Abstract This study addresses the understanding gap concerning the factors that influence the continuous learning intention of adult learners on online education platforms. The uniqueness and significance of this study stem from its dual focus on both platform features, such as service quality, and course features, including perceived interactivity and added value, aspects often overlooked in previous research. Rooted in Expectation Confirmation Theory, the study constructs a comprehensive model to shed light on the complex interplay of these factors. Empirical evidence collected from a survey of 1592 adult learners robustly validates the effectiveness of this model. The findings of the study reveal that platform service quality, perceived interactivity, and perceived added value significantly amplify adult learners' expectation confirmation and perceived usefulness. These elements subsequently enhance learner satisfaction, fostering their ongoing intention to use online education platforms. These insights offer practical guidance for online education providers, emphasizing the necessity to enhance platform service quality and course features to meet adult learners' expectations and perceived usefulness. The study provides valuable perspectives for devising strategies to boost user satisfaction and stimulate continuous usage intention among adult learners in the intensely competitive online education market. This study enriches the literature by uncovering the relationships among platform features, course features, expectation confirmation, perceived usefulness, and continuous usage intention. By proposing a comprehensive model, this study provides a novel theoretical basis for understanding how platform and course features impact adult learners' ongoing intention to use online education platforms, thereby aiding the evolution and refinement of relevant theories.

The fluctuation of the tide is closely related to the production and life of people in coastal areas, and the change in the tide level will have a significant impact on the safety of infrastructure, ship travel, ecological environment, and other issues. Therefore, it is of great significance to analyze, study, and forecast the change in tide level. Aiming at the complex characteristics of nonlinearity, time-varying dynamics, and uncertainty generated by celestial bodies’ movements and influenced by geographical as well as hydrometeorological factors, this paper proposes a combined model based on variational mode decomposition (VMD) and long short-term memory neural networks (LSTM). A tidal level prediction procedure is proposed by combining the harmonic analysis method with a neural network and takes the point tide data of Luchao Port from 2021 to 2022 as the applied data. First, the VMD algorithm decomposes the tidal level data into model components. Then, the LSTM model is used to predict each component. Finally, the predicted value of each component is superposed to obtain the final prediction result. Standard evaluation indexes were used to analyze the performance of the proposed model. The model’s RMSE, MAE, MAPE, and R2 were 0.0385, 0.0267, 5.8327, and 99.91%, respectively, superior to other compared models (BP, SVM, and LSTM). This study can provide a reference for tidal level prediction. These results show that the VMD-LSTM model is an effective and reliable tidal level prediction tool with considerable potential in offshore engineering and maritime management.

Graphene has promising applications in agriculture and forestry. In the current study, six different concentrations of graphene (0mg/L, 0.01mg/L, 0.10mg/L, 1.00mg/L, 10.00mg/L, and 100.00mg/L) were used to investigate its effect on the growth and development of V. angularis plants in soil culture. The results showed that the group treated with 1.00mg/L graphene (G-1) had significantly increased plant height (19.86%), stem diameter (24.33%), and leaf area (13.69%), compared to the control group (CK). Moreover, all concentrations of graphene had positive effects on the total root length, total root surface area, and the number of root tips of V. angularis. Compared to the CK group, the G-1 group had significantly increased leaf water potential (37.89%), leaf conductivity (2.25%), and SOD, POD, and CAT activities (47.67%, 35.22%, and 199.3%, respectively). The G-1 group also showed improved leaf net photosynthetic rate, chlorophyll content, and soluble sugar content (51.28%, 24.25%, and 38.35%, respectively), compared to the CK group. Additionally, 1.00mg/L graphene led to a 23.88% increase in the podding rate and a 17.04% increase in the yield of V. angularis plants. The rhizosphere soil of V. angularis treated with 1.00mg/L graphene had a 25.14% increase in hydrolyzable nitrogen content and a 66.67% increase in available phosphorus content. RNA-seq data indicated that 1.00mg/L graphene induced the expression of photosynthesis and nitrogen transmembrane transport genes, including ATP synthase subunit b, photosystem I reaction center subunit XI, photosystem I reaction center subunit IV A, ferredoxin, and psbP-like protein 1, as well as genes for photosynthesis antenna proteins, glutamine synthetase, glutamate dehydrogenase 1, cyanate hydratase, protein fluG-like, and NRT1/PTR family, suggesting that graphene promoted the growth and development of V. angularis by enhancing the photosynthesis and nitrogen metabolism processes in V. angularis plants. Our results indicated that a suitable concentration of graphene could significantly promote the growth of V. angularis plants in soil.

The Protein Arginine Methyltransferases (PRMTs) family is involved in various biological processes, including gene transcription, pre-mRNA splicing, mRNA translation, and protein stability. Recently, mounting evidence has shown that PRMTs also play critical roles in regulating the host antiviral immune response, either in an enzymatic activity dependent or independent manner. This review aims to provide an overview of the recent findings regarding the function and regulatory mechanisms of PRMTs in the antiviral response. These findings have the potential to aid in the discovery and design of novel therapeutic strategies for viral infections.

Traditional univariate drought indices may not be sufficient to reflect comprehensive information on drought. Therefore, this paper proposes a new composite drought index that can comprehensively characterize meteorological and hydrological drought. In this study, the new drought index was established by combining the standardized precipitation index (SPI) and the standardized baseflow index (SBI) for the Jiaojiang River Basin (JRB) using the copula function. The prediction model was established by training random forests on past data, and the driving force behind the combined drought index was explored through the LIME algorithm. The results show that the established composite drought index combines the advantages of SPI and SBI in drought forecasting. The monthly and annual droughts in the JRB showed an increasing trend from 1991 to 2020, but the temporal characteristics of the changes in each subregion were different. The accuracies of the trained random forest model for heavy drought in Baizhiao (BZA) and Shaduan (SD) stations were 83% and 88%, respectively. Furthermore, the Local Interpretable Model-Agnostic Explanations (LIME) interpretation identified the essential precipitation, baseflow, and evapotranspiration features that affect drought. This study provides reliable and valid multivariate indicators for drought monitoring and can be applied to drought prediction in other regions.

Baseflow is the part of streamflow that is mainly replenished by groundwater. The protection of the biological environment and the growth of its water resources greatly depend on the spatial and temporal evolution of baseflow. Therefore, the Baizhiao (BZA) and Shaduan (SD) catchments of the Jiaojiang River Basin (JRB) in the Zhejiang province of China were selected as study areas. The ABCD model and Eckhardt method were used to calculate baseflow and baseflow index (BFI). The temporal and spatial evolution patterns of baseflow were analyzed through statistical analysis and the Mann–Kendall test. The results showed that the ABCD model performs well in simulating overall hydrological processes on the monthly streamflow at BAZ and SD stations with NSE (Nash–Sutcliffe Efficiency) values of 0.82 and 0.83 and Pbias (Percentage Bias) values of 9.2% and 8.61%, respectively. The spatial–temporal distribution of the BFI indicates the higher baseflow contribution in upstream areas compared to downstream areas at both stations. The baseflow and BFI had significant upward trends at the BZA and SD stations in the dry season, while their trends were not uniform during the wet period. These findings are essential guidance for water resource management in the JRB regions.

Colon adenocarcinoma (COAD) is among the most prevalent cancers worldwide, ranking as the third most prevalent malignancy in incidence and mortality. The somatostatin receptor (SSTR) family comprises G-protein-coupled receptors (GPCRs), which couple to inhibitory G proteins (Gi and Go) upon binding to somatostatin (SST) analogs. GPCRs are involved in hormone release, neurotransmission, cell growth inhibition, and cancer suppression. However, their roles in COAD remain unclear. This study used bioinformatics to investigate the expression, prognosis, gene alterations, functional enrichment, and immunoregulatory effects of the SSTR family members in COAD. SSTR1-4 are differentially downregulated in COAD, and low SSTR2 expression indicates poor survival. Biological processes and gene expression enrichment of the SSTR family in COAD were further analyzed using the Kyoto Encyclopedia of Genes and Genomes and Gene Ontology. A strong correlation was observed between SSTR expression and immune cell infiltration. We also quantified SSTR2 expression in 25 COAD samples and adjacent normal tissues using quantitative real-time polymerase chain reaction. We analyzed its correlation with the dendritic cell–integrin subunit alpha X marker gene. The biomarker exploration of the solid tumors portal was used to confirm the correlation between SSTR2 with immunomodulators and immunotherapy responses. Our results identify SSTR2 as a promising target for COAD immunotherapy. Our findings provide new insights into the biological functions of the SSTR family and their implications for the prognosis of COAD.

Erianin is an active constituent of Dendrobium candidum. In this work, 18F-fluoroethoxylerianin ([18F]FEE), a 18F-Labeled erianin analogue, was designed and synthesized to evaluate the properties of erianin and related analogues by in vivo PET imaging. The initial product was separated and purified by liquid-phase separation module Explora LC and simple homemade solid-phase extraction, and high purity [18F]FEE was finally obtained. The radiochemical purity of [18F]FEE was determined by Radio-TLC and Radio-HPLC. [18F]FEE showed good stability in normal saline and serum, and could be quickly eliminated from mice. Cell experiments, biological distribution, and small-animal PET/CT further showed that [18F]FEE had a high uptake rate in HepG2 tumor cells, and showed good imaging ability in a HepG2 tumor model. The results of this study indicate that the synthesized 18F-labeled erianin analogue is an effective new probe for positron emission tomography (PET) imaging of HepG2 hepatocellular carcinoma, which provides an intuitive and reliable theoretical basis for the development of erianin as an anticancer drug.

Indium can efficiently mediate the Reformatsky reaction of iododifluoroacetylketones with aldehydes to afford the corresponding α,α-difluoro-β-hydroxyketones in high yield in pure water This reaction has excellent substrate suitability and functional group selectivity and provides an efficient approach for the synthesis of bioactive molecules containing the α,α-difluoro-β-hydroxyketone pharmacophore.

Metastasis is a major challenge in the treatment of cancer. Exosomes are a class of small extracellular vesicles (EVs) that play critical roles in several human diseases, especially cancer, by transferring information (e.g., DNA, RNA, and protein) via cell-to-cell communication. Numerous recent studies have shown that exosomal long non-coding RNAs (lncRNAs) play crucial regulatory roles in cancer metastasis in the tumor microenvironment by altering the expression of several key signaling pathways and molecules. Due to their specificity and sensitivity, exosomal lncRNAs have potential as novel tumor markers and therapeutic targets in the treatment of cancer metastasis. In this review, we aim to summarize the roles of exosomal lncRNAs in cancer metastasis, the mechanisms underlying their roles, and their potential clinical applications.

Chinese water chestnut (CWC) is one of the most popular hydrophytic vegetables in China due to its unique aroma and taste. The volatile compounds (VOCs) of CWC with different processing methods were evaluated. The results of headspace-gas chromatography–ion mobility spectrometry (HS-GC-IMS) showed that alcohols, aldehydes, aromatics, and esters were the main VOCs, and the headspace solid-phase microextraction-gas chromatography–mass spectrometry (HS-SPME-GC–MS) results indicated that esters, amines and aromatics were the main flavor components. Compared with other processes, the VOC alcohol levels in fresh cutting (FF) decreased, while esters and sulfide increased. The flavor of CWC in the cooking with peeling (PC) and steaming with peeling (PS) were similar, the FF and steaming with skin (WPC) are very different from the other samples. This study is the first to provide results regarding differences in the aroma compounds of CWC processed with different methods, and 146 aroma compounds were characterized.

The SMYD family is a unique class of lysine methyltransferases (KMTases) whose catalytic SET domain is split by a MYND domain. Among these, Smyd1 was identified as a heart- and skeletal muscle-specific KMTase and is essential for cardiogenesis and skeletal muscle development. SMYD1 has been characterized as a histone methyltransferase (HMTase). Here we demonstrated that SMYD1 methylates Skeletal muscle-specific splice variant of the Nascent polypeptide-Associated Complex (skNAC) transcription factor. SMYD1-mediated methylation of skNAC targets K1975 within the carboxy-terminus region of skNAC. Catalysis requires physical interaction of SMYD1 and skNAC via the conserved MYND domain of SMYD1 and the PXLXP motif of skNAC. Our data indicated that skNAC methylation is required for the direct transcriptional activation of myoglobin (Mb), a heart- and skeletal muscle-specific hemoprotein that facilitates oxygen transport. Our study revealed skNAC as a methylation target of SMYD1, illuminates the molecular mechanism by which SMYD1 cooperates with skNAC to regulate transcriptional activation of genes crucial for muscle functions and implicates the MYND domain of the SMYD-family KMTases as an adaptor to target substrates for methylation.

The transcriptional co-activator with PDZ-binding motif (TAZ) is a downstream effector of the Hippo pathway. It has been identified as an oncogene in certain tumor types; however, the function and role of TAZ in colorectal cancer (CRC) has not been illustrated. Here, we aimed to analyze the expression and role of TAZ in CRC. In this study, we investigated the expression level of TAZ in 127 CRC and matched adjacent normal tissues by immunohistochemistry (IHC) and analyzed its correlation with clinicopathological characteristics in CRC. Moreover, we further analyzed the role of TAZ in the CRC-associated immunology using integrative bioinformatic analyses. The cBioPortal and WebGestalt database were used to analyze the co-expressed genes and related pathways of TAZ in CRC by gene ontology (GO) and KEGG enrichment analyses. Meanwhile, the correlations between TAZ and the infiltrating immune cells and gene markers were analyzed by TIMER database. Our study revealed that TAZ expression is higher in CRC tissues than in matched adjacent non-tumor tissues. In addition, CRC patients with higher TAZ expression demonstrated poor overall survival (OS) and recurrent-free survival rates as compared to CRC patients with lower expression of TAZ. Furthermore, the TAZ expression was identified to closely associate with the immune infiltration of CD4 + T, CD8 + T, and B cells. Taken together, our findings suggest that TAZ may serve as a promising prognostic biomarker and therapeutic target in CRC.

Abstract Background To explore the 131I-SPECT/CT characteristics of remnant thyroid tissue (RTT) in differentiated thyroid cancer (DTC), further assess the risk factors and clinical significance. Methods 52 DTC patients after total thyroidectomy had undergone neck 131I-SPECT/CT before 131I ablation. The diagnosis of RTT was based on SPECT/CT and follow-up at least 3 months. The anatomic locations and features of SPECT/CT of RTT were assessed by reviewers. The risk factors of RTT with CT positive were analyzed by the chi-square test. Results A total of 80 lesions of RTT were diagnosed in this study, most of them were mainly located in the regions adjacent to trachea cartilage (37/80) or lamina of thyroid cartilage (17/80). On SPECT/CT of RTT, low, moderate and high uptake were respectively noted in 10, 24 and 46 lesions, definite positive, suspected positive and negative CT findings were respectively noted in 10, 21 and 49. The RTT lesions with definite positive CT findings were mainly located adjacent to lamina of thyroid cartilage (5/10). Primary thyroid tumor (P = 0.029) and T stage (P = 0.000) were the effective risk factors of CT positive RTT. Conclusions RTT has certain characteristic distribution and appearances on SPECT/CT. Most of RTT with definite CT abnormalities located adjacent to lamina of thyroid cartilage, which suggest surgeons should strengthen the careful removal in this region, especially primary thyroid tumor involving bilateral and T4 stage. This study can provide a certain value for the improvement of thyroidectomy quality in DTC patients.

Abstract The root of Angelica sinensis (Oliv.) Diels, a well-known Chinese herbal medicine, has been used historically as hematopoietic and anti-inflammatory agents for thousands of years. Recent phytochemistry and pharmacological studies have proved that polysaccharides are one of the major active ingredients in A. sinensis. It has been demonstrated that ASPs (A. sinensis polysaccharides) had various important biological activities, such as hematopoietic, hepatoprotective, hypoglycemic, anti-inflammatory, antitumor, and antioxidant activities. The purpose of this present review is to appraise previous and current literatures on the extraction, purification, structural characterization and biological activities of ASPs. In addition, the structure–activity relationship will be further explored and discussed. We believe that this review will provide a useful bibliography for the investigation, production, and application of ASPs in functional foods and therapeutic agents. Moreover, this review also highlights the challenges of investigation and future considerations for holistic utilization.

Soil degradation resulting from human trampling in urban forest parks can negatively impact the taxonomic diversity and function of soil microbial communities. In this study, we established long-term, fixed large plots in Zijin Mountain Urban Forest Park in Nanjing, China, to assess the level of trampling pressure. Soil samples were collected from depths of 0–10 cm, 10–20 cm, and 20–30 cm for light trampling (LD), moderate trampling (MD), severe trampling (SD), extreme trampling (ED), and a no-trampling control (CK). The effects of different trampling pressures on soil were studied, including soil nutrient indices, microbial biomass, and the taxonomic diversity of fungi and bacteria. ANOVA and structural equation modeling (SEM) were employed to investigate the impacts of human trampling on the microbial community structure and function. The results indicated that soil organic carbon, ammonium, and acid phosphatase activity were the primary driving factors of microbial community change. Soil microbial diversity initially increased and then decreased with increasing trampling intensity. The changes in soil microbial function and classification were found to be associated with the intensity of trampling. Moderate trampling could enhance the diversity of the soil microbial community. The succession pattern of the fungi and bacteria communities was distinct, and the composition of the bacteria community remained relatively stable. Trampling impacts vegetation and soil structure, which then affects the structure and function of the microbial community. This study provides an essential foundation for the restoration of compacted soil in urban forest parks through targeted monitoring and management efforts.

Abstract The technological revolution of long-awaited energy-saving and vision-friendly displays represented by bistable display technology is coming. Here we discuss methods, challenges, and opportunities for implementing bistable displays in terms of molecular design, device structure, further expansion, and required criteria, hopefully benefiting the light-related community.

Summary: Meiotic crossover (CO) recombination between homologous chromosomes regulates chromosome segregation and promotes genetic diversity. Human females have different CO patterns than males, and some of these features contribute to the high frequency of chromosome segregation errors. In this study, we show that CO covariation is transmitted to progenies without detectable selection in both human males and females. Further investigations show that chromosome pairs with longer axes tend to have stronger axis length covariation and a stronger correlation between axis length and CO number, and the consequence of these two effects would be the stronger CO covariation as observed in females. These findings reveal a previously unsuspected feature for chromosome organization: long chromosome axes are more coordinately regulated than short ones. Additionally, the stronger CO covariation may work with human female-specific CO maturation inefficiency to confer female germlines the ability to adapt to changing environments on evolution.

Pancreatic adenocarcinoma (PAAD) is a malignant tumor with high morbidity and mortality worldwide. Members from the structural maintenance of chromosomes (SMC) gene family function as oncogenes in various tumor types, but their roles in PAAD have not been elucidated. In this study, we aimed to explore the role of the SMC family in tumor progression and cancer immune infiltration in PAAD using integrative bioinformatic analyses. The results showed that the SMC 1A, 2, 3, 4, and 6 were overexpressed in PAAD tissues; of these, SMC 1A, 4, 5, and 6 could be potential prognostic biomarkers for PAAD. The expression of SMC genes was found to be strongly associated with immune cell infiltration. According to the infiltrative status of various immune cells, the mRNA expression of SMC genes in PAAD was associated with the overall and recurrence-free survival of patients. In conclusion, the SMC gene family is associated with PAAD and may be involved in tumorigenesis and cancer-immune interactions; thus, members from this gene family may serve as promising prognostic and therapeutic biomarkers of PAAD.

In order to improve poor anti-offset capability of rotary transformer in electric excitation motor, a coaxial nested rotary wireless power transfer (CNR-WPT) system has been proposed in this paper. Firstly, considering the spatial geometric relationship of the coils and the power transmission efficiency of the CNR-WPT, the preliminary coil structure has been proposed. Secondly, through theoretical derivation, the specific relationship between the mutual inductance of the coils and the offset have been studied to verify the feasibility of the preliminary design. Thirdly, aiming at the problem that the CNR-WPT is susceptible to steel interference, the magnetic field has been optimized by adding ferrite and introducing a protective casing in this paper. Finally, an experimental platform for CNR-WPT system has been built. The experimental results verify that the power transmission efficiency of the CNR-WPT system can reach 90% when the radial offset and axial offset are below 5 mm, and the angular offset is below 5°. The energy losses can be reduced by adding ferrite and protecting the casing. The CNR-WPT system thereof can also be applied to other rotary power transmission occasions.

Abstract Light‐responsive color‐switching materials (LCMs) are long‐lasting hot fields. However, non‐ideal comprehensive performance (such as color contrast and retention time cannot be combined, unsatisfactory repeatability, and non‐automated coloring mode) significantly hinder their development toward high‐end products. Herein, the development of LCMs that exhibit long retention time, good color contrast, repeatability, and the property of automatic coloring is reported. The realization of this goal stems from the adoption of a bio‐inspired multi‐component collaborative step‐by‐step coloring strategy. Under this strategy, a conventional one‐step photochromic process is divided into a “light+heat” controlled multi‐step process for the fabrication of the desired LCMs. The obtained LCMs can effectively resist the long‐troubled ambient‐light interference and avoid its inherent yellow background, thereby achieving the longest retention time and good repeatability. Multiple colors are generated and ultra‐fast imaging compatible with the laser‐printing technology is also realized. The application potential of the materials in short‐term reusable identity cards, absorptive readers, billboards, and shelf labels is demonstrated. The results reported herein can potentially help in developing and designing various high‐performance, switchable materials that can be used for the production of high‐end products.

Chinese water chestnut (CWC) is popular among consumers due to its unique flavor and crisp and sweet taste. Thus far, the key substances affecting the taste compound of CWC are still unclear. In this study, we used UPLC-MS/MS and an electronic tongue system to study the effects of four typical steaming and cooking methods, cooking without peel for 10 min (PC), steaming without peel for 15 min (PS), cooking with peel for 30 min (WPC), steaming with peel for 30 min (WPS), on the taste compound generation and variation of CWC, and revealed the secret of its crisp and sweet taste. The results show that the electronic tongue can effectively identify the taste profile of CWC, and the effective tastes of CWC were umami, bitterness, saltiness, and sweetness. We screened 371 differential compounds from 640 metabolic species. Among them, nucleotides and their derivatives, carbohydrates, organic acids and their derivatives, and amino acids and their derivatives are closely related to the key taste of CWC, and these compounds affected the taste of CWC through six related metabolic pathways: oxidative phosphorylation and purine metabolism; alanine, aspartate, and glutamate; bile secretion; amino sugar and nucleotide sugar metabolism; the phenylpropane pathway; and toluene degradation. This study reveals the potential metabolic causes of taste compound generation and variation in the taste of CWC.

Understanding the impact of climate change and human activities on the hydrological cycle of any watershed can provide a scientific basis for regional water resource planning, flood management, and disaster mitigation. An improved three-parameter hydrological model (CM) based on monthly water balance using an exponential equation to depict the distribution of groundwater storage capacity was developed and evaluated. The model uses Asian Precipitation Highly Resolved Observational Data Integration Towards Evaluation (APHRODITE) rainfall data as input, with the Zhejiang Province as the case application, and the effects of climate change and human activities on streamflow changes were assessed by separating environmental variables in this study. The results indicate that APHRODITE data has excellent monthly accuracy, with a mean correlation coefficient (CC) of more than 0.96 and an average absolute percentage bias (Pbais) of less than 5%. The three models are relatively close in their ability to simulate high flows, but the CM simulated low flow is better than the other two models. Positive and negative Pbais phenomena occur in the CM model in each catchment, and absolute levels are regulated by 5%. Furthermore, the CM model’s average Nash efficiency coefficient (NSE) is greater than 0.9, indicating that it can correctly fulfill the water balance. The results are more consistent throughout multiple catchments in each watershed using Budyko-based and hydrological model technique to evaluate the influence of climate change and human activities on streamflow. Climate change dominated streamflow variations in 18 of the 21 catchments in Zhejiang Province, whereas human activities dominated the rest. The findings of the study will be used to influence the management, development, and usage of water resources in the watershed.

Worldwide, hepatocellular carcinoma (HCC) is one of the most malignant cancers with poor prognosis. The structural maintenance of chromosomes (SMC) gene family has been shown to play important roles in human cancers. Nevertheless, the role of SMC members in HCC is not well-understood. In this study, we comprehensively explored the role of the SMC family in HCC using a series of bioinformatic analysis tools. Studies have demonstrated that the mRNA expression levels of SMC1A, SMC1B, SMC2, SMC4, and SMC6 are significantly overexpressed in HCC, and the protein levels of SMC1A, SMC2, SMC3, SMC4, SMC5, and SMC6 are similarly elevated. Moreover, HCC patients with high SMC2 and SMC4 expression levels exhibit poor survival. Using KEGG and GO analyses, we analyzed the enrichment of gene expression in the biological functions and pathways of the SMC family in HCC. Immune infiltration analysis revealed that the expression of the SMC family is closely associated with B cells, CD4+ T cells, CD8+ T cells, macrophages, neutrophils, and DCs. In conclusion, our findings will enhance a more thorough understanding of the SMC family in HCC progression and provide new directions for the diagnosis and treatment of HCC in the future.

Lactate dehydrogenase A (LDHA) is an important glycolytic enzyme that promotes glycolysis and plays a crucial role in cancer cell invasion and immune infiltration. However, the relevance of LDHA in colon adenocarcinoma (COAD) remains unclear. In this study, we analyzed the correlation between the expression of LDHA and clinicopathological characteristics in COAD using immunohistochemistry analysis, and then used integrative bioinformatics analyses to further study the function and role of LDHA in COAD. We found that LDHA was highly expressed in COAD tissues compared with adjacent normal tissues, and that COAD patients with high LDHA expression levels showed poor survival. In addition, LDHA expression was closely associated with the immune infiltrating levels of CD8+ T cells, neutrophils, and dendritic cells. Our findings highlight the potential role of LDHA in the tumorigenesis and prognosis of COAD. Furthermore, our results indicate that COAD is a novel immune checkpoint in the diagnosis and treatment of COAD.

Colorectal cancer (CRC) is one of the most common gastrointestinal cancers, with extremely high rates of morbidity and mortality. The main cause of death in CRC is distant metastasis; it affects patient prognosis and survival and is one of the key challenges in the treatment of CRC. Long non-coding RNAs (lncRNAs) are a group of non-coding RNA molecules with more than 200 nucleotides. Abnormal lncRNA expression is closely related to the occurrence and progression of several diseases, including cancer. Recent studies have shown that numerous lncRNAs play pivotal roles in the CRC metastasis, and reversing the expression of these lncRNAs through artificial means can reduce the malignant phenotype of metastatic CRC to some extent. This review summarizes the major mechanisms of lncRNAs in CRC metastasis and proposes lncRNAs as potential therapeutic targets for CRC and molecular markers for early diagnosis.

In this study, a sensitive monoclonal antibody (mAb) 3D5 against sparfloxacin (SPFX) was generated. Based on the mAb 3D5, an indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) method was developed for the detection of SPFX in honey. The half maximal inhibitory concentration and limit of detection for the ic-ELISA method were 0.12 and 0.02 ng/mL, respectively. The experiments showed a negligible cross-reaction with other drugs. The average recovery rates for spiked SPFX honey extracts ranged from 90% to 101%, indicating an accepted accuracy. Furthermore, a lateral-flow immunochromatographic assay strip method with a cut-off value of 2 ng/mL was developed. Therefore, both of the developed methods are suitable for future use as rapid screening methods to detect and control the content of SPFX residues in honey samples.

Variations in coastline geometry caused by coastal engineering affect tides, storm surges, and storm tides. Three cluster land reclamation projects have been planned for construction in the Jiaojiang Estuary during the period from 2011 to 2023. They will cause significant changes in coastline geometry. In this study, a surge-tide coupled model was established based on a three-dimensional finite-volume coastal ocean model (FVCOM). A series of numerical experiments were carried out to investigate the effects of variations in coastline geometry on tides, storm surges, and storm tides. This model was calibrated using data observed at the Haimen and Ruian gauge stations and then used to reproduce the tides, storm surges, and storm tides in the Jiaojiang Estuary caused by Typhoon Winnie in 1997. Results show that the high tide level, peak storm surge, and high storm tide level at the Haimen Gauge Station increased along with the completion of reclamation projects, and the maximum increments caused by the third project were 0.13 m, 0.50 m, and 0.43 m, respectively. The envelopes with maximum storm tide levels of 7.0 m and 8.0 m inside the river mouth appeared to move seaward, with the latter shifting 1.8 km, 3.3 km, and 4.4 km due to the first project, second project, and third project, respectively. The results achieved in this study contribute to reducing the effects of, and preventing storm disasters after the land reclamation in the Jiaojiang Estuary.

Bovine parainfluenza virus 3 (BPIV3) is a crucial causative agent of respiratory disease in young and adult cattle. No specific therapies are available for BPIV3 infection. Understanding the internalization pathway of the virus will provide a new strategy for the development of antiviral therapy. Here, the mechanism of BPIV3 entry into HeLa cells was analyzed using RNA silencing and pharmacological inhibitors. Treatment of HeLa cells with hypertonic medium prevented BPIV3 internalization. These results indicated that BPIV3 entered HeLa cells via receptor-mediated endocytosis. Moreover, removing cell membrane cholesterol through MβCD treatment hampered viral penetration but not viral replication. In addition, BPIV3 infection was inhibited by pretreatment with dynasore or chlorpromazine (CPZ) or knockdown of dynamin II or clathrin heavy chain. However, virus entry was unaffected by nystatin, EIPA, wortmannin, or cytochalasin D treatment or caveolin-1 knockdown. These data demonstrated that the entry of BPIV3 into HeLa cells was dependent on clathrin-mediated endocytosis but not on caveolae-mediated endocytosis or the macropinocytosis pathway. Many viruses are transported to endosomes, which provide an acidic environment and release their genome upon separation from primary endocytic vesicles. However, we found that BPIV3 infection required endosomal cathepsins, but not a low pH. In summary, we show, for the first time, that BPIV3 enters HeLa cells through the clathrin-mediated endocytosis pathway, presenting novel insights into the invasion mechanism of Paramyxoviridae.

Hepatitis B virus (HBV) replicates its DNA genome through reverse transcription of a viral RNA pregenome. We report herein that the interferon (IFN) stimulated exoribonuclease gene of 20 KD (ISG20) inhibits HBV replication through degradation of HBV RNA. ISG20 expression was observed at basal level and was highly upregulated upon IFN treatment in hepatocytes, and knock down of ISG20 resulted in elevation of HBV replication and attenuation of IFN-mediated antiviral effect. The sequence element conferring the susceptibility of HBV RNA to ISG20-mediated RNA degradation was mapped at the HBV RNA terminal redundant region containing epsilon (ε) stem-loop. Furthermore, ISG20-induced HBV RNA degradation relies on its ribonuclease activity, as the enzymatic inactive form ISG20D94G was unable to promote HBV RNA decay. Interestingly, ISG20D94G retained antiviral activity against HBV DNA replication by preventing pgRNA encapsidation, resulting from a consequence of ISG20-ε interaction. This interaction was further characterized by in vitro electrophoretic mobility shift assay (EMSA) and ISG20 was able to bind HBV ε directly in absence of any other cellular proteins, indicating a direct ε RNA binding capability of ISG20; however, cofactor(s) may be required for ISG20 to efficiently degrade ε. In addition, the lower stem portion of ε is the major ISG20 binding site, and the removal of 4 base pairs from the bottom portion of ε abrogated the sensitivity of HBV RNA to ISG20, suggesting that the specificity of ISG20-ε interaction relies on both RNA structure and sequence. Furthermore, the C-terminal Exonuclease III (ExoIII) domain of ISG20 was determined to be responsible for interacting with ε, as the deletion of ExoIII abolished in vitro ISG20-ε binding and intracellular HBV RNA degradation. Taken together, our study sheds light on the underlying mechanisms of IFN-mediated HBV inhibition and the antiviral mechanism of ISG20 in general.

The zinc finger antiviral protein (ZAP) is a mammalian host restriction factor that inhibits the replication of a variety of RNA viruses, including retroviruses, alphaviruses and filoviruses, through interaction with the ZAP-responsive elements (ZRE) in viral RNA, and recruiting the exosome to degrade RNA substrate. Hepatitis B virus (HBV) is a pararetrovirus that replicates its genomic DNA via reverse transcription of a viral pregenomic (pg) RNA precursor. Here, we demonstrate that the two isoforms of human ZAP (hZAP-L and -S) inhibit HBV replication in human hepatocyte-derived cells through posttranscriptional down-regulation of viral pgRNA. Mechanistically, the zinc finger motif-containing N-terminus of hZAP is responsible for the reduction of HBV RNA, and the integrity of the four zinc finger motifs is essential for ZAP to bind to HBV RNA and fulfill its antiviral function. The ZRE sequences conferring the susceptibility of viral RNA to ZAP-mediated RNA decay were mapped to the terminal redundant region (nt 1820-1918) of HBV pgRNA. In agreement with its role as a host restriction factor and as an innate immune mediator for HBV infection, ZAP was upregulated in cultured primary human hepatocytes and hepatocyte-derived cells upon IFN-α treatment or IPS-1 activation, and in the livers of hepatitis B patients during immune active phase. Knock down of ZAP expression increased the level of HBV RNA and partially attenuated the antiviral effect elicited by IPS-1 in cell cultures. In summary, we demonstrated that ZAP is an intrinsic host antiviral factor with activity against HBV through down-regulation of viral RNA, and that ZAP plays a role in the innate control of HBV replication. Our findings thus shed light on virus-host interaction, viral pathogenesis, and antiviral approaches.

Background: Adolescence is a crucial period for the development of depression, and previous studies have suggested that the Behavioral Activation System (BAS) plays a significant role. However, little is known about the underlying mechanisms. This study aimed to explore the mediating role of anhedonia in the relationship between BAS and depressive symptoms among Chinese adolescents. Method: A total of 1,023 high-school students aged 15-18 years participated in the study, with 916 continuing their participation three months later. All participants completed the Behavioral Inhibition System/Activation System (BIS/BAS) scale, Dimensional Anhedonia Rating Scale (DARS), Children's Depression Inventory (CDI), and the State-Trait Anxiety Inventory (STAI-S/T). Pathway model analysis was performed to examine the concurrent and prospective mediating effects of anhedonia and the potential moderating effect of sex. Result: Anhedonia in the domains of social activities, hobbies and sensory experiences significantly mediated the cross-sectional relationship between BAS and depressive level three months later. Furthermore, the beta-value of the mediating effect of social activities was significantly higher than that of the other domains of hedonic capacity cross-sectionally and longitudinally. However, sex showed no significant moderating effect. Conclusion: Our findings underscore the importance of hedonic capacity, especially within the social domain, in the development of depressive symptoms. Bigelovin (BigV), as traditional Chinese medicine, has been shown to inhibit the malignant progression of hepatocellular carcinoma (HCC). This study aimed to investigate whether BigV affects the development of HCC by targeting the MAPT and Fas/FasL pathway. Human HCC cell lines HepG2 and SMMC-7721 were used for this study. Cells were treated with BigV, sh-MAPT, and MAPT. The viability, migration, and apoptosis of HCC cells were detected by CCK-8, Transwell, and flow cytometry assays, respectively. Immunofluorescence and immunoprecipitation were used to verify the relationship between MAPT and Fas. Subcutaneous xenograft tumor and tail vein-injected lung metastases mouse models were constructed for histological observation. Hematoxylin-eosin staining was used to assess lung metastases in HCC. Western blotting was used to measure the expression of migration, apoptosis, and epithelial-mesenchymal transition (EMT) marker proteins, as well as Fas/FasL pathway-related proteins. BigV treatment inhibited the proliferation, migration, and EMT of HCC cells, whereas enhanced cell apoptosis. Moreover, BigV downregulated MAPT expression. The negative effects of sh-MAPT on HCC cell proliferation, migration, and EMT were enhanced by BigV treatment. Conversely, BigV addition attenuated the positive effects of MAPT overexpression on the malignant progression of HCC. In vivo experiments showed that BigV and/or sh-MAPT reduced tumor growth and lung metastasis while promoting tumor cell apoptosis. Furthermore, MAPT could act with Fas and inhibit its expression. sh-MAPT upregulated the expression of Fas/FasL pathway-associated proteins, which were enhanced by BigV administration. [Objectives] To explore the role and molecular mechanisms of aerobic exercise in post-stroke depression. [Methods] Wistar rats were randomly divided into sham, sedentary' and aerobic exercise (Ex) groups. The rats in sedentary and Ex groups received middle cerebral artery occlusion (MCAO) surgery. Aerobic exercise was performed 24 h after MCAO for four weeks in Ex group. The sucrose preference test (SPT), open field test (OFT), tail suspension test (TST) was performed, respectively. The levels of IL-lß, IL-6, TNF-a were detected by ELISA. [Results] Aerobic exercise successfully prevented depression-like symptom induced by ischemic stroke w'ith sedentary behavior as indicated by significant increase in sucrose preference (P <0.01) during SPT and decrease in immobility time (P <0. 01) during FST and TST. Aerobic exercise significantly reduced the high levels of IL-lß, IL-6, TNF-a induced by ischemic stroke and sedentary' behavior in the ischemic hippocampus (P <0.05). [Conclusions] Aerobic exercise after ischemic stroke can reverse depression and improve the inflammatory microenvironment in the hippocampus. [ABSTRACT FROM AUTHOR]

Objectives: To explore the effects of high-intensity intermittent training ( HIT) on the degree of depression, neuronal morphology, and the expreesion of bran-derived neurotrophic factor ( BDNF) in the hippocampal dentate gyrus ( DG) of ischemic hemisphere in rats with post-stroke depreesion (PSD), and to investigate the potential mechanisms. #Methods: 40 male Wistar rats were randomly divided into control group, sham-operation group (Sham), PSD model group, moderate intensity aerobic training group (MIAT) and HIT group (n =8 for each group ). Except the control and Sham group, all other groups were given the middle cerebral arter occlusion ( MCAO ) using the intraluminal thread method, and chronic unpredictable mild strees (CUMS) was applied. After succusful modeling, training programs of diferent intensity were perormed in the MIAT and the HIT group. Rats in the five groups were subjected to behavioral tests ( open field test and tail suspension test)) before and 28 d after training. Nisl staining was used to obsere the morphological changes of neurons in the left hippocampal DG area. The expreesions of BDNF protein were detected by western blotting and immunohistochemistry. Results : After 28 d of training, compared with the other groups, the PSD group's pause time of the central lattice was prolonged ; the immobility time increased in tail suspension test; the expreesion of BDNF decreased in the left hippocampal DG area. The diference was statisticaUy significant ( 8<0.01). Neurons of hippocampal DG neurons were loosely afranged ; cells were miising. Nisl body was lightly stained and even disolved ; the etiological findings in MIAT and HIT rats were revered ; the exprusion of BDNF was elevated. Nisl staining showed that the cells were aaranged neatly ; the layers were dense, and the Nisl body was deeply stained. The changes in HIT group were more significant than that in MIAT group (8 <0.05). Conclusions: Compared with the traditional medium-intensity aerobic training, high-intensity interval training can improve the depression-like behavior in rats with post-stroke depreesion. The potential mechanism might be that HIT increase the neuroplasticity and ex- preesion of BDNF in hippocampal DG area. [ABSTRACT FROM AUTHOR]
