Polyomavirus productively infects mouse cells, transforms rat fibroblasts in culture, and induces a broad spectrum of tumors when inoculated into newborn mice. The expression of large, middle, and small T antigen are necessary for virus growth and oncogenic transformation. We have generated a small deletion in a region common to both large and middle T antigen that encodes three consecutive prolines. In this report we characterize this mutant virus in terms of its ability to replicate in mouse cells, transform rat fibroblasts, and induce tumors in the mouse. We find that the virus immortalizes primary cells and that viral DNA replication is not impaired, indicating that these functions of large T antigen are not altered. However, the ability of the virus to transform rat fibroblasts is defective. The mutant virus makes fewer foci and the foci are weaker in appearance. The mutant middle T still associates with PI 3-kinase and shc, suggesting that the overall structure of the protein has not been disrupted. When inoculated into newborn C3H mice, the mutant virus induces fewer overall tumors with a longer latency than wild-type virus. These results indicate that this proline-rich domain in middle T antigen is important for oncogenesis in a wide variety of tissues and cell types.

Results in a previous report established the presence of dominant genetic factor(s) in C3H/BiDa mice which contributed to the unusually high susceptibility of this strain to polyoma virus-induced tumors (Freund et al. (1992) Virology 191, 724-731). Here we show that C57BR/cdJ mice, while identical to C3H/BiDa at the H-2 locus, are almost completely lacking in susceptibility. Analysis of crosses between these two H-2 k strains has shown that susceptibility is due to a single dominant autosomal gene, designated Pyv S . On an H-2 k background, Pyv S acts in a highly penetrant manner to confer susceptibility to induction of a broad range of tumors by the virus. Analysis of F1 mice between C3H/BiDa and the highly resistant C57BL/6 (H-2 b ) strain shows that Pyv S can partially overcome the immunologically mediated resistance associated with an H-2 b haplotype. Pyv S does not encode cell receptors for the virus, nor does it affect levels of virus replication or anti-viral humoral responses in the host. Studies with early passage embryo fibroblasts in culture show that Pyv S does not affect intracellular events essential for either productive infection or cell transformation by the virus. Pyv S thus determines a generalized susceptibility of the host to polyoma-induced tumors but apparently does not act at the level of target cells for tumor induction.

A mutant strain of polyoma virus encoding a truncated middle T protein has been studied for its ability to replicate and induce tumors following inoculation into newborn mice. Virus replication in the acute period prior to expected onset of tumors as well as persistence of virus in older animals were followed. The mutant virus proved to be defective in replication and persistence and failed to induce tumors. These results demonstrate that middle T plays an essential role in productive viral infection in the animal. Since the mutant virus encodes normal large and small T proteins, the results also indicate that functions associated with these T antigens, including large T binding of the retinoblastoma tumor suppressor gene product and the ability to immortalize, are insufficient to cause development of tumors in this system.

We have examined effects of mutations of the minor capsid proteins VP2 and VP3 of polyoma virus. Infection of cells by mutants blocked in expression of either VP2 or VP3 by alterations of their initiator methionine codons led to the rapid selection of wild-type revertants. This reversion suggests a strong, if not absolute, requirement for both proteins in virus growth. A mutant virus, VP2*, in which alanine was substituted for glycine-2, expressed a nonmyristylated form of VP2 that was incorporated into virions. Studies of VP2* revealed a 15- to 20-fold lower specific infectivity and a delay in growth in both primary and established mouse cells compared to wild-type virus. Analysis of the growth delay indicated a defect in an early step of infection, at or prior to uncoating. Synthesis of viral DNA and VP1 was delayed by about 9 hr in mutant compared to that in wild-type infected cells. No evidence was obtained for an effect on either virus assembly (encapsidation) or stability of virions. Infection of mice by the VP2* mutant virus resulted in attenuated virus replication and tumor induction which were much more severely affected following intranasal inoculation than intraperitoneal inoculation.

We have determined the tumor responses of nine inbred mouse strains and of two F1 hybrids following inoculation with polyoma virus. The results showed wide variations in the frequencies of mice developing tumors. Correlations with H-2 haplotype were evident. C3H/BiDa mice of H-2 k type were fully susceptible, while C57BL/6 mice of H-2 b type were completely resistant. DBA/2 and BALB/c mice, both H-2 d , were of low susceptibility. Contributions of the major histo-compatibility complex were demonstrated using pairs of H-2 congenic mice. Substitution of H-2 k on either a BALB/c or a C57BL/10 background resulted in an increase in the frequencies and kinds of tumors induced. Susceptibilities of BALB and C57BL mice bearing the H-2 k haplotype were still well below those of C3H/BiDa mice, however. Crosses of C3H/BiDa with BALB/c or BALB.K (H-2 k ) produced Fls that were of moderate and full susceptibility, respectively. These results indicate two distinct classes of host genes, one at the H-2 locus where different alleles confer varying degrees of resistance and the other a dominant susceptibility gene(s) carried in C3H/BiDa mice.

We have examined the state and expression of polyoma viral DNA in representative epithelial and mesenchymal tumors, using a combination of biochemical and in situ methods. Results showed wide variations among tumor types and also in different regions within individual tumors, with respect to copy number of viral DNA, presence or absence of deletions, and expression of early and late viral proteins. Epithelial tumors showed the greatest heterogeneity. High copy free viral DNA, frequently with deletions, was found in all such tumors. A portion of free viral DNA was recoverable as transcriptionally active minichromosomes. Three distinct subpopulations of cells were distinguished by in situ analyses. Type 1 cells showed high copy free viral DNA and expressed the major viral capsid protein VP1; these cells appeared to be at various stages of productive (lytic) viral infection. Some productively infected cells were able to undergo mitosis; in a portion of these cells, VP1 was found in close association with the mitotic spindle. Type 2 cells contained high copy free DNA but did not express VP1; by some unknown mechanism, these cells manifest a post-replication block to late gene expression and lytic infection. Type 3 cells contained only low copy, presumably integrated, viral DNA and expressed no VP1; they thus resemble cells transformed in vitro by the virus. Epithelial tumors contained variable mixtures of these subpopulations, while mesenchymal tumors were composed of Type 3 cells only. Differences in virus-cell interactions are discussed in terms of their possible implications in tumor development

hr-t mutants of polyomavirus are defective in virus growth as well as in cell transformation, and have genetic alterations that invariably affect both the middle and small T proteins. We have examined the growth properties of three site-directed mutants that either eliminate or alter the middle T without affecting the small T protein. Mutant 808A encodes large and small T proteins but no middle T; it grew poorly in NIH 3T3 cells. In contrast, mutants 1387T and 1178T which express altered middle T along with normal large and small T proteins grew nearly as well as wild-type virus. Thus, although the altered middle T proteins encoded by 1387T and 1178T are defective for cell transformation, they retained the ability to induce expression of a cellular permissivity factor(s) required for virus production. At the biochemical level, the induction of permissivity by middle T was manifested primarily in terms of phosphorylation of VP1 on threonine and in efficient encapsidation of viral DNA to form infectious virus. The natural role of middle T involves regulation of phosphorylation events, and can be enacted, at least in part, independently of interactions with pp60c-src.

Middle T (mT), the oncogene of murine polyomavirus, causes transformation of rat fibroblasts by activating a number of signal transducing pathways usually used by polypeptide growth factors and their receptors. Here, we report data regarding the activation of signal transducing pathways involving phospholipase D (PL-D). The hydrolysis of phospholipids by PL-D produces phosphatidic acid (PA), a compound with multiple biological effects. The PA content of cells expressing wild-type mT, introduced via a number of different methods, is approximately 50% higher than their untransformed counterparts. This increase in cellular PA content is associated with an approximately 65% increase in PL-D activity in cells expressing wild-type mT. We have also examined the effects of a number of site-directed mutants of mT, on both cellular PA levels and on PL-D activity. Mutants that do not produce mT (Py808A) or that produce a truncated, nonmembrane bound mT (Py1387T) have PA levels similar to that of control cells. Cells expressing the 322YF mutant of mT (which abolishes interaction of mT with phospholipase C γ 1 ) show increases in both PA levels and PL-D activity that are similar to those seen with wild-type mT. Expression of mutants that abolish the interaction of mT with either shc or with phosphatidylinositol 3-kinase (250YS and 315YF, respectively) cause an increase in PL-D activity comparable to that seen with wild-type mT. However, the PA content of cells expressing these mutants is not elevated. These results suggest that mT causes activation of cellular PL-D, but this activation alone is not sufficient to cause an increase in cellular PA content. Therefore, wild-type mT must affect another, as yet unknown, step in PA metabolism.
