Abstract Protein aggregation plays key roles in age-related degenerative diseases, but how different proteins coalesce to form inclusions that vary in composition, morphology, molecular dynamics and confer physiological consequences is poorly understood. Here we employ a general reporter based on mutant Hsp104 to identify proteins forming aggregates in human cells under common proteotoxic stress. We identify over 300 proteins that form different inclusions containing subsets of aggregating proteins. In particular, TDP43, implicated in Amyotrophic Lateral Sclerosis (ALS), partitions dynamically between two distinct types of aggregates: stress granule and a previously unknown non-dynamic (solid-like) inclusion at the ER exit sites (ERES). TDP43-ERES co-aggregation is induced by diverse proteotoxic stresses and observed in the motor neurons of ALS patients. Such aggregation causes retention of secretory cargos at ERES and therefore delays ER-to-Golgi transport, providing a link between TDP43 aggregation and compromised cellular function in ALS patients.

Abstract The main purpose of this paper is to introduce a new general-type proximal point algorithm for finding a common element of the set of solutions of monotone inclusion problem, the set of minimizers of a convex function, and the set of solutions of fixed point problem with composite operators: the composition of quasi-nonexpansive and firmly nonexpansive mappings in real Hilbert spaces. We prove that the sequence x n which is generated by the proposed iterative algorithm converges strongly to a common element of the three sets above without commuting assumption on the mappings. Finally, applications of our theorems to find a common solution of some nonlinear problems, namely, composite minimization problems, convex optimization problems, and fixed point problems, are given to validate our new findings.

Abstract Background The association of macrophage activation syndrome and primary Sjögren’s syndrome has been rarely reported in the literature. We report the first observation of this association in Africa, south of the Sahara, and we discuss the diagnosis and therapeutic challenge. Case presentation A 26-year-old Mauritanian and Berber woman was followed for primary Sjögren’s syndrome. After a voluntary cessation of her usual background treatment, she was admitted to our department for an outbreak of her illness. A clinical examination revealed anemic syndrome, peripheral polyarthritis, coughing rales at both pulmonary bases, and fever at 39.5 °C. On biologic examination, there was bicytopenia with anemia at 5.70 g/dl, lymphopenia at 796/mm3, a biological inflammatory syndrome with a sedimentation rate at 137 mm in the first hour, C-reactive protein at 97 mg/l, hyperferritinemia at 1778 mg/l (9 normal value), and hypergammaglobulinemia at 20.7 g/l of polyclonal appearance. The triglycerides were 1.95 g/l (1.4 normal value) and the lactate dehydrogenase level was 491 IU/l (1.5 normal value). Cytological examination of a medullary puncture revealed an image of hemophagocytosis. An infectious screening was negative. Thoracic computed tomography showed non-specific interstitial lung disease. A diagnosis of macrophage activation syndrome complicating primary Sjögren’s syndrome was selected with a probability of 97.2%, according to H-Score. The evolution was favorable under a treatment including etoposide (VP-16). Conclusion Macrophage activation syndrome is a rare entity, rarely reported during primary Sjögren’s syndrome. Its spontaneous evolution is invariably fatal. There is no consensus on therapeutic treatment. Etoposide is a therapeutic option especially in forms refractory to corticosteroid therapy.

The adhesion of tungsten (W) dust particles on W tokamak plasma-facing surfaces is investigated. When spherical dust hits the tokamak walls, its shape is altered as a contact area is created by the adhesion work and irreversible plastic deformation. The van der Waals adhesion force between such a deformed grain and a flat surface is estimated, and the link between the force and the impact velocity is determined. We show that adhesion can be increased by several orders of magnitude when the impact velocity approaches the adhesion velocity. Finally, the effects of surface roughness are incorporated.

The ability of tungsten (W) dust to be remobilized from rough tokamak plasma-facing surfaces is investigated in this study. Atomic Force Microscopy is used to evaluate the adhesion force distribution between W spheroids of ∼1−10 µm radius and a rough W substrate. Our results confirm that the Rabinovich model describes reasonably well the mean adhesion force in tokamak-relevant surface roughness regimes. The external (electric and ion drag) forces are estimated as well using a simplified sheath model. The results reveal that micron-size dust can be resuspended by attainable electric fields in tokamak conditions.

BackgroundIncreasingly studies show that immigrants have different perinatal health outcomes compared to native-born women. Nevertheless, we lack a detailed examination of the combined effects of maternal immigrant trajectory and socioeconomic status on perinatal outcomes. Our objective was to analyze the influence of time since naturalization on low birth weight and maternal socioeconomic status in Belgium.MethodsThe data came from the linkage between the Brussels birth and death registers, the national register of migrant trajectories and the social security register for the years 2004-2010. We used logistic regression to estimate the odds ratios of the associations between low birth weight (LBW) and time since naturalization, by nationality groups, taking into account socioeconomic status (SES), parity and maternal age.ResultsData relate to all singleton births to Belgian, Maghrebi, Sub-Saharan African and Turkish women (n = 76 312). The results show an U-shaped of LBW according to time since naturalization for all migrant groups. LBW declines for women naturalized since less than one year and increases significantly thereafter (pConclusionOur results show that, despite an improvement of their SES, LBW increases among Maghrebi, Sub-Saharan African and Turkish women with time since naturalization. Mothers from Maghreb have lower rates of LBW compared to Belgians and maintain their protection even after more than 10 years of having acquired the Belgian nationality. Additional studies need to be carried out in order to gain a better understanding of the association between migration trajectories, SES and perinatal health of immigrants.

During the 2006 and 2007 special observing periods of the African Monsoon Multidisciplinary Analysis campaign an original experimental system has been implemented in Banizoumbou (Niger) for measuring the size-resolved dust emission flux in natural conditions and documenting the possible influence of wind speed on its size distribution. The instrumental set-up, associated methodology, and the quality tests applied to the data set are described before the results acquired during 2 events of the Monsoon type and 1 of the convective type are analyzed in detail. In good agreement with the theory of sandblasting, it is found in all cases that saltation must take place for a vertical emission flux to be detected. During a particular erosion event, the magnitude of the vertical flux is controlled by the surface roughness, which conditions the saltation threshold, and by the wind friction velocity. The dust flux released by the high energy convective event is also found to be much richer in very fine (u*. This is interpreted as a possible result of the rather long duration (15 min) over which wind fluctuations must be averaged for computing u*, which could make it an inadequate parameter for representing the very short response-time physical processes that are at the origin of fine dust emission at the measurement sites.
