Abstract

A study of sensory rhodopsin I (SRI) homologue from the archaeon Haloarcula vallismortis (HvSRI) expressed as a recombinant protein which uses all-trans-retinal as a chromophore is presented. The identical structural changes observed in Haloarcula vallismortis (HvSRI) and, Halobacterium salinarum SRI (HsSRI) and Salinibacter ruber SRI (SrSRI) revealed that HvSRI could be very useful protein model to better understand the molecular mechanism of the dual photosensing by SRI.

Abstract

Attenuated total reflection Fourier-transform infrared (ATR-FTIR) spectroscopy was used to directly observe binding of Na(+) to carboxylates in the PomA-PomB complex, including the functionally essential residue Asp24. Based on the results obtained it was concluded that Asp24 and at least two other carboxylates constitute Na(+) interaction sites in the PomA-PomB complex.

Abstract

The characterization and cloning of Salinibacter sensory rhodopsin I (SrSRI), the first sensory rhodopsin I (SRI)-like protein identified in eubacteria, is reported. The comparison of the photochemical properties of the SrSRI-SrHtrI complex to SrSRI without SrHtrl via use of time-resolved laser flash spectroscopy and other spectroscopic techniques is also reported.

Abstract

Fourier transform infrared (FTIR) spectroscopy is used for studying sensory rhodopsin I from eubacterium Salinibacter ruber (SrSRI) and the spectral changes upon formation of the K and M intermediates. The structure and structural changes of SrSRI are discussed based on the infrared spectral comparisons with other rhodopsins.

Abstract

The synthesis and incorporation of several different monodeuterated all-trans retinal analogues into sensory rhodopsin II (SRII) are discussed. The steric constraint in the primary photoproduct of SRII is shown to be a prerequisite for light-signal transfer to the cognate transducer protein (HtrII).
