Purpose: The aim of the study explores probable toxic effects of vancomycin on kidney and analysis of the probable protective effects of melatonin.Materials and Methods: In this study, rats were randomly divided into 4 groups: the control group; the melatonin (10 mg/kg/day) group; the vancomycin-treated (200 mg/kg) group; and the vancomycin (200 mg/kg) + melatonin (10 mg/kg/day) group. Rats in the treatment group were given two doses of vancomycin a day with an interval of seven consecutive days and melatonin (10 mg/kg/day) once daily for seven consecutive days. The experiment was continued for 15 days. In each group, seven rats were grouped together. 15 days after the experiment, the rats were sacrificed under anesthesia and among all groups. Kidney tissues were collected and processed for further TNF- expression analysis, as well as histological analyses such as hematoxylin and eosin (H&E), Masson's tricrom, and Periodic acid schiff (PAS) staining to assess pathological severity. In addition, a terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay was performed to evaluate apoptosis.Results: While vancomycin upregulated TNF-α expression, melatonin reduced levels of TNF-α immunoreactivity intensity and clearly improved pathological severity in rat kidneys. Further, melatonin significantly inhibited vancomycin-induced TUNEL-positive cell numbers. Conclusion: Melatonin has protective activity against vancomycin-induced pro-inflammatory and proapoptotic effects in kidneys during organ preservation time and improves kidney function., Amaç: Bu çalışmanın amacı, vankomisinin böbrek üzerindeki olası toksik etkilerinin belirlenmesi ve melatoninin olası koruyucu etkilerini araştırmaktır.Gereç ve Yöntem: Bu çalışmada, sıçanlar rastgele 4 gruba ayrıldı: kontrol grubu, melatonin (10 mg/kg/gün) grubu, vankomisin uygulanan (200 mg/kg) grup ve vankomisin (200 mg/kg) +melatonin (10 mg/kg/gün) grubu. Tedavi grubundaki sıçanlara art arda yedi gün boyunca günde iki kez vankomisin ve ardından yedi gün boyunca günde bir kez melatonin (10 mg/kg/gün) verildi. Her grupta yedi sıçan yer aldı ve deneye 15 gün devam edildi. Deneyden 15 gün sonra, sıçanlar anestezi altında sakrifiye edildi. Sıçanlara ait böbrek dokuları alındı ve patolojik şiddeti değerlendirmek TNF-α ekspresyon analizi, hematoksilin ve eozin (H&E), Masson's tricrom ve Periodic acid schiff (PAS) gibi histolojik analizler yapıldı. Ek olarak, apoptozu değerlendirmek için Terminal deoksinükleotidil transferaz dUTP nick-end etiketlemesi (TUNEL) yöntemi uygulandı.Bulgular: Vankomisin, TNF-α ekspresyonunu yükseltirken; melatonin, TNF-α immünoreaktivite yoğunluğunu azalttı ve sıçanların böbreklerinde açıkça patolojik şiddeti iyileştirdi. Ayrıca melatonin, vankomisinin neden olduğu TUNEL pozitif hücre sayılarını önemli ölçüde inhibe etti.Sonuç: Bulgularımız, melatoninin organ koruma süresi boyunca böbreklerde vankomisinin neden olduğu proinflamatuar ve proapoptotik etkilere karşı koruyucu ve böbrek fonksiyonlarını iyileştirici etkiye sahip olduğunu gösterdi.

Amaç: Bu çalışmanın amacı, diyabetik akciğer fibrozisi üzerine histopatolojik ve immünohistokimyasal olarak STZ (Streptozotosin) kaynaklı diyabet modeli üzerindeki etki ve potansiyel mekanizmaları araştırmaktır.Gereç ve Yöntem: Bu çalışmada, 14 yetişkin dişi Wistar albino sıçan, 7 rastgele hayvandan oluşan gruplara ayrıldı: kontrol ve STZ ile indüklenen diyabet grupları. Çalışmada kan şekerinin 200 mg/dl'nin üzerinde olması diyabet olarak kabul edildi. Deneyden dokuz gün sonra sıçanlar anestezi altında sakrifiye edildi ve her birinin akciğer örnekleri alındı. Örneklerin histopatolojik görünümleri değerlendirildi ve histopatolojik hasar skoru yapıldı. Dokulardaki apoptoz ve inflamasyon, immünohistokimyasal olarak kaspaz-3 ve IL-1β ile değerlendirildi.Bulgular: Histopatolojik incelemede STZ grubunun histopatolojik hasar skoru kontrol grubuna göre daha yüksekti ve damar tıkanıklığı, alveol duvarında kalınlaşma, inflamatuar hücre infiltrasyonu gibi bulgular vardı. Kaspaz-3 immünohistokimyasında, STZ grubunda akciğer dokularının boyanması kontrol grubuna göre daha yüksekti. Apoptosis and inflammation in tissues were evaluated with caspase-3 and IL-1β immunohistochemically.Results: In the histopathological examination, the STZ group had a higher histopathologic damage score than the control group, and there were findings such as vascular congestion, thickened alveolar wall, and inflammatory cell infiltration. In the caspase-3 immunohistochemistry, staining of the lung tissues of the STZ group was higher than the control group. This difference was also significant in terms of IL-1β immunoreactivity intensity. Conclusion: This study determined that lung complications and damage due to diabetes-induced by STZ occurred.

7. Protective effects of melatonin on female rat ovary treated with nonylphenol.

We investigated using histochemistry and immunohistochemistry ovarian damage caused by nonylphenol (NP) and the protective effect of melatonin treatment of NP induced ovarian damage. We used 21 female rats divided randomly into three groups: control, NP and melatonin + NP. Histopathological examination of the ovaries, and counting and classification of follicles were performed using Masson's trichrome staining. Expression of anti-Mullerian hormone (AMH), Bax, Bcl-2 and caspase-3 was detected in the ovaries using immunohistochemistry. Melatonin had an ameliorative effect on NP induced follicular atresia and absence of corpora lutea. More follicles were observed in the ovaries of animals treated with melatonin prior to treatment with NP. AMH immunoreactivity was significantly lower in the NP group than in the melatonin + NP group. NP increased immunostaining for Bax, Bcl-2 and caspase-3. Melatonin significantly reduced the increased expression of Bax, Bcl-2 and caspase-3 due to NP exposure. We found that pretreatment with melatonin is beneficial for protecting the ovaries from damage by NP. [ABSTRACT FROM AUTHOR]

Aim: Vancomycin is a commonly used antibiotic with potent activity against Gram-positive organisms, but prolonged use and high doses can lead to toxicity. While vancomycin-associated nephrotoxicity is widely reported, few cases of neurotoxicity have been described. Presupplementation with melatonin, a powerful antioxidant for nervous tissues, protects the sciatic nerve against all of the changes. Materials and Methods: Melatonin, a neurosecretory product of the pineal gland, functions as an antioxidant in vivo and in vitro. Therefore, in this study the effect of melatonin in rats treated with vancomycin on the sciatic nerve was investigated. 28 wistar albino female rats were divided into four groups, each containing 7 rats. The first group was used as a control. The second group, melatonin (10 mg/kg/day) was injected intraperitoneally into rats for 7 days, respectively. The rats in the third group were injected with vancomycin (200 mg/kg) for 7 days, respectively. The fourth group, vancomycin (200 mg/kg)+melatonin (10 mg/kg/day) were received vancomycin for 7 days and than melatonin into rats for 7 days, respectively. The experiment was continued for 15 days. The sciatic nerve tissues were examined under light microscopes. Results: According to our findings, S100 expression was lowest in the vancomycin-treated group. In addition, the immunoreactivity intensity of S100 was significantly increased in the vancomycin+melatonin group, compared to the vancomycin group (p < 0.01). However, the immunoreactivity intensity of UCHL in the vancomycin group was no statistically significantly than the immunoreactivity intensity of UCHL in the other groups (p > 0.05). Conclusion: In this study, it was found that melatonin treatment effects positively regeneration of sciatic nerve injury caused by vancomycin in the sciatic nerve of rats. [ABSTRACT FROM AUTHOR]

- Abstract
Objective: Although autogenous grafting is accepted as the gold standard in intraoral grafting, xenogenous grafts are frequently used in sinus lift surgeries due to their osteoinductive and osteoconductive properties. This study aimed to investigate the efficacy of fish spine-derived xenogenic grafts in sinus augmentation surgery., Material and Methods: In this study, a fish spine-derived xenogenic graft was produced for comparison with autogenous graft and bovine derived xenogenic grafts. Twenty-one New Zealand rabbits were used. Autogenous grafts (AG- Group 1), as well as bovine-derived (bHAP - Group 2) and fish spine-derived (fHAP - Group 3) xenogenic grafts were placed in the right and left sinuses of rabbits. The animals were sacrificed at the 4th and 8th weeks. New bone formation (NBF) was evaluated through histological examination, while bone volume (BV), new bone surface/bone volume (BS-BV), new bone surface/tissue volume (BS-TV), and trabecular separation (Tb-Sp) were assessed via Micro-CT. Statistical significance was considered at p<0.05., Results: Histological examination revealed a significant difference in NBF between AG-bHAP (p<0.001), as well as between fHAP-bHAP (p<0.001) in the fourth-week group. No significant difference was found in the eighth-week group (p=0.130). In the eighth-week group, a statistically significant difference was found between fHAP and bHAP in terms of BV. (p=0.007)., Conclusion: Although both graft materials used in this study showed positive effects on bone regeneration, fHAP and AG presented similar effects on bone regeneration and were superior to bHAP.

15. The effect of oral hydroxychloroquine on chondral defect: An experimental study.

Objectives: This study aims to evaluate the time- and dose-dependent effects of oral hydroxychloroquine (HCQ) on focal full-thickness knee chondral defect healing in a rabbit model., Materials and Methods: Cartilage defects of 4x4 mm
2 were created on both medial femoral condyles of 24 New Zealand rabbits. The rabbits were divided into six groups (A-F) according to HCQ administration and sacrifice time: A (three-week control) and B (six-week control) received no additional interventions; C (20 mg/kg HCQ, three weeks); D (20 mg/kg HCQ, six weeks); E (40 mg/kg HCQ, three weeks); and F (40 mg/kg HCQ, six weeks). Osteochondral specimens were evaluated macroscopically, histologically, and immunohistochemically. The terminal deoxynucleotidyl-transferase (TdT)-mediated dUTP nick end labeling (TUNEL) method was used to detect apoptotic cells., Results: The International Cartilage Repair Society (ICRS) scores were significantly higher in the experimental groups than in the controls (p<0.001). The Wakitani scores in Group D showed a significant improvement compared to those in Group B (p<0.01). The 20 mg/kg HCQ treatment groups showed better recovery than the controls (p<0.01). High-dose HCQ (40 mg/kg) treatment significantly reduced the intensity of collagen type 2 immunoreactivity compared to that in the groups receiving 20 mg/kg of HCQ (p<0.01). Collagen type 2 expression in Group F was significantly lower than that in Group D (p<0.01). There were more TUNEL-positive cells in the repair sites of Groups E and F than in the lower-dose experimental groups and untreated experimental groups (p<0.001)., Conclusion: A low dose of HCQ improved cartilage repair, while higher doses of HCQ exerted a negative effect on cartilage regeneration in rabbits. In the presence of defective cartilage, the use of HCQ at an appropriate dose and time is important for cartilage health.- Published
