Abstract

The total synthesis of kinamycins C, F and J is described by a convergent strategy that is both enantioseletive and expedient. This chemistry has provided an expedient and flexible entry into the kinamycin family of antitumor antibiotics and is found to be useful in the total synthesis of the lomaiviticins.

Abstract

The first highly enantioselective Friedel-Crafts reaction of indoles with imines with a chiral organic catalyst is reported. This reaction provides a direct and broadly useful catalytic enantioselective approach toward 3-indolyl methanamines, which should facilitate the asymmetric synthesis of biologically interesting indole compounds.

The first highly enantioselective catalytic conjugate addition to vinyl sulfones are developed. The synthetic utility of the optically active chiral sulfones is demonstrated in the development of a versatile catalytic enantioselective approach toward alpha, alpha-disubstituted amino acids.

Abstract

A novel approach to hemiaminal synthesis via palladium-catalyzed C–N coupling with chiral bisphosphine mono-oxides is described. This efficient new method exhibits a broad scope, provides a highly efficient synthesis of HCV drug candidate elbasvir, and has been applied to the synthesis of chiral N,N-acetals.

The development of an asymmetric Diels-Alder reaction of 2-pyrones by using cinchona alkaloid-derived bifunctional organic crystals is described. The possibility of using these catalysts to control the endo/exo selectivity in a Diels-Alder reaction is demonstrated.

Abstract

The reactions of 2-pyrones with electron-deficient dienophiles constitute a synthetically useful class of Diels−Alder reaction. By exploring cinchona alkaloid-derived organic molecules as acid−base bifunctional catalysts, we successfully developed the first highly enantioselective and diastereoselective catalytic Diels−Alder reaction with 2-pyrones. Furthermore, we demonstrated the possibility of using such catalysts to control the endo/exo selectivity in a Diels−Alder reaction.

Abstract

The development of readily accessible bifunctional chiral catalysts is a desirable yet challenging goal in catalytic asymmetric synthesis. In this communication, we describe the development of a new class of readily accessible chiral bifunctional organic catalysts that could be derived in one or two steps in high yield from either quinidine or quinine. These catalysts have been shown to catalyze a highly enantioselective conjugate addition of methyl and ethyl malonates and beta-ketoesters to a broad range of beta-substituted nitroalkenes, an synthetically important C-C bond-forming reaction utilizing readily available starting materials. This new catalytic asymmetric reaction proceeds in 91-98% ee and 71-99% yield.

Abstract

The kinamycins are a series of naturally occurring compounds endowed with intriguing molecular architectures and potent biological properties, including antibiotic and antitumor activities. These novel diazofluorene-containing compounds defied chemical synthesis since their initial disclosure by Omura et al. in 1970 until the first total synthesis of kinamycin C by Porco et al. in late 2006, and of methyl-kinamycin C by the Kumamoto−Ishihawa group in 2007. We now report our enantioselective total synthesis of kinamycins C, F, and J through a convergent strategy that is both enantioselective and expedient.

Abstract

The article focuses on the enantioselective nitroaldol reaction of cinchona alkaloid-catalyzed alpha-ketotesters. Significantly, the nitroaldol reaction is an important class of C-C bond forming reactions that provide access to important synthetic intermediates from readily accessible nitroalkanes and carbonyl compounds. There is no broadly effective chiral organic catalyst has been developed for the direct asymmetric nitroaldol reaction eventhough effective chiral organic catalysts have been reported for enantioselective reactions. Finally, the researchers have developed the first efficient organocatalytic enantioselective nitroaldol reaction.
