Atomic nuclei are finite quantum systems composed of two distinct types of fermion-protons and neutrons. In a manner similar to that of electrons orbiting in an atom, protons and neutrons in a nucleus form shell structures. In the case of stable, naturally occurring nuclei, large energy gaps exist between shells that fill completely when the proton or neutron number is equal to 2, 8, 20, 28, 50, 82 or 126 (ref. 1). Away from stability, however, these so-called 'magic numbers' are known to evolve in systems with a large imbalance of protons and neutrons. Although some of the standard shell closures can disappear, new ones are known to appear. Studies aiming to identify and understand such behaviour are of major importance in the field of experimental and theoretical nuclear physics. Here we report a spectroscopic study of the neutron-rich nucleus 54Ca (a bound system composed of 20 protons and 34 neutrons) using proton knockout reactions involving fast radioactive projectiles. The results highlight the doubly magic nature of 54Ca and provide direct experimental evidence for the onset of a sizable subshell closure at neutron number 34 in isotopes far from stability. Basaltic lavas erupted at some oceanic intraplate hotspot volcanoes are thought to sample ancient subducted crustal materials. However, the residence time of these subducted materials in the mantle is uncertain and model-dependent, and compelling evidence for their return to the surface in regions of mantle upwelling beneath hotspots is lacking. Here we report anomalous sulphur isotope signatures indicating mass-independent fractionation (MIF) in olivine-hosted sulphides from 20-million-year-old ocean island basalts from Mangaia, Cook Islands (Polynesia), which have been suggested to sample recycled oceanic crust. Terrestrial MIF sulphur isotope signatures (in which the amount of fractionation does not scale in proportion with the difference in the masses of the isotopes) were generated exclusively through atmospheric photochemical reactions until about 2.45 billion years ago. Therefore, the discovery of MIF sulphur in these young plume lavas suggests that sulphur-probably derived from hydrothermally altered oceanic crust-was subducted into the mantle before 2.45 billion years ago and recycled into the mantle source of Mangaia lavas. These new data provide evidence for ancient materials, with negative Δ33S values, in the mantle source for Mangaia lavas. Our data also complement evidence for recycling of the sulphur content of ancient sedimentary materials to the subcontinental lithospheric mantle that has been identified in diamond-hosted sulphide inclusions. This Archaean age for recycled oceanic crust also provides key constraints on the length of time that subducted crustal material can survive in the mantle, and on the timescales of mantle convection from subduction to upwelling beneath hotspots. The snowball Earth hypothesis postulates that the planet was entirely covered by ice for millions of years in the Neoproterozoic era, in a self-enhanced glaciation caused by the high albedo of the ice-covered planet. In a hard-snowball picture, the subsequent rapid unfreezing resulted from an ultra-greenhouse event attributed to the buildup of volcanic carbon dioxide (CO2) during glaciation. High partial pressures of atmospheric CO2 (; from 20,000 to 90,000?p.p.m.v.) in the aftermath of the Marinoan glaciation (?635?Myr ago) have been inferred from both boron and triple oxygen isotopes. These values are 50 to 225 times higher than present-day levels. Here, we re-evaluate these estimates using paired carbon isotopic data for carbonate layers that cap Neoproterozoic glacial deposits and are considered to record post-glacial sea level rise. The new data reported here for Brazilian cap carbonates, together with previous ones for time-equivalent units, provide estimates lower than 3,200?p.p.m.v.-and possibly as low as the current value of ?400?p.p.m.v. Our new constraint, and our re-interpretation of the boron and triple oxygen isotope data, provide a completely different picture of the late Neoproterozoic environment, with low atmospheric concentrations of carbon dioxide and oxygen that are inconsistent with a hard-snowball Earth. The stable isotope ratios of atmospheric CO2 (18O/16O and 13C/12C) have been monitored since 1977 to improve our understanding of the global carbon cycle, because biosphere-atmosphere exchange fluxes affect the different atomic masses in a measurable way. Interpreting the 18O/16O variability has proved difficult, however, because oxygen isotopes in CO2 are influenced by both the carbon cycle and the water cycle. Previous attention focused on the decreasing 18O/16O ratio in the 1990s, observed by the global Cooperative Air Sampling Network of the US National Oceanic and Atmospheric Administration Earth System Research Laboratory. This decrease was attributed variously to a number of processes including an increase in Northern Hemisphere soil respiration; a global increase in C4 crops at the expense of C3 forests; and environmental conditions, such as atmospheric turbulence and solar radiation, that affect CO2 exchange between leaves and the atmosphere. Here we present 30 years' worth of data on 18O/16O in CO2 from the Scripps Institution of Oceanography global flask network and show that the interannual variability is strongly related to the El Niño/Southern Oscillation. We suggest that the redistribution of moisture and rainfall in the tropics during an El Niño increases the 18O/16O ratio of precipitation and plant water, and that this signal is then passed on to atmospheric CO2 by biosphere-atmosphere gas exchange. We show how the decay time of the El Niño anomaly in this data set can be useful in constraining global gross primary production. Our analysis shows a rapid recovery from El Niño events, implying a shorter cycling time of CO2 with respect to the terrestrial biosphere and oceans than previously estimated. Our analysis suggests that current estimates of global gross primary production, of 120 petagrams of carbon per year, may be too low, and that a best guess of 150-175 petagrams of carbon per year better reflects the observed rapid cycling of CO2. Marine and ice-core records show that the Earth has experienced a succession of glacials and interglacials during the Quaternary (last ?2.6 million years), although it is often difficult to correlate fragmentary terrestrial records with specific cycles. Aminostratigraphy is a method potentially able to link terrestrial sequences to the marine isotope stages (MIS) of the deep-sea record. We have used new methods of extraction and analysis of amino acids, preserved within the calcitic opercula of the freshwater gastropod Bithynia, to provide the most comprehensive data set for the British Pleistocene based on a single dating technique. A total of 470 opercula from 74 sites spanning the entire Quaternary are ranked in order of relative age based on the extent of protein degradation, using aspartic acid/asparagine (Asx), glutamic acid/glutamine (Glx), serine (Ser), alanine (Ala) and valine (Val). This new aminostratigraphy is consistent with the stratigraphical relationships of stratotypes, sites with independent geochronology, biostratigraphy and terrace stratigraphy. The method corroborates the existence of four interglacial stages between the Anglian (MIS 12) and the Holocene in the terrestrial succession. It establishes human occupation of Britain in most interglacial stages after MIS 15, but supports the notion of human absence during the Last Interglacial (MIS 5e). Suspicions that the treeless 'optimum of the Upton Warren interstadial' at Isleworth pre-dates MIS 3 are confirmed. The release of methane into the atmosphere through destabilization of clathrates is a positive feedback mechanism capable of amplifying global warming trends that may have operated several times in the geological past. Such methane release is a hypothesized cause or amplifier for one of the most drastic global warming events in Earth history, the end of the Marinoan 'snowball Earth' ice age, ∼635 Myr ago. A key piece of evidence supporting this hypothesis is the occurrence of exceptionally depleted carbon isotope signatures (δ13CPDB down to −48‰; ref. 8) in post-glacial cap dolostones (that is, dolostone overlying glacial deposits) from south China; these signatures have been interpreted as products of methane oxidation at the time of deposition. Here we show, on the basis of carbonate clumped isotope thermometry, 87Sr/86Sr isotope ratios, trace element content and clay mineral evidence, that carbonates bearing the 13C-depleted signatures crystallized more than 1.6 Myr after deposition of the cap dolostone. Our results indicate that highly 13C-depleted carbonate cements grew from hydrothermal fluids and suggest that their carbon isotope signatures are a consequence of thermogenic methane oxidation at depth. This finding not only negates carbon isotope evidence for methane release during Marinoan deglaciation in south China, but also eliminates the only known occurrence of a Precambrian sedimentary carbonate with highly 13C-depleted signatures related to methane oxidation in a seep environment. We propose that the capacity to form highly 13C-depleted seep carbonates, through biogenic anaeorobic oxidation of methane using sulphate, was limited in the Precambrian period by low sulphate concentrations in sea water. As a consequence, although clathrate destabilization may or may not have had a role in the exit from the 'snowball' state, it would not have left extreme carbon isotope signals in cap dolostones. Diffusion in condensed phases is a ubiquitous but poorly understood phenomenon. For example, chemical diffusion, which is the transport of matter associated with chemical concentration gradients (Fick's law), is treated as a separate process from thermal transport (the Soret effect), which is mass transport induced by temperature gradients. In the past few years, large variations in the proportions of isotopes of Mg, Ca, Fe, Si and O found in silicate melts subject to thermal gradients have been found, but no physical mechanism has been proposed. Here we present a model of diffusion in natural condensed systems that explains both the chemical and isotopic fractionation of Mg, Ca and Fe in high-temperature geochemical melts. Despite the high temperatures associated with these melts (T > 1,000 °C), we find that consideration of the quantum-mechanical zero-point energy of diffusing species is essential for understanding diffusion at the isotopic level. Our model explains thermal and chemical mass transport as manifestations of the same underlying diffusion mechanism. This work promises to provide insights into mass-transport phenomena (diffusion and evaporation) and associated isotopic fractionations in a wide range of natural condensed systems, including the atmospheric water cycle, geological and geochemical systems and the early Solar System. Widespread anoxia in the ocean is frequently invoked as a primary driver of mass extinction as well as a long-term inhibitor of evolutionary radiation on early Earth. In recent biogeochemical studies it has been hypothesized that oxygen deficiency was widespread in subsurface water masses of later Cambrian oceans, possibly influencing evolutionary events during this time. Physical evidence of widespread anoxia in Cambrian oceans has remained elusive and thus its potential relationship to the palaeontological record remains largely unexplored. Here we present sulphur isotope records from six globally distributed stratigraphic sections of later Cambrian marine rocks (about 499 million years old). We find a positive sulphur isotope excursion in phase with the Steptoean Positive Carbon Isotope Excursion (SPICE), a large and rapid excursion in the marine carbon isotope record, which is thought to be indicative of a global carbon cycle perturbation. Numerical box modelling of the paired carbon sulphur isotope data indicates that these isotope shifts reflect transient increases in the burial of organic carbon and pyrite sulphur in sediments deposited under large-scale anoxic and sulphidic (euxinic) conditions. Independently, molybdenum abundances in a coeval black shale point convincingly to the transient spread of anoxia. These results identify the SPICE interval as the best characterized ocean anoxic event in the pre-Mesozoic ocean and an extreme example of oxygen deficiency in the later Cambrian ocean. Thus, a redox structure similar to those in Proterozoic oceans may have persisted or returned in the oceans of the early Phanerozoic eon. Helium is a powerful tracer of primitive material in Earth’s mantle. Extremely high 3He/4He ratios in some ocean-island basalts suggest the presence of relatively undegassed and undifferentiated material preserved in Earth’s mantle. However, terrestrial lavas with high 3He/4He ratios have never been observed to host the primitive lead-isotopic compositions that are required for an early (roughly 4.5 Gyr ago) formation age. Here we show that Cenozoic-era Baffin Island and West Greenland lavas, previously found to host the highest terrestrial-mantle 3He/4He ratios, exhibit primitive lead-isotope ratios that are consistent with an ancient mantle source age of 4.55–4.45?Gyr. The Baffin Island and West Greenland lavas also exhibit 143Nd/144Nd ratios similar to values recently proposed for an early-formed (roughly 4.5 Gyr ago) terrestrial mantle reservoir. The combined helium-, lead- and Nd-isotopic compositions in Baffin Island and West Greenland lavas therefore suggest that their source is the most ancient accessible reservoir in the Earth’s mantle, and it may be parental to all mantle reservoirs that give rise to modern volcanism. The phenomenon of thermal diffusion (mass diffusion driven by a temperature gradient, known as the Ludwig–Soret effect) has been investigated for over 150 years, but an understanding of its underlying physical basis remains elusive. A significant hurdle in studying thermal diffusion has been the difficulty of characterizing it. Extensive experiments over the past century have established that the Soret coefficient, ST (a single parameter that describes the steady-state result of thermal diffusion), is highly sensitive to many factors. This sensitivity makes it very difficult to obtain a robust characterization of thermal diffusion, even for a single material. Here we show that for thermal diffusion experiments that span a wide range in composition and temperature, the difference in ST between isotopes of diffusing elements that are network modifiers (iron, calcium and magnesium) is independent of the composition and temperature. On the basis of this finding, we propose an additive decomposition for the functional form of ST and argue that a theoretical approach based on local thermodynamic equilibrium holds promise for describing thermal diffusion in silicate melts and other complex solutions. Magmatic production on Earth is dominated by asthenospheric melts of basaltic composition that have mostly erupted at mid-ocean ridges. The timescale for segregation and transport of these melts, which are ultimately responsible for formation of the Earth’s crust, is critically dependent on the permeability of the partly molten asthenospheric mantle, yet this permeability is known mainly from semi-empirical and analogue models. Here we use a high-pressure, high-temperature centrifuge, at accelerations of 400g–700g, to measure the rate of basalt melt flow in olivine aggregates with porosities of 5–12 per cent. The resulting permeabilities are consistent with a microscopic model in which melt is completely connected, and are one to two orders of magnitude larger than predicted by current parameterizations. Extrapolation of the measurements to conditions characteristic of asthenosphere below mid-ocean ridges yields proportionally higher transport speeds. Application of these results in a model of porous-media channelling instabilities yields melt transport times of ∼1–2.5 kyr across the entire asthenosphere, which is sufficient to preserve the observed 230Th excess of mid-ocean-ridge basalts and the mantle signatures of even shorter-lived isotopes such as 226Ra (refs 5,11–14). Accretion left the terrestrial planets depleted in volatile components. Here I examine evidence for the hypothesis that the Moon and the Earth were essentially dry immediately after the formation of the Moon—by a giant impact on the proto-Earth—and only much later gained volatiles through accretion of wet material delivered from beyond the asteroid belt. This view is supported by U–Pb and I–Xe chronologies, which show that water delivery peaked ∼100 million years after the isolation of the Solar System. Introduction of water into the terrestrial mantle triggered plate tectonics, which may have been crucial for the emergence of life. Geochemical data suggest that oxygenation of the Earth’s atmosphere occurred in two broad steps. The first rise in atmospheric oxygen is thought to have occurred between ∼2.45 and 2.2 Gyr ago, leading to a significant increase in atmospheric oxygen concentrations and concomitant oxygenation of the shallow surface ocean. The second increase in atmospheric oxygen appears to have taken place in distinct stages during the late Neoproterozoic era (∼800–542 Myr ago), ultimately leading to oxygenation of the deep ocean ∼580 Myr ago, but details of the evolution of atmospheric oxygenation remain uncertain. Here we use chromium (Cr) stable isotopes from banded iron formations (BIFs) to track the presence of Cr(VI) in Precambrian oceans, providing a time-resolved picture of the oxygenation history of the Earth’s atmosphere–hydrosphere system. The geochemical behaviour of Cr is highly sensitive to the redox state of the surface environment because oxidative weathering processes produce the oxidized hexavalent [Cr(VI)] form. Oxidation of reduced trivalent [Cr(III)] chromium on land is accompanied by an isotopic fractionation, leading to enrichment of the mobile hexavalent form in the heavier isotope. Our fractionated Cr isotope data indicate the accumulation of Cr(VI) in ocean surface waters ∼2.8 to 2.6 Gyr ago and a likely transient elevation in atmospheric and surface ocean oxygenation before the first great rise of oxygen 2.45–2.2 Gyr ago (the Great Oxidation Event). In ∼1.88-Gyr-old BIFs we find that Cr isotopes are not fractionated, indicating a decline in atmospheric oxygen. Our findings suggest that the Great Oxidation Event did not lead to a unidirectional stepwise increase in atmospheric oxygen. In the late Neoproterozoic, we observe strong positive fractionations in Cr isotopes (δ53Cr up to +4.9‰), providing independent support for increased surface oxygenation at that time, which may have stimulated rapid evolution of macroscopic multicellular life. Primary production in over half of the world’s oceans is limited by fixed nitrogen availability. The main loss term from the fixed nitrogen inventory is the production of dinitrogen gas (N2) by heterotrophic denitrification or the more recently discovered autotrophic process, anaerobic ammonia oxidation (anammox). Oceanic oxygen minimum zones (OMZ) are responsible for about 35% of oceanic N2 production and up to half of that occurs in the Arabian Sea. Although denitrification was long thought to be the only loss term, it has recently been argued that anammox alone is responsible for fixed nitrogen loss in the OMZs. Here we measure denitrification and anammox rates and quantify the abundance of denitrifying and anammox bacteria in the OMZ regions of the Eastern Tropical South Pacific and the Arabian Sea. We find that denitrification rather than anammox dominates the N2 loss term in the Arabian Sea, the largest and most intense OMZ in the world ocean. In seven of eight experiments in the Arabian Sea denitrification is responsible for 87–99% of the total N2 production. The dominance of denitrification is reproducible using two independent isotope incubation methods. In contrast, anammox is dominant in the Eastern Tropical South Pacific OMZ, as detected using one of the isotope incubation methods, as previously reported. The abundance of denitrifying bacteria always exceeded that of anammox bacteria by up to 7- and 19-fold in the Eastern Tropical South Pacific and Arabian Sea, respectively. Geographic and temporal variability in carbon supply may be responsible for the different contributions of denitrification and anammox in these two OMZs. The isotope 146Sm undergoes α-decay to 142Nd, with a half-life of 103 million years. Measurable variations in the 142Nd/144Nd values of rocks resulting from Sm–Nd fractionation could therefore only have been produced within about 400 million years of the Solar System’s formation (that is, when 146Sm was extant). The 142Nd/144Nd compositions of terrestrial rocks are accordingly a sensitive monitor of the main silicate differentiation events that took place in the early Earth. High 142Nd/144Nd values measured in some Archaean rocks from Greenland hint at the existence of an early incompatible-element-depleted mantle. Here we present measurements of low 142Nd/144Nd values in 1.48-gigayear-(Gyr)-old lithospheric mantle-derived alkaline rocks from the Khariar nepheline syenite complex in southeastern India. These data suggest that a reservoir that was relatively enriched in incompatible elements formed at least 4.2 Gyr ago and traces of its isotopic signature persisted within the lithospheric root of the Bastar craton until at least 1.48 Gyr ago. These low 142Nd/144Nd compositions may represent a diluted signature of a Hadean (4 to 4.57 Gyr ago) enriched reservoir that is characterized by even lower values. That no evidence of the early depleted mantle has been observed in rocks younger than 3.6 Gyr (refs 3, 4, 7) implies that such domains had effectively mixed back into the convecting mantle by then. In contrast, some early enriched components apparently escaped this fate. Thus, the mantle sampled by magmatism since 3.6 Gyr ago may be biased towards a depleted composition that would be balanced by relatively more enriched reservoirs that are ‘hidden’ in Hadean crust, the D′′ layer of the lowermost mantle or, as we propose here, also within the roots of old cratons. Mass spectrometry is a powerful technology for the analysis of large numbers of endogenous proteins. However, the analytical challenges associated with comprehensive identification and relative quantification of cellular proteomes have so far appeared to be insurmountable. Here, using advances in computational proteomics, instrument performance and sample preparation strategies, we compare protein levels of essentially all endogenous proteins in haploid yeast cells to their diploid counterparts. Our analysis spans more than four orders of magnitude in protein abundance with no discrimination against membrane or low level regulatory proteins. Stable-isotope labelling by amino acids in cell culture (SILAC) quantification was very accurate across the proteome, as demonstrated by one-to-one ratios of most yeast proteins. Key members of the pheromone pathway were specific to haploid yeast but others were unaltered, suggesting an efficient control mechanism of the mating response. Several retrotransposon-associated proteins were specific to haploid yeast. Gene ontology analysis pinpointed a significant change for cell wall components in agreement with geometrical considerations: diploid cells have twice the volume but not twice the surface area of haploid cells. Transcriptome levels agreed poorly with proteome changes overall. However, after filtering out low confidence microarray measurements, messenger RNA changes and SILAC ratios correlated very well for pheromone pathway components. The oxygen isotope ratio (δ18O) of cellulose is thought to provide a record of ambient temperature and relative humidity during periods of carbon assimilation. Here we introduce a method to resolve tree-canopy leaf temperature with the use of δ18O of cellulose in 39 tree species. We show a remarkably constant leaf temperature of 21.4 ± 2.2 °C across 50° of latitude, from subtropical to boreal biomes. This means that when carbon assimilation is maximal, the physiological and morphological properties of tree branches serve to raise leaf temperature above air temperature to a much greater extent in more northern latitudes. A main assumption underlying the use of δ18O to reconstruct climate history is that the temperature and relative humidity of an actively photosynthesizing leaf are the same as those of the surrounding air. Our data are contrary to that assumption and show that plant physiological ecology must be considered when reconstructing climate through isotope analysis. The recent discovery of diamond–graphite inclusions in the Earth’s oldest zircon grains (formed up to 4,252 Myr ago) from the Jack Hills metasediments in Western Australia provides a unique opportunity to investigate Earth’s earliest known carbon reservoir. Here we report ion microprobe analyses of the carbon isotope composition of these diamond–graphite inclusions. The observed δ13CPDB values (expressed using the PeeDee Belemnite standard) range between -5 per mil and -58 per mil with a median of -31 per mil. This extends beyond typical mantle values of around -6 per mil to values observed in metamorphic and some eclogitic diamonds that are interpreted to reflect deep subduction of low-δ13CPDB biogenic surface carbon. Low δ13CPDB values may also be produced by inorganic chemical reactions, and therefore are not unambiguous evidence for life on Earth as early as 4,250 Myr ago. The emplacement of the 2.05-billion-year-old Bushveld complex, the world’s largest layered intrusion and platinum-group element (PGE) repository, is a singular event in the history of the Kaapvaal craton of southern Africa, one of Earth’s earliest surviving continental nuclei. In the prevailing model for the complex’s mineralization, the radiogenic strontium and osmium isotope signatures of Bushveld PGE ores are attributed to continental crustal contamination of the host magmas. The scale of the intrusion and lateral homogeneity of the PGE-enriched layers, however, have long been problematical for the crustal contamination model, given the typically heterogeneous nature of continental crust. Furthermore, the distribution of Bushveld magmatism matches that of seismically anomalous underlying mantle, implying significant interaction before emplacement in the crust. Mineral samples of the ancient 200-km-deep craton keel, encapsulated in macrodiamonds and entrained by proximal kimberlites, reveal the nature of continental mantle potentially incorporated by Bushveld magmas. Here we show that sulphide inclusions in ∼2-billion-year-old diamonds from the 0.5-billion-year-old Venetia and 1.2-billion-year-old Premier kimberlites (on opposite sides of the complex) have initial osmium isotope ratios even more radiogenic than those of Bushveld sulphide ore minerals. Sulphide Re–Os and silicate Sm–Nd and Rb–Sr isotope compositions indicate that continental mantle harzburgite and eclogite components, in addition to the original convecting mantle magma, most probably contributed to the genesis of both the diamonds and the Bushveld complex. Small isotopic differences in the atomic abundance of neodymium-142 (142Nd) in silicate rocks represent the time-averaged effect of decay of formerly live samarium-146 (146Sm) and provide constraints on the timescales and mechanisms by which planetary mantles first differentiated. This chronology, however, assumes that the composition of the total planet is identical to that of primitive undifferentiated meteorites called chondrites. The difference in the 142Nd/144Nd ratio between chondrites and terrestrial samples may therefore indicate very early isolation (<30 Myr from the formation of the Solar System) of the upper mantle or a slightly non-chondritic bulk Earth composition. Here we present high-precision 142Nd data for 16 martian meteorites and show that Mars also has a non-chondritic composition. Meteorites belonging to the shergottite subgroup define a planetary isochron yielding an age of differentiation of 40 ± 18 Myr for the martian mantle. This isochron does not pass through the chondritic reference value (100 × ε142Nd = -21 ± 3; 147Sm/144Nd = 0.1966). The Earth, Moon and Mars all seem to have accreted in a portion of the inner Solar System with ∼5 per cent higher Sm/Nd ratios than material accreted in the asteroid belt. Such chemical heterogeneities may have arisen from sorting of nebular solids or from impact erosion of crustal reservoirs in planetary precursors. Biosignatures and structures in the geological record indicate that microbial life has inhabited Earth for the past 3.5 billion years or so. Research in the physical sciences has been able to generate statements about the ancient environment that hosted this life. These include the chemical compositions and temperatures of the early ocean and atmosphere. Only recently have the natural sciences been able to provide experimental results describing the environments of ancient life. Our previous work with resurrected proteins indicated that ancient life lived in a hot environment. Here we expand the timescale of resurrected proteins to provide a palaeotemperature trend of the environments that hosted life from 3.5 to 0.5 billion years ago. The thermostability of more than 25 phylogenetically dispersed ancestral elongation factors suggest that the environment supporting ancient life cooled progressively by 30 °C during that period. Here we show that our results are robust to potential statistical bias associated with the posterior distribution of inferred character states, phylogenetic ambiguity, and uncertainties in the amino-acid equilibrium frequencies used by evolutionary models. Our results are further supported by a nearly identical cooling trend for the ancient ocean as inferred from the deposition of oxygen isotopes. The convergence of results from natural and physical sciences suggest that ancient life has continually adapted to changes in environmental temperatures throughout its evolutionary history. The evolution of the Earth’s atmosphere is marked by a transition from an early atmosphere with very low oxygen content to one with an oxygen content within a few per cent of the present atmospheric level. Placing time constraints on this transition is of interest because it identifies the time when oxidative weathering became efficient, when ocean chemistry was transformed by delivery of oxygen and sulphate, and when a large part of Earth’s ecology changed from anaerobic to aerobic. The observation of non-mass-dependent sulphur isotope ratios in sedimentary rocks more than ∼2.45 billion years (2.45 Gyr) old and the disappearance of this signal in younger sediments is taken as one of the strongest lines of evidence for the transition from an anoxic to an oxic atmosphere around 2.45 Gyr ago. Detailed examination of the sulphur isotope record before 2.45 Gyr ago also reveals early and late periods of large amplitude non-mass-dependent signals bracketing an intervening period when the signal was attenuated. Until recently, this record has been too sparse to allow interpretation, but collection of new data has prompted some workers to argue that the Mesoarchaean interval (3.2–2.8 Gyr ago) lacks a non-mass-dependent signal, and records the effects of earlier and possibly permanent oxygenation of the Earth’s atmosphere. Here we focus on the Mesoarchaean interval, and demonstrate preservation of a non-mass-dependent signal that differs from that of preceding and following periods in the Archaean. Our findings point to the persistence of an anoxic early atmosphere, and identify variability within the isotope record that suggests changes in pre-2.45-Gyr-ago atmospheric pathways for non-mass-dependent chemistry and in the ultraviolet transparency of an evolving early atmosphere. Chlorine in the Earth is highly depleted relative to carbonaceous chondrites and solar abundances. Knowledge of the Cl concentrations and distribution on Earth is essential for understanding the origin of these depletions. Large differences in the stable chlorine isotope ratios of meteoritic, mantle and crustal materials have been used as evidence for distinct reservoirs in the solar nebula and to calculate the relative proportions of Cl in the mantle and crust. Here we report that large isotopic differences do not exist, and that carbonaceous chondrites, mantle and crust all have the same 37Cl/35Cl ratios. We have further analysed crustal sediments from the early Archaean era to the Recent epoch and find no systematic isotopic variations with age, demonstrating that the mantle and crust have always had the same δ37Cl value. The similarity of mantle, crust and carbonaceous chondrites establishes that there were no nebular reservoirs with distinct isotopic compositions, no isotopic fractionation during differentiation of the Earth and no late (post-core formation) Cl-bearing volatile additions to the crustal veneer with a unique isotopic composition. Atmospheric moisture cycling is an important aspect of the Earth’s climate system, yet the processes determining atmospheric humidity are poorly understood. For example, direct evaporation of rain contributes significantly to the heat and moisture budgets of clouds, but few observations of these processes are available. Similarly, the relative contributions to atmospheric moisture over land from local evaporation and humidity from oceanic sources are uncertain. Lighter isotopes of water vapour preferentially evaporate whereas heavier isotopes preferentially condense and the isotopic composition of ocean water is known. Here we use this information combined with global measurements of the isotopic composition of tropospheric water vapour from the Tropospheric Emission Spectrometer (TES) aboard the Aura spacecraft, to investigate aspects of the atmospheric hydrological cycle that are not well constrained by observations of precipitation or atmospheric vapour content. Our measurements of the isotopic composition of water vapour near tropical clouds suggest that rainfall evaporation contributes significantly to lower troposphere humidity, with typically 20% and up to 50% of rainfall evaporating near convective clouds. Over the tropical continents the isotopic signature of tropospheric water vapour differs significantly from that of precipitation, suggesting that convection of vapour from both oceanic sources and evapotranspiration are the dominant moisture sources. The Palaeocene/Eocene thermal maximum represents a period of rapid, extreme global warming ∼55 million years ago, superimposed on an already warm world. This warming is associated with a severe shoaling of the ocean calcite compensation depth and a >2.5 per mil negative carbon isotope excursion in marine and soil carbonates. Together these observations indicate a massive release of 13C-depleted carbon and greenhouse-gas-induced warming. Recently, sediments were recovered from the central Arctic Ocean, providing the first opportunity to evaluate the environmental response at the North Pole at this time. Here we present stable hydrogen and carbon isotope measurements of terrestrial-plant- and aquatic-derived n-alkanes that record changes in hydrology, including surface water salinity and precipitation, and the global carbon cycle. Hydrogen isotope records are interpreted as documenting decreased rainout during moisture transport from lower latitudes and increased moisture delivery to the Arctic at the onset of the Palaeocene/Eocene thermal maximum, consistent with predictions of poleward storm track migrations during global warming. The terrestrial-plant carbon isotope excursion (about -4.5 to -6 per mil) is substantially larger than those of marine carbonates. Previously, this offset was explained by the physiological response of plants to increases in surface humidity. But this mechanism is not an effective explanation in this wet Arctic setting, leading us to hypothesize that the true magnitude of the excursion—and associated carbon input—was greater than originally surmised. Nuclear-magnetic-resonance spectroscopy can determine the three-dimensional structure of proteins in solution. However, its potential has been limited by the difficulty of interpreting NMR spectra in the presence of broadened and overlapping resonance lines and low signal-to-noise ratios. Here we present stereo-array isotope labelling (SAIL), a technique that can overcome many of these problems by applying a complete stereospecific and regiospecific pattern of stable isotopes that is optimal with regard to the quality and information content of the resulting NMR spectra. SAIL uses exclusively chemically and enzymatically synthesized amino acids for cell-free protein expression. We demonstrate for the 17-kDa protein calmodulin and the 41-kDa maltodextrin-binding protein that SAIL offers sharpened lines, spectral simplification without loss of information, and the ability to rapidly collect the structural restraints required to solve a high-quality solution structure for proteins twice as large as commonly solved by NMR. The origins of the building blocks of the Solar System can be studied using the isotopic composition of early planetary and meteoritic material. Oxygen isotopes in planetary materials show variations at the per cent level that are not related to the mass of the isotopes; rather, they result from the mixture of components having different nucleosynthetic or chemical origins. Isotopic variations reaching orders of magnitude in minute meteoritic grains are usually attributed to stellar nucleosynthesis before the birth of the Solar System, whereby different grains were contributed by different stars. Here we report the discovery of abundant silica-rich grains embedded in meteoritic organic matter, having the most extreme 18O/16O and 17O/16O ratios observed (both ∼10-1) together with a solar silicon isotopic composition. Both O and Si isotopes indicate a single nucleosynthetic process. These compositions can be accounted for by one of two processes: a single exotic evolved star seeding the young Solar System, or irradiation of the circumsolar gas by high energy particles accelerated during an active phase of the young Sun. We favour the latter interpretation, because the observed compositions are usually not expected from nucleosynthetic processes in evolved stars, whereas they are predicted by the selective trapping of irradiation products. Marine records of sediment oxygen isotope compositions show that the Earth's climate has gone through a succession of glacial and interglacial periods during the past million years. But the interpretation of the oxygen isotope records is complicated because both isotope storage in ice sheets and deep-water temperature affect the recorded isotopic composition. Separating these two effects would require long records of either sea level or deep-ocean temperature, which are currently not available. Here we use a coupled model of the Northern Hemisphere ice sheets and ocean temperatures, forced to match an oxygen isotope record for the past million years compiled from 57 globally distributed sediment cores, to quantify both contributions simultaneously. We find that the ice-sheet contribution to the variability in oxygen isotope composition varied from ten per cent in the beginning of glacial periods to sixty per cent at glacial maxima, suggesting that strong ocean cooling preceded slow ice-sheet build-up. The model yields mutually consistent time series of continental mean surface temperatures between 40 and 80° N, ice volume and global sea level. We find that during extreme glacial stages, air temperatures were 17 ± 1.8 °C lower than present, with a 120 ± 10 m sea level equivalent of continental ice present. [ABSTRACT FROM AUTHOR]

The marine sedimentary record exhibits evidence for episodes of enhanced organic carbon burial known as‘oceanic anoxic events’(OAEs). They are characterized by carbon-isotope excursions in marine and terrestrial reservoirs and mass extinction of marine faunas. Causal mechanisms for the enhancement of organic carbon burial during OAEs are still debated, but it is thought that such events should draw down significant quantities of atmospheric carbon dioxide. In the case of the Toarcian OAE (~183?million years ago), a short-lived negative carbon-isotope excursion in oceanic and terrestrial reservoirs has been interpreted to indicate raised atmospheric carbon dioxide caused by oxidation of methane catastrophically released from either marine gas hydrates or magma-intruded organic-rich rocks. Here we test these two leading hypotheses for a negative carbon isotopic excursion marking the initiation of the Toarcian OAE using a high-resolution atmospheric carbon dioxide record obtained from fossil leaf stomatal frequency. We find that coincident with the negative carbon-isotope excursion carbon dioxide is first drawn down by 350±100?p.p.m.v. and then abruptly elevated by 1,200±400?p.p.m.v, and infer a global cooling and greenhouse warming of 2.5±0.1?°C and 6.5±1?°C, respectively. The pattern and magnitude of carbon dioxide change are difficult to reconcile with catastrophic input of isotopically light methane from hydrates as the cause of the negative isotopic signal. Our carbon dioxide record better supports a magma-intrusion hypothesis, and suggests that injection of isotopically light carbon from the release of thermogenic methane occurred owing to the intrusion of Gondwana coals by Toarcian-aged Karoo-Ferrar dolerites. The discovery in primitive components of meteorites of large oxygen isotopic variations that could not be attributed to mass-dependent fractionation effects has raised a fundamental question: what is the composition of the protosolar gas from which the host grains formed? This composition is probably preserved in the outer layers of the Sun, but the resolution of astronomical spectroscopic measurements is still too poor to be useful for comparison with planetary material. Here we report a precise determination of the oxygen isotopic composition of the solar wind from particles implanted in the outer hundreds of nanometres of metallic grains in the lunar regolith. These layers of the grains are enriched in16O by>20±4‰relative to the Earth, Mars and bulk meteorites, which implies the existence in the solar accretion disk of reactions-as yet unknown-that were able to change the17O/16O and18O/16O ratios in a way that was not dependent strictly on the mass of the isotope. Photochemical self-shielding of the CO gas irradiated by ultraviolet light may be one of these key processes, because it depends on the abundance of the isotopes, rather than their masses. Identifying the origin of primordial volatiles in the Earth's mantle provides a critical test between models that advocate magma-ocean equilibration with an early massive solar-nebula atmosphere and those that require subduction of volatiles implanted in late accreting material. Here we show that neon isotopes in the convecting mantle, resolved in magmatic CO2 well gases, are consistent with a volatile source related to solar corpuscular irradiation of accreting material. This contrasts with recent results that indicated a solar-nebula origin for neon in mantle plume material, which is thought to be sampling the deep mantle. Neon isotope heterogeneity in different mantle sources suggests that models in which the plume source supplies the convecting mantle with its volatile inventory require revision. Although higher than accepted noble gas concentrations in the convecting mantle may reduce the need for a deep mantle volatile flux, any such flux must be dominated by the neon (and helium) isotopic signature of late accreting material. The origin of the isotopic signature of Indian mid-ocean ridge basalts has remained enigmatic, because the geochemical composition of these basalts is consistent either with pollution from recycled, ancient altered oceanic crust and sediments, or with ancient continental crust or lithosphere. The radiogenic isotopic signature may therefore be the result of contamination of the upper mantle by plumes containing recycled altered ancient oceanic crust and sediments, detachment and dispersal of continental material into the shallow mantle during rifting and breakup of Gondwana, or contamination of the upper mantle by ancient subduction processes. The identification of a process operating on a scale large enough to affect major portions of the Indian mid-ocean ridge basalt source region has been a long-standing problem. Here we present hafnium and lead isotope data from across the Indian-Pacific mantle boundary at the Australian-Antarctic discordance region of the Southeast Indian Ridge, which demonstrate that the Pacific and Indian upper mantle basalt source domains were each affected by different mechanisms. We infer that the Indian upper-mantle isotope signature in this region is affected mainly by lower continental crust entrained during Gondwana rifting, whereas the isotope signature of the Pacific upper mantle is influenced predominantly by ocean floor subduction-related processes. It is generally assumed that solid hydrogen will transform into a metallic alkali-like crystal at sufficiently high pressure. However, some theoretical models have also suggested that compressed hydrogen may form an unusual two-component (protons and electrons) metallic fluid at low temperature, or possibly even a zero-temperature liquid ground state. The existence of these new states of matter is conditional on the presence of a maximum in the melting temperature versus pressure curve (the‘melt line’). Previous measurements of the hydrogen melt line up to pressures of 44?GPa have led to controversial conclusions regarding the existence of this maximum. Here we report ab initio calculations that establish the melt line up to 200?GPa. We predict that subtle changes in the intermolecular interactions lead to a decline of the melt line above 90?GPa. The implication is that as solid molecular hydrogen is compressed, it transforms into a low-temperature quantum fluid before becoming a monatomic crystal. The emerging low-temperature phase diagram of hydrogen and its isotopes bears analogies with the familiar phases of3He and4He (the only known zero-temperature liquids), but the long-range Coulomb interactions and the large component mass ratio present in hydrogen would result in dramatically different properties. Two deep ice cores from central Greenland, drilled in the 1990s, have played a key role in climate reconstructions of the Northern Hemisphere, but the oldest sections of the cores were disturbed in chronology owing to ice folding near the bedrock. Here we present an undisturbed climate record from a North Greenland ice core, which extends back to 123,000 years before the present, within the last interglacial period. The oxygen isotopes in the ice imply that climate was stable during the last interglacial period, with temperatures 5°C warmer than today. We find unexpectedly large temperature differences between our new record from northern Greenland and the undisturbed sections of the cores from central Greenland, suggesting that the extent of ice in the Northern Hemisphere modulated the latitudinal temperature gradients in Greenland. This record shows a slow decline in temperatures that marked the initiation of the last glacial period. Our record reveals a hitherto unrecognized warm period initiated by an abrupt climate warming about 115,000 years ago, before glacial conditions were fully developed. This event does not appear to have an immediate Antarctic counterpart, suggesting that the climate see-saw between the hemispheres (which dominated the last glacial period) was not operating at this time. Not all noise in experimental measurements is unwelcome. Certain fundamental noise sources contain valuable information about the system itself—a notable example being the inherent voltage fluctuations (Johnson noise) that exist across any resistor, which allow the temperature to be determined1,2. In magnetic systems, fundamental noise can exist in the form of random spin fluctuations3,4. For example, statistical fluctuations of N paramagnetic spins should generate measurable noise of order √N p spins, even in zero magnetic field5,6. Here we exploit this effect to perform perturbation-free magnetic resonance. We use offresonant Faraday rotation to passively7,8 detect the magnetization noise in an equilibrium ensemble of paramagnetic alkali atoms; the random fluctuations generate spontaneous spin coherences that precess and decay with the same characteristic energy and timescales as the macroscopic magnetization of an intentionally polarized or driven ensemble. Correlation spectra of the measured spin noise reveal g-factors, nuclear spin, isotope abundance ratios, hyperfine splittings, nuclear moments and spin coherence lifetimes—without having to excite, optically pump or otherwise drive the system away from thermal equilibrium. The availability of nitrogen is important in regulating biological productivity in marine environments. Deepwater nitrate has long been considered the major source of new nitrogen supporting primary production in oligotrophic regions of the open ocean, but recent studies have showed that biological N2 fixation has a critical role in supporting oceanic new production. Large colonial cyanobacteria in the genus Trichodesmium and the heterocystous endosymbiont Richelia have traditionally been considered the dominant marine N2 fixers, but unicellular diazotrophic cyanobacteria and bacterioplankton have recently been found in the picoplankton and nanoplankton community of the North Pacific central gyre, and a variety of molecular and isotopic evidence suggests that these unicells could make a major contribution to the oceanic N budget. Here we report rates of N2 fixation by these small, previously overlooked diazotrophs that, although spatially variable, can equal or exceed the rate of N2 fixation reported for larger, more obvious organisms. Direct measurements of 15N2 fixation by small diazotrophs in various parts of the Pacific Ocean, including the waters off Hawaii where the unicellular diazotrophs were first characterized, show that N2 fixation by unicellular diazotrophs can support a significant fraction of total new production in oligotrophic waters. In conventional superconductors, the electron pairing that allows superconductivity is caused by exchange of virtual phonons, which are quanta of lattice vibration. For high-transition-temperature (high-Tc) superconductors, it is far from clear that phonons are involved in the pairing at all. For example, the negligible change in Tc of optimally doped Bi2Sr2CaCu2O8+d (Bi2212; ref. 1) upon oxygen isotope substitution (16O ? 18O leads to Tc decreasing from 92 to 91?K) has often been taken to mean that phonons play an insignificant role in this material. Here we provide a detailed comparison of the electron dynamics of Bi2212 samples containing different oxygen isotopes, using angle-resolved photoemission spectroscopy. Our data show definite and strong isotope effects. Surprisingly, the effects mainly appear in broad high-energy humps, commonly referred to as ‘incoherent peaks’. As a function of temperature and electron momentum, the magnitude of the isotope effect closely correlates with the superconducting gap-that is, the pair binding energy. We suggest that these results can be explained in a dynamic spin-Peierls picture, where the singlet pairing of electrons and the electron-lattice coupling mutually enhance each other. The deep-ocean circulation is responsible for a significant component of global heat transport. In the present mode of circulation, deep waters form in the North Atlantic and Southern oceans where surface water becomes sufficiently cold and dense to sink. Polar temperatures during the warmest climatic interval of the Cenozoic era (~ 65 to 40 million years (Myr) ago) were significantly warmer than today, and this may have been a consequence of enhanced oceanic heat transport. However, understanding the relationship between deep-ocean circulation and ancient climate is complicated by differences in oceanic gateways, which affect where deep waters form and how they circulate. Here I report records of neodymium isotopes from two cores in the Pacific Ocean that indicate a shift in deep-water production from the Southern Ocean to the North Pacific ~65 Myr ago. The source of deep waters reverted back to the Southern Ocean 40 Myr ago. The relative timing of changes in the neodymium and oxygen isotope records indicates that changes in Cenozoic deep-water circulation patterns were the consequence, not the cause, of extreme Cenozoic warmth. Solar luminosity on the early Earth was significantly lower than today. Therefore, solar luminosity models suggest that, in the atmosphere of the early Earth, the concentration of greenhouse gases such as carbon dioxide and methane must have been much higher. However, empirical estimates of Proterozoic levels of atmospheric carbon dioxide concentrations have not hitherto been available. Here we present ion microprobe analyses of the carbon isotopes in individual organic-walled microfossils extracted from a Proterozoic (~ 1.4-gigayear-old) shale in North China. Calculated magnitudes of the carbon isotope fractionation in these large, morphologically complex microfossils suggest elevated levels of carbon dioxide in the ancient atmosphere-between 10 and 200 times the present atmospheric level. Our results indicate that carbon dioxide was an important greenhouse gas during periods of lower solar luminosity, probably dominating over methane after the atmosphere and hydrosphere became pervasively oxygenated between 2 and 2.2 gigayears ago. The abundances of elements and their isotopes in our Galaxy show wide variations, reflecting different nucleosynthetic processes in stars and the effects of Galactic evolution. These variations contrast with the uniformity of stable isotope abundances for many elements in the Solar System, which implies that processes efficiently homogenized dust and gas from different stellar sources within the young solar nebula. However, isotopic heterogeneity has been recognized on the subcentimetre scale in primitive meteorites, indicating that these preserve a compositional memory of their stellar sources. Small differences in the abundance of stable molybdenum isotopes in bulk rocks of some primitive and differentiated meteorites, relative to terrestrial Mo, suggest large-scale Mo isotopic heterogeneity between some inner Solar System bodies, which implies physical conditions that did not permit efficient mixing of gas and dust. Here we report Mo isotopic data for bulk samples of primitive and differentiated meteorites that show no resolvable deviations from terrestrial Mo. This suggests efficient mixing of gas and dust in the solar nebula at least to 3?au from the Sun, possibly induced by magnetohydrodynamic instabilities.
