Botulinum toxin, frequently used to manage focal limb spasticity, has been reported to affect both extrafusal and intrafusal fibers of the injected muscle. Since most studies have used spinal reflexes, it is difficult to isolate the intrafusal effects from extrafusal and central effects. In a paper by On et al. [7], both stretch and H-reflexes were used to examine the intrafusal effects of botulinum toxin injections. Revisiting the data from On et al. [7] presented a unique opportunity to describe a novel method of measuring the effect of botulinum toxin-A on muscle spindle activity in patients with spasticity. H-reflex, maximum M-wave, and Achilles tendon reflex were serially assessed in ten patients with stroke pre-, 2, 4, and 12 weeks post-botulinum. In order to assess the intrafusal effects, we subtracted the %change in H-reflex amplitude from baseline (representing extrafusal and central effects) from the %change in Achilles tendon reflex amplitude from baseline (representing intrafusal, extrafusal and central effects). Using this formula, our results suggest that botulinum induces significant chemodenervation of the intrafusal muscle fibers (33% decreases). Intrafusal effects were greatest at 2 weeks, but tapered off by 12 weeks post-botulinum (p0.017). We found a significant positive correlation between the intrafusal effects of botulinum toxin and changes in modified Ashworth scale. Our method of assessing the effects of botulinum toxin shows significant effect on intrafusal fibers, which correlates with clinical manifestation of spasticity. Future studies need to investigate ways to maximize intrafusal effects and minimize extrafusal effects of botulinum therapy.

Stereotyped responses must be suppressed at certain times during daily life, which can be difficult for patients with lesions in the frontal cortices. Neuropsychologists have used the rock-paper-scissors (RPS) task to evaluate patients' ability to suppress a stereotyped response. In this study, we measured functional magnetic resonance imaging signals to investigate how frontal cortex activities change corresponding to subjects' performance as they tried to lose (successfully inhibiting the typical response to win) when presented with a gesture signifying rock, paper, or scissors. Performance rates ranged from 50% to 100%, and results indicated that activation in the bilateral anterior part of the prefrontal cortex increased parametrically corresponding to subjects' successful performance. This result implies that the anterior prefrontal cortex plays a key role in the successful completion of a modified RPS task and may play a role in the suppression of stereotyped responses.

Highlights • Simultaneous MEG recordings from two persons during live interaction. • Left-lateralized involvement of sensorimotor cortex during natural conversation. • Phasic modulation of sensorimotor rhythm indexing preparation to own speaking turn., Although the main function of speech is communication, the brain bases of speaking and listening are typically studied in single subjects, leaving unsettled how brain function supports interactive vocal exchange. Here we used whole-scalp magnetoencephalography (MEG) to monitor modulation of sensorimotor brain rhythms related to the speaker vs. listener roles during natural conversation. Nine dyads of healthy adults were recruited. The partners of a dyad were engaged in live conversations via an audio link while their brain activity was measured simultaneously in two separate MEG laboratories. The levels of ∼10-Hz and ∼20-Hz rolandic oscillations depended on the speaker vs. listener role. In the left rolandic cortex, these oscillations were consistently (by ∼20%) weaker during speaking than listening. At the turn changes in conversation, the level of the ∼10 Hz oscillations enhanced transiently around 1.0 or 2.3 s before the end of the partner’s turn. Our findings indicate left-hemisphere-dominant involvement of the sensorimotor cortex during own speech in natural conversation. The ∼10-Hz modulations could be related to preparation for starting one’s own turn, already before the partner’s turn has finished.

Highlights • We used human fMRI and a novel computer-based task to study the effects of visual threat uncertainty on brain activity. • Lack of visual threat information increased activity in hippocampus, ventromedial prefrontal cortex and amygdala (regions involved in anxiety). • Presence of visual threat information increased activity in periaqueductal gray (involved in fear). • High trait-anxiety participants anticipated hippocampal activation when visual threat information was not provided., Recent theories distinguish anxiety from fear in the brain. Anxiety is associated with activation in ventromedial prefrontal cortex and hippocampus, while fear is associated with activation in periaqueductal gray, with amygdala involved in processing aspects of both emotional responses. These theories propose that the amount of information available about threat determines which of the two defensive responses is elicited, with fear and anxiety associated with well-defined and uncertain threats respectively. However, a direct test of this hypothesis is lacking. Here we provide such a test using fMRI to record participants’ brain activity while they performed a computer-based task which required to press a button to move an artificial agent to a target position while an artificial predator chased the agent. In one condition (associated with fear) the predator was visible, while in another condition (associated with anxiety) the predator was invisible. Ventromedial prefrontal cortex, hippocampus, and amygdala showed increased activity when the predator was invisible compared to visible, while the opposite effect was observed in periaqueductal gray. We also observed that participants with high but not low trait-anxiety showed an hippocampal activation with invisible threat at an earlier time stage during the trial. These findings help clarify the neural mechanisms that underlie different defensive emotions and shed light on how these mechanisms may contribute to exaggerated anxiety.

FOXO3a, the main isoform of FOXO transcription factors, mediates the cellular response to oxidative stress by regulating the expression of genes involved in DNA repair and glutamine metabolism, including glutamine synthetase (GS). Immunohistochemical investigation of the population-based neuropathology cohort of the Medical Research Council’s Cognitive Function and Ageing Study (MRC CFAS) demonstrates that nuclear retention of FOXO3a significantly correlates with a DNA damage response and with GS expression by astrocytes. Furthermore, we show that GS expression correlates with increasing Alzheimer-type pathology in this ageing cohort. Our findings suggest that in response to oxidative stress, the nuclear retention of FOXO3a in astrocytes upregulates expression of GS as a neuroprotective mechanism. However, the activity of GS may be compromised by increasing levels of oxidative stress in the ageing brain resulting in dysfunctional enzyme activity, neuronal excitotoxic damage and cognitive impairment.

The present study shows that postnatal handling (H: consisting of removing the pups from the nest twice daily and placing them individually in plastic cages lined with paper towel for a period of 10 min, between postnatal days 1 and 21) and/or environmental enrichment (E: for a period of 6 months) of Roman high- and low-avoidance (RHA/Verh and RLA/Verh) rats induced long-lasting decreases in emotional reactivity (i.e. reduced defecations in the open field, OF, and hole-board, HB, tests) as well as increases in exploratory behavior (i.e. head-dipping) in a manner dependent upon the rat line (there were ‘line × H’ and ‘line × E’ interactions). It is reported for the first time that RHA/Verh rats show more head-dipping behavior than RLA/Verh rats, and that the environmental treatments can increase head-dipping of RLA/Verh animals to the level shown by RHA/Verh rats.

A boiled extract of porcine cerebral cortex was fractionated on Sephadex G-75 and LH-20 followed by paper chromatography of the active fraction having inhibitory activity towards [3H]GABA binding to rat brain synaptic membranes. A ninhydrin-negative substance migrating more quickly than authentic GABA was identified as a copper-GABA complex. The complex inhibited specific [3H]GABA binding (IC50 approximately equal to 1 microM) and antagonized the anticonvulsant effect exerted by intraamygdaloid injection of diazepam. The effect of a synthetic copper-GABA complex was compared and found to be similar to the endogenous complex. Cu2+ alone has no affinity for the GABA recognition site but antagonizes the anticonvulsant effect of diazepam. Therefore, Cu2+ is suggested to be the pharmacologically active principle of the endogenous substance. Cu2+ does not seem to function via the recognition site of the GABA receptor.

Contains fulltext : 216041.pdf (Publisher’s version ) (Open Access) Experimental treatments for treating neuropathic pain include transcranial magnetic stimulation (TMS) and invasive electric motor cortex stimulation (iMCS) of the primary motor cortex (M1). Mechanisms of action of both methods, however, remain largely elusive. Within this paper, we focus on animal-based experiments in order to investigate the biological mechanisms that are involved in alleviating pain by use of TMS and/or iMCS. Therefore, this paper systematically reviewed the animal-based evidence on these mechanisms. Multiple online databases were systematically searched and retrieved articles were assessed using predefined inclusion and exclusion criteria. Twenty-three suitable articles were included; six on TMS and seventeen on iMCS. In general, iMCS and TMS were found to impact the primary motor cortex structure and function in animals. Furthermore, structural and functional changes within the thalamus, striatum, periaqueductal grey, rostral ventromedial medulla and dorsal horn were reported to occur. Although widespread, all areas in which structural and functional changes occurred after TMS and iMCS have been found to be interconnected anatomically. This could provide a rationale for future investigations of treating neuropathic pain by use of neuromodulation. 11 p.

Background With the tremendous advances in the field of brain–computer interfaces (BCI), the literature in this field has grown exponentially; examination of highly cited articles is a tool that can help identify outstanding scientific studies and landmark papers. This study examined the characteristics of 100 highly cited BCI papers over the past 10 years. Methods The Web of Science was searched for highly cited papers related to BCI research published from 2006 to 2015. The top 100 highly cited articles were identified. The number of citations and countries, and the corresponding institutions, year of publication, study design, and research area were noted and analyzed. Results The 100 highly cited articles had a mean of 137.1(SE: 15.38) citations. These articles were published in 45 high-impact journals, and mostly in TRANSACTIONS ON BIOMEDICAL ENGINEERING (n = 14). Of the 100 articles, 72 were original articles and the rest were review articles. These articles came from 15 countries, with the USA contributing most of the highly cited articles (n = 52). Fifty-seven institutions produced these 100 highly cited articles, led by Duke University (n = 7). Conclusions This study provides a historical perspective on the progress in the field of BCI, allows recognition of the most influential reports, and provides useful information that can indicate areas requiring further investigation.

International audience; This paper deals with a spontaneous, bursting neuronal activity which can not be altered by any stimulation in the periphery or voluntary actions or by cognitive tasks. An initial description of such units led to the conclusion that this activity was generated ectopically at the site of a previous or present impalement of a nerve fibre. The aim of the current study was to record a larger number of these units by using microneurography, in order to characterise their firing properties and particularly, see if any subtypes of units could be identified. In conclusion, this paper suggests that some of these discharges could be related to an injury of the nerve fibre, however most of them could not. Mirror neurons are a class of neurons in the ventral pre-motor cortex (area F5) and inferior parietal lobule (area PFG) that respond during the execution as well as the observation of goal-directed motor acts. These intriguing response properties stirred an intense debate in the scientific community with respect to the possible cognitive role of mirror neurons. The aim of the present review is to contribute to this debate by providing, in a single paper, an extended summary of 20 years of neurophysiological research on mirror neurons in the macaque. To this end, I provide a comprehensive description of the methodology and the main results of each paper about mirror neurons published since their first report in 1992. Particular care was devoted in reporting the different response characteristics and the percentages of neurons exhibiting them in relation to the total number of studied neurons. Furthermore, I also discuss recent results indicating that mirror neurons might not be confined to areas F5 and PFG and that "mirroring" might not be limited to action observation. Finally, I offer a unifying framework for many of the results discussed here by speculating that a potential functional role of mirror neurons might be, during action observation, to generalize from the particular grasping movement being observed to the "concept" of grasping.

Several data indicate that hypothalamic fatty acid synthesis pathway plays an important role in the control of food intake and energy expenditure in rodents. However, the confirmation of its physiological relevance in regulation of feeding in human remains incomplete. For fatty acid synthesis pathway to function as regulator of energy balance in human hypothalamus, acetyl-CoA carboxylase (ACC), fatty acid synthase (FAS) and other lipogenic enzymes activities must be present. The presence of FAS in human hypothalamic neurons has been shown by immunohistochemistry, but quantitative studies on FAS activity there has not been performed so far. There is no available data concerning ACC activity in human hypothalamus. Thus, we investigated ACC and FAS (as well as other lipogenic enzymes) activities in human hypothalamus of subjects who died in car accidents. The results presented in this paper indicate that ACC and FAS activities are present in human hypothalamus and that these activities are 2- to 3-fold lower than in rat hypothalamus. Moreover, our data presented in this paper indicate that other lipogenic enzymes activities are also present in human hypothalamus. The activity of FAS, ACC and other lipogenic enzymes in human hypothalamus suggests that fatty acid synthesis actively occurs there. Therefore, it is likely, that in human this pathway may be relevant to hypothalamic functioning as food intake and energy expenditure regulator, similarly as it was suggested in rodents.

Diffusion magnetic resonance imaging tractography allows investigating brain structural connections in a noninvasive way and has been widely used for understanding neurological disease. Quantification of brain connectivity along with its length by dividing a fiber bundle into multiple segments (node) is a powerful approach to assess biological properties, which is termed as tractometry. However, current tractometry methods face challenges in node identification along with the length of complex bundles whose morphology is difficult to summarize. In addition, the anatomic measure reflecting the macroscopic fiber cross-section has not been followed in previous tractometry. In this paper, we propose an automated fiber bundle quantification, which we refer to as ClusterMetric. The ClusterMetric uses a data-driven approach to identify fiber clusters corresponding to subdivisions of the white matter anatomy and identify consistent space nodes along the length of clusters across individuals. The proposed method is demonstrated by applicating to our collected dataset including 23 Alzheimer's disease (AD) patients and 22 healthy controls (HCs) and a public dataset of ADNI including 53 AD patients and 85 HCs. The altered white matter tracts in AD group are observed using both datasets, which involve several major fiber tracts including the corpus callosum, corona-radiata-frontal, arcuate fasciculus, inferior occipito-frontal fasciculus, uncinate fasciculus, thalamo-frontal, superior longitudinal fasciculus, inferior cerebellar peduncle, cingulum bundle, and extreme capsule. These fiber clusters represent the white matter connections that could be most affected in AD, suggesting the ability of our method in identifying potential abnormalities specific to local regions within a fiber cluster.

Subsequent recordings collected during a follow-up of our studieson blink-related EEG have not produced results overlapping thosereported in the previous paper [3]. In particular, the blink-relateddelta oscillation, that was a biphasic negative-positive waveformin the above-mentioned paper, has turned out to be reversed inpolarity (i.e., positive-negative) in the recent recordings.The only difference in the EEG data processing with respect tothe previous recordings was in the export file format: EDF (recent)vs ASCII (previous). We believe that the polarity originating fromthe EDF files (i.e., positive-negative) should be considered correct,because EEG data exported in the form of EDF files, according tothe specifications of the format itself, faithfully represent originalraw data [6,7]. On the contrary, data exported in the form of filetext (with an ASCII code) only allow reproducing signals visual-izedontherecordingdevicescreen.But,indoingso,thewaveformis reconstructed referring to an internal code, according to whichnegative values are assigned to the points situated in the inferiorpart of the picture (with respect to an equatorial line of symme-try). This poses a serious problem when the reference coordinatesof the recording device, concerning vice versa the polarity of thedeflections with respect to the isoelectric line, are such that posi-tive deflections are downward. In this case, in fact, the procedureof exporting data as ASCII files, by assigning negative values to thesamedeflections(becausetheyarereallydownwardfromaspatialpoint of view), gives rise to an error of inverted polarity. Hearing loss is a common neurodegenerative disease that can start at any stage of life. Misalignment of the auditory neural impairment may impose challenges in processing incoming auditory stimulus that can be measured using electroencephalography (EEG). The electrophysiological behaviour response emanated from EEG auditory evoked potential (AEP) requires highly trained professionals for analysis and interpretation. Reliable automated methods using techniques of machine learning would assist the auditory assessment process for informed treatment and practice. It is thus highly required to develop models that are more efficient and precise by considering the characteristics of brain signals. This study aims to provide a comprehensive review of several state-of-the-art techniques of machine learning that adopt EEG evoked response for the auditory assessment within the last 13 years. Out of 161 initially screened articles, 11 were retained for synthesis. The outcome of the review presented that the Support Vector Machine (SVM) classifier outperformed with over 80% accuracy metric and was recognized as the best suited model within the field of auditory research. This paper discussed the comprehensive iterative properties of the proposed computed algorithms and the feasible future direction in hearing impaired rehabilitation.

Computer aided diagnosis (CAD) methods for Parkinson's disease (PD) can assist clinicians in diagnosis and treatment. Magnetic resonance imaging (MRI) based CAD methods can help reveal structural changes in brain. Classifier is a key component in CAD system, which directly affects the classification performance. Privileged information (PI) can assist to train the classifier by providing additional information, which makes test samples have less error and improves the classification accuracy. In this paper, we proposed a PI based kernel ridge regression plus (KRR+) in diagnosis of PD. Specifically, morphometric features and brain network features are extracted from MRI. Then, Empirical Kernel Mapping (EKM) feature expression method is used to make the data separable in the high-dimensional space. Besides, we introduce self-paced learning (SPL) that can adaptively select the sample input training model, which can further improve the classification effect. The experimental results show that the proposed method is effective for PD diagnosis, its performance is superior to existing classification model. This method is helpful to assist clinicians in the diagnosis of PD and to find out possible neuroimaging biomarkers.

Secretory immunoglobulin A (sIgA) has been suggested to be a potential marker of stress in the human and the dog. We describe a simple method of saliva collection from rats for quantification of salivary sIgA in the first study of sIgA as a possible stress marker in the rat. Three groups of adult male rats were housed under different conditions (singly housed, paired with a female, and group housed). The animals were conditioned for the saliva collection by presenting a chocolate reward after every session and saliva was collected by soaking filter paper discs (5 mm in diameter) with saliva directly in the rats' oral cavity. The levels of salivary sIgA were determined by a novel paper disc rocket immunoelectrophoresis using sheep anti rat IgA antiserum. The resulting rockets had similar morphology as rockets in a standard rocket immunoelectrophoresis. Males housed singly showed stable sIgA levels with little variation while those housed singly with a female showed an initial decrease in sIgA followed by a steady increase. Males housed in a group (n = 6) showed a steady decline in sIgA levels. This indicates that sIgA levels are reduced by social stress and that this molecule may be a useful marker for well-being in the rat.

While the psychoactive stimulant nicotine has been the subject of extensive research, considerably less attention has focused on other compounds found in either tobacco smoke or that are nicotine metabolites. Recent papers have suggested that some of the compounds in question may either alter nicotine's effects or have reinforcing properties themselves, although they would only be experienced after consumption of tobacco. The potential for these compounds to function as reinforcers or to potentiate the reinforcing properties of nicotine merits investigation. To pursue this line of inquiry, we examined cotinine in a planarian model of environmental place preference. In the present study, planarians demonstrated that the compound cotinine, which is present in tobacco smoke, and is also the principal nicotine metabolite, establishes a conditioned place preference. These data represent the first ever demonstration that cotinine will establish a conditioned place preference in planarians and possibly contribute to the addictive properties of nicotine.

In a previous paper it has been reported that the repetitive conjunctive stimulation of both a granule cell and an inferior-olivary neuron depresses the synaptic transmission between a cerebellar granule cell and a Purkinje cell, and the repetitive stimulation of only a granule cell potentiates the transmission in a simple culture preparation. This paper demonstrates, by experiments controlling the membrane potential of a postsynaptic Purkinje cell, that the postsynaptic depolarization of a Purkinje cell during activation of a granule cell is both necessary and sufficient to induce the depression, and that the potentiation is induced without the postsynaptic depolarization.

As the most common cause of dementia, Alzheimer's disease (AD) is progressively neurodegenerative disease. In the initial stage, Alzheimer's disease is related to the memory disorder, followed by a serious progressive decline in cognitive function, and finally died. Neurofibrillary tangles (NFTs) deposited in neurons form one of the histopathological features of AD. NFTs are composed of abnormally modified forms, such as hyperphosphorylation, of tau protein. DNA methylation on Tau protein related genes in the brains of AD patients plays an important role in AD pathogenesis. In this paper, the process and role of gene methylation in abnormal Tau modification and aggregation in the development of Alzheimer's disease were discussed. The effect of DNA methylation on tau protein in the brain of patients with Alzheimer's disease will help to find new targets in the development of drugs for treating Alzheimer's disease.

Stress is a common seizure trigger in persons with epilepsy. The body's physiological response to stress is mediated by the hypothalamic-pituitary-adrenal (HPA) axis and involves a hormonal cascade that includes corticotropin releasing hormone (CRH), adrenocorticotropin releasing hormone (ACTH) and the release of cortisol (in humans and primates) or corticosterone (in rodents). The prolonged exposure to stress hormones may not only exacerbate pre-existing medical conditions including epilepsy, but may also increase the predisposition to psychiatric comorbidities. Hyperactivity of the HPA axis negatively impacts the structure and function of the temporal lobe of the brain, a region that is heavily involved in epilepsy and mood disorders like anxiety and depression. Seizures themselves damage temporal lobe structures, further disinhibiting the HPA axis, setting off a vicious cycle of neuronal damage and increasing susceptibility for subsequent seizures and psychiatric comorbidity. Treatments targeting the HPA axis may be beneficial both for epilepsy and for associated stress-related comorbidities such as anxiety or depression. This paper will highlight the evidence demonstrating dysfunction in the HPA axis associated with epilepsy which may contribute to the comorbidity of psychiatric disorders and epilepsy, and propose treatment strategies that may dually improve seizure control as well as alleviate stress related psychiatric comorbidities.

The paper is aimed to investigate the changes in cardiac function and peripheral neurotoxicity caused by postoperative chemotherapy in patients with breast cancer infected with human papillomavirus (HPV) by echocardiogram information technology. 68 patients with breast cancer undergoing anthracycline (ATC) drug chemotherapy were selected as the research subjects. The routine echocardiography was performed before and after each patient's chemotherapy. The MoCA score of the T2DM group was positively correlated with the node degree (r = 0.400, p = 0.043) and the node efficiency (r = 0.452, p = 0.021) of the right straight back. FBG is positively correlated with the node degree of the left occipital gyrus (r = 0.422, p = 0.023); HbAlc is related to the node degree of the left occipital gyrus (r = 0.372, p = 0.043) and the node degree of the left occipital gyrus (r = 0.382, p = 0.037) was positively correlated with the node intermediary (r = 0.388, p = 0.034) at the back of the right cingulate gyrus. The topological properties of the resting brain function network of T2DM patients with negative MRI findings have changed compared with normal people, indicating that T2DM is an important factor leading to brain function damage, further explaining the rs-fMRI technology and complex brain networks based on graph theory Analysis theory can be used as an effective method to study the changes of brain function in T2DM patients.

There are isolated instances in the literature that suggest the 'onion skin' phenomenon is not always present. That is, newly recruited high threshold motor units (MU) have higher discharge rates than previously recruited low threshold MUs. Therefore, the purpose of this paper was to investigate the presence of the 'onion skin' phenomenon in a large sample of intramuscular myoelectric recordings from low to maximal force outputs. Forty-eight participants performed rapid isometric dorsiflexion contractions at 20, 40, 60, 80 and 100 % MVC while intramuscular electrical activity was recorded. A bivariate frequency-distribution of the motor unit discharge rate and motor unit action potential peak-to-peak (P-P) amplitude was assessed. There was a significant difference in bivariate frequency-distribution across force levels (D's = 0.1083-0.3094, p's < 0.001). Newly recruited high threshold MUs did have lower discharge rates, but there was an increase in the presence of high threshold, large P-P amplitude MUs with higher discharge rates than lower threshold MUs (reverse onion skin) during the stable portion of the force output. The recruitment of high threshold MUs with higher discharge rates decreased the level of common drive from the cross-correlation (Rxy) = 0.79 at 20 % MVC to Rxy = 0.68 at 100 % MVC (p < 0.01), but it remained high. As the interference pattern becomes more complex with the recruitment of more motor units at higher force outputs, intramuscular electrodes may be more discriminating while recording motor unit activity leading to the identification of both the 'reverse onion skin' and 'onion skin' phenomenon being present.

Purpose Functional MRI has played a fundamental role in Parkinson's disease(PD) study. In this paper, we performed an independent component analysis (ICA) based on functional networks to reveal the intricate variations on the morphology and functional properties of brain. Our analysis aims at discovering the differences between PD patients with sensorimotor function impairment and normal controls(NC), thus helping to understand the coordination neurological function degeneration in PD objectively. Method We investigated the blood oxygen level dependent(BOLD) functional MRI obtained at a 3.0 T MRI scanner. 30 PD patients and 28 NC subjects underwent the scan in resting state. The signals of sensory and motor coordinative control areas in the sensorimotor, insula and cerebellum networks acquired by ICA(Independent Component Analysis)were applied to analyze the functional alterations. Decreased connections were detected between sensory and motor cortex in sensorimotor network and between cerebellum and insula network by FNC analysis in PD patients as well. Discussion Parkinson's disease derives from the degeneration of the dopaminergic neurons in substantia nigra, and results in decreased secretion of inhibitory neurotransmitter. The significant differences between PD and NC groups in our research maybe explain the clinical manifestations of prominent bradykinesia and multiple extrapyramidal symptoms.

It was accepted for a long time that in mammals there is only retinofugal neuronal connection between the eye and the pineal body (PB). In our previous paper we described that nerve cells were present in hamster PB and these neurons could establish a reverse connection with the retina through a transsynaptic pathway. In adult albino rats neuronal perikarya were not found. In this present experiment it was examined whether the lack of these nerve cells in the PB of adult rats is the result of an apoptotic phenomenon or the lack of migration during the fetal period. Green fluorescence protein expressing pseudorabies virus, spreading only in retrograde direction, was injected into the vitreous body of rats at various postnatal ages. Virus labeled cell bodies were not observed in the PB of adult rats; however, labeling with gradually decreasing number of cells was present in animals aged 3-6, 13-14, 20, 35 and 41 postnatal days. Injection of virus, spreading in anterograde direction (expressing red fluorescence protein), into the PB of young prepubertal animals resulted in labeling in the retina. This observation indicates that the pinealo-retinal connection in prepubertal period is active. Immunostaining revealed that some of the labeled neuronal perikarya showed activated caspase-3 (an apoptotic marker) immunoreactivity. Our results clearly show that the neurons migrate to the PB and later, during the prepubertal period, they disappear. Caspase-3 immnoreactivity indicates that these cells die off by apoptosis.

Background and Objective An accurate detection of neurodegenerative diseases (NDDs) definitely improves the life of patients and has attracted growing attention. Methods In this paper, a general automatic method for detection of Parkinson’s disease (PD), Amyotrophic lateral sclerosis (ALS) and Huntington’s disease (HD) has been proposed based on the localized time–frequency information of gait signals. The new main part of the detection method is to obtain a small set of sparse coefficients for the local representation of gait signals with appropriate time and frequency resolution. For this purpose, a hybrid feature set based on sparse matching pursuit decomposition and two sets of nonlinear and linear features has been developed. Then, principal components of the proposed feature have been analyzed using a sparse coding classifier. Results The proposed approach has achieved high average accuracy rates of 93%, 94%, and 97% for PD, ALS, and HD detection, respectively. Conclusions The obtained results have indicated that combination of time and frequency information of the gait signals through adaptive localized window length in MP makes it more efficient in comparison with the existing time, frequency or other time–frequency gait parameters. The great potential of nonlinear sparse features for PD and HD detection and linear ones for ALS detection has also been shown.

Innovative payment methods have been getting worldwide attention. Exploring the mechanisms behind consumers’ purchase behaviors modulated by different payment methods was critical but challenging. In this paper, we proposed a 2 (payment methods: cash payment vs mobile payment) × 2 (price levels of products: high vs low) Event-Related Potentials (ERPs) experiment to study the difference of cash payment and mobile payment on consumers' purchase intention of products in different price level from a neuroscience perspective. Greater P200 amplitude was found in mobile payment condition, which meant that mobile payment captured more early attention resources than cash. Larger N270 amplitude was found in cash payment condition as the participants had to spend more cognitive resources and struggled more when using cash. Moreover, lower N270 amplitude was found in high-price product condition, which indicated that when there were limited cognitive resources, the affective process played a dominating role. Specifically, buying the high-price products with mobile payment, the consumers would experience the pleasure of consumption more rather than the pain of paying. The study offers insights on the cognitive process of consumers when they pay with different methods.

In recent years complexity of the brain structure in healthy and disordered subjects has been studied increasingly. But to the best of the authors' knowledge, researchers so far have investigated the structural complexity only in the context of two restricted networks known as Small-World and Scale-free networks; whereas other aspects of the structural complexity of brain activities may be affected by aging and neurodegenerative disorders such as the Alzheimer's disease and autism spectrum disorder. In this study, two general complexity metrics of graphs, Graph Index Complexity and Offdiagonal Complexity are proposed as general measures of complexity, not restricted to SWN only. They are adopted to measure the structural complexity of the weighted graphs instead of the common binary graphs. Fuzzy Synchronization Likelihood is applied to the EEGs and their sub-bands, as a functional connectivity metric of the brain, to construct the functional connectivity graphs. Two applications are used to evaluate the efficacy of the complexity measures: diagnosis of autism and aging, both based on EEG. It was discovered that the Graph Index Complexity of gamma band is discriminative in distinguishing autistic children from non-autistic children. Also, Offdiagonal Complexity of theta band in young subjects was observed to be significantly different than old subjects. This study shows that changes in the structure of functional connectivity of brain in disorders and different healthy states can be revealed by unrestricted metrics of graph complexity. While the applications presented in this paper are based on EEG, the approach is general and can be used with other modalities such as fMRI, MEG, etc. Further, it can be used to study every other neurological and psychiatric disorder.

Cross frequency coupling is used to study the cross talk between brain oscillations. In this paper we focus on a special type of frequency coupling between brain and body oscillations, which is reflected by the numerical ratio (r) between two frequencies (m and n; n > m). This approach is motivated by theoretical considerations, indicating that during alert wakefulness brain-body oscillations form a coupled hierarchy of frequencies with integer relationships that are binary multiples (r = n:m = 1:2, 1:4, 1:8…..). During sleep we expect an irrational relationship (r = n/m = irrational number) between brain and body oscillations that reflects decoupling. We analyzed alpha frequency, heart rate, breathing frequency during performance of a memory tasks and in addition spindle frequency from data collected by the SIESTA sleep research group. As predicted, our results show a binary multiple frequency relationship between alpha, heart rate and breathing frequency during task performance but an irrational relationship between spindle frequency, heart rate and breathing frequency during sleep.

The neuropathic pain phenotype is the consequence of functional and morphological reorganization of the PNS and CNS. This reorganization includes DRGs and the spinal cord, and extends to multiple supraspinal areas including the limbic and reward systems. Several recent papers show that acute manipulation of cortical and subcortical brain areas causally correlates with the cognitive, emotional and sensory components of neuropathic pain, yet mechanisms responsible for pain chronification remain largely unknown. Here we show that nucleus accumbens expression of ΔFos-B, a transcription factor that plays a critical role in addiction and in the brain response to stress, is reduced long term following peripheral neuropathic injury. Conversely, boosting ΔFos-B expression in the nucleus accumbens by viral transfection causes a significant and long-lasting improvement of the neuropathic allodynia. We suggest that ΔFos-B in the nucleus accumbens is a key modulator of long term gene expression leading to pain chronification.

The relative sequence of morphological decomposition and whole-word recognition during morphologically complex word (MCW) processing has attracted much attention in recent years. However, most studies have only focused on inflected and derived words while disregarding compound words, and have mainly examined the differences between native speakers and L2 learners without addressing language proficiency levels. This paper investigates the language proficiency effect on L2 English learners’ processing of all the three types of MCWs in a masked transposed letter priming paradigm. Results showed that the high proficiency learners adhered to the Post-lexical Model in general, while the low proficiency learners presented a blurred tendency due to their poor whole-word memory and overall processing efficiency. Different morphological types caused gradable priming effects with compounds on the top of the continuum. In sum, language proficiency as well as morphological types impacts L2 learners’ MCW processing mechanisms.

Lidocaine hydrochloride (LC-HCl) and morphine hydrochloride (Mor-HCl) are two kinds of most prevalently used anesthetics. However, their influences on electrical excitability of hippocampal neuronal networks and hippocampal brain slices were rarely studied. Previously, our group have assessed the influence of acetylcholine, alcohol and temperature change on the excitability of neural networks with the so-called Voltage Threshold Measurement Method (VTMM) based on microelectrode array (MEA). In this paper, we will study the influence of LC-HCl and Mor-HCl on the electrical excitability of neural networks and the morphological features of neurons, and discuss the relations between the changes of electrical excitability of neural networks and the morphological changes of neurons. The results of VTMM showed: The voltage threshold (VTh) of hippocampal neuronal networks and hippocampal brain slices first increased and then decreased as the LC-HCl concentration increased. The VTh of hippocampal neuronal networks and hippocampal brain slices increased as the Mor-HCl concentration increased. The results of HCS experiments showed: The neurite length change of cultured hippocampal neuronal networks increased first and then decreased with increased LC-HCl concentration, but decreased as the Mor-HCl concentration increased. The combined analysis of VTMM and HCS experiments showed that under effects of the two drugs, the VTh and the hippocampal neurite length were strongly negatively correlated.

International audience; Changes in microglial development and morphology can be induced by inflammatory conditions and associated with eating or mood disorders, such as hyperphagia or depression. In a previous paper in the minipig model, we showed that maternal Western diet during gestation and lactation decreased hippocampus neurogenesis and food-rewarded cognitive abilities in the progeny. Whether these alterations are concomitant with a central inflammatory process in brain structures involved in learning and memory (hippocampus, HPC), cognitive (prefrontal cortex, PFC), or hedonic (orbitofrontal cortex, OFC) control of food intake is still unknown. EEG signal has been widely used in emotion recognition. However, too many channels and extracted features are used in the current EEG-based emotion recognition methods, which lead to the complexity of these methods This paper studies on feature extraction on EEG-based emotion recognition model to overcome those disadvantages, and proposes an emotion recognition method based on empirical mode decomposition (EMD) and sample entropy. The proposed method first employs EMD strategy to decompose EEG signals only containing two channels into a series of intrinsic mode functions (IMFs). The first 4 IMFs are selected to calculate corresponding sample entropies and then to form feature vectors. These vectors are fed into support vector machine classifier for training and testing. The average accuracy of the proposed method is 94.98% for binary-class tasks and the best accuracy achieves 93.20% for the multi-class task on DEAP database, respectively. The results indicate that the proposed method is more suitable for emotion recognition than several methods of comparison.

Human endogenous retrovirus W family (HERV-W) envelope (env) is known to be associated with neurological and psychiatric disorders, such as multiple sclerosis and schizophrenia. Previous studies showed that overexpression of HERV-W env could induce brain-derived neurotrophic factor (BDNF) gene expression. In human and rat cells, BDNF-mediated signal transduction might be modulated by glycogen synthase kinase 3β (GSK3β). Both BDNF and GSK3β are schizophrenia-related genes. In this paper, we investigated whether GSK3β was involved in the HERV-W env-induced expression of BDNF. We found that HERV-W env increased phosphorylation of GSK3β at Ser9 (p-GSK3β (Ser9)) and the ratio of p-GSK3β (Ser9) to total GSK3β (p0.05) in U251 cells. Overexpression of HERV-W env led to a 36.2% reduction in GSK3β activity compared to control (p0.05). The levels of β-catenin, cyclin D1 and TSC2 mRNAs were upregulated (p0.05). These data suggested that overexpression of HERV-W env might activate the GSK3β signaling pathway in U251 cells. Further, knockdown of GSK3β reduced the expression of total GSK3β, p-GSK3β (Ser9), and the ratio of p-GSK3β (Ser9) to total GSK3β by 28.6%, 50.4%, and 30.2%, respectively (p0.05). Levels of β-catenin, cyclin D1 and TSC2 mRNAs were also reduced (p0.05). Interestingly, GSK3β activity increased (p0.05). Knockdown of GSK3β also decreased mRNA and protein expression of BDNF by 49.9% and 48.5% respectively (p0.05). These results indicated that phosphorylation of GSK3β at Ser9 might be involved in HERV-W env-induced BDNF expression, and will hopefully improve our understanding of the role of HERV-W env in neurological and psychiatric diseases (schizophrenia, etc).

Genetic variants of AKT1 have been shown to influence brain function of Parkinson's disease (PD) patients, and in this paper our aim is to investigate the association between the three single-nucleotide polymorphisms (rs2498799; rs2494732; rs1130214) and PD in Han Chinese. 413 Han Chinese PD patients and 450 healthy age and gender-matched controls were genotyped using the Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method. Both the patient and control groups show similar genotype frequencies at the three loci: rs2498799, rs2494732 and rs1130214. We are able to identify a significant difference in the frequencies of genotype (p=0.019) and G allele (OR=0.764, 95% CI=0.587-0.995, p=0.045) both at rs2498799 between the patient and control groups. Furthermore, the association of subjects with GG genotypes versus those with GA+AA genotype remain significant after adjusting for age in the Han Chinese female cohort (OR=0.538, 95%CI=0.345-0.841, p=0.006), which is especially evident in the late-onset cohort (OR=0.521, 95%CI=0.309-0.877, p=0.012). In contrast, allele frequencies at rs2494732 and rs1130214 were similar between patients and controls in all subgroup analyses. These results suggest that polymorphism of AKT1 locus is associated with risk of PD and that the G allele at rs2498799 may decrease the risk of PD in the North-eastern part of Han Chinese female population.

The vascular hypothesis of Alzheimer's disease postulates that disruption of the brain microvasculature is important for the accumulation of amyloid beta and increased neuroinflammation. Liver X Receptor agonist, GW3965, has been demonstrated to successfully modulate neuroinflammation and lipid metabolism in murine models of AD. This is partially due to increased expression of ApoE levels and increased mobility of endothelial progenitor cells. This paper analyzes changes in the neurovascular unit and in astrocytes and microglia markers following oral administration of GW3965 in a very old triple transgenic AD mice (3xTg-AD mice). We found that astrogliosis, but not activation of microglia, decreased in very old (24 months) 3xTg-AD mice treated with GW965. In addition, GW3965 increased LRP1 levels in neuron-like cells and partially restored microvascular morphology by decreasing tortuosity and increasing length as shown by Lectin immunostaining. Interestingly, these changes were associated with decreased Aβ in blood vessels. In conclusion, short-term treatment of 3xTg-AD mice with GW3965 restored microvascular architecture which may be important in the cognitive improvement previously shown.

The basal ganglia (BG) is involved in higher order motor control such as movement planning and execution of complex motor synergies. Neuroimaging study on stroke patients specifically with BG lesions would help to clarify the consequence of BG damage on motor control. In this paper, we performed a longitudinal study in the stroke patients with lesions in BG regions across three motor recovery stages, i.e., less than 2week (Session 1), 1-3m (Session 2) and more than 3m (Session 3). The patients showed an activation shift from bilateral hemispheres during early sessions (3m) to the ipsilesional cortex in late session (3m), suggesting a compensation effect from the contralesional hemisphere during motor recovery. We found that the lateralization of cerebellum(CB) for affected hand task correlated with patients' concurrent Fugl-Meyer index (FMI) in Session 2. Moreover, the cingulate cortex lateralization index in Session 2 was shown to significantly correlate with subsequent FMI change between Session 3 and Session 2, which serves as a prognostic marker for motor recovery. Our findings consolidated the close interactions between BG and CB during the motor recovery after stroke. The dominance of activation in contralateral cingulate cortex was associated with a better motor recovery, suggesting the important role of ipsilesional attention modulation in the early stage after BG stroke.

Objective To study the CT and MRI imaging manifestations of central and intraventricular central neurocytoma in the ventricle. Methods In this paper, 39 patients with central nervous cell tumour treated in our hospital from August 2015 to June 2018 were selected. All patients were performed plain CT scans using GE Highspeed CT; GE signa Twin speed 1.5 T superconducting magnet Resonance scanners were used to perform MRI plain scans on all patients. Observe the specific location, size, and morphology of tumours in the supra- and sub-ventricular ventricles of 39 central nervous cell tumour patients, and compare the performance of CT examination with the performance of MRI examination. Results Of the 39 patients with central neurocytoma, 13 were in the right lateral ventricle, 9 were in the left lateral ventricle, 11 patients had tumours in both lateral ventricles, and 6 patients had bilateral ventricles and bilateral ventricles. There were tumours in the third ventricle; 39 patients had an average tumour size of 52 mm; 36 patients had irregular lobes, and 3 patients had blurred tumour boundaries; some tumours had different degrees of calcification. Conclusions The location and characteristics of the imaging manifestations of central nervous cell tumours are typical. Both CT scans and MRI scans can effectively detect central neuroblastomas, and MRI imaging examinations can effectively improve the diagnosis accuracy of tumour is better than that of CT.

In this paper, we report the results of treating cells with an effective small molecule, (+)4-cholesten-3-one (PubChem CID: 91477), which can promote neural stem cell(NSC) differentiation into dopaminergic neurons. This study used rat neural stem cells stimulated with two different concentrations (7.8 μM and 78 μM) of (+)4-cholesten-3-one. Cell phenotypic analysis showed that (+)4-cholesten-3-one induced NSC differentiation into dopaminergic neurons, and the level of tyrosine hydroxylase(TH), which is specific for dopaminergic cells, was significantly increased compared with that of the drug-free control group. Furthermore, in this study, we found that this effect may be related to the transcription factor fork-head box a2 (FoxA2) and ten-eleven translocation 1 (TET1). The expression of TET1 and FoxA2 was upregulated after treatment with (+)4-cholesten-3-one. To verify the relationship between (+)4-cholesten-3-one and these genes, we found that the binding rate of TET1 and FoxA2 increased after the application of (+)4-cholesten-3-one, as confirmed by a coimmunoprecipitation (Co-IP) assay. With a small interfering RNA (siRNA) experiment, we found that only when Tet1 and Foxa2 were not silenced was the mRNA level of Th increased after (+)4-cholesten-3-one treatment. Taken together, these data show that (+)4-cholesten-3-one can promote the differentiation of NSCs into dopaminergic neurons by upregulating the expression of TET1 and FoxA2 and by increasing their binding. Thus, (+)4-cholesten-3-one may help address the application of neural stem cell replacement therapy in neurodegenerative diseases.

Objective Fatal central nervous system (CNS) malformation is one of the most common congenital malformations, and is an important cause of infant mortality. Ultrasound diagnostic technology has the advantages of fast, safe, economic, convenient and real-time dynamic imaging, and it is the first choice for imaging diagnosis of the foetus. This paper studies the diagnosis of fatal central nervous system malformations based on prenatal Doppler ultrasound, and further accumulates clinical experience for prenatal diagnosis. Methods 320 pregnant women who underwent prenatal diagnosis in our hospital from August 2017 to December 2018. Ultrasound showed abnormal fatal craniocerebral development and MRI examination of fatal cranial brain was performed on the same day or the next day. The PHILIPS IU22 colour Doppler ultrasound system was used to scan pregnant women. Ultrasound showed 320 cases of fatal craniocerebral abnormalities and MRI diagnosis on the same day or the next day, of which 122 cases were consistent with MRI results, accounting for 38%; 173 cases were supplemented with MRI diagnostic information, accounting for 54%; MRI corrected diagnostic information There were 16 cases, accounting for 5%; 9 cases were abnormal, but 3 cases were not revealed by MRI. Conclusion Ultrasound examination during pregnancy has a high detection rate for the diagnosis of fatal CNS malformations. As the preferred imaging method for prenatal diagnosis, it is essential for timely and early screening of fatal malformations. MRI has a higher diagnostic value for fatal CNS malformations, and can play an important role in supplementing and misdiagnosing cases that have been missed by ultrasound. Therefore, in the prenatal diagnosis, attention should be paid to combining these two inspection methods, each taking its advantages and applying it to clinical.

Objective To analyse the traditional MRI signs, diffusion weighted imaging (DWI), perfusion weighted imaging (PWI), and hydrogen proton magnetic resonance spectroscopy (1H-MRS) of intracranial primary central nervous system lymphoma in immunocompetent people. To explore its diagnostic value. Methods The paper retrospectively analysed the MRI signs of 12 patients with primary central nervous system lymphoma confirmed by surgery and pathology. Results Twelve patients were all B-cell lymphoma. A total of 13 nodules were detected, 11 were single and 1 was multiple. Lesions T1WI showed low or equal signal shadows, T2WI showed equal, slightly higher, high signal shadows, DWI showed high and low signal shadows; the boundaries of the lesions were clearer, and mild to moderate enema was visible around them; most of the lesions showed obvious uniform enhancement and "gap" Signs "and" spike signs "; 1H-MRS showed a moderate decrease in N-acetyl aspartate (NAA) peak, an increase in choline (Cho) peak, and a slight decrease in creatine (Cr) peak, which can be seen Huge lipid peak. Perfusion-weighted imaging suggested that the lesions were hypoperfusion nodules. Conclusion The combined application of traditional magnetic resonance imaging, diffusion weighted imaging, perfusion weighted imaging, and 1H-MRS can improve the diagnosis rate of primary central nervous system lymphoma.

The paper is aimed to detect atrial fibrillation (AF) based on convolutional neural network (CNN) in artificial intelligence environment and to analyze its cause location. The paper was to study the infection factors of chronic glomerular disease and peripheral nerve conduction under information health data to deepen the understanding of the disease. By collecting the data of existing patients, the basic situation of infection in patients with chronic glomerular disease was analyzed, and the infection factors of chronic glomerular disease were analyzed. At the same time, the electron microscope was used for assisted observation to understand the situation of different kidney diseases. The results showed that the infection site was mainly lung (81.1%), followed by the blood line (7.9%). Pathogen infection was mainly bacterial infection (38.9%), and the virus (22%) was second. Mild cognitive impairment (MCI) is an early sign of Alzheimer's disease (AD) which is the fourth leading disease mostly found in the aged population. Early intervention of MCI will possibly delay the progress towards AD, and this makes it very important to diagnose early MCI (EMCI). However, it is very difficult since the subtle difference between EMCI and cognitively normal control (NC). For improving classification performance, this paper presents a deep learning based diagnosis approach using structure MRI images for exploiting deeply embedded diagnosis features; then a feature selection strategy is performed to eliminate redundant features. A Support Vector Machine (SVM) is further employed to distinguish EMCI from NC. Experiments were performed on the publicly available ADNI dataset with a total of 120 subjects. The classification results demonstrate the superior performance of the proposed method with accuracy of 89.4% for EMCI versus NC.

Multiple studies provide evidence to support dysfunction of glutamate neurotransmission in the pathogenesis of drug dependence. Pharmacogenetic investigation of glutamate-related genes has provided further support for the involvement of this neurotransmitter in the risk of, and consequences of, drug abuse and dependence. This paper aims to provide a brief review of these association studies. Findings involving single nucleotide polymorphisms (SNPs) in glutamate receptor genes (GRIN, GRIA) and glutamate transporter genes (SLC1A, SLC17A) are reviewed as potential risk factors. As yet a clear perspective of the functional consequences and interactions of the various reported findings is lacking.

Progress in PTSD pharmacotherapy has lagged far behind that of other major mental illnesses. Unfortunately, due to the enormous costs and lengthy process involved in bringing drugs to market, delivering new treatments to patients with PTSD in the near future will remain a challenge. However, by capitalizing on recent advances in the pharmacogenetics of antidepressants, precision psychiatry approaches can be leveraged to optimize the delivery of currently-available medications in a fraction of the time and cost required to develop novel therapeutics. This paper provides a review of the pharmacogenetics of the four serotonin reuptake inhibitors (SRIs) that are currently endorsed for the treatment of PTSD (paroxetine, sertraline, fluoxetine and venlafaxine). It focuses on genes involved in SRI pharmacokinetics (including the liver enzyme genes CYP2D6 and CYP2C19 and blood-brain barrier-relevant gene ABCB1) as well as those implicated in both SRI pharmacodynamics and the pathophysiology of PTSD and related conditions (e.g., BDNF, FKBP5, HTR1A, HTR2A, TPH2). The review concludes with an overview of emerging commercial platforms for pharmacogenetic-guided prescription and a discussion of challenges and directions for future pharmacogenetic research on PTSD.
