A routine multiresidue method developed for the detection and quantification of veterinary drug residues in animal-based food was used to analyze sheep (ovine) liver. Unlike when working with previously validated matrices (e.g., bovine liver), some of the analytes of interest chromatographed in the form of split- or even fully baseline separated peaks. In other cases a significantly longer retention times (tR) was observed. A detailed investigation led to the elucidation of taurocholic acid as the causative agent. This compound is present in sheep liver at significantly higher concentrations than in most other animal tissues. Taurocholic acid is a zwitterionic compound and likely acts as an ion pairing agent, which modifies the selectivity of the stationary phase in a highly spatial and dynamic way. Injecting smaller volumes of matrix extract or the use of a significantly higher formic acid concentration in the mobile phase reduced or even completely eliminated the peak splitting. A more detailed examination led to the observation that the problem is not restricted to this particular matrix and extraction procedure or the used stationary phase. In fact, a higher formic acid concentration (e.g., 1.0 % versus 0.1 %) significantly improves the peak shape of many analytes present in fortified matrix samples as well as in pure standard solutions. In addition, analytical column aging was observed as being slower with a higher formic acid concentration. Finally the peak shape of analytes interacting with the metallic parts along the flow path of the liquid chromatograph was also significantly improved. Use of 0.1 % acid in mobile phases is often taken for granted in LC-MS. Regardless of the stationary phase, a higher ionic strength better stabilizes the pH and reduces unwanted interactions, which ultimately improves the method robustness. Flow injection experiments often show that 0.1 % acid concentrations produce the highest analyte signals. Yet, the use of 1 % acid in the mobile phase often leads to narrower and therefore taller chromatographic peaks, which may lead to lower detection limits for many analytes and to an improved separation efficiency. In this contribution, a multiplicative effects model for generalized multiple-internal-standard method (MEM GMIS ) was proposed to solve the signal instability problem of LC–MS over time. MEM GMIS model seamlessly integrates the multiple-internal-standard strategy with multivariate calibration method, and takes full use of all the information carried by multiple internal standards during the quantification of target analytes. Unlike the existing methods based on multiple internal standards, MEM GMIS does not require selecting an optimal internal standard for the quantification of a specific analyte from multiple internal standards used. MEM GMIS was applied to a proof-of-concept model system: the simultaneous quantitative analysis of five edible artificial colorants in two kinds of cocktail drinks. Experimental results demonstrated that MEM GMIS models established on LC–MS data of calibration samples prepared with ultrapure water could provide quite satisfactory concentration predictions for colorants in cocktail samples from their LC–MS data measured 10 days after the LC–MS analysis of the calibration samples. The average relative prediction errors of MEM GMIS models did not exceed 6.0%, considerably better than the corresponding values of commonly used univariate calibration models combined with multiple internal standards. Liquid chromatography–mass spectrometry (LC–MS) is now a main stream technique for large-scale metabolic phenotyping to obtain a better understanding of genomic functions. However, repeatability is still an essential issue for the LC–MS based methods, and convincing strategies for long time analysis are urgently required. Our former reported pseudotargeted method which combines nontargeted and targeted analyses, is proved to be a practical approach with high-quality and information-rich data. In this study, we developed a comprehensive strategy based on the pseudotargeted analysis by integrating blank-wash, pooled quality control (QC) sample, and post-calibration for the large-scale metabolomics study. The performance of strategy was optimized from both pre- and post-acquisition sections including the selection of QC samples, insertion frequency of QC samples, and post-calibration methods. These results imply that the pseudotargeted method is rather stable and suitable for large-scale study of metabolic profiling. As a proof of concept, the proposed strategy was applied to the combination of 3 independent batches within a time span of 5 weeks, and generated about 54% of the features with coefficient of variations (CV) below 15%. Moreover, the stability and maximal capability of a single analytical batch could be extended to at least 282 injections (about 110 h) while still providing excellent stability, the CV of 63% metabolic features was less than 15%. Abstract: We performed a comprehensive study to assess the fit for purpose of four chromatographic conditions for the determination of six groups of marine lipophilic toxins (okadaic acid and dinophysistoxins, pectenotoxins, azaspiracids, yessotoxins, gymnodimine and spirolides) by LC–MS/MS to select the most suitable conditions as stated by the European Union Reference Laboratory for Marine Biotoxins (EURLMB). For every case, the elution gradient has been optimized to achieve a total run-time cycle of 12min. We performed a single-laboratory validation for the analysis of three relevant matrices for the seafood aquaculture industry (mussels, pacific oysters and clams), and for sea urchins for which no data about lipophilic toxins have been reported before. Moreover, we have compared the method performance under alkaline conditions using two quantification strategies: the external standard calibration (EXS) and the matrix-matched standard calibration (MMS). Alkaline conditions were the only scenario that allowed detection windows with polarity switching in a 3200 QTrap mass spectrometer, thus the analysis of all toxins can be accomplished in a single run, increasing sample throughput. The limits of quantification under alkaline conditions met the validation requirements established by the EURLMB for all toxins and matrices, while the remaining conditions failed in some cases. The accuracy of the method and the matrix effects where generally dependent on the mobile phases and the seafood species. The MMS had a moderate positive impact on method accuracy for crude extracts, but it showed poor trueness for seafood species other than mussels when analyzing hydrolyzed extracts. Alkaline conditions with EXS and recovery correction for OA were selected as the most proper conditions in the context of our laboratory. Abstract: A protocol for the analysis of multiple prostaglandins and leukotrienes in cell culture media by using multiple internal standards was validated. A two-factor Doehlert design was used to determine the behaviour of the relationship analyte/internal standard (namely: PGE2/PGE2-d 4, PGE3/PGE2-d 4, LTB4/LTB4-d 4 and LTB5/LTB4-d 4) and to select the optimal amounts of deuterated internal standards for quantifying simultaneously the prostaglandins and leukotrienes in cell culture media by LC–MS/MS. The selection of optimal amounts of internal standards was based on mathematical models that allow visualizing concentration regions where the response factors remain constant over a wide range of analytical concentrations. The linearity of the calibration curves for each analyte at the optimal levels suggested by the mathematical models was statistically confirmed by means of the ratio lack-of-fit to pure error. The validated protocol was successfully applied in the simultaneous quantification of pro- and anti-inflammatory eicosanoids in stimulated cod head kidney cell culture media. The two-factor Doehlert design has permitted to estimate the experimental response as a function of six variables (PGE2, PGE3, LTB4, LTB5, PGE2-d 4 and LTB4-d 4) which represents a substantial reduction of resources, time and experiments of approximately 84% (7×3 experiments) when compared with the full six-factor Doehlert design (43×3 experiments). Abstract: The aim of this research was to compare the extraction efficiencies of two modes of three-phase hollow fiber microextraction (HF-LLLME) based on aqueous and organic acceptor phases for analysis of tricyclic antidepressant (TCA) drugs. High-performance liquid chromatography with photodiode array detection (HPLC-DAD) was applied for determination of the drugs. In order to examine the ability of the new concept of HF-LLLME based on organic acceptor solvent in comparison with aqueous acceptor phase to extract the analytes, four TCAs were selected. The effect of different extraction conditions (i.e., type of acceptor phase, hollow fiber length, ionic strength, stirring rate, and extraction time) on the extraction efficiency of the TCAs was investigated and optimized using central composite design (CCD) as a powerful tool. Both methods were characterized by good linearity and high repeatability, but HF-LLLME with organic acceptor provided higher extraction efficiency and thus lower limits of detection (LODs). Calibration curves were linear (r 2 >0.996) in the range of 0.2–200μgL−1. LODs for all the TCAs ranged from 0.08 to 0.2μgL−1 using HPLC-DAD. Also an improvement in sensitivity of several orders of magnitude was achieved using single-ion monitoring GC–MS analyses (0.04μgL−1) due to compatibility of this technique with GC instrument. The applicability of the proposed HF-LLLME/GC–MS and HPLC-DAD methods was demonstrated by analyzing the drugs in spiked urine and plasma samples. The obtained recoveries of the drugs in the range of 87.9–109.2% indicated the excellent capability of the developed method for extraction of TCAs from complex matrices. [Copyright &y& Elsevier]

Abstract: A simple solvent microextraction method termed vortex-assisted liquid–liquid microextraction (VALLME) coupled with gas chromatography micro electron-capture detector (GC-μECD) has been developed and used for the pesticide residue analysis in water samples. In the VALLME method, aliquots of 30μL toluene used as extraction solvent were directly injected into a 25mL volumetric flask containing the water sample. The extraction solvent was dispersed into the water phase under vigorously shaking with the vortex. The parameters affecting the extraction efficiency of the proposed VALLME such as extraction solvent, vortex time, volumes of extraction solvent and salt addition were investigated. Under the optimum condition, enrichment factors (EFs) in a range of 835–1115 and limits of detection below 0.010μgL−1 were obtained for the determination of target pesticides in water. The calculated calibration curves provide high levels of linearity yielding correlation coefficients (r 2) greater than 0.9958 with the concentration level ranged from 0.05 to 2.5μgL−1. Finally, the proposed method has been successfully applied to the determination of pesticides from real water samples and acceptable recoveries over the range of 72–106.3% were obtained. Abstract: The primary objective of this review is to discuss recent technological developments in the field of solid-phase microextraction that have enhanced the utility of this sample preparation technique in the field of bioanalysis. These developments include introduction of various new biocompatible coating phases suitable for bioanalysis, such as commercial prototype in vivo SPME devices, as well as the development of sampling interfaces that extend the use of this methodology to small animals such as mice. These new devices permit application of in vivo SPME to a variety of analyses, including pharmacokinetics, bioaccumulation and metabolomics studies, with good temporal and spatial resolution. New calibration approaches have also been introduced to facilitate in vivo studies and provide fast and quantitative results without the need to achieve equilibrium. In combination with the drastic improvement in the analytical sensitivity of modern liquid chromatography–tandem mass spectrometry instrumentation, full potential of in vivo SPME as a sample preparation tool in life sciences can finally be explored. From the instrumentation perspective, SPME was successfully automated in 96-well format for the first time. This opens up new opportunities for high-throughput applications (>1000 samples/day) such as for the determination of unbound and total drug concentrations in complex matrices such as whole blood with no need for sample pretreatment, studies of distribution of drugs in various compartments and/or determination of plasma protein binding and other ligand–receptor binding studies, and this review will summarize the progress in this research area to date. Abstract: An analytical method combining both a simple, fast and efficient solvent microextraction and a sensitive and selective monitoring mode, based on ion isolation ion-trap mass spectrometry (MS), was developed for analysis of perfluorinated compounds (PFCs) in biota. The method involved the vortex-shaking of 0.2g of tissue sample and 800μL of tetrahydrofuran (THF):water (75:25, v/v) for 7min, subsequent centrifugation for 13min and direct quantitation of PFCs in the extract against solvent-based calibration curves. Selection of solvent composition was based on Hildebrand solubility parameters and their components (i.e. dispersion, dipole–dipole and hydrogen bonding forces). Recoveries in samples for PFCs with hydrocarbon chain lengths between C4 and C14 ranged from 85 to 111%, with relative standard deviations between 1 and 11%. The ion isolation monitoring mode, proposed for the first time for ion-trap-MS quantitation, proved to be effective in avoiding space-charge effects caused by co-eluting matrix components while keeping the sensitivity of full scan MS operation. Detection limits of the method were in the range 0.8−6ngg−1 for perfluoroalkyl carboxylates (PFACs) and 0.4–0.8ngg−1 for perfluoroalkyl sulfonates (PFASs) in wet weight samples. The method was validated using a reference material made up of flounder muscle and by comparison with triple quadrupole MS measurements and it was applied to the determination of PFCs in liver and muscle samples from sea birds and fishes. Only PFASs were found in samples at quantifiable levels (2.9 and 13.1ngg−1) while PFACs were below the respective quantitation limits. Abstract: A supramolecular solvent made up of reverse micelles of decanoic acid, dispersed in a continuous phase of THF:water, was proposed for the simple, fast and efficient microextraction of OTA in wheat prior to liquid chromatography-fluorescence determination. The method involved the stirring of 300 mg-wheat subsamples (particle size 50μm) and 350μL of supramolecular solvent for 15min, subsequent centrifugation for 15min and the direct quantitation of OTA in the extract, previous 5.7-fold dilution with ethanol/water/acetic acid (49.5/49.5/1), against solvent-based calibration curves. No clean-up of the extracts or solvent evaporation was needed. Interactions between the supramolecular solvent and major matrix components in the wheat (i.e. carbohydrates, lipids and proteins) were investigated. The reverse micelles in the extractant induced gluten flocculation but only in the coacervation region of lower analytical interest (i.e. at percentages of THF above 11%). The quantitation of OTA was interference-free. Representativity of the 300 mg-wheat subsamples was proved by analysing a reference material. OTA recoveries in wheat ranged between 84% and 95% and the precision of the method, expressed as relative standard deviation, was 2%. The quantitation limit of the method was 1.5μgkg−1 and was below the threshold limit established for OTA in raw cereals by EU directives (5.0μgkg−1). The method developed was validated by using a certified reference material and it was successfully applied to the determination of OTA in different wheat varieties from crops harvested in the South of Spain. OTA was not detected in any of the analysed samples. This method allows quick and simple microextraction of OTA with minimal solvent consumption, while delivering accurate and precise data. Abstract: A new RP-HPLC method for the separation and quantification of the most common genetic variants of bovine milk proteins is described. A reversed-phase analytical column C8 (Zorbax 300SB-C8 RP, 3.5μm, 300Å, 150×4.6 I.D.) was used. All the most common casein (CN) and whey protein genetic variants, including β-CNI, were detected and separated simultaneously in less then 40min, with the exception of αS1-CNB and CNC variants. Purified protein genetic variants were employed in calibration and showed different absorbances at 214nm. The procedure was developed using 40 raw individual milk samples of cows belonging to four different breeds and certified skim milk powder BCR-063R. Method validation consisted in testing linearity, repeatability, reproducibility and accuracy. A linear relationship (R 2 >0.99) between the concentrations of proteins and peak areas was observed over the concentration range, with low detection limits. Repeatability and reproducibility were satisfactory for both retention times and peak areas. The RSD of peak areas ranged from 0.92 to 4.32% within analytical day and from 0.85 to 9.52% across analytical days. Abstract: A new calibration method for high-performance liquid chromatography was validated. The method was called single-injection calibration approach (SICA) because it allowed to obtain a complete calibration curve by means of a single injection of a standard solution containing several non-volatile and semi-volatile organic compounds at different concentration levels. The compounds studied included carboxylic acids, polyalcohols, carbohydrates and water-soluble vitamins. This method allowed a 1–7-fold reduction in the analysis time with regard to conventional calibration methods. The method was applied to three different chromatographic detection methods: refractive index (RI) detection, diode array detection (DAD) and inductively coupled plasma atomic emission detection (ICP-AED). Good linearity was achieved (r 2 >0.999) for the three detection methods but signal correction was required for RI detection and DAD. This fact demonstrated that ICP-AES was the most universal because the signal obtained for non-volatile and semi-volatile organic compounds was not a function of the chemical nature of the compound and only depended on the mass content of carbon. The method was validated by analyzing a reference non-fat milk powder sample as well as several real food samples (three fruit juices, four wines, three candies and a multivitamin complex). Abstract: A bi-dimensional (non-orthogonal) chromatographic system made of two columns of different length (longer along the first direction and shorter on the second) but packed with identical (or similar) materials has been employed to perform automated on-line fraction analysis and peak deconvolution of multicomponent overloaded band profiles. Multicomponent overloaded peaks, eluted from the first column, are sampled through a remotely-controlled switching valve and fractions are injected, under analytical conditions, directly into the second direction. This set-up serves a twofold purpose. First of all, it has been possible to use linear calibration curves for detector calibration (DC). Secondly, since single component peaks were resolved on the second direction, individual concentration profiles have been obtained beneath the envelop of overlapping bands. All the information necessary to operate the bi-dimensional system can be achieved by analyzing the chromatographic behavior of the mixture on the shorter column under linear conditions. Therefore, solutions of unknown concentration can be prepared and their composition reconstructed with a simple chromatographic run. Abstract: A screening method for analysis of perfluoroalkylated substances (PFAS) in biota samples has been developed and validated using liver samples from polar cod (Boreogadus saida) and glaucous gull (Larus hyperboreus). The method was based on extraction of target compounds from homogenised samples into the solvent mixture used as mobile phase in high-performance liquid chromatography (HPLC), i.e. methanol/water (50:50; 2mM ammonium acetate). The extract was filtered and directly injected into a HPLC/time-of-flight mass spectrometry (TOF-MS) system. Quantification was performed using 7H-perfluoroheptanoic acid as internal standard and a calibration standard solution dissolved in sample extract for each matrix type (matrix-matched calibration standard). The method is very time and cost efficient. Except for long-chain compounds and perfluorooctane sulfonamide (which cannot be covered by this method), recoveries were between 60% and 115% and method detection limits were in the range 0.04–1.3ng/g wet weight. Blank values could be neglected with the exception of perfluorooctane sulfonate (PFOS), perfluorohexanoic acid (PFHxA) and perfluorooctanoic acid (PFOA). One of the major challenges in PFAS analysis is ionisation disturbance by co-eluting matrix in the ion source of the mass spectrometer. Both matrix and analyte specific signal enhancement and suppression was observed and quantified. Repeated extractions (n =3) gave relative standard deviations (RSD) <35% for all PFAS. Accuracy was examined by comparing the screening method to the generally applied ion pair extraction (IPE) method. PFAS concentration values of a glaucous gull liver sample deviated by less than 30% for the two methods, provided that matrix-matched calibration standards were employed in both methods. Abstract: Interactive liquid chromatography (iLC) for polymer analysis is usually applied to the characterisation of distributions other than molar mass. In particular, its use for the determination of chemical-composition, functionality-type and tacticity distributions has been demonstrated. The application of iLC for the determination of molar mass distributions (MMDs), however, has not yet been fully explored. An expanded version of the reversed-phase liquid chromatography model has been developed to describe and predict how the retention behaviour of polydisperse polystyrene samples changes with molar mass. The relationship between molar mass and the parameters of the model has been investigated in some detail and non-linear correlations were found. From the model and the relationships between the model parameters and molar mass, calibration curves (retention time versus molar mass) were constructed to predict changes in chromatographic selectivity across a given molar mass range. These calibration curves were compared to experimentally obtained curves and, in most cases, excellent agreement was found. The dramatic enhancement in selectivity that can be obtained with iLC in comparison to size-exclusion chromatography (SEC) was illustrated by measuring matrix-assisted laser desorption ionisation (MALDI) MS spectra of fractions collected during a gradient-LC separation. In the low-molar mass range, essentially monodisperse fractions were obtained. Calibration curves, predicted by the model and validated experimentally using narrow-dispersity standards and MALDI-MS spectra of fractions, were used to determine the molar mass distribution of some narrowly distributed polystyrene samples. Molar mass distributions for such standards were found to be somewhat lower than the values reported by the manufacturers. The results also deviated from those obtained by MALDI-MS. Abstract: The aim of this study was to evaluate the applicability of ECHO technique in pesticide residue analysis using LC/MS/MS instruments with atmospheric pressure chemical (APCI) and electrospray (ESI) ionization. The technique is based on simultaneous injections of reference standards and samples in one run. First and second injections are made ahead and behind a precolumn, respectively, thus resulting in a short difference of retention times between standard and sample peak. The obtained couple of peaks were applied to the easy detection of pesticides and simultaneous estimation of the residue content in real samples in a single run. If residues were not observed, the second sample peak did not occur and the ECHO peaks were used to demonstrate instrument performance in each run and for each analyte. Another advantage of ECHO technique is its potential to compensate matrix effects. The occurrence and compensation of matrix effects using APCI was tested with four matrix types (water containing, acidic, dry and sugar containing) and 22 pesticides. The same matrix types but 58 pesticides were used tests with electrospray ionization. Most often matrix effects had been observed with lemon. The percentage of pesticides showing significant matrix effects did not differ between APCI and ESI. But these effects caused signal enhancement in APCI measurements and signal suppression, when ESI was used. A commercially available evaporative light-scattering detection (ELSD) system was adapted for micro and capillary LC. Therefore the various parameters involved in the droplet formation during the nebulization step in the ELSD system were studied. It was shown that the velocity term in the Nukiyama Tanasawa equation remains constant, leading to droplets of the same order of magnitude for narrow bore and capillary columns. Consequently, the ELSD modification was performed by decreasing the internal diameter of the effluent capillary tube in the nebulizer nozzle and by keeping its external diameter constant. Next, response curves for a conventional and the developed micro and capillary LC were compared as to investigate why a linear ELSD response is often obtained when used in micro or capillary LC. By splitting the flow rate post column, we showed that the nebulization process was not at the origin of the phenomenon. For ceramide III and tripalmitin, the response curves were found to be non-linear. However the curvature was less significant when the columns internal diameter decreased. Calculated particle size profiles for micro or capillary LC suggest that the particle entering the detection chamber are bigger than under conventional LC conditions. Last, triethylamine and formic acid were used to increase the response of the detector. The emission of platinum from automobile catalytic exhaust converters is well known and the accumulation of Pt in road dust has been studied by the analysis of total Pt contents. However, there are few studies on the speciation of the emitted Pt-compounds in the environment. A separation method with HPLC-UV has been developed by Nachtigall et al. [Chromatogr. A 775 (1997) 197] with aqueous standards of Pt(II)- and Pt(IV)-chloro complexes. Due to the limited selectivity and sensitivity of the UV-detection this method is not suitable for Pt-speciation in road dust extracts. Therefore, in this study the method of Nachtigall et al. was modified to realize an on-line coupling to ICP-MS with the advantage of Pt-specific detection. Calibration was performed with aqueous standards and spiked road dust extracts. Calibration curves were linear with low residual standard deviation (1.3–3.1% for the aqueous standards) and approximately 10-times lower detection limits compared to the HPLC-UV-method. Moreover, the stability of the model species was investigated using isocratic as well as gradient separation. Isotopic ratios of 194Pt, 195Pt, 196Pt and 198Pt were determined for quality control. A new mathematical correction method for the HfO-interference on the 195Pt-signal was developed. Additionally, the elution pattern of further elements in the road dust extracts was monitored and total element contents were determined in unspiked road dust extracts. [Copyright &y& Elsevier]

For the faster evaluation of selectivity in reversed-phase liquid chromatography of siloxane-bonded silica columns using the solvation parameter model a minimal set of calibration compounds is described suitable for mobile phase composition from 20-70% (v/v) methanol-, acetonitrile-, or tetrahydrofuran-water. The Kennard-Stone uniform mapping algorithm is used to select the calibration compounds from a larger database of compounds with known retention properties used earlier for column selectivity evaluation. Thirty-five compounds are shown to be necessary to minimize the standard deviation of the system constants and to minimize the difference between the system constants determined by conventional calibration and the values obtained for the reduced calibration compounds. The models for SunFire C 18 with methanol-, acetonitrile- and tetrahydrofuran-water mobile phase compositions and XBridge Shield RP18, XBridge C 8 , XBridge Phenyl and Discovery HS F5 with methanol- and acetonitrile-water mobile phase compositions had an average coefficient of determination of 0.996 (standard deviation = 0.003, n = 11) and average standard error of the estimate 0.025 (standard deviation = 0.005, n = 11) for the reduced calibration compounds. Some octadecylsiloxane-bonded silica stationary phases with a high bonding density and methanol-water mobile phase compositions containing ≤ 30% (v/v) methanol exhibit extreme retention factors (log k > 2.5) for the low-polarity, two-ring aromatic compounds in the thirty-five compound calibration set. Alternative calibration compounds with more favorable retention properties are suggested as replacements in these cases. The predictive capability of the calibration models is validated using external test sets characterized by the average error, average absolute error and root mean square error of prediction. For the thirty-five calibration compounds sets the average absolute error 0.026 (standard deviation = 0.009, n = 11) and root mean square error of prediction 0.032 (standard deviation = 0.010, n = 11) confirm the suitability of the calibration models for column selectivity evaluation. System maps for XBridge Shield RP18 for 20-70% (v/v) methanol-water and Synergi Hydro-RP and 50% (v/v) methanol-water at temperatures from 25-65 °C together with a correlation diagram for XBridge Shield RP18 and SunFire C 18 are presented as representative applications of the reduced calibration compounds for column selectivity evaluation. Abstract: Matrix matching is used in analysis to compensate for matrix effects that influence analytical response. It has been a widely discussed topic in electro-spray mass spectrometry where the ionization suppression is a major problem in accurate quantitative analysis. However, the unique strength of mass spectrometry to detect and quantify accurately a co-eluting stable isotope labelled internal standard offers an easy solution to the ionization suppression problem. Given the fact that it is impossible to match the matrix of the calibration standards with all samples, mass spectrometry allows accurate quantitation without the need for matrix matching, as long as the internal standard co-elutes with the analyte of interest. If the analyte and internal standard co-elute, the slope of the calibration curve analyte response/internal standard vs. analyte concentration is independent of the matrix composition, eliminating the need for matrix matching.
