This data paper presents a survey conducted in a participatory manner in the territory of Santarém, in the Brazilian Amazon. The aim is to understand how global changes are affecting family farmers. In the study area, family farming has been confronted over the past 20 years with the rapid expansion of large-scale monocultures, especially soybean. As part of the Odyssea socio-environmental observatory, academic researchers and family farming organizations entered into a partnership to co-produce data that could be strategically useful for these organizations. A process of co-construction of the expectations allowed priorities to be established and the data collection strategy to be defined. Three levels of analysis were chosen in order to allow an integrated understanding of the dynamics of change: the Santarem Plateau territory, the rural community level (living place recognized by the farmers) and the farmers’ households. Twenty-one farmers, called community researchers, organized in 3 teams, applied a questionnaire through the KoboCollect smartphone application to 544 families in the municipalities of Santarém, Mojuí dos Campos and Belterra. Meetings were previously held in the rural communities and questionnaires were applied with representatives of 32 communities. Data was collected between April and June 2019. The community researchers and academic researchers then came together for two collective sessions of data analysis and interpretation in July and October 2019. Data was standardized and cleaned using SPSS software, between September and December 2019. The metadata and databases are available on the CIRAD dataverse.

Objectives Increasingly, laboratories flag low serum alkaline phosphatase (sALP) that are age-and sex-specific in paediatrics. The aim of this study was to report clinical manifestations of paediatric patients with age-and sex-specific low sALP, thereby increasing awareness of its potential aetiologies. Methods This retrospective Canadian tertiary care paediatric hospital study assessed all sALP of ambulatory patients aged less than 18 years from 2015 to 2017. The hospital used a Beckman Coulter AU assay to measure sALP and compared values to the Canadian age-and sex-specific reference intervals from CALIPER. All children who had at least one subnormal age-and sex-specific sALP were evaluated. A review of medical charts of included patients was performed and demographic characteristics, medical history and diagnosis were collected, and categorized under groups of medical disorders. Results Of 11,874 included patients, 1,001 patients (9.2%) had low sALP. Of those, 48% (485/1,001) had transient low sALP activity and 9.6% (96/1,001) had persistently low sALP. Prolonged immobilization and inflammatory bowel disease represented the main aetiologies for persistently low sALP. Interestingly, 13.5% (13/96) of patients with persistently low sALP had no apparent aetiology. Conclusions Our results report aetiologies of low sALP in a Canadian paediatric population using age-and sex-specific Canadian reference ranges. This study highlights that healthcare providers should be aware that a low sALP may have clinical significance and should be repeated if warranted based on further clinical assessment. Adequate nutrient supply is crucial for the proper development of the embryo. Although nutrient supply is determined by maternal diet, the gut microbiota also influences nutrient availability. While currently there is no cure for neural tube defects (NTDs), their prevention is largely amenable to maternal folic acid and inositol supplementation. The gut microbiota also contributes to the production of these nutrients, which are absorbed by the host, but its role in this context remains largely unexplored. In this study, we performed a functional and morphological analysis of the intestinal tract of loop-tail mice (Vangl2 mutants), a mouse model of folate/inositol-resistant NTDs. In addition, we investigated the changes in gut microbiota using 16S rRNA gene sequencing regarding (1) the host genotype; (2) the sample source for metagenomics analysis; (3) the pregnancy status in the gestational window of neural tube closure; (4) folic acid and (5) D-chiro-inositol supplementation. We observed that Vangl2+/Lp mice showed no apparent changes in gastrointestinal transit time or fecal output, yet exhibited increased intestinal length and cecal weight and gut dysbiosis. Moreover, our results showed that the mice supplemented with folic acid and D-chiro-inositol had significant changes in their microbiota composition, which are changes that could have implications for nutrient absorption. The meiotic recombination checkpoint reinforces the order of events during meiotic prophase I, ensuring the accurate distribution of chromosomes to the gametes. The AAA+ ATPase Pch2 remodels the Hop1 axial protein enabling adequate levels of Hop1-T318 phosphorylation to support the ensuing checkpoint response. While these events are localized at chromosome axes, the checkpoint activating function of Pch2 relies on its cytoplasmic population. In contrast, forced nuclear accumulation of Pch2 leads to checkpoint inactivation. Here, we reveal the mechanism by which Pch2 travels from the cell nucleus to the cytoplasm to maintain Pch2 cellular homeostasis. Leptomycin B treatment provokes the nuclear accumulation of Pch2, indicating that its nucleocytoplasmic transport is mediated by the Crm1 exportin recognizing proteins containing Nuclear Export Signals (NESs). Consistently, leptomycin B leads to checkpoint inactivation and impaired Hop1 axial localization. Pch2 nucleocytoplasmic traffic is independent of its association with Zip1 and Orc1. We also identify a functional NES in the non-catalytic N-terminal domain of Pch2 that is required for its nucleocytoplasmic trafficking and proper checkpoint activity. In sum, we unveil another layer of control of Pch2 function during meiosis involving nuclear export via the exportin pathway that is crucial to maintain the critical balance of Pch2 distribution among different cellular compartments. Author summary: Meiosis is a specialized cell division essential for sexual reproduction because it is responsible for halving the number of chromosomes during gametogenesis. Meiosis involves a series of events that culminate in the interaction between maternal and paternal chromosomes, and the exchange of genetic material by recombination. These processes not only generate genetic diversity, but also, and more important, are essential to promote proper distribution of chromosomes to the gametes. Meiotic cells possess surveillance mechanisms, called checkpoints, that reinforce the proper order of events during the meiotic cell cycle, ensuring the accurate segregation of chromosomes and preventing the formation of aneuploid gametes. The Pch2 protein is an evolutionarily-conserved ATPase that functions in the yeast meiotic recombination checkpoint. Pch2 localizes in the nucleolus, chromosomes, and the cytoplasm, performing distinct and, in some cases, opposite functions in these different subcellular compartments. In this work, we uncover the mechanism that Pch2 uses to travel from the nucleus to the cytoplasm; in particular, the Crm1 exportin pathway. We also identify and characterize a nuclear export signal in the non-catalytic domain of Pch2 required for its nucleocytoplasmic trafficking. Escherichia coli is a key indicator of food hygiene, and its monitoring in meat samples points to the potential presence of antimicrobial-resistant strains capable of causing infections in humans, encompassing resistance profiles categorized as serious threats by the Centers for Disease Control and Prevention (CDC), such as Extended-Spectrum Beta-Lactamase (ESBL)—a problem with consequences for animal, human, and environmental health. The objective of the present work was to isolate and characterize ESBL-producing E. coli strains from poultry, pork, and beef meat samples, with a characterization of their virulence and antimicrobial resistance profiles. A total of 450 meat samples (150 chicken, 150 beef, and 150 pork) were obtained from supermarkets and subsequently cultured in medium supplemented with cefotaxime. The isolated colonies were characterized biochemically, followed by antibiogram testing using the disk diffusion technique. Further classification involved biofilm formation and the presence of antimicrobial resistance genes (blaCTX-M, AmpC-type, mcr-1, and fosA3), and virulence genes (eaeA, st, bfpA, lt, stx1, stx2, aggR, iss, ompT, hlyF, iutA, iroN, fyuA, cvaC, and hylA). Statistical analysis was performed via the likelihood-ratio test. In total, 168 strains were obtained, with 73% originating from chicken, 22% from pork, and 17% from beef samples. Notably, strains exhibited greater resistance to tetracycline (51%), ciprofloxacin (46%), and fosfomycin (38%), apart from β-lactams. The detection of antimicrobial resistance in food-isolated strains is noteworthy, underscoring the significance of antimicrobial resistance as a global concern. More than 90% of the strains were biofilm producers, and strains carrying many ExPEC genes were more likely to be biofilm formers (OR 2.42), which increases the problem since the microorganisms have a greater chance of environment persistence and genetic exchange. Regarding molecular characterization, bovine samples showed a higher prevalence of blaCTX-M-1 (OR 6.52), while chicken strains were more likely to carry the fosA3 gene (OR 2.43, CI 1.17–5.05) and presented between 6 to 8 ExPEC genes (OR 2.5, CI 1.33–5.01) compared to other meat samples. Concerning diarrheagenic E. coli genes, two strains harbored eae. It is important to highlight these strains, as they exhibited both biofilm-forming capacities and multidrug resistance (MDR), potentially enabling colonization in diverse environments and causing infections. In conclusion, this study underscores the presence of β-lactamase-producing E. coli strains, mainly in poultry samples, compared to beef and pork samples. Furthermore, all meat sample strains exhibited many virulence-associated extraintestinal genes, with some strains harboring diarrheagenic E. coli (DEC) genes. [ABSTRACT FROM AUTHOR]

The aim of this work was a complete thermodynamic study on the partitioning of Remazol Yellow l, Remazol Brilliant Blue, and Remazol Red dyes in isopropyl alcohol + sodium phosphate + water aqueous two‐phase systems at different temperatures. The free energy change of transfer, enthalpy change of transfer, and entropy change of transfer were evaluated. The effects of the structure of the dye, tie line length, and temperature on the partitioning were studied. The results indicate that these aqueous two‐phase systems have high capacity to extract the dyes from aqueous matrices to the organic phase of these systems and are an interesting alternative to effluent treatment process. The aim of this study was to evaluate whether high-fat (HF) diet intake during puberty can program obesity as well as generate glucose imbalance and hepatic metabolic dysfunctions in adult life. Male Wistar rats were randomly assigned into two groups: rats fed standard chow (NF) and rats fed a HF from postnatal 30-day-old (PND30) until PND60. Then, both groups were fed a standard chow from PND60 until PND120. Euthanasia and samples collections occurred at PND120. HF animals were overweight (+11%) and had increased adiposity, hyperphagia (+12%), hyperglycaemia (+13%), hyperinsulinemia (+69%), and hypertriglyceridemia (+34%). Plasma glucose levels during intravenous glucose tolerance test (ivGTT) and intraperitoneal insulin tolerance test (ipITT) were also higher in the HF group, whereas Kitt was significantly lower (–34%), suggesting reduced insulin sensitivity. In the same sense, HF animals present pancreatic islets hypertrophy and high β-cell mass. HF animals also had a significant increase in blood glucose levels during pyruvate tolerance test, indicating increased gluconeogenesis. Hepatic morphology analyses showed an increase in lipid inclusion in the HF group. Moreover, PEPCK and FAS protein expression were higher in the livers of the HF animals (+79% and + 37%, respectively). In conclusion, HF during puberty causes obese phenotype leading to glucose dyshomeostasis and nonalcoholic fatty liver disease, which can be related to the overexpression of proteins PEPCK and FAS. The efficacy of 5-((4-methoxyphenyl)thio)benzo[c][1,2,5] thiodiazole (MTDZ) in mitigating paclitaxel (PTX)-induced peripheral neuropathy was investigated in male and female Swiss mice. The study examined the effects of MTDZ on various pathways, including transient receptor potential cation channel subfamily V member 1 (TRPV1), glutamatergic, nitrergic, guanylate cyclase (cGMP), serotonergic, and opioidergic. Mice received intraperitoneal PTX (2 mg/kg) or vehicle on days 1, 2, and 3, followed by oral MTDZ (1 mg/kg) or vehicle from days 3 to 14. Mechanical and thermal sensitivities were assessed using Von Frey and hot plate tests on days 8, 11, and 14. The open field test evaluated locomotion and exploration on day 12. On day 15, nitrite and nitrate (NOx) levels and Ca2+−ATPase activity in the cerebral cortex and spinal cord were measured after euthanizing the animals. MTDZ administration reversed the heightened mechanical and thermal sensitivities induced by PTX in male and female mice without affecting locomotion or exploration. MTDZ also modulated multiple pathways, including glutamatergic, NO/L−arginine/cGMP, serotonergic (5−HT1A/1B), opioid, and TRPV1 pathways. Additionally, MTDZ reduced NOx levels and modulated Ca2+−ATPase activity. In conclusion, MTDZ effectively alleviated PTX−induced peripheral neuropathy and demonstrated multi-targeted modulation of pain-related pathways. Its ability to modulate multiple pathways, reduce NOx levels, and modulate Ca2+−ATPase activity makes it a potential pharmacological candidate for peripheral neuropathy, acute nociceptive, and inflammatory conditions. A mathematical model is proposed to describe and simulate microalgae growth from emissions in an outdoor airlift photobioreactor. To this end, the influences of the concentration of total nitrogen, total phosphorus, CO2 present in the exhaust gases, temperature, and light intensity were considered in biomass production. The model was tested under different experimental conditions with biomass concentrations ranging from 2 g L−1 to 4.5 g L−1. Furthermore, the accuracy of the airlift photobioreactor simulations was greater than 90 %. Thus, the developed model was able to satisfactorily describe the experimental data, so that it can be applied in other microalgae systems for optimizing biomass production, both on laboratory and pilot scales. The C–S cross‐coupling reaction is a very efficient and powerful tool for constructing C–S bonds. Despite the well‐reported C–S cross‐coupling protocols using aryl halides, this reaction using sp2‐hybridized nonaromatic halides is still challenging. Herein, we report light‐assisted C–S cross‐coupling between thiols and 5‐chlorobenzofurazan under blue LED irradiation. We used a new nickel catalyst to synthesize 11 unpublished compounds. Furthermore, we assessed the mechanism and the influence of light irradiation on this reaction and proposed a plausible mechanism that is mediated by the excited state of the nickel catalyst promoted by irradiation with blue light and the C–S coupling product formed. Notably, C–S cross‐coupling under light irradiation was performed for the first time using a nickel catalyst in the absence of an external photocatalyst. We demonstrated that because nickel has a low energy barrier for oxidative addition, this protocol proceeded efficiently using chlorine as a leaving group, and the excited state of the nickel catalyst promoted reductive elimination with a lower energy barrier. It is important to mention that the procedure was tolerated by many functional groups within the thiol, affording a greater than 99% yield after reacting for 6 h. [ABSTRACT FROM AUTHOR]

Background and Objectives: Although peak oxygen uptake (VO2peak) is one of the most important measures in clinical practice, the high cost and time consumption have led to the search for simpler devices and the development of the estimating cardiopulmonary fitness (eCPF) equation. Since the lungs are one of the sites most affected by rheumatoid arthritis (RA), this study aimed to create a predictive equation for VO2peak obtained by simple sampling technology in women with RA‐associated interstitial lung disease (RA‐ILD). Methods: This cross‐sectional study evaluated 47 women with RA‐ILD. The participants underwent the following evaluations: computed tomography (CT); evaluation of disease activity through the Clinical Disease Activity Index (CDAI); measurement of physical function using the Health Assessment Questionnaire disability index (HAQ‐DI); pulmonary function testing, including spirometry, diffusing capacity for carbon monoxide (DlCO), nitrogen single‐breath washout (N2SBW) test, and impulse oscillometry; and cardiopulmonary exercise testing (CPET) using FitMate™. Results: VO2peak was correlated with age (r = −0.550, p < 0.0001), rheumatoid factor (r = −0.443, p = 0.002), anti‐cyclic citrullinated peptide antibodies (r = −0.410, p = 0.004), CDAI (r = −0.462, p = 0.001), HAD‐DI (r = −0.486, p = 0.0005), forced vital capacity (r = 0.491, p = 0.0004), DlCO (r = 0.621, p < 0.0001), phase III slope of N2SBW (r = −0.647, p < 0.0001), resonance frequency (Fres, r = −0.717, p < 0.0001), integrated low‐frequency reactance (r = −0.535, p = 0.0001), and the inhomogeneity of respiratory system resistance between 4 and 20 Hz (r = −0.631, p < 0.0001). In the CT examination, patients with extensive ILD had significantly lower VO2peak than patients with limited ILD (p < 0.0001). In the stepwise forward regression analysis, Fres, DlCO and age explained 61% of the VO2peak variability. Conclusions: As assessed by CPET, women with RA‐ILD show reduced cardiopulmonary fitness, which can be explained at least in part by the presence of small airway disease, deterioration of pulmonary gas exchange, and advanced age. These associations of pulmonary variables with eCPF may be clinically important and support the use of the eCPF equation to improve patient outcomes. [ABSTRACT FROM AUTHOR]

To fully leverage the benefits of augmented and mixed reality (AR/MR) in supporting users, it is crucial to establish a consistent and well-defined situated visualization (SV) model. SV encompasses visualizations that adapt based on context, considering the relevant visualizations within their physical display environment. Recognizing the potential of SV in various domains such as collaborative tasks, situational awareness, decision-making, assistance, training, and maintenance, AR/MR is well-suited to facilitate these scenarios by providing additional data and context-driven visualization techniques. While some perspectives on the SV model have been proposed, such as space, time, place, activity, and community, a comprehensive and up-to-date systematization of the entire SV model is yet to be established. Therefore, there is a pressing need for a more comprehensive and updated description of the SV model within the AR/MR framework to foster research discussions. [ABSTRACT FROM AUTHOR]

Virtual reality shows great potential as an alternative to traditional therapies for motor rehabilitation given its ability to immerse the user in engaging scenarios that abstract them from medical facilities and tedious rehabilitation exercises. This paper presents a virtual reality application that includes three serious games and that was developed for motor rehabilitation. It uses a standalone headset and the user's hands without the need for any controller for interaction. Interacting with an immersive virtual reality environment using only natural hand gestures involves an interaction that is similar to that of real life, which would be especially desirable for patients with motor problems. A study involving 28 participants (4 with motor problems) was carried out to compare two types of interaction (hands vs. controllers). All of the participants completed the exercises. No significant differences were found in the number of attempts necessary to complete the games using the two types of interaction. The group that used controllers required less time to complete the exercise. The performance outcomes were independent of the gender and age of the participants. The subjective assessment of the participants with motor problems was not significantly different from the rest of the participants. With regard to the interaction type, the participants mostly preferred the interaction using their hands (78.5%). All four participants with motor problems preferred the hand interaction. These results suggest that the interaction with the user's hands together with standalone headsets could improve motivation, be well accepted by motor rehabilitation patients, and help to complete exercise therapy at home.
