Piperazines are a class of new psychoactive substances with hallucinogenic effects that affect the central nervous system by affecting the level of monoamine neurotransmitters. Abuse of piperazines will produce stimulating and hallucinogenic effects, accompanied by headache, dizziness, anxiety, insomnia, vomiting, chest pain, tachycardia, hypertension and other adverse reactions, and may even cause cardiovascular diseases and multiple organ failure and lead to death, seriously affecting human physical and mental health and public safety. The abuse of new psychoactive substance piperazines has attracted extensive attention from the international community. The study of its pharmacological toxicology and analytical methods has become a research hotspot in the field of forensic medicine. This paper reviews the in vivo processes, sample treatment and analytical methods of existing piperazines, in order to provide reference for forensic identification.

Talent is one of the basic and strategic supports for building a modern socialist country in all aspects. Since the 1980s, the establishment of forensic medicine major and the cultivation of innovative talents in forensic medicine have become hot topics in higher education in forensic medicine. Over the past 43 years, the forensic medicine team of Shanxi Medical University has adhered to the joint education of public security and colleges, and made collaborative innovation, forming a training mode of “One Combination, Two Highlights, Three Combinations, Four in One” for innovative talents in forensic medicine. It has carried out “5+3/X” integrated reform, and formed a relatively complete talent training innovation mode and management system in teaching, scientific research, identification, major, discipline, team, platform and cultural construction. It has made a historic contribution to China’s higher forensic education, accumulated valuable experience for the construction of first-class major and first-class discipline of forensic medicine, and provided strong support for the construction of the national new forensic talent training system. The popularization of this training mode is conducive to the rapid and sustainable development of forensic science, and provides more excellent forensic talents for national building, regional social development and the discipline construction of forensic science.

ObjectiveTo establish a method for the detection of carbamazepine and its metabolites 10,11-dihydro-10,11-epoxycarbamazepine and 10,11-dihydro-10-hydroxycarbamazepine in blood samples by liquid chromatography-tandem mass spectrometry （LC-MS/MS）.MethodsThe blood samples were treated with 1-butyl-3-methylimidazolium hexafluorophosphate as an extraction solvent. The samples were extracted by ultrasound-assisted extraction and separated by ZORBAX Eclipse Plus C18， 95Å column. The mobile phase A aqueous solution containing 0.1% formic acid and 10 mmol/L ammonium acetate, and mobile phase B mixed organic solvent containing acetonitrile/methanol （Vacetonitrile∶Vmethanol=2∶3） were used for gradient elution at the flow rate of 1.00 mL/min. An electrospray ion source in positive mode was used for detection in the multiple reaction monitoring.ResultsThe linearities of carbamazepine and its metabolites 10,11-dihydro-10,11-epoxycarbamazepine and 10,11-dihydro-10-hydroxycarbamazepine in blood samples were good within the corresponding range, with correlation coefficients （r） greater than 0.995 6. The limits of detection were 3.00， 0.40 and 1.30 ng/mL，respectively. The limit of quantitation were 8.00，1.00 and 5.00 ng/mL，respectively. The extraction recoveries ranged from 76.00% to 106.44%. The relative standard deviations of the intra-day and inter-day precisions were less than 16%. Carbamazepine and its main metabolite 10,11-dihydro-10,11-epoxycarbamazepine were detected in blood samples of death cases with a mass concentration of 2.71 μg/mL and 252.14 ng/mL，respectively.ConclusionThis method has high sensitivity and good selectivity， which is suitable for the detection of carbamazepine and its metabolites in blood samples， and can be used for carbamazepine-related forensic identifications.

ObjectiveTo establish a carbofuran intragastric administration death model in rabbits, and to observe the postmortem distribution and postmortem redistribution of carbofuran-7-phenyl glucuronic acid （Glu-7PH） in rabbits.MethodsThe postmortem distribution: Rabbits were given an administration of 1/2LD50, LD50, 2LD50 carbofuran. Dead rabbits were dissected immediately. Rabbits that had remained alive 2 hours were sacrificed by carbon dioxide （CO2） inhalation and dissected immediately. The myocardium, cardiac blood, liver, spleen, lung, kidney, brain and right hindlimb muscle were collected. The postmortem redistribution: After giving an administration of 4LD50 carbofuran, the myocardium, cardiac blood, liver, spleen, lung, kidney, brain, and right hindlimb muscle were collected at 0, 12, 24, 48, and 72 h postmortem in supine position at 15 ℃ room temperature. The quantity of Glu-7PH was determined by LC-MS/MS.ResultsThe postmortem distribution: Among the three dose groups, there were significant differences in the quantities of Glu-7PH in different tissues. The postmortem redistribution: There was no significant difference in the Glu-7PH quantities in cardiac blood, mycardium, spleen, kidney, brain and right hindlimb muscle, but there was a significant difference in the Glu-7PH quantities in the liver and lung.ConclusionThe mycardium, cardiac blood, liver, lung, kidney, brain and hindlimb muscle of rabbits can be used as appropriate samples for Glu-7PH detection. However, it should be noted that Glu-7PH was redistributed postmortem in rabbit liver and lung.

ObjectiveTo explore the differential expression of messenger RNA （mRNA） in myocardial tissues of rats with sudden coronary death （SCD）, and to provide ideas for the forensic identification of SCD.MethodsThe rat SCD model was established, and the transcriptome sequencing was performed by next-generation sequencing technology. Differentially expressed genes （DEGs） in myocardial tissues of SCD rats were screened by using the R package limma. A protein-protein interaction （PPI） network was constructed by using the STRING database and Cytoscape 3.8.2 on DEG, and hub genes were screened based on cytoHubba plug-in. Finally, the R package clusterProfiler was used to analyze the biological function and signal pathway enrichment of the selected DEG.ResultsA total of 177 DEGs were associated with SCD and were mainly involved in the renin-angiotensin system and PI3K-Akt signaling pathway. The genes including angiotensinogen （AGT）, complement component 4a （C4a）, Fos proto-oncogene （FOS） and others played key roles in the development of SCD.ConclusionGenes such as AGT, C4a, FOS and other genes are expected to be potential biomarkers for forensic identification of SCD. The study based on mRNA expression profile can provide a reference for forensic identification of SCD.

ObjectiveTo explore the mRNA differential expressions and the sequential change pattern in acute myocardial infarction （AMI） mice.MethodsThe AMI mice relevant dataset GSE4648 was downloaded from Gene Expression Omnibus （GEO）. In the dataset， 6 left ventricular myocardial tissue samples were selected at 0.25， 1， 4， 12， 24 and 48 h after operation in AMI group and sham control group， and 6 left ventricular myocardial tissue samples were selected in blank control group， a total of 78 samples were analyzed. Differentially expressed genes （DEGs） were analyzed by R/Bioconductor package limma， functional pathway enrichment analysis was performed by clusterProfiler， protein-protein interaction （PPI） network was constructed by STRING database and Cytoscape software， the key genes were identified by Degree topological algorithm， cluster sequential changes on DEGs were analyzed by Mfuzz.ResultsA total of 1 320 DEGs were associated with the development of AMI. Functional enrichment results included cellular catabolic process， regulation of inflammatory response， development of muscle system and vasculature system， cell adhesion and signaling pathways mainly enriched in mitogen-activated protein kinase （MAPK） signaling pathway. The key genes of AMI included MYL7， TSC22D2， HSPA1A， BTG2， NR4A1， RYR2 were up-regulated or down-regulated at 0.25-48 h after the occurrence of AMI.ConclusionThe functional signaling pathway of DEGs and the sequential expression of key genes in AMI may provide a reference for the forensic identification of AMI.

Objective To explore the postmortem diffusion rule of Aconitum alkaloids and their metabolites in poisoned rabbits, and to provide a reference for identifying the antemortem poisoning or post-mortem poisoning of Aconitum alkaloids. Methods Twenty-four rabbits were sacrificed by tracheal clamps. After 1 hour, the rabbits were administered with aconitine LD50 in decocting aconite root powder by intragastric administration. Then, they were placed supine and stored at 25 °C. The biological samples from 3 randomly selected rabbits were collected including heart blood, peripheral blood, urine, heart, liver, spleen, lung and kidney tissues at 0 h, 4 h, 8 h, 12 h, 24 h, 48 h, 72 h and 96 h after intragastric administration, respectively. Aconitum alkaloids and their metabolites in the biological samples were analyzed by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Results At 4 h after intragastric administration, Aconitum alkaloids and their metabolites could be detected in heart blood, peripheral blood and major organs, and the contents of them changed dynamically with the preservation time. The contents of Aconitum alkaloids and their metabolites were higher in the spleen, liver and lung, especially in the spleen which was closer to the stomach. The average mass fraction of benzoylmesaconine metabolized in rabbit spleen was the highest at 48 h after intragastric administration. In contrast, the contents of Aconitum alkaloids and their metabolites in kidney were all lower. Aconitum alkaloids and their metabolites were not detected in urine. Conclusion Aconitum alkaloids and their metabolites have postmortem diffusion in poisoned rabbits, diffusing from high-content organs (stomach) to other major organs and tissues as well as the heart blood. The main mechanism is the dispersion along the concentration gradient, while urine is not affected by postmortem diffusion, which can be used as the basis for the identification of antemortem and postmortem Aconitum alkaloids poisoning. Effects of gap sizes (50, 100 and 150 m2) on seasonal dynamics of soil organic carbon (SOC) and microbial biomass carbon (MBC) in soil surface (0-15 cm) and subsurface (15-30 cm) were investigated in a 50-year old Picea asperata plantation in Miyaluo forest, Western Sichuan, China. In the four seasons, the SOC and MBC contents were higher in the soil surface than in soil subsurface in the four treatments, and varied insignificantly in different seasons. Compared with the control, the SOC content increased by 35.4%, 21.2% and 10.3% in soil surface, and by 45.5%, 25.0% and 12.1% in soil subsurface with the forest gaps of 50, 100 and 150 m2, respectively. The MBC content increased by 26.7%, 16.7%, 11.3% and 24.4%, 12.6%, 7.3% in soil surface and subsurface, respectively. There were significant negative relationship between soil SOC content and the soil pH and moisture, and significant positive relationship between soil SOC content and soil temperature. Significant effects of SOC content, soil pH, soil temperature, soil moisture on MBC content were observed, suggesting that forest gaps could significantly affect environmental conditions, and increasing gaps could reduce microbial activity and decomposition rate and decrease soil SOC and MBC contents. Fine root in forest ecosystems plays an important role in global C cycle. In this study, a measurement was made on the fine root biomass and carbon storage in the surface soil (0-30 cm) of a 31 year-old Cinnamomum camphora plantation in the Rainy Area of West China in November, 2010-December, 2011. The total biomass and carbon storage of the fine roots (living and dead) in the surface soil were 1592.29 kg•hm-2 and 660.68 kg C•hm-2, in which, living fine roots accounted for 91. 1% and 91.8% respectively. The total biomass and carbon storage of the first five order living roots and dead roots decreased significantly with increasing soil depth, and the living root biomass and carbon storage increased significantly with root order. The sum of the biomass and carbon storage of living and dead fine roots was the largest in autumn and the smallest in winter, but the biomass and carbon storage of the dead fine roots were the largest in winter and the smallest in summer. The biomass and carbon storage of the first two order roots were the largest in summer and the smallest in winter, while those of the last three order roots were the largest in autumn and the smallest in winter. The spatial heterogeneity of soil moisture and nutrients was the main factor affecting the fine root biomass and carbon storage. [ABSTRACT FROM AUTHOR]

Drug abuse is very easy to cause the residues of harmful substances in food of animal origin, which not only does direct harm to human health, but also does great harm to the development of animal husbandry and the ecological environment. Therefore, the detection of residual drugs in animal-derived food is of great significance to ensure food safety. In this study, 28 drugs in lambs were simultaneous determined by modified QuEChERs combined with ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) based on magnetic multi-walled carbon nanotubes (Fe3O4-MWCNTs). Under the optimized separation and detection conditions, the effects of different dehydrated salts (MgSO4, Na2SO4) and metal chelating agents (Na2EDTA) on the drug recoveries were investigated. Then, the Fe3O4-MWCNTs adsorbent dosages on the purification efficiency were evaluated in terms of drug recoveries and residual matrix masses as well as decoloration effects. The Fe3O4-MWCNTs showed better or equivalent purification performance results than commercial purification adsorbents (C18, PSA, GCB). The results showed that the linear relationship was good for all monitored drugs in the range of 15-500 μg/kg, the correlation coefficient was better than 0.999. By constructing matrix-matched curves and solvent standard curves, the calculated matrix effects (MEs) were located within -9.7%-14.2%, demonstrating its good matrix purification efficiency. Under three spiked concentrations (20, 100, 200 μg/kg), satisfactory recoveries were obtained in the range of 60.2%-114.3%, 68.9%-106.7% and 60.4%-94.8%, respectively, which demonstrated the good accuracy of the method in quantitative analysis. The method precision was validated by intra- and inter-day repetitive experiments on the same day or on the three consecutive days. The obtained intra- and inter-day relative standard deviations (RSDs) were no more than 9.9% and 13.0%, respectively, which indicated the good precision and robustness of the developed method. The limits of detection (LODs) and limits of quantitation (LOQs) were obtained in the range of 0.1-5.0 μg/kg and 0.3-12.0 μg/kg, respectively, which were sensitive enough to analyze the monitored drugs with concentrations exceeding their corresponding maximum residue limits. This method has good applicability in qualitative and quantitative analysis of 28 drug residues in lambs, and can provide a strong technical support for rapid detection of food hazards in other animal origins.

Parabacteroides distasonis is a gram-negative bacterium initially isolated from a clinical specimen in the 1930s. The strain was re-classified to form the new genus Parabacteroides in 2006. P. distasonis can regulate intestinal barrier function and plays a key role in immune response and metabolic regulation of bodies. Traditional Chinese medicine(TCM) is closely related to the intestinal microbiota. Polysaccharides, saponins, and other ingredients of TCM can treat diseases by interacting with P. distasonis, but the specific mechanisms underlying these processes are still unclear, requiring further exploration. This study reviewed the roles and related mechanisms of P. distasonis in inflammatory-immune diseases, metabolic diseases, cardiovascular disease, neuropsychiatric diseases, cancer, and other diseases and summarized the relevant research results of TCM to prevent and treat diseases by regulating P. distasonis. This study provides a reference for subsequent exploration of P. distasonis and research on the interaction between TCM and intestinal microbiota.

This study explores the reparative effect of Qixiong Zuogui Compound Prescription(QXZG) intervention on the blood-brain barrier(BBB) in the aging brain with middle cerebral artery occlusion(MCAO) in rats mediated by bone marrow stem cells(BMSCs)-derived exosomes, as well as its anti-aging mechanism. An aging MCAO composite model was established using D-galactose-induced aging combined with line embolism. Rats were divided into young sham surgery group, aging sham surgery group, model group, exosome group, and exosome with traditional Chinese medicine(TCM) intervention group. SA-β-gal staining combined with TTC staining were used to evaluate the model establishment. Exosomes group or exosomes with TCM intervention group were injected via the tail vein 2 h post-surgery and 48 and 96 h post-ischemia reperfusion. Neurological deficit scores were assessed at 2 h, 3 d, and 7 d post-modeling. Cognitive function was evaluated using novel object recognition tests on the 7th day, and brain infarct volume was observed via MRI. Evans Blue staining was used to evaluate BBB permeability. Western blot was performed to detect the expression of tight junction proteins ZO-1 and Occludin in brain tissue. ELISA was used to measure serum matrix metalloproteinase(MMP)-9 and MMP-2 levels. SA-β-gal staining was used to assess cellular senescence. RT-qPCR was performed to detect p21 mRNA expression, and qPCR was used to measure relative telomere length. The results showed that QXZG intervention-derived BMSCs exosomes significantly reduced brain infarct volume in aging MCAO rats, improved neurological deficit scores, and enhanced cognitive function. These exosomes suppressed MMP-2 and MMP-9 expression, promoted he expression of tight junction proteins ZO-1 and Occludin, reduced BBB permeability, lowered p21 mRNA expression, elongated telomeres, and delayed cell senescence. In conclusion, this study confirms that QXZG intervention-derived BMSC exosomes may repair the BBB by reducing MMP-2 and MMP-9 levels and increasing ZO-1 and Occludin expression. Moreover, its anti-aging mechanism may involve reducing p21 mRNA expression and limiting telomere shor-tening.

Objectives: To establish a rapid screening method for 34 emerging contaminants in surface water by ultra-high performance liquid chromatography-quadrupole-time of flight mass spectrometry (UHPLC-Q-TOF-MS)., Methods: The pretreatment conditions of solid phase extraction (SPE) were optimized by orthogonal experimental design and the surface water samples were concentrated and extracted by Oasis ® HLB and Oasis ® MCX SPE columns in series. The extracts were separated by Kinetex ® EVO C18 column, with gradient elution of 0.1% formic acid aqueous solution and 0.1% formic acid methanol solution. Q-TOF-MS 'fullscan' and 'targeted MS/MS' modes were used to detect 34 emerging contaminants and to establish a database with 34 emerging contaminants precursor ion, product ion and retention times., Results: The 34 emerging contaminants exhibited good linearity in the concentration range respectively and the correlation coefficients ( r ) were higher than 0.97. The limit of detection was 0.2-10 ng/L and the recoveries were 81.2%-119.2%. The intra-day precision was 0.78%-18.70%. The method was applied to analyze multiple surface water samples and 6 emerging contaminants were detected, with a concentration range of 1.93-157.71 ng/L., Conclusions: The method is simple and rapid for screening various emerging contaminants at the trace level in surface water.

Objectives: To investigate the toxicokinetic differences of 3,4-methylenedioxy- N -methylamphetamine (MDMA) and its metabolite 4,5-methylene dioxy amphetamine (MDA) in rats after single and continuous administration of MDMA, providing reference data for the forensic identification of MDMA., Methods: A total of 24 rats in the single administration group were randomly divided into 5, 10 and 20 mg/kg experimental groups and the control group, with 6 rats in each group. The experimental group was given intraperitoneal injection of MDMA, and the control group was given intraperitoneal injection of the same volume of normal saline as the experimental group. The amount of 0.5 mL blood was collected from the medial canthus 5 min, 30 min, 1 h, 1.5 h, 2 h, 4 h, 6 h, 8 h, 10 h, 12 h after administration. In the continuous administration group, 24 rats were randomly divided into the experimental group (18 rats) and the control group (6 rats). The experimental group was given MDMA 7 d by continuous intraperitoneal injection in increments of 5, 7, 9, 11, 13, 15, 17 mg/kg per day, respectively, while the control group was given the same volume of normal saline as the experimental group by intraperitoneal injection. On the eighth day, the experimental rats were randomly divided into 5, 10 and 20 mg/kg dose groups, with 6 rats in each group. MDMA was injected intraperitoneally, and the control group was injected intraperitoneally with the same volume of normal saline as the experimental group. On the eighth day, 0.5 mL of blood was taken from the medial canthus 5 min, 30 min, 1 h, 1.5 h, 2 h, 4 h, 6 h, 8 h, 10 h, 12 h after administration. Liquid chromatography-triple quadrupole tandem mass spectrometry was used to detect MDMA and MDA levels, and statistical software was employed for data analysis., Results: In the single-administration group, peak concentrations of MDMA and MDA were reached at 5 min and 1 h after administration, respectively, with the largest detection time limit of 12 h. In the continuous administration group, peak concentrations were reached at 30 min and 1.5 h after administration, respectively, with the largest detection time limit of 10 h. Nonlinear fitting equations for the concentration ratio of MDMA and MDA in plasma and administration time in the single-administration group and continuous administration group were as follows: T =10.362 C -1.183 , R 2 =0.974 6; T =7.397 3 C -0.694 , R 2 =0.961 5 ( T : injection time; C : concentration ratio of MDMA to MDA in plasma)., Conclusions: The toxicokinetic data of MDMA and its metabolite MDA in rats, obtained through single and continuous administration, including peak concentration, peak time, detection time limit, and the relationship between concentration ratio and administration time, provide a theoretical and data foundation for relevant forensic identification.

Neurodegenerative diseases are a collective term for a large group of diseases caused by degenerative changes in nerve cells. Aging is the main risk factor for neurodegenerative diseases. The neurovascular unit(NVU) is the smallest functional unit of the brain, which regulates brain blood flow and maintains brain homeostasis. Accelerated aging of NVU cells directly impairs NVU function and leads to the occurrence of various neurodegenerative diseases. The intrinsic mechanisms of NVU cell aging are complex and involve oxidative stress damage, loss of protein homeostasis, DNA damage, mitochondrial dysfunction, immune inflammatory response, and impaired cellular autophagy. In recent years, studies have found that traditional Chinese medicine(TCM) can inhibit NVU aging through multiple pathways and targets, exerting a brain-protective effect. Therefore, this article aimed to provide a theoretical basis for further research on TCM inhibition of NVU cell aging and references for new drug development and clinical applications by reviewing its mechanisms of anti-aging, such as regulating relevant proteins, improving mitochondrial dysfunction, reducing DNA damage, lowering inflammatory response, antioxidant stress, and modulating cellular autophagy.

This study aims to systematically evaluate the efficacy and safety of Shuxuetong Injection in the treatment of stroke in progressive. Randomized controlled trials of Shuxuetong Injection in the treatment of stroke in progressive were searched from CNKI, Wanfang, VIP, CMB, PubMed and EMbase. After strict literature screening, data extraction and quality evaluation, a total of 22 articles were included for analysis by RevMan 5.3. The Meta-analysis showed that Shuxuetong Injection combined with conventional treatment was superior to the conventional treatment alone in the major outcome indicators including effective rate(RR=1.27, 95%CI[1.20, 1.33], Z=9.18, P<0.000 01), deterioration rate(RR=0.38, 95%CI[0.22, 0.68], Z=3.31, P=0.000 9), NIHSS scores(MD=-3.89, 95%CI[-4.34,-3.43], Z=16.83, P<0.000 01), CSS scores(MD=-5.59, 95%CI[-6.42,-4.76], Z=13.20, P<0.000 01) and activity of daily living scores(MD=12.02, 95%CI[10.31, 13.72], Z=13.83, P<0.000 01), mortality during treatment was not increased(RR=0.40, 95%CI[0.13, 1.26], Z=1.56, P=0.12). Moreover, Shuxuetong Injection combined with conventional treatment further reduced the secondary outcome indicators including fibrinogen(MD=-0.35, 95%CI[-0.58,-0.13], Z=3.09, P=0.002), triglyceride(MD=-0.38, 95%CI[-0.67,-0.10], Z=2.65, P=0.008), low density lipoprotein cholesterol(MD=-0.72, 95%CI[-0.83,-0.61], Z=12.64, P<0.000 01), serum hypersensitive C-reactive protein(MD=-4.41, 95%CI[-6.96,-1.86], Z=3.38, P=0.000 7), and interleukin-6(MD=-5.43, 95%CI[-6.91,-3.96], Z=7.22, P<0.000 01). GRADE evaluation results showed that the major outcome indicators had low quality of evidence. Shuxuetong Injection in the treatment of stroke in progressive can improve the clinical effective rate, reduce the deterioration rate, improve the neurological function and activity of daily living, down-regulate the levels of fibrinogen, triglyceride, low density lipoprotein cholesterol and alleviate the inflammatory response. Although most studies have reported no adverse reactions, there are selective reports. The safety of Shuxuetong Injection needs to be further verified by more high-quality randomized controlled trial.

Post-incision pain often occurs after surgery and is emergent to be treated in clinic. It hinders the rehabilitation of patients and easily leads to various types of postoperative complications. Acupuncture-combined anesthesia (ACA) is the combination of traditional acupuncture and modern anesthesia, which means acupuncture is applied at acupoints with general anesthesia. It was testified that ACA strengthened the analgesic effect and reduced the occurrence of postoperative pain, but its mechanism was not clear. Numerous reports have shown that chemokine receptor CX3CR1 is involved in the development and progression of many pathological pains. The present study was aimed to reveal whether ACA played the analgesic roles in the post-incision pain by affecting CX3CR1. A model of toe incision pain was established in C57BL/6J mice. The pain threshold was detected by behavioral test, and the expression of CX3CR1 protein was detected by immunohistochemical method and Western blot. The results showed that the significant mechanical allodynia and thermal hyperalgesia were induced by paw incision in the mice. Mechanical allodynia was significantly suppressed by ACA, but thermal hyperalgesia was not changed. CX3CR1 was mainly expressed in microglia in the spinal cord dorsal horn, and its protein level was significantly increased at 3 d after incision compared with that of naïve C57BL/6J mice. ACA did not affect CX3CR1 protein expression at 3 d after incision in the toe incision model mice. Paw withdrawal threshold was significantly increased at 3 d after incision in CX3CR1 knockout (KO) mice compared with that in the C57BL/6J mice. But the analgesic effect of ACA was disappeared in CX3CR1 KO mice. Accordingly, it was also blocked when neutralizing antibody of CX3CR1 was intrathecally injected (i.t.) 1 h before ACA in the C57BL/6J mice. These results suggest that CX3CR1 in microglia is involved in post-incision pain and analgesia of ACA.

Effects of gap sizes (50, 100 and 150 m2) on seasonal dynamics of soil organic carbon (SOC) and microbial biomass carbon (MBC) in soil surface (0-15 cm) and subsurface (15-30 cm) were investigated in a 50-year old Picea asperata plantation in Miyaluo forest, Western Si- chuan, China. In the four seasons, the SOC and MBC contents were higher in the soil surface than in soil subsurface in the four treatments, and varied insignificantly in different seasons. Compared with the control, the SOC content increased by 35.4%, 21.2% and 10.3% in soil surface, and by 45.5%, 25.0% and 12.1% in soil subsurface with the forest gaps of 50, 100 and 150 m2, respectively. The MBC content increased by 26.7%, 16.7%, 11.3% and 24.4%, 12.6%, 7.3% in soil surface and subsurface, respectively. There were significant negative relationship between soil SOC content and the soil pH and moisture, and significant positive relationship between soil SOC content and soil temperature. Significant effects of SOC content, soil pH, soil temperature, soil moisture on MBC content were observed, suggesting that forest gaps could significantly affect envi- ronmental conditions, and increasing gaps could reduce microbial activity and decomposition rate and decrease soil SOC and MBC contents.

Fine root in forest ecosystems plays an important role in global C cycle. In this study, a measurement was made on the fine root biomass and carbon storage in the surface soil (0-30 cm) of a 31 year-old Cinnamomum camphora plantation in the Rainy Area of West China in November, 2010-December, 2011. The total biomass and carbon storage of the fine roots (living and dead) in the surface soil were 1592.29 kg x hm(-2) and 660.68 kg C x hm(-2), in which, living fine roots accounted for 91.1% and 91.8% respectively. The total biomass and carbon storage of the first five order living roots and dead roots decreased significantly with increasing soil depth, and the living root biomass and carbon storage increased significantly with root order. The sum of the biomass and carbon storage of living and dead fine roots was the largest in autumn and the smallest in winter, but the biomass and carbon storage of the dead fine roots were the largest in winter and the smallest in summer. The biomass and carbon storage of the first two order roots were the largest in summer and the smallest in winter, while those of the last three order roots were the largest in autumn and the smallest in winter. The spatial heterogeneity of soil moisture and nutrients was the main factor affecting the fine root biomass and carbon storage.

Objective: To determine the duration of the resection of pulmonary lobe (RPL) and the effect of video-assisted thoracoscopic surgery (VATS) procedure on the duration of RPL., Methods: The records of 126 patients who had VATS lobectomy from October 2009 to October 2010 with a single-direction thoracoscopic procedure in one hospital were reviewed., Results: The patients included 65 (52%) men and 61 (48%) women with a mean age of 56 years (range, 15 years to 82 years). Four cases were excluded because of intraoperative complication. The average length of operation of RPL in the 122 patients without intraoperative complications was 38.98 min +/- 13.243 min (range, 17 min to 100 min), and the average length of the entire operation was 153.36 min +/- 39.64 min (range, 70 min to 245 min). Regardless which lobes, the average length of a single RPL remained the same (P > 0.05). The RPL of bilobectomies took longer than a single lobectomy (P < 0.05). However, no significant differences in the length of the entire operations were found between bilobectomy and single lobectomy (P > 0.05)., Conclusion: The length of RPL reflects the difficulties of the core step of thoracoscopic lobectomy. Single-direction thoracoscopic lobectomy is an easy procedure to manipulate.

Objective: To compare acute inflammatory responses and immunosuppression to lobectomy in lung cancer patients with video-assisted thoracoscopic surgery (VATS) and posterolateral thoracotomy (PLT)., Methods: A total of 103 patients who underwent either a VATS (n = 51) or a PLT (n = 52) lobectomy for early non-small cell lung cancers (NSCLC, stage I ) were recruited for this study. Blood samples of the participants were taken preoperatively and at 24 h and 72 h post-operatively for analyses of C-reactive protein (CRP), interleukin (IL)-6, IL-2 receptors (IL-2R), and serum amyloid A (SAA). Blood samples taken pre-operatively and at 2 d and 7 d post operations were also analyzed for total lymphocytes, NK cells, CD4+ T, and CD8+ T., Results: Patients in the VATS group lost significantly less blood than those in the PLT group (P = 0.001). Patients in the PLT group had significantly higher serum SAA than those in the VATS group (P = 0. 006). Significant reduction of CD8+ T was found in the patients with PLT after operations (P < 0.01). Patients in the PLT group had significantly lower at CD8+ T 7 d post operations than those in the VATS group (P = 0.015)., Conclusion: VATS pulmonary lobectomy is associated with reduced acute inflammatory responses and immunosuppression compared with the PLT approach.

Objective: To modify and improve the procedures of video-assisted thoracoscopic surgery (VATS) bronchial sleeve lobectomy for lung cancers., Methods: From December 2010 to July 2012, 11 patients with nonsmall cell lung cancers underwent VATS bronchial sleeve lobectomy in our department, which included 7 cases of right upper lobectomy, 3 cases of left upper lobectomy and 1 case of left lower lobectomy. We modified the surgical procedures in relation to the distribution of VATS ports, the techniques of dissecting hilar structures and the method of bronchial reconstruction. The position of incisions for the left side differed from the right side. The lobe was resected with the technique of "hollow out" and the bronchus was reconstructed using a running prolene stitch., Results: Blood loss of these 11 patients ranged from 50-400 (median 200) mL. The operations were completed within 200-320 (median 235) minutes and the duration needed for bronchial reconstruction ranged from 35 to 60 (median 50) minutes. A total of 8-28 (median 15) lymph nodes were dissected. There was no conversion to thoracotomy or blood transfusion. Chest drainage tubes were removed within 2-12 (median 3) days after surgery. The patency of bronchial anastomosis was confirmed by bronchoscopy. Postoperative complications occurred in three patients, which included one case of bronchial pleural fistula (BPF) and two cases of pneumonia. The patient suffered from BPF died 49 days after surgery from intrabronchial bleeding. The postoperative hospital stay of those patients ranged from 8 to 49 (median 8) days. The reconstructed bronchus continued to work well during the three month follow-up visits., Conclusion: VATS bronchial sleeve lobectomy is feasible and safe. The modifications of the surgical procedures may promote the use of this complex operation.

Objective: To evaluate the safety and feasibility of video-assisted thoracic surgery (VATS) anatomic segmentectomy for pulmonary diseases., Methods: Between November 2009 and July 2011, 20 patients received consecutive VATS anatomic segmentectomy by single surgical group. There were 3 male and 17 female, aging from 32 to 81 years with a mean of 53 years. The patients included 12 cases of non-small cell lung cancer (NSCLC) (5 cases of bronchioloalveolar carcinoma, 6 cases of adenocarcinoma, and 1 case of squamous cell carcinoma), 7 cases of benign diseases (3 cases of bronchiectasis, 2 cases of inflammatory pseudotumor, 1 case of tuberculosis, and 1 case of sclerosing hemangioma) and 1 case of metastasis tumor. The locations of resected segments included 1 anterior segment, 3 posterior segments, 4 apical segments, and 2 superior segments in the right side; and 5 lingular segments, 3 trisegments, and 2 superior segments in the left side. Simultaneously, 3 patients with bronchiectasis underwent segmentectomy and lobectomy, 1 of 6 patients with adenocarcinoma underwent lingulectomy and thymectomy. The pathological TNM stages of 12 NSCLC patients were 9 cases of T1aN0M0, 1 case of T1bN0M0 and 2 cases of T2aN0M0., Results: Of these 20 patients, the median operative time was 155 minutes (range, 120 to 235 minutes), the median blood loss was 50 ml (range, 10 to 600 ml), the median drainage duration was 3 d (range, 1 to 6 d), and the median hospital stay was 6 d (range, 3 to 9 d). One patient who had undergone lingulectomy had a 600 ml intraoperative bleeding from lingular artery, and the bleeding was controlled by suturing the rupture under VATS. Bloody sputum occurred in 2 patients, prolonged air leak occurred in one patient for 5 days, and one patient developed subcutaneous emphysema that spontaneously resolved. No mortality was observed for 30 days after the surgery., Conclusions: VATS anatomic segmentectomy is a feasible and safe technique with acceptable operative time, less blood loss, fewer complications, and shorter hospital stay.

Objective: To investigation the clinical characteristics in myasthenia gravis (MG) patients with thymomas., Methods: A total of 856 MG patients admitted to the department during 2008.7 - 2010.12 were reviewed retrospectively. The patients with MG were divided into two groups based on thymic pathology, which were 162 cases with thymoma and 694 cases without thymoma. We compared the different clinical features including the gender, age of onset, MG symptoms and the incidence rate of myasthenia crisis. And the relationship between the WHO types, Maosaoka stages of thymoma and the severe of MG was also studied., Results: The percentage of thymoma-associated MG patients was 18.9 percent of hospitalized MG patients at the same period. Of the 162 thymoma-associated patients, 94 were male and 68 were female, with a ratio of 1.38:1 and a mean age of (42.9 ± 12.4) years old. Thymoma was more frequent in middle-old aged patients than in children. Compared with non-thymoma MG, more thymomatous patients showed generalised MG, but not only ocular muscles weakness (90.1% vs 62.4%, P < 0.001). There were significant differences of the incidence rate of myasthenic crisis in the two groups (14.8% vs 2.3%). (2) WHO type B2 and Maosaoka I, II thymoma were the commonest types among all potentially MG-associated thymoma. No differences of Osserman MG classification was found in thymomatous patients with different pathologic changes., Conclusions: The thymomatous MG patients had its distinctive clinical features: thymomas occurred in about 19.7% of MG patients with more men than women, more common in generalized, higher incidence of myasthenia crisis, with B2 type thymic pathology and Maosaoka I, II stages. No correlation was found between pathologic and clinical stages.

Objective: To investigate the stability of estazolam in biological samples preserved in formaldehyde solution., Methods: The dog was given intragastric administration of estazolam with a dose of 37.6 mg/kg and killed 2 h later. Heart, liver, kidney and brain of the dog were cut up into 1 g and preserved in 4% formaldehyde solution respectively. The content of estazolam in biological samples and formaldehyde solution were analyzed by HPLC at different times., Results: The content of estazolam in heart, liver, kidney and brain or in formaldehyde solution reduced gradually followed with the extention of preservation time. At the 63rd day, estazolam content in four tissues were 0.8%, 1.7%, 1.0% and 2.2% of the original content respectively., Conclusion: Estazolam in tissues can diffuse into formaldehyde solution and decomposed quickly, so biological samples contained estazolam should not be preserved in formaldehyde solution.

Objective: To study on the decomposition kinetics of bupivacaine in brain, blood and urine, which were collected from dogs executed by bupivacaine and stored in different conditions., Methods: Dogs were given arachnoid cavity anesthesia with bupivacaine. Then the brain, blood and urine were collected and divided equally to three groups stored in 20, 4 and -20 degrees C respectively. The concentrations of bupivacaine at different days were determined by the GC. The equation and half-time period of decomposition kinetics were imitated and calculated with WinNolin program., Results: The decomposition kinetics of bupivacaine in the dogs' brain, blood and urine were fit to the first order kinetics. The common equation was lgC = lgCo-kt/2.303 and k was the decomposition constant of first order reaction., Conclusion: Bupivacaine in the brain, blood and urine specimens were found to be decomposed at various environments for storage. The higher temperature for storage, the faster of decomposition reaction.

Objective: To analyze the learning curve of single-direction complete video-assisted thoracoscopic surgery (cVATS) for lung cancer., Methods: From May 2006 to April 2009, 125 cases of cVATS for lung cancer were performed by two dedicated surgeons. Clinical data were collected prospectively and analyzed retrospectively. The patients operated by different surgeon were divided into 2 groups (group A, n = 24; group B, n = 101), and group B was further divided sequentially into 4 subgroups (B1, B2, B3 and B4) by the number of patients. The patients in group A and B were operated by the surgeons with 2-year and 5-year experience of VATS respectively. The operating time, blood loss, number of resected lymph nodes (NLN), rate of thoracotomy conversion (RTC) and postoperative complications (POC) were compared., Results: Compared with group B, the operating time of group A was significantly prolonged [(237 ± 85) min vs. (187 ± 43) min, P = 0.013], but there were no significant differences in blood loss, NLN, RTC and POC. Comparing group A with B1, the same results were got. From group B1 to B4, the operating time was gradually reduced and blood loss decreased, but the difference was not statistically significant. And in group B, there was a significant reduction of blood loss for the last 51 cases compared to the first 50 cases [(122 ± 141) ml vs. (87 ± 81) ml, P = 0.009]., Conclusions: At the early stage of cVATS resection of lung cancer, the duration of operation was longer, which it was more significant for the surgeons with short carrier of thoracoscopic experience. But the morbidity of operation related complications did not increase. The indicator of proficiency in this operation is achievement of 50 cases of complete thoracoscopic resection of lung cancer.

Objectives: To observe the clinical outcome of invasive thymoma, and analyze how the surgical methods, Masaoka staging, adjuvant radiotherapy and/or chemotherapy affect the prognosis., Methods: The clinical data of 59 surgical patients with invasive thymoma and conducted follow-up from January 2000 to December 2009 was analyzed retrospectively. There were 34 male and 25 female, aged from 18 to 72 years with a mean age of 49 years. Forty-four cases underwent radical resection while the other 15 cases underwent palliative resection or biopsy. Masaoka staging: 18 cases with stage II, 30 cases with stage III, 11 cases with stage IV. Patients with stage II didn't undergo further adjuvant radiotherapy or chemotherapy after surgery. Among the patients with stage III and stage IV, 26 patients received adjuvant radiotherapy and/or chemotherapy after surgery, while the other 15 patients did not receive any further therapy. The relationship between the prognosis and the different surgical methods, Masaoka staging, adjuvant radiotherapy and or chemotherapy was evaluated., Results: Fifty-nine patients had been followed up for 1 to 111 months with an average of 54 months. Three cases were lost with the rate of 6.1%. Nineteen patients suffered local recurrence or systemic metastasis, and 14 of them died. The 3-year and 5-year survival rates were 86.8% and 70.8% respectively. Univariate analysis indicated that patients with early Masaoka staging and who received radical resection, adjuvant radiotherapy and/or chemotherapy after surgery had better survival (P < 0.05). Multivariate analysis indicated that radical resection, adjuvant radiotherapy and or chemotherapy were the most significant prognostic factors which could remarkably improve the survival of patients (P < 0.05). For patients with resectable recurrence, reoperation could also improve survival., Conclusions: The Masaoka staging is related to the prognosis of patients with invasive thymoma. Radical resection, adjuvant radiotherapy, chemotherapy can significantly improve the survival of patients with invasive thymoma. Reoperation can improve the survival of some patients with recurrence.

Objective: To explore the feasibility and safety of single-direction video-assisted thoracoscopic lobectomy in the treatment of benign pulmonary diseases., Methods: From May 2006 to Dec 2009, 60 patients with benign pulmonary diseases were treated by single-direction complete video-assisted thoracoscopic lobectomy. Nine cases were bronchiectasis, 7 bronchial cyst, 7 tuberculoma, 11 inflammatory pseudotumor, 6 sclerosing hemangioma, 4 hamartoma, 2 atelectasis with bronchial stenosis, 2 pulmonary aspergillosis, 2 pulmonary sequestration, 8 multiple bullae and 2 cases of mid-lobe syndrome. In the 60 patients, there were 18 cases received right upper lobectomy, and right middle lobectomy in 11 cases, right lower lobectomy in 7 cases, left upper lobectomy in 6 cases, left lower lobectomy in 18 cases., Results: There was one conversion to open surgery. One patient died of postoperative pneumonia. The average operation time was 169 min (range, 70-265 min), average blood loss was 125 mL (range, 10-70 mL), average drainage was 860 mL (range, 20-3500 mL) and average duration of drainage was 4 days (range, 2-16 days). The average postoperative hospital stay was 7.9 days (range, 4-19 days)., Conclusion: Single-direction complete video-assisted thoracoscopic lobectomy is feasible and safe in the treatment of benign pulmonary diseases.

Objective: To establish the models of postmortem redistribution(PMR) in dogs with epidural anesthesia and to investigate the effect of temperature on the PMR of Bupivacaine., Methods: Eighteen male dogs were executed by epidural anesthesia with a dose of 5 mg/kg bupivacaine hydrochloride and randomly divided into three groups, room temperature (20-23 degrees C) group, 4 degrees C group and -20 degrees C group. The cardiac blood, peripheral blood, liver and cerebrum were collected at 0, 2, 4, 8, 24, 48, 72, 96, 120h postmortem. The contents of bupivacaine in those samples were analyzed by GC-NPD and GC-MS, the difference among three groups were compared., Results: The bupivacaine PMR of room temperature group was evident and complex in cardiac blood, peripheral blood and cerebrum. The PMR of 4 degrees C group was weaker and slower than that of normal temperature group. The bupivacaine PMR of the -20 degrees C group was the weakest in three groups., Conclusion: PMR of bupivacaine will happen in epidural anesthesia death dogs, but it could be delayed or prevent by low temperature storage.

Objective: The stability of ketamine in biological samples was studied under different storage temperature and time., Methods: The rabbits were given intragastric administration of ketamine with a dose of 150 mg/kg and were sacrificed after 30 minutes. Blood, liver, kidney and brain of the rabbits were stored at room temperature (between 18 degrees C and 24 degrees C) and -20 degrees C. The specimens were analyzed at different times by GC-MS and GC-NPD., Results: At -20 degrees C, the concentration of ketamine decreased in all of samples (P < 0.05) within 30 days. The concentration of ketamine increased in all of samples stored at room temperature after 5 days(P < 0.05)., Conclusion: The stability of ketamine in biological samples stored at -20 degrees C was better than that at room temperature. The samples suspected containing ketamine should be stored at -20 degrees C and should be tested as soon as possible.

In the present paper, the fluorescence reaction of cationic surface-active agents (CSAA) with Tetrabromofluorescein sodium (TBF) in aqueous solution was investigated. It was found that the fluorescence quenching of TBF appears when it reacts with the cation monomer of a CSAA and a new stronger fluorescence is obtained when the ion-associates react with the micellate of CSAA. The authors investigated the condition of energy transfer between acidic fluorescent dyes in micelles of cetyl trimethyl ammonium bromide or hexadecyl pyridinium bromide. It was indicated that in the micelles formed by cationic surface active agent with dyes embedded (cationic surface active agent and dyes are charge opposite), the effective energy transfer between anionic dyes could occur. When the concentration of cationic surface active agent reached certain value, the energy transfer could occur. With the value of two thirds of critical micelle concentration, the efficiency of energy transfer reached the maximum; when the concentration of cationic surface active agent increased, the efficiency of energy transfer would be decreased. The authors also deduced the model of energy transfer between dyes in micelles and laws of it.

A complex composed of ciprofloxacin and terbium (Tb3+) in the solution of acetic acid-sodium acetate buffer (pH 6.2) was studied by fluorescence spectroscopy and ultra-violet absorption spectroscopy. The emission peak of Tb3+ appeared at 490, 545, 590 nm, (the sensitized fluorescence was enforced mightily) and the intensity of 545 nm emission peak was increased obviously. In its acute emission spectrum, the strongest emission peak of Tb3+ was at 545 nm, which was close to the wavelength of the biggest absorption peak of RB, 552 nm. Therefore, as the basic dye rhodamine B(RB) was added, the fluorescence intensity of 545 nm emission peak decreased regularly, indicating that there was a great quenching process. The result showed that the course was statistic. Based on the mechanism of the Förster energy transfer, the efficiency of energy transfer and the distance between the acceptor RB and the complex were found. Thereby, it was indicated that the course of action was single static quenching and the mechanism of quenching was based on energy transfer.

Objective: To observe the curative effect of integrated Chinese and Western medicine on acute cerebral infarction (ACI)., Methods: Two hundred and seventy-nine ACI patients were assigned to two groups. The control group (140 cases) was treated with Western medicine by staging and the treated group (139 cases) was given TCM therapy according to syndrome differentiation on the basis of Western medicine. The end point was set at the 90th day of the administration. The curative effect was assessed by Chinese stroke scale (CSS), National Institutes of health stroke scale score (NIHSS), Rankin scale and Barthel index (BI)., Results: At the end point of the trial, the total effective rate was 73.38% in the treated group and 61.43% in the control group, the former was superior to the latter (P<0.05). There were 66 cases (47.14%) in the control group and 80 cases (57.55%) in the treated group with improvement rate of NIHSS > or =40%, 65 cases (46.43%) with their Rankin scale within 0-2 grade in the control group and 78 cases (56.12%) in the treated group, 60 cases (42.86%) in the control group and 71 cases (51.08%) in the treated group with BI > or =85, 61 cases (43.57%) in the control group and 72 cases (51.80%) in the treated group with improvement rate of CSS > or = 46%, comparison between them showed significant differences (P < 0.05)., Conclusion: TCM therapy accord-ing to syndrome differentiation combined with Western medicine by staging shows better curative efficacy on ACI.

Based on the records of carboxyhemoglobin in blood samples stored for recent years, the stability of carboxyhemoglobin in these samples could be affected by the containers, the storage temperatures, the volumes of air above the blood, the saturation of the initial carboxyhemoglobin and preservatives added in these blood samples, among which the storage temperatures, the volumes of air above the blood and the saturation of the initial carboxyhemoglobin are the major influence factors.

Objective: To establish a rapid and simple gas chromatographic-mass spectric method for qualitative and quantitative analysis of lidocaine in blood and cerebrospinal fluid(CSF)., Methods: Following an acidification of HCl, blood or CSF was alkalinized with NaOH (pH=9) and extracted with ether for two times. Evaporated in a water bath and with an air velocity of nitrogen gas, extract was dissolved with ethanol and analyzed by a gas chromatographic-mass spectrum method, lidocaine was analyzed qualitatively and quantitatively by GC/MS (SIM:86, 58, 72, 87)., Results: Linear range of lidocaine detected in blood or CSF by this method is 1.0-60.0 microg x mL(-1) (r=0.9999), the minimum detected concentration of lidocaine was 0.02 microg x mL(-1) (S/N=3), recovery is at 85%-103%. This method was used in the determination of lidocaine in dog model died of the anesthesia with lidocaine., Conclusion: This study provided a gas chromatographic-mass spectric analysis for lidocaine in blood and CSF. This method was more selective, little interferefering, more sensitivities and simpler. It could be used in the detection of lidocaine in biological fluids.

We study the polymorphism at DYS605 ,a new tetranucleotide Y-STR locus,in a Chinese Han population of Shanxi to meet the need of more genetic markers in forensic practice and genetic analysis. DNA were extracted from 128 unrelated male venous blood, and amplified using GDB primers. PCR products were detected using non-denaturing polyacrylamide gel electrophoresis and silver staining. Five alleles named 22,21,20,19,18 were observed with frequency of 0.0156;0.1797;0.4531;0.2891;0.0625. PCR products were not found in female DNA. Using a long enough gel for a long electrophoresis time is strongly encouraged because the rung between allele 20 and allele19 is smaller than expected. Allele sequences show that the repetitive units of DYS605 were composed with the variant units and non-variant units.

In order to provide the evidence for the application of DYS413(YCAIII) locus in forensic medicine and population genetics by investigating its polymorphism in the Mongolians lived in the Inner Mongolia Autonomous Region, China, to collect 120 unrelated male Mongolians' venous blood, who lived in the Inner Mongolia Autonomous Region, and the blood is anticoagulant with EDTA, then to extract DNA by using the method of phenol-chloroform and amplify the DYS413 locus with PCR. The products were separated with 6% non-denatured polyacrylamide/bisacrylamide gels(19:1) electrophoresis and stained with 1 per thousand nitrate of silver(AgNO(3)). There are total 30 different haplotypes in Mongolians, the frequencies are from 0.0007 to 0.01361, the HD(haplotype diversity) is 0.94054. It shows that DYS413 locus is high valuable in the application of forensic medicine and population genetics.
