2. Abrogation of KLF5 sensitizes BRCA1-proficient pancreatic cancer to PARP inhibition

Abstract

Poly ADP-ribose polymerase (PARP) inhibitor monotherapies are selectively effective in patients with pancreatic, breast, prostate, and ovarian cancers with BRCA1 mutations. Cancer patients with more frequent wild-type BRCA show poor responses to PARP inhibitors. Moreover, patients who are initially sensitive to these inhibitors eventually respond poorly to drugs. In the present study, we discover that abrogation of Kruppel-like factor 5 (KLF5) significantly inhibits homologous recombination, which is the main mechanism for DNA double-stranded repair. Furthermore, the downregulation of KLF5 expression promotes the DNA damage induced by olaparib and significantly reduces the IC50 of the RARP inhibitor in pancreatic cancer cells. Overexpression of BRCA1 reverses the above effects caused by silencing of KLF5. Olaparib combined with a KLF5 inhibitor has an enhanced cytotoxic effect. Mechanistically, we identify BRCA1 as a KLF5 target gene. BRCA1 is positively correlated with KLF5 in PDAC tissue. Our results indicate that inhibition of KLF5 may induce BRCAness in a larger pancreatic cancer subset with proficient BRCA. The combination of KLF5 inhibitors and PARP inhibitors provides a novel treatment strategy to enhance the sensitivity of BRCA1-proficient pancreatic cancer to PARP inhibitors.

26. Investigating the role of FcRL4⁺ B cells at the site of epithelial inflammation

Abstract

The slow and progressive deterioration of the small bile ducts is a hallmark of primary biliary cholangitis (PBC). This damage is mediated by infiltrating lymphocytes which form aggregates surrounding the small bile ducts eventually leading to cholestasis and cirrhosis. A subset of memory B cells expressing the surface marker Fc receptor like 4 (FcRL4) have been implicated in the pathogenesis of rheumatoid arthritis and primary Sjogren's syndrome. Here, we addressed the hypothesis that the FcRL4⁺ B cells are involved in the lymphoepithelial lesion that leads to the destruction of bile ducts in PBC patients and explored their role in the disease. In explant livers from patients undergoing liver transplantation, FcRL4⁺ B cells were identified in PBC livers and were largely absent in other end-stage liver diseases. Within the PBC liver, these cells are highly proliferative and can be seen within immune infiltrates that surround and infiltrate the damaged biliary epithelium. FcRL4⁺ B cells isolated from the PBC liver are enriched for CD27⁺IgD⁻ class-switched memory cells and CD11c⁺CD21⁻T-bet⁺ age associated B cells (ABCs). FcRL4 is a receptor for IgA aggregates. FcRL4⁺ B cells isolated from end-stage PBC liver have IgA captured on their surface. In vitro, FcRL4 expressing cells can bind IgA from patient derived bile. High levels of expression of co-stimulatory molecules CD80 and CD86 by FcRL4⁺ suggest that they can act as antigen presenting cells. FcRL4⁺ B cells preferentially express IgA BCR, suggesting a mucosal origin. Together with their ability to capture IgA immune complexes, these findings suggest that FcRL4⁺ B cells may act as a link between mucosal immunity and immune mediated damage of the small bile ducts in PBC.

Abstract

Using ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF-MS), we separated and identified 148 non-volatile organic compounds including flavonoids, organic acids and amino acids in navel orange pulp in 37 min. The volatile organic compounds (VOCs) in the pulp of navel oranges from four major production regions in southern Jiangxi Province, Ruijin, Xinfeng, Anyuan and Nankang were detected by headspace solid phase microextraction combined with gas chromatography-mass spectrometry (HS-SPME-GC-MS). In total, 97 VOCs were identified including esters, alcohols, aldehydes and terpenes. The results of this study provide detailed data for further research on chemical substances in the pulp of Gannan navel oranges.

35. Genetic alterations of KRAS and TP53 in intrahepatic cholangiocarcinoma associated with poor prognosis

Abstract

The aim of this study is to investigate certain genetic features of intrahepatic cholangiocarcinoma (ICCA). A total of 12 eligible ICCA patients were enrolled, and tumor tissues from the patients were subjected to next-generation sequencing of a multi-genes panel. Tumor mutation burden (TMB), mutated genes, copy number variants (CNVs), and pathway enrichment analysis were performed. The median TMB was 2.76 Mutation/Mb (range, 0–36.62 Mutation/Mb) in ICCA patients. The top two most commonly mutated genes in ICCA were KRAS (33%) and TP53 (25%). The co-mutations of KRAS and TP53 were 16.7% (2/12) in ICCA patients. Notably, patient P6 with the highest TMB did not have KRAS and TP53 mutations. Additionally, TP53 and/or KRAS alterations were significantly associated with poor progression-free survival than those with wild type (1.4 months vs 18 months). DNA damage repair and homologs recombinant repair deficiencies were significantly associated with high TMB in ICCA cases. In conclusion, we found that certain genetic mutations of TP53 and KRAS could predict poor prognosis in ICCA patients.
