In this work, we carried out a comparative study of optical property on unintentionally doped and Sn-doped β-Ga2O3 crystals by edge-defined film-fed growth (EFG) method with a cylindrical iridium (Ir) die. The non-polarized Raman spectra showed the anisotropy in Raman peak position and intensity for three different planes (i.e., (100), (010) and (001)) of β-Ga2O3 crystals, independent of the Sn dopant. A difference in the Raman spectra between the unintentionally doped and Sn-doped β-Ga2O3 crystals was observed in the (001) plane. The strong in-plane anisotropy of the monoclinic β-Ga2O3 crystal related to the crystallographic orientation was demonstrated by the angle-resolved polarized Raman spectroscopy. The Sn dopant cannot change the optical transmittance and bandgap values of the unintentionally doped and Sn-doped β-Ga2O3 crystals with the (100), (010) and (001) orientations. However, the Sn-doped β-Ga2O3 crystal with the (010) orientation showed the higher valence band maximum and lower surface barrier height values. The X-ray photoelectron spectroscopy of the unintentionally doped and Sn-doped β-Ga2O3 crystals with the (010) orientation was also measured, which showed no obvious difference. The different luminescence centers and level of electron ionization of the unintentionally doped and Sn-doped β-Ga2O3 crystals were observed by the cathodoluminescence spectroscopy and electron paramagnetic resonance, respectively. Based on the comprehensive comparison, it can be concluded that the anisotropy in optical property of the unintentionally doped and Sn-doped β-Ga2O3 crystals with (100), (010) and (001) orientations apparently exist.

Erythropoietin-producing hepatocyte receptor A2 (EphA2) is an attractive target for immunotherapy due to its high expression in a variety of solid tumors including prostate cancer. Among various types of immunotherapeutics, chimeric antigen receptor T (CAR-T) cell therapy has made promising progress in hematological and solid tumors. Here, we detected the expression of EphA2 in prostate cancer cells and developed a second-generation CAR targeting EphA2 with CD28 as a co-stimulatory receptor to explore its tumor suppressive potential for prostate cancer in vitro and in vivo. EphA2 was highly expressed on the surface of PC3 and DU145 cells. EphA2 CART cells effectively inhibited prostate cancer growth in an antigen-dependent manner in vitro and in vivo. In addition, tumor cells could stimulate the proliferation of CAR-T cells and the release of cytokine IFN-γ in vitro. These findings shed light on EphA2 as a potential target for prostate cancer, promising EphA2 specific CAR-T cells for the treatment of prostate cancer.

Under the dual-carbon target, hydrogen energy, as a zero-carbon secondary energy source, has great scope for replacing fossil feedstocks in the fields of energy, transportation and industry. However, the current research on the competitiveness of hydrogen energy in various fields is not sufficiently addressed. In this paper, we use the LEAP model to predict the future scale of hydrogen use and the two-factor learning curve to predict the trend of hydrogen price change from 2025 to 2050, using Qinghai Province as the research background. At the same time, considering the carbon emission reduction benefits and raw material costs, the competitiveness of hydrogen energy in energy, transportation, industry and other fields in the future is compared. The results show that: 1) The hydrogen load scale in Qinghai Province will grow fast from 2025 to 2030. From 2030 to 2040, it slows under the steady and basic scenarios but remains high under the accelerated one. By 2040, the consumption scales are 1.057 million, 649,000 and 442,000 tons respectively. 2) The price of hydrogen energy will drop rapidly from the current 28 CNY/kg to about 20 CNY/kg in the next 5 years. By 2040, the price of hydrogen energy will be reduced to about 17 CNY/kg. 3) In terms of hydrogen energy competitiveness, when carbon emissions are not taken into account, hydrogen energy is currently competitive in the transportation field. During 2032–2038, it will be competitive in the field of methanol synthesis. By 2040, hydrogen energy will not be competitive in the fields of ammonia synthesis and power/heating. When considering carbon emissions, the competitiveness of hydrogen energy in the transportation field will become greater. The competitive year in the field of methanol synthesis will be 1–2 years ahead. By 2040, it will not be competitive in the field of synthetic ammonia and power/heating, but the gap will be significantly reduced due to the consideration of carbon emissions.

Strawberry is frequently attacked by mites, which directly affects the yield and quality of this fruit species. The WRKY Group III transcription factors (TFs) play an important role in plant tolerance to biotic sources of stress, such as pathogens and insect pests. In this study, six Group III WRKY TFs (FaWRKY25, FaWRKY31, FaWRKY32, FaWRKY43, FaWRKY44, and FaWRKY45) were identified in strawberry. A phylogenetic analysis showed that the six WRKY III TFs were divided into two clades and all had a conserved WRKYGQK domain and the C-X7-C-X23-H-T-C zinc finger motif. An interaction network analysis revealed that FaWRKY44 was co-expressing with FaWRKY25 and FaWRKY45. The expression patterns showed that the WRKY Group III genes responded to plant hormones and mite infestation in strawberry. To further verify the role of FaWRKY25 in plant resistance to mites, we cloned the FaWRKY25 gene and overexpressed it in transgenic plants. An in vivo subcellular localization analysis indicated that the FaWRKY25 protein was localized in the nucleus. Fewer mites were also detected on the wild-type plants than on FaWRKY25-overexpressing transgenic plants, suggesting that FaWRKY25 negatively regulates the resistance of strawberry to mites. The present study advances our understanding on a potential target that mites use to manipulate host plant defenses.

IntroductionCancer represents a significant global public health concern. In recent years, the incidence of cancer has been on the rise worldwide due to various factors, including diet, environment, and an aging population. Simultaneously, advancements in tumor molecular biology and genomics have led to a shift from systemic chemotherapy focused on disease sites and morphopathology towards precise targeted therapy for driver gene mutations. Therefore, we propose a comprehensive review aimed at exploring the research hotspots and directions in the field of Kirsten rat sarcoma viral oncogene homolog (KRAS)-mutant cancers over the past decade, providing valuable insights for cancer treatment strategies. Specifically, we aim to present an intellectual landscape using data obtained from the Web of Science (WoS) regarding KRAS mutation.MethodsBibliometrix, VOSviewer, CiteSpace, and HistCite were employed to conduct scientometric analyses on national publications, influential authors, highly cited articles, frequent keywords, etc.ResultsA total of 16,609 publications met the screening criteria and exhibited a consistent annual growth trend overall. Among 102 countries/regions, the United States occupied the vast majority share of the published volume. The journal Oncotarget had the highest circulation among all scientific publications. Moreover, the most seminal articles in this field primarily focus on biology and targeted therapies, with overcoming drug resistance being identified as a future research direction.ConclusionThe findings of the thematic analysis indicate that KRAS mutation in lung cancer, the prognosis following B-Raf proto-oncogene, serine/threonine kinase (BRAF) or rat sarcoma (RAS) mutations, and anti-epidermal growth factor receptor (EGFR)-related lung cancer are the significant hotspots in the given field. Considering the significant advancements made in direct targeting drugs like sotorasib, it is anticipated that interest in cancers associated with KRAS mutations will remain steadfast.

Abstract Posterior capsule opacification (PCO) is the most common complication after cataract surgery. Drug-eluting intraocular lens (IOLs) is a promising concept of PCO treatment in modern cataract surgery. However, the large dose of drugs in IOL leads to uncontrollable and unpredictable drug release, which inevitably brings risks of overtreatment and ocular toxicity. Herein, a low-power NIR-triggered thermosensitive IOL named IDG@P(NIPAM-co-AA)-IOL is proposed to improve security and prevent PCO by synergetic controlled drug therapy and simultaneous photo-therapy. Thermosensitive polymer brushes Poly(N-isopropylacrylamide-co-Acrylic acid) (P(NIPAM-co-AA)) is prepared on IOL via surface-initiated reversible addition-fragmentation chain transfer (SI-RAFT) polymerization. Then, Doxorubicin (DOX) and Indocyanine green (ICG) co-loaded Gelatin NPs (IDG NPs) are loaded in P(NIPAM-co-AA) by temperature control. The IDG NPs perform in suit photodynamic & photothermal therapy (PTT&PDT), and the produced heat also provides a trigger for controllable drug therapy with a cascade effect. Such functional IOL shows excellent synergistic drug-phototherapy effect and NIR-triggered drug release behavior. And there is no obvious PCO occurrence in IDG@P(NIPAM-co-AA) IOL under NIR irradiation compared with control group. This proposed IDG@P(NIPAM-co-AA)-IOL serves as a promising platform that combines phototherapy and drug-therapy to enhance the therapeutic potential and medication safety for future clinical application of PCO treatment.

The synergetic strategy has created tremendous advantages in drug-resistance bacterial infection treatment, whereas challenges related to novel compound discovery and identifying drug-binding targets still remain. The mechanisms of antimicrobial resistance involving β-lactamase catalysis and the degradation of β-lactam antibiotics are being revealed, with relevant therapies promising to improve the efficacy of existing major classes of antibiotics in the foreseeable future. In this study, it is demonstrated that nordalbergin, a coumarin isolated from the wood bark of Dalbergia sissoo, efficiently potentiated the activities of β-lactam antibiotics against methicillin-resistant Staphylococcus aureus (MRSA) by suppressing β-lactamase performance and improving the bacterial biofilm susceptibility to antibiotics. Nordalbergin was found to destabilize the cell membrane and promote its permeabilization. Moreover, nordalbergin efficiently improved the therapeutic efficacy of amoxicillin against MRSA pneumonia in mice, as supported by the lower bacterial load, attenuated pathological damage, and decreased inflammation level. These results demonstrate that nordalbergin might be a promising synergist of amoxicillin against MRSA infections. This study provided a new approach for developing potentiators for β-lactam antibiotics against MRSA infections.

Abstract B7-H3 is an attractive target for immunotherapy because of its high expression across multiple solid tumors, including prostate cancer, and restricted expression in normal tissues. Among various types of tumor immunotherapy, chimeric antigen receptor T (CAR-T) cell therapy has shown remarkable success in hematological tumors. However, the potency of CAR-T cell therapy in solid tumors is still limited. Here, we examined the expression of B7-H3 in prostate cancer tissues and cells and developed a second-generation CAR that specifically targets B7-H3 and CD28 as costimulatory receptor to explore its tumoricidal potential against prostate cancer in vitro and in vivo. The high expression of B7-H3 was detected on both the surface of PC3, DU145 and LNCaP cells and prostate cancer tissues. B7-H3 CAR-T cells efficiently controlled the growth of prostate cancer in an antigen-dependent manner in vitro and in vivo. Moreover, tumor cells could induce the proliferation of CAR-T cells and the release of high levels of cytokines of IFN-γ and TNF-α in vitro. Results demonstrated that B7-H3 is a potential target for prostate cancer therapy that supports the clinical development of B7-H3 specific CAR-T cells for prostate cancer.

IntroductionBreast cancer remains a significant global health challenge, accounting for 2.3 million new cases in 2020 and ranking as the most prevalent cancer by incidence and the fourth in cancer-related mortality worldwide. In China, breast cancer also rapidly increases incidence and burden. The research of exosomes in breast cancer has attracted more and more attention and has a rapid development. Recognizing the pivotal role of exosomes in breast cancer research, we have undertaken a comprehensive scientometric analysis of pertinent scholarly articles published over the past decade to elucidate the current research landscape for researchers.MethodsIn this study, we gathered all pertinent publications from the Web of Science. Biblioshiny (a web interface for Bibliometrix), VOSviewer software, and CiteSpace software were used to analyze the information on publications, including global trends, countries, institutions, journals, authors, keywords, and citations.ResultsA total of 1,239 articles and 625 review articles were retrieved. The annual global publication output has an increased trend in recent decades overall. China contributed the most articles. The publications of the USA had the most total link strength. Nanjing Medical University had the most total link strength. The most relevant source was the International Journal of Molecular Sciences. Tang JH contributed the most articles and had the highest H-index, G-index, and total link strength. The most cited document was “Tumor exosome integrins determine organotropic metastasis”, with 2730 citations. The basic themes included “exosomes”, “expression”, “cells”, “identification”, “biomarkers”, and “serum”. The keyword “membrane vesicle” had the strongest bursts. The keywords “target”, “biology”, “suppressor cell”, “molecular mechanism”, “tumor progression”, “inhibitor”, and “model” appeared as prominent focal points in current research and active areas of exploration.ConclusionOver the past decade, exosome research in breast cancer has undergone a discernible evolution, shifting from broader investigations of exosome roles to focused exploration of specific pathways relevant to breast cancer. Notably, the emphasis has extended to the clinical application of exosomes as biomarkers and potential therapeutic agents in breast cancer treatment.

Accurate determination of protein localization, levels, or protein−protein interactions is pivotal for the study of their function, and in situ protein labeling via homologous recombination has emerged as a critical tool in many organisms. While this approach has been refined in various model fungi, the study of protein function in most plant pathogens has predominantly relied on ex situ or overexpression manipulations. To dissect the molecular mechanisms of development and infection for Verticillium dahliae, a formidable plant pathogen responsible for vascular wilt diseases, we have established a robust, homologous recombination-based in situ protein labeling strategy in this organism. Utilizing Agrobacterium tumefaciens-mediated transformation (ATMT), this methodology facilitates the precise tagging of specific proteins at their C-termini with epitopes, such as GFP and Flag, within the native context of V. dahliae. We demonstrate the efficacy of our approach through the in situ labeling of VdCf2 and VdDMM2, followed by subsequent confirmation via subcellular localization and protein-level analyses. Our findings confirm the applicability of homologous recombination for in situ protein labeling in V. dahliae and suggest its potential utility across a broad spectrum of filamentous fungi. This labeling method stands to significantly advance the field of functional genomics in plant pathogenic fungi, offering a versatile and powerful tool for the elucidation of protein function.

Background: Numerous studies have demonstrated a close relationship between antioxidant-rich diets and comorbidities as well as mortality. However, the relationship between such diets and aging remains unclear. The purpose of this study was to investigate the association between the Composite Dietary Antioxidant Index (CDAI) and aging. Methods: All participants were from the National Health and Nutrition Examination Survey (NHANES) 2001–2010. Phenotypic age was calculated using a formula and subtracted from the chronological age to determine the aging. When the phenotypic age exceeded the chronological age, it was considered as aging. A weighted logistic regression model was employed to explore the relationship between CDAI and aging. Restricted cubic splines (RCSs) were used to examine the potential nonlinear relationship between them. Subgroup analysis and joint analysis were conducted to explore the effect of modifiers in these relationships. Results: A total of 19,212 participants (weighted: 165,285,442 individuals) were included in this study. The weighted logistic regression model showed a significant correlation between CDAI and the risk of aging (OR = 0.90, 95% CI: 0.84–0.96). RCS analysis revealed an L-shaped dose–response relationship between CDAI and the risk of aging. Subgroup analysis indicated that the association between CDAI and aging was more pronounced in middle-aged individuals and non-smokers. The joint analysis demonstrated that although smoking accelerated aging among participants, a high CDAI diet could still offset these damages. Conclusions: The association between high CDAI and reduced risk of aging is particularly significant in young and middle-aged individuals and non-smokers. Consuming foods rich in CDAI components may potentially lower the risk of aging.

Objective Recurrent disease flare is one of the key problems in lupus patients. A Chinese Flare-Prevention Lupus Initiative Cohort (FLIC) was established. Risk factors of disease flare were evaluated accordingly.Methods Patients with low-grade disease activity (the Safety of Estrogens in Lupus Erythematosus National Assessment–SLE Disease Activity Index (SELENA-SLEDAI) =≤6, daily prednisone ≤20 mg, no British Isles Lupus Assessment Group A or no more than one B organ domain score) from January 2014 to August 2020 were included in the FLIC. Disease flares were defined by the modified SELENA-­SLEDAI Flare Index. Low disease activity status (LDAS) and remission were also assessed. The cumulative flare rate was estimated by an event per 100 person-years analysis. Cox proportional hazards models were performed to identify risk factors of subsequent disease flares after adjusting clinical confounders. Survival was assessed with the Kaplan-Meier method.Results 448 eligible patients with low-grade disease activity were included in FLIC. During a mean follow-up of 30.4 months, 170 patients flared. The cumulative lupus flare rate was 22.2 events per 100 patient-years. Compared with patients without flare, those with lupus flares were taking more prednisone, had higher disease activity index and with less patients attained LDAS/remission at baseline. They also had higher rates of antiphospholipid antibodies (aPLs) and antiribosomal P antibody. Cox regression analysis confirmed that attainment of either LDAS or remission at baseline were independent protective factors against subsequent disease flare (LDAS but not in remission: HR 0.58, 95% CI 0.38~0.88; remission: HR 0.46, 95% CI 0.30~0.69), while aPL was a risk factor of lupus flares (HR 1.95, 95% CI 1.36~2.78). Kaplan-Meier curves indicated that attaining LDAS or remission and absence of aPL at baseline had the least flare risk.Conclusions In our real-world cohort study, not attaining LDAS or remission at baseline and aPL positivity was associated with higher risk of disease flares in patients with low-grade SLE.

Objective To evaluate the risk of major infections and the relationship between major infections and mortality in patients with newly diagnosed SLE.Methods A newly diagnosed (10, peripheral lymphocyte count 104 µmol/L was established to identify patients at low risk (3%–5%) or high risk (37%–39%) of major infections within the first 4 months.Conclusions Newly onset active SLE is susceptible to major infections, which is probably due to underlying profound immune disturbance. Identifying high-risk patients using an appropriate prediction tool might lead to better tailored management and better outcome.

Abstract Background Systemic lupus erythematosus is an autoimmune disease characterized by an overproduction of autoantibodies resulting from dysregulation in multiple immune cell types. D-mannose is a C− 2 epimer of glucose that exhibits immunoregulatory effects in models of autoimmune diseases, such as type 1 diabetes, induced rheumatoid arthritis, and airway inflammation. This study was conducted to evaluate the efficacy of D-mannose treatment in mouse models of lupus. Results Firstly, the effect of D-Mannose was evaluated by flow cytometry on the in vitro activation of non-autoimmune C57BL/6 (B6) bone marrow-derived dendritic cells (BMDCs) and their ability to induce antigen-specific CD4+ T cell proliferation and activation. D-mannose inhibited the maturation of BMDCs and their induction of antigen-specific T cell proliferation and activation. In vivo, D-mannose increased the frequency of Foxp3+ regulatory T cells in unmanipulated B6 mice. To assess the effect of D-mannose in mouse models of lupus, we used the graft-versus-host disease (cGVHD) induced model and the B6.lpr spontaneous model. In the cGVHD model, D-mannose treatment decreased autoantibody production, with a concomitant reduction of the frequency of effector memory and follicular helper T cells as well as germinal center B cells and plasma cells. These results were partially validated in the B6.lpr model of spontaneous lupus. Conclusion Overall, our results suggest that D-mannose ameliorates autoimmune activation in models of lupus, at least partially due to its expansion of Treg cells, the induction of immature conventional dendritic cells and the downregulation of effector T cells activation. D-Mannose showed however a weaker immunomodulatory effect in lupus than in other autoimmune diseases.

Here the authors identify a unilateral neural circuit from the CamKII-positive neurons in the anterior insular cortex to the vGluT2-positive neurons in the lateral hypothalamus that controls feeding responses to visceral aversive stimuli.

Ce-doped strontium bismuth titanate (SrBi4Ti4O15, SBT) powders were prepared by glycine-nitrate process at 400–500 °C and the ceramics were sintered at 980 °C. The phase composition, morphology and electric properties were investigated. It was found that the calcined powders consist of a single phase SBT and the calcination temperature is lower than that for the conventional solid state method. The morphology of Cedoped SBT is flake-like and the layer size decreases with Ce-addition from 12 to 2 µm confirming that the addition of Ce3+ inhibits grain growth. The Curie temperature of Ce-doped SBT increased for about 20 °C compared to the pure SBT. The tan δ was ∼0.005 at 35 °C and even below 0.05 up to 400 °C. The temperature coefficient of dielectric constant was ∼0.012 and the rate of frequency change was 0.01–0.04, which indicated the high stability of dielectric properties of the Ce-doped SBT. Impedance analysis revealed that the conduction mechanism of the Ce-doped SrBi4Ti4O15 ceramics is mainly grain conduction.

Objective To confirm that metformin prevents flares in patients with SLE with low disease activity, we performed a post hoc analysis combining our previous two randomised trials.Methods Post hoc analyses were performed on data from the open-labelled proof-of-concept trial (n=113, ChiCTR-TRC-12002419) and placebo-controlled ‘Met Lupus’ trial (n=140, NCT02741960) comparing the efficacy of metformin versus placebo/nil add-on to standard therapy in patients with SLE with low disease activity (SELENA-SLEDAI ≤4). The primary endpoint was defined by the SELENA-SLEDAI Flare Index at 12-month follow-up. A subgroup analysis was performed.Results Overall, 201 eligible patients were included, with 99 allocated to metformin group and 102 allocated to the placebo/nil group. By 12 months of follow-up, 21 patients (21.2%) flared in the metformin group, as compared with 36 (35.3%) in the placebo/nil group (p=0.027, risk ratio=0.68, 95% CI 0.46 to 0.96). Fin field-effect transistor (FinFET) technology has been introduced to the mainstream complementary metal-oxide semiconductor (CMOS) manufacturing for low-power and high-performance applications. However, advanced FinFET nodes are facing significant challenges to enhance the device performance due to the increasingly prominent parasitic resistance and capacitance. In this study, for the first time, we demonstrate methods of enhancing p-channel FinFET (pFET) performance on a fully integrated advanced FinFET platform via source/drain (S/D) cavity structure optimization. By modulating the cavity depth and proximity around the optimal reference point, we show that the trade-off between the S/D resistance and short channel effect, as well as the impact on the parasitic capacitance must be considered for the S/D cavity structure optimization. An extra process knob of applying cavity implant on the desired cavity structure was also demonstrated to modify the S/D junction profile for device performance enhancement.

Abstract Development of conformal n-channel organic phototransistor (OPT) array is urgent for future applications of organic complementary circuits in portable and wearable electronics and optoelectronics. In this work, the ultrathin conformal OPT array based on air-stable n-type PTCDI-C13H27 was fabricated. The OPT array shows excellent electrical and photoelectrical performance, good device uniformity, and remains stable in electron mobility by 83% after 90 days compared to the initial values. Eventhough mobility, on-state current, off-state current, and photocurrent of PTCDI-C13H27 thin film phototransistor show slight decrease with the decreased bending radius, the device still remains the stable photosensitivity as high as 104 when the device is freely adhered on the 2D surfaces and 3D hemispherical sphere, which is in a class with the highest photosensitivity for perylene diimide derivatives. These results present the promising application potential of our conformable air-stable n-type PTCDI-C13H27 OPTs as the photodetection system of curved artificial compound eyes in wearable and portable electronics and optoelectronics.

CD4+ T cells have numerous features of over-activated cellular metabolism in lupus patients and mouse models of the disease. This includes a higher glycolysis than in healthy controls. Glucose transporters play an essential role in glucose metabolism by controlling glucose import into the cell from the extracellular environment. We have previously shown that treatment of lupus-prone mice with 2-deoxy-D-glucose, which inhibits the first step of glycolysis was sufficient to prevent autoimmune activation. However, direct targeting of glucose transporters has never been tested in a mouse model of lupus. Here, we show that CG-5, a novel glucose transporter inhibitor, ameliorated autoimmune phenotypes in a spontaneous lupus-prone mouse model, B6.NZM2410.Sle1.Sle2.Sle3 (Triple-congenic, TC), and in a chronic graft- vs. host-disease (cGVHD) model of induced lupus. In vitro, CG-5 blocked glycolysis in CD4+ T cells, and limited the expansion of CD4+ T cells induced by alloreactive stimulation. CG-5 also modulated CD4+ T cell polarization by inhibiting Th1 and Th17 differentiation and promoting regulatory T (Treg) induction. Moreover, CG-5 treatment reduced lupus phenotypes including the expansion of germinal center B (GC B) cells, as well as the production of autoantibodies in both TC mice and cGVHD models. Finally, CG-5 blocked glycolysis in human T cells. Overall, our data suggest that blocking glucose uptake with a small molecule inhibitor ameliorates autoimmune activation, at least partially due to its inhibition of glycolysis in CD4+ T cells.

The pharmacokinetics of our previous prepared oxfendazole nanosuspension was studied in sheep to understand its enhancement of bioavailability. Ten sheep were randomly divided into two groups, each of which received a singledose (5mg/kg) oral oxfendazole nanosuspension and oxfendazole granules. After intragastric administration, oxfendazole rapidly reached peak concentrations of 11.25 μg/mL and 5.50 μg/mL at 5.6 h and 6.6 h in the nanosuspension group and granules group and the concentrations gradually reduced to below the detection limit at 96 h and 72h, respectively. The main pharmacokinetic parameters of oxfendazole after administration to the sheep: The Tmax, Cmax, AUClast, MRTlast, T1/2 and Ke of in nanosuspension were 5.6 h, 11.26 μg/mL, 179.22 μg*h/mL, 17.82 h, 87.22 h and 0.012 1/h, respectively, while the these main pharmacokinetic parameters in the OFZ granules group were 6.6 h, 5.50 μg/mL, 105.28 μg*h/mL, 16.55 h, 35.04 h, and 0.020 1/h, respectively. Compared with the granules group, the Cmax and AUClast of the nanosuspension group were increased by 2.05 and 1.70 times, respectively, and the Cmax and AUClast of the total active compound in the nanosuspension group were increased by 1.85 times and 1.71 times, respectively. Seed dormancy is a natural phenomenon in plants. It ensures that seeds complete the grain-filling stage before germination and prevents germination in unsuitable ecological conditions. In this study, we determined the previously unknown function of the rice (Oryza sativa) gene GERMIN-LIKE PROTEIN 2-1 (OsGLP2-1) in seed dormancy. Using artificial microRNA and CRISPR/CAS9 approaches, suppression of OsGLP2-1 expression in rice resulted in the release of dormancy in immature seeds. Conversely, overexpression of OsGLP2-1 driven by the OsGLP2-1 native promoter led to greater seed dormancy. Seed scutellum-specific expression of OsGLP2-1 was increased by exogenous abscisic acid, but decreased with gibberellic acid treatment. We provide evidence that OsGLP2-1 is antagonistically controlled at the transcriptional level by ABA INSENSITIVE5 and GAMYB transcription factors. We conclude that OsGLP2-1 acts as a buffer, maintaining appropriate equilibrium for the regulation of primary dormancy during seed development in rice. [ABSTRACT FROM AUTHOR]

In this paper, we present surface urban heat island (SUHI) analysis of Shanghai (China) based on the change in land use and land cover using satellite Landsat images from 2002 to 2013. With the rapid development of urbanization, urban ecological and environmental issues have aroused widespread concern. The urban heat island (UHI) effect is a crucial problem, as its generation and evolution are closely related to social and economic activities. Land-use and land-cover change (LUCC) is the key in analyzing the UHI effect. Shanghai, one of China’s major economic, financial and commercial centers, has experienced high development density for several decades. A tremendous amount of farmland and vegetation coverage has been replaced by an urban impervious surface, leading to an intensive SUHI effect, especially in the city’s center. Luckily, the SUHI trend has slowed due to reasonable urban planning and relevant green policies since the 2010 Expo. Data analyses demonstrate that an impervious surface (IS) has a positive correlation with land surface temperature (LST) but a negative correlation with vegetation and water. Among the three factors, impervious surface is the most relevant. Therefore, the policy implications of land use and control of impervious surfaces should pay attention to the relief of the current SUHI effect in Shanghai. [ABSTRACT FROM AUTHOR]

Real-time indoor localization based on supporting infrastructures like wireless devices and QR codes are usually costly and labor intensive to implement. In this study, we explored a cheap alternative approach based on images for indoor localization. A user can localize him/herself by just shooting a photo of the surrounding indoor environment using the mobile phone. No any other equipment is required. This is achieved by employing image-matching and searching techniques with a dataset of pre-captured indoor images. In the beginning, a database of structured images of the indoor environment is constructed by using image matching and the bundle adjustment algorithm. Then each image's relative pose (its position and orientation) is estimated and the semantic locations of images are tagged. A user's location can then be determined by comparing a photo taken by the mobile phone to the database. This is done by combining quick image searching, matching and the relative orientation. This study also try to explore image acquisition plans and the processing capacity of off-the-shell mobile phones. During the whole pipeline, a collection of indoor images with both rich and poor textures are examined. Several feature detectors are used and compared. Pre-processing of complex indoor photo is also implemented on the mobile phone. The preliminary experimental results prove the feasibility of this method. In the future, we are trying to raise the efficiency of matching between indoor images and explore the fast 4G wireless communication to ensure the speed and accuracy of the localization based on a client-server framework. Fraxetin is one of the main constituents of the traditional medicinal plant Fraxinus rhynchophylla. The inhibitory effect of fraxetin on various bacterial strains has been extensively reported, however, its mechanism of action on bacterial cells remains to be elucidated. In the present study, the antibacterial mechanism of fraxetin on Staphylococcus aureus was systematically investigated by examining its effect on cell membranes, protein synthesis, nucleic acid content and topoisomerase activity. The results indicated that fraxetin increased the permeability of the cell membrane but did not render it permeable to macromolecules, such as DNA and RNA. Additionally, the quantity of protein, DNA and RNA decreased to 55.74, 33.86 and 48.96%, respectively following treatment with fraxetin for 16 h. The activity of topoisomerase I and topoisomerase II were also markedly inhibited as fraxetin concentration increased. The result of the ultraviolet-visible spectrophotometry demonstrated that the DNA characteristics exhibited a blue shift and hypochromic effect following treatment with fraxetin. These results indicated that fraxetin had a marked inhibitory effect on S.aureus proliferation. Further mechanistic studies showed that fraxetin could disrupt nucleic acid and protein synthesis by preventing topoisomerase from binding to DNA. [ABSTRACT FROM AUTHOR]

Mesoporous ZrO2 with surface area of 100–180 m2/g and pore size of 2.5–3.5 nm can be prepared by copying the middle phase template formed in the trialkyl phosphine oxide(TRPO)–kerosene/Zr(IV)–HCl extraction system. The microstructure of the middle phase template is related to the initial composition of the extraction system. Excessive solvent, extractant, and polar additives(such as long-chain alcohol) solubilized in the middle phase cause the bi-continuous structure to be changed, or even destroyed forming a w/o microemulsion. The key to obtain mesoporous ZrO2 successfully is to prepare the middle phase template with an optimized bi-continuous structure. [ABSTRACT FROM AUTHOR]
