Artificial intelligence-powered deep learning methods are being used to diagnose brain tumors with high accuracy, owing to their ability to process large amounts of data. Magnetic resonance imaging stands as the gold standard for brain tumor diagnosis using machine vision, surpassing computed tomography, ultrasound, and X-ray imaging in its effectiveness. Despite this, brain tumor diagnosis remains a challenging endeavour due to the intricate structure of the brain. This study delves into the potential of deep transfer learning architectures to elevate the accuracy of brain tumor diagnosis. Transfer learning is a machine learning technique that allows us to repurpose pre-trained models on new tasks. This can be particularly useful for medical imaging tasks, where labelled data is often scarce. Four distinct transfer learning architectures were assessed in this study: ResNet152, VGG19, DenseNet169, and MobileNetv3. The models were trained and validated on a dataset from benchmark database: Kaggle. Five-fold cross validation was adopted for training and testing. To enhance the balance of the dataset and improve the performance of the models, image enhancement techniques were applied to the data for the four categories: pituitary, normal, meningioma, and glioma. MobileNetv3 achieved the highest accuracy of 99.75%, significantly outperforming other existing methods. In the Indo-Gangetic Plains (IGP), rice-wheat cropping system (RWCS) predominates, producing large quantity of crop residue and its management is major concern. Farmers usually burn the residue to clear the field for succeding crop, and burning damages soil microbes, resulted in loss of soil organic matter. Hence, current study was conducted to assess the impact of different Happy seeder based residue management options on changes in microbial dynamics, enzyme activities and soil organic matter content and also to know that alternative method for attaining sustainable wheat productivity in sandy loam soils of Haryana, India. Results revealed that Zero tillage wheat (ZTW) with partial and full residue retention treatments sown with Happy seeder (after using chopper and spreader), and ZTW with anchored stubbles significantly enhanced soil microbial count by 47.9-60.4%, diazotropic count by 59.0-73.1% and actinomycetes count by 47.3-55.2%, grain yield by 9.8-11.3% and biomass yield by 7.4-9.6% over conventional tilled (CT) residue burning and residue removal plots. ZTW sown with surface retention of rice crop residue increased the organic carbon by 0.36-0.42% and the soil moisture content by 13.4-23.6% over CTW without residue load. Similarly, ZTW sown with Happy seeder with full residue enhanced alkaline phosphatase activity from 95.3 µg TPF g -1 soil 24 h -1 in 2018-2019 to 98.6 µg TPF g -1 soil 24 h -1 in 2019-2020 over control plots. Likely, microbial population and enzymatic activity showed strong positive correlation under variable residue retention practices. However, increased microbial population reduced the soil pH from 7.49 to 7.27 under ZTW with residue retention plots. The wheat yield enhanced by 9.8-11.3% during 2018-2019 and 2019-2020 under ZTW with Happy seeder with full residue load over residue burning and residue removal plots. ZTW sown with Happy seeder under full residue retention, achieved maximum net return 43.16-57.08 × 10 3 ₹ ha -1 ) and B-C ratio (1.52 to 1.70) over CTW without residue. Therefore, rice residue needs to be managed by planting wheat using appropriate machinery under ZT for sustaining higher productivity in RWCS and improve soil health and environment under IGP regions., (© 2024. We report the present day GNSS velocities (2015-2021) and strain rates in Himachal, Kashmir and Ladakh Himalaya covering the rupture zones of the 2005 Muzaffarabad earthquake and the 1905 Kangra earthquake. Geodetic strain rates estimated from GPS velocities of about 58 sites spanning last two decades of measurements indicate a mean compression rate of - 32.5 ns/year (nanostrain/year) and dilatation of - 37.3 ns/year. Seismic strain rates are estimated using both the instrumental period (1964-2021) and historical earthquakes since 1500 AD in this region. Seismic strain rates during the instrumental period of the past 50 + years indicate a mean compression rate of - 28.1 ns/year and it slightly decreases to - 21.7 ns/year after including the historical earthquakes of the past 520 years. The Azimuth of the seismic strain tensor for the instrumental and historic periods and geodetic strain tensor is broadly consistent with orientation of major faulting in this region suggesting uniform compression over a long-time interval justifying combined analysis of the strain rate field to determine the seismic potential of the region. Composite analysis of geodetic and seismic strain rates and the associated moments estimate the accumulated strain budget of ~ 1E + 21 Nm in the past 520 years which has a potential of generating future earthquake of M w  > 8 in this segment of Northwest Himalaya., (© 2023. Traumatic brain injury (TBI) is a leading neurological cause of death and disability across the world. Early characterization of TBI severity could provide a window for therapeutic intervention and contribute to improved outcome. We hypothesized that granular electronic health record data available in the first 24 h following admission to the intensive care unit (ICU) can be used to differentiate outcomes at discharge. Working from two ICU datasets we focused on patients with a primary admission diagnosis of TBI whose length of stay in ICU was ≥ 24 h (N = 1689 and 127). Features derived from clinical, laboratory, medication, and physiological time series data in the first 24 h after ICU admission were used to train elastic-net regularized Generalized Linear Models for the prediction of mortality and neurological function at ICU discharge. Model discrimination, determined by area under the receiver operating characteristic curve (AUC) analysis, was 0.903 and 0.874 for mortality and neurological function, respectively. Model performance was successfully validated in an external dataset (AUC 0.958 and 0.878 for mortality and neurological function, respectively). Principal Component Analysis (PCA) projects high-dimensional genotype data into a few components that discern populations. Ancestry Informative Markers (AIMs) are a small subset of SNPs capable of distinguishing populations. We integrate these two approaches by proposing an algorithm to identify necessary informative loci whose removal from the data deteriorates the PCA structure. Unlike classical AIMs, necessary informative loci densely cover the genome, hence can illuminate the evolution and mixing history of populations. We conduct a comprehensive analysis to the genotype data of the 1000 Genomes Project using necessary informative loci. Projections along the top seven principal components demarcate populations at distinct geographic levels. Millions of necessary informative loci along each PC are identified. Population identities along each PC are approximately determined by weighted sums of minor (or major) alleles over the informative loci. Variations of allele frequencies are aligned with the history and direction of population evolution. The population distribution of projections along the top three PCs is recapitulated by a simple demographic model based on several waves of founder population separation and mixing. Informative loci possess locational concentration in the genome and functional enrichment. Genes at two hot spots encompassing dense PC 7 informative loci exhibit differential expressions among European populations. The mosaic of local ancestry in the genome of a mixed descendant from multiple populations can be inferred from partial PCA projections of informative loci. Finally, informative loci derived from the 1000 Genomes data well predict the projections of an independent genotype data of South Asians. Metabolic syndrome is characterized by central obesity, insulin resistance, elevated blood pressure, and dyslipidemia. Metabolic syndrome is a significant risk factor for several common cancers (e.g., liver, colorectal, breast, pancreas). Pharmacologic treatments used for the components of the metabolic syndrome appear to be insufficient to control cancer development in subjects with metabolic syndrome. Murine models showed that cancer has the slowest progression when there is no food consumption during the daily activity phase. Intermittent fasting from dawn to sunset is a form of fasting practiced during human activity hours. To test the anticancer effect of intermittent fasting from dawn to sunset in metabolic syndrome, we conducted a pilot study in 14 subjects with metabolic syndrome who fasted (no eating or drinking) from dawn to sunset for more than 14 h daily for four consecutive weeks. We collected serum samples before 4-week intermittent fasting, at the end of 4th week during 4-week intermittent fasting and 1 week after 4-week intermittent fasting. We performed serum proteomic analysis using nano ultra-high performance liquid chromatography-tandem mass spectrometry. We found a significant fold increase in the levels of several tumor suppressor and DNA repair gene protein products (GP)s at the end of 4th week during 4-week intermittent fasting (CALU, INTS6, KIT, CROCC, PIGR), and 1 week after 4-week intermittent fasting (CALU, CALR, IGFBP4, SEMA4B) compared with the levels before 4-week intermittent fasting. We also found a significant reduction in the levels of tumor promoter GPs at the end of 4th week during 4-week intermittent fasting (POLK, CD109, CAMP, NIFK, SRGN), and 1 week after 4-week intermittent fasting (CAMP, PLAC1) compared with the levels before 4-week intermittent fasting. Fasting from dawn to sunset for four weeks also induced an anti-diabetes proteome response by upregulating the key regulatory proteins of insulin signaling at the end of 4th week during 4-week intermittent fasting (VPS8, POLRMT, IGFBP-5) and 1 week after 4-week intermittent fasting (PRKCSH), and an anti-aging proteome response by upregulating H2B histone proteins 1 week after 4-week intermittent fasting. Subjects had a significant reduction in body mass index, waist circumference, and improvement in blood pressure that co-occurred with the anticancer, anti-diabetes, and anti-aging serum proteome response. These findings suggest that intermittent fasting from dawn to sunset actively modulates the respective genes and can be an adjunct treatment in metabolic syndrome. Further studies are needed to test the intermittent fasting from dawn to sunset in the prevention and treatment of metabolic syndrome-induced cancers.

We present GPS velocities in Kashmir valley and adjoining regions from continuous Global Positioning System (cGPS) network during 2008 to 2019. Results indicate total arc normal shortening rates of ~ 14 mm/year across this transect of Himalaya that is comparable to the rates of ~ 10 to 20 mm/year reported else-where in the 2500 km Himalaya Arc. For the first time in Himalayas, arc-parallel extension rate of ~ 7 mm/year was recorded in the Kashmir valley, pointing to oblique deformation. Inverse modeling of the contemporary deformation rates in Kashmir valley indicate oblique slip of ~ 16 mm/year along the decollement with locking depth of ~ 15 km and width of ~ 145 km. This result is consistent with the recorded micro-seismicity and low velocity layer at a depth of 12 to 16 km beneath the Kashmir valley obtained from collocated broadband seismic network. Geodetic strain rates are consistent with the dislocation model and micro-seismic activity, with high strain accumulation (~ 7e-08 maximum compression) to the north of Kashmir valley and south of Zanskar ranges. Assuming the stored energy was fully released during 1555 earthquake, high geodetic strain rate since then and observed micro-seismicity point to probable future large earthquakes of Mw ~ 7.7 in Kashmir seismic gap.

The high fat diet alters intestinal microbiota due to increased intestinal permeability and susceptibility to microbial antigens leads to metabolic endotoxemia. But probiotic juices reported for various health benefits. In this background we hypothesized that pectinase treated probiotic banana juice has diverse effects on HFD induced obesity and non-alcoholic steatohepatitis. 20 weeks fed HFD successfully induced obesity and its associated complications in experimental rats. The supplementation of probiotic banana juice for 5 months at a dose of 5 mL/kg bw/day resulted significant decrease (p < 0.05) in body weight (380 ± 0.34), total fat (72 ± 0.8), fat percentage (17 ± 0.07) and fat free mass (165 ± 0.02). Reduction (p < 0.05) in insulin resistance (5.20 ± 0.03), lipid profile (TC 120 ± 0.05; TG 160 ± 0.24; HDL 38 ± 0.03), liver lipid peroxidation (0.7 ± 0.01), hepatic enzyme markers (AST 82 ± 0.06; ALT 78 ± 0.34; ALP 42 ± 0.22), and hepatic steatosis by increasing liver antioxidant potential (CAT 1.4 ± 0.30; GSH 1.04 ± 0.04; SOD 0.82 ± 0.22) with normal hepatic triglycerides (15 ± 0.02) and glycogen (0.022 ± 0.15) contents and also showed normal liver size, less accumulation of lipid droplets with only a few congestion. It is concluded that the increased intestinal S. cerevisiae yeast can switch anti-obesity, antidiabetic, antioxidative stress, antioxidant and anti-hepatosteatosis effect. This study results will have significant implications for treatment of NAFLD.

Children diagnosed with brain tumors have the lowest overall survival of all pediatric cancers. Recent molecular studies have resulted in the discovery of recurrent driver mutations in many pediatric brain tumors. However, despite these molecular advances, the clinical outcomes of high grade tumors, including H3K27M diffuse midline glioma (H3K27M DMG), remain poor. To address the paucity of tissue for biological studies, we have established a comprehensive protocol for the coordination and processing of donated specimens at postmortem. Since 2010, 60 postmortem pediatric brain tumor donations from 26 institutions were coordinated and collected. Patient derived xenograft models and cell cultures were successfully created (76% and 44% of attempts respectively), irrespective of postmortem processing time. Histological analysis of mid-sagittal whole brain sections revealed evidence of treatment response, immune cell infiltration and the migratory path of infiltrating H3K27M DMG cells into other midline structures and cerebral lobes. Sequencing of primary and disseminated tumors confirmed the presence of oncogenic driver mutations and their obligate partners. Our findings highlight the importance of postmortem tissue donations as an invaluable resource to accelerate research, potentially leading to improved outcomes for children with aggressive brain tumors.

The exciting discovery of the semiconducting-like properties of deoxyribonucleic acid (DNA) and its potential applications in molecular genetics and diagnostics in recent times has resulted in a paradigm shift in biophysics research. Recent studies in our laboratory provide a platform towards detecting charge transfer mechanism and understanding the electronic properties of DNA based on the sequence-specific electronic response, which can be applied as an alternative to identify or detect DNA. In this study, we demonstrate a novel method for identification of DNA from different shrimp viruses and bacteria using electronic properties of DNA obtained from both negative and positive bias regions in current-voltage (I-V) profiles. Characteristic electronic properties were calculated and used for quantification and further understanding in the identification process. Aquaculture in shrimp industry is a fast-growing food sector throughout the world. However, shrimp culture in many Asian countries faced a huge economic loss due to disease outbreaks. Scientists have been using specific established methods for detecting shrimp infection, but those methods do have their significant drawbacks due to many inherent factors. As such, we believe that this simple, rapid, sensitive and cost-effective tool can be used for detection and identification of DNA from different shrimp viruses and bacteria.

Current research is based on the identification of novel inhibitors of α-amylase enzyme. For that purpose, new hybrid molecules of hydrazinyl thiazole substituted chromones 5-27 were synthesized by multi-step reaction and fully characterized by various spectroscopic techniques such as EI-MS, HREI-MS, 1 H-NMR and 13 C-NMR. Stereochemistry of the iminic bond was confirmed by NOESY analysis of a representative molecule. All compounds 5-27 along with their intervening intermediates 1-4, were screened for in vitro α-amylase inhibitory, DPPH and ABTS radical scavenging activities. All compounds showed good inhibition potential in the range of IC 50  = 2.186-3.405 µM as compared to standard acarbose having IC 50 value of 1.9 ± 0.07 µM. It is worth mentioning that compounds were also demonstrated good DPPH (IC 50  = 0.09-2.233 µM) and ABTS (IC 50  = 0.584-3.738 µM) radical scavenging activities as compared to standard ascorbic acid having IC 50  = 0.33 ± 0.18 µM for DPPH and IC 50  = 0.53 ± 0.3 µM for ABTS radical scavenging activities. In addition to that cytotoxicity of the compounds were checked on NIH-3T3 mouse fibroblast cell line and found to be non-toxic. In silico studies were performed to rationalize the binding mode of compounds (ligands) with the active site of α-amylase enzyme.

We estimate a new angular velocity for the India plate and contemporary deformation rates in the plate interior and along its seismically active margins from Global Positioning System (GPS) measurements from 1996 to 2015 at 70 continuous and 3 episodic stations. A new India-ITRF2008 angular velocity is estimated from 30 GPS sites, which include stations from western and eastern regions of the plate interior that were unrepresented or only sparsely sampled in previous studies. Our newly estimated India-ITRF2008 Euler pole is located significantly closer to the plate with ~3% higher angular velocity than all previous estimates and thus predicts more rapid variations in rates and directions along the plate boundaries. The 30 India plate GPS site velocities are well fit by the new angular velocity, with north and east RMS misfits of only 0.8 and 0.9 mm/yr, respectively. India fixed velocities suggest an approximate of 1-2 mm/yr intra-plate deformation that might be concentrated along regional dislocations, faults in Peninsular India, Kachchh and Indo-Gangetic plain. Relative to our newly-defined India plate frame of reference, the newly estimated velocities for 43 other GPS sites along the plate margins give insights into active deformation along India's seismically active northern and eastern boundaries.

The goal of this pilot study was to determine whether HDL glycoprotein composition affects HDL's immunomodulatory function. HDL were purified from healthy controls (n = 13), subjects with metabolic syndrome (MetS) (n = 13), and diabetic hemodialysis (HD) patients (n = 24). Concentrations of HDL-bound serum amyloid A (SAA), lipopolysaccharide binding protein (LBP), apolipoprotein A-I (ApoA-I), apolipoprotein C-III (ApoC-III), α-1-antitrypsin (A1AT), and α-2-HS-glycoprotein (A2HSG); and the site-specific glycovariations of ApoC-III, A1AT, and A2HSG were measured. Secretion of interleukin 6 (IL-6) in lipopolysaccharide-stimulated monocytes was used as a prototypical assay of HDL's immunomodulatory capacity. HDL from HD patients were enriched in SAA, LBP, ApoC-III, di-sialylated ApoC-III (ApoC-III 2 ) and desialylated A2HSG. HDL that increased IL-6 secretion were enriched in ApoC-III, di-sialylated glycans at multiple A1AT glycosylation sites and desialylated A2HSG, and depleted in mono-sialylated ApoC-III (ApoC-III 1 ). Subgroup analysis on HD patients who experienced an infectious hospitalization event within 60 days (HD+) (n = 12), vs. those with no event (HD-) (n = 12) showed that HDL from HD+ patients were enriched in SAA but had lower levels of sialylation across glycoproteins. Our results demonstrate that HDL glycoprotein composition, including the site-specific glycosylation, differentiate between clinical groups, correlate with HDL's immunomodulatory capacity, and may be predictive of HDL's ability to protect from infection.

The Shuttle Radar Topography Mission (SRTM) Digital Terrain Elevation Data (DTED) are used with the consensus view that it has a minimum vertical accuracy of 16 m absolute error at 90% confidence (Root Mean Square Error (RMSE) of 9.73 m) world-wide. However, vertical accuracy of the data decreases with increase in slope and elevation due to presence of large outliers and voids. Therefore, studies using SRTM data "as is", especially in regions like the Himalaya, are not statistically meaningful. New data from ~200 high-precision static Global Position System (GPS) Independent Check Points (ICPs) in the Himalaya and Peninsular India indicate that only 1-arc X-Band data are usable "as is" in the Himalaya as it has height accuracy of 9.18 m (RMSE). In contrast, recently released (2014-2015) "as-is" 1-arc and widely used 3-arc C-Band data have a height accuracy of RMSE 23.53 m and 47.24 m and need to be corrected before use. Outlier and void filtering improves the height accuracy to RMSE 8 m, 10.14 m, 14.38 m for 1-arc X and C-Band and 3-arc C-Band data respectively. Our study indicates that the C-Band 90 m and 30 m DEMs are well-aligned and without any significant horizontal offset implying that area and length computations using both the datasets have identical values., Competing Interests: The authors declare no competing financial interests.

Human milk oligosaccharides (HMOs) play an important role in the health of an infant as substrate for beneficial gut bacteria. Little is known about the effects of HMO composition and its changes on the morbidity and growth outcomes of infants living in areas with high infection rates. Mother's HMO composition and infant gut microbiota from 33 Gambian mother/infant pairs at 4, 16, and 20 weeks postpartum were analyzed for relationships between HMOs, microbiota, and infant morbidity and growth. The data indicate that lacto-N-fucopentaose I was associated with decreased infant morbidity, and 3'-sialyllactose was found to be a good indicator of infant weight-for-age. Because HMOs, gut microbiota, and infant health are interrelated, the relationship between infant health and their microbiome were analyzed. While bifidobacteria were the dominant genus in the infant gut overall, Dialister and Prevotella were negatively correlated with morbidity, and Bacteroides was increased in infants with abnormal calprotectin. Mothers nursing in the wet season (July to October) produced significantly less oligosaccharides compared to those nursing in the dry season (November to June). These results suggest that specific types and structures of HMOs are sensitive to environmental conditions, protective of morbidity, predictive of growth, and correlated with specific microbiota.
