ABSTRACT: This study evaluated the effects of dietary butyric acid (BA) on the Japanese quail' performance, immunology, lipid profile, cecal microbiota, and antioxidant levels. 250 unsexed, one-week-old quail chicks were divided into 5 groups, each with fifty chicks (5 replicates of 10 chicks). The first group was given the basal diet (BD), while the 2nd to 5th groups were fed BD with 50, 100, 150, and 200 mg BA/kg, respectively. The results indicated that BA improved weight gain and FCR (p < 0.05) and decreased total FI. The 200 mg BA/kg of diet showed the lowest FI (p < 0.05) and the best FCR (p > 0.05). BA boosted immunity through increasing IgA, IgM, IgG, and Complement 3. Significantly lower alanine aminotransferase (ALT), aspartate aminotransferase (AST), and lactate dehydrogenase (LDH) were observed at 150 and 200 mg BA/kg (P < 0.05) than the control group. The BA-supplemented quail showed lower total cholesterol (TC), triglyceride (TG), low-density lipoprotein (LDL), and very low-density lipoprotein (VLDL) than the control one. This effect was more pronounced for 100 and 200 mg of BA/kg. However, high low-density lipoprotein (HDL) did not differ from the control group (p > 0.05). BA at ≥100 mg/kg diet reduced malondialdehyde (MDA) and induced greater levels of superoxide dismutase (SOD), total antioxidant capacity (TAC), glutathione peroxidase (GPX), globulin, total protein, digestive enzymes than the control group (P < 0.05). BA decreased cecal E. coli, Salmonella, Enterococcus, and Coliforms and increased Lactic acid bacteria (p < 0.05) compared to non-supplemented group. Collectively, the inclusion of 100 mg BA/kg diet is ideal for Japanese quail production and health.

ABSTRACT: Brevibacillus laterosporus S62-9 has been shown to improve broiler growth performance and immunity. In the present study, we aimed to evaluate the effects of B. laterosporus S62-9 on the immunity and lipid metabolism of broilers by means of transcriptomic analysis. A total of 160 1-day-old broilers were randomly allocated to a S62-9 group, the diet of which was supplemented with 106 CFU/g B. laterosporus S62-9 daily, and a control group, which was not. After 42 d of feeding, the broilers in the S62-9 group had higher body mass (7.2%) and feed conversion ratio (5.19%) than the control group. Supplementation with B. laterosporus S62-9 resulted in lower serum total cholesterol and low-density lipoprotein-cholesterol concentrations and higher high-density lipoprotein-cholesterol concentrations. An analysis of the fatty acid composition of the broiler's thigh muscles revealed that the proportions of the unsaturated fatty acids myristoleic acid (C14:1) and arachidonic acid (C20:1) were higher for birds in the S62-9 group. Transcriptomic analysis also showed an upregulation of immunity-related genes in the S62-9 group. Gene Ontology functional enrichment analysis showed that the mitogen-activated protein kinase pathway was enriched in the liver, the defense response was enriched in the duodenum, and immunoglobulin-related entries were enriched in the jejunum of the S62-9 group. Furthermore, the expression of key genes involved in unsaturated fatty acid synthesis (SCD, encoding stearoyl-CoA desaturase) and fatty acid metabolism (HACD2, encoding 3-hydroxyacyl-CoA dehydratase 2) was upregulated in the liver, and the expression of genes associated with fat biosynthesis and accumulation, such as PLIN1, encoding perilipin 1, and FABP4, encoding fatty acid binding protein 4, was upregulated in the ileum of the birds in the S62-9 group. In summary, supplementation with B. laterosporus S62-9 could improve immune defense and the fatty acid metabolism of broiler chickens, thereby enhancing their disease resistance and promoting growth and development.

ABSTRACT: This study aimed to evaluate the effects of rosehip leaves (RL) supplementation on the production performances and egg quality of laying hens during the first stage of laying. For that, 90 Lohmann Brown Classic hens, aged 26 wk, were randomly assigned to 3 dietary treatments as follows: a control diet (RL0), an experimental diet containing 0.5% rosehip leaves (RL0.5), and an experimental diet containing 1% rosehip leaves (RL1). The laying hens were accommodated in an experimental hall with identical pens (3.96 m2), and controlled microclimate conditions. The dietary treatments were administered for a 6-wk experimental period (26–32 d of age). During this time, production performance of hens was recorded. To assess the impact of RL on egg weight and its components evolution, egg yolk and albumen color, and primary chemical composition of egg constituents were determined every 2 wk. The effect on egg yolk fatty acids profile was determined only at the end of the trial. The results showed that RL0.5 and RL1 dietary treatments significantly improved laying rate, feed conversion ratio and egg mass compared with RL0 group. A significant effect on egg weight and its components was observed only over time, showing that RL has no effect on these parameters. Significant changes in egg yolk coloration were observed over time, with significant increases in L*, a* and b* chromomeric parameters in both RL0.5 and RL1 compared to RL0. Significant alterations were obtained from the sum of egg yolk fatty acids. The RL1 egg yolk presented significantly higher n-3, and significantly lower n-6 fatty acids, compared with both RL0 and RL0.5. Further research is warranted to explore the long-term effects of rosehip leaf supplementation on laying hens performance and egg quality.

ABSTRACT: Live vaccines containing Eimeria oocysts are commercially available to protect against avian coccidiosis. Additionally, probiotics (PRO) and prebiotics (PRE) improve the poultry productivity and health and can be used as anticoccidial substitutes. However, the impact of PRO and PRE on reproductive potential, lesion score, intestinal health, and immunization outcomes of the live coccidia vaccines has not received adequate attention. Five groups of unsexed 1-day-old broiler chicks were used as follows: negative control (NC); challenged control (CC); vaccinated and challenged (VC); vaccinated, PRO-treated, and challenged (V-PRO); and vaccinated, PRE-treated, and challenged (V-PRE). At 21 d post-vaccination (pv), the vaccine increased the count of cecal anaerobes (P ≤ 0.05) and coliforms (P > 0.05) as well as harmed body weight gain (WG) (P ≤ 0.05), cecal lactic acid bacteria (P ≤ 0.05), and plasma carotenoid level (P > 0.05). None of the additives decreased oocyst shedding after vaccination, although they lowered the middle intestine and cecal lesion scores (P > 0.05). Compared to VC (2.68 ± 0.12) and V-PRE (2.66 ± 0.05), the V-PRO group showed an improved carotenoid level pv (2.96 ± 0.05) (P ≤ 0.05). V-PRE exhibited higher WG (822.95 ± 18.25) (P > 0.05) and FI (1153.01 ± 10.02) (P ≤ 0.05) than VC (781.86 ± 25.16 and 1109.85 ± 33.68) and V-PRO pv (787.61 ± 19.92 and 1077.43 ± 15.99). Following the homologous coccidia challenge, coccidia-vaccinated broilers adminstered the PRO or PRE continued to exhibit protection levels comparable to those received the vaccine alone. During 2 weeks post-challenge, VC, V-PRO and V-PRE improved bird performance and reduced oocyst shedding and lesion scores compared to CC. Ultimately, PRO and PRE treatments did not significantly reverse the reduction in growth performance in broiler chickens vaccinated against coccidia during the 1st three weeks of age.

ABSTRACT: In the first study, an in vitro culture system was developed to investigate the effects of carnosine on macrophage proinflammatory cytokine response using an established chicken macrophage cell line (CMC), gut integrity using a chicken intestinal epithelial cell line (IEC), muscle differentiation in quail muscle cells (QMCs) and primary chicken embryonic muscle cells (PMCs), and direct anti-parasitic effect against Eimeria maxima sporozoites. Cells to be tested were seeded in 24-well plates and treated with carnosine at 4 different concentrations (0.1, 1.0, and 10.0 µg). After 18 h of incubation, cells were harvested to measure gene expression of proinflammatory cytokines in CMC, tight junction (TJ) proteins in IECs, and muscle cell growth markers in QMCs and PMCs. In vivo trials were conducted to investigate the effect of dietary carnosine on disease parameters in broiler chickens challenged with E. maxima. One hundred and twenty male broiler chickens (0-day-old) were allocated into 4 treatment groups: 1) basal diet without infection (NC), 2) basal diet with E. maxima infection (PC), 3) carnosine at 10.0 mg/kg feed with PC (HCS), and 4) carnosine at 1.0 mg/kg feed with PC (LCS). All groups except NC were orally infected with E. maxima on d 14. Jejunal samples were collected for lesion scoring and jejunum gut tissues were used for transcriptomic analysis of cytokines and TJ proteins. In vitro, carnosine treatment significantly decreased IL-1β gene expression in CMC following LPS stimulation. In vivo feeding studies showed that dietary carnosine increased BW and ADG of chickens in E. maxima-infected groups and reduced the jejunal lesion score and fecal oocyst shedding in HCS group. Jejunal IL-1β, IL-8, and IFN-γ expression were suppressed in the HCS group compared to PC. The expression levels of claudin-1 and occludin in IECs were also increased in HCS following carnosine treatment. In conclusion, these findings highlight the beneficial effects of dietary carnosine supplementation on intestinal immune responses and gut barrier function in broiler chickens exposed to E. maxima infection.

ABSTRACT: The effects of rearing Cobb500 broiler chickens under 3 different housing systems (floor litter, floor plastic, and batteries) without or with feed additive (Butinov) on broiler performance, blood parameters and carcass traits were evaluated. Three hundred 1-day-old chicks were distributed in a 3 × 2 factorial arrangement (6 treatments each of 5 replicates). The results showed that reared broilers on litter or plastic floors had high values (P ≤ 0.01) of bird's weight (BW), weight gain (WG), and feed intake (FI) throughout the entire study period (1–42 d of age) compared with rearing on batteries. Rearing broilers in the different housing systems and with or without feed additives did not affect (P ≥ 0.05) total feed conversion (FCR). Different rearing systems or feed additives did not influence broiler chicks' carcass traits and some serum blood parameters. The plastic floor system significantly increased (P ≤ 0.05) blood serum corticosterone compared with litter and batteries. Feed additive (Butinov) decreased the level of T4 (P ≤ 0.05) in blood serum. Plastic floors or batteries significantly increased (P ≤ 0.01) the level of antibody titer against avian influenza virus (HIAV) compared to chicken reared on a litter floor. The results suggested that using housing systems of litter or plastic floors could improve broiler growth performance without adversely affecting carcass traits and blood characteristics compared with rearing in batteries. Also, broiler diets' feed additive (Butinov) and their interaction with rearing systems did not improve broiler growth performance.

ABSTRACT: Addition of vitamins and antioxidants has been long associated with increased immunity and are commonly used in the poultry industry; however, less is known regarding their use in broiler breeder hens. The objective of this study was to determine if feeding a complex of protected biofactors and antioxidants composed of vitamins and fermentation extracts to broiler breeder hens conferred resistance against Salmonella enterica serovar Enteritidis (S. Enteritidis) in the progeny chicks. Three-day-old chicks from control- and supplement-fed hens were challenged with S. Enteritidis and necropsied 4- and 11-days postchallenge (dpc) to determine if there were differences in invasion and colonization. Serum and jejunum were evaluated for various cytokine and chemokine production. Fewer (P = 0.002) chicks from supplement-fed hens had detectable S. Enteritidis in the ceca (32.6%) compared to chicks from control-fed hens (64%). By 11 dpc, significantly (P < 0.001) fewer chicks from supplement-fed hens were positive for S. Enteritidis (liver [36%]; ceca [16%]) compared to chicks from the control hens (liver [76%]; ceca [76%]). The recoverable S. Enteritidis in the cecal content was also lower (P = 0.01) at 11 dpc. In additional to the differences in invasion and colonization, cytokine and chemokine production were distinct between the 2 groups of chicks. Chicks from supplement-fed hens had increased production of IL-16, IL-6, MIP-3α, and RANTES in the jejunum while IL-16 and MIP-1β were higher in the serum of chicks from the control-fed hens. By 11 dpc, production of IFN-γ was decreased in the jejunum of chicks from supplement-fed hens. Collectively, these data demonstrate adding a protected complex of biofactors and antioxidants to the diet of broiler breeder hens offers a measure of transgenerational protection to the progeny against S. Enteritidis infection and reduces colonization that is mediated, in part, by a robust and distinct cytokine and chemokine response locally at the intestine and systemically in the blood.

ABSTRACT: Infection by Salmonella Typhimurium, a food-borne pathogen, can reduce the poultry production efficiency. The objective of this study was to investigate the effects of tannic acid (TA) supplementation on growth performance, Salmonella colonization, gut barrier integrity, serum endotoxin levels, antioxidant capacity, gut health, and immune function in broilers infected with the Salmonella enterica serovar Typhimurium nalidixic acid resistant strain (STNR). A total of 546 one-day-old broilers were arbitrarily allocated into 6 treatments including 1) Sham-challenged control (SCC; birds fed a basal diet and administrated peptone water); 2) Challenged control (CC; birds fed a basal diet and inoculated with 108 STNR); 3) Tannic acid 0.25 (TA0.25; CC + 0.25 g/kg TA); 4) TA0.5 (CC + 0.5 g/kg TA); 5) TA1 (CC + 1 g/kg TA); and 6) TA2 (CC + 2 g/kg TA). On D 7, supplemental TA linearly reduced STNR colonization in the ceca (P < 0.01), and TA1 and TA2 group had significantly lower reduced STNR colonization in the ceca (P < 0.01). On D 7 to 21, average daily gain tended to be linearly increased by supplemental TA (P = 0.097). The serum endotoxin levels were quadratically decreased by supplemental TA on D 21 (P < 0.05). Supplemental TA quadratically increased ileal villus height (VH; P < 0.05), and the TA0.25 group had higher ileal VH compared to the CC group (P < 0.05). Supplemental TA linearly increased percentage of peripheral blood CD8+ T cells on D 18 (P < 0.01). The TA0.5 group had significantly lower lymphocyte numbers compared to the CC groups (P < 0.05). The abundance of monocytes linearly increased with TA supplementation (P < 0.01). Therefore, broilers fed TA had reduced STNR colonization, increased growth performance, decreased serum endotoxin levels, enhanced gut health in the broilers, and stimulated the immune system in broilers infected with STNR. Supplementation of TA (1–2 g/kg) enhanced growth performance and gut health via antimicrobial and immunostimulatory effects in broilers infected with STNR.

As the demand for alternatives to antibiotic growth promoters (AGP) increases in food animal production, phytobiotic compounds gain popularity because of their ability to mimic the desirable bioactive properties of AGP. Chestnut tannins (ChT) are one of many phytobiotic compounds used as feed additives, particularly in South America, for broilers because of its favorable antimicrobial and growth promotion capabilities. Although studies have observed the microbiological and immunologic effects of ChT, there is a lack of studies evaluating the metabolic function of ChT. Therefore, the objective of this study was to characterize the cecal metabolic changes induced by ChT inclusion and how they relate to growth promotion. A total of 200 day-of-hatch broiler chicks were separated into 2 feed treatment groups: control and 1% ChT. The ceca from all the chicks in the treatment groups were collected on day 2, 4, 6, 8, and 10 after hatch. The cytokine mRNA quantitative RT-PCR was determined using TaqMan gene expression assays for IL-1B, IL-6, IL-8, IL-10, and interferon gamma quantification. The cytokine expression showed highly significant increased expressions of IL-6 and IL-10 on day 2 and 6, whereas the other proinflammatory cytokines did not have significantly increased expression. Attenuation of host IL-10 activity during Eimeria infection may elicit a robust Th1 response to eliminate the parasite from the gut epithelium. An experiment was conducted to study the effects of feeding IL-10 neutralizing antibody delivered via a dried egg product (DEP) on growth performance, immune responsivity, and gut health outcomes during a severe challenge with either Eimeria acervulina (study 1) or Eimeria tenella (study 2) following FDA CVM #217 protocol to test anticoccidial products. A total of 720 male Ross 308 chicks were used in each study, with 15 replicate cages of 12 birds and the following 4 treatments: sham-inoculated (uninfected) control diet (UCON), Eimeria-infected control diet (ICON), and Eimeria-infected control diet supplemented with DEP at 2 levels (165 [I-165] or 287 [I-287] U/tonne in study 1 and 143 [I-143] or 287 [I-287] U/tonne in study 2). ABSTRACT: Aflatoxin B1 (AFB1) is one of the most toxic mycotoxins. It has been reported that dietary exposure to AFB1 is related to the low growth performance, immunosuppression, and high susceptibility to infectious diseases of chickens. The aim of the present study was to evaluate the protective effects of Lactobacillus salivarius on broiler chickens challenged with AFB1. First, AFB1 degradation ability of Lactobacillus salivarius was measured by a high-performance liquid chromatography (HPLC) method. Then, the Arbor Acres broiler chickens were randomly assigned to experimental groups. The effects of Lactobacillus salivarius supplementation on the growth performance, liver function, and meat quality were measured, and immune response was also determined after vaccination with attenuated infectious bursal disease virus (IBDV) vaccine of broilers challenged with AFB1. Besides, resistance to Salmonella Pullorum infection along with AFB1 exposure was determined in broilers. The results showed that Lactobacillus salivarius could effectively degrade AFB1. Lactobacillus salivarius supplementation improved growth performance, liver function, and meat quality of broilers challenged with AFB1. In addition, Lactobacillus salivarius supplementation resulted in enhanced specific antibody and IFN-γ production, and lymphocyte proliferation in broilers challenged with AFB1 after IBDV vaccine immunization. Furthermore, Lactobacillus salivarius supplementation enhanced Salmonella Pullorum infection resistance in broilers challenged with AFB1. Our results revealed a tremendous potential of Lactobacillus salivarius as feed additive to degrading AFB1 and increasing broilers production performance in poultry production.

ABSTRACT: Carotenoids, which are pigments known to have many health benefits, such as their antioxidant properties, are being researched for their potential as a feed additive for production animals. These pigments are found in varying quantities in different breeds of corn, and their impact on the chicken microbiome requires further investigation. This 35 d laying hen (Novagen White) feeding trial involved varying the levels and composition of carotenoids by changing the corn source: white (0.9 µg total carotinoids/g total diet), yellow (5.7 µg/g), and orange (24.9 µg/g). For each of the three corn diet treatments, 6 replicate cages were randomly assigned. The cecal microbial community composition of the hens was then studied by 16S rRNA gene amplicon sequencing. The composition of the cecal bacterial community, as determined by Bray-Curtis dissimilarity, was different (P < 0.05) in chickens fed the orange corn diet, compared to chickens on the white corn diet, but there was no statistical difference between animals fed yellow corn compared to the white or orange corn groups. There was no change in the alpha diversity between any of the groups. Within Lactobacillus, which is one of the most abundant genera, 2 amplicon sequence variants (ASVs) were decreased and one ASV was increased in the orange corn group compared to both the white and yellow corn groups. While previous studies showed that orange corn did not alter the community composition in broilers, it appears that orange corn based feed may alter the community composition of laying hens.

During the first week after hatch, young chicks are vulnerable to pathogens as the immune system is not fully developed. The objectives of this study were to determine if supplementing the starter diet with a microencapsulated feed additive containing citric and sorbic acids, thymol, and vanillin affects in vitro functional activity of peripheral blood leukocytes (PBLs). Day-old chicks (n = 800) were assigned to either a control diet (0 g/metric ton [MT]) or a diet supplemented with 500 g/MT of the microencapsulated additive. At 4 D of age, peripheral blood was collected (100 birds per treatment), and heterophils and monocytes isolated (n = 4). Heterophils were assayed for the ability to undergo degranulation and production of an oxidative burst response while nitric oxide production was measured in monocytes. Select cytokine and chemokine mRNA expression levels were also determined. Statistical analysis was performed using Student t test comparing the supplemented diet to the control (P ≤ 0.05). Heterophils isolated from chicks fed the microencapsulated citric and sorbic acids, thymol, and vanillin had higher (P ≤ 0.05) levels of degranulation and oxidative burst responses than those isolated from chicks on the control diet. Heterophils from the supplemented chicks also had greater (P ≤ 0.05) expression of IL10, IL1β, and CXCL8 mRNA than those from control-fed chicks. Similarly, nitric oxide production was significantly (P ≤ 0.05) higher in monocytes isolated from birds fed the supplement. The cytokine and chemokine profile in monocytes from the supplement-fed chicks showed a significant (P ≤ 0.05) drop in IL10 mRNA expression while IL1β, IL4, and CXCL8 were unchanged. In conclusion, 4 D of supplementation with a microencapsulated blend made up of citric and sorbic acids, thymol, and vanillin enhanced the in vitro PBL functions of degranulation, oxidative burst, and nitric oxide production compared with the control diet. Collectively, the data suggest feeding broiler chicks a diet supplemented with a microencapsulated blend of citric and sorbic acids, thymol, and vanillin may prime key immune cells making them more functionally efficient and acts as an immune-modulator to boost the inefficient and undeveloped immune system of young chicks.

ABSTRACT: The novel chelator, L-glutamic acid, N,N-diacetic acid (GLDA) can be used as a dietary ingredient to safely reduce Zn supplementation in complete feed, without compromising the Zn status of farm animals. The objective of this study was to study dietary tolerance, bioaccumulation, and evaluate the safety of GLDA when supplemented in broiler diets at 0, 100, 300, 1000, 3,000, and 10,000 mg/kg. A total of 480 one-day-old Ross 308 male broilers were randomly allocated to 48 pens and fed one of the 6 experimental diets. Production performance was used to assess tolerance to the additive. At trial end, toxicity was evaluated using hematology, plasma biochemistry (n = 144) and gross necropsy (n = 48). Residue levels of GLDA were assessed in liver, kidney and breast tissue of birds used for necropsy. Performance showed an increase (P < 0.05) in body weight for GLDA inclusion at 300 mg/kg. A decrease on the measured performance parameters was found for the 10,000 mg/kg GLDA inclusion level (P < 0.05). The additive was added as a tetra-sodium salt, leading to sodium levels being 2.5 times higher in the latter treatment compared to the control diet which may have led to impaired intestinal barrier function. Mortality was not different between treatments. Residue levels for GLDA at the highest inclusion indicate that 0.0005% of total GLDA consumption is accumulated in breast tissue. Higher values of GLDA were found in kidney and liver at the highest inclusion level, potentially confirming that the small fraction of GLDA absorbed was readily excreted by the animal. At 100 and 300 mg/kg GLDA inclusion there were negligible amounts of GLDA present in all tissues measured. The present experiment demonstrated a high dietary tolerance to GLDA in broilers and indicated that GLDA does not pose a significant risk to food safety when supplemented below 3,000 mg/kg.

Live vaccines containing Eimeria oocysts are commercially available to protect against avian coccidiosis. Additionally, probiotics (PRO) and prebiotics (PRE) improve the poultry productivity and health and can be used as anticoccidial substitutes. However, the impact of PRO and PRE on reproductive potential, lesion score, intestinal health, and immunization outcomes of the live coccidia vaccines has not received adequate attention. Five groups of unsexed 1-day-old broiler chicks were used as follows: negative control (NC); challenged control (CC); vaccinated and challenged (VC); vaccinated, PRO-treated, and challenged (V-PRO); and vaccinated, PRE-treated, and challenged (V-PRE). At 21 d post-vaccination (pv), the vaccine increased the count of cecal anaerobes (P ≤ 0.05) and coliforms (P > 0.05) as well as harmed body weight gain (WG) (P ≤ 0.05), cecal lactic acid bacteria (P ≤ 0.05), and plasma carotenoid level (P > 0.05). None of the additives decreased oocyst shedding after vaccination, although they lowered the middle intestine and cecal lesion scores (P > 0.05). Compared to VC (2.68 ± 0.12) and V-PRE (2.66 ± 0.05), the V-PRO group showed an improved carotenoid level pv (2.96 ± 0.05) (P ≤ 0.05). V-PRE exhibited higher WG (822.95 ± 18.25) (P > 0.05) and FI (1153.01 ± 10.02) (P ≤ 0.05) than VC (781.86 ± 25.16 and 1109.85 ± 33.68) and V-PRO pv (787.61 ± 19.92 and 1077.43 ± 15.99). Following the homologous coccidia challenge, coccidia-vaccinated broilers adminstered the PRO or PRE continued to exhibit protection levels comparable to those received the vaccine alone. During 2 weeks post-challenge, VC, V-PRO and V-PRE improved bird performance and reduced oocyst shedding and lesion scores compared to CC. ABSTRACT: This paper presents attempts to enrich hens eggs with ions of copper, manganese, and zinc through the use of new feed additives (19 mg Cu2+; 124 mg Mn2+ and 85 mg Zn2+) such as biomass of alfalfa and goldenrod after extraction with supercritical carbon dioxide enriched with microelements via biosorption. Mechanical parameters of eggs (shell thickness and strength, Haugh unite), hen's laying performance, microelements content in albumen and yolk were examined and the transfer factor from feed to eggs was determined. The highest transfer of microelements content in albumen occurred in the group of hens fed with enriched goldenrod in a 100% dose (daily dose of microelements from biomass; Cu2+ 106%; Mn2+ 104%; Zn2+ 104% more in comparison to the inorganic salt group), while the highest yolk enrichment with microelements manifested itself for hens fed with enriched goldenrod in a 50% dose (daily dose of microelements from biomass; Cu2+ 32%; Zn2+ 22% more in comparison to the inorganic salt group). These groups also had the highest total microelements concentration. Mechanical properties of eggs varied insignificantly during the trial. Production parameters did not differ statistically among all experimental group. Eggs produced with need additives had better organoleptic parameters than fed with conventional premixes, which is why they were preferred by the respondents. The presented technology allows obtaining low-cost feed materials characterized by high bioavailability of components. The produced feed additives can serve as potential material for biofortification of eggs with nutrients.

ABSTRACT: Spotty liver disease (SLD) is a serious condition affecting extensively housed laying hens. The causative bacterium was described in 2015 and characterized in 2016 and named Campylobacter hepaticus. Antibiotics are the only tool currently available to combat SLD. However, antimicrobial resistance has already been detected, so finding therapeutic alternatives is imperative. Isoquinoline alkaloids (IQA), such as sanguinarine and chelerythrine, have been shown to have immunomodulatory effects. It has been hypothesized that IQA could ameliorate some of the deleterious effects of SLD. This study aimed to address that hypothesis in an experimental disease induction model. Birds were fed with diets containing 2 different doses of an IQA containing product, 100 mg of product/kg of feed (0.5 ppm of sanguinarine) and 200 mg of product/kg of feed (1.0 ppm of sanguinarine). Two additional groups remained untreated (a challenged positive control and an unchallenged negative control). After 4 wk of treatment, birds from all groups except the negative control group were exposed to C. hepaticus strain HV10. The IQA treated groups showed a reduction in the number of miliary lesions on the liver surface and reduced lesion scores compared with untreated hens. A significant reduction of egg mass was detected 6 d after exposure to C. hepaticus in the untreated group (P = 0.02). However, there was not a significant drop in egg-mass in the IQA groups, especially those fed with a high dose of IQA (P = 0.93). IQA supplementation did not produce significant changes in intestinal villus height and crypt depth but did result in a significant reduction in the proinflammatory cytokine, interleukin-8, in the blood (P < 0.01). Microbiota analysis showed that IQA treatment did not alter the alpha diversity of the cecal microbiota but did produce changes in the phylogenetic structure, with the higher dose of IQA increasing the Firmicutes/Bacteroidetes ratio.Other minor changes in production indicators included an increase in feed consumption (P < 0.01) and an increase in body weight of the treated hens (P < 0.0001). The present study has demonstrated that IQA confers some protection of chickens from the impact of SLD.

The effects of rearing Cobb500 broiler chickens under 3 different housing systems (floor litter, floor plastic, and batteries) without or with feed additive (Butinov) on broiler performance, blood parameters and carcass traits were evaluated. Three hundred 1-day-old chicks were distributed in a 3 × 2 factorial arrangement (6 treatments each of 5 replicates). The results showed that reared broilers on litter or plastic floors had high values (P ≤ 0.01) of bird's weight (BW), weight gain (WG), and feed intake (FI) throughout the entire study period (1–42 d of age) compared with rearing on batteries. Rearing broilers in the different housing systems and with or without feed additives did not affect (P ≥ 0.05) total feed conversion (FCR). Different rearing systems or feed additives did not influence broiler chicks' carcass traits and some serum blood parameters. The plastic floor system significantly increased (P ≤ 0.05) blood serum corticosterone compared with litter and batteries. Feed additive (Butinov) decreased the level of T 4 (P ≤ 0.05) in blood serum. Plastic floors or batteries significantly increased (P ≤ 0.01) the level of antibody titer against avian influenza virus (HIAV) compared to chicken reared on a litter floor. The results suggested that using housing systems of litter or plastic floors could improve broiler growth performance without adversely affecting carcass traits and blood characteristics compared with rearing in batteries. An experiment was conducted to investigate the toxicity and tissue distribution of methylsulfonylmethane (MSM) following oral gavage in broilers. A total of four hundred and thirty-two 15-day-old Ross 308 male broilers were allotted to 6 treatments with 6 replicates of 12 birds per replicate and administered a single oral dose of MSM at 0, 50, 100, 300, 1,000, or 2,000 mg/kg BW (Study 1). Another one hundred and sixty-eight 3-day-old chicks were allotted to either control or test group (Study 2) and administered a daily oral gavage of either 0 or 1, 500 mg/kg BW of MSM for 21 D consecutively. Blood and tissue samples were collected over a 48 h (Study 1) or 21 D (Study 2) period and analyzed for MSM concentrations. Toxicity was assessed through changes in hematology and clinical blood chemistry. In Study 1, plasma MSM concentrations were below 167 μg/mL at all time-points in birds receiving up to 300 mg/kg BW, and were significantly higher (P < 0.05) in birds receiving 1,000 or 2,000 mg/kg BW. Similarly, only the highest 2 MSM dosages elicited increased lymphocyte and decreased heterophil counts at 8 h (P < 0.003) and decreased hematocrit at 48 h (P = 0.015). Growth performance variables were unaffected by MSM in Study 2, and plasma and tissue MSM concentrations were highest on study day 21, with MSM-dosed birds always exhibiting higher (P < 0.03) concentrations compared with the control. Birds in Study 2 that were dosed with MSM had decreased liver enzyme concentrations at day 7 and 21 and decreased glucose and phosphorus at day 7. Importantly, MSM was detected in plasma and all tissues of control groups, confirming that MSM is synthesized de novo in chickens. In conclusion, oral MSM at either acute (single dose at 1,000 to 2,000 mg/kg BW) or sub-chronic (1,500 mg/kg BW daily for 21 D) concentrations did not cause any adverse effects on growth or clinical outcomes and appeared to be absorbed and distributed throughout the body. The aim of this study was to evaluate the effect of ampicillin, an organic acid-based feed additive and a synbiotic preparation on the prevalence of antibiotic-resistant E. coli in the ceca of broilers. A total of 2000 broiler chickens (Ross 708) were randomly assigned to 5 groups with 8 replicates. The negative control group was the only group that was not subjected to avian pathogenic E. coli challenge, while all the other 4 groups received a multi-resistant E. coli strain that was resistant to ampicillin, cephalexin, and nalidixic acid as an oral challenge. The second group served as a challenge control, and the third group received the antibiotic ampicillin via water for 5 d. The fourth group received a feed additive based on organic acids and cinnamaldehyde, and the fifth group received a synbiotic preparation via feed and water. On day 17 and 38 of the trial, cecal samples from 3 birds from each of the 40 pens were obtained, and the E. coli counts and abundances of antibiotic-resistant E. coli were determined. Oral challenge with an avian pathogenic E. coli strain did not influence the performance, and there was no significant difference in growth performance between groups. The total E. coli count was lower (P < 0.05) in the group supplemented with the synbiotic than in the challenge control group on day 38 of the trial. Administration of an antibiotic for 5 d led to a significant increase in the abundance of E. coli strains resistant to ampicillin, amoxicillin-clavulanic acid, cefoxitin, and ceftriaxone. There was no increase in the abundance of antibiotic-resistant E. coli observed in the groups that received feed supplemented with an organic acid/cinnamaldehyde-based feed additive or a synbiotic. Moreover, the effects of the tested feed additives on the prevalence of resistant E. coli are demonstrated by the lower ceftriaxone minimal inhibitory concentration values for this group than for the antibiotic group. Antibiotic growth promoters have been widely used in poultry to improve overall performance. The emergence of antibiotic resistance has resulted in sanctions imposed on the use of antibiotics in poultry diets, and alternatives such as herbal extracts are being considered to improve growth performance. The aim of this study was to compare the performance and feed digestibility of the feed supplement Novacid, which contains organic acids, glucomannan, and phytochemicals, with that of the antibiotic growth promoter bacitracin methylene disalicylate (BMD) in commercial broiler chickens. Six hundred 1-d-old Ross × Ross 308 male broiler chicks were randomly and equally assigned to six treatment groups with five replicates each (20 chicks per replicate). The chicks were fed a corn–soybean meal basal diet, and divided into two groups: unchallenged and challenged with E. coli (400 mg/kg Escherichia coli inoculation). Each of these groups was divided into three study groups: untreated, treated with 0.05% Novacid, and treated with 400 mg/kg BMD. At day 42, inclusion of Novacid or BMD significantly (P < 0.05) improved the performance in the unchallenged groups relative to the control group. However, in E. coli -challenged groups, Novacid and BMD did not improve performance. Ileal digestibility of crude fat, crude protein, and gross energy were reduced in the Novacid group (P < 0.05). BMD and Novacid were equally effective in controlling ileal nutrient digestibility and feed coliform count (P < 0.05). Novacid reduced cecal E. coli and Salmonella count compared to BMD and control. Thus, a phytochemical feed supplement with organic acids and glucomannan could be an effective substitute for antibiotic growth promoters in broiler diets, but cannot replace antibiotics to counter potent infectious agents such as E. coli. Antrodia cinnamomea, a precious and unique medical fungus existing exclusively in Taiwan, exhibits antioxidant and immunomodulatory properties. This study was conducted to evaluate the beneficial effects of A. cinnamomea powder (ACP) and to further illuminate its underlying antioxidant and immunomodulation molecular mechanisms in broilers. The functional compounds of ACP—crude triterpenoids, crude polysaccharides, and total phenolic content—were assayed, respectively. Two-hundred-forty one-day-old broilers (Ross 308) were assigned to 4 treatment groups receiving dietary supplementation with ACP at 0, 0.1, 0.2, and 0.4% for 35 days. Each group had 4 replicate pens, with 15 birds per pen. During 1 to 21- and 22 to 35-day periods, chickens on ACP-supplemented diet demonstrated increased body weight gain, compared to those on the control diet, resulting in increased weight gain throughout the entire experimental period with an increased tendency in feed consumption yet no significant difference in FCR. Blood antioxidant potentiality, superoxide dismutase (SOD), increased in birds fed the supplemented diet at both 21 and 35 d, accompanied by higher catalase (CAT) activity at 21 days. In vivo peripheral blood mononuclear cells (PBMC) exposed to lipopolysaccharide (LPS) and 2,2΄-Azobis(2-amidinopropane) dihydrochloride (AAPH) capability showed that the diminished cell viability caused by both challenge factors was improved in ACP-supplemented groups. Antioxidant genes dominated by Nrf2 genes, such as HO-1 and GCLC, were up-regulated in 35-day-old birds. Inflammatory-related genes, such as IL-1β and IL-6, ruled mainly by NF-κB, were rather down-regulated by 0.2% ACP addition at 21 and 35 days. Protein expression of Nrf2 and NF-κB in the liver supported the mRNA results, demonstrating that all ACP-supplemented groups showed significantly higher Nrf2 expression, whereas the NF-κB was inhibited. In conclusion, preferable microbial balance may putatively indicate the improvement of immunomodulatory-related capacity by ACP. Furthermore, ACP could induce the Nrf2-dependent pathway and decrease the NF-κB-dominated inflammatory signaling pathway. Antioxidant and immune capacity in terms of antioxidant enzymes and cell tolerance also was elevated by ACP. Reducing Campylobacter spp. carriage in poultry is challenging, but essential to control this major cause of human bacterial gastroenteritis worldwide. Although much is known about the mechanisms and route of Campylobacter spp. colonization in poultry, the literature is scarce on antibiotic-free solutions to combat Campylobacter spp. colonization in poultry. In vitro and in vivo studies were conducted to investigate the role of TYPLEX® Chelate (ferric tyrosine), a novel feed additive, in inhibiting Campylobacter jejuni (C. jejuni) biofilm formation and reducing C. jejuni and Escherichia coli (E. coli) colonization in broiler chickens at market age. In an in vitro study, the inhibitory effect on C. jejuni biofilm formation using a plastic bead assay was investigated. The results demonstrated that TYPLEX® Chelate significantly reduces biofilm formation. In an in vivo study, 800 broilers (one d old) were randomly allocated to 4 dietary treatments in a randomized block design, each having 10 replicate pens with 20 birds per pen. At d 21, all birds were challenged with C. jejuni via seeded litter. At d 42, cecal samples were collected and tested for volatile fatty acid (VFA) concentrations and C. jejuni and E. coli counts. The results showed that TYPLEX® Chelate reduced the carriage of C. jejuni and E. coli in poultry by 2 and 1 log10 per gram cecal sample, respectively, and increased cecal VFA concentrations. In a market undergoing constant evolution, the production of chicken meat that consumers would perceive as "natural" and "animal friendly" is crucial. The use of probiotics in rurally reared chickens could represent a major opportunity to achieve mutual benefit for both the industry and consumers. A total of 264 male Kabir chicks were randomly distributed to one of 2 dietary treatments: the L group received a commercial feed supplemented with 2.0 g/100 kg of Lactobacillus acidophilus D2/CSL, while the C group received the same basal diet without the additive. To assess the effects of probiotic supplementation in the chickens' diet, productive performance was evaluated at d 21 and 42, whereas microbiological analyses of the intestinal content and intestinal histology and morphometry were performed at the end of the trial (d 42). At d 21 and 42, L birds showed better (P < 0.001) performance in terms of body weight, average daily gain, and feed conversion ratio. Enterococci, staphylococci, and Escherichia coli populations were not influenced by dietary treatment. On the contrary, Lactobacillus population increased (P = 0.032) in the L group. Furthermore, a tendency (P = 0.069) was observed for the coliforms to be influenced by diet, with lower values in the L group in comparison to the C group. Histological techniques revealed that the number of goblet cell containing neutral mucins was lower in the C group. Morphometric evaluations demonstrated that the probiotic supplementation increased the height of the mucosal layer by improving (P = 0.040) villus height, while crypt depth was unaffected. In conclusion, the results obtained in this study demonstrate that it is possible to use Lactobacillus acidophilus D2/CSL (CECT 4529) in rurally reared chicken breeds with positive effects on performance and gut health. Carotenoids have been used for many years as an added pigment to enhance egg yolk color. One such carotenoid, astaxanthin, has a strong antioxidant activity and is produced by several microorganisms, including the bacterium Paracoccus marcusii, which has shown promise to be used as a feed additive. Therefore, this study investigated the use of P. marcusii as a possible source of pigmentation in layer hen feed to enhance egg yolk color. Paracoccus marcusii was fed to hens in a sucrose solution (10% m/v). The hens were fed daily and all eggs were collected for analysis. Dilutions of egg contents were plated onto selective media to detect the presence of known food pathogens (E. coli, Listeria, and Salmonella). In the feeding trial, there was no negative effect on hen body weight, egg production, or overall egg quality. There was a significant increase (P < 0.05) in yolk color as well as an increase in whole egg and yolk weight. There were also no known food pathogens detected in any of the egg samples. To counter the ill effects of synthetic dyes, bacterial pigment production as an alternative is now one of the promising and emerging fields of research. This study was conducted to evaluate the applicability of Deinococcus genus on the egg quality traits in laying hens. In study I, 24 single comb White Leghorn layers were fed with various 1 wt % Deinococcus bacterial strains for 10 d. In study II, 84 brown Hendrix layers were fed with one of 4 diets containing 0, 0.2, 1, or 5 wt % Deinococcus sp. GKB-Aid 1995 powder for 12 wk. In study III, 60 White Leghorn laying hens were fed either with or without 1 wt % Deinococcus sp. GKB-Aid 1995 powder, 1 wt % Deinococcus sp. GKB-Aid 1995 granules, or 1 wt % Deinococcus sp. GKB-Aid 1995 oily granules for 10 successive d. In all of the experiments, feeding Deinococcus powder did not affect egg quality traits except for the yolk color. In particular, supplementation with all Deinococcus powder treatments changed the yolk color (P < 0.05) in study I, with the best pigmentation score obtained by D. grandis and Deinococcus sp. Corn with naturally occurring aflatoxin (AF), wheat with naturally occurring doxynivalenol (DON), and barley with naturally occurring zearalenone (ZEA) were used to make rations for feeding turkey hen poults to 6 weeks of age. Control rations with equal amounts of corn, wheat, and barley were also fed. The control rations did contain some DON while both sets of rations contained ZEA. Within each grain source, there were 4 treatments: the control ration plus 3 rations each with a different feed additive which were evaluated for the potential to lessen potential mycotoxin effects on bird performance and physiology. The additives were Biomin BioFix (2 lb/ton), Kemin Kallsil (4 lb/ton), and Nutriad UNIKE (3 lb/ton). The mycotoxin rations reduced poult body weight (2.31 vs. 2.08 ± 0.02 kg) and increased (worsened) poult feed conversion (1.47 vs. 1.51 ± 0.01) at 6 wk. Feeding the poults the mycotoxin feed also resulted in organ and physiological changes typical of feeding dietary aflatoxin although a combined effect of AF, DON, and ZEA which cannot be dismissed. The feed additives resulted in improved feed conversion to 6 wk in both grain treatment groups. The observed physiological effect of feeding the additives was to reduce relative gizzard weight for both groups and to lessen the increase in relative kidney weight for the birds fed the mycotoxin feed. In conclusion, the feed additives used in this study did alleviate the effect of dietary mycotoxins to some degree, especially with respect to feed conversion. Addition of vitamins and antioxidants has been long associated with increased immunity and are commonly used in the poultry industry; however, less is known regarding their use in broiler breeder hens. The objective of this study was to determine if feeding a complex of protected biofactors and antioxidants composed of vitamins and fermentation extracts to broiler breeder hens conferred resistance against Salmonella enterica serovar Enteritidis ( S. Enteritidis) in the progeny chicks. Three-day-old chicks from control- and supplement-fed hens were challenged with S. Enteritidis and necropsied 4- and 11-days postchallenge (dpc) to determine if there were differences in invasion and colonization. Serum and jejunum were evaluated for various cytokine and chemokine production. Fewer (P = 0.002) chicks from supplement-fed hens had detectable S. Enteritidis in the ceca (32.6%) compared to chicks from control-fed hens (64%). By 11 dpc, significantly (P < 0.001) fewer chicks from supplement-fed hens were positive for S. Enteritidis (liver [36%]; ceca [16%]) compared to chicks from the control hens (liver [76%]; ceca [76%]). The recoverable S. Enteritidis in the cecal content was also lower (P = 0.01) at 11 dpc. In additional to the differences in invasion and colonization, cytokine and chemokine production were distinct between the 2 groups of chicks. Chicks from supplement-fed hens had increased production of IL-16, IL-6, MIP-3α, and RANTES in the jejunum while IL-16 and MIP-1β were higher in the serum of chicks from the control-fed hens. By 11 dpc, production of IFN-γ was decreased in the jejunum of chicks from supplement-fed hens. Collectively, these data demonstrate adding a protected complex of biofactors and antioxidants to the diet of broiler breeder hens offers a measure of transgenerational protection to the progeny against S. Enteritidis infection and reduces colonization that is mediated, in part, by a robust and distinct cytokine and chemokine response locally at the intestine and systemically in the blood.
