a new potential phyto-insecticide (decomposed leaf litter) were compared under different conditions against three detritivorous larval mosquito types. Bioassays performed under standard conditions indicated differential tolerance levels according to the xenobiotic and the larval type. Bioassays performed under different conditions of xenobiotic dose and geometry of the water column indicated differential effects of those parameters on mortality rates. This allowed us to distinguish the performances of temephos versus those of Bti and leaf litter. These toxicological performances were examined as indicators for analysis of xenobiotic bioavailability for mosquito larvae in environmental water, and also for their comparative interest in field mosquito control. The ability of mosquito larvae to tolerate toxic compounds (temephos, Bacillus thuringiensis var. israelensis, toxic vegetable leaf litter) was examined on a laboratory larval strain of Aedes aegypti L. Bioassays and detoxifying enzyme activity measurements were performed to compare tolerance/resistance capacities. The possibility of a functional plasticity of detoxifying equipment was investigated through experimental determination of the inductive effect of each xenobiotic within a given generation. In the same way, the selective effect of a toxic leaf litter was also investigated along successive generations. Results revealed that differential cytochrome P450 monooxygenase, esterase, and glutathione S-transferase activity levels correlated with the bioassay results. Both induction and selection increased larval tolerance to the xenobiotics used and increased the levels of larval detoxifying enzyme activities. [ABSTRACT FROM AUTHOR]

The larvicidal properties of the dietary leaf litter originating from the vegetation surrounding the subalpine mosquito breeding sites were investigated by using 10-month decomposed alder leaf litter against different field collections of culicine taxa of various ecological origin (Aedes cantans, Aedes caspius, Aedes cataphylla, Aedes detritus, Aedes punctor, Aedes pullatus, Aedes rusticus, Anopheles claviger,Culex hortensis, Culex pipiens, Culiseta morsitans). Larvae originating from sites with polyphenol-poor vegetation appeared more sensitive to ingested leaf litter than those originating from sites with polyphenol-rich vegetation. Within a given taxon (e.g., A. rusticus, A. cataphylla, C. hortensis), the overall levels of cytochrome P450 monooxygenase and esterase activities appeared higher in larvae able to feed on leaf litter than in pupae and adults unable to feed on leaf litter. This suggests the involvement of these enzymes in the detoxification mechanisms responsible for larval tolerance to polyphenols of the dietary leaf litter. Such a tolerance of the larval stage thus appears as fundamental in the ecotoxicological adaptation of mosquito taxa to the polyphenolic profiles of the riparian vegetation. [ABSTRACT FROM AUTHOR]
