Genetic markers in forensic DNA typing experienced the variable number of tandem repeats （VNTR） sequences and the short tandem repeats （STR） sequences. With the emerge of sequencing technology, the third generation of genetic markers were found out, which usually have two alleles including single nucleotide polymorphism （SNP） and insertion/deletion （InDel）, also known as biallelic genetic markers. Because of the insertions or deletions of DNA fragments, InDel genetic marker reveals DNA fragment length polymorphism and widely distributes across the whole genome. InDel genetic marker is numerous and has the characteristics of STR and SNP genetic markers, which has been applied in the fields of genetics and anthropology. This review focuses on the research progress of InDel genetic marker in forensic science, aiming to review and summarize the main research findings in recent years and provide clues for future researches.

To address the issues of insufficient receptive fields for extracting low-level features and the lack of reinforcement for local key features in lightweight networks, this paper proposed a multistage feature distillation-weighted lightweight image super-resolution network LMSWN. Firstly, a pyramid- like module is employed to expand the receptive field during shallow feature extraction, integrate feature information of different scales, and enrich the information flow of the network. Secondly, a multi-stage residual distillation-weighted module is designed to enhance the ability of square convolution to extract local key features, recover more detailed information, and improve reconstruction performance. At the same time, the combination of channel separation and 1x1 convolution realizes gradual distillation of features, reducing the number of network parameters. Finally, two adaptive parameters are introduced to jointly learn the features of the two branches of the multi-stage residual distillationweighted module, enhancing the attention to different levels of feature information and further enhancing the representation ability of the network. Aiming at the inaccurate phoneme segmentation problem caused by the lack of consideration of Lao language tone changes and audio diversity in existing methods, this paper proposes an unsupervised phoneme segmentation method for Lao language with multi-feature interaction fusion. Firstly, self-supervised features, spectral features and pitch features are independently coded to avoid the insufficiency of a single feature. Secondly, multiple independent features are gradually fused based on the attention mechanism, so that the model can more comprehensively capture the information of Lao language tone changes and phoneme boundaries. Finally, a learnable framework is adopted to optimize the phoneme segmentation model. The experimental results show that the proposed method improves the R-value by 27.88% on the Lao phoneme segmentation task compared with the baseline methods. [ABSTRACT FROM AUTHOR]

Text normalization (TN) is an indispensable work in the front-end analysis task of speech synthesis text. Lao text normalization is to convert non-standard words (NSW) in Lao text into spoken-form words (SFW). At present, the task of text normalization has not yet been carried out in Lao, which mainly faces the problems of difficult acquisition of training data, diversified language expression and text regularization with ambiguity. A text normalization task in Lao is carried out. This ask is completed as a sequence tagging task, and neural networks are used to predict NSW with ambiguity in combination with context. The corpus of the Lao text normalization task is constructed, the model results is predicted through the neural network, the self-attention mechanism is increased to deepen the relationship between the sequence characters, and different strategies are explored to introduce the pre-trained language model. Environmental risk is typically characterized by its spatial distance, while the study on the mechanism and mode of how spatial distance influence risk perception is quite limited. This paper formulated a framework from the background of environmental risk, proposed an axiomatic set of preferences and derived a proposition of probability-spatial distance tradeoff, with a form of exponential function. Finally, a spatial discount model was built to describe the manner of individuals' environmental risk assessment. The model suggests that individuals psychologically map a remote risky event into a proximate one, while the spatial distance results in a discounting effect on the estimation of the seriousness of impact as well as the possibility of its occurrence. This model contributes in explicitly formalizing the connection between probability and spatial distance based on a general environmental risk, and providing a useful framework for considering risk in a spatial context. Objective To investigate the factors influencing interbody cage subsidence after stand-alone oblique lumbar interbody fusion (OLIF). Methods A total of 65 patients (72 lumbar vertebrae) who underwent OLIF in our hospital from Ja-nuary 2019 to January 2023 were selected and divided into subsidence group (n=19) and non-subsidence group (n=53) according to the loss of disc height. The clinical data of the two groups were collected and compared, including gender, age, body mass index, number of lumbar vertebrae requiring surgery, responsible vertebral endplate sclerosis, cage height and length, preoperative CT values of the responsible vertebral body and its upper and lower endplates, preoperative multifidus muscle cross-sectional area at responsible vertebral body, degree of multifidus muscle atrophy, and degree of preoperative articular process degeneration. Logistic regression analysis was used to explore the factors influencing cage subsidence. Results There were significant differences between the two groups in the preoperative CT values of responsible vertebral body and its upper endplate, the proportion of patients with preoperative responsible vertebral body endplate sclerosis, the preoperative multifidus muscle cross-sectional area at responsible vertebral body, the proportion of patients with preoperative severe atrophy of the multifidus muscle, the proportion of patients with cage height 14 mm, the proportion of patients with cage length 55 mm, and the proportion of patients with articular process degeneration of degree 3 (t=2.332-3.097,χ2=4.545-6.246,P

Woodblock printing activities of chieftain are worthy of attention in the history of publishing in China. The woodblock printing activities of the Mu (木氏) chieftains of Lijiang (麗江) is a representative case. This paper first clarifies some relevant problems which could not be solved by previous studies. According to the research statistics, there are up to 34 block-printed editions published by chieftain Mu. The woodblock printing activities of chieftain Mu took place during the reign of Six Masters in the Mu Family (木氏六公). The locations where these books were made were not fixed. The character of these editions is Suzhou (蘇州) style appreciated by Chinese traditional literati. Originating from Three Immortal Deeds of Confucianism and Buddhist concept of merits, these books can be divided into Mu’s works, historical materials for family and religious books. Chinese characters and minority writings both were applied in these books, which is different from other private printing. The woodblock printing activities of chieftain Mu not only holds a place in the history of publishing in China, but also is an important sign of cultural identity in Tusi (土司) areas. As a bibliography compiled by the government, Mi Shu Sheng Xu Bian Dao Si Ku Que Shu Mu (秘書省續編到四庫闕書目; hereafter Mi Mu秘目), which of great textual value, has a definite source. But there is almost no attention to this bibliography. The important reason is that the edit of Mi Mu is not perfect, while the precondition of the edit is to decide the lineage of the editions of Mi Mu. There are two known edition systems of Mi Mu, the one is the transmitted edition system, another is the incomplete edition system. The editions which can be seen presently all belong to the transmitted edition system. This article is to study the transmitted edition of Mi Mu. By investigation, there are 12 existing editions of Mi Mu in this system. Through collation and evidential research, we can divide these 12 editions into four sub-systems. These four sub-systems are Ji Rui Lou Ben (稽瑞樓本) edition system, Zhang Fu Chuan Cang Ben (張芙川藏本) edition system, Ni En Fu Ben (倪恩福本) edition system, and Ti Zhang Jin Wu Ben (題張金吾本) edition system. Objective To investigate the genetic polymorphism of 21 autosomal STR loci and DYS391 locus of SiFaSTRTM 23plex DNA ID system in Han population of eastern China and to evaluate its application value in forensic science. Methods Typing test of 2 000 unrelated individuals was performed using SiFaSTRTM 23plex DNA ID system. The population genetic parameters of STR loci were statistically analysed. A total of 3 198 parentage confirmed cases were detected with that system and the mutation conditions were observed in 21 autosomal STR loci. Results All the 21 autosomal STR loci showed no significant departure from Hardy-Weinberg equilibrium （P>0.05）. The Ho ranged from 0.617 5 to 0.927 0. The DP ranged from 0.796 4 to 0.986 9, as well as the PIC distributed from 0.561 1 to 0.912 3. The CDP was 0.999 999 999 999 999. The CPEduo was 0.999 997 431 701 961, while CPEtrio was 0.999 999 999 654 865. Five alleles were detected in DYS391 locus, with the allele frequency from 0.004 0 to 0.729 0, and GD was 0.418 9. Except D13S317 and D10S1248, seventy-six mutation events were observed at the rest nineteen autosomal STR loci. Among them, seventy-five （98.68%） were one step mutation, and only one （1.32%） was three steps mutation. The mutation rate ranged from 0.246 5×10-3 to 2.711 4×10-3, and the averaged mutation rate was 0.892 1×10-3 （95% CI： 0.70×10-3-1.10×10-3）. In 33 trio mutation cases, the proportion of the paternal mutation and the maternal mutation was 2.09∶1. Conclusion The involved STRs are highly polymorphic in Eastern Han population with acceptable mutation rates by the SiFaSTRTM 23plex DNA ID system, which is suitable for paternity testing and individual identification.

Utilization of carbon dioxide (CO 2 ) is a huge challenge for global sustainable development. Biological carbon fixation occurs in nature, but the low energy efficiency and slow speed hamper its commercialization. Physical-chemical carbon fixation is efficient, but relies on high energy consumption and often generates unwanted by-products. Combining the advantages of biological, physical and chemical technologies for efficient utilization of CO 2 remains to be an urgent scientific and technological challenge to be addressed. Here, based on the development of Tianjin Institute of Industrial Biotechnology, Chinese Academy of Sciences in the past decade, we summarize the important progress in the design and construction of functional parts, pathways and systems for artificial bioconversion of carbon dioxide, including the breakthrough on the artificial synthesis of starch from CO 2 . Moreover, we prospect how to further develop the technologies for artificial bioconversion of carbon dioxide. These progress and perspectives provide new insight for achieving the goal of "carbon peaking and carbon neutrality".

Based on the land use data of Hexi Corridor region from 1980 to 2020, we calculated the ecological environment quality index and analyzed the spatial and temporal evolution characteristics, change frequency, patterns, circles of the long time series land use type changes and its ecological environment quality, using the spatial analysis method of ArcGIS. The results showed that unused land was the main land use type in the study area during 1980-2020. The area of construction land, forest land, grassland, and water changed more, and the transition among land use types was obvious. The trends of deterioration and improvement of ecological environment quality in the study area occurred simultaneously and offset each other under certain conditions. The ecological environment quality index evolved in a 'U' shape, first decreasing and then increasing, with little change in overall ecological environment quality fluctuations. The spatial clustering of ecological quality was obvious, which was higher in the southeast and lower in the northwest, without high frequency and large change. The ecological quality in the southeast quadrant of the study area was the worst, followed by the southwest and northwest quadrants, while the ecological quality in the northeast quadrant was the best. The area of the continuously decline zone in ecological environment quality type conversion was larger than that of the continuously rising zone, indicating that ecological environmental protection in the Hexi Corridor had been effective during the study period. The hot spots of ecological quality change were mainly distributed in the central and southwestern part of the Hexi Corridor, and the cold spots were mainly distributed in the northwestern part.

The remediation of metalliferous mine tailings remains a challenge in many regions of the world. A field experiment was conducted on representative Pb-Zn mine tailings with different species richness (1-, 4-, 8-, and 16-species) to evaluate the potential roles of species diversity in the phytoremediation of metalliferous mine tailings. The main results were ① high species diversity greatly enhanced vegetation cover and biomass. For example, the average vegetation cover and biomass were 33.4% and 66.7 g ·m -2 in 1-species plots and reached 78.4% and 183.8 g ·m -2 in 16-species plots, respectively. ② Plant species diversity had significant effects on nutrient accumulation. Total organic carbon (TOC), water organic carbon (SOC), total nitrogen (TN) and total phosphorus (TP) significantly increased with the species diversity (TOC: r =0.30, P <0.001; SOC: r =0.20, P <0.05; TN: r =0.24, P <0.05; TP: r =0.20, P <0.05). ③ Species diversity not only reduced the concentration of diethylenetriamine pentaacetate (DTPA)-extracted metals in the mine tailings, but also decreased heavy metal transfer and accumulation in the soil-plant system. With enhanced species diversity, DTPA-extracted Cd, Cu, Pb, and Zn decreased significantly (DTPA-Cd: r =0.20, P <0.05; DTPA-Cu: r =0.19, P <0.05; DTPA-Pb: r =0.23, P <0.05; DTPA-Zn: r =0.23, P <0.05). With increasing species diversity, a slightly decreasing trend was observed for Cd, Cu, Pb, and Zn concentrations in the aboveground parts of plants (Cd: r =-0.16, P <0.01; Cu: r =-0.23, P <0.001; Pb: r =-0.15, P <0.05; Zn: r =-0.18, P <0.001). In conclusion, plant diversity can play an important role in mine reclamation.

Prunus mume &quot;Langmei&quot; is a relict tree species, which fruit has good medicinal value. To understand the biosynthesis pathway of citric acid, the Illumina HiSeq XTen high-throughput sequencing technology was used to get the transcriptome from &quot;Langmei&quot;. A total of 38 936 unigenes were obtained by assembling the fruit transcripts, of which 28 311 unigenes were successfully annotated in public databases, 15 193 unigenes were mapped to 265 KEGG metabolic pathways, and 18 908 unigenes were classified into 59 GO functional subclasses, 103 unigenes encoding 15 key enzymes involved in citric acid synthesis pathway were identified and analyzed. The structural model of citrate synthetase in &quot;Langmei&quot; showed that it was a homodimer and the secondary structure of each monomer was mainly composed of alpha helixes. Moreover, the residues in the active site of the citrate synthetase were highly conserved. This study provides a valuable resource for identifying candidate genes involved in the citric acid biosynthesis pathway, and will promote the development and sustainable utilization of genetic resources of &quot;Langmei&quot;.

Objective: To observe the effect of wrist-ankle acupuncture (WA) stimulation at "R 4 "- "R 5 " - "R 6 " on the expression of glutamate (Glu) and phosphorylated protein NMDAR1(p-NMDAR1) of the spinal dorsal horn in spared nerve injury (SNI) rats, so as to explore its mechanism underlying improvement of SNI., Methods: A total of 36 SD rats were randomly divi-ded into sham operation, model and WA groups, with 12 rats in each group. The SNI procedure comprised an axotomy and ligation of the tibial and common peroneal nerves leaving the sural nerve intact. Rats of the WA group were treated by acupuncture at "R 4 "-"R 5 "-"R 6 " points from the 5 th day to the 14 th day after modeling. The mechanical pain thresholds were measured before and 5, 10 and 14 d after SNI, respectively. The cold allodynia was dectected by Acetone solution dropped onto the lateral plantar surface of the paw. Glu content and p-NMDAR1 expression of spinal dorsal horn were detected by 1 H-MRS, ELISA and immunohistochemistry Methods., Results: Compared with the sham operation group, the mechanical pain threshold of the model group was significantly decreased ( P <0.01), the duration of cold stimulation foot contraction was increased ( P <0.01), and the Glu content and p-NMDAR1 expression in the spinal dorsal horn were significantly increased ( P <0.05, P <0.01). After WA intervention, the mechanical pain threshold was significantly increased ( P <0.01), the duration of cold stimulation was significantly shortened ( P <0.01), and Glu content and p-NMDAR1 protein expression of spinal dorsal horn were decreased significantly ( P <0.05, P <0.01) in the WA group compared with the model group., Conclusion: WA can reduce pain sensitivity in rats with neuropathic pain, possibly by inhibiting the expression of Glu and p-NMDAR1 in the spinal dorsal horn.

Steroidal saponins, which are the characteristic and main active constituents of Polygonatum, exhibit a broad range of pharmacological functions, such as regulating blood sugar, preventing cardiovascular and cerebrovascular diseases and anti-tumor. In this study, we performed RNA sequencing(RNA-Seq) analysis for the flowers, leaves, roots, and rhizomes of Polygonatum cyrtonema using the BGISEQ-500 platform to understand the biosynthesis pathway of steroidal saponins and study their key enzyme genes. The assembly of transcripts for four tissues generated 129 989 unigenes, of which 88 958 were mapped to several public databases for functional annotation, 22 813 unigenes were assigned to 53 subcategories and 64 877 unigenes were annotated to 136 pathways in KEGG database. Furthermore, 502 unigenes involved in the biosynthesis pathway of steroidal saponins were identified, of which 97 unigenes encoding 12 key enzymes. Cycloartenol synthase, the first key enzyme in the pathway of phytosterol biosynthesis, showed conserved catalytic domain and substrate binding domain based on sequence analysis and homology modeling. Differentially expressed genes(DEGs) were identified in rhizomes as compared to other tissues(flowers, leaves or roots).The 2 437 unigenes annotated by KEGG showed rhizome-specific expression, of which 35 unigenes involved in the biosynthesis of steroidal saponins. Our results greatly extend the public transcriptome dataset of Polygonatum and provide valuable information for the identification of candidate genes involved in the biosynthesis of steroidal saponins and other important secondary metabolites.

A rapid analysis method based on ultraviolet-visual(UV-Vis) spectroscopy, near infrared(NIR) spectroscopy and multivariable data analysis was established for quality evaluation of Shengxuebao Mixture. The contents of eight active ingredients of Shengxuebao Mixture including albiflorin, paeoniflorin, 2, 3, 5, 4'-tetra-hydroxy-stilbene-2-O-β-D-glucopyranoside, specnuezhenide,ecliptasaponin D, emodin, calycosin-7-glucoside and astragaloside Ⅳ were simultaneously detected by using this method. HPLC-UV-MS was used as a reference method for determining the contents of these ingredients. Partial least squares(PLS) analysis was implemented as a linear method for multivariate models calibrated between UV spectrum/NIR spectrum and contents of 8 ingredients. Finally, the performance of the model was evaluated by 24 batches of test samples. The results showed that both UV-Vis and NIR models gave a good calibration ability with an R~2 value above 0.9, and the prediction ability was also satisfactory, with an R~2 value higher than 0.83 for UV-Vis model and higher than 0.79 for NIR model. The overall results demonstrate that the established method is accurate, robust and fast, therefore, it can be used for rapid quality evaluation of Shengxuebao Mixture.

Agkistrodon acutus is a traditional Chinese herb medicine which has immunological regulation,anti-tumor,anti-inflammatory and analgesic effects,which is mainly used for the treatment of rheumatoid arthritis,ankylosing spondylitis,sjogren's syndrome and tumors. In order to excavate more important functional genes from A. acutus,the transcriptome of the venom gland was sequenced by the Illumina Hi Seq 4000,and 32 862 unigenes were assembled. Among them,26 589 unigenes were mapped to least one public database. 2 695 unigenes were annotated and assigned to 62 TF families,and 5 920 SSR loci were identified. The majority of mapped unigenes was from Protobothrops mucrosquamatus in the NR database,which revealed their closest homology. Three secretory phospholipase A&#95;2 with different amino acid sequences showed similar spatial structures and all had well-conserved active sites. The 3 D structural models of C-type lectin showed conserved glycosylation binding sites( Asn45). This study will lay the foundation for the further study of the function of snake venom protein,and promoting the development and utilization of genome resources from A. acutus.

A field trial was conducted with abandoned Pb-Zn mine tailings to evaluate the effectiveness of amendments with different C/N/P ratios on plant growth, soil nutrients and enzyme activities, and heavy metal concentrations in plant tissues and the mine tailings. The following results were noted. ①The application of amendments with different C/N/P ratios promoted plant growth and development. The vegetation cover and biomass increased from 2.0%-20.0% and 9.4-115 g·m -2 at 6 months to 62.5%-98.5% and 389-2358.3 g·m -2 at 30 months, respectively. ②When compared with the control tailings, the mean values of organic carbon, water organic carbon, nitrate nitrogen, and available phosphorus in the treatments with different C/N/P ratios increased 6.0%-93.3%, 1.3%-49.3%, 12.3%-214.7%, and 2.7%-81.3%, respectively. Similarly, the addition of amendments with different C/N/P ratios enhanced the soil enzyme activities of dehydrogenase, β -glucosidase, urease, and phosphatase 0.3-2.8, 0.1-1.4, 0.1-0.6, and 0.1-0.5 times those in the tailings. ③The addition of amendments with different C/N/P ratios decreased the concentrations of diethylenetriamine pentaacetate (DTPA)-extracted Cd, Cu, Pb, and Zn in the mine tailings and the accumulation of Cd, Cu, Pb, and Zn in plant tissues in different degrees. DTPA-Cd, DTPA-Cu, DTPA-Pb, and DTPA-Zn decreased 2.5%-40.2%, 1.4%-25.6%, 1.4%-15.2%, and 0.4%-24.9%, respectively, compared with the control tailings. ④Pearson's correlation coefficients showed that the vegetation cover and biomass were correlated positively with the soil nutrient elements and enzyme activities and negatively with DTPA-extractable metal concentrations. No correlations were observed between the plant metal concentrations and soil DTPA-extractable metal concentrations, nutrient elements, and enzyme activities. Generally, amendments with different C/N/P ratios aided phytostabilization of some types of mine tailings is the preferred option for full remediation of these mine wastelands.

Chalcone synthase( CHS) and chalcone isomerase( CHI) are key enzymes in the biosynthesis pathway of flavonoids. In this study,unigenes for CHS and CHI were screened from the transcriptome database of Arisaema heterophyllum. The open reading frame( ORFs) of chalcone synthase( Ah CHS) and chalcone isomerase( Ah CHI) were cloned from the plant by RT-PCR. The physicochemical properties,expression and structure characteristics of the encoded proteins Ah CHS and Ah CHI were analyzed. The ORFs of Ah CHS and Ah CHI were 1 176,630 bp in length and encoded 392,209 amino acids,respectively. Ah CHS functioned as a symmetric homodimer. The N-terminal helix of one monomer entwined with the corresponding helix of another monomer. Each CHS monomer consisted of two structural domains. In particular,four conserved residues define the active site. The tertiary structure of Ah CHI revealed a novel open-faced β-sandwich fold. A large β-sheet( β4-β11) and a layer of α-helices( α1-α7) comprised the core structure. The residues spanning β4,β5,α4,and α6 in the three-dimensional structure were conserved among CHIs from different species. Notably,these structural elements formed the active site on the protein surface,and the topology of the active-site cleft defined the stereochemistry of the cyclization reaction. The homology comparison showed that Ah CHS had the highest similarity to the CHS of Anthurium andraeanum,while Ah CHI had the highest similarity to the CHI of Paeonia delavayi. This study provided the basis for the functional study of Ah CHS and Ah CHI and the further study on plant flavonoid biosynthesis pathway.

Objective: To study the chemical constituents from Mitrasacme pygmaea., Methods: The compounds were isolated and purified by column chromatography and their structure were identified by NMR and MS,and comparison spectral data with literature., Results: Eleven compounds were isolated and identified as tricin-7-O-β-D-glucopyranoside( 1),massonianoid A( 2),kaempferol( 3),cinnamic acid( 4),quercetin( 5),tiliroside( 6),tricin( 7),β-sitosterol( 8),adenosine( 9),α-tocopherolquinone( 10)and β-daucosterol( 11)., Conclusion: All the compounds are isolated from this genus for the first time.

Objective: To study the chemical constituents from Periploca forrestii., Methods: The constituents were separated by column chromatography and their structures were elucidated by spectroscopic methods., Results: Seven compounds were isolated from Periploca forrestii and identified as wogonin( 1),negletein( 2),vanilline( 3),isovanilline( 4),periplocoside L( 5),β-sitosterol( 6) and β-daucosterol( 7)., Conclusion: Compounds 1 and 2 are obtained from this genus for the first time,and compounds 3 ~ 5 are isolated from this plant for the first time.

A field trial was conducted in an abandoned Pb/Zn mine tailings to evaluate the effectiveness of three industrial wastes [sweet sorghum vinasse (SSV), medicinal herb residues (MHR) and spent mushroom compost (SMC)] as organic amendments on plant growth, soil nutrients and enzyme activities, and heavy metal concentrations in plant tissues and the mine tailings. (1) The main findings were as follows: (1) The mean concentrations of diethylene-triamine-pentaacetic acid (DTPA)-extractable Cd, Cu, Pb and Zn in SSV, MHR and SMC treatments decreased by 24.2%-27.3%, 45.7%-48.3%, 18.0%-20.9% and 10.1%-14.2% as compared to the control tailings. When compared to the control tailings, the mean values of organic matter, ammonium-N and available P in SSV, MHR and SMC treatments increased by 2.27-2.32, 12.4-12.8 and 4.04-4.74 times, respectively. Similarly, the addition of SSV, MHR and SMC significantly enhanced soil enzyme activities (dehydrogenase, beta-glucosidase, urease and phosphatase), being 5.51-6.37, 1.72-1.96, 6.32-6.62 and 2.35-2.62 times higher than those in the control tailings. (2) The application of these wastes promoted seed germination and seedling growth. The vegetation cover reached 84%, 79% and 86% at SSV, MHR and SMC subplots. For Lolium perenne and Cynodon dactylon, the addition of SSV, MHR and SMC led to significant increases in the shoot biomass yields with 4.2-5.6 and 15.7-17.3 times greater than those in the tailings. Moreover, the addition of SSV, MHR and SMC significantly reduced the concentrations of Cd, Cu, Pb and Zn in the shoots of L. perenne and C. dactylon in comparison with the control tailings. (3) Pearson's correlation coefficients showed that the vegetation cover and biomass were positively correlated with soil nutrient elements and enzyme activities. Significant negative correlations were observed between DTPA-extractable metal concentrations and vegetation cover and biomass. The metal concentrations in plants were positively correlated with soil DTPA-extractable metal concentrations and negatively correlated with soil nutrient elements and enzyme activities. The results obtained in this field study confirmed that the three organic-rich industrial wastes could be used as amendments for aided phytostabilization of some types of mine tailings.

The present work is to investigate the chemical constitutions of Hyptis rhomboidea and their antifungal activities. The compounds were isolated by Toyopearl HW-40, Sephadex LH-20, MCI-Gel CHP-20, RP-18, PTLC and silica column chromatographic methods and subjected to evaluate some monomers antifungal activity of eight kinds of plant pathogenic bacteria. Eleven compounds were isolated and identified as ethyl caffeate (1), ursolic acid (2), oleanolic acid (3), vanillactic acid (4), methyl rosmarinate (5), kaempferol 3-O-alpha-L-rhamnopyranosyl-(1 --> 6) -beta-D-glucopyranoside (6), kaempferol 3-O-alpha-L-rhamnopyranosyl-(1 --> 6)-beta-D-glucopyranoside (7), ilexgenin A (8), beta-amyrin (9), kaempferol 3-O-beta-D-glucopyranoside (astrgalin, 10) and cholest-5-ene-3beta, 4beta-diol (11). Compound 1 showed the strongest inhibitory effect on Sclerotinia sclerotiorum with the MIC 16.2 mg x L(-1), and compound 5 showed the strongest inhibitory effect on S. minor and Exserohilum turcicum with MIC 16.2, 8.1 mg x L(-1), respectively. All compounds were isolated from the H. rhomboidea for the first time, and compounds 1 and 5 showed antifungal activity.

Objective: To explore the effect of Wnt signaling suppression on proliferation of non small cell lung cancer to gefitinib, and its related mechanisms., Methods: PC9 and PC9/AB2 cells of both gefitinib sensitive and resistant were treated with different concentrations of gefitinib, and the proliferation index was measured using CCK8 kit. The members of Wnt signaling pathway were detected by Western blot. Dual luciferase reportor gene assay (TOP Flash) was used to document the transcriptional level of β-catenin. β-catenin siRNA was transfected into PC9/AB2 cells to suppress the Wnt signaling transcription, followed by treatment with different concentrations of gefitinib. Western blot was then used to detect the expression of EGFR and its downstream signaling after inhibit the expression of β-catenin., Results: Treating with different concentrations of gefitinib, the resistance of PC9/AB2 cells to gefitinib was significantly increased (P < 0.05). The members of Wnt signaling expressed at higher level in PC9/AB2 cells than in PC9 cells (t = 24.590, P = 0.000). TOP Flash examination showed that the endogenous transcriptional activity of Wnt signaling was higher in PC9/AB2 cell than that in PC9 cell (t = 4.983, P = 0.008). Compared with the negative control group, apoptotic rate and sensitivity to gefitinib significantly increased in interfered group (P < 0.05). The expression of p-ERK1/2 significantly decreased after Wnt signaling suppression, although other proteins showed no significant alterations., Conclusion: Suppressing the activity of Wnt signaling can partly reverse the celluar resistance to gefitinib in non small cell lung cancer.

Objective: To explore the liver disease spectrum in patients with type 2 diabetes (T2DM) and the risk factors of non-alcoholic fatty liver disease (NAFLD)., Methods: From September 2009 to October 2011, 1069 hospitalized patients with T2DM in Department of Endocrinology and Metabolism were involved in the study. The history informations, results of laboratory examination, hepatic ultrasound and hepatic proton magnetic resonance spectrum ((1)H MRS) of all patients were collected to analysis., Results: (1) The detectable rate of raised liver enzymes in T2DM patients was 28.7% (307/1069), composed mainly of NAFLD (39.4%, 121/307). After excluding the factors such as alcoholic abuse, viral hepatitis, the detect rate of raised liver enzymes in T2DM patients was 26.9% (185/688). (2) The detectable rate of fatty liver by ultrasound in T2DM patients was 56.7% (500/882), composed mainly of NAFLD (72.6%, 363/500), and the detectable rate of NAFLD was 58.0% (363/626). (3) The detectable rate of fatty liver by hepatic (1)H MRS was 72.8% (227/312), composed mainly of NAFLD (69.6%, 158/227). The detectable rate of NAFLD was 69.6% (158/227). (4) Of the three methods for diagnosing NAFLD, (1)H MRS had the highest detectable rate, followed by ultrasound, and the hepatic enzymes was the lowest. Set the hepatic (1)H MRS as gold diagnosing standard of NAFLD, the combination of hepatic enzymes and ultrasound increase the sensitivity. The optional cut-off points of ALT were 19.7 U/L (male, ROCAUC = 0.689, P < 0.01) and 17.0 U/L (female, ROCAUC = 0.727, P < 0.01). (5) Logistic stepwise regression analysis showed sex, BMI, hemoglobin, fasting C-peptide and uric acid (OR = 3.803, 1.195, 1.037, 2.896, 1.011, all P < 0.05) were positively correlated with NAFLD, and diabetes duration (OR = 0.948, P < 0.05) was positively correlated with NAFLD independently., Conclusions: The detectable rate of fatty liver was high in T2DM which was composed mainly of NAFLD. High abnormal liver enzymes detectable rate indicated that NAFLD with T2DM are prone to NASH.

Objective: To explore the diagnostic values of endobronchial ultrasound-guided transbronchial needle aspiration (EBUS-TBNA) in patients with hilar and mediastinal tumors., Methods: A total of 837 patients with chest CT or PET/CT confirmed mediastinal/hilar lymphadenopathy and or tumors in (or around) the trachea or bronchi, were evaluated by EBUS-TBNA examination. Pathological study or follow-up visit was carried out in the same period to make a final diagnosis, and therefore to verify the accuracy of EBUS-TBNA., Result: The study punctured 1631 lymph nodes totally, with an average of 1.95 times per case. The 4R group and the 7th group of lymph nodes accounted for 43% and 34%, respectively. The success rate of TBNA was 100%. The diagnosis rates of lung cancer was 94.11%, of which squamous cell carcinoma accounted for 19.22% (89 cases), adenocarcinoma 32.40% (150 cases), small cell carcinoma 23.54% (109 cases), non-small cell carcinoma of unknown histological type 9.50% (44 cases), adenosquamous carcinoma 7.56% (35 cases), and other types 7.78% (36 cases). The diagnosis rate of tuberculosis was 85.50%, and that of sarcoidosis was 55.88%. The overall sensitivity of EBUS-TBNA was 94.02%, specificity 100%, positive predictive value 100%, negative predictive value 61.91%, and the accuracy was 94.56%. Besides mild bleeding in the puncture site, no other complications occurred, and there were no severe complications such as pneumothorax, pneumomediastinum, or major vascular injury., Conclusion: EBUS-TBNA is of high value as a minimally invasive, convenient, and low-risk procedure for the diagnosis of mediastinal and hilar lymphadenopathy and tumors.

Objective: To investigate the ultrastructural features of sputum deposition (SD) and its value in the diagnosis of pulmonary alveolar proteinosis (PAP)., Methods: Seven patients with PAP diagnosed by lung biopsy and cytology were enrolled in this study. The patients consisted of 5 men and 2 women, whose median age was 48 years (range 36 to 73). SD and bronchoalveolar lavage fluid (BALF) sediment were made into ultrathin sections and observed under transmission electron microscope (TEM), respectively. Seven cases of control group composed of 4 men and 3 women whose median age was 49 years (range 39 to 68) including 3 cases of bacterial pneumonia, two cases of COPD and 2 cases of exudative pulmonary tuberculosis. Each SD was made into ultrathin section, and compared with the experimental group., Results: In PAP group, Periodic acid-schiff (PAS) staining was performed on 7 sputum smears and none of them was tested positive for any components with diagnostic interest. Four cases from the 7 paraffin-embed sections of BALF sediment by microscopic examination suggested PAP. Under TEM, BALF sediment showed that many lamellar bodies existed in and outside alveolar epithelial cells, and 5 specimens were consistent with PAP diagnosis. Compared with BALF sediment, SD had apparent degeneration with more myelin phagosomes in the cytoplasm of macrophages, more lamellar bodies in alveolar epithelial cells, and lots of lamellar bodies in the shape of concentric circle in the extracellular spaces. Four from the 7 SD samples were consistent with the diagnosis of PAP. No significant difference was found between SD and BALF in the diagnosis of PAP by electronic examination (P > 0.05). In the 7 cases of control group no drifting osmiophilic lamellar bodies in extracellular space were detected., Conclusions: The osmiophilic lamellar bodies with diagnostic value were found in SD and BALF of patients with PAP. TEM of SD in combination with clinical manifestations and radiologic findings can make a definitive diagnosis of PAP, especially for those patients who have contraindications to lung biopsy and lung lavage.

Heavy metal pollution is a major environmental problem of mine wasteland. Finding out the tolerant plants, which can adapt to the local climate and the soil conditions, is the premise of vegetation restoration. An extensive vegetation survey and sampling were conducted in Huayuan Mn and Pb/Zn mineland, 76 species belonging to 69 genera and 39 families were recorded. The main dominant species and their associated soils were determined for heavy metal concentrations. The results showed that soil Pb, Zn and Cd levels exceeded the threshold levels of Class II of China Environmental Quality Standard for Soils, which suggested minesoils might be polluted by the three elements. The main dominant plants can adapt to the unfavorable edaphic conditions of mineland and were tolerant to heavy metals. There were great variations of metal uptake and accumulation among different plant species. They were classified into three types according to the metal concentrations in the plant shoots and roots: the accumulator, e. g. Camellia oleifera and Dicranopteris dichotoma, absorbed a large amount of heavy metals by the roots and transported to the shoots, which can be used to clean up the soils containing light to moderate toxic metal concentration and with high-value; the root compartment, e. g. Rubus tephrodes, R. corchorifolius, R. chroosepalus, Artemisia princeps and Pteridium aquilinum also absorbed a large amount of heavy metals but held in the roots; and the excluder, e. g. Miscanthus sinensis, Imperata cylindrica, Indocalamus tessellatus and Toddalia asiatica, absorbed less heavy metals than the accumulators. The root compartment and the excluder were more suitable for remediation of the mine wastelands with high heavy metal concentration, low-value and extensive area.

Objective: To investigate the pathogen spectrum constitution of HEV related to acute meningitis/encephalitis syndrome (AMES) and to analyze the genetic characterization of the echovirus 30 (Echo30) isolates., Methods: Cerebrospinal fluid, stool, and/or throat swab specimens from 101 AMES cases were collected for virus isolation with RD and HEp-2 cell lines in Linyi city from April to December, 2010. After typing by neutralization test, the entire VP1 gene of the isolates were amplified by RT-PCR and sequenced. Homologous comparison was carried out and phylogenetic analysis were performed., Results: HEVs were isolated from 47 specimens of 34 patients (4 from cerebrospinal fluid, 18 from throut swab and 25 from stool), with 11 Echo30 isolates identified by microneutralization assay and molecular typing method. In alignment of VP1 sequences with global Echo30 isolates, relative high nucleotide homologies (91.2% - 95.4%) with Taian, Zhangqiu and Jiangsu isolates and high divergences (17.9% -19.6%) with prototype strain were observed. Nucleotide divergences among Linyi isolates were 0 - 10.2%, and 3 lineages were revealed via phylogenetic analysis, reflecting 3 transmission chains of Echo30 co-circulated in Linyi city, 2010., Conclusion: Echo30 was the predominant serotype of AMES cases in Linyi city, 2010. These isolates possessed considerable divergence with Echo30 from other countries.

Aim: To observe the change and the clinical significance of S100β protein level in cerebrospinal fluid and serum from the patients with cerebral hemorrhage (CH)., Methods: ELISA was used to detect the expression of S100β protein in CSF and serum from CH patients control with Inguinal Hernia or great saphenous varix patients. Meanwhile, rabbit CH model at 6 h, 12 h, 24 h, 48 h, 72 h and 96 h ., Results: The levels of CSF S100β protein at acute stage of CH patients increased significantly compared with those at recovery stage of CH patients and control group(P<0.01). The levels of S100β protein in CSF from CH patients increased significantly compared with those in serum (P<0.01).The levels of S100β protein in CSF of rabbit experimental group increased significantly compared with those of sham operation group at different time points(P<0.01)., Conclusion: The level of S100β protein in CSF from CH patients increases. It may be a biomarker as reflecting degree of pathogenetic and predicting outcome in the CH patients.

To investigate the genetic characteristics of poliovirus isolates from environmental sewage surveillance in Shandong province, we collected sewage samples in Jinan and Linyi City. Serotyping and VP1/ 3D sequencing were performed on polioviruses isolated from the concentrated sewage samples, and VP1 mutation and recombination were analyzed. Thirty-two of sewage samples were collected, and polioviruses were detected in 10 of the samples with a positive rate of 31.3%. Eighteen Sabin strains were isolated including three type 1, nine type 2, and six type 3 polioviruses, and the number of nucleotide substitutions in VP1 coding region varied from 0 to 4. Recombination was found in three Sabin 2 and four Sabin 3 polioviruses. Analysis of neurovirulence sites of VP1 revealed that one Sabin 1 vaccine strain had a nucleotide change of A to G at nt 2749, one Sabin 2 strain had a nucleotide change of A to G at nt 2908, three Sabin 2 strains had a nucleotide change of U to C at nt 2909, and all six Sabin 3 strains had a nucleotide change of C to U at nt 2493. Poliovirus vaccine strains could be isolated from environmental sewage with a high rate of gene recombination and back mutation of neuvirulence-associated sites. None of wild-type poliovirus or vaccine-derived poliovirus was detected.

Objective: To study the relationship between liver fat content (LFC) and liver enzymes in individuals with various statuses of glucose metabolism., Methods: A total of 109 subjects including with impaired glucose regulation (IGR) (n = 31), newly diagnosed type 2 diabetes (NT2DM) (n = 31) and normal glucose tolerance (NGT) (n = 47) were recruited. The level of LFC was measured by (1)H magnetic resonance spectroscopy ((1)H-MRS) to study the relationship between liver fat content (LFC) and alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (AKP) and γ-glutamyltransferase (GGT). The receiver operating characteristic curve (ROC) was employed to obtain the optimal cut-off point of ALT to predict the occurrence of nonalcoholic fatty liver disease (NAFLD)., Results: (1) The levels of LFC were progressively raised in NGT, IGR and NT2DM groups respectively [3.83 (2.35 - 7.59)%, 12.82 (8.10 - 21.37)% and 21.99 (11.89 - 34.43)%, P < 0.01]; (2) the subjects were divided into four subgroups by the method of LFC quartile. And quartile subgroups Q1-4 were associated with the increase of LFC. Waist, BMI, systolic blood pressure, triglyceride, total cholesterol, fasting plasma glucose, OGTT 2 h postprandial glucose and HOMA-IR had a rising trend from Q2. But HDL-C showed a declining trend from Q2; (3) ALT and GGT significantly increased from Q3 (P < 0.01) while AST and AKP significantly increased in Q4 (P < 0.01); (4) adjusted by gender, age and body mass index (BMI), LFC was positively correlated with AST (r = 0.329, P < 0.05), ALT (r = 0.454) and GGT (r = 0.378) (All P < 0.01). But it was negatively correlated with AST/ALT (r = -0.364, P < 0.01); (5) the analysis of stepwise regression demonstrated that LFC was a predictor of ALT, AST, GGT and AST/ALT; (6) ALT had a ROC(AUC) of 0.813 (male) and 0.769 (female) (All P < 0.01). The optimal cut-off point of 23.5 U/L (male) and 17.5 U/L (female) might predict the occurrence of NAFLD., Conclusions: Liver enzymes are correlated with LFC even in normal range. The optimal cut-off point of 23.5 U/L (male) and 17.5 U/L (female) might predict the occurrence of NAFLD. The current used ALT upper limit could underestimate the NAFLD.

Objective: To study the expression of integrin beta1 in squamous cell carcinoma (SCC) and explore the relationship between stem cell marker and SCC., Methods: The expressions of integrin beta1 in SCC tissues and SCC cell strain A431 were detected with immunohistochemical methods and cell staining method. The differentiation of SCC cells were induced with all-trans-retinoic acid (ATRA). The changes of integrin beta1 levels before and after induction were detected with RT-PCR., Results: In highly differentiated SCC tissues, integrin beta1 was constantly expressed in the basal-like cells in the edge of tumor; some cells inside arranged as island also showed positive integrin beta1 expression. In poorly differentiated SCC tissues, island-like integrin beta1-positive cells remarkably increased and distributed in a diffuse way. In SCC A431 cells, integrin beta1 was expressed unevenly in tumor cells. After treatment by ATRA, level of integrin beta1 mRNA in A431 cells significantly decreased compared with untreated control (P < 0.05), and the ratios between the intensity values of integrin beta1 to beta-actin were 0.071 +/- 0.025 and 0.029 +/- 0.018 at 24 h and 48 h, respectively, whereas in controls were 0.148 +/- 0.027 and 0.136 +/- 0.011 (P < 0.05)., Conclusions: Integrin beta1 is heterogeneously expressed in both SCC tissues and SCC A431 cells. The expression of Integrin beta1 decreases when the differentiation level of tumor cells increase, indicating that integrin beta1 is closely related with the initiation of SCC and potential cancer stem cells in SCC.

A quantitative real-time PCR assay was developed to measure the proviral load of human T-lymphotropic virus type I (HTLV-I) in peripheral blood. The technology utilizes special primers and Taqman MGB fluorescence probe to measure amplification products from the gag-pro-pol polyprotein gene of HTLV-I. HTLV-I copy number was normalized to the amount of cellular DNA by quantitation of the beta-actin gene, The amplification system was sensitive to detect 5 copy/microL. The standard curve had a good linearity when the quantity for the gene was between 10(3) and 10(7) copy/microL (R2 = 0.999). Good reproducibility was observed in each intra- and inter-assay. We also measured proviral load in peripheral blood in 12 HTLV-I seropositive former blood donors. Proviral load for HTLV-I infected donors ranged from 0.015 to 12.819 copy/cell in WBC with the mean of 3.116 copy/cell.

Objective: To investigate the southern region of Zhejiang hepatitis E virus (HEV) infection., Methods: A cluster sampling strategy was used to sample all blood donors from February to October in 2008 in Wenzhou blood center. Their blood was tested for IgG and IgM antibody against HEV. Reverse transcriptase-polymerase chain reaction(RT-PCR) and sequencing were applied to detect its genotype and sequence homology in HEV IgM-positive specimen., Results: The prevalence of anti-HEV IgG in 3044 cases of blood donors was 33.28%. IgG increased with age. There are certain increase in positive rates between the 20-year-old group and over 40 years of age group from 21.16% to 50.36%. The positive rate of IgM was 0.92%. The ratio of infection among different age group was the highest in the age range from 31 to 40 years and up to 1.90%. IgG and IgM through their negative and positive analysis of samples found in their group with donors age, sex and blood type does not significantly related to each other. Nucleic acids were found in three cases through PCR amplification in all 28 cases of HEV IgM positive samples. The total positive rate was one-thousandth, of which two cases for gene 4, 1 cases of infection for gene 1., Conclusion: The results indicate that there was a certain percentage of HEV virus in voluntary blood donors in south Zhejiang.

Occult hepatitis B virus (HBV) infection status of blood donors in a southern city in China was investigated by immunological assays and nucleic acid testing. Overall, 17 (0.19%, 95% CI: 0.11%-0.30%) of the 9023 HBsAg negative samples were found to be positive for the presence of HBV DNA. "A" epitope sequences were obtained from 14 among them. Mutation(s) in aa124-aa147 existed in 6 (42.9%, 6/14) samples and 4 (66.7%, 4/6)were G145R mutation. Ratio of genotype C in occult donors (10/17) was statistically higher than HBs-positive donors (0/15, P<0.01), which implied that HBV genotype C leaded to occult infection more easily.

The concentrations of 7 heavy metals (Ni, Cu, Cr, Pb, Cd, As, Hg) and organic matters in the surface sediments of Liao River were determined by atomic absorption spectrometry (AAS) and atomic fluorescence spectrometry (AFS), and sediments pollution assessment was carried out using geoaccumulation index. Moreover, the sources of heavy metal contamination were estimated with the principal component analysis (PCA). The results demonstrate that the heavy metal concentrations of Liao River are markedly higher than that of Liao River in 1998 and lay at medium level compared with other rivers in China. The average concentrations (mg/kg) obtained were: Ni 26.5 mg x kg(-1), Cu 37.9 mg x kg(-1), Cr 90.3 mg x kg(-1), Pb 32.9 mg x kg(-1), Cd 0.49 mg x kg(-1), As 12.3 mg x kg(-1), Hg 0.14 mg x kg(-1). The results of geoaccumulation index reveal that sediments of Liao River are uncontaminated with Ni, As, lightly contaminated with Cu, Cr, Pb and moderately contaminated with Cd, Hg. The order of the analyzed heavy metals, arranged from the highest to lowest pollution degree, is as follows Cd > Hg > Cu > Cr > Pb > As > Ni. By estimating the sources of heavy metal contamination with the Principal component analysis (PCA), it was found that the first two components account for 58.74% and 17.18% of the total variance, respectively. The industrial and living wastewater, degradation of organic matter atmospheric precipitation and geochemical changes are the main sources of heavy metal contamination.

Objective: To investigate the impact of erlotinib as a second or third line treatment on the symptoms and quality of life (QOL) in patients with advanced non-small cell lung cancer (NSCLC)., Methods: Fifty patients with stage III b and IV NSCLC, treated previously with at least one regimen of platinum-based chemotherapy, received 150 mg of erlotinib orally, once a day till disease progression. QOL was assessed by European Organization for Research and Treatment of Cancer QLQ-C30 and the lung cancer module (QLQ-LC13). The primary end points for QOL analysis were time to deterioration of three common lung cancer symptoms: cough, dyspnea and pain., Results: Among 47 evaluable cases, there were partial remission (PR) in 18 cases, stable disease (SD) in 21 cases, and progressive disease (PD) in 8 cases. After two cycles of treatment, the mean scores of global QOL and all 5 functioning scales except the cognitive function increased significantly (P < 0.05). Mean scores of major general symptoms, hypodynamia and anorexia, and disease-related symptoms alleviated significantly. Both response rates of five functioning and global QOL were more than 44% after erlotinib treatment. Response rates of major general symptoms and disease-related symptoms varied from 14% to 76%. Patients with complete or partial response likely had improvement in the QOL response (P < 0.05), and the time to major symptom deterioration in those were significantly longer (P < 0.001) than that in patients with stable or even progressive disease., Conclusion: Erlotinib is effective to improve not only survival, but also tumor-related symptoms and quality of life in patients with advanced NSCLC previously treated with cisplatin-contained regimens. The improvement in the quality of life is positively correlated with objective tumor response.

Western blot, capture-PCR, blocking ELISA and synthetic polypeptides were used to systematically study the recognition epitopes on HEV ORF2 of 23 anti-HEV monoclonal antibodies(McAbs) which were previously generated in our laboratory directed against HEV ORF2. Results showed that seven McAbs recognized linear epitopes that located at aa408-458 of HEV ORF2 and 16 conformation-dependent McAbs, most of which recognized the surface epitopes of native HEV, located at aa459-606 of HEV ORF2. The systematical study of the recognition epitopes of anti-HEV McAbs on HEV ORF2 provides important information for the investigation of virus receptor and HEV infection mechanism, as well as its vaccine and diagnostics development.

Objective: To investigate the effect of a novel retinoid CD437 and all-trans retinoic acid (ATRA) in inducing cell apoptosis and inhibiting the proliferation of human epidermoid carcinoma A431 cells and normal human epidermal keratinocytes., Methods: MTT assay was used to determine the inhibitory effects of CD437 and ATRA on the growth of A431 cells and normal human epidermal keratinocytes, and the cell morphological changes were observed microscopically. Flow cytometry was used to investigate the effect of CD437 and ATRA on the cell cycle and apoptosis., Results: CD437 was more effective than ATRA in inhibiting the proliferation of A431 cells and normal human epidermal keratinocytes. CD437 increased the percentage of sub-G1 populations in A431 cells and induced G1 arrest in normal human epidermal keratinocytes. ATRA appeared to be relatively ineffective for inducing apoptosis in A431 cells as compared to CD437. CD437 did not duce obvious apoptosis in normal human epidermal keratinocytes., Conclusion: CD437 is more effective than ATRA in inhibiting the proliferation and inducing apoptosis in A431 cells and shows selective apoptosis-inducing effect against malignant keratinocytes, suggesting its potential in the prevention or treatment of cutaneous carcinoma.

Objective: To evaluate the value of MRI in preoperative staging of rectal cancer., Methods: The data of 156 rectal cancer patients,undergone MRI scans from December 2004 to June 2006 in our hospital, were analyzed retrospectively. Findings of MRI were compared with postoperative pathological examinations., Results: Intracavitary localized parenchyma tumors were seen in 72 cases, and intestinal wall abnormal incrassation or stricture in other 84 cases from MRI scan. Sixteen cases had rectal polyps simultaneously, 2 cases ovarian cysts, 13 cases anterior-sacral metastases and 2 cases bone metastases. The sensitivity and specificity of T(1-2), T(3) and T(4) rectal cancer by MRI examination were 25%(8/32), 93.3%(84/90), 94.1%(32/34) and 100%(124/124), 57.6%(38/66), 96.7%(118/122) respectively. In MRI imaging, metastatic para-rectal lymph node was diagnosed as the diameter >5 mm or abnormal border or mixed resonance, with a sensitivity of 85.1%(80/94) and specificity of 45.2%(28/62)., Conclusion: MRI has high accuracy for preoperative staging of rectal cancer, and is useful to detect the serosal infiltration and lymph node metastasis.

Objective: To evaluate the accuracy of proton magnetic resonance spectroscopy (1H MRS) in quantitative measurement of intra-hepatic triglyceride (TG) stores in fatty liver at the levels of animal model and human body., Methods: 40 male Wistar rats were randomly divided into 2 equal groups: fatty liver model group, fed with high fat diet for 16 weeks, and control group, fed with standard diet. Every 4 weeks 5 rats from each group underwent 1H MRS to examine the intra-hepatic triglyceride contents and then were sacrificed. The liver tissues were stained with Sudan III] to detect the fat content. Eleven hospitalized patients with fatty liver suspected or confirmed by CT and with indications of liver puncture underwent 1H MRS and then liver puncture within 7 days. Linear regression was conducted to analyze the correlation between the results of 1H MRS and liver pathology., Results: Intrahepatic lipid was detected in all the rat models, some rats of the control group, and the patients with fatty live. The lipid peak area and the ratio of the CH2 lipid peak area relative to the water peak area increased along with the duration of high fat diet feeding. In the rat models there was a close positive correlation between the hepatic TG content measured in vivo by 1H MRS and pathological measurement of liver fat content [r = 0.899; P <0001; slope 14.45; gamma-intercept -0.51]. The similar correlation existed in the patients too [r =0.814; P <0001; slope 33.72; gamma-intercept 27.04]. When the data of the rats and patients were analyzed together, the percentage of liver fat was highly positively correlated with the ratio of area under fat peak to the area under water peak (r = 0.878; P <0001; slope 56.33; gamma-intercept -0.68)., Conclusion: A relative quantitative method to measure intrahepatic lipid content by 1H MRS, a safe, non-invasive method, is primarily established at the animal level and has been verified in humans with a good correlation with the pathological examination.

Objective: To investigate the mutations of ATP2C1 gene in Chinese patients with Hailey-Hailey disease (HHD)., Methods: Genomic DNA was extracted from peripheral blood leukocytes. PCR and direct DNA sequencing were used to detect the mutations in all 27 exons of ATP2C1 gene in patients of two Chinese families and a sporadic patient with HHD., Results: Three mutations in ATP2C1 gene were found, including 1 nonsense mutation, 1 deletion/frameshift mutation and 1 missense mutation. All of them were novel mutations., Conclusion: All the three mutations could affect the transcription and translation, and further the function of protein encoded by ATP2C1 gene.

Objective: To investigate the anatomical study and clinical applications of sural neuron-myocutaneous flap transposition for repairing the special patients with soft tissue defect in foot and ankle., Methods: The branches, distributions and anastomoses of the vessels and nerves lie in superficial layer of the posterior crural region were observed on 30 sides of adult cadaver lower limb specimens perfused with red latex. Since February 2004, distally based sural neuron-myocutaneous flap was applied for repairing 7 cases of soft tissue defect in foot and ankle., Results: The nutrient vessels of sural nerve, small saphenous vein and posterior femoral cutaneous nerve anastomosed permanently with the musculocutaneous perforators of medial and lateral head of gastrocnemius. There were 2 - 3 anastomoses found respectively. The musculocutaneous perforators pierced the two heads of gastrocnemius muscle (1.8 +/- 0.5) cm medially and (3.7 +/- 0.9) cm laterally away from the groove of the muscle. The medial anastomoses more closed to the middle groove and their diameters were found larger than the lateral ones. In operation, we routinely observed the compound flap for 15 to 20 minutes and found actively errhysis on the muscle, so the fine blood circulation in the flap was demonstrated. All flap survived after operation and the cases were followed up 2 to 6 months with cured osteomyelitis and satisfied flap outline., Conclusions: Distally based sural neuro-myocutaneous flap can live. The operative method is simple. The flap offers an excellent donor site for repairing the soft tissue defect in foot and ankle in special cases.

Objective: To evaluate the ability of multidetector computed tomography (MDCT) in differentiating ovarian tumors from non-ovarian masses., Methods: Forty-two cases with pelvic masses were examined with 16-row MDCT. All source image of each case was put into workstation for multi-planar reconstruction (MPR) and curved planar reconstruction(CPR). Axial image combined with 2D image was used for determining the relationship of the mass to ovarian vascular pedicle and identifying the normal ovary, which was compared with postoperative pathologic result and the finding during operation. All the data was compared using Fisher's exact test., Results: There were 28 ovarian tumors and 14 non-ovarian tumors in this series. If the ovarian vascular pedicle sign was used for determining whether the tumor was from the ovary or not, the sensitivity, specificity, positive predictive value, negative predictive value and accuracy was 89.3%, 85.7%, 92.6%, 80.0% and 88.1%, respectively, with a significant difference in differentiating the tumor from the ovary or non-ovarian organs (P <0.05). If the identification of full normal ovary was used to determine non-ovarian origin of the tumor, the sensitivity, specificity, positive predictive value, negative predictive value and accuracy was 50.0%, 100.0%, 100.0%, 80.0% and 83.3%, respectively, also with a significant difference in differentiating the tumors from the ovary or non-ovarian organs (P <0.05)., Conclusion: MDCT can clearly show the relationship of the tumor to the normal ovary and its vascular pedicle, which is very helpful in differentiating the ovarian tumors from a non-ovarian masses.

HEV is classified into H (human) group and Z (zoonosis) group according to its compatible host. H group contains genotype 1 and genotype 2 HEV isolates which infect human only; Z group contains genotype 3 and genotype 4 HEV isolates which infect both human and animals. After analysis of amino acid sequences between ORF2 aa368 and aa606, four group-conserved sites that were all located in the neutralization region of ORF2 were identified. They are aa483, aa492, aa497 and aa599. Mutation analysis and capture PCR were then performed on these sites with a group of monoclonal antibodies. Results showed that the difference of the aa497 between H and Z groups was responsible for the maintenance of their group-specific immunodominant epitopes, probably through confirmation-dependent epitope changes. Thus, aa497 and its related change on the surface structure of HEV may play important roles in host selection by H and Z groups of HEV.

Objective: To investigate the role of MRI in evaluating the peripancreatic vessel invasion and resectability of pancreatic carcinoma based on the comparison of MRI image with surgical exploration, and try to establish the criteria for assessment of the sensitivity, specificity and accuracy of resectability., Methods: Forty-one pancreatic carcinoma patients confirmed by pathology received preoperative plain and contrast enhanced MRI scan, and 37 of them had additional coronal MRA scan. Peripancreatic vessel invasion was preoperatively assessed based on MRI features, and the vessel invasion degree from the uninvolved to the severely involved was divided into 6 grades represented by 1, 2a, 2b, 3a, 3b and 4, respectively. Compared with the findings during the surgery, the sensitivity and specificity of each vessel invasion grade were studied and the receiver operator characteristic curve (ROC) was drawn. The sensitivity, specificity, positive predictive value, negative predictive value and accuracy of resectability evaluation based on 2 and 2a degree were calculated respectively. The resectability of involved arteries and veins of grade 2 were also analyzed., Results: Of the 41 patients, 22 had resectable tumor, with 20 curative resection and 2 palliative. Compared with the findings during surgery, seven vessels including three arteries and four veins were not correctly interpreted by MRI. If grade 1,2a,2b,3a and 3b was used as the resectable standard,respectively, the sensitivity to predict the unresectbility was 78.3%, 84.8%, 67.4%, 56.5% and 47.8%, respectively. Receiver operator characteristic curve demonstrated that grade 2a was the optimal critical point. The sensitivity, specificity, positive predictive value, negative predictive value and accuracy of grade 2a in predicting the unresectbility were 84.8%, 98.5%, 92.9%, 96.6% and 95.9%., Conclusion: Our data showed that grade 2a (tumor involvement < 2 cm long and < 1/2 circumference of the vessel) may be more sensitive and accurate in predicting the resectability, which may be considered as the line of demarcation between the respectable and unresectable cases in clinical practice.
