2. A comparative study of glutathione-coated iron oxide and glutathione-coated core-shell magnetic nanoparticles for their antiviral activities

Iron oxide nanoparticles and its nanocomposites have attracted attention because of their potential applications in biomedicine. Here, firstly the Fe3O4 nanoparticles were prepared and then Ag was deposited by reducing the Ag salt onto the surface of the Fe3O4 nanoparticles. This way, bimetallic nanoparticles were obtained. The synthesized nanoparticles were characterized using ultraviolet-visible absorption spectroscopy, transmission electron microscopy and X-ray diffraction and the size and surface charge of the nanoparticles were determined by the dynamic light scattering (DLS) and zeta potential. The spectrographic data demonstrated the size of the glutathione-coated Fe3O4 nanoparticles to be 4.48 nm and glutathione-coated core-shell magnetic nanoparticles to be 7.98 nm with the spherical morphology and well monodispersed. This study was also designed to investigate the inhibitory effect of Ag@Fe3O4-GSH, Fe3O4-GSH and glutathione (GSH) against Human Herpes Simplex Virus Type 1 (HSV-1), Human Adenovirus Type 5, Human Poliovirus Type 1, and Bovine coronavirus. The significant inhibition of Ag@Fe3O4-GSH was observed against Poliovirus (4 Log), Adenovirus (3 Log), and HSV-1 (2 Log), respectively. GSH showed remarkable antiviral effect against Bovine coronavirus (3 Log) while it exhibited log reduction (1 Log) against HSV-1 and poliovirus. Fe3O4-GSH showed a reduction of 1 Log only for RNA viruses such as poliovirus and bovine coronavirus. These results demonstrate promising antiviral activity, highlighting the potential of these nanoparticles in combating viral infections.

3. Detecting viable but non-culturable lactic acid bacteria following spray-drying and during storage

Microencapsulation with various materials has been used as an efficient method to improve the viability of probiotic bacteria in multiple food products and the human gastrointestinal tract. Although plate count agar is the most commonly used method for evaluating the viability of encapsulated bacteria, it is still far from providing reliable information about the intermediate state between viable and dead bacteria. This study optimized a tetrazolium salt-based colorimetric method for the detection of viable but non-culturable state within encapsulated Lactobacillus rhamnosus and Lactobacillus plantarum probiotic strains. The viability of encapsulated bacteria was assessed after different spray-drying conditions and also during two months of storage at room temperature. The ability to reduce tetrazolium salts of two lactic acid bacteria was verified and calibrated according to the experimental conditions (strains, incubation time, and microencapsulation material). The loss of bacterial cultivability was species-specific and more problematic throughout the processing than during the storage period. An outlet temperature of 73-75 °C yielded a higher viable but non-culturable state level than at 68-69 °C, especially in maltodextrin and trehalose powders. Whey protein was statistically the best carrier in preserving viable and culturable encapsulated bacteria after spray-drying and during storage, as compared to sugar-based carriers. The tetrazolium-optimized method was more sensitive and accurate for the evaluation of viable bacteria in microcapsules as compared to the conventional plate count methods available. It showed the high variability of CFU counts on Man–Rogosa–Sharpe (MRS) agar. This colorimetric technique could be considered a real-time, simple, cost-effective, and reliable alternative to culture-based methods in evaluating probiotic microencapsulation efficiency.

5. The Potential Role of Boron in the Modulation of Gut Microbiota Composition: An In Vivo Pilot Study

Background/Objectives: The role of the gut microbiome in the development and progression of many diseases has received increased attention in recent years. Boron, a trace mineral found in dietary sources, has attracted interest due to its unique electron depletion and coordination characteristics in chemistry, as well as its potential role in modulating the gut microbiota. This study investigates the effects of inorganic boron derivatives on the gut microbiota of mice. Methods: For three weeks, boric acid (BA), sodium pentaborate pentahydrate (NaB), and sodium perborate tetrahydrate (SPT) were dissolved (200 mg/kg each) in drinking water and administered to wild-type BALB/c mice. The composition of the gut microbiota was analyzed to determine the impact of these treatments. Results: The administration of BA significantly altered the composition of the gut microbiota, resulting in a rise in advantageous species such as Barnesiella and Alistipes. Additionally, there was a decrease in some taxa associated with inflammation and illness, such as Clostridium XIVb and Bilophila. Notable increases in genera like Treponema and Catellicoccus were observed, suggesting the potential of boron compounds to enrich microbial communities with unique metabolic functions. Conclusions: These findings indicate that boron compounds may have the potential to influence gut microbiota composition positively, offering potential prebiotic effects. Further research with additional analyses is necessary to fully understand the interaction between boron and microbiota and to explore the possibility of their use as prebiotic agents in clinical settings.

6. Synergistic anti-cancer effect of sodium pentaborate pentahydrate, curcumin and piperine on hepatocellular carcinoma cells

Abstract Hepatocellular carcinoma (HCC) is a leading cause of cancer-related death in the world. Poor prognosis of HCC patients is a major issue, thus, better treatment options for patients are required. Curcumin (Cur), hydrophobic polyphenol of the plant turmeric, shows anti-proliferative, apoptotic, and anti-oxidative properties. Boron is a trace element which is essential part of human nutrition. Sodium pentaborate pentahydrate (NaB), a boron derivative, is an effective agent against cancer. In the current study, we performed in vitro experiments and transcriptome analysis to determine the response of NaB, Cur, piperine (Pip) and their combination in two different HCC cell lines, HepG2 and Hep3B. NaB and Cur induced cytotoxicity in a dose and time dependent manner in HepG2 and Hep3B, whereas Pip showed no significant toxic effect. Synergistic effect of combined treatment with NaB, Cur and Pip on HCC cells was observed on cytotoxicity, apoptosis and cell cycle assay. Following in vitro studies, we performed RNA-seq transcriptome analysis on NaB, Cur and Pip and their combination on HepG2 and Hep3B cells. Transcriptome analysis reveals combined treatment of NaB, Cur and Pip induces anti-cancer activity in both of HCC cells.

7. Dual- or single rinse? The tubular sealer penetration of endodontic chelating agents.

IntroductionIn this study, we aimed to compare the effectiveness of various chelating agents, ethilenediaminetetraacetic acid (EDTA), citric acid (CA), and etidronic acid (HEDP) mixed in two different forms, in removing the smear layer and promoting the penetration of an endodontic sealer into the dentinal tubules of extracted single-rooted teeth.MethodsThe study used 75 teeth divided into five groups: 17% EDTA, 10% CA, 9% HEDP + NaOCl, 9% HEDP + distilled water (DW), and a control (DW) group. Scanning electron microscopy was used to assess smear layer removal and confocal laser microscopy was used to evaluate tubular sealer penetration at different depths from the apical tip.ResultsSealer penetration was highest with 17% EDTA and 10% CA as compared with the other agents (pConclusionWithin the limitations of this study, highest values were seen in EDTA and CA in terms of sealer penetration and smear layer removal. In the light of these findings, the use of strong chelating agents highlights better clinical efficiency than dual-rinse or single HEDP irrigation.

Abstract The SARS-CoV-2 virus caused the most severe pandemic around the world, and vaccine development for urgent use became a crucial issue. Inactivated virus formulated vaccines such as Hepatitis A and smallpox proved to be reliable approaches for immunization for prolonged periods. In this study, a gamma-irradiated inactivated virus vaccine does not require an extra purification process, unlike the chemically inactivated vaccines. Hence, the novelty of our vaccine candidate (OZG-38.61.3) is that it is a non-adjuvant added, gamma-irradiated, and intradermally applied inactive viral vaccine. Efficiency and safety dose (either 1013 or 1014 viral RNA copy per dose) of OZG-38.61.3 was initially determined in BALB/c mice. This was followed by testing the immunogenicity and protective efficacy of the vaccine. Human ACE2-encoding transgenic mice were immunized and then infected with the SARS-CoV-2 virus for the challenge test. This study shows that vaccinated mice have lowered SARS-CoV-2 viral RNA copy numbers both in oropharyngeal specimens and in the histological analysis of the lung tissues along with humoral and cellular immune responses, including the neutralizing antibodies similar to those shown in BALB/c mice without substantial toxicity. Subsequently, plans are being made for the commencement of Phase 1 clinical trial of the OZG-38.61.3 vaccine for the COVID-19 pandemic.

Abstract COVID-19 outbreak caused by SARS-CoV-2 created an unprecedented health crisis since there is no vaccine for this novel virus. Therefore, SARS-CoV-2 vaccines have become crucial for reducing morbidity and mortality. In this study, in vitro and in vivo safety and efficacy analyzes of lyophilized vaccine candidates inactivated by gamma-irradiation were performed. The candidate vaccines in this study were OZG-3861 version 1 (V1), an inactivated SARS-CoV-2 virus vaccine, and SK-01 version 1 (V1), a GM-CSF adjuvant added vaccine. The candidate vaccines were applied intradermally to BALB/c mice to assess toxicity and immunogenicity. Preliminary results in vaccinated mice are reported in this study. Especially, the vaccine models containing GM-CSF caused significant antibody production with neutralization capacity in absence of the antibody-dependent enhancement feature, when considered in terms of T and B cell responses. Another important finding was that the presence of adjuvant was more important in T cell in comparison with B cell response. Vaccinated mice showed T cell response upon restimulation with whole inactivated SARS-CoV-2 or peptide pool. This study shows that the vaccines are effective and leads us to start the challenge test to investigate the gamma-irradiated inactivated vaccine candidates for infective SARS-CoV-2 virus in humanized ACE2 + mice.

10. Antileishmanial Activity of BNIPDaoct- and BNIPDanon-loaded Emulsomes on Leishmania infantum Parasites

Among bisnaphthalimidopropyl (BNIP) derivatives, BNIPDaoct and BNIPDanon recently came forward with antileishmanial activities beyond the standard, commercialized antileishmanial therapies. However, high-level toxicity on macrophages plus poor aqueous solubility and poor bioavailability of the compounds limit their application in therapies. Addressing these limitations, the present study introduces BNIPDaoct- and BNIPDanon-loaded emulsomes as lipid-based nanocarrier systems. Accordingly, emulsome formulations were prepared with the presence of BNIP compounds. The average diameters of BNIPDaoct- and BNIPDanon-loaded emulsomes were found as 363.1 and 337.4 nm, respectively; while empty emulsomes differed with a smaller average particle diameter, i.e., 239.1 nm. All formulations exhibited a negative zeta potential value. The formulations achieved the encapsulation of BNIPDaoct and BNIPDanon at approximately 0.31 mg/ml (501 µM) and 0.24 mg/ml (387 µM), respectively. The delivery of BNIP within the emulsomes improved the antileishmanial activity of the compounds. BNIPDaoct-loaded emulsome with 50% inhibitory concentration (IC50) value of 0.59 ± 0.08 µM was in particular effective against Leishmania infantum promastigotes compared to free BNIPDaoct (0.84 ± 0.09 µM), free BNIPDanon (1.85 ± 0.01 µM), and BNIPDanon-loaded emulsome (1.73 ± 0.02 µM). Indicated by at least ≥ 2-fold higher 50% cytotoxic concentration (CC50) values, the incorporation of BNIP into emulsomes significantly reduced the toxicity of BNIPs against macrophages, corresponding to up to 16-fold improvement in selectivity index (CC50/IC50) for L. infantum promastigotes. The infection rates of macrophages were determined using dual-fluorescent flow cytometry as 68.6%. Both BNIP formulations at concentration of 1.87 µM reduced the parasitic load nearly to 40%, whereas BNIPDaoct-loaded emulosmes could further decrease the parasitic load below 20% at 7.5 µM and above. In conclusion, the incorporation of BNIPDaoct and BNIPDanon into emulsomes results in water-soluble dispersed emulsome formulations that do not only successfully facilitate the delivery of BNIP compounds into the parasites and the Leishmania-infected macrophages in vitro but also enhance antileishmanial efficacy as proven by the decline in IC50 values. The selectivity of the formulation for L. infantum parasites further contributes to the challenging safety profile of the compounds. The promising in vitro antileishmanial efficacy of BNIP-loaded emulsomes highlights the potential of the system for the future in vivo studies.

11. Curcumin- and Piperine-Loaded Emulsomes as Combinational Treatment Approach Enhance the Anticancer Activity of Curcumin on HCT116 Colorectal Cancer Model

Combination chemotherapy, administrating two chemotherapeutic agents concurrently, comes into prominence, as the heterogeneity or the level of the disease necessitates a collaborative action. Curcumin, isolated from turmeric, and piperine, isolated from black long pepper, are two dietary polyphenols studied for their intrinsic anti-cancer properties against various cancer types including colorectal cancer (CRC). Furthermore, piperine improves the therapeutic effect of curcumin. Addressing this mutual behavior, this study combines curcumin and piperine within emulsome nanoformulations. Curcumin- (CurcuEmulsomes) and piperine-loaded emulsomes (PiperineEmulsomes) have established a uniform, stable, spherical dispersion with average diameters of 184.21 and 248.76 nm, respectively. The solid tripalmitin inner core achieved encapsulation capacities of up to 0.10 mg/ml curcumin and 0.09 mg/ml piperine content. While piperine treatment alone – in its both free and emulsome forms – showed no inhibition in the proliferation of HCT116 cells in vitro, its presence as the second drug agent enhanced curcumin’s effect. Combination of 7 μM PiperineEmulsome and 25 μM CurcuEmulsome concentrations was found to be most effective with an inhibition of cell proliferation of about 50% viability. Cell cycle arrest at G2/M phase and induced apoptosis verified the improved anti-cancer characteristics of the therapy. While CurcuEmulsomes achieved a fourfold increase in Caspase 3 level, combination of treatment with PiperineEulsomes achieved a sixfold increase in the level of this apoptotic marker. Combinational treatment of HCT116 cells with CurcuEmulsomes and PiperineEmulsomes improved the anticancer activity of the compounds and highlighted the potential of the approach for further in vivo studies.

New formulation of glycerol and sodium pentaborate may be able to solve the problem. Method. Female Wistar albino rats were randomly assigned into four equal groups. The adhesion model was created on the caecum anterior wall and covered with 2 ml 0.9% NaCl, 3% glycerol, 3% sodium pentaborate, and 3% glycerol plus 3% sodium pentaborate solutions in the groups, respectively. OBJECTIVE: The aim of this study was to compare the cytotoxic effects of endodontic cements on human tooth germ stem cells (hTGSCs). MTA Fillapex, a mineral trioxide aggregate (MTA)-based, salicylate resin containing root canal sealer, was compared with iRoot SP, a bioceramic sealer, and AH Plus Jet, an epoxy resin-based root canal sealer. MATERIAL AND METHODS: To evaluate cytotoxicity, all materials were packed into Teflon rings (4 mmµ3 mm) and co-cultured with hTGSCs with the aid of 24-well Transwell permeable supports, which had a pore size of 0.4 µm. Coverslips were coated with MTA Fillapex, iRoot SP and AH Plus Jet and each coverslip was placed onto the bottom of one well of a six-well plate for scanning electron microscopy (SEM) analysis. Before the cytotoxicity and SEM analysis, all samples were stored at 37ºC and at 95% humidity and 5% CO2 for 24 hours to set. The cellular viability was analyzed using MTS test (3-(4,5-dimethyl-thiazol-2-yl)-5-(3-carboxy-methoxy-phenyl)-2-(4-sulfo-phenyl)-2H-tetrazolium). The cytotoxic effects and SEM visualization of the tested materials were analyzed at 24-hour, 72-hour, one-week and two-week periods. RESULTS: On the 1st day, only MTA Fillapex caused cytotoxicity compared to negative control (NC) group (p0.05). Bean diseases caused by different pathogens reduce important yield and quality of beans in different bean growing regions in both Turkey and the other bean producing countries. In the present study, bacterial pathogens in the species of Pseudomonas savastanoi pv. phaseolicola (Burkholder) Garden et al. and Xanthomonas axonopodis pv. Phaseoli (Smith) Vauterin et al. causing economically important disease on bean plants growing in the commercial fields of Erzurum and Erzincan provinces located in the Eastern Anatolia region of Turkey has been isolated and identified. Totally thirty-six bean genotypes and two commercial cultivars commonly grown in the region have been screened for resistance to these pathogens both in greenhouse and field condition during 2001–2002. Disease severity in the field condition reduced seed quality and quantity of bean. Among the thirty-eight genotypes tested, only 36K was found to be resistant to both of the pathogens.

Objective. The effect of oral boric acid intake on reducing body weight has been previously demonstrated although the mechanism has been unclear. This research study reveals the mechanism. Subjects. Twelve mice were used, in groups of six each in the control and study groups. For five days, control group mice drank standard tap water while during the same time period the study group mice drank tap water which contains 0.28 mg/250 mL boric acid. After a 5-day period, gene expression levels for uncoupling proteins (UCPs) in the white adipose tissue (WAT), brown adipose tissue (BAT), and skeletal muscle tissue (SMT) and total body weight changes were analyzed. Results. Real time PCR analysis revealed no significant change in UCP3 expressions, but UCP2 in WAT (: 0.0317), BAT (: 0.014), and SMT (: 0.0159) and UCP1 in BAT (: 0.026) were overexpressed in the boric acid group. In addition, mice in the boric acid group lost body weight (mean 28.1%) while mice in the control group experienced no weight loss but a slight weight gain (mean 0.09%, ). Conclusion. Oral boric acid intake causes overexpression of thermogenic proteins in the adipose and skeletal muscle tissues. Increasing thermogenesis through UCP protein pathway results in the accelerated lipolysis and body weight loss.

Abstract The primary role of the parathyroid glands is to maintain calcium homeostasis through the secretion of parathyroid hormone (PTH). The limited proliferative capacity and differentiation of parathyroid cells hinder the generation of cell therapy options. In this study, parathyroid organoids are successfully generated from human‐induced pluripotent stem cells (hiPSCs). At the end of the 20 days of differentiation, the parathyroid organoids exhibited distinct parathyroid morphology. Stereomicroscope, scanning electron microscopy (SEM), and transmission electron microscopy (TEM) analysis demonstrated the 3D arrangement of the cell layers in which intracellular structures of parathyroid cells resemble human parathyroid cellular morphology. Comprehensive molecular analyses, including RNA sequencing (RNA‐Seq) and liquid chromatography/mass spectrometry (LC‐MS/MS), confirmed the expression of key parathyroid‐related markers. Protein expression of CasR, CxCr4, Gcm2, and PTH are observed in parathyroid organoids. Parathyroid organoids secrete PTH, demonstrate active intercellular calcium signaling, and induce osteogenic differentiation via their secretome. The tissue integration potential of parathyroid organoids is determined by transplantation into parathyroidectomized rats. The organoid transplanted animals showed significant elevations in PTH‐related markers (CasR, CxCr4, Foxn1, Gcm2, and PTH). PTH secretion is detected in organoid‐transplanted animals. The findings represent a significant advancement in parathyroid organoid culture and may offer a cellular therapy for treating PTH‐related diseases, including hypoparathyroidism.

Purpose: The boron and fluoride mainly accumulate in the bones and teeth of the human body. The purpose of this study is to determine boron or fluoride levels in the whole tooth, to evaluate the correlation between their levels and to compare these levels in primary/permanent, carious, and non-carious groups. Materials and Methods: The boron and fluoride levels of thirty-six teeth, separated such as primary carious (n=9) and non-carious (n=9), permanent carious (n=9) and non-carious (n=9), were determined by ICP-MS and ion-selective electrode, respectively. Results: While boron levels were between 0.001 and 5.88 ppm, the fluoride levels were between 21.24 and 449.22 ppm. The boron level of non-carious teeth was higher than those of carious teeth in primary and permanent tooth groups. However, this difference was not statistically significant (p>0.05). The fluoride level of non-carious teeth was higher than those of carious teeth in primary (p=0.062) and permanent teeth groups (p=0.046). Negative correlation, found between boron and fluoride in all groups, was significant only in non-carious teeth group (r=-0.488, p=0.040). Conclusion: The results of our study proved the importance of fluoride as a protective factor for dental caries once more. The boron levels in non-carious teeth were also higher than carious teeth. However, it was not significant. Moreover, there was negative correlation between teeth boron and fluoride levels. Therefore, it is necessary to conduct more detailed studies on the tooth boron level and its relation with caries formation and with fluoride levels.

Phragmites communis L. is a halophytic plant that can withstand harsh environmental conditions such as salinity. In the present study, we isolated a bacterium associated with the rhizosphere of Phragmites communis L. Based on morphological and biochemical as well as MALDI-TOF and 16S rRNA sequencing results, the strain S6 was identified as Klebsiella aerogenes with a score of 2.34 for MALDI-TOF and a similarity of 99.86% with type strain Klebsiella aerogenes KCTC 2190T. Furthermore, we demonstrated that strain S6 tolerated up to 2.5 M of NaCl and exhibited resistance to only three antibiotics that are frequently used for Enterobacteriaceae. Plant growth-promoting (PGP) results showed that the strain S6 had multiple traits, it was able to fix atmospheric nitrogen, solubilized inorganic phosphate (Ca3(PO4)2 and potassium in addition to the production of siderophores, ammonia, cellulase, protease and amylase. Indole acetic acid (IAA) production was also detected, and the strain S6 showed a positive ACC deaminase activity. Moreover, when the strain S6 was used as an inoculum for pepper seeds, increases in different parameters including shoot and root lengths and fresh and dry weights were shown under normal and saline conditions. We reported in this study, that the inoculation of pepper (Capsicum annuum L.) seeds with the Klebsiella sp. strain S6 was efficient for pepper growth in normal and salt stress conditions. The results of this study thus indicate that natural plants from saline habitats present a good source for isolating beneficial PGPR to grow crops like pepper under saline conditions.

Here we present self-assembled polymeric micelles as potential delivery systems for therapeutic agents with highly tunable properties. The major goal of this study is to design breast and prostate cancer specific targeting peptide modified PEtOx-co-PEI-b-PCL block copolymer based micelles as a targetable carrier system in cancer treatment. For this, a series of micelles based on poly(2-ethyl-2-oxazoline)-co-polyethyleneimine-block-poly(epsilon-caprolactone) [P(EtOx-co-EI)-b-PCL] copolymers with two different proportions of PEI (30% and 60% hydrolysis degrees of PEtOx) were successfully prepared. The block copolymers were synthesized using a combination of living cationic ring-opening polymerization and a copper(i)-catalyzed azide-alkyne cycloaddition (CuAAC) click reaction. Then, peptide 18 and peptide 563 were conjugated to P(EtOx-co-EI)-b-PCL through a thiol-ene click-type reaction to obtain the desired tumor-targeting. The structural properties of the copolymers were confirmed by H-1 NMR, FT-IR, UV-Vis spectrometry and GPC. Peptide and non-peptide-conjugated micelles with particle sizes between 82 +/- 0.6 and 170 +/- 10.7 nm were obtained by self-assembly with two different chain lengths of PEI blocks. The micelles containing the 60% PEI block showed increased zeta potential values. The cytotoxicity of the copolymers was evaluated under in vitro conditions. Overall, our results indicate that the micelles prepared with peptide-conjugated block copolymers can be used as potential nanocarriers for targeted therapeutic delivery systems. Aim To investigate whether circulating T cells including regulatory T cells (Treg) and derived cytokines contribute to the immune imbalance observed in schizophrenia. Methods Forty patients with schizophrenia and 40 age, sex, body mass index, education, and smoking status-matched healthy controls (HC) are included in the study. We stained cells with anti-CD14, anti-CD3, anti-CD4, anti-CD8, anti-CD19, anti-CD20, and anti-CD16/56. Peripheral blood mononuclear cells (PBMCs) were isolated and stained with the human FoxP3 kit containing anti-CD4/anti-CD25 and intracellular anti-Foxp3. PBMCs were cultured for 72 h and stimulated with anti-CD3/anti-CD28. Cytokines (IL-2, IL-4, IL-6, IL-10, IFN-gamma, TNF-alpha, and IL-17A) were measured from the culture supernatant and plasma using the Th1/Th2/Th17 cytokine bead array kit. Results In comparison with HC, Treg percentages in schizophrenia were higher (1.17 +/- 0.63 vs 0.81 +/- 0.53, P = 0.005) in unstimulated but lower in the stimulated condition (0.73 +/- 0.69 vs 0.97 +/- 0.55, P = 0.011). Activated T cell percentages were higher in schizophrenia than HC in unstimulated (2.22 +/- 0.78 vs 1.64 +/- 0.89, P = 0.001) and stimulated (2.25 +/- 1.01 vs 1.72 +/- 1.00, P = 0.010) conditions. The culture supernatant levels of IL-6 (7505.17 +/- 5170.07 vs 1787.81 +/- 1363.32, P < 0.001), IL-17A (191.73 +/- 212.49 vs 46.43 +/- 23.99, P < 0.001), TNF-alpha (1557 +/- 1059.69 vs 426.57 +/- 174.62, P = 0.023), and IFN-gamma (3204.13 +/- 1397.06 vs 447.79 +/- 270.13, P < 0.001); and plasma levels of IL-6 (3.83 +/- 3.41vs 1.89 +/- 1.14, P = 0.003) and IL-17A (1.20 +/- 0.84 vs 0.83 +/- 0.53, P = 0.033) were higher in patients with schizophrenia than HC. Conclusion Our explorative study shows reduced level of Foxp3 expressing Treg in a stimulated condition with induced levels of proinflammatory cytokines in patients with schizophrenia.

In the present study, three entomopathogenic strains of Bacillus sphaericus (MBI5, MBI6, and MBI7) were isolated from greenhouse pest and larval habitat of Istanbul, Turkey. All of the MBI groups tested in laboratory bioassays were able to kill larvae, but not pupae and adults of Culex spp. in water. The most efficient larvicidal activity of the strains was found in the presence of two toxin genes, binA and binB for MBI5, MBI6, and MBI7 strains. Each B. sphaericus strains identified in this study were characterized as unique and novel in terms of fatty acid methyl ester (FAME) profiles and 16S rRNA sequencing data. The results of this study suggest that the three strains of B. sphaericus may be new sources of potential biocontrol agent of mosquito.

Background. Postoperative peritoneal adhesions (PPA) are a serious problem for abdominal surgery. An effective remedy has not been found yet. New formulation of glycerol and sodium pentaborate may be able to solve the problem. Method. Female Wistar albino rats were randomly assigned into four equal groups. The adhesion model was created on the caecum anterior wall and covered with 2 ml 0.9% NaCl, 3% glycerol, 3% sodium pentaborate, and 3% glycerol plus 3% sodium pentaborate solutions in the groups, respectively. Two weeks later, the rats were sacrificed. PPA were graded macroscopically and microscopically. Results. Total adhesion scores of the 3% glycerol + 3% sodium pentaborate group were statistically different from the other groups for macroscopic and also microscopic evaluations (p<0.001). Conclusion. 3% glycerol plus 3% sodium pentaborate as a new formulation has preventive effects on PPA with a synergistic mechanism.

Melatonin, a neuro-differentiation factor, may play a role in the neurodevelopmental origins of schizophrenia. Cognitive impairment and decreased melatonin are reported in schizophrenia; however, the relationship between them remains unclear. We hypothesised that patients with schizophrenia would have lower concentrations of circulating melatonin than healthy controls and that melatonin levels would be associated with cognitive impairment. This study included 47 patients with schizophrenia and 40 healthy controls (HC). Serum melatonin concentrations were measured using the enzyme-linked immunosorbent assay. Positive and Negative Syndrome Scales (PANSS), The Morningness-Eveningness Questionnaire (MEQ), Pittsburgh Sleep Quality Index (PSQI), the Stroop and Oktem verbal memory processes (VMPT) tests were applied. Patients with schizophrenia had lower levels of melatonin compared to the HC group (p = 0.016), also after controlling for age, sex, and body mass index (BMI) (p = 0.024). In patients with schizophrenia, melatonin concentrations were associated with higher BMI (rho = 0.34, p = 0.01) and lower MEQ score (rho = -0.29, p = 0.035). The patient sample was split into low and high melatonin categories by using the median melatonin concentration in HC as the cut-off. Patients in the low melatonin group had poorer performance in VMPT-Recognition (p = 0.026) and Stroop-Colour Error (p = 0.032). Notwithstanding its limitations, the findings of this exploratory study suggest that decreased serum melatonin concentrations observed in schizophrenia might also be associated with cognitive impairment and circadian preferences. Mesenchymal Stem Cells (MSCs) are adult stem cells; isolated from various body parts including bone marrow, adipose tissue and dental tissue, have been characterized well and used in regenerative medicine applications. The promising potential of MSCs makes them great candidates in many disorders. It has been well known in the literature that MSCs interact with cancer cells and regulate the carcinogenesis process at different stages. The dual role of MSCs in cancer progression should be clearly identified at the physiological and molecular level to identify clinical potential in cancer treatment. The promoting or suppressive role of MSCs in cancer is controlled by various growth factors, cytokines and chemokines which affect the cell proliferation, angiogenesis and metastasis. Although many studies have been conducted to explore MSC-cancer cell interactions, it is still unclear how MSCs communicate with cancer cells and tumor microenvironment. Further studies are required to investigate secreted factors and paracrine effects, tumor stroma environment, molecular regulators and downstream pathways that are involved in MSC-cancer interaction loop. MSC type, cancer type and stage specific phenotypic and transcriptomic profile changes should be identified in detail to improve clinical use of MSCs in cancer either as a target or as a tool. In the current book chapter, we review the literature to summarize current information about the MSC-cancer cell interactions in terms of soluble factors, angiogenesis, metastasis and drug resistance. The aim of this study was to determine efficiency of a new molecule that was obtained by linking boric acid with ampicillin in treating intra-abdominal infection.Following intraperitoneal E. coli injection totwenty-one female Wistar albino rats, group 1 was administered boron-linked ampicillin, group 2 was administered only ampicillin and group 3 was injected intraperitoneally with physiological serum. IL-6, and a white blood cell analysis was performed from the blood before and on the seventh day of treatment.No statistically significant difference in blood WBC levels after treatment was found among the groups. There was no statistically significant difference in the IL-6 values of group 2 and group 3 before and after the treatment (p=0.195 and 0.193, respectively); however, the reduction in the serum IL-6 values of group 1 was statistically significant (p=0.003).Boric acid-linked ampicillin is a more effective intra-abdominal infection treatment compared with ampicillin alone. © 2019 Pakistan Journal of Pharmaceutical Sciences. Novel thiourea (5a, 5b) and thiazolidinone derivatives (6a, 6b) were synthesized by hybridizing molecules starting from the compound 6-(4-phenylpiperazin-1-yl)pyridin-3-amine (4) which is known to show anticancer activity. The synthesis of the leading compound was carried out by using 1-(5-nitropyridin-2-yl)-4-phenylpiperazine (3) which was obtained by a novel method of the reaction of 2-chloro-5-nitropyridine (1) and N-phenylpiperazine (2). The structures of the compounds were confirmed using FTIR, 1H NMR, 13C NMR, HRMS spectroscopic methods and elemental analysis. The organic molecules were tested for their anticancer activities against prostate cancer (PC) cell lines: DU 145, PC-3 and LNCaP. As the compound 5a exerted the highest cytotoxic activity, IC50 concentrations of compound 5a were further investigated in terms of morphology, colony-forming ability, RNA expression, fragmented DNA and cell cycle distributions of PC cell lines. Food packaging technology has been advancing to provide safe and high quality food products and to minimize food waste. Moreover, there is a dire need to replace plastic materials in order to reduce environmental pollution. The aim of this study was to prepare biodegradable antimicrobial packaging films from gelatin. Boric acid, disodium octaborate tetrahydrate, and sodium pentaborate were incorporated as the antimicrobial agents. Films containing boric acid and its salts showed antibacterial effect against Staphylococcus aureus and Pseudomonas aeruginosa, as well as antifungal and anticandidal effects against Aspergillus niger and Candida albicans. The mechanical strength of the films was mostly enhanced by the addition of boron derivatives. The rheological measurements and Fourier-transform infrared spectroscopy results suggest that boron derivatives did not interfere with the network formation during gelling. The morphology of boron-added antimicrobial films was found to be similar to the morphology of the control. In conclusion, the newly developed gelatin films containing 10% or 15% disodium octaborate (g/g gelatin) might be good candidates for biodegradable antimicrobial packaging materials. © TÜBİTAK.

Background: The topical application of boron has been significantly associated with intensifying wound healing. Using 3% boric acid in deep wounds significantly contributes to wound healing and reduces the duration of hospitalization in the intensive care. The objective of this study was to assess the therapeutic impact of a topical gel containing sodium pentaborate pentahydrate on the management of wounds resulting from grade 1 to 3 hemorrhoids. Methods: In this randomized double-blind placebo-controlled trial, we applied a topical gel consisting of sodium pentaborate pentahydrate 3% on 206 eligible patients with the diagnosis of grade 1, 2, and 3 hemorrhoid diseases. Then patients were randomly allocated to two groups of sodium pentaborate pentahydrate or placebo gel with a ratio of 1:1 and received the allocated gel for four weeks. Patient hemorrhoid symptoms severity, hemorrhoid degree, and anoscopy findings were compared before and after the trial. Results: Before the intervention, symptom severity (p > 0.05) and anoscopy findings (p = 0.815) were similar between the two groups. Subsequent to the intervention, a majority of patients in the intervention group experienced a reduction in anal itching compared to the placebo group [adjusted mean difference (aMD) 95% CI: -1.98 (−2.2 to −1.8), p = 0.007]. Moreover, resting pain [aMD (95% CI): -1.37 (−1.6 to −1.1), p = 0.015], pain during defecation [aMD (95% CI): -2.19 (−2.4 to −2.0), p = 0.005], feeling a lump in the anus (aMD (95% CI): -0.71 (−1.2 to −0.2), p = 0.011), bleeding during defecation (41.7% vs. 66.9%, p = 0.027), and hemorrhoid degree (p

Tumors consists of subpopulation of cells in which each subtype has contributes to tumor progression. Specifically one subtype known as cancer stem cells are associated with the initiation, progression, resistance to conventional therapies and metastasis. Metastasis is leading cause of cancer related deaths. Overall it is important to consider cancer as a whole in which a mutated cell proliferating indefinitely and forming its hierarchy consisting of subgroups with different molecular signatures. To be able to target this disease we need to evaluate every step including initiation, progression, survival, angiogenesis and finally migration and repopulation. Cancer stem cells do play vital roles in each step however when metastasis can be stopped or eliminated we talk about saving a life or improving its quality. Considering how deeply these cancer stem like cells affect the tumor life and metastasis it is crucial to develop effective strategies against them. Metastatic cascade can also be directed by membrane derived vesicles specifically exosomes. Several studies show the role of exosomes in mediating cellular migration and pre-metastatic niche formation. During this chapter we wanted to explain in detail how the metastasis occur in tumor and how cancer stem cells contribute into the development of metastatic cascade and possibly suggest therapeutic approaches against cancer stem cells. © Springer Nature Switzerland AG 2018.

Background The oral cavity can be an extra-gastric reservoir for Helicobacter pylori (H.pylori). This can play a role in the pathogenesis of halitosis, glossitis, recurrent aphthous stomatitis, and dental caries. The present study was conducted to detect the presence of H.pylori within the dental biofilm and in saliva samples collected from children suffering from dyspepsia and children without any gastrointestinal complaints. Associations with gastric infection, halitosis, and some oral parameters were also evaluated. Methods Seventy children (aged between 5–16) with dyspepsia were selected for the study group and control group composed of 30 healthy children without dyspepsia were also included in the study. After detailed oral and clinical examinations for oral parameters, saliva, and supragingival dental biofilm samples were collected for 16S rRNA and 23S rRNA genes detection by real-time polymerase chain reaction (RT-PCR). The presence of gastric H.pylori was evaluated in endoscopic biopsy specimens histopathologically. Halitosis was evaluated by benzoyl-DL-arginine-naphthylamid (BANA) test. Salivary S.mutans and Lactobacilli sp. counts were also carried out by commercial kits. Results H.pylori was histopathologically detected amongst 83% of the children with the dyspeptic condition. The detection rate of this bacteria in dental biofilm and saliva samples and halitosis were found relatively higher in the dyspeptic children rather than the control group (p 0.05). In the gastric H.pylori positive group with dyspepsia, DMFT/S and dmft/s numbers and plaque indices were found higher than the control group (p 0.05). Comparing to those with negative for both genes, in children whose dental biofilm and saliva samples were positive for both 16S rRNA and 23S rRNA genes, significantly higher results for halitosis, and DMFS numbers and significantly lower results for dmfs numbers and pH values were found (p

WOS: 000408296600003 PubMed ID: 28669743 Conflicting data in the literature about the function of P2X7R in survival following ischemia necessitates the conductance of in-depth studies. To investigate the impacts of activation vs inhibition of the receptor on neuronal survival as well as the downstream signaling cascades, in addition to optic nerve transection (ONT), 30 min and 90 min of middle cerebral artery occlusion (MCAo) models were performed in mice. Intracellular calcium levels were assessed in primary cortical neuron cultures. Here, we show that P2X7R antagonist Brilliant Blue G (BBG) decreased DNA fragmentation, infarct volume, brain swelling, neurological deficit scores and activation of microglial cells after focal cerebral ischemia. BBG also significantly increased the number of surviving retinal ganglion cells (RGCs) after ONT and the number of surviving neurons following MCAo. Importantly, receptor agonist BzATP resulted in increased activation of microglial cells and induced phosphorylation of ERK,AKT and JNK. These results indicated that inhibition of P2X7R with BBG promoted neuronal survival, not through the activation of survival kinase pathways, but possibly by improved intracellular Ca2+ overload and decreased the levels of Caspase 1, IL-1 beta and Bax proteins. On the other hand, BzATP-mediated increased number of activated microglia and increased survival kinase levels in addition to increased caspase-1 and IL-1 beta levels indicate the complex nature of the P2X7 receptor-mediated signaling in neuronal injury. In conjunction with an increasing public awareness of infectious diseases, the textile industry and scientists are developing hygienic fabrics by the addition of various antimicrobial and antiviral compounds. In the current study, sodium pentaborate pentahydrate and triclosan are applied to cotton fabrics in order to gain antimicrobial and antiviral properties for the first time. The antimicrobial activity of textiles treated with 3 % sodium pentaborate pentahydrate, 0.03 % triclosan, and 7 % Glucapon has been investigated against a broad range of microorganisms including bacteria, yeast, and fungi. Moreover, modified cotton fabrics were tested against adenovirus type 5 and poliovirus type 1. According to the test results, the modified textile goods attained very good antimicrobial and antiviral properties. Thus, the results of the present study clearly suggest that sodium pentaborate pentahydrate and triclosan solution-treated textiles can be considered in the development of antimicrobial and antiviral textile finishes. © 2016, Springer Science+Business Media New York.

Invasive fungal infections are one of the major challenges especially for immunosuppressed patients since they are drug resistant and pathogen to patients. Therefore, developing new, efficient and nonresistant antifungal agents have been a primary focus of international research. In the current study, a novel Schiff base [hetero-dinuclear copper(II) Mn(II) complex] (SB) derivative was investigated for its anticandidal activity against Candida albicans and possible mechanisms inducing cell death. The results revealed that SB treatment induces apoptotic and necrotic pathways in C. albicans ATCC10231 strain. Intracellular reactive oxygen species production determined by 2',7'-dichlorofluorescein diacetate staining was triggered by SB and amphotericin B administrations in a dose-dependent manner. Gene expression analysis demonstrated that SB exposure resulted in regulation of critical development and stress related gene expressions. SB treatment directly upregulated expression of stress related genes, DDR48 and RIM101, while suppressed important cell signaling and antibiotic resistance acquiring related genes such as HSP90, ERG11 and EFG1. Furthermore, CaMCA1 mRNA levels were found to be significantly high in SB-treated yeast cells, indicating possible caspase-like mechanism activation. Scanning electron microscopy analysis confirmed that SB treatment led to severe cell wall integrity disruption and wrinkling. The study will encourage development of SB-based anticandidal regimens but further studies are highly warranted to understand limitations and the extended use in the routine. © 2017, Springer Science+Business Media B.V.

Crop production in salty-sodic soils causes poor crop growth due to ion imbalance, water stress, low hormonal and enzymatic balances. The use of rhizobacteria promoting plant growth in saline and sanitary areas (PGPR) is an economical and environmentally friendly approach that increases and eliminates the tolerance of plants to salt. Field experiments were conducted to determine the response of barley, maize and sorghum crop yields to the application of the bacteria (Halobacillus sp), which was preliminarily experimented by a greenhouse study, on two big soil groups (Haplustept and Fluvaquent). The experiment was established with four different treatments; T0 (control-no application), T1 (the gypsum application), T2 (the PGPR application), and T3 (the gypsum + PGPR application) on the high saline and sodic soils in a semi-arid region. The observations of the present study indicate positive impacts of PGPR applications on crop yield and soil productivity. The gypsum + PGPR treatment was the most effective treatment obtaining higher yields owing to significant improvement in the plant photosynthetic activity, chlorophyll content, stomatal conductance, enzymes, hormones, amino and organic acids, and crop-soil productivity. The seed inoculation with the Halobacillus sp bacteria cultures help in alleviation of stress and enhance crop productivity under the short-term gypsum application on Haplustept and Fluvaquent saline-sodic soils. Objective: Complementary and alternative medicine (CAM) has an increasing usage in the last decades throughout the world. Herbal medicine, the most preferred CAM, is used in the treatment of various disorders by approximately eighty percent of the world's population. In this study, it was aimed to determine the antiviral activity and wound healing potential of a special herbal oil blend prepared from Olea europaea, Nigella sativa and Rosmarinus officinalis that has been used for centuries in Middle and East Anatolia. Materials and Methods: The nontoxic concentration of herbal blend was determined by MTS assay. This concentration was investigated for its wound healing potential using in vitro scratch assay. A scratch was made on cultured keratinocyte cell layer and the herbal blend was added to medium. Pictures of cells were taken at different time points. The antiviral activity was determined using Adenovirus type 5 and Poliovirus type 1. Virus titer was calculated by Spearman-Karber method. Results: The nontoxic concentration of the blend was found to show statistically higher wound healing rate in compare to control group at the end of 12, 24 and 36 hours. According to antiviral efficacy test, four log of reduction in virus titer was seen, which meant that the mixture was quite effective against the viruses used in the study. Conclusion: The study shows that the special herbal blend speed up wound healing rate and it also has antiviral activity.

Ex vivo dermal toxicology analyses are crucial for replacement of in vivo test methods and have been of interest in recent years for testing cosmetics, drugs, and chemicals. Development of an appropriate reconstructed epidermis model might overcome the limitations of monolayer culture systems. In the current study, we used the immortalized human keratinocyte cell line (HaCaT) to develop an ex vivo 3D cell culture system for keratinocyte-based toxicity analysis. Mouse embryonic fibroblast-conditioned medium and Matrigel matrix-based 3D HaCaT cell culture systems expressed skin-related genes and proteins in culture. The 3D HaCaT cultures demonstrated a skin-like phenotype and response against selected test compounds. Reliable results were obtained compared to monolayer HaCaT cells which were exposed to selected chemicals for 1 h and 24 h. Gene expression profiles of 3D HaCaT cell cultures and monolayer cultures were completely different after administration of the test compounds.Overall, our results showed that a 3D HaCaT cell culture system generated in Matrigel matrix exerted a skin epidermis-like phenotype. Consequently, 3D HaCaT cell cultures may be an acceptable test method for conducting in vitro toxicology experiments.

Here we present the integration of boron (NaB) with graphene oxide (GO) to develop a new class of membranes which are biocompatible and cost-effective for cell and tissue culture studies. Ethanol (EtOH) assisted the uniform dispersion of GO flakes on top of a glass substrate. We investigated the effect of a GO + NaB membrane on the growth and proliferation of hASCs. hASCs showed better cell viability on NaB integrated GO membranes compared to their respective controls. The concentrations of NaB and GO are 0.02% and 1/20 of stock (0.024%) respectively. To our knowledge this is the first time that enhanced cell proliferation and attachment ability of hASCs with NaB integrated GO membranes has been observed. Our study provides a platform for the development of 3D-GO scaffold systems combined with NaB in tissue engineering. © 2016 The Royal Society of Chemistry.

Background: Boric acid is widely used in biology, but its body weight reducing effect is not researched.Methods: Twenty mice were divided into two equal groups. Control group mice drank standard tap water, but study group mice drank 0.28mg/250ml boric acid added tap water over five days. Total body weight changes, major organ histopathology, blood biochemistry, urine and feces analyses were compared.Results: Study group mice lost body weight mean 28.1% but in control group no weight loss and also weight gained mean 0.09% (pConclusion: Low dose oral boric acid intake cause serious body weight reduction. Blood and urine analyses support high glucose, lipid and middle protein catabolisms, but the mechanism is unclear.

Currently, use of cannabinoids is limited to improve adverse effects of chemotherapy and their palliative administration during treatment is curiously concomitant with improved prognosis and regressed progression in patients with different tumor types. Although, non-psychoactive cannabidiol (CBD) and cannabigerol (CBG) display antineoplastic effects by repressing tumor growth and angiogenesis both in cell line and animal models, their use as chemotherapeutic agents is awaiting further investigation. Both clinical and epidemiological evidence supported by experimental findings suggest that micronutrients such as curcumin and piperine may present a safer strategy in preventing tumorigenesis and its recurrence. Recent studies demonstrated that piperine potentiates curcumin’s inhibitory effect on tumor progression via enhancing its delivery and therapeutic activity. In this study, we investigated a plausible therapeutic synergism of a triple combination of CBD/CBG, curcumin, and piperine in the colon adenocarcinoma using HCT116 and HT29 cell lines. Potential synergistic effects of various combinations including these compounds were tested by measuring cancer cell proliferation and apoptosis. Our findings revealed that different genetic backgrounds of HCT116 and HT29 cell lines resulted in divergent responses to the combination treatments. Triple treatment showed synergism in terms of exhibiting anti-tumorigenic effects by activating the Hippo YAP signaling pathway in the HCT116 cell line.

A two-year irrigated field study was conducted to determine the effects of plant growth-promoting rhizobacteria (PGPR; Bacillus subtilis OSU-142 and Bacillus megaterium M3) as biofertilizer, and in combination with a chemical nitrogen (N) fertilizer, on turf color and clipping yield, and interaction of biofertilizer and chemical N fertilizers in perennial ryegrass (Lolium perenne L.), tall fescue (Festuca arundinacea L. Schreb.), and Kentucky bluegrass (Poa pratensis L.). The three turf species were tested separately in split-plot design experiments with three replications. Three fertilizer sources (ammonium nitrate only, ammonium nitrate + B. megaterium M3, and ammonium nitrate + B. subtilis OSU-142) were the main plots. N applications with monthly applications of 0.0, 2.5, 5.0, and 7.5g N/m(2) were the subplots. Color ratings and clipping yields increased with increasing chemical N fertilizers in all species. Both Bacillus sp. significantly increased color ratings and clipping yields in perennial ryegrass and tall fescue. However, there were no significant differences among the three fertilizer sources in color and clipping yield of Kentucky bluegrass. The experiments showed that there is a small but significant benefit from applying biofertilizers for turf color, and that N fertilization may be reduced in some turf species when biofertilization are made for this purpose.

WOS: 000364895700004 The usage of magnesite strictly depends on its impurities. CaCO3 is the major impurity in magnesite ores. The amount of CaCO3 should be less than 1% for the quality and economical use of magnesite ores. Some magnesite ores in Turkey cannot be used due to high amount of CaCO3 (>= 3%). This study aims to investigate an alternative biotechnological method for decalcification of magnesite ores by using a microorganism (Lactococcus spp.). The amount of CaCO3 was reduced from 2.94% to 0.57%. This reduction improved the quality of magnesite ore by approximately 80.61%. The results showed that Lactococcus spp. can be used safely in industrial decalcification of magnesite ore as the microorganism has no pathogenic properties. (C) 2015 Published by Elsevier B.V.

The essential oils of S. hortensis and C. nepeta were isolated by the hydrodistillation method and the chemical composition of the essential oils were analyzed by GC-MS. The antibacterial properties of the essential oils were evaluated against 20 phytopathogenic bacteria through Disc diffusion assay and micro dilution assay. The results revealed that the essential oils of S. hortensis and C. nepeta have significant antibacterial activity. Furthermore, the findings of the study are valuable for future investigations focusing on the alternative natural compounds to control plant diseases.

To compare the cellular viability of diced, crushed, and morselized cartilage used in nasal surgeries. In this study, cartilage was extracted from the ears of seven New Zealand rabbits and was subsequently either diced, crushed or morselized to an amorphous state, or left unmodified. The four types of grafts were then implanted in the back regions of the rabbits. After 3 months, the cellular viability from four groups was compared to a control group using confocal microscopy. Analysis of the data obtained from the enumeration of live cells showed no statistically significant difference between the unmodified graft group and the control group. The diced, crushed, and morselized cartilage groups did show a statistically significant difference in terms of live cell count with the highest number of live cells in diced cartilage group. A statistically significant decrease in live cell count was detected in crushed cartilage group. Our study shows that the viability of cells in diced cartilage grafts is greater than those in crushed or morselized cartilage grafts. © 2014, Springer-Verlag Berlin Heidelberg. Sakarya Üniversitesi

Obesity is a popular topic due to both its mortality and morbidity rates and related diseases such as cardiovascular diseases, diabetes and cancer. Cancer development and progression relate to many factors one of which is dysfunctional adipocytes found in the tumor microenvironment. However, underlying molecular mechanisms of the obesity-cancer link have not been fully understood. In the current study, condition media (CM) obtained from differentiated pre-adipocytes was used to set an indirect co-culture system with the prostate cancer cell line. Firstly, adipogenesis of 3T3F44-2A was checked by oil red o staining and expressions of specific genes. CM of differentiated 3T3F44-2A was applied on prostate cancer cells (PC3). Cell viability, migration capacity and related gene expression levels of the cells were evaluated. 20% CM increased cell viability after 48h. The administration also induced proliferation, migration and epithelial-mesenchymal transition (EMT)-related gene expressions. The results presented the roles of adipocytes found in the tumor microenvironment and this could allow new therapeutic developments. As a new perspective to evaluate the obesity-cancer link, our model experiment may also be useful for other cancer types.
